1. Comparison of bacterial diversity in Bactrocera cucurbitae (Coquillett) ovaries and eggs based on 16S rRNA sequencing

Abstract

Abstract Next-generation sequencing allows for fine-scale studies of microbial communities. Herein, 16S ribosomal RNA high-throughput sequencing was used to identify, classify, and predict the functions of the bacterial communities in the eggs and ovaries of Bactrocera cucurbitae (Coquillett) (Diptera: Tephritidae), which is a pest that infests a variety of cucurbit fruits at different developmental stages. Taxonomic analyses indicate that bacteria associated with B. cucurbitae represent 19 phyla, which were spread across different developmental stages. Specifically, the egg microbiota had a higher alpha diversity than those of microbiota in the primary and mature ovaries. Significant differences were not observed between the primary and mature ovaries in terms of their microbiota’s alpha diversities. Pseudomonadota, Deinococcota, Bacteroidota, Bacillota, and Actinomycetota were the dominant phyla in all three developmental stages of B. cucurbitae, and Pseudomonadaceae and Enterobacteriaceae were the most abundant families. Owing to the unique physiological environment of the ovaries, the diversity of their bacterial community was significantly lower than that in the eggs. This study provides new insights into the structure and abundance of the microbiota in B. cucurbitae at different developmental stages and contributes to forming management strategies for this pest.

Abstract

The red palm weevil, Rhynchophorus ferrugineus is one of the alien species of high risk in China and a key pest of palm trees. This paper presents the first status report of the damage caused to sugarcane by R. ferrugineus in China. The first observation of damage was recorded in Changling farm in Shangsi County, Guangxi Province, on a small plot in October 2017 and the recorded damage level, exceeded 80% with cane stalks completely destroyed. In September 2021, R. ferrugineus was found in sugarcane fields (11.85 hm2) in Liucheng County, in Liuzhou City, but the percentage of plant damaged was sporadic and didn't exceed 1%. The mean percent of damaged stalks and the length of damaged tunnels in plants recorded were 0.76%, and 19.54 cm, respectively when the canes were harvested. This discovery allowed us to prevent potential establishment and spread of the red palm weevil in the sugarcane regions in China, as well as serve a warning notice for other sugarcane regions worldwide. Abstract

Understanding the most effective host switch patterns in parasitic wasps, specifically Aenasius bambawalei (AB), is crucial for effectively controlling pests like Penacoccus solenopsis (PSS). This study aims to elucidate AB's ideal host switch pattern and assess its utility in maintaining synchronization between AB and PSS, thereby aiding in PSS control. We examined various host switch patterns and cycles to evaluate their impact on AB's offspring's parasitism rates and fitness in laboratory conditions. Concurrently, we assessed the fitness of both PSS and AB on tomato plants using different banker plant systems to maintain field synchronization. Results indicate that the three-repeat T1 host switch pattern of PSS-Penacoccus solani (PSI)-PSS was the most effective. Additionally, a specific banker plant system, "System B", which provided succulent plants hosting PSI, was optimal for synchronizing AB and PSS in a summer greenhouse setting. Our findings underscore the importance of employing specific host switch patterns and banker plant systems to effectively control PSS in the field. Abstract

Acute lung injury (ALI) is one of the representative "lung heat syndromes" in traditional Chinese medicine (TCM). Scutellaria baicalensis is an herbal medicine used in TCM for treating lung diseases, due to its remarkable anti-inflammatory and antiviral effects. When used in TCM, S. baicalensis root is divided into two categories: S. baicalensis pith-not-decayed root (SN) and S. baicalensis pith-decayed root (SD). Compared to SN, SD has a better effect on lung diseases. We constructed a lipopolysaccharide (LPS)-induced acute lung injury (ALI) mouse model to study the pharmacodynamic mechanism of SD. The ethanolic extract of Scutellaria baicalensis pith-decayed root (EESD) significantly affected LPS-induced ALI by reducing alveolar interstitial thickening, pulmonary edema, and other pathological symptoms, decreasing the infiltration of inflammatory cells, especially macrophages, and inhibiting IL-1 β , TNF- α , and IL-6 transcription and translation. Furthermore, in the THP-1 macrophage model induced by LPS, EESD inhibited the expression of phosphorylated nuclear factor inhibitory protein alpha (p-I κ B α), phosphorylated nuclear factor- κ B P65 (p-p65), cleaved-caspase-1, cleaved-IL-1 β protein, and the release of inflammatory factors in the NF- κ B/NLRP3 pathway, inhibiting macrophage function. In vivo experiments yielded similar results. Therefore, the present study clarified the potential of EESD in the treatment of ALI and revealed its potential pharmacodynamic mechanism by inhibiting the NF- κ B/NLRP3 inflammasome pathway and suppressing the pro-inflammatory phenotype activation of lung tissue macrophages. Abstract

Background The oriental fruit fly Bactrocera dorsalis is one of the most destructive pests of fruits and vegetables. The sterile insect technique (SIT) is an effective and environmentally friendly approach to the control of tephritid fruit flies. The pro-apoptotic gene head involution defective (hid) has been used as an effective lethal effector in SIT. It initiates an interaction cascade including activation of caspase-like proteases. However, the biological role of caspase activity in tephritid fruit flies has yet to be explored. Results In this study, the B. dorsalis caspase-1 gene (Bdcp-1) was cloned and characterized. Sequence comparison showed that Bdcp-1 protein shared highly homology with Drosophila effector caspases Drice and Dcp-1. It is predicted to contain a short pro-domain because two proteolytic cleavage sites (Asp16 and Asp223 ) are present. Expression patterns indicated that Bdcp-1 is highly transcribed in embryos and expression was upregulated during metamorphosis and upon ultraviolet irradiation. RNA interference showed that Bdcp-1 is essential for ovarian development and female fertility. For example, knockdown of Bdcp-1 caused transcriptional downregulation of expression of the yolk protein-1 gene (Bdyp-1) and delayed ovarian development. The percentage of spawning females and female fecundity were significantly reduced. Conclusion This study illustrates the function of the Bdcp-1 gene and provides an attractive method to develop a biological way to control the oriental fruit fly through the control of caspases. © 2020 Society of Chemical Industry.
