17. Cauliflower-shaped Pleurotus ostreatuscultivated in an atmosphere with high environmental carbon dioxide concentration

Abstract

ABSTRACTCommercial aspects, physiological properties, and nutritional characteristics of Pleurotus ostreatusgrown under various environmental carbon dioxide concentration ([CO2]e) conditions were assessed. As [CO2]eincreased, the activity of antioxidant enzymes (catalase, peroxidase, and superoxide dismutase) in fruiting body increased, activities of succinate dehydrogenase and cytochrome coxidase were inhibited, and malondialdehyde and adenosine triphosphate (ATP) syntheses were reduced, leading to incomplete development of pilei and stipes, or even absence of pilei. Under high [CO2]e(≥1.00%), fruiting body of P. ostreatuswas morphologically altered to assume cauliflower shape. This cultivation condition resulted in high total contents of crude protein, crude fiber, and amino acids, increased levels of umami- and sweet-tasting amino acids, and reduced levels of bitter-tasting amino acids, thus enhancing the flavor of the product. In conclusion, a novel “cauliflower-shaped” mushroom (P. ostreatus) was successfully cultivated at high (≥1.00%) environmental CO2concentration. The product has a delicious taste and high nutritional value, is relatively easy to transport and store, and has excellent potential for commercial development.

Abstract

An evaluation of genetic variation and genetic structure of 17 wild populations of Callicarpa kwangtungensis in China were studied using morphological traits, biochemical and ISSR markers. The 17 accessions of C. kwangtungensis genotypes exhibited considerable variation for morphological characters, which plant height, ground diameter, leaf length, leaf width and leaf area were the main traits according to the principal coordinate analysis (PCA). High performance liquid chromatography (HPLC) results show that poliumoside, forsythiaside B are higher than the criterion standard for Callicarpae caulis et folium in Chinese Pharmacopoeia. ISSR results revealed that the 20 selected primers generated a total of 217 clear and distinguishable bands fragments, 187 bands (85.69%) of which were polymorphic. There was a high level of inter-population genetic variability, relatively low genetic diversity within populations, which indicated the same trend by Gst (0.438). This was also confirmed by the limited gene flow among populations (Nm 0.642), which indicated the difficulty in genetic exchange among populations. Mantel test revealed a low association between the ISSR data and geographic position (r = 0.390), ISSR and morphological traits (0.283), and a weak correlation between ISSR and content of phytochemicals (0.078). Finally, a preliminary proposal for conservation and breeding of this germplasm was put forward based on these findings.

Abstract

The electrochemical characterization of boron-doped polycrystalline diamond thin-film (BDF) electrodes was studied using the anodic scan after concentrating lead in 0.1 mol/L KCl - 41 micromol/L Hg(NO(3))(2) and 0.1 mol/L KNO(3) - 0.01 mol/L HNO(3) - 41 micromol/L Hg(NO(3))(2); accumulation voltage was -0.90 V. The results obtained were compared with those given by glassy carbon (GC) electrodes and proved that the BDF electrodes offered high sensitivity, good precision and extreme stability over a 2-month period. These electrodes provided good resolving power for the determination of lead and cadmium and gave satisfactory results in the analysis of a pure water sample.
