16. Radiogenomics in Hepatocellular Carcinoma: Machine Learning Based CT Texture Analysis in Predicting β-Catenin Mutation Status

Abstract

OBJECTIVE. The purpose of this study is to evaluate the potential value of CT radiomics in predicting the mutation status of β-catenin in patients with hepatic cell cancer (HCC).MATERIALS AND METHODS.In this retrospective study, 43 patients with hepatic cell HCC (18 without β-catenin mutation and 15 with β-catenin mutation) were identified in The Cancer Genome Atlas–hepatic liver Cell Carcinoma database (TCGA-LIHC). To create stable models, the data were augmented to a total of 202 labeled samples (131 without β-catenin mutation and 73 with β-catenin mutation) by obtaining up to five different samples per patient. Extraction of large amounts of image features from portal phase contrast-enhanced CT images had been performed on an open-source software package (Pyradiomics, version 2.1.2.). Reproducibility analysis (intraclass correlation, run ICCs in SPSS 18.0) was performed by two radiologists. Classification problem is about β-catenin gene mutation status. Machine Learning based classifications were performed using the Pycaret (version 2.1.2) software. The main performance metric was the AUC value.RESULTS. Of 828 extracted texture features, 759 had excellent reproducibility. Using 10 selected features, the Extra Trees Classifier algorithm correctly classified 93.4% of the HCCs in terms of β-catenin mutation status (AUC value, 0.9741); the CatBoost Classifier algorithm correctly classified 91.9% of the HCCs (AUC value, 0.9692); Gradient Boosting Classifier algorithm correctly classified 91.1% ( AUC value, 0.9722). All the three advanced algorithms performed above 90% accuracy.CONCLUSION. Machine Learning-based high-dimensional quantitative CT radiomics analysis might be a feasible and potential method for predicting β-catenin mutation status in patients with HCC.

18. Prediction of KRAS, NRAS and BRAF status in colorectal cancer patients with liver metastasis using a deep artificial neural network based on radiomics and semantic features

Abstract

There is a critical need for development of improved methods capable of accurately predicting the RAS (KRAS and NRAS) and BRAF gene mutation status in patients with advanced colorectal cancer (CRC). The purpose of this study was to investigate whether radiomics and/or semantic features could improve the detection accuracy of RAS/BRAF gene mutation status in patients with colorectal liver metastasis (CRLM). In this retrospective study, 159 patients who had been diagnosed with CRLM in two hospitals were enrolled. All patients received lung and abdominal contrast-enhanced CT (CECT) scans prior to radiation therapy and chemotherapy. Semantic features were independently assessed by two radiologists. Radiomics features were extracted from the portal venous phase (PVP) of the CT scan for each patient. Seven machine learning algorithms were used to establish three scores based on the semantic, radiomics and the combination of both features. Two semantic and 851 radiomics features were used to predict the mutation status of RAS and BRAF using an artificial neural network method (ANN). This approach performed best out of the seven tested algorithms. We constructed three scores which were based on radiomics, semantic features and the combined scores. The combined score could distinguish between wild-type and mutant patients with an AUC of 0.95 in the primary cohort and 0.79 in the validation cohort. This study proved that the application of radiomics together with semantic features can improve non-invasive assessment of the gene mutation status of RAS (KRAS and NRAS) and BRAF in CRLM.

25. Inhibition of Jak-STAT3 pathway enhances bufalin-induced apoptosis in colon cancer SW620 cells

Abstract

Abstract Background The purpose of the research is to investigate the roles of Jak-STAT3 signaling pathway in bufalin-induced apoptosis in colon cancer SW620 cells. Methods The inhibitory effects of bufalin on cell proliferation were determined by MTT (Methyl thiazolyltetrazolium) assay. The morphological changes of cells were measured by Wright-Giemsa staining. The cell cycle arrest and apoptosis were tested by flow cytometry analysis. Western Blot was used to determine the protein expression of the apoptosis inhibitors livin and caspase-3, the apoptosis-related proteins Bax and Bcl-2, as well as the key protein kinases in the Jak-stat3 signaling pathway, stat3 and p-stat3. Results (1) Bufalin inhibited the proliferation of SW620 cells. IC50 at 24 h, 48 h and 72 h were 76.72 ± 6.21 nmol/L, 34.05 ± 4.21 nmol/L and 16.7 ± 6.37 nmol/L. (2) Bufalin induced SW620 cell cycle arrest and apoptosis, indicated by the appearance of apoptotic bodies; (3) The results from flow cytometry demonstrated that there was cell cycle G2/M phase arrest in 20 nmol/L bufalin treatment group (36.29 ± 2.11% vs 18.39 ± 1.74%, P Conclusions Bufalin not only inhibited the growth of colon cancer SW620 cells, but also induced apoptosis of SW620 cells. Activation of caspase-3, up-regulation of Bax, down-regulation of livin and Bcl-2, as well as inhibition of Jak-stat3 signaling pathway might be the important mechanisms for the bufalin-induced apoptosis.

Abstract

Introduction Erythropoietin (EPO) has been shown to have beneficial effects on peripheral nerve damage, but its mechanism of action remains incompletely understood. In this study we hypothesized that EPO promotes peripheral nerve repair via neurotrophic factor upregulation. Material and methods Thirty adult male Wistar rats were employed to establish a sciatic nerve injury model. They were then randomly divided into two groups to be subjected to different treatment: 0.9% saline (group A) and 5000 U/kg EPO (group B). The walking behavior of rats was evaluated by footprint analysis, and the nerve regeneration was assessed by electron microscopy. The expression of insulin-like growth factor-1 (IGF-1) in the injured sciatic nerves was detected by immunohistochemical analysis. Results Compared to saline treatment, EPO treatment led to the growth of myelin sheath, the recovery of normal morphology of axons and Schwann cells, and higher density of myelinated nerve fibers. Erythropoietin treatment promoted the recovery of SFI in the injured sciatic nerves. In addition, EPO treatment led to increased IGF-1 expression in the injured sciatic nerves. Conclusions Erythropoietin may promote peripheral nerve repair in a rat model of sciatic nerve injury through the upregulation of IGF-1 expression. These findings reveal a novel mechanism underlying the neurotrophic effects of EPO.
