The linear variable differential transformer is a key component for measuring vibration noise and active vibration isolation. The nonlinear output associated with increased differential displacement in LVDT constrains the measurement range. To extend the measurement range, this paper proposes an advanced Snake Optimization–Tangential Functional Link Artificial Neural Network (ASO-TFLANN) model to extend the linear range of LVDT. First, the Latin hypercube sampling method and the Levy flight method are introduced into the snake optimization (SO) algorithm, which enhances the global search ability and diversity preservation ability of the SO algorithm and effectively solves the common overfitting and local optimal problems in the training process of the gradient descent method. Second, a voltage–displacement test bench is constructed, collecting the input and output data of the LVDT under four different main excitation conditions. Then, the collected input and output data are fed into the ASO-TFLANN model to determine the optimal weight vectors of the tangential functional link Artificial Neural Network (TFLANN). Finally, by comparing with the simulation experiments of several algorithms, it is proven that the ASO proposed in this paper effectively solves the common overfitting and local optimization problems in the training process of the gradient descent method. On this basis, through offline simulation comparison experiments and online tests, it is proven that the method effectively reduces ϵfs while expanding the linear range of LVDT and significantly improves the measurement range, which provides a reliable basis for improving measurement range and accuracy.

Abstract Background The basic helix-loop-helix (bHLH) gene family is one of plants’ largest transcription factor families. It plays an important role in regulating plant growth and abiotic stress response. Results In this study, we determined that the PavbHLH28 gene participated in cold resistance. The PavbHLH28 gene was located in the nucleus and could be induced by low temperature. Under the treatment of ABA, PEG, and GA3, the transcript level of PavbHLH28 was affected. At low temperature, overexpression of the PavbHLH28 gene enhanced the cold resistance of plants with higher proline content, lower electrolyte leakage (EL) and malondialdehyde (MDA) content. Compared with the WT plants, the transgenic plants accumulated fewer reactive oxygen species (ROS), and the activity and expression levels of antioxidant enzymes were significantly increased. The expression of proline synthesis enzyme genes was up-regulated, and the transcripts levels of degradation genes were significantly down-regulated. The transcripts abundance of the cold stressed-related genes in the C-repeat binding factor (CBF) pathway was not significantly different between WT plants and transgenic plants after cold stress. Moreover, the PavbHLH28 could directly bind to the POD2 gene promoter and promote its gene expression. Conclusions Overall, PavbHLH28 enhanced the cold resistance of transgenic plants through a CBF-independent pathway, which may be partly related to ROS scavenging.

Small auxin up RNA (SAUR) is a large gene family that is widely distributed among land plants. In this study, a comprehensive analysis of the SAUR family was performed in sweet cherry, and the potential biological functions of PavSAUR55 were identified using the method of genetic transformation. The sweet cherry genome encodes 86 SAUR members, the majority of which are intron-less. These genes appear to be divided into seven subfamilies through evolution. Gene duplication events indicate that fragment duplication and tandem duplication events occurred in the sweet cherry. Most of the members mainly underwent purification selection pressure during evolution. During fruit development, the expression levels of PavSAUR16/45/56/63 were up-regulated, and conversely, those of PavSAUR12/61 were down-regulated. Due to the significantly differential expressions of PavSAUR13/16/55/61 during the fruitlet abscission process, they might be the candidate genes involved in the regulation of physiological fruit abscission in sweet cherry. Overexpression of PavSAUR55 in Arabidopsis produced earlier reproductive growth, root elongation, and delayed petal abscission. In addition, this gene did not cause any change in the germination time of seeds and was able to increase the number of lateral roots under abscisic acid (ABA) treatment. The identified SAURs of sweet cherry play a crucial role in fruitlet abscission and will facilitate future insights into the mechanism underlying the heavy fruitlet abscission that can occur in this fruit crop.

Background: Enabling community-led health initiatives will contribute to reducing the burdens on the healthcare system. Implementing such initiatives successfully in high and upper-middle income Asian countries is poorly understood and documented. We undertook a Rapid Review, systematically synthesising the evidence to develop implementation guidelines to address this gap. Methods: Eligible studies focused on community movements or affiliated constructs in upper-middle and high-income Asian countries, conducted between 2014 and 2021. Studies were sought from either electronic databases – Cochrane and Campbell Collaboration, PubMed, Embase, CINAHL, SCOPUS, APA Psycinfo, Web of Science, Google Scholar – or recommendation from experts. Extraction was undertaken according to mid-level programme goals, termed Intermediate Results. These were conceptualized by a cross-disciplinary team and iteratively reworked as analysis progressed. Framework analysis was undertaken and structured according to the IRs. 28 studies (9 mixed methods, 9 quantitative, 7 qualitative and 3 case studies) were included and synthesised. Results: The MovEMENTs checklist and related strategies were elicited through the review. The six Intermediate Results include to: (1) Move the community to be recruited and retained (2) Engage capacity and build capability; (3) Maintain emotional resonance; (4) Embed participatory approaches; (5) Nurture network building and partnerships; (6) Team up to improve commissioning and funding structures. Sixteen strategies and related implementation guidelines underpinning the Intermediate Results are extracted from the evidence-base of included studies. Conclusion: The MovEMENTs for Health checklist is developed to serve as a guide for implementers and proposed to be adaptable to various contexts. Abstract Background The shedding of premature sweet cherry (Prunus avium L) fruitlet has significantly impacted production, which in turn has a consequential effect on economic benefits. Result To better understand the molecular mechanism of sweet cherry fruitlet abscission, pollen viability and structure had been observed from the pollination trees. Subsequently, the morphological characters of the shedding fruitlet, the plant hormone titers of dropping carpopodium, the transcriptome of the abscising carpopodium, as well as the HD-ZIP gene family were investigated. These findings showed that the pollens giving rise to heavy fruitlet abscission were malformed in structure, and their viability was lower than the average level. The abscising fruitlet and carpopodium were characterized in red color, and embryos of abscising fruitlet were aborted, which was highly ascribed to the low pollen viability and malformation. Transcriptome analysis showed 6462 were significantly differentially expressed, of which 2456 genes were up-regulated and 4006 down-regulated in the abscising carpopodium. Among these genes, the auxin biosynthesis and signal transduction genes (α-Trp, AUX1), were down-regulated, while the 1-aminocyclopropane-1-carboxylate oxidase gene (ACO) affected in ethylene biosynthesis, was up-regulated in abscising carpopodium. About genes related to cell wall remodeling (CEL, PAL, PG EXP, XTH), were up-regulated in carpopodium abscission, which reflecting the key roles in regulating the abscission process. The results of transcriptome analysis considerably conformed with those of proteome analysis as documented previously. In comparison with those of the retention fruitlet, the auxin contents in abscising carpopodium were significantly low, which presumably increased the ethylene sensitivity of the abscission zone, conversely, the abscisic acid (ABA) accumulation was considerably higher in abscising carpopodium. Furthermore, the ratio of (TZ + IAA + GA3) / ABA also obviously lower in abscising carpopodium. Besides, the HD-ZIP gene family analysis showed that PavHB16 and PavHB18 were up-regulated in abscising organs. Conclusion Our findings combine morphology, cytology and transcriptional regulation to reveal the molecular mechanism of sweet cherry fruitlet abscission. It provides a new perspective for further study of plant organ shedding.

Cherries are one of the important fruit trees. The growth of cherry is greatly affected by abiotic stresses such as drought, which hinders its development. Chalcone synthase (CHS, EC 2.3.1.74) is a crucial rate-limiting enzyme in the flavonoid biosynthetic pathway that plays an important role in regulating plant growth, development, and abiotic stress tolerance. In the current study, three genes encoding chalcone synthase were identified in the genome of sweet cherry (Prunus avium L.). The three genes contained fewer introns and showed high homology with CHS genes of other Rosaceae members. All members are predicted to localize in the cytoplasm. The conserved catalytic sites may be located at the Cys163, Phe214, His302, and Asn335 residues. These genes were differentially expressed during flower bud dormancy and fruit development. The total flavonoid content of Chinese cherry (Cerasus pseudocerasus Lindl.) was highest in the leaves and slightly higher in the pulp than in the peel. No significant difference in total flavonoid content was detected between aborted kernels and normally developing kernels. Overexpression of Chinese cherry CpCHS1 in tobacco improved the germination frequency of tobacco seeds under drought stress, and the fresh weight of transgenic seedlings under drought stress was higher than that of the wild type, and the contents of SOD, POD, CAT, and Pro in OE lines were significantly increased and higher than WT under drought stress. These results indicate cherry CHS genes are conserved and functionally diverse and will assist in elucidating the functions of flavonoid synthesis pathways in cherry and other Rosaceae species under drought stress.
