Celangulin V is a natural

The discovery of new highly active molecules from natural products is a common method to create new pesticides. Celangulin V targeting Mythimna separate (M. separate) midgut V-ATPase H subunit, has received considerable attention for its excellent insecticidal activity and unique mechanism of action. Therefore, combined with our preliminary work, thirty-seven sulfonamide derivatives bearing propargyloxy or pyridine groups were systematically synthesized to search for insecticidal candidate compounds with low cost and high efficiency on the H subunit of V-ATPase. Bioactive results showed that compounds A2-A4 and A6-A7 exhibited a better bioactivity with median effective concentration (LC50) values (2.78, 3.11, 3.34, 3.54 and 2.48 mg/mL, respectively) against third-instar larvae of M. separate than Celangulin V (LC50 = 18.1 mg/mL). Additionally, molecular docking experiments indicated that these molecules may act on the H subunit of V-ATPase. Based on the above results, these compounds provide new ideas for the discovery of insecticides.

In our previous studies, a kind of novel benzenesulfonamides was found to be a candidate insecticidal compounds. It was shown that propargyloxy and sulfonamide groups are pharmacodynamic groups. One hundred and twenty-six (126) naphthalenesulfonamides derivatives with propargyloxy functionality were designed and synthesized, and their insecticidal activities were determined. Some of them showed outstanding activity, with LC50 values as low as 0.202 mg ml-1, much lower than that of the positive control celangulin V (23.9 mg ml-1). In addition, the structure-activity relationships were discussed, and molecular docking was used to verify the binding mode of the compound and the target receptor.

Steering fracturing is the key technology applied to stimulate and enhance oil recovery in tight sandstone reservoirs. In this paper, based on the integration of theoretical background of the porous media fluid mechanics, rock mechanics, and thermodynamics, a heat-fluid-solid three-field coupling stress model is established to simulate the steering fracturing, considering the adjacent well production and rock deformation. To solve the coupling flow equations and coupling temperature field equations, the finite difference method is adopted, while the finite element method is applied to solve the rock deformation equations and provide an explicit alternative approach to the whole model. The model is used to predict stress variations during production/injection processes and multistage artificial fracturing based on the heat-fluid-solid three-field coupling effect. The impact of multiple-field induced stresses on steering fracturing in wells containing vertical fractures can be quantitatively analyzed and simulated accordingly. Finally, a case study is performed to verify the model according to the characteristics of fiber steering fracturing, failure pressure distribution around the well-bore, characteristics analysis of G-function, and interpretation results of microseismic monitoring.

Background After storing blood for a period of time, the structure and properties of the red blood cells (RBC) will change, which results in a decrease in the oxygen-carrying capacity, and further has a certain impact on their exosomes. Objectives Effective oxygen uptake (Q), P50, 2,3-DPG, and Na+-K+-ATP of RBC after different storage times were detected. Electron microscopy was used to observe the morphology of RBC and the characteristics of secreting exosomes. Western blot was used to detect the expression of phenotypes CD63 and CD81 of exosomes, and the expression of mitochondrial riboprotein MRPS35 of exosomes was also detected to explore the mechanism of decreased function of RBC with the extension of preservation time. Material and methods After the RBC suspension was prepared, the effective oxygen-carrying capacity (Q) and P50, as well as 2,3-DPG and Na+-K+-ATP were prepared. This was followed by morphology observation of erythrocyte exosomes using transmission electron microscope (TEM), and by western blot analysis of exosome phenotypes CD63 and CD81. Results Erythrocytes secrete exosomes, which results in abnormal expression of related proteins in mitochondria. This leads to increased ROS production, mitochondrial apoptosis and, finally, changes in or damage to erythrocytes. Conclusions Changes in the rheological properties and oxygen-carrying functions of erythrocytes during preservation are all observable manifestations, and underlying these manifestations are mechanisms of damage to erythrocytes at a molecular level. Erythrocytes secrete exosomes, which results in abnormal expression of related proteins in mitochondria, increasing ROS production, mitochondrial apoptosis and, finally, changes or damage to erythrocytes.

ContextGlucocorticoids have potent effects on the central nervous system. However, while patients with Cushing syndrome frequently report impairments in cognitive function, studies investigating cognitive function in patients with autonomous cortisol secretion (ACS) in adrenal incidentalomas (AIs) are scarce.ObjectiveThe aim of the present study was to evaluate neurocognitive function in patients with ACS.MethodsWe prospectively recruited 63 patients with AI, 36 patients with nonfunctional adrenal adenoma (NFA) (46.5 ± 10.5 years), and 27 patients with ACS (48.6 ± 9.1 years); these patients underwent a battery of validated neuropsychological tests. ACS was diagnosed when serum cortisol levels after a 1-mg dexamethasone suppression test (cortisol1 mg DST) ≥ 50 nmol/L.ResultsPatients with ACS had higher frequency of subjective memory complaints (40.7% vs 13.9%, P < 0.05) and higher proportion of mild cognitive impairment (22.2% vs 2.8%, P < 0.05) than patients with NFA. Furthermore, patients with ACS had worse performance on working memory and the visuospatial/constructional domain than patients with NFA (all P < 0.05). Serum cortisol1 mg DST was negatively correlated with working memory and visuospatial/constructional domains (r = −0.307 and −0.306, respectively, all P < 0.05). Performance on working memory and visuospatial/constructional domains gradually deteriorated with increases in serum cortisol1 mg DST (all P values for trend < 0.05). Multivariate linear regression analysis showed that serum cortisol1 mg DST was a significant risk factor for impairment of working memory and visuospatial/constructional domains (B = −0.006 and −0.043, respectively, all P < 0.05).ConclusionThis study is the first to report that ACS is accompanied by impaired cognitive function. Consequently, cognitive function assessment should be incorporated into the clinical evaluation of patients with ACS.Clinical trial registration numberNCT05357456.

A novel palladium-catalyzed three-component reaction of 3-iodochromones with bridged olefins and aziridine is described, which employs a [2 + 2 + 1] domino annulation strategy to produce chromone fused methylenecyclopentanes in moderate to good yields via a Heck coupling/C(sp2)–H activation/imine–enamine tautomerization/retro-aza-Michael addition sequence. Significantly, aziridine was used as a vinylidene unit by cleavage of two C–N bonds and was successfully introduced into the products for the first time.

Background Red blood cell (RBC) transfusion is associated with systemic inflammation and immune suppression as adverse outcomes. Objectives To investigate the immunomodulatory function of the transfused autologous RBC in altering pro-inflammatory and immunosuppressive effects. Material and methods A total of 24 Sprague Dawley male rats were randomly divided into 3 groups (n = 8 in each group). Group 1 did not receive blood transfusions, while the other 2 groups of rats separately received transfusion of RBC stored for 14 days (group 2) and 35 days (group 3). The rats were treated with HO-1 inhibitor, HO-1 inducer and nuclear factor erythroid 2-related factor 2 (Nrf2) activator after they separately received autologous transfusion of RBC that were cryopreserved for 14 days or 35 days. The blood samples of the rats were collected 12 h after the transfusion, and the macrophage phenotype of M1 and M2 were analyzed with flow cytometry (FCM). Also, the surface protein expression of CD68 and CD200R in macrophages were analyzed and the inflammatory signals in the serum were measured with enzyme-linked immunosorbent assay (ELISA). Moreover, the location and expression of proteins heme oxygenase 1 (HO-1), arginine 1 (Arg-1) and nitric oxide synthase 2 (NOS2) in macrophage were detected with immunofluorescence (IF). Results Autologous transfusion of long-time stored ("old") RBC promoted macrophage polarization to M2 phenotype and upregulated the expression of its surface proteins CD68 and CD200R. The pro-inflammatory cytokines tumor necrosis factor α (TNF-α), interleukin (IL)-6, IL-1β, and IL-18 were inhibited, and the secretion of NOS isoforms (iNOS) in serum was reduced with blood transfusion; contrarily, the production of IL-10 and CCL22 was increased. Additionally, HO-1, Arg-1 and NOS2 proteins were located in the cytoplasm, and HO-1 and Arg-1 proteins were highly expressed in macrophage, while the expression of protein NOS2 was low. Moreover, Nrf2, HO-1 and Arg-1 proteins were upregulated in macrophage after receiving "old" RBC transfusion. Conclusions Autologous transfusion of "old" RBC drove the macrophage phenotype toward M2 macrophages and induced immunosuppressive effects through the IL-10-NRF2-HO-1 signals.

Disinfection is essential for the microbial safety of reclaimed water. Traditional chlorine disinfection leads to secondary problems such as disinfection by-products and chlorine-resistant bacteria. Ferrate (Fe(VI)) is a novel green disinfectant. However, research on the disinfection characteristics of Fe(VI) remains insufficient. This study compared the disinfection efficacy between Fe(VI) and chlorine in secondary effluent, including the inactivation efficiency of coliforms and heterotrophic bacteria and the control effect on typical chlorine-resistant bacteria. The results showed that Fe(VI) was more effective than chlorine in inactivating Escherichia coli and total coliforms at low doses, whereas chlorine was more effective than Fe(VI) in inactivating heterotrophic bacteria. A severe trailing phenomenon was observed in Fe(VI) disinfection. Based on bacterial community structure analysis, Fe(VI) was also found to be capable of controlling the relative abundance of some chlorine-resistant bacteria such as Sphingomonas, Bacillus, Mycobacterium and Legionella except for Pseudomonas. The results of this study could have implications in evaluating Fe(VI) disinfection ability and optimizing Fe(VI) dosing for disinfection.

This study aimed to explore the molecular mechanisms underlying the differential quality of tea made from leaves at different development stages. Fresh Camellia sinensis (L.) O. Kuntze "Sichuan Colonial" leaves of various development stages, from buds to old leaves, were subjected to transcriptome sequencing and metabolome analysis, and the DESeq package was used for differential expression analysis, followed by functional enrichment analyses and protein interaction analysis. Target metabolome analysis indicated that the contents of most compounds, including theobromine and epicatechin gallate, were lowest in old leaves, and transcriptome analysis revealed that DEGs were significantly involved in extracellular regions and phenylpropanoid biosynthesis, photosynthesis-related pathways, and the oleuropein steroid biosynthesis pathway. Protein-protein interaction analysis identified LOC114256852 as a hub gene. Caffeine, theobromine, L-theanine, and catechins were the main metabolites of the tea leaves, and the contents of all four main metabolites were the lowest in old leaves. Phenylpropanoid biosynthesis, photosynthesis, and brassinosteroid biosynthesis may be important targets for breeding efforts to improve tea quality.

Background:The guidewire seldom breaks inside the coronary artery during percutaneous coronary intervention. Herein we report a rare case of spontaneous fracture of the guidewire when used for contralateral angiography, in which multiple percutaneous attempts at retrieval failed. Use of the twisting wire technique for this purpose led to coronary perforation and perfusion. The entrapped fragment was removed during emergency surgery to prevent further complications. This report also discusses potential causes of guidewire breakage and management strategies, and highlights the potential risk of the twisting wire technique when used for retrieving a fractured guidewire from the coronary artery.Case presentation: A guidewire that breaks inside the coronary artery during an intervention can cause immediate complications as well as adverse events over time. It must be removed. Retrieval methods to extract a guidewire fragment include deep engagement of the guiding catheter, or techniques such as the loop snare or twisting wire. The latter used to retrieve the fragment threatens coronary perforation, as the broken end of the guidewire is hard and sharp and can easily penetrate the coronary artery wall. In the present case, several pullback maneuvers using the twisting wire technique were performed to withdraw a guidewire fragment from the ostial right coronary artery (RCA), but coronary perforation occurred. When all attempts at percutaneous retrieval failed, surgical removal was necessary and successful. The patient had an uneventful recovery without signs or complications, as of the 3-month follow-up. Conclusions: We conclude that fracture of the guidewire remains a dangerous and unpredictable complication of percutaneous coronary intervention, and we especially highlight the potential risk of the twisting wire technique when used for retrieving the fractured wire from the coronary artery. Although the twisting wire technique is suitable to withdraw a fractured guidewire in selected cases, it must be used with great caution. For our patient, not only did the twisting wire technique fail to retrieve the broken guidewire fragment, but a most serious consequence was the penetration of the wall of the coronary artery.

Over the past few decades, the study of epidemic propagation has caught widespread attention from many areas. The field of graphs contains a wide body of research, yet only a few studies explore epidemic propagation’s dynamics in “signed” networks. Motivated by this problem, in this paper we propose a new epidemic propagation model for signed networks, denoted as S-SIS. To explain our analysis, we utilized the mean field theory to demonstrate the theoretical results. When we compare epidemic propagation through negative links to those only having positive links, we find that a higher proportion of infected nodes actually spreads at a relatively small infection rate. It is also found that when the infection rate is higher than a certain value, the overall spreading in a signed network begins showing signs of suppression. Finally, in order to verify our findings, we apply the S-SIS model on Erdös–Rényi random network and scale-free network, and the simulation results is well consist with the theoretical analysis.

Background. Excessive endoplasmic reticulum (ER) stress in intestinal epithelial cells (IEC) may lead to impaired intestinal mucosal barrier function and then participate in the pathogenesis of ulcerative colitis (UC). Jianpi Qingchang decoction (JPQCD) has been shown to have protective effects on UC. However, further studies are needed to determine whether JPQCD regulates PERK/eIF2α/ATF4/CHOP pathways to play a role in treating UC. Methods. IL-10−/− mice were randomly assigned into five groups: control, model, low-dose JPQCD (JPQCD L), middle-dose JPQCD (JPQCD M), and high-dose JPQCD (JPQCD H). All groups except for the control group were given model feed containing 200 ppm piroxicam for 10 d to induce colitis. As a comparison, we used wild-type mice that were the progeny of IL-10+/− matings, bred in the same facility. The control group and wild-type mice were fed with common feed. At the same time, mice in each group were given corresponding drugs by gavage for 14 d. The disease activity index of mice in each group was evaluated daily. Colon tissues of mice were collected, colon length was measured, and pathological changes and ultrastructure of colon epithelial cells were observed. The effects of JPQCD on the PERK/eIF2α/ATF4/CHOP pathways were evaluated by western blotting and reverse transcription-polymerase chain reaction (RT-PCR). The expression of CHOP in colon tissue was detected by tissue immunofluorescence assay. The expression of NF-κB, p-NF-κB p65 protein was analyzed by western blotting; the level of IL-17 in colon tissue was detected by enzyme-linked immunosorbent assay (ELISA) and verified by examining NF-κB and IL-17 mRNA levels by RT-PCR. Results. Compared with the control group, the model group showed significant colitis symptoms and severe colonic tissue damage. The results showed that JPQCD significantly reduced body weight loss, ameliorated disease activity index, and restored colon length in IL-10−/− mice with piroxicam-induced colitis. Western blotting and RT-PCR showed that the PERK/eIF2α/ATF4/CHOP pathway was activated in colon tissue of model mice, suggesting that the pathway is involved in the pathogenesis of ulcerative colitis (UC) and could become a potential therapeutic target. The JPQCD treatment inhibited the activation of the PERK/eIF2α/ATF4/CHOP pathway, alleviated the ER stress, and played a role in preventing and treating UC. In addition, JPQCD can also downregulate the protein of NF-κB, p-NF-κB p65, downregulate the mRNA expression of NF-κB, and reduce the content of IL-17 and its mRNA expression in colon tissues. Conclusion. JPQCD may play a protective role in UC by regulating the PERK/eIF2α/ATF4/CHOP signaling pathway and relieving endoplasmic reticulum stress.

Adsorption and coagulation were commonly used to alleviate reverse osmosis (RO) membrane fouling caused by dissolved organic matters (DOM), but the effects of changed composition and structure of DOM in dyeing wastewater after adsorption and coagulation on RO membrane fouling have seldom been studied. This study aimed at resolving the mechanism how the RO membrane fouling during dyeing wastewater treatment was alleviated by using adsorption and coagulation. The dyeing wastewater caused serious RO membrane fouling. Pretreatment with granular activated carbon (GAC), polyferric sulfate (PFS) and polyaluminum chloride (PACl) were conducted. It was shown that GAC could remove most of the DOM (95%) and preferred to adsorb protein, hydrophobic neutrals and fluorescent compounds. Both coagulants of PFS and PACl preferred to remove polysaccharides (the removal rate was 9-19% higher than that of DOM), high-MW compounds and these compounds with high fouling potential. Afterwards, the RO membrane fouling potential of the dyeing wastewater was tested. The GAC and PFS performed well to alleviate fouling. After GAC treatment, the decline rate of RO flux was similar to that of raw wastewater after 6-fold dilution. With pretreatment by PFS or PACl, the fouling potential of dyeing wastewater was much lower than that of raw wastewater after diluted to the same DOM content. Changes in polysaccharides content in the DOM had more effects on RO membrane fouling than that of proteins after these pretreatment. Although the DOM changed significantly after pretreatment, the fouling type was still intermediate blocking.

Dissolved organic matter(DOM) is an important component of the organic matter in the environment. This study explored the characteristics of DOM. The components and spectral characteristics of rainwater DOM were measured using Ultraviolet-visible absorption spectrum(UV-vis) and three-dimensional fluorescence spectrum in the Hebei province. The results showed significant differences for DOM molecular weight in the northern, central, and southern region; the DOM of the northern region exhibited the lowest molecular weight. Introduction.Malasseziafolliculitis (MF) and pityriasis versicolor (PV) are common dermatoses caused byMalasseziaspecies. Their molecular epidemiology, drug susceptibility and exoenzymes are rarely reported in China.Aim.To investigate the molecular epidemiology, drug susceptibility and enzymatic profile ofMalasseziaclinical isolates.Methodology.Malasseziastrains were recovered from MF and PV patients and healthy subjects (HS) and identified by sequencing analysis. The minimum inhibitory concentrations (MICs) of nine antifungals (posaconazole, voriconazole, itraconazole, fluconazole, ketoconazole, miconazole, bifonazole, terbinafine and caspofungin) and tacrolimus, the interactions between three antifungals (itraconazole, ketoconazole and terbinafine) and tacrolimus, and the extracellular enzyme profile were evaluated using broth and checkerboard microdilution and the Api-Zym system, respectively.Results.Among 392Malasseziaisolates from 729 subjects (289 MF, 218 PV and 222 HS),Malassezia furfurandMalassezia globosaaccounted for 67.86 and 18.88 %, respectively.M. furfurwas the major species in MF and PV patients and HS. Synergistic effects between terbinafine and itraconazole or tacrolimas and antifungals may be irrelevant to clinical application. Bone loss and osteoporosis are commonly described as extra-intestinal manifestations of inflammatory bowel disease (IBD). Jianpi Qingchang Bushen decoction (JQBD) is a prescription used in clinical practice. However, further studies are needed to determine whether JQBD regulates the receptor activator of nuclear factor kappa B (NF-κB) (RANK)/receptor activator of NF-κB ligand (RANKL)/ osteoprotegerin (OPG) pathways and could play a role in treating IBD-induced bone loss.To evaluate the therapeutic effect of JQBD in IBD-induced bone loss and explore the underlying mechanisms.An IBD-induced bone loss model was constructed by feeding 12 6-to-8-wk-old interleukin-10 (IL-10)-knockout mice with piroxicam for 10 d. The mice were randomly divided into model and JQBD groups. We used wild-type mice as a control. The JQBD group was administered the JQBD suspension for 2 wk by gavage, while the control and model groups were given normal saline at the corresponding time points. All mice were killed after the intervention. The effect of JQBD on body weight, disease activity index (DAI), and colon length was analyzed. Histopathological examination, colon ultrastructure observation, and micro-computed tomographic scanning of the lumbar vertebrae were performed. The gene expression of NF-κB, tumor necrosis factor-α (TNF-α), IL-1β, IL-6, and IL-8 in the colon was evaluated by real-time polymerase chain reaction. Colon samples were assessed by Western blot for the expression of RANKL, OPG, RANK, and NF-κB proteins.The model group lost body weight, had a shorter colon, and showed a dramatic increase in DAI score, whereas JQBD had protective and therapeutic effects. Treatment with JQBD significantly improved inflammatory cell infiltration and reduced crypt abscess and ulcer formation. Three-dimensional imaging of the vertebral centrum in the model group revealed a lower bone mass, loose trabeculae, and "rod-shaped" changes in the structure compared to the control group and JQBD groups. The bone volume/total volume ratio and bone mineral density were significantly lower in the model group than in the control group. JQBD intervention downregulated the NF-κB, TNF-α, IL-1β, IL-6, and IL-8 mRNA expression levels. The RANKL and OPG protein levels were also improved.JQBD reduces inflammation of the colonic mucosa and inhibits activation of the RANK/ RANKL/OPG signaling pathway, thereby reducing osteoclast activation and bone resorption and improving bone metabolism.

Ulcerative colitis (UC) is a chronic nonspecific inflammation that mainly affects the mucosa and submucosa of the rectum and colon. Numerous studies have shown that endoplasmic reticulum stress (ERS)-induced autophagy plays a vital role in the pathogenesis of UC. ERS is the imbalance of internal balance caused by misfolded or unfolded proteins accumulated in the endoplasmic reticulum (ER).Excessive ERS triggers the unfolded protein response (UPR), an increase in inositol-requiring enzyme 1, and a Ca2+ overload, which activates the autophagy pathway. Autophagy is an evolutionarily conserved method of cellular self-degradation. Dysregulated autophagy causes inflammation, disruption of the intestinal barrier, and imbalance of intestinal homeostasis, therefore increasing the risk of colonic diseases. This review summarizes the pathogenesis of ERS, UPR, and ERS-related autophagy in UC, providing potential new targets and more effective treatment options for UC.

Chlorine disinfection is often used as a pretreatment technology to control biofouling of reverse osmosis (RO) membranes. However, previous studies showed that biofouling of the RO system was aggravated after chlorine disinfection. Chlorine-resistant bacteria (CRB) were presumed to be closely related to the aggravation of fouling caused by chlorine disinfection. In order to analyze the membrane fouling mechanisms of CRB, 5 CRB strains were isolated from the surface of fouled RO membranes for wastewater reclamation, and 3 reference bacterial strains, Sphingopyxis soli BM1-1, Pseudomonas aeruginosa PAO1 and Escherichia coli CGMCC1.3373, were selected for comparative study. The chlorine resistance, membrane fouling potential, secretion and adhesion characteristics of these strains were evaluated. Dissolved organic matter (DOM) plays an important role in the evolution of microbial communities. Meanwhile, ecological water delivery is an important feature of Baiyangdian Lake. To explore how the structure of the aerobic denitrification bacteria community responds to DOM during the water delivery period, the DOM components of water were examined and high-throughput sequencing of aerobic denitrification bacteria was performed. The results showed significant differences in DOM concentration in Baiyangdian Lake, with the estuary area exhibiting lower DOM concentrations. The water exhibited strong autogenous source, while DOM in the estuary area had a higher molecular weight and degree of humification. Three protein-like substances (C1, C2, and C4) and one humic-like substance (C3) were identified through PARAFAC. The protein-like substances accounted for the major proportion of DOM, which was consistent with the results of fluorescence regional integration (FRI). In order to reveal the interaction of overlying water-interstitial water nitrogen and phosphorus nutrient salt in summer at the entrance region of Baiyangdian Lake, this study sampled six main rivers in the region during July 2019. An analysis of the overlying water and interstitial water quality characteristics and the diffusion flux of applied nutrients at the sediment-water interface revealed the effects of nutrient diffusion on sediments and overlying water. The overlying water analysis showed that the water quality was slightly alkaline in the Baiyangdian Lake. Chinese vine tea can improve glucose and lipid metabolic disorders. However, its protective effects in non-alcoholic steatohepatitis (NASH) and its underlying molecular mechanisms remain unclear. Liver X receptor α (LXRα) inhibition and adenosine monophosphate-(AMP)-activated protein kinase (AMPK) activation can enhance control of NASH. AMPK activators have also been shown to inactivate LXRα. Here, the anti-NASH effects of vine tea extract (VTE) dosed at 1 g.100 g−1 diet were investigated using NASH mice challenged with a methionine and choline-deficient l-amino acid diet (MCDD) and a high-fat diet (HFD). Pharmacological mechanisms of VTE were explored using TUNEL staining, AMPK inhibition, Western blot, reporter assays, qRT-PCR analyses, and immunofluorescence. VTE treatment improved fatty liver in HFD-induced mice, while it alleviated the progression of NASH including protecting against liver lipid accumulation, steatosis, endoplasmic reticulum stress, apoptosis, inflammation, and functional injury in MCDD-fed mice. VTE reduced the action of hepatic lipogenic genes, F4/80, pro-inflammatory cytokines, CHOP, and cleaved Caspase-3 expression, while promoting expression of fatty acid oxidation genes CPT1α, ß. VTE also enhanced AMPK and blocked LXRα signaling in mouse livers. In vitro results indicated that VTE increased AMPK phosphorylation and reduced LXRα activity in HepG2 cells. Conversely, the antagonistic effect of VTE on LXRα was decreased through AMPK inhibition. Our data suggests that VTE may improve diet-induced NASH, which involves the pharmacological modulation of the AMPK-LXRα signaling pathway.

Disk tube reverse osmosis (DTRO), a modified module RO, has received growing attention in wastewater treatment. However, there is no comprehensive review of DTRO applications for wastewater treatment. In this study, China was found to be a major contributor to DTRO investigations. Specifically, 46 full- and 17 pilot-scale DTRO applications in China from the literature are summarized. The cumulative reported DTRO application scale in wastewater treatment amounted to ~16,500 m3/d by 2020 in China. Leachate and flue gas desulfurization (FGD) wastewater had the highest reported frequencies. Two-stage DTRO and "biological treatment + DTRO/two-stage DTRO" were the most reported processes in leachate treatment. Chemical oxygen demand (COD) and NH4+-N were removed at 99.00-99.95% and 98.00-99.98%, respectively, in full scales in leachate treatment. DTRO was primarily utilized in the concentration unit in hypersaline wastewater treatment (e.g., FGD wastewater). Total dissolved solids (TDS) were removed at 94.69-96.87% and 85.95-96.5% in the full- and pilot-scales, respectively. The overall operating costs were 17.50-60.61 CNY/m3 and 0.69-8.75 CNY/kgCOD for leachate treatment and 26.94-52.28 CNY/m3 and 0.71-3.61 CNY/kgTDS for FGD wastewater treatment. The major components of operating costs were chemical costs (13.09 CNY/m3, 1.63 CNY/kgCOD) for two-stage DTRO and electricity costs (19.73 CNY/m3, 1.67 CNY/kgCOD) for the "biological treatment + DTRO/two-stage DTRO" process. DTRO has shown promising prospects for wastewater treatment.

Ultrafiltration (UF) was often used as pretreatment in front of reverse osmosis (RO) unit because of its high rejection efficiency of microbes and particles. However, in some cases UF pretreatment might show adverse effects on the RO membrane flux. In this study, the effects of UF pretreatment on secondary effluent water quality and its RO membrane fouling characteristics were explored. There was almost no change of water quality after UF with different molecular weight cut-off (MWCO) membranes (100, 30 and 10 kDa), including total dissolved solid (TDS), alkalinity, conductivity, ion concentrations, etc., while pH increased a little and dissolved organic carbon (DOC) declined by about 1 mg/L. On the contrary, the RO membrane flux of UF permeates presented clear decline in comparison to the secondary effluent. The membrane fouling velocity and steady-state flux of secondary effluent was 0.052 and 0.656, while fouling velocity increased (0.077, 0.071, 0.067) and steady-state flux decreased significantly (0.397, 0.416, 0.448) after 100, 30, 10 kDa UF membrane pretreatment. This study aimed to investigate the protective effects of erythrocyte-mediated endoplasmic reticulum (ER) stress in macrophages in hemorrhagic shock. An hemorrhagic shock model was established in male BALB/c mice. Animals were randomly divided into three groups ( n = 8): control group (A), erythrocyte reinfusion group (B), and TLR9 inhibition group (C). Eight healthy BALB/c mice were also included as group N ( n = 8). Mice in group A were not treated, while mice in groups B and C were transfused with red blood cells separated from the blood of mice in group N. Flow cytometry was used to detect the expression of erythrocyte surface protein TLR9 in each group. Immunofluorescence assay was used to analyze the distribution and relative expression of protein STING in macrophages. Flow cytometry was used to analyze the expression of STING, ATF6, and IRE1 in macrophages. Enzyme-linked immunosorbent assay was used to analyze the levels of inflammatory signal molecules, including IFN-α, IFN-β, IL-6, CCL4, CCL5, and IL-6. FITC-Annexin V was used to analyze the apoptosis of immune cells (macrophages) in mouse blood samples and to detect the concentration of calcium ions in erythrocyte cytoplasm. The results showed that the expression of erythrocyte surface protein TLR9; the distribution of STING-positive cells in macrophages; the expressions of STING, ATF6, and IRE1 in macrophages; the levels of inflammatory signal molecules; the apoptosis rate of macrophages; and the intracellular calcium concentration in erythrocytes in group B were higher than those in group A, followed by group C. These results suggest that TLR9 regulates ER stress in macrophages of mice with hemorrhagic shock through the TLR9-cGAS-STING-IFN signaling pathway. Increased expression of TLR9 enhanced macrophage activity, reduced apoptosis, enhanced inflammatory response and immune response, and restored electrolyte level, which might be a therapeutic option for the treatment of hemorrhagic shock.

Extracellular polymeric substances (EPS) were the main cause of membrane biofouling in advanced treatment of wastewater. Disinfection was usually used to reduce microbial concentration in the feed water and prevent biofouling. However, the disinfection residual bacteria (DRB) could regrow to produce EPS, and there was still a paucity of information on EPS productivity and properties from DRB and their further interaction with membranes. In this study, three commonly-used disinfection methods (free chlorine, chloramine and ozone disinfection) were used to investigate their effects on microbial EPS production and the further effects on membrane fouling. It was found all the disinfection treatment increased the relative abundance of typical bacterial genus with high EPS production ability, and thus significantly enhanced EPS volumetric productivity (by 26%–82%). The molecular weight and fluorescence intensity of EPS produced by DRB also increased. The fouling potential of these EPS on ultrafiltration (UF) membranes and reverse osmosis (RO) membranes was then tested. EPS of DRB showed significantly higher fouling potential. Compared with the control group, the time for transmembrane pressure (TMP) increased to 3 bar decreased by 43%–49% in the constant-flow UF system in the disinfection groups. As for the constant-pressure RO systems, the final flux decreased by 33%–41% in the disinfection groups compared with the control group. The thickness of foulants on UF and RO membranes in the disinfection groups was all higher than that in the control group. The extended Derjaguin–Landau–Verwey–Overbeek (XDLVO) theory further showed that the EPS of DRB had significantly higher interaction energy with membranes than that in the control group. Therefore, caution should be exercised when oxidizing disinfection was used as pre-treatment for membrane filtration in the wastewater reclamation process.

Soluble microbial products (SMP) are one of the main causes of reverse osmosis (RO) membrane fouling in wastewater reclamation, and coagulation and adsorption are commonly used to prevent such fouling. However, the mechanism by which typical coagulants and adsorbents affect the RO membrane fouling caused by SMP remains unclear. In this study, two bacterial strains isolated from fouled RO membranes in a full-scale wastewater reclamation plant were used to produce SMP. Both bacterial strains were found to generate high SMP yield (0.23–0.25 g/g), which caused severe RO membrane fouling. Coagulation by polyaluminum chloride (PACl) and ferric chloride (FeCl3) and adsorption by granular activated carbon (GAC) were applied as pretreatment methods to alleviate the membrane fouling caused by SMP. Compared with PACl, FeCl3 performed better in removing SMP, decreasing fluorescence intensity, and reducing molecular weight (MW). GAC preferentially adsorbed protein and humic substances in SMP and unselectively and efficiently removed fluorescent compounds; however, it could only remove a limited amount of the high-MW compound. The RO membrane fouling potential of treated SMP was investigated, and FeCl3 and GAC were found to effectively alleviate the fouling. In addition to a decrease in the dissolved organic carbon content of SMP, a decrease in specific fluorescence intensity, MW, and polysaccharide content caused by coagulation or adsorption could result in lower RO membrane fouling.

The closure of artificial fractures is one of the most complicated problems in the development of unconventional oil and gas reservoirs. Aiming at modifying the current theoretical models, an analytical model of the contact process was proposed with due regard for elastoplastic deformation. The constructed model is based on Hertz’s elastic contact equations and can be used to interpolate an exponential function. A numerical analysis method was used to derive the equation of state to determine the rigidity and asperity of the alastoplastic phase for all the deformation phases. The correlation between average contact pressure and fracture closure of the entire rough surface was established using probability statistics. Compared with the classical models, this analytical model is easier to consruct and use. As the new model takes into account all the deformation phases, it describes the fracture surface contact behavior well with increasing normal stress. Comparison has shown that the results calculated by the developed model are in agreement with the finite-element-based Kogut-Etison (KE) model.

A new cycloartane triterpene, yunnanterpene G (1), containing an oxaspiro[5.4]decane moiety, was purified from the roots of Cimicifuga foetida. The new structure was determined from spectroscopic data and the X-ray diffraction method. Biological evaluations revealed that compound 1 significantly inhibited the mRNA expression of the atherosclerosis-related adhesion molecule CD147 (extracellular matrix metalloproteinase inducer, EMMPRIN), and proteolytic enzymes matrix metalloproteinase 2 (MMP-2), MMP-9 and MMP-14, in a dose-dependent manner in phorbol-12-myristate-13-acetate-induced human monocytic THP-1 cells by quantitative real-time PCR method. At the same time, the migration ability of the induced THP-1 cells was potently inhibited. Furthermore, western blot experiments showed that compound 1 at 25 μM strongly suppressed phosphorylation of NF-κB p65 and p38 MAPK in the differentiated THP-1 cells.
