Abstract CD73, a cell surface-bound nucleotidase, serves as a crucial metabolic and immune checkpoint. Several studies have shown that CD73 is widely expressed on immune cells and plays a critical role in immune escape, cell adhesion and migration as a costimulatory molecule for T cells and a factor in adenosine production. However, recent studies have revealed that the protumour effects of CD73 are not limited to merely inhibiting the antitumour immune response. Nicotinamide adenine dinucleotide (NAD+) is a vital bioactive molecule in organisms that plays essential regulatory roles in diverse biological processes within tumours. Accumulating evidence has demonstrated that CD73 is involved in the transport and metabolism of NAD, thereby regulating tumour biological processes to promote growth and proliferation. This review provides a holistic view of CD73-regulated NAD + metabolism as a complex network and further highlights the emerging roles of CD73 as a novel target for cancer therapies.

Abstract We analyzed the symptoms composition of Interstitial Cystitis (IC), the regularity of the evolution of symptoms before and after treatment, and the visualization of the community network, to provide a reference for clinical diagnosis and treatment of Interstitial Cystitis. Based on the outpatient electronic case data of 552 patients with Interstitial Cystitis, we used a complex network community discovery algorithm, directed weighted complex network, and Sankey map to mine the data of the symptoms composition of Interstitial Cystitis, the evolution of symptoms before and after treatment and the visualization of the community network, to analyze the epidemiological characteristics of interstitial cystitis symptoms in the real world. By the community division of the complex network of interstitial cystitis symptoms, We finally obtained three core symptom communities. Among them, symptom community A (bladder‐related symptoms) is the symptom community with the highest proportion of nodes (60.00%) in the complex network of Interstitial Cystitis, symptom community B (non‐bladder‐related symptoms 1) ranks second (32.00%) in a complex network of Interstitial Cystitis, and symptom community C (non‐bladder‐related symptoms 2) has the lowest proportion (8.00%). There is a complex evolutionary relationship between the symptoms of Interstitial Cystitis before and after treatment. Among the single symptoms before and after treatment, the decreased rate of Day frequency is 93.22%, and the reduced urgency rate is 93.07%. The decline rate of Nocturia was 82.33%. From the perspective of different communities, the overall symptoms of symptom community A decreased by 34.39% after treatment, the general symptoms of symptom community B decreased by 35.37%, and the prevalent symptoms of symptom community C decreased by 71.43%. In the case of using diet regulation treatment to treat bladder pain, the cure rate of bladder pain can reach 22.67%. The cure rate of burning in bladders can get 15.38% with Percutaneous Sacral neuromodulation, 96.95% with diet regulation treatment, and 100% with Percutaneous Sacral neuromodulation. When using behavioral physiotherapy to treat bladder pain, 3.57% of the patient's symptoms change to bladder discomfort; 4% of the patient's symptoms change to bladder discomfort when using oral medicine to treat bladder pain.Symptom research methods based on community findings can effectively explore the rule of symptom outcome of Interstitial Cystitis before and after treatment, and the results are highly interpretable by professionals. Abstract Objective This study seeks to investigate independent risk factors affecting the prognoses of patients with bladder pain syndrome/interstitial cystitis (BPS/IC) following hydrodistention surgery and to develop a column chart model and a random forest model to help predict clinical outcomes. Method A retrospective analysis was conducted on the clinical data of 1006 BPS/IC patients who visited the urology department of the Fifth Hospital of Shanxi Medical University (Shanxi Provincial People's Hospital) between June 2012 and June 2022. The patients were randomly divided into a model group (n = 704) and a validation group (n = 302). In the model group, logistic regression analysis was used to identify independent risk factors, which were used to construct a prognostic nomogram. The nomogram was evaluated by analyzing the area under the curve (AUC), calibration curve, and decision curve. These results were subsequently validated via consistency analysis (n = 302). And based on the random forest algorithm, we calculate the same data and construct a random forest model. The Hutuo River Drinking Water Source Area is an important water source of Shijiazhuang (North China Plain). Knowing the characteristics of groundwater chemistry/quality is essential for the protection and management of water resources. However, there are few studies focused on the groundwater chemistry evolution over the drinking water area. In this study, total of 160 groundwater samples were collected in November 2021, and the spatial distribution of groundwater chemistry and related controlling factors were analyzed using hydrological and multivariate analysis. The entropy-weighted water quality index (EWQI) was introduced to assess the groundwater quality. The results show that the hydrogeochemical types of groundwater are Ca-HCO3 (78.1%), mixed Ca-Mg-Cl (20%), and Ca-Cl (1.9%) in the area. Graphical and binary diagrams indicate that groundwater hydrochemistry is mainly controlled by water–rock interaction (i.e., rock weathering, mineral dissolution, and ion exchange). Five principal components separated from the principal component analysis represent the rock–water interaction and agricultural return, redox environment, geogenic sources, the utilization of agricultural fertilizer, the weathering of aluminum silicates, and dissolution of carbonates, respectively. More than 70% of the samples are not recommended for irrigation due to the presence of high salt content in groundwater. EWQI assessment demonstrates that the quality of the groundwater is good. The outcomes of this study are significant for understanding the geochemical status of the groundwater in the Hutuo River Drinking Water Source Area, and helping policymakers to protect and manage the groundwater.

Vegetable production is an important area of focus in China’s agricultural structural adjustment plans, and it serves as one of the essential industries in the rural economy. Several studies have investigated how to optimize vegetable yield and quality through proper irrigation and fertilization to ensure efficient and sustainable development. The main objective of this paper is to examine the impact of different combinations of irrigation and nitrogen inputs on facility-grown vegetables under irrigation and fertilization conditions. Additionally, we aim to identify the optimal irrigation and fertilization regime that can enhance yield quality while also promoting environmental benefits. In this study, we focused on a white radish–tomato facility as the main research object. Using multiple regression and spatial analysis methods, we established three irrigation levels (W1: 100% ET0, W2: 85% ET0, W3: 70% ET0) and four nitrogen application levels (N0: no nitrogen, N1: high nitrogen, locally recommended nitrogen, N2: medium nitrogen, 85% N1, N3: low nitrogen, 70% ET0). We analyzed the effects of an irrigation nitrogen application on vegetable yield, nitrogen bias productivity, soil nitrogen surplus, and integrated N1 warming potential. Our experimental results showed that irrigation volume and nitrogen application had a considerable impact on the yield of facility-grown vegetables, and there was a positive correlation between irrigation water and fertilizer application and yield. By moderately reducing the irrigation volume and increasing nitrogen application, soil nitrogen surplus and nitrogen fertilizer bias productivity can be effectively improved. In addition, our study found that the integrated warming potential and the bias productivity of nitrogen fertilizer showed a quadratic relationship, which indicated that the integrated warming potential and nitrogen fertilizer bias productivity would first become larger and then decrease under the condition of increasing the irrigation volume and nitrogen application rate. By analyzing the difference between W2N2 and W1N1, we found that moderate water-saving and nitrogen reduction did not affect yield. Furthermore, it effectively improved the bias productivity of nitrogen fertilizer. Therefore, it is recommended that when the irrigation volume is between 560 and 650 mm and the nitrogen application rate is between 325 and 400 kg/hm2 and more than 90% of the maximum value of yield, nitrogen fertilizer bias productivity can be achieved at the same time while also having a lower integrated warming potential. This range of irrigation and nitrogen application intervals is close to optimal. Our study provides a guiding basis for rotational soil nitrogen balance, optimal water, and nitrogen management of facility-grown vegetables. We propose an optimal water and nitrogen management strategy that is more efficient and sustainable under the plant culture model. This strategy provides a new way of thinking and methodology for high-quality production that is water-saving and fertilizer-saving while addressing the water and soil resource problems that exist in the current development of the vegetable industry.

To explore the causal relationship between maternal smoking around birth and childhood asthma using Mendelian randomization (MR). Using the data from large-scale genome-wide association studies, we selected independent genetic loci closely related to maternal smoking around birth and maternal diseases as instrumental variables and used MR methods. In this study, we considered the inverse variance weighted method (MR-IVW), weighted median method, and MR-Egger regression. We investigated the causal relationship between maternal smoking around birth and maternal diseases in childhood asthma using the odds ratio (OR) as an evaluation index. Multivariable MR (MVMR) included maternal history of Alzheimer's disease, illnesses of the mother: high blood pressure and illnesses of the mother: heart diseaseas covariates to address potential confounding. Sensitivity analyses were evaluated for weak instrument bias and pleiotropic effects. It was shown with the MR-IVW results that maternal smoking around birth increased the risk of childhood asthma by 1.5% (OR = 1.0150, 95% CI: 1.0018–1.0283). After the multivariable MR method was used to correct for relevant covariates, the association effect between maternal smoking around birth and childhood asthma was still statistically significant (P < 0.05). Maternal smoking around birth increases the risk of childhood asthma.

Studies have suggested that the progress of most kidney diseases from occurrence to course and subsequent related complications are closely related to inflammatory reaction. Increased common leukocytes count in the family (neutrophils, eosinophils, basophils, lymphocytes, etc.) are also involved in the tissue damage of kidney diseases. However, these studies are only traditional observational studies, which cannot prove whether there is a causal relationship between these four kinds of leukocytes count and kidney diseases. We aim to explore the causal relationship between these four kinds of leukocytes count and kidney diseases by Mendelian randomization (MR). Large sample size of the genome-wide association database of four cell traits (neutrophil, basophil, lymphocyte, and eosinophil cell counts) in the leukocyte family were used as exposure variables. The outcome variables were various renal diseases (including chronic renal failure, acute renal failure, hypertensive heart or/and kidney disease, hypertensive renal disease, disorders resulting from impaired renal tubular function, and type 1 diabetes with renal complications). The covariates used in multivariable MR are also four cell traits related to blood cells (neutrophil, basophil, lymphocyte, and eosinophil cell counts). Instrumental variables and single nucleotide polymorphic loci were identified (P < 5 × 10−8. Linkage disequilibrium R2 < 0.001). The causal relationships were studied by inverse variance weighted (IVW), weighted median, and MR-Egger regression. Sensitivity analysis was also performed. In our study, IVW analysis results showed that increased neutrophil cell count was a risk factor for chronic renal failure (OR = 2.0245861, 95% CI = 1.1231207–3.649606, P = 0.01896524), increased basophil cell count was a risk factor for chronic renal failure (OR = 3.975935, 95% CI = 1.4871198–10.62998, P = 0.005942755). Basophil cell count was not a risk factor for acute renal failure (OR = 1.160434, 95% CI = 0.9455132–1.424207, P = 0.15448828). Increased basophil cell count was a protective factor for hypertensive heart and/or renal disease (OR = 0.7716065, 95% CI = 0.6484979–0.9180856, P = 0.003458707). Increased basophil cell count was a risk factor for disorders resulting from impaired renal tubular function (OR = 1.648131, 95% CI = 1.010116–2.689133, P = 0.04546835). Increased lymphocyte cell count was a risk factor for hypertensive renal disease (OR = 1.372961, 95% CI = 1.0189772–1.849915, P = 0.03719874). Increased eosinophil cell count was a risk factor for type 1 diabetes with renal complications (OR = 1.516454, 95% CI = 1.1826453–1.944482, P = 0.001028964). Macrophage inflammatory protein 1b levels was a protective factor for renal failure (OR = 0.9381862, 95% CI = 0.8860402–0.9934013, P = 0.02874872). After multivariable MR was used to correct covariates (neutrophil, basophil, and lymphocyte cell counts), the correlation effect between increased eosinophil cell counts and type 1 diabetes with renal complications was still statistically significant (P = 0.02201152). After adjusting covariates (neutrophil, basophil, and eosinophil cell counts) with multivariable MR, the correlation effect between increased lymphocyte cell counts and hypertensive renal disease was still statistically significant (P = 0.02050226). This study shows that increased basophils can increase the relative risk of chronic renal failure and renal tubular dysfunction, and reduce the risk of hypertensive heart disease and/or hypertensive nephropathy, while increased basophil cell count will not increase the relative risk of acute renal failure, increased neutrophil cell count can increase the risk of chronic renal failure, increased lymphocyte cell count can increase the relative risk of hypertensive nephropathy, and increased eosinophil cell count can increase the relative risk of type 1 diabetes with renal complications.
