Zingiber roseum (Roxb.) Roscoe, a perennial herb from the Zingiberaceae family, has a long history of traditional use in the treatment of several ailments including pain, inflammation, fever, cough, arthritis, skin diseases, and liver infections. This study sought to confirm the efficacy of Zingiber roseum (Roxb.) Roscoe leaves methanol extract (ZrlME) as reported in traditional usage by evaluating its analgesic, anti-inflammatory, and antipyretic capabilities. In addition, in silico molecular docking of the metabolites identified in ZrlME was studied to verify the experimental outcomes. ZrlME demonstrated strong dose-dependent analgesic efficacy against all analgesic tests. ZrlME (400 mg/kg) showed higher antiinflammatory activity than the standard in the carrageenan-induced paw edema test model. A significant reduction of rectal temperature (3.97°F↓) was also recorded at the same dose of ZrLME after 24 h of treatment. Seven polyphenolic metabolites were identified and quantified by HPLC-DAD analysis, including 3, 4-dihydroxy benzoic acid, (-) epicatechin, rutin hydrate, p-coumaric acid, trans-ferulic acid, rosmarinic acid, and myricetin. Strong binding affinities (ranges from -5.8 to -8.5 Kcal/mol) between the aforesaid polyphenols and cyclooxygenase-2 were discovered. Moreover, molecular dynamics simulations (MDS) demonstrated that these polyphenols exhibit significant COX-2 inhibitory activity due to their high stability in the COX-2 active site. In computational prediction, the polyphenols were also found to be nontoxic, and a variety of biological activities, such as antioxidant, analgesic, anti-inflammatory, antipyretic, and hepatoprotective, were observed. Introduction: A perennial, aromatic, tuberose plant Zingiber roseum (Roscoe.) (Zingiberaceae), flourishes in tropical and subtropical climates. In Traditional Chinese Medicine, several pharmacological properties of Zingiber roseum have been reported its antiseptic, antivertigo, and antidiarrheal activities. Therefore, the present article aims to provide insights into the ethnomedicinal, phytochemistry, and pharmacology of Zingiber roseum. Methods: The literature was compelled after systematically searching scientific databases, including Scopus, PubMed, Google Scholar, and Research Gate. The selection criteria for the plant comprised the therapeutic potential of Zingiber roseum and its active components. Moreover, to explore anti-diabetic activity, ligands of interest from Z. roseum were evaluated for their affinity towards PPAR-α and PPAR-γ. Results and discussions: Out of 200 articles, 140 were selected for the current study, and from the para-topic literature, it was found that Zingiber roseum has numerous pharmacological properties due to the presence of phytoconstituents like flavonoids, alkaloids, phenolic chemicals, terpenoids, saponins, and phytosterols. Furthermore, in silico studies were carried out using PyRx. It was found that rosmarinic acid (-8.3 kcal/mol) and stigmasterol (-11.12 kcal/mol) exhibited the highest binding affinities for PPAR-α and PPAR-γ, respectively, when compared to standard Rosiglitazone. Conclusion: It may be concluded that Z. roseum has several therapeutic activities. Moreover, in silico studies revealed the anti-diabetic action of Z. roseum via modulation of PPAR-α and PPAR-γ.

Introduction: The rhizomes of Zingiber roseum plant have been used in traditional medicine to treat several ailments. Regardless of worth, no research has accounted its toxicity potential. So, the study was designed to determine safety and toxicity potential of Zingiber roseum rhizomes (ZRR) in acute oral toxicity model in swiss albino mice. Methods: Acute oral toxicity was assessed as per the guidelines of ‘The Brazilian Agency of National Health Surveillance'. In an acute toxicity investigation, 300 mg/kg, 600 mg/kg, and 1200 mg/kg of ZRR extract was orally administered to mice. Thereafter, the animals were monitored for 14 days. To analyze any potential toxicity, general behavior of animals, clinical symptoms of poisoning, body weight, biochemical and hematological marker, and liver histology was carried out. Results: Oral dosing of 300, 600, and 1200 mg/kg of crude extract did not produce mortality or any adverse effect in the laboratory animals. The control and treatment groups of mice exhibited similar behavioral characteristics, neurological signs and total body weight during the treatment period of 14 days. The markers of liver damage i.e., ALT and AST, total serum protein, and albumin did not show any significant change between extract-treated and control mice. The extracts also significantly suppressed ALP activity as compared to control. Kidney function was assessed in mice by measuring creatinine and urea level and no change was observed in level of creatinine and urea in experimental animals. Moreover, no alterations were observed in hematological markers and lipid profile (triglyceride and total cholesterol level). In addition, the liver showed normal architecture and no significant adverse consequences on histopathology analysis. Discussion & conclusion: These outcomes propose that LD50 of rhizomes of Z. roseum is higher than 1200 mg/kg b.w. and might be possibly safe for consumption.

Aim: To evaluate the antidiabetic potential of β-sitosterol from Zingiber roseum., Background: Diabetes mellitus is a cluster of metabolic disorders, and 90% of diabetic patients are affected with Type II diabetes (DM2). For the treatment of DM2, thiazolidinedione drugs (TZDs) were proposed, but recent studies have shown that TZDs have several detrimental effects, such as weight gain, kidney enlargement (hypertrophy), fluid retention, increased risk of bone fractures, and potential harm to the liver (hepatotoxicity). That is why a new molecule is needed to treat DM2., Objective: The current research aimed to assess the efficacy of β-Sitosterol from methanolic extract of Zingiber roseum in managing diabetes via PPARγ modulation., Methods: Zingiber roseum was extracted using methanol, and GC-MS was employed to analyze the extract. Through homology modeling, PPARγ structure was predicted. Molecular docking, MD simulation, free binding energies, QSAR, ADMET, and bioactivity and toxicity scores were all used during the in-depth computer-based research., Results: Clinically, agonists of synthetic thiazolidinedione (TZDs) have been used therapeutically to treat DM2, but these TZDs are associated with significant risks. The rhizome of Zingiber roseum has been used as traditional medicines for the treatment of gastric ulcer, skin diseases, and liver infections. Zerumbone is an essential sesquiterpenoid previously isolated from different species of Zingiberaceae and possesses numerous pharmacological properties. Based on previous reports, the aim of the current study was to investigate the antimicrobial activity of extract, fractions, and isolation of zerumbone from Zingiber roseum. The antimicrobial activity was determined using disc diffusion method. The serial tube dilution technique was used to determine Minimum Inhibitory Concentration (MIC) of the samples. Vacuum Liquid Chromatography (VLC) and open column chromatography were used for separation. The methanol extract exhibited maximum antimicrobial activity against Escherichia coli, Saccharomyces cerevisiae, and Aspergillus niger in both methods. VLC of the extract yielded five fractions (Fr. 1 to Fr. 5). Fr. 1 exhibited maximum antimicrobial activity compared to the other fractions. Column chromatography of Fr.1 generated 47 fractions. Fraction 1 to 9 were combined and crystallized in n-hexane to isolate zerumbone as the most abundant source (yield 6.5 % from dried MeOH extract). Zerumbone showed good activity against all tested bacteria. The results of our study might validate the ethnobotanical use of Zingiber roseum rhizomes in the treatment of infectious diseases. Moreover, Zingiber roseum could be one of the most abundant sources for the isolation of natural zerumbone to study different pharmacological studies in the field of new drug discovery. Plant extracts are considered as a promising alternative to chemical pest control agents. The chemistry of plant extracts varies with several environmental as well as geographical factors. Zingiber roseum (Roxb.) Roscoe also known as "Rosy Ginger" and " Jangli Adrak" is one of the most important aromatic and medicinal plant of family Zingiberaceae. The present study aimed to investigate the impact of altitude on the chemical composition, nematicidal and herbicidal activity of Z. roseum rhizome and seed extracts. The plants were collected from three different altitudes of Kumaun, Uttarakhand representing different environmental conditions. The extracts were prepared in acetone using cold percolation method. The compositional variation in chemical constituents of the extracts was determined by gas chromatography-mass spectrometry (GC–MS) analysis. The yield and chemical composition of extracts showed a significant altitudinal impact. Nematicidal activity of extracts was performed against Meloidogyne incognita. Results showed that the extracts exhibited significant nematode mortality irrespective of the altitudinal gradients. A significant correlation of altitudinal gradient was observed for the nematode egg hatchability reduction for the rhizome extracts. It was observed that plants growing at higher altitudes had better egg hatchability reduction (86.42 ± 2.30% at 1000 μg/mL after 96 h), compared with those growing at lower elevations. The herbicidal activity of rhizome and seed extracts was performed against radish and chicory seeds and the extracts showed different level of intensity of germination percentage, root and shoot length inhibition. In silico molecular docking was conducted to screen the nematicidal activity of major and potent compounds with the crystalline structure of Glutathione S-transferases (GSTs) of Caenorhabditis elegans and Wuchereria bancrofti , and Glutathione S-transferase-1 (GST-1) of M. incognita. The present study focussed on phytochemical analysis of Zingiber roseum through GC & GC-MS, antifungal and antioxidant activities of methanolic extracts of seed, rhizome and perianth of the plant. GC-MS analysis was done by using GC MS-QP 2010 Plus equipment revealed diverse chemical make-up of the constituents. Among identified constituents ascorbic acid 2, 6-dihexadecanoate, 2-(4-hydroxy-3-methoxyphenyl)-3,7-dimethoxy-4h-chromen-4-one, hexadecanoic acid metyl ester, 9,12-octadecadienoic acid (Z,Z) and β-citronellal were found as major compounds in methanolic extracts from seeds, rhizome and petals respectively. The antifungal activity was done by disc diffusion methods for which extracts were tested against Fusarium accuminata. Among them the extracts were found to be remarkable fungicide according to its inhibition action against Fusarium accuminata. The in vitro antioxidant activity of the methanol extracts were evaluated by DPPH radical scavenging, FRAP assay and metal chelating activity. The antioxidant assay revealed good to moderate antioxidant potential with different IC50 values with respect to Fe3+ reducing power (200.34-438.79 μg/ml), Fe2+ metal chelating ability (310.80-469.69 μg/ml) and DPPH anion scavenging activities (215.99-884.40 μg/ml) in comparison to known standards. Zingiber roseum is a perennial herb in the Zingiberaceae family. The plant is native to Bangladesh, and rhizomes are frequently used in traditional medicine to cure gastric ulcers, asthma, wounds, and rheumatic disorders. Therefore, the present study aimed to analyse the antipyretic, anti-inflammatory, and analgesic properties of Z. roseum rhizome to confirm its efficacy in traditional applications. After 24 h of treatment, ZrrME (400 mg/kg) showed a considerable drop in rectal temperature (3.42°F) compared to standard paracetamol (5.26°F). At both doses (200 and 400 mg/kg), ZrrME showed a substantial dose-dependent decrease in paw oedema. However, after 2, 3 and 4 h of testing, the extract (200 mg/kg) had a lower anti-inflammatory response than standard indomethacin, whereas the higher dose (400 mg/kg) of rhizome extract had a more robust response compared to standard. ZrrME also showed substantial analgesic activity against all in vivo analgesic test models. The in vivo findings were further evaluated by in silico study of our previously identified compounds of ZrrME with the cyclooxygenase-2 enzyme (3LN1). The substantial binding energy (ranges from-6.2 to-7.7 Kcal/mol) of the polyphenols (excluding catechin hydrate) to the COX-2 enzyme affirm the in vivo test results of the present studies. In addition, the compounds were found effective as antipyretic, anti-inflammatory, and analgesic agents, according to the biological activity prediction software. Both in vivo and in silico results demonstrated promising antipyretic, anti-inflammatory, and pain-relieving effects of Z. roseum rhizome extract, which corroborate the claim of its traditional uses. Abstract: A combination of GC and GC–MS revealed the presence of 60 compounds out of which 47 compounds (96.2% of the oil) were identified in the essential oil of seeds of Zingiber roseum Rosc., the major compounds being α-pinene, β-pinene, limonene, p-cymene, α-terpineol and verticiole. The unique feature is the dominant presence of mono- and sesquiterpene hydrocarbons which make about 82% of the oil. The oil showed myorelaxant activity on isolated rat duodenal smooth muscle. The Zingiber roseum seed essential oil (ZRSEO) relaxed both cabachol- and KCl-induced contractile responses with IC50 of non-significant difference. The results confirm a common mechanism in both the types of action of ZRSEO and suggests a probable inhibitory effect of ZRSEO on influx of Ca2+ through cell membrane of rat duodenal smooth muscle. Histopathological examination also revealed the evidence of recovery from CCL 4 induced cellular damage when pretreated with Z. roseum rhizome extract. The in-vivo hepatoprotective effects were further investigated by the in-silico study of the aforementioned compounds with liver-protective enzymes such as superoxide dismutase (SOD), peroxiredoxin, and catalase. The strong binding affinities (ranging from −7.3359 to −9.111 KCal/mol) between the phenolic compounds (except trans-cinnamic acid) and oxidative stress enzymes inhibit ROS production during metabolism. The compounds were also found non-toxic in computational prediction, and a series of biological activities like antioxidant, anticarcinogen, cardio-protectant, hepato-protectant have been detected.
