In this paper, we introduced a new double inertial subgradient extragradient method for solving a variational inequality problem in Hilbert space. In our method, the mapping needed not to satisfy any assumption of monotonicity and two different self-adaptive step sizes were used for avoiding the need of Lipschitz constant of the mapping. The strong convergence of the proposed method was proved under some new conditions. Finally, some numerical examples were presented to illustrate the convergence of our method and compare with some related methods in the literature.

Abstract Background The mental health of university students during the COVID-19 pandemic has attracted the attention of researchers. For the present study researchers constructed a mediation model to explore the relationship between psychological resilience and post-traumatic growth, the mediating role of negative emotions and the moderating role of deliberate rumination in students. Methods The Psychological Resilience Scale, Posttraumatic Growth Inventory, Depression-Anxiety-Stress Scale (DASS-21) and Event Related Rumination Inventory were used in a survey of 881 college students. The data were analyzed using SPSS 26.0 and the PROCESS plugin (version 3.3). Results (1) Psychological resilience is positively related with post-traumatic growth. Deliberate rumination is positively related to psychological resilience, posttraumatic growth, and negative emotions. Psychological resilience, post-traumatic growth and negative emotions are negatively related. (2) Negative emotions mediated the relationship between psychological resilience and post-traumatic growth. (3) Deliberate rumination plays a moderating role in psychological resilience affecting negative emotions. Deliberate rumination plays a moderating role in the extent to which psychological resilience influences PTG through negative emotions. Conclusions Psychological resilience affects post-traumatic growth directly and also indirectly through negative emotions. With the increase of mental resilience, the level of negative emotion tended to decrease. When individuals are experiencing negative emotions, high levels of active rumination are more likely to promote post-traumatic growth. This study helps to explore the factors affecting the mental health of college students during the epidemic, thus providing guidance for appropriate mental health interventions.

Background: Ischemic stroke (IS) is a leading cause of long-term disability and even mortality, threatening people's lives. Yinao Fujian (YNFJ) formula is a Traditional Chinese Medicine formula that has been widely used to treat patients with IS. However, the molecular mechanism of YNFJ for the treatment of IS is still elusive. Our study aimed to explore the potential protective effect and the underlying mechanisms of YNFJ on IS using a network pharmacology approach coupled with experimental validation. Materials and methods: Effective compounds of YNFJ were collected from BATMAN-TCM and TCMSP databases, while IS targets were obtained from GeneCards, OMIM, TTD and DrugBank databases. The protein-protein interaction (PPI) network was constructed to further screen the hub targets of YNFJ in IS treatment. GO and KEGG enrichment analyses were used to identify the critical biological processes and signaling pathways of YNFJ for IS. Moreover, Nissl staining, HE, TTC staining and Tunel staining were used in the MCAO model to prove the neuroprotective effect of YNFJ. Oxidative damage, inflammatory factor release and related pathways were tested in MCAO rat model and hypoxia-induced BV2 cell model, respectively. Results: We found that YNFJ treatment significantly alleviated MCAO-induced nerve damage and apoptosis. Then, network pharmacology screening combined with literature research revealed IL6, TNF, PTGS2, NFKBIA and NFE2L2 as the critical targets in a PPI network. Moreover, the top 20 signaling pathways and biological processes associated with the protective effects of YNFJ on IS were enriched through GO and KEGG analyses. Further analysis indicated that NF-κB and Nrf2/HO-1 signaling pathways might be highly involved in the protective effects of YNFJ on IS. Finally, in vitro and in vivo experiments confirmed that YNFJ inhibited the release of inflammatory factors (IL-6 and TNF-α) and MDA content, and increased the activity of SOD. In terms of the mechanism, YNFJ inhibited the release of inflammatory factors by suppressing the NF-κB pathway and decreased the expression of iNOS and COX-2 to protect microglia from inflammation damage. In addition, YNFJ initiated the dissociation of Keap-1 and Nrf2, and activated the downstream protein HO-1, NQO1, thus decreasing oxidative stress. Conclusion: Taken together, the findings in our research showed that the protective effects of YNFJ on IS were mainly achieved by regulating the NF-κB and Nrf2/HO-1 signaling pathways to inhibit oxidative stress damage and inflammatory damage of microglia.

Hepatocellular carcinoma (HCC) is a fatal malignancy that has limited treatment options. This study focused on the potential therapeutic effects of curcumin (CUR) and berberine (BBR) on the miR-221/SRY-box transcription factor 11 (SOX11) axis in HCC. We investigated the combined effects of CUR and BBR on HEPG2 and Huh7 cell survival and miR-221 expression using Cell Counting Kit-8 assays and RT-qPCR, respectively. Western blotting was used to detect changes in the apoptosis-related caspase-3/9 protein levels. We performed bioinformatics analysis and dual-luciferase assays and measured apoptotic protein levels to assess the role of the miR-221/SOX11 axis in mediating the effects of CUR-BBR. Both CUR and BBR suppressed HCC cell growth in a dose-dependent manner, with the most potent combined effect observed at a 2:1 ratio. CUR-BBR treatment significantly downregulated miR-221 expression, and miR-221 overexpression partially reversed the CUR-BBR-mediated decrease in cell survival. In addition, SOX11 was found to be a direct target of miR-221. CUR-BBR treatment upregulated SOX11 expression, and overexpression of SOX11 restored the inhibitory effects of CUR-BBR on cell growth, migration, and invasion and promoted apoptosis in the presence of miR-221. Furthermore, CUR-BBR activated pro-apoptotic proteins caspase-3/9 through the miR-221/SOX11 axis. The combined effect of CUR-BBR played an important role in inhibiting the growth of HCC cells. This combined effect was achieved by regulating the miR-221/SOX11 axis and activating the synthesis of pro-apoptotic proteins. Our findings highlight a promising combined therapeutic approach for HCC and underscore the importance of targeting the miR-221/SOX11 axis.

Abstract The fast-developing synthetic biology (SB) has provided many genetic tools to reprogram and engineer cells for improved performance, novel functions, and diverse applications. Such cell engineering resources can play a critical role in the research and development of novel therapeutics. However, there are certain limitations and challenges in applying genetically engineered cells in clinical practice. This literature review updates the recent advances in biomedical applications, including diagnosis, treatment, and drug development, of SB-inspired cell engineering. It describes technologies and relevant examples in a clinical and experimental setup that may significantly impact the biomedicine field. At last, this review concludes the results with future directions to optimize the performances of synthetic gene circuits to regulate the therapeutic activities of cell-based tools in specific diseases.

Abstract The joint detection and re-identification (re-ID) strategy shares network features of detection and re-ID, sacrifices the complex probability graph model pairing strategy, and consolidates a two-stage video tracking process into a one-stage, making the multi-object tracking process simple, fast, and accurate. In dense scenes, identified transfer is a major challenge for joint detection and re-ID. To this end, a probability graph model suitable for joint detection and re-ID is presented. The proposed model abandons the idea of matching candidate detections with historical detections in a classical probability graph, uses a scheme to calculate the degree of matching between candidate detections and historical trajectories, and transforms task of ID matching in re-ID process into an energy minimization problem of a conditional random field (CRF). However, the solution space of general CRF is complex and requires an iterative search. To achieve efficient online tracking, the original CRF problem is approximately transformed into a combination of multiple CRF problems with closed-form solutions. Moreover, the proposed algorithm has been applied in practical applications using an edge-cloud model that maintains the balance between performance and efficiency. Extensive experiments on the well-known MOTchallenge benchmark demonstrate the superior performance of the proposed algorithm.

Abstract Background The aim of the study was to use a moderated mediation model to understand and examine the relationship between negative emotions and creativity among college students during the COVID-19 pandemic, using psychological resilience as a mediator and posttraumatic growth as a moderator. Methods A sample of 881 college students in mainland China completed a self-report questionnaire that included four scales: the Depression-Anxiety-Stress Scale, Psychological Resilience Scale, Runco Ideational Behavior Scale and Posttraumatic Growth Inventory. Results Findings indicated that:(1) negative emotions were a strong predictor of creativity; (2) psychological resilience partially mediated the association between negative emotions and creativity; and (3) posttraumatic growth moderated the positive effect of psychological resilience, such that the indirect effect between negative emotions and creativity via psychological resilience was stronger for someone with a low level of resilience. Conclusion The findings further clarify the mechanisms that affect the relationship between negative emotions and creativity among college students.

Highlights 1. An IDD-related ceRNA network involving four circRNAs, three miRNAs and 11 mRNAs was constructed. 2. The expression levels of miR-185-5p, miR-486-5p, FOXO1, SMAD2 and TGFB1 were consistent in different databases. 3. Our study identified IDD-related circRNA–miRNA–mRNA interaction axes, including circRNA_100086–miR-509-3p–MAPK1.

After androgen deprivation, prostate cancer frequently becomes castration resistant (CRPC), with intratumoral androgen production from extragonadal precursors that activate the androgen receptor pathway. 3β-Hydroxysteroid dehydrogenase-1 (3βHSD1) is the rate-limiting enzyme for extragonadal androgen synthesis, which together lead to CRPC. Here, we show that cancer-associated fibroblasts (CAFs) increased epithelial 3βHSD1 expression, induced androgen synthesis, activated the androgen receptor, and induced CRPC. Unbiased metabolomics revealed that CAF-secreted glucosamine specifically induced 3βHSD1. CAFs induced higher GlcNAcylation in cancer cells and elevated expression of the transcription factor Elk1, which induced higher 3βHSD1 expression and activity. Elk1 genetic ablation in cancer epithelial cells suppressed CAF-induced androgen biosynthesis in vivo. In patient samples, multiplex fluorescent imaging showed that tumor cells expressed more 3βHSD1 and Elk1 in CAF-enriched areas compared with CAF-deficient areas. Our findings suggest that CAF-secreted glucosamine increases GlcNAcylation in prostate cancer cells, promoting Elk1-induced HSD3B1 transcription, which upregulates de novo intratumoral androgen synthesis to overcome castration.

Prostate cancer is highly dependent on androgens and the androgen receptor (AR). Hormonal therapies inhibit gonadal testosterone production, block extragonadal androgen biosynthesis, or directly antagonize AR. Resistance to medical castration occurs as castration-resistant prostate cancer (CRPC) and is driven by reactivation of the androgen-AR axis. 3β-hydroxysteroid dehydrogenase-1 (3βHSD1) serves as the rate-limiting step for potent androgen synthesis from extragonadal precursors, thereby stimulating CRPC. Genetic evidence in men demonstrates the role of 3βHSD1 in driving CRPC. In postmenopausal women, 3βHSD1 is required for synthesis of aromatase substrates and plays an essential role in breast cancer. Therefore, 3βHSD1 lies at a critical junction for the synthesis of androgens and estrogens, and this metabolic flux is regulated through germline-inherited mechanisms. We show that phosphorylation of tyrosine 344 (Y344) occurs and is required for 3βHSD1 cellular activity and generation of Δ4, 3-keto-substrates of 5α-reductase and aromatase, including in patient tissues. BMX directly interacts with 3βHSD1 and is necessary for enzyme phosphorylation and androgen biosynthesis. In vivo blockade of 3βHSD1 Y344 phosphorylation inhibits CRPC. These findings identify what we believe to be new hormonal therapy pharmacologic vulnerabilities for sex-steroid dependent cancers.

Background: Krüppel like factor 10 (KLF10), which is also known as TGF-β Inducible Early Gene-1 (TIEG1), plays a crucial role in regulating cell proliferation, cell apoptosis and inflammatory reaction in human carcinoma cells. Moreover, KLF10 knockout in mice leads to severe defects associated with muscle, skeleton and heart etc. However, the function of KLF10 in intervertebral disc degeneration (IVDD) has not been reported yet. Methods: The relationship between KLF10 and IVDD were investigated in nucleus pulposus (NP) tissues from human and rats. The role of KLF10 in NP cells was explored via loss or gain of function experiments. IVDD rat models were constructed through needle puncture and the effects of KLF10 in IVDD model of rats were investigated via intradiscal injection of KLF10. Results: We first found that KLF10 was lowly expressed in degenerative NP tissues and the level of KLF10 showed negative correlation with the disc grades of IVDD patients. Loss or gain of function experiments demonstrated that KLF10 could inhibit apoptosis and enhance migration and proliferation of IL-1β induced NP cells. And KLF10 overexpression reduced extracellular matrix (ECM) degeneration and enhanced ECM synthesis, whereas knockdown of KLF10 resulted in adverse effects. These positive effects of KLF10 could be reversed by the inhibition of TGF-β signaling pathway. In vivo, KLF10 overexpression alleviated IVDD. Conclusions: This is the first study to reveal that KLF10 was dysregulated in IVDD and overexpressed KLF10 could alleviate IVDD by regulating TGF-β signaling pathway both in vitro and in vivo, which were involved in prohibiting apoptosis, promoting proliferation and migration of NP cells.The translational potential of this article: Overexpression of KLF10 might be an effective therapeutic strategy in the treatment of IVDD.

Rosa rugosa is a famous Chinese traditional flower with high ornamental value and well environmental adapt ability. The cultivation of new colorful germplasms to improve monotonous flower color could promote its landscape application. However, the mechanism of flower color formation in R. rugosa remains unclear. In this study, combined analyses of the chemical and transcriptome were performed in the R. rugosa germplasms with representative flower colors. Among the identified anthocyanins, cyanidin 3,5-O-diglucoside (Cy3G5G) and peonidin 3,5-O-diglucoside (Pn3G5G) were the two dominant anthocyanins in the petals of R. rugosa. The sum content of Cy3G5G and Pn3G5G was responsible for the petal color intensity, such as pink or purple, light- or dark- red. The ratio of Cy3G5G to Pn3G5G was contributed to the petal color hue, that is, red or pink/purple. Maintaining both high relative and high absolute content of Cy3G5G may be the precondition for forming red-colored petals in R. rugosa. Cyanidin biosynthesis shunt was the dominant pathway for anthocyanin accumulation in R. rugosa, which may be the key reason for the presence of monotonous petal color in R. rugosa, mainly pink/purple. In the upstream pathway of cyanidin biosynthesis, 35 differentially expressed structural genes encoding 12 enzymes co-expressed to regulate the sum contents of Cy3G5G and Pn3G5G, and then determined the color intensity of petals. RrAOMT, involved in the downstream pathway of cyanidin biosynthesis, regulated the ratio of Cy3G5G to Pn3G5G via methylation and then determined the color hue of petals. It was worth mentioning that significantly higher delphinidin-3,5-O-diglucoside content and RrF3’5’H expression were detected from deep purple-red-flowered 8-16 germplasm with somewhat unique and visible blue hue. Three candidate key transcription factors identified by correlation analysis, RrMYB108, RrC1, and RrMYB114, might play critical roles in the control of petal color by regulating the expression of both RrAOMT and other multiple structural genes. These results provided novel insights into anthocyanin accumulation and flower coloration mechanism in R. rugosa, and the candidate key genes involved in anthocyanin biosynthesis could be valuable resources for the breeding of ornamental plants in future.

The most recent 3D object detectors for point clouds rely on the coarse voxel-based representation rather than the accurate point-based representation due to a higher box recall in the voxel-based Region Proposal Network (RPN). However, the detection accuracy is severely restricted by the information loss of pose details in the voxels. Different from considering the point cloud as voxel or point representation only, we propose a point-to-voxel feature learning approach to voxelize the point cloud with both the point-wise semantic and local spatial features, which maintains the voxel-wise features to build the high-recall voxel-based RPN and also provides the accurate point-wise features for refining the detection results. Another difficulty in object detection for point cloud is that the visible part varies a lot against the full view of object because of the perspective issues in data acquisition. To address this, we propose an attentive corner aggregation module to attentively aggregate the features of local point cloud surrounding a 3D proposal from the perspectives of eight corners in the proposal 3D bounding box. The experimental results on the competitive KITTI 3D object detection benchmark show that the proposed method achieves state-of-the-art performance.

Any sort of wound injury leads to the destruction of skin integrity and wound formation, causing millions of deaths every year and accounting for 10% of death rate insight into various diseases. The ideal biological wound dressings are expected to possess extraordinary mechanical characterization, cytocompatibility, adhesive properties, antibacterial properties, and conductivity of endogenous electric current to enhance the wound healing process. Recent studies have demonstrated that biomedical hydrogels can be used as typical wound dressings to accelerate the whole healing process due to them having a similar composition structure to skin, but they are also limited by ideal biocompatibility and stable mechanical properties. To extend the number of practical candidates in the field of wound healing, we designed a new structural zwitterion poly[3-(dimethyl(4-vinylbenzyl) ammonium) propyl sulfonate] (SVBA) into a poly-acrylamide network, with remarkable mechanical properties, stable rheological property, effective antibacterial properties, strong adsorption, high penetrability, and good electroactive properties. Both in vivo and in vitro evidence indicates biocompatibility, and strong healing efficiency, indicating that poly (AAm-co-SVBA) (PAS) hydrogels as new wound healing candidates with biomedical applications.

Irregular text recognition of natural scene is a challenging task due to large span of character angles and morphological diversity of a word. Recent work first rectifies curved word region, and then employ sequence algorithm to complete the recognition task. However, this strategy largely depends on rectification quality of the text region, and cannot be applied to large difference between tilt angles of character. In this work, a novel anchor-free network structure of rotating character detection is proposed, which includes multiple sub-angle domain branch networks, and the corresponding branch network can be selected adaptively according to character tilt angle. Meanwhile, a curvature Adaptive Text linking method is proposed to connect the discrete strings detected on the two-dimensional plane into words according to people's habits. We achieved state-of-the-art performance on two irregular texts (TotalText, CTW1500), outperforming state-of-the-art by 2.4% and 2.7%, respectively. The experimental results demonstrate the effectiveness of the proposed algorithm.

Due to high performance and low power consumption, heterogeneous processors are widely used in many real-time systems. In these systems, if tasks are not completed before deadline, it will cause disastrous consequences, and thus it is important to provide fault-tolerance. This paper proposes a novel, adaptive and transient fault-tolerant scheduling algorithm to solve the fault-tolerant problem in heterogeneous real-time systems, aiming to improve system reliability within a given deadline. Since task replication is efficient in improving system reliability, the proposed algorithm supports multiple replicas for each primary task and allows the primary tasks and their replicas to be scheduled on the same processor to increase reliability and lower latency. Also, the algorithm can dynamically adjust the number of replicas for each task to accommodate the deadline and ensure higher reliability. Simulated results show that the proposed algorithm can achieve higher reliability in comparison with existing and related fault-tolerant algorithms. To be specific, the proposed algorithm can obtain the reliability of 89.37% whereas the two existing algorithms DB-FTSA and FTSA obtain the reliability of 47.05% and 84.75% for the benchmark of sixty tasks, respectively, to be detailed in Fig. 4 in experiment.

Crowd behavior refers to a collective behavior composed of two or more individuals who influence, interact, and depend on each other for a specific goal. Compared with an ordinary crowd behavior, the probability of a dangerous crowd behavior is much smaller. Video-based crowd behavior recognition can be categorized as one multi-label classification task, which is characterized by complex scenes and imbalanced samples. Aimed at tackling problems of imbalanced samples and multi-label task, a classification method of associative subspace is proposed. For a single category (called main category) with fewer samples, this paper generates a special subspace wherein it is relatively easy to distinguish these samples by association with other categories. A classifier that can weaken the main category and strengthen relationship between the main category and other categories is designed in the subspace. Therefore, the main category can contribute to reducing dependence on the number of samples with the above-mentioned classifier in the corresponding subspace. In order to make full use of the relevant information concerning categories, multi-label information is further injected into spatio-temporal features of video action representation. Experiments on a challenging WWW dataset show that both the proposed subspace method and multi-label information fusion mechanism are efficient.

Recent studies showed that Escherichia coli (E. coli) strains isolated from captive giant pandas have serious resistance to antibiotics and carry various antibiotic resistance genes (ARGs). ARGs or virulence-associated genes (VAGs) carried by antibiotic-resistant E. coli are considered as a potential health threat to giant pandas, humans, other animals and the environment. In this study, we screened ARGs and VAGs in 84 antibiotic-resistant E. coli strains isolated from clinically healthy captive giant pandas, identified the association between ARGs and VAGs and analyzed the phylogenetic clustering of E. coli isolates. Our results showed that the most prevalent ARG in E. coli strains isolated from giant pandas is blaTEM (100.00%, 84/84), while the most prevalent VAG is fimC (91.67%, 77/84). There was a significant positive association among 30 pairs of ARGs, of which the strongest was observed for sul1/tetC (OR, 133.33). A significant positive association was demonstrated among 14 pairs of VAGs, and the strongest was observed for fyuA/iroN (OR, 294.40). A positive association was also observed among 45 pairs of ARGs and VAGs, of which the strongest was sul1/eaeA (OR, 23.06). The association of ARGs and mobile gene elements (MGEs) was further analyzed, and the strongest was found for flor and intI1 (OR, 79.86). The result of phylogenetic clustering showed that the most prevalent group was group B2 (67.86%, 57/84), followed by group A (16.67%, 14/84), group D (9.52%, 8/84) and group B1 (5.95%, 5/84). This study implied that antibiotic-resistant E. coli isolated from captive giant pandas is a reservoir of ARGs and VAGs, and significant associations exist among ARGs, VAGs and MGEs. Monitoring ARGs, VAGs and MGEs carried by E. coli from giant pandas is beneficial for controlling the development of antimicrobial resistance.

In mobile edge computing (MEC), mobile devices can choose to offload their tasks to edge servers for execution, thereby effectively reducing the completion time of tasks and energy consumption of mobile devices. However, most of the data transfer brought by offloading relies on wireless communication technology, making the private information of mobile devices vulnerable to eavesdropping and monitoring. Privacy leakage, especially the location and association privacies, can pose a significant risk to users of mobile devices. Therefore, protecting the privacy of mobile devices during task offloading is important and cannot be ignored. This paper considers both location privacy and association privacy of mobile devices during task offloading in MEC and targets to reduce the leakage of location and association privacy while minimizing the average completion time of tasks. To achieve these goals, we design a privacy-preserving task offloading scheme to protect location privacy and association privacy. The scheme is mainly divided into two parts. First, we adopt a proxy forwarding mechanism to protect the location privacy of mobile devices from being leaked. Second, we select the proxy server and edge server for each task that needs to be offloaded. In the proxy server selection policy, we make a choice based on the location information of proxy servers, to reduce the leakage risk of location privacy. In the edge server selection strategy, we consider the privacy conflict between tasks, the computing ability, and location of edge servers, to reduce the leakage risk of association privacy plus the average completion time of tasks as much as possible. Simulated experimental results demonstrate that our scheme is effective in protecting the location privacy and association privacy of mobile devices and reducing the average completion time of tasks compared with the-state-of-art techniques.

Coal gasification coarse slag (CGCS) is a by-product of coal gasification. Despite its abundance, CGCS is mostly used in boiler blending, stacking, and landfill. Large-scale industrial applications of CGCS can be environment-friendly and cost saving. In this study, the application of CGCS as a substitute for river sand (RS) with different replacement ratios in ultra-high performance concrete (UHPC) was investigated. The effects of CGCS replacement ratios on the fluidity and mechanical properties of specimens were examined, and the effect mechanisms were explored on the basis of hydration products and the multi-scale (millimetre-scale and micrometre-scale) microstructure analysis obtained through X-ray diffraction (XRD), scanning electron microscopy, and X-ray energy-dispersive spectroscopy. With an increase in the CGCS replacement ratio, the water–binder ratio (w/b), flexural strength, and compressive strength decreased. Specimens containing CGCS of ≤25% can satisfy the strength requirement of non-structural UHPC, with flexure strength of 29 MPa and compressive strength of 111 MPa at day 28. According to the XRD results and multi-scale microstructure analysis, amorphous glass beads in CGCS positively influenced ettringite generation due to the pozzolanic activity. Porous carbon particles in CGCS showed strong interfacial bonding with cement slurry due to internal hydration; this bonding was conducive to improving the mechanical strength. However, CGCS hindered hydration in the later curing stage, leading to an increase in the unreacted cement and agglomeration of fly ash; in addition, at a CGCS replacement ratio of up to 50%, an apparent interfacial transition zone structure was observed, which was the main contributor to mechanical strength deterioration.

In this paper, a series of numerical experiments are carried out on the anti-scour device combined with perforated baffle and ring-wing plate. In addition, the optimal dimensions and location of the combined device are obtained: The perforated ratio of the baffle is S = 20%, the distance from the center of the bridge pier is L = 2d (d is the diameter of the bridge pier), and the ring-wing plate is located at H = 1/3h (h is the water depth). To verify the effect of the anti-scour device, the scour characteristics and flow field are further investigated. Compared with single pier and single ring-wing plate, the results revealed that the combined device with the optimal dimensions is of great anti-scour performance. Moreover, the maximum scour depth at the front and side of the pier reduced by 84.20% and 78.95%, which is better than the single ring-wing plate and other combined conditions in the orthogonal experiments. Due to the diversion of perforated baffle and ring-wing plate, the flow velocity at the pier side near the bed surface decreases by 30.7%, and the down-flow is almost eliminated on the vertical plane. Furthermore, the turbulent kinetic energy at different horizontal and vertical planes is reduced due to the reduction in horseshoe vortex and wake flow. Based on the investigation presented herein, the combined device is a promising tool for mitigating scour around the bridge pier.

Purpose: This study used a moderated mediation model to explore the relationship between general self-efficacy (GSE) and psychological resilience (PR) and the associated mechanisms, the mediating role of posttraumatic growth (PTG), and the moderating role of deliberate rumination (DR) during the coronavirus disease 2019 (COVID-19) pandemic. Knowledge of the relationship between these four variables examined further understanding of the PR improvement mechanism of college students and even the general public.Methods: The college students who participated in this study came from an independent college in Guangdong Province, China. A total of 918 college students completed the survey, and the final data sample size was 881. SPSS 23.0 and PROCESS (version 3.3) were used to conduct Pearson's correlation analysis and hierarchical regression linear analysis on the data.Results: (1) The correlation analysis showed that GSE and PR were positively correlated and that PTG was positively correlated with GSE and PR. DR was positively correlated with GSE, PTG, and PR. (2) The results of mediation analysis showed that GSE had a direct predictive effect on DR, and PTG partially mediated the relationship between the two. (3) The results of moderating effect analysis showed that DR hindered the effect of GSE on PTG but enhanced the positive impact of PTG on PR.Conclusions: General self-efficacy can improve PR under the mediating influence of PTG. DR played a positive moderating role in the relationship between GSE and PTG, and played a negative moderating role in the relationship between PTG and PR. These results advance the understanding of the mechanism between GSE and PR.

Dielectric elastomers can generate large voltage-induced deformation and have been widely used as soft actuators. Dielectric elastomer multilayer structures function as bending-type actuators, enhancing their stiffness and ability to resist external loads. Modeling a dielectric elastomer multilayer bending actuator allows analyzing its bending performance, and obtaining the stress and strain distributions in each layer of the actuator. It is of practical importance to develop a simple, yet accurate model. This work develops a model to analyze the electromechanical behavior of the dielectric elastomer multilayer bending actuator with various pre-stretch, material, and structural parameters. We also propose a simplified model for a bilayer slender beam, which is a typical multilayer structure. The simplified model describes the relation between the beam bending curvature and the applied voltage, achieving fast calculation with little accuracy sacrificed. Both the model for the dielectric elastomer multilayer and the simplified model for the bilayer beam agree well with the experiment results, validating the models for practical applications in soft robots, flexible sensors, and smart structures.

Abstract Recurrent complex spinal tuberculosis accompanied by sinus tract formation is one of the most common and complex types of recurrent tuberculosis. To date, only very few studies have reported the strategies and effectiveness of surgical treatments on postoperative recurrent spinal tuberculosis accompanied by sinus tract formation. In this study, 21 recurrent patients out of 87 patients with complex spinal tuberculosis were reviewed. The data of the patients, including age, gender, existence of drug-resistant mycobacterium tuberculosis, postoperative standard chemotherapy, completeness of lesion debridement, reconstruction of the spinal stability, nutritional status, accompaniment by tuberculosis in other regions, timing of the operations, and areas of the lesions, were collected for single- and multiple-factor analyses. The clinical outcomes were evaluated by long-term follow-ups. The results showed that 7 factors were significantly associated with postoperative recurrence of complex spinal tuberculosis accompanied by sinus tract formation. This suggested that when we treat complex spinal tuberculosis, we should pay more attention to these seven indicators to avoid postoperative recurrence, and our clinical outcomes suggested that it is difficult to treat recurrent complicated spinal tuberculosis combined with sinus tract formation. The key for successful surgical treatment includes effective chemotherapy, radical debridement and proper reconstruction of spinal stability.

Cervicothoracic spinal tuberculosis is a rare disease. Due to its difficult and challenging surgical exposure, its surgical treatment approach remains inconclusive. Long-term follow-up studies to address this puzzling issue are rarely seen in the literature. The purpose of this study was to explore the selection of surgical treatment approaches for cervicothoracic spinal tuberculosis through a 10-year case review.From January 2003 to January 2013, 45 patients suffering from cervicothoracic spinal tuberculosis were treated surgically. According to the relation between the tuberculosis lesion segments and the suprasternal notch on sagittal MRI, 19 patients were treated with a single-stage anterior debridement, fusion and instrumentation approach, and the other 26 patients were treated with a single-stage anterior debridement and fusion, posterior fusion and instrumentation approach. The clinical efficacy was evaluated using statistical analysis based on the Cobb angle of kyphosis, the Neck Disability Index (NDI) and the Japanese Orthopedic Association (JOA) scoring system. The neurofunctional recovery was assessed by the American Spinal Injury Association (ASIA) system.All patients were followed up for 6.6 years on average (range 3-13 years). No instrumentation loosening, migration or breakage was observed during the follow-up. Prostate cancer is driven by androgen stimulation of the androgen receptor (AR). The next-generation AR antagonist, enzalutamide, prolongs survival, but resistance and lethal disease eventually prevail. Emerging data suggest that the glucocorticoid receptor (GR) is upregulated in this context, stimulating expression of AR-target genes that permit continued growth despite AR blockade. However, countering this mechanism by administration of GR antagonists is problematic because GR is essential for life. We show that enzalutamide treatment in human models of prostate cancer and patient tissues is accompanied by a ubiquitin E3-ligase, AMFR, mediating loss of 11β-hydroxysteroid dehydrogenase-2 (11β-HSD2), which otherwise inactivates cortisol, sustaining tumor cortisol concentrations to stimulate GR and enzalutamide resistance. Remarkably, reinstatement of 11β-HSD2 expression, or AMFR loss, reverses enzalutamide resistance in mouse xenograft tumors. Together, these findings reveal a surprising metabolic mechanism of enzalutamide resistance that may be targeted with a strategy that circumvents a requirement for systemic GR ablation.

Background/Aims: Osteoporosis is a metabolic bone disorder that tortures about millions of people worldwide. Recent study demonstrated agents derived from picrasma quassioides is a promising drug for targets multiple signaling pathways. However its potential in treatment of bone loss has not been fully understood. Methods: The bone marrow macrophages (BMMs) were cultured and induced with M-CSF and RANKL followed by picrasidine I (PI) treatment. Then the effects of PI on osteoclast formation were evaluated by counting tartrate-resistant acid phosphatase (TRAP)-positive multinucleated cells. Moreover, effects of PI on bone resorption activity of mature osteoclast were studied through bone resorption pit counting and actin ring structure analysis. Further, the involved potential signaling pathways cross-talking were investigated by performed Western blotting and quantitative real-time PCR examination. Results: Results demonstrated PI strongly inhibited RANKL induced osteoclast formation from its precursors. Mechanistically, the inhibitory effect of PI on osteoclast differentiation was due to the suppression of osteoclastogenic transcription factors, c-Fos and NFATc1. Moreover, PI markedly blocked the RANKL-induced osteoclastogenesis by attenuating MAPKs and NF-κB signaling pathways. In addition, PI decreased the ROS generation in osteoclast and osteoblast. Conclusion: Taken together our data demonstrate that PI has antiosteoclastogenic effect by inhibiting inflammation induced activation of MAPKs, NF-κB and ROS generation followed by suppressing the gene expression of c-Fos and NFATc1 in osteoclast precursors.
