Wound infections caused by Multidrug-resistant Staphylococcus aureus (MRSA) have been regarded as a challenging problem in clinic for the long time. In this study, based on the excellent antimicrobial effect of prodigiosin(PG) and the ability of hydrogel dressing in terms of tissue repair and regeneration, we prepared the PG hydrogel as a treatment for the wound infection induced by MRSA. Rheological tests indicated that PG hydrogel as a semi-solid gel had good mechanical properties. In ex vitro drug permeation studies and dermatokinetic studies showed that PG hydrogel had high PG permeability and were capable of short-term retention in the skin. In addition, in vivo experiments for mouse skin wounds showed that the serum levels of inflammatory factors including IL-β and other inflammatory factors were reduced, the inflammatory infiltration of tissues was reduced, the transcript levels of genes such as COL1A1 were up-regulated at different stages of wound healing, and the relative abundance of genera such as Desulfovibrio was lowered after treatment with PG hydrogel, which facilitated wound healing in mice. Our study would provide a new solution to the clinical shortage of drugs for the treatment of MRSA infection and provide a research basis for improving the comprehensive values of PG.

INTRODUCTION: Significant advances have been made in photovoltaic (PV) systems, resulting in the development of new Maximum Power Point Tracking (MPPT) methods. The output of PV systems is heavily influenced by the varying performance of solar-facing PV panels under different weather conditions. Partial shading (PS) conditions pose additional challenges, leading to multiple peaks in the power-voltage (P-V) curve and reduced output power. Therefore, controlling MPPT under partial shading conditions is a complex task. OBJECTIVES: This study aims to introduce a novel MMPT algorithm based on the ant colony incorporated bald eagle search optimization (AC-BESO) method to enhance the efficiency of PV systems. METHODS: The effectiveness of the proposed MPPT algorithm was established through a series of experiments using MATLAB software, tested under various levels of solar irradiance. RESULTS: Compared to existing methods, the proposed AC-BESO algorithm stands out for its simplicity in implementation and reduced computational complexity. Furthermore, its tracking performance surpasses that of conventional methods, as validated through comparative analyses. CONCLUSION: This study confirms the efficacy of the AC-BESO method over traditional strategies. It serves as a framework for selecting an MPPT approach when designing PV systems.

INTRODUCTION: Photovoltaic (PV) energy sources frequently experience issues, including fragmentation, open-circuit, short-circuiting, and other common and hazardous problems. The current focus of PV research is on fault detection within solar arrays. Traditional models encounter challenges in identifying errors due to uncertainties in panel settings and the complex nature of the actual PV structure. OBJECTIVES: This study aims to introduce a novel Extreme Gradient Boosting (XGBoost) approach for fault diagnosis in PV arrays. METHODS: The XGBoost algorithm is trained using collected PV array defect data samples. Data preprocessing is performed to manage missing values and remove noisy data. Feature extraction is conducted using Linear Discriminant Analysis (LDA) to improve detection accuracy. To further enhance XGBoost’s performance, the World Cup Optimization (WCO) approach is applied to select optimal features from the extracted data. Fault detection is then conducted using the XGBoost algorithm on the processed data. Various indicators are utilized for performance assessment within the Python environment. RESULTS: The comparative analysis demonstrates that this research improves fault detection efficiency in PV arrays compared to existing methodologies. CONCLUSION: The study presents an effective method for enhancing fault detection in PV systems, showcasing the advantages of the XGBoost and WCO-based approach over conventional methods.

Reliable long-term settlement prediction of a high embankment relates to mountain infrastructure safety. This study developed a novel hybrid model (NHM) that combines a joint denoising technique with an enhanced gray wolf optimizer (EGWO)-ν-support vector regression (ν-SVR) method. High-embankment field measurements were preprocessed using the joint denoising technique, which includes complete ensemble empirical mode decomposition, singular value decomposition, and wavelet packet transform. Furthermore, high-embankment settlements were predicted using the EGWO-ν-SVR method. In this method, the standard gray wolf optimizer (GWO) was improved to obtain the EGWO to better tune the ν-SVR model hyperparameters. The proposed NHM was then tested in two case studies. Finally, the influences of the data division ratio and kernel function on the EGWO-ν-SVR forecasting performance and prediction efficiency were investigated. The results indicate that the NHM suppresses noise and restores details in high-embankment field measurements. Simultaneously, the NHM outperforms other alternative prediction methods in prediction accuracy and robustness. This demonstrates that the proposed NHM is effective in predicting high-embankment settlements with noisy field measurements. Moreover, the appropriate data division ratio and kernel function for EGWO-ν-SVR are 7:3 and radial basis function, respectively.

Abstract The high and steep slopes along a high-speed railway in the mountainous area of Southwest China are mostly composed of loose accumulations of debris with large internal pores and poor stability, which can easily induce adverse geological disasters under rainfall conditions. To ensure the smooth construction of the high-speed railway and the subsequent safe operation, it is necessary to master the stability evolution process of the loose accumulation slope under rainfall. This article simulates rainfall using the finite element analysis software’s hydromechanical coupling module. The slope stability under various rainfall situations is calculated and analysed based on the strength reduction method. To validate the simulation results, a field monitoring system is established to study the deformation characteristics of the slope under rainfall. The results show that rainfall duration is the key factor affecting slope stability. Given a constant amount of rainfall, the stability of the slope decreases with increasing duration of rainfall. Moreover, when the amount and duration of rainfall are constant, continuous rainfall has a greater impact on slope stability than intermittent rainfall. The setting of the field retaining structures has a significant role in improving slope stability. The field monitoring data show that the slope is in the initial deformation stage and has good stability, which verifies the rationality of the numerical simulation method. The research results can provide some references for understanding the influence of rainfall on the stability of loose accumulation slopes along high-speed railways and establishing a monitoring system.

Summary: HS3ST3B1-IT1 was identified as a downregulated long noncoding RNA in osteoarthritic cartilage. However, its roles and mechanisms in the pathogenesis of osteoarthritis (OA) are unclear. In this study, we demonstrated that the expressions of HS3ST3B1-IT1 and its maternal gene HS3ST3B1 were downregulated and positively correlated in osteoarthritic cartilage. Overexpression of HS3ST3B1-IT1 significantly increased chondrocyte viability, inhibited chondrocyte apoptosis, and upregulated extracellular matrix (ECM) proteins, whereas HS3ST3B1-IT1 knockdown had the opposite effects. In addition, HS3ST3B1-IT1 significantly ameliorated monosodium-iodoacetate-induced OA in vivo. Mechanistically, HS3ST3B1-IT1 upregulated HS3ST3B1 expression by blocking its ubiquitination-mediated degradation. Knockdown of HS3ST3B1 reversed the effects of HS3ST3B1-IT1 on chondrocyte viability, apoptosis, and ECM metabolism. AlkB homolog 5 (ALKBH5)-mediated N6-methyladenosine (m6A) demethylation stabilized HS3ST3B1-IT1 RNA. Together, our data revealed that ALKBH5-mediated upregulation of HS3ST3B1-IT1 suppressed OA progression by elevating HS3ST3B1 expression, suggesting that HS3ST3B1-IT1/HS3ST3B1 may serve as potential therapeutic targets for OA treatment.

Prodigiosin (PG) is a secondary metabolite produced by Serratia marcescens which has a promising future in food, textile, and other industries due to its bright color and diverse biological activities. Currently, the production of PG is mainly restricted by the components of the fermentation medium and large losses during its extraction process, making large-scale industrial production impossible. In this study, a Box–Behnken design (BBD) was used to optimize the response surface of the fermentation medium of S. marcescens. The optimum medium composition was found to be sucrose, 16.29 g/L; peptone, 11.76 g/L; and tween 80, 2.64 g/L. This composition produced a PG amount of 1653.95 ± 32.12 mg/L, which is a 64-fold increase compared to the initial medium. A Box–Behnken design (BBD) was then used to optimize the response surface of the extraction process of PG, aiming to reduce loss during extraction. The optimal extraction conditions were determined to be a solvent fermentation liquid ratio of 9.12:1, an extraction temperature of 25.35 °C, and an extraction time of 30.33 min. These conditions resulted in a final PG production amount of 2142.75 ± 12.55 mg/L, which was nearly 84 times higher than the initial production amount of PG. These results provide essential theoretical and experimental support for the industrial production of PG.

The measurement of the pressure in the gas generator (GG) plays a decisive role in the closed-loop regulation of the gas flow in a solid ducted rocket ramjet (SDR). Therefore, the fault detection and isolation (FDI) of the pressure sensors is particularly significant. Especially in the GG with double pressure feedback, there are issues such as less available data and sensor fault transmission, which make the FDI more difficult. In this paper, for the GG with double pressure feedback, firstly, the “Consistency Index” was constructed based on the principal component analysis (PCA) algorithm, which made it easier to evaluate the consistency of the measured values. Secondly, based on Kalman’s principle, the redundancy information of the system was used to estimate the pressure in the GG. Finally, the gap metric between the measured pressure and the estimated pressure was employed to characterize the health of the sensors. Compared with the MWPCA algorithm, it was shown that our proposed algorithm was more accurate in fault diagnosis and could avoid the problem of missing alarm when two sensors had consistent faults, which would provide strong support for the safe operation of the SDR and could further promote its application in long-endurance aircrafts.

To ensure efficient railroad operation and maintenance management, the accurate reconstruction of railroad BIM models is a crucial step. This paper proposes a workflow for automated segmentation and reconstruction of railroad structures using point cloud data, without relying on intensity or trajectory information. The workflow consists of four main components: point cloud adaptive denoising, scene segmentation, structure segmentation combined with deep learning, and model reconstruction. The proposed workflow was validated using two datasets with significant differences in railroad line point cloud data. The results demonstrated significant improvements in both efficiency and accuracy compared to existing methods. The techniques enable direct automated processing from raw data to segmentation results, providing data support for parameterized modeling and greatly reducing manual processing time. The proposed algorithms achieved an intersection over union (IoU) of over 0.9 for various structures in a 450-m-long railroad line. Furthermore, for single-track railroads, the automated segmentation time was within 1 min per kilometer, with an average mean intersection over union (MIoU) and accuracy of 0.9518 and 1.0000, respectively.

Autonomous underwater manipulation is very important for the robotic and intelligence operations of oceanic engineering. However, a small target often involves limited features and results in inaccurate visual matching. In order to improve visual measurement accuracy, this paper has proposed an improved unsharp masking algorithm to further enhance the weak texture region of blurred and low contrast images. Moreover, an improved ORB feature-matching method with adaptive threshold, non-maximum suppression and improved random sample consensus has also been proposed. To overcome unknown underwater disturbances and uncertain system parameters in the underwater robotic manipulations, an adaptive non-singular terminal sliding mode controller has been proposed with a quasi-barrier function to suppress the chattering problem and improve grasp accuracy for small target. Oceanic experiments have been conducted to prove the performance of the proposed method.

The genetic adaptations to harsh climatic conditions in high altitudes and genetic basis of important agronomic traits are poorly understood in Elymus sibiricus L. In this study, an association population of 210 genotypes was used for population structure, selective sweep analysis, and genome-wide association study (GWAS) based on 88,506 single nucleotide polymorphisms (SNPs). We found 965 alleles under the natural selection of high altitude, which included 7 hub genes involved in the response to UV, and flavonoid and anthocyanin biosynthetic process based on the protein–protein interaction (PPI) analysis. Using a mixed linear model (MLM), the GWAS test identified a total of 1,825 significant loci associated with 12 agronomic traits. Based on the gene expression data of two wheat cultivars and the PPI analysis, we finally identified 12 hub genes. Especially, in plant height traits, the top hub gene (TOPLESS protein) encoding auxins and jasmonic acid signaling pathway, shoot apical meristem specification, and xylem and phloem pattern formation was highly overexpressed. These genes might play essential roles in controlling the growth and development of E. sibiricus. Therefore, this study provides fundamental insights relevant to hub genes and will benefit molecular breeding and improvement in E. sibiricus and other Elymus species.

Flowering is a significant stage from vegetative growth to reproductive growth in higher plants, which impacts the biomass and seed yield. To reveal the flowering time variations and identify the flowering regulatory networks and hub genes in Elymus sibiricus, we measured the booting, heading, and flowering times of 66 E. sibiricus accessions. The booting, heading, and flowering times varied from 136 to 188, 142 to 194, and 148 to 201 days, respectively. The difference in flowering time between the earliest- and the last-flowering accessions was 53 days. Furthermore, transcriptome analyses were performed at the three developmental stages of six accessions with contrasting flowering times. A total of 3,526 differentially expressed genes (DEGs) were predicted and 72 candidate genes were identified, including transcription factors, known flowering genes, and plant hormone-related genes. Among them, four candidate genes (LATE, GA2OX6, FAR3, and MFT1) were significantly upregulated in late-flowering accessions. LIMYB, PEX19, GWD3, BOR7, PMEI28, LRR, and AIRP2 were identified as hub genes in the turquoise and blue modules which were related to the development time of flowering by weighted gene co-expression network analysis (WGCNA). A single-nucleotide polymorphism (SNP) of LIMYB found by multiple sequence alignment may cause late flowering. The expression pattern of flowering candidate genes was verified in eight flowering promoters (CRY, COL, FPF1, Hd3, GID1, FLK, VIN3, and FPA) and four flowering suppressors (CCA1, ELF3, Ghd7, and COL4) under drought and salt stress by qRT-PCR. The results suggested that drought and salt stress activated the flowering regulation pathways to some extent. The findings of the present study lay a foundation for the functional verification of flowering genes and breeding of new varieties of early- and late-flowering E. sibiricus.

Performing object tracking tasks and efficiently perceiving the underwater environment in real time for underwater vehicles is a challenging task due to the complex nature of the underwater environment. A hybrid excitation model based lightweight Siamese network is proposed to solve the mismatch between underwater objects with limited characteristics and complex deep learning models. The lightweight neural network is applied to the residual network in the Siamese network to reduce the computational complexity and cost of the model while constructing a deeper network. In addition, to deal with the changeable complex underwater environment and consider the timing of video tracking, the global excitation model (HE module) is introduced. The model adopts the excitation methods of space, channel, and motion to improve the accuracy of the algorithm. Based on the designed underwater vehicle, the underwater target tracking and target grabbing experiments are carried out, and the experimental results show that the proposed tracking algorithm has a high tracking success rate.

Variable-throat adjustment is the most practical flow regulation method of solid ducted rocket ramjet (SDR). The high-fidelity mathematical model of the interstage valve is the basis for realizing high-precision gas flow and thrust regulation. In this paper, the complex effect of gas was divided into load and throat deformation effect. The load was mainly determined by the clearance, friction torque, and pneumatic torque that the valve was subjected to during operation. And the throat deformation was determined primarily by the deposition and ablation of the valve faced in the gas. Therefore, we could divide the valve model into three parts: the servo motor model, the load characteristic model, and the deformation model of the actual acting throat (referred to as the throat). Given, we have designed a cold-air experiment program, using cold air to equalize the valve load. Furthermore, we analyzed its mechanism of action and established the load model using the experimental data and neural network. Finally, the deformation mechanism of the throat was investigated, and simultaneously, the deformation model was shown based on the flight test data. Compared with the traditional interstage valve model, the model established in this paper is closer to the actual working conditions, which is helpful to carry out the more comprehensive and practical ground simulation. It has essential reference value for further realizing the precise regulation of gas flow.

The solid ducted rocket ramjet (SDR) system faces many disturbances in the process of operation, and the linear active disturbance rejection controller (LADRC) has been widely used in engineering to solve such problems. However, the SDR also has strong nonlinearity, which brings a great challenge to the application of the LADRC in the gas flow regulation of the SDR. And the problem of fast adjustment of the “compensation factor” is one of the main difficulties in LADRC. In this paper, under the LADRC frame, the gas generator system’s closed-loop stability of the SDR was analyzed and the range of compensation factors had been calculated, and then, the “gap factor” was introduced and the “cross-iteration” method was used to quickly map out the “compensation factor” in the multilinear model controller based on the variation of the zero-point position of the system and the gap metric between adjacent set points. This greatly simplifies the parameter tuning process of the LADRC when it was applied to strongly nonlinear systems. Finally, through the comparison of simulation with adaptive PI controller and model-assisted LADRC (M-LADRC), the results have shown that the control method designed in this paper can obtain satisfactory performance and has a good engineering application prospect.

The Rover-mounted Subsurface Penetrating Radar (RoSPR) is one of the scientific payloads onboard China’s first independent Mars exploration mission, Tianwen-1. The radar aims to characterize the thickness of the upper Martian soil and investigate the subsurface stratigraphy by collecting and processing the data. This article is mainly divided into two parts, the introduction of data pre-processing and analysis of pre-processed radar signals, aiming at helping scientists make more effective use of radar data. The first part describes the operating principle of the RoSPR and the procedure of radar data pre-processing at all levels. Data pre-processing is mainly designed to transfer the raw data format to a common PDS (Planetary Data System) and eliminate the influence of the instrument. In the signal analysis part, the performances of both self-check signals and echo signals of low- and high-frequency channels are analyzed, which indicate a stable radar system and are useful for background removal. Phase and time calibration is of great importance for improving data quality and making the radar data more accurate. Moreover, further processing is required to obtain clear radar images, such as filtering, background removal and gain setting.

ObjectiveOur aim was to conduct a systematic review and meta-analysis to assess the effectiveness and safety of tea supplements for patients with polycystic ovary syndrome (PCOS).MethodsWe conducted searches of the published literature in PubMed, EMBASE, Cochrane Library, Web of Science, Chinese Biomedical Literature Database, Chinese National Knowledge Infrastructure (CNKI), VIP database, and Wanfang Database in 1985 to September 2021. Data from randomized controlled trials (RCTs) were obtained to assess the effects of tea versus placebo in women with PCOS. Weighted mean differences (WMDs) were pooled using a random-effects model or risks ratios (RRs) using a random-effects model.ResultsSix RCTs (235 participants) were included in our systematic review. Tea supplements as adjuvant therapy led to greater improvement in body weight (WMD −2.71, 95% CI −4.95 to −0.46, P = 0.02, I2 = 0%), fasting blood glucose (FBG: WMD −0.40, 95% CI −0.59 to −0.20, P < 0.0001, I2 = 0%) and fasting insulin (FINS: WMD −3.40, 95% CI −4.76 to −2.03, P < 0.00001, I2 = 0%) when compared with placebo. There were no significant differences of body mass index, waist circumference, hip circumference, waist-to-hip ratio (WHR), body fat rate, total testosterone, free testosterone (FT), dehydroepiandrosterone, luteinizing hormone or follicular-stimulating hormone (FSH) between the two groups. In addition, subgroup analysis suggested that green tea was effective on body weight, FINS, FBG, FT, and FSH, and herbal tea can also reduce FT levels, tea supplements had a significant impact on FBG and FSH in trials with intervention duration ≥ 3 months, and intervention lasting less than 3 months can improve FINS. Tea had significant effect on reducing WHR, FBG and FSH in Asian PCOS patients, but not in Caucasians. And there was no statistically significant effect of tea on weight and FINS in Asians, but it was effective for Caucasian participants. Compared with placebo, tea supplements did not cause significant adverse reactions (RR 1.45, 95% CI 0.30 to 6.90, P = 0.65, I2 = 0%).ConclusionThis meta-analysis suggests that consumption of tea supplementation in women with PCOS could significantly decrease the levels of FBG and FINS as well as reduce body weight. Especially green tea, not only has the above effects, but also has a significant effect on improving a variety of reproductive hormone indexes. Furthermore, tea supplementation is a relatively safe therapy for PCOS patients. Abstract Background Elymus sibiricus is an ecologically and economically important perennial, self-pollinated, and allotetraploid (StStHH) grass, widely used for forage production and animal husbandry in Western and Northern China. However, it has low seed yield mainly caused by seed shattering, which makes seed production difficult for this species. The goals of this study were to construct the high-density genetic linkage map, and to identify QTLs and candidate genes for seed-yield related traits. Results An F2 mapping population of 200 individuals was developed from a cross between single genotype from “Y1005” and “ZhN06”. Specific-locus amplified fragment sequencing (SLAF-seq) was applied to construct the first genetic linkage map. The final genetic map included 1971 markers on the 14 linkage groups (LGs) and was 1866.35 cM in total. The length of each linkage group varied from 87.67 cM (LG7) to 183.45 cM (LG1), with an average distance of 1.66 cM between adjacent markers. The marker sequences of E. sibiricus were compared to two grass genomes and showed 1556 (79%) markers mapped to wheat, 1380 (70%) to barley. Phenotypic data of eight seed-related traits (2016–2018) were used for QTL identification. A total of 29 QTLs were detected for eight seed-related traits on 14 linkage groups, of which 16 QTLs could be consistently detected for two or three years. A total of 6 QTLs were associated with seed shattering. Based on annotation with wheat and barley genome and transcriptome data of abscission zone in E. sibiricus, we identified 30 candidate genes for seed shattering, of which 15, 7, 6 and 2 genes were involved in plant hormone signal transcription, transcription factor, hydrolase activity and lignin biosynthetic pathway, respectively. Conclusion This study constructed the first high-density genetic linkage map and identified QTLs and candidate genes for seed-related traits in E. sibiricus. Results of this study will not only serve as genome-wide resources for gene/QTL fine mapping, but also provide a genetic framework for anchoring sequence scaffolds on chromosomes in future genome sequence assembly of E. sibiricus.

Abstract Background Elymus L. is the largest genus in the tribe Triticeae Dumort., encompassing approximately 150 polyploid perennial species widely distributed in the temperate regions of the world. It is considered to be an important gene pool for improving cereal crops. However, a shortage of molecular marker limits the efficiency and accuracy of genetic breeding for Elymus species. High-throughput transcriptome sequencing data is essential for gene discovery and molecular marker development. Results We obtained the transcriptome dataset of E. sibiricus, the type species of the genus Elymus, and identified a total of 8871 putative EST-SSRs from 6685 unigenes. Trinucleotides were the dominant repeat motif (4760, 53.66%), followed by dinucleotides (1993, 22.47%) and mononucleotides (1876, 21.15%). The most dominant trinucleotide repeat motif was CCG/CGG (1119, 23.5%). Sequencing of PCR products showed that the sequenced alleles from different Elymus species were homologous to the original SSR locus from which the primer was designed. Different types of tri-repeats as abundant SSR motifs were observed in repeat regions. Two hundred EST-SSR primer pairs were designed and selected to amplify ten DNA samples of Elymus species. Eighty-seven pairs of primer (43.5%) generated clear and reproducible bands with expected size, and showed good transferability across different Elymus species. Finally, thirty primer pairs successfully amplified ninety-five accessions of seventeen Elymus species, and detected significant amounts of polymorphism. In general, hexaploid Elymus species with genomes StStHHYY had a relatively higher level of genetic diversity (H = 0.219, I = 0.330, %P = 63.7), while tetraploid Elymus species with genomes StStYY had low level of genetic diversity (H = 0.182, I = 0.272, %P = 50.4) in the study. The cluster analysis showed that all ninety-five accessions were clustered into three major clusters. The accessions were grouped mainly according to their genomic components and origins. Conclusions This study demonstrated that transcriptome sequencing is a fast and cost-effective approach to molecular marker development. These EST-SSR markers developed in this study are valuable tools for genetic diversity, evolutionary, and molecular breeding in E. sibiricus, and other Elymus species.

Variable flow ducted rockets (VFDRs) are promising candidates for propulsion systems in hypersonic vehicles because of their inherent advantages, such as high specific impulse, low weight, and high speed. The control of gas flow is essential for optimal VFDRs performance. However, the characteristics of gas flow regulation systems, such as anti-regulation, non-linearity, and parameter variation, make it difficult to construct gas flow controllers. Aiming at the above problems, we propose a compound control strategy integrating a novel second-order fuzzy adaptive tracking differentiator (SOA-TD) and an intelligent proportional-integral controller based on adaptive neuro-fuzzy inference system (ANFIS). First, a mathematical model of a gas flow regulation system was developed to analyze the control characteristics of VFDRs. Next, an ANFIS-based proportional-integral controller to developed to respond to the system’s time-varying characteristics. In addition, a novel SOA-TD was constructed to optimize the “arrange transient process” of instructions, which effectively suppressed anti-regulation of the gas flow without increasing response time. Finally, a hardware in loop (HIL) simulation device for VFDRs was established, and serial HIL simulation tests were carried out to verify the validation of the controller. The HIL simulation results indicate that our strategy exhibited a superior performance compared to traditional controllers in terms of adaptability, ability to suppress anti-regulation, and robustness, which is hoped to fulfill VFDRs’ thrust control requirements for a wide range of altitudes and Mach numbers in future engineering applications.

The Chinese Chang’E-5 probe landed in the Mons Rümker of Oceanus Procellarum on the near side of the Moon. The lunar regolith penetrating radar (LRPR) carried by the Chang’E-5 probe allows for the determination of in situ lunar regolith dielectric properties, which are probably related to the age and chemical composition of the regolith. In this paper, we analyze the Chang’E-5 LRPR data with the frequency shift method to estimate the loss tangent of the lunar regolith within a depth of ∼2.8 m. The loss tangent of the Chang’E-5 landing site is constrained to be 0.0148 ± 0.0016, which is substantially higher than that of the typical lunar regolith. The high loss tangent is found to be characteristic of the young basalt age (∼2.0 Ga) and high TiO2+FeO content (28.21 ± 1.57%) of the Chang’E-5 landing site. Integrated analysis of results from Chang’E-3, Chang’E-4, and Chang’E-5 show that the younger is the geologic age of the mare unit, the greater is the loss tangent of the lunar regolith, and the weaker is the radar electromagnetic signal penetrating ability.

A multi-channel pressure scanner is an essential tool for measuring and acquiring various pressure parameters in aerospace applications. It is important to note, however, that the pressure sensor of each of these channels will drift significantly with the increase in the temperature range of the pressure measurement, and the output voltage of each of these channels will show nonlinear characteristics, which will constrain the improvements in the accuracy of the measurement. In the regression fitting process, it is difficult to fit nonlinear data with the traditional least-squares method, which leaves pressure measurement accuracy unsatisfactory. A temperature compensation method based on an improved cuckoo search optimizing a BP neural network for a multi-channel pressure scanner is proposed in this paper to improve pressure measurement accuracy in a wide temperature range. Using the chaotic simplex algorithm, we first improved the cuckoo search algorithm, then optimized the connection weights and thresholds of the BP neural network, and finally constructed an experimental calibration system to investigate the temperature compensation of the multi-channel pressure scanning valves in the −40 °C to 60 °C temperature range. The compensation test results show that the algorithm has a better compensation effect and is more suitable for the temperature compensation of multi-channel pressure scanners than the traditional least-squares method and the standard RBF and BP neural networks. The maximum full-scale error of all 32 channels is 0.02% FS (full-scale error) and below, which realizes its high-accuracy multi-point pressure measurement in a wide temperature range.

Abstract Background Elymus nutans and E. sibiricus are two important forage grasses of the genus Elymus. But they are difficult to grow for commercial seed production due to serious seed shattering. We conducted a comparative transcriptome analysis of abscission zone to find possible transcription changes associated with seed shattering, explore candidate genes involved in seed shattering and identify candidate gene-based EST-SSR markers for germplasm evaluation. Results cDNA libraries from abscission zone (AZ) and non-abscission zone (NAZ) tissues of E. nutans were constructed and sequenced. A total of 111,667 unigenes were annotated and 7644 differentially expressed transcripts (DETs) were predicted, corresponding to 6936 up-regulated in AZ and 708 down-regulated in NAZ. We identified 489 candidate genes related to transcription factor, cell wall hydrolysis or modification, hydrolase activity, phytohormone signaling and response, lignin biosynthesis, and signal transduction or protein turnover. Eleven similar candidate genes involved in polygalacturonase activity, hydrolase activity, and mitogen-activated protein kinase were up-regulated in the abscission zone of the two Elymus species, suggesting these genes may have specific function for abscission zone development and seed shattering. A total of 67 polymorphic EST-SSR markers were developed and characterized based on the sequences of these candidate genes. Fourteen polymorphic EST-SSR primers were finally used to study genetic diversity in 48 E. nutans genotypes with contrasting seed shattering habit. The dendrogram based on molecular data showed that most accessions with similar seed shattering degree tended to group together. Conclusions The expression data generated from this study provides an important resource for future molecular biological research. Many DETs were associated with abscission zone development, and EST-SSR loci related to candidate genes may have potential application in identifying trait-associated markers in E. nutans in the future.

Abstract Background Anterior cervical discectomy and fusion (ACDF) is the classic surgical treatment for symptomatic cervical degenerative disc disease (CDDD). However, there is controversy over the best surgical management in patients with two noncontiguous symptomatic levels of CDDD. Methods From April 2011 to May 2014, 44 patients with two noncontiguous symptomatic levels of CDDD underwent skip-level ACDFs. In Group NoPlate, 23 cases underwent 2 noncontiguous levels of ACDF using zero-profile anchored spacer; and in Group Plate, 21 cases underwent 2 noncontiguous levels of ACDF using cages and plates. Operation-related paraeters for each group were recorded and compared. Japanese Orthopedic Association (JOA) scores and Neck Disability Index (NDI) scores at preoperation and postoperation were compared with at least a 2-year follow-up. Cervical lordosis was analyzed before surgery, 1 month after surgery, 3 months after surgery, and at final follow-up. Results Mean follow-up was 35.4 ± 6.5 (range 24–48) months. Abstract Background Very severe osteoporotic vertebral compression fractures (vsOVCFs) are osteoporotic vertebral compression fractures with vertebral body collapse to less than one third of their original height. Few data are available about the use of percutaneous kyphoplasty (PKP) in treating vsOVCFs with spinal canal compromise. The aim of this study was to evaluate the safety and efficacy of percutaneous kyphoplasty (PKP) for the treatment of vsOVCFs with spinal canal compromise. Methods Thirty-five patients who suffered vsOVCFs with spinal canal compromise but without neurological deficits were treated by PKP between January 2009 and October 2014. The MOSIR (Mars Orbiter Subsurface Investigation Radar) is one of the scientific payloads carried by the Tianwen-1 orbiter. MOSIR conducted a ground experiment in the desert near Dengkou County, northern China, before the launch of the Tianwen-1 satellite. The MOSIR prototype was suspended from a hot air balloon and flew over a flat region at an altitude of 2500–3300 m. This experiment aimed to verify the system performance and data processing. The data collected in subsurface sounding mode is performed range compression, and the altitude measurement data removes invalid data. After processing, the altitude measurement results of two operating modes are analyzed and compared with that of the Global Position System (GPS), which verifies the accuracy of the altitude measurement.

Abstract Background Elymus sibiricus is an important native grass in Qinghai-Tibetan Plateau. Seed shattering is a serious problem for E. sibiricus, especially at harvest time. Cross breeding is an effective way to create new varieties with beneficial characteristic or improved traits, and to broaden genetic base. Results In this study, we created five hybrid populations by crossing seven E. sibiricus genotypes that have seed shattering variation. Then, nine EST-SSR primers were used for hybrid identification based on DNA fingerprinting, and genetic diversity analysis of hybrid populations and their respective parents. A total of 15 hybrids were identified. An analysis of amplified polymorphic bands among genuine hybrids and their respective parents revealed mainly two types of markers: 1) hybrids shared bands exclusively amplified in both parents; 2)hybrids shared bands exclusively amplified in male parents. For each hybrid population, the total number of amplified bands ranged from 37 to 57, the percentage of polymorphism varied from 65.12% to 75.68%, with an average of 70.51%. Novel bands found in each hybrid population varied from 0 to 9 bands, suggesting an occurrence of rearrangements in the hybrid population. The structure analysis revealed that all hybrid populations and parents were assigned to eight groups. The principal coordinate analysis (PCoA) showed similar results. Conclusions In general, this study proved EST-SSR markers are efficient for hybrid identification, and suggested more genetic variation could be captured in hybrid populations by crossing breeding.

Abstract Background Elymus sibiricus is an important forage grass in semi-arid regions, but it is difficult to grow for commercial seed production due to high seed shattering. To better understand the underlying mechanism and explore the putative genes related to seed shattering, we conducted a combination of morphological, histological, physiochemical and transcriptome analysis on two E. sibiricus genotypes (XH09 and ZhN03) that have contrasting seed shattering. Results The results show that seed shattering is generally caused by a degradation of the abscission layer. Early degradation of abscission layers was associated with the increased seed shattering in high seed shattering genotype XH09. Two cell wall degrading enzymes, cellulase (CE) and polygalacturonase (PG), had different activity in the abscission zone, indicating their roles in differentiation of abscission layer. cDNA libraries from abscission zone tissue of XH09 and ZhN03 at 7 days, 21 days and 28 days after heading were constructed and sequenced. A total of 86,634 unigenes were annotated and 7110 differentially expressed transcripts (DETs) were predicted from “XH09-7 vs ZhN03-7”, “XH09-21 vs ZhN03-21” and “XH09-28 vs ZhN03-28”, corresponding to 2058 up-regulated and 5052 down-regulated unigenes. The expression profiles of 10 candidate transcripts involved in cell wall-degrading enzymes, lignin biosynthesis and phytohormone activity were validated using quantitative real-time PCR (qRT-PCR), 8 of which were up-regulated in low seed shattering genotype ZhN03, suggesting these genes may be associated with reduction of seed shattering. Conclusions The expression data generated in this study provides an important resource for future molecular biological research in E. sibiricus.

Accurate relative permittivity is essential to the further analysis of lunar regolith. The traditional hyperbola fitting method for the relative permittivity estimation using the lunar penetrating radar generally ignored the effect of the position and geometry of antennas. This paper proposed a new approach considering the antenna mounting height and spacing in more detail. The proposed method is verified by numerical simulations of the regolith models. Hence the relative permittivity of the lunar regolith is calculated using the latest high-frequency radar image obtained by the Yutu-2 rover within the first 24 lunar days. The simulation results show that the relative permittivity is underestimated when derived by the traditional method, especially at the shallow depth. The proposed method has improved the accuracy of the estimated lunar regolith relative permittivity at a depth of 0–3 m, 3–6 m, and 6–10 m by 35%, 14%, and 9%, respectively. The thickness of the lunar regolith at the Chang’E 4 landing site is reappraised to be 11.1 m, which improved by ~8% compared with previous studies.

Elymus sibiricus L. is an important cold-season grass with excellent cold and drought tolerance, good palatability, and nutrition. Flowering time is a key trait that affects forage and seed yield. Development of EST-SSR (expressed sequence tag simple sequence repeat) markers based on flowering genes contributes to the improvement of flowering traits. In the study, we detected 155 candidate genes related to flowering traits from 10,591 unigenes via transcriptome sequencing in early- and late-flowering genotypes. These candidate genes were mainly involved in the photoperiodic pathway, vernalization pathway, central integrator, and gibberellin pathway. A total of 125 candidate gene-based EST-SSRs were developed. Further, 15 polymorphic EST-SSRs closely associated to 13 candidate genes were used for genetic diversity and population structure analysis among 20 E. sibiricus accessions, including two contrasting panels (early-flowering and late-flowering). Among them, primer 28366, designed from heading date 3a (HD3a), effectively distinguished early- and late-flowering genotypes using a specifically amplified band of 175 bp. The polymorphic information content (PIC) value ranged from 0.12 to 0.48, with an average of 0.25. The unweighted pair group method analysis (UPGMA) cluster and structure analysis showed that the 20 E. sibiricus genotypes with similar flowering times tended to group together. These newly developed EST-SSR markers have the potential to be used for molecular markers assisted selection and germplasm evaluation of flowering traits in E. sibiricus.

Siberian wild rye (Elymus sibiricus L.) is an important native grass in the Qinghai-Tibet Plateau of China. It is difficult to grow for commercial seed production, since seed shattering causes yield losses during harvest. Assessing the genetic diversity and relationships among germplasm from its primary distribution area contributes to evaluating the potential for its utilization as a gene pool to improve the desired agronomic traits. In the study, 40 EST-SSR primers were used to assess the genetic diversity and population structure of 36 E. sibiricus accessions with variation of seed shattering. A total of 380 bands were generated, with an average of 9.5 bands per primer. The polymorphic information content (PIC) ranged from 0.23 to 0.50. The percentage of polymorphic bands (P) for the species was 87.11%, suggesting a high degree of genetic diversity. Based on population structure analysis, four groups were formed, similar to results of principal coordinate analysis (PCoA). The molecular variance analysis (AMOVA) revealed the majority of genetic variation occurred within geographical regions (83.40%). Two genotypes from Y1005 and ZhN06 were used to generate seven F1 hybrids. The molecular and morphological diversity analysis of F1 population revealed rich genetic variation and high level of seed shattering variation in F1 population, resulting in significant improvement of the genetic base and desired agronomic traits.

Sir2, a NAD-dependent deacetylase, modulates lifespan in yeasts, worms and flies. The SIRT1, mammalian homologue of Sir2, regulates signaling for favoring survival in stress. But whether SIRT1 has the function to influence cell viability and senescence under non-stressed conditions in human diploid fibroblasts is far from unknown. Our data showed that enforced SIRT1 expression promoted cell proliferation and antagonized cellular senescence with the characteristic features of delayed Senescence-Associated beta-galactosidase (SA-beta-gal) staining, reduced Senescence-Associated Heterochromatic Foci (SAHF) formation and G1 phase arrest, increased cell growth rate and extended cellular lifespan in human fibroblasts, while dominant-negative SIRT1 allele (H363Y) did not significantly affect cell growth and senescence but displayed a bit decreased lifespan. Western blot results showed that SIRT1 reduced the expression of p16(INK4A) and promoted phosphorylation of Rb. Our data also exposed that overexpression of SIRT1 was accompanied by enhanced activation of ERK and S6K1 signaling. These effects were mimicked in both WI38 cells and 2BS cells by concentration-dependent resveratrol, a SIRT1 activator. It was noted that treatment of SIRT1-.transfected cells with Rapamycin, a mTOR inhibitor, reduced the phosphorylation of S6K1 and the expression of Id1, implying that SIRT1-induced phosphorylation of S6K1 may be partly for the decreased expression of p16(INK4A) and promoted phosphorylation of Rb in 2BS. It was also observed that the expression of SIRT1 and phosphorylation of ERK and S6K1 was declined in senescent 2BS. These findings suggested that SIRT1-promoted cell proliferation and antagonized cellular senescence in human diploid fibroblasts may be, in part, via the activation of ERK/ S6K1 signaling.

BACKGROUND: p16(INK4a) tumor suppressor protein has been widely proposed to mediate entrance of the cells into the senescent stage. Promoter of p16(INK4a) gene contains at least five putative GC boxes, named GC-I to V, respectively. Our previous data showed that a potential Sp1 binding site, within the promoter region from -466 to -451, acts as a positive transcription regulatory element. These results led us to examine how Sp1 and/or Sp3 act on these GC boxes during aging in cultured human diploid fibroblasts. METHODOLOGY/PRINCIPAL FINDINGS: Mutagenesis studies revealed that GC-I, II and IV, especially GC-II, are essential for p16(INK4a) gene expression in senescent cells. Electrophoretic mobility shift assays (EMSA) and ChIP assays demonstrated that both Sp1 and Sp3 bind to these elements and the binding activity is enhanced in senescent cells. Ectopic overexpression of Sp1, but not Sp3, induced the transcription of p16(INK4a). Both Sp1 RNAi and Mithramycin, a DNA intercalating agent that interferes with Sp1 and Sp3 binding activities, reduced p16(INK4a) gene expression. In addition, the enhanced binding of Sp1 to p16(INK4a) promoter during cellular senescence appeared to be the result of increased Sp1 binding affinity, not an alteration in Sp1 protein level. CONCLUSIONS/SIGNIFICANCE: All these results suggest that GC- II is the key site for Sp1 binding and increase of Sp1 binding activity rather than protein levels contributes to the induction of p16(INK4a) expression during cell aging.
