In this process, how Modernity penetrates people with discursive formations is handled through the concretization of “time”, which is an important concept for Modernity, through the institute, which is also one of the important institutions of Modernity. The Time Regulation Institute, which was founded out of a need, gains legitimacy through discursive formations. Thus, it begins to permeate society. The institution, functioning as a part of discursive formations, is finally dissolved with the change of discourse. The ironic language of the novel is extremely important at this point. With the absurd picture it paints, the novel ironically reveals the situations that people cannot comprehend while living, thus enabling people to look at the reality they live in from an outside perspective., Ahmet Hamdi Tanpınar tarafından yazılan Saatleri Ayarlama Enstitüsü, Türkiye’nin modernleşme sürecini Osmanlı’nın son döneminden başlatıp romanın yazıldığı 1954’lere kadar getirerek ele alır. Bu süreçte Modernite’nin söylemsel oluşumlarla insanlara nasıl nüfuz ettiği, Modernite için önemli bir kavram olan “zaman”ın, yine Modernite’nin önemli kurumlarından olan enstitü aracılığıyla somutlaştırılması yolu ile işlenir. Bir ihtiyaçtan kaynaklanmadan kurulan Saatleri Ayarlama Enstitüsü, kuruluşunu önce söylemsel bir zemine oturtarak meşruiyet kazanır. Böylece topluma nüfuz etmeye başlar. Çalışma süresi boyunca söylemsel oluşumların bir parçası olarak işleyen kurum, nihayet mevcut söylemin değişmesiyle lağvedilir. Romanın ironik dili bu noktada son derece önemlidir. Roman, insanın yaşarken idrak edemediği hâl ve durumları ironik bir dille ortaya koyarak resmettiği absürt tablo ile insana içinde yaşadığı gerçekliğe dışarıdan bakabilmesini sağlar.

Türk edebiyatındapopüler romanlar, genellikle edebî değerden yoksun olmaları nedeniyle horgörülmüş, çok okunan romanlar olmalarına rağmen akademik çalışmalarda ihmaledilmiştir. Bugün bu önyargı yavaş yavaş kırılmaya, popüler romanlar da çeşitliaçılardan incelenmeye başlanmıştır. Okuma yazma oranının düşük olduğu Tanzimatve Meşrutiyet yıllarında popüler romanlar, okuma-yazma oranının artmasınısağlamış, tarihî popüler romanlar ayrıca tarihteki değerleri hatırlatarak vetarihten hareketle aktüel mesajlar vererek millî bilincin uyanmasında önemlirol üstlenmişlerdir. Toplumda görülen millî temayüller, Cumhuriyet rejimininkurulmasıyla birlikte yeni devletin temel ideolojisi hâline gelmiştir. Popülertarihî romanlar da bu dönemden sonra milli değerlerin yerleşmesine hizmet eder.Popüler tarihî roman yazarlarından Murat Sertoğlu da romanlarında bu amacıtaşır. Bu çalışmada 56 yıl gibi uzun bir süre yazdıklarıyla Türk halkınıpeşinden sürükleyen Murat Sertoğlu’nun popüler edebiyat içindeki yeri üzerindedurulacaktır., Popular novels inTurkish literature were often despised because of their lack of literary value,and although they were widely read novels, they were ignored in academia. Todaythis prejudice is gradually being broken, and popular novels have also beenstudied from various perspectives. In Tanzimat and Meşrutiyet periods whenliteracy rate is low, popular novels have increased the literacy rate. Inaddition, evoking the historical values and hereby giving actual messages,historical popular novels played an important role in the awakening of nationalconsciousness. National tendency in society has become the basic ideology ofthe Republic of Turkey. Popular historical novels serve to establish nationalvalues ​​hereupon. Murat Sertoğlu, one of the popular historical novelists, hasthis aim in his novels. In this study, the place of Murat Sertoğlu in popularliterature will be examined.

Ayna yüzyıllar boyunca özel ve kamusal olarak görüntüyü yansıtmak amacıyla kullanılan ve kendisine bu amacının çok ötesinde dinî, metafizik ve toplumsal anlamlar yüklenen bir nesne olmuştur. Genellikle kadınlarla özdeşleştirilen ayna, kişisel bir eşya ve mekanların önemli birer nesnesi olmasının yanı sıra dinî ritüellerde, falcılıkta da kullanılmış, zaman zaman tılsımlı olduğuna inanılmıştır. İnsanlara her zaman ilginç gelen ve hayatlarının önemli bir parçası olarak görülen ayna, çok farklı yönleriyle edebî metinlerde de farklı anlamlarda çeşitli amaçlarla kullanılmıştır. Efsanelerden romanlara uzanan bu yelpazede aynaya atfedilen anlam ve önem de değişmiştir. Modern edebiyatta, eski kullanımlar bir yandan korunurken, öte yandan yeni bağlamlar üretilmiş, aynanın anlam ve kullanım yelpazesi, özellikle bireyle ve toplumla ilişkisi bakımından sürekli genişlemiştir. Şemsettin Sami’den Halid Ziya Uşaklıgil’e, Ahmet Hamdi Tanpınar’dan Leyla Erbil’e, Murathan Mungan’a, Orhan Pamuk’a uzanan çizgide ayna, zaman zaman tekil metinlerin, zaman zaman yazarın külliyatının özünü temsil edebilecek kadar merkezî bir yerde durabilmektedir. Türk Edebiyatında aynanın izini sürmek, edebiyatın aslî hedeflerinden biri olan insanı ve toplumu anlamaya yarayacak önemli veriler sunmaktadır. Yeni Türk Edebiyatında aynayı nesne olarak alıp, bunun üzerinden geniş çerçeveli bir literatür taraması içeren çalışma henüz yoktur. Bu tez bu eksikliği kapatmayı hedeflemektedir. Malzemenin bolluğu nedeniyle tür ve zaman açısından kısıtlamaya gitmek zaruridir; bu nedenle en çeşitli ve bol malzemeyi vermesi itibarıyla roman türüne odaklanılmış, modern Türk Edebiyatının başlangıcı kabul edilen 1872’den 1973’e dek yazılan romanlar ele alınmıştır. Bununla birlikte diğer türlere giren ve/veya sınırlanan zamanı aşan diğer eserlere gerek görüldükçe, özellikle dipnotlarda referans verilecektir. The mirror has been an object used to reflect the image both privately and publicly for centuries and attributed to religious, metaphysical, and social meanings. The mirror generally identified with women is a personal belonging and an important object of places, and it is believed to be magic from time to time. Mirror, which is always fascinating to people and seen as an important part of their lives, thereby has been used for various purposes in different meanings in literary texts. Ranging from legends to novels, the meaning and importance attributed to mirrors have also changed. In modern literature, while the old uses are preserved, new contexts also have been created, especially in terms of its relation to the individual and society. From Şemsettin Sami to Halid Ziya Uşaklıgil, from Ahmet Hamdi Tanpınar to Leyla Erbil, Murathan Mungan, and Orhan Pamuk, the mirror can at times stand in the center, representing the essence of individual texts and sometimes of the author's corpus. Tracing the mirror in Turkish literature provides important data to understand people and society, which is one of the main goals of literature. In Modern Turkish Literature, there is no study taking the mirror as an object and including a wide-framed literature review over novels. This dissertation aims to fill this gap. Due to the abundance of the material, the focus has been on the genre of novels, as it gives the most diverse and abundant material, and the novels written from 1872 to 1973, are handled with.

Ayna, insanin kendini butunuyle gorebilmesini saglayan, boylece insanin kendisini kesfetmesine aracilik eden bir nesnedir. Ayna, oznenin kendini kurmasiyla yakindan iliskili oldugu icin bireyin hikâyesini anlatan romanlarda gercek birer nesne ya da metafor olarak cokca kullanilmistir. Halid Ziya Usakligil’in romanlarinda da aynalar, ozellikle romanin ana karakteri konumunda olan kadinlarin kisiliklerini aciga cikarma noktasinda son derece onemli rol oynar. Kadin karakterler aynada daha once farkinda olmadiklari ya da yadsidiklari benliklerini kesfeder ve kabul eder; boylece kendilerini bu goruntu araciligiyla yeniden tanimlamaya baslar. Dolayisiyla aynada olusan goruntunun tasviri aynaya bakan kisinin kendini nasil gorduguyle veya gormek istedigiyle ilgili ipuclari verir. Bu calismada, Halid Ziya’nin ilk romanindan baslayarak Halid Ziya kulliyatinda ayna sahnesinin zirvesi kabul edilebilecek olan Ask-i Memnu’ ya kadar olan romanlar kadin-ayna iliskisi acisindan incelenmistir.

Objective: The aim of this study was to investigate the relationships between F216L (rs28942112), R496W (rs374603772), S127R (rs28942111), and D374Y (rs137852912) PCSK9 gain-of-function (GOF) mutations and primary dyslipidemia and serum lipid levels in patients with primary dyslipidemia. Methods: In this case-control study, DNA was isolated from blood samples collected from patients diagnosed with primary dyslipidemia in cardiology outpatient clinic of Ege University (n=200) and healthy individuals (n=201). F216L, R496W, S127R, and D374Y GOF mutations in the PCSK9 gene were evaluated and genotyped according to the results of melting curve analysis performed in a real-time polymerase chain reaction (PCR) 480 instrument using specific primers for each mutation. Results: There were statistically significant differences between the patient and individuals in control groups in the R496W and D374Y mutations (χ2=10.742 p=0.005; χ2=6.078 p=0.048, respectively). In addition, triglyceride levels in patients with primary dyslipidemia heterozygous for R496W and D374Y mutations were 12.8-fold (p=0.015) and 3.4-fold (p=0.03) higher than that in mutant and wild-type genotype, respectively. Additionally, in the entire study group (n=401), PCSK9 R496W and D374Y mutation carriers had increased total cholesterol (p=0.021), triglycerides (p=0.0001), HDL cholesterol (p=0.028), and low-density lipoproteins (LDL) cholesterol (p=0.028) levels. However, F216L (rs28942112) and S127R (rs28942111) mutations were not detected in patients with primary dyslipidemia and healthy controls. Conclusion: We conclude that the PCSK9 R496W (rs374603772) and D374Y (rs137852912) GOF mutations may be significant risk factors in the development of primary dyslipidemia and may have significant impact on lipid parameters in general population.

Amaç: t(4;11), MLL-AF4 translokasyonu sonucu oluşan, 4q21 kromozomal bandına yerleşim gösteren AF4 geninin 11q23 kromozomal bandına yerleşim gösteren MLL genine füzyonu sonucu gelişen kromozomal bir anomalidir. Bu çalışmada, retrospektif olarak 2009-2013 yılları arasındaki akut lenfoblastik lösemi (ALL) hastalarındaki t(4;11) MLL- AF4 translokasyonunun analiz sonuçlarının incelenmesi amaçlandı. Gereç ve Yöntem: Ege Üniversitesi Tıp Fakültesi Tıbbi Biyoloji Anabilim Dalı’na 2009-2013 yılları arasında akut lösemi ön tanısıyla 176 çocuk (70 kız, 106 erkek) ve 144 yetişkin (60 kadın, 84 erkek) olgunun kan veya kemik iliği örnekleri incelendi. Bu olgulara ait 71 kan ve 473 kemik iliği örneğinin t(4;11) translokasyon RNA sonuçları, gerçek zamanlı RT-PCR yöntemi ile kantitatif olarak değerlendirildi. İlk aşamada, kan ve kemik iliği örneklerinden izole edilen total RNA veya mRNA’dan konvansiyonel bir PCR cihazı ile komplementer DNA sentezlendi. İkinci aşamada, gerçek zamanlı PCR cihazı ile t(4;11) kantitasyonu gerçekleştirildi. Olguların kantitatif olarak değerlendirilmesi, pozitif kontrol ve negatif kontrolün karşılaştırılması ile örneklerin negatif yada pozitif (pozitif olgu kopya sayısının referans kopya sayısına oranı) olması şeklinde yapıldı. Bulgular: Çalışmamızda 98’i takip hastası olmak üzere toplam 320 hasta t(4;11) MLL-AF4 translokasyonu için değerlendirildi. Çalışmaların sonucunda toplam 34 olgu (24 çocuk, 10 yetişkin) pozitif ve diğer örnekler negatif olarak bulundu. Sonuç: Bu değerlendirmenin sonuçları, RT-PCR yöntemi ile ALL hastalarında yeni tanı döneminde ve tedavi sürecinde t(4;11) MLL-AF4 translokasyonunun kantitatif tayini, hem tanının kesinleştirilmesinde hem de moleküler remisyon sağlanmasına yönelik tedaviyi yönlendirmesinde değerli bir yöntem olduğunu desteklemektedir., Aim: t(4,11) is a chromosomal abnormality formed by the translocation MLL-AF4, which is the result of the fusion of the AF4 gene, localized on 4q21 chromosomal band, to the MLL gene, localized on 11q23 chromosomal band. The aim of this study is to examine the results of the analysis of t (4;11) MLL-AF4 translocation in acute lymphoblastic leukemia (ALL) patients retrospectively. Materials and Methods: Peripheral blood or bone marrow samples of 176 children (70 girls, 106 boys) and 144 adults (60 women, 84 men) with a preliminary diagnosis of acute leukemia between 2009-2013 were analyzed in the Medical Biology Department of Ege University Faculty of Medicine. The translocation RNA results of 71 peripheral blood and 473 bone marrow samples of these patients were evaluated quantitatively for t(4;11) with real-time RT- PCR. t(4;11) quantitation was performed by real-time qRT-PCR instrument after the synthesis of complementary DNA with conventional PCR from total RNA or mRNA isolated from blood and bone marrow. Quantitative analysis of the patients was performed by comparing positive and negative controls and samples classified as positive or negative (the ratio of the number of positive copies to the number of reference copies). Results: A total of 320 patients, with 98 having also follow-ups, were evaluated for t(4;11) translocation. Totally 34 patients (24 children and 10 adults) were found positive and the other samples were negative. Conclusion: The assessment of these results supports that, quantitative determination of t(4;11) with RT-PCR method among newly diagnosed ALL patients and ALL patients undergoing treatment, is a valuable method for both confirming the diagnosis and guiding the treatment intended to achieve molecular remission.

Amac: Bu calismada, Ege Universitesi Tip Fakultesi Tibbi Biyoloji Anabilim Dali'na 2009-2013 yillari arasinda akut myeloid losemi (AML) on tanisi ile basvuran 402 olgunun (322 yetiskin, 80 cocuk) kan veya kemik iligi orneklerinin inv16 kantitasyon analizlerinin RT-PCR yontemi ile degerlendirilmesi amaclandi. Gerec ve Yontem: Hastalardan alinan kan ve kemik iligi orneklerinden total RNA/mRNA izolasyonunu takiben cDNA'lari elde edilerek revers-transkriptaz-polimeraz zincir reaksiyonu (RT-PCR) yontemiyle inv16 kantitasyonu LightCycler2 cihazinda gerceklestirildi. Bulgular: Dort yuz iki olgu, inv(16) CBFBETA-MYH11 inversiyonu acisindan degerlendirildi. Yapilan analiz sonucunda 12 (% 4) eriskin ve 7 (% 9) cocuk olmak uzere toplam 19 olguda inv16 pozitifligi saptandi. Sonuc: Inv16 kantitatif analizi, AML hastalarinin klinik degerlendirilmesinde, hastaligin seyri ve remisyon saglanmasina yonelik tedavi protokollerinin uygulanmasinda etkili bir yontem oldugunu desteklemektedir. AML hasta grubunu iceren daha genis kapsamli calismalar yapilarak elde edilen sonuclar desteklenmelidir.

AML hasta grubunu içeren daha geniş kapsamlı çalışmalar yapılarak elde edilen sonuçlar desteklenmelidir., Aim: In this study, it was aimed to evaluate the analysis of inv16 blood and bone marrow samples quantitive ly by reve rse transcriptase polymerase chain reaction (RT-PCR ) method of 402 patients (322 adult, 80 children) prediagnosed as acute myeloid leukemia (AML), who were admitted to Ege University Faculty of Medicine, Department of Medical Biology between years 2009 -201 3. Materials and Methods: cDNA’s were obtained following the total RNA/mRNA isolation which were taken from patients’ blood and bone marrow samples and quantitaion of inv16 were performed by reverse transcriptase RT-PCR method at LightCycler2. Results: Four hundred and two cases were evaluated in terms of inv(16) CBFBETA -MYH11 inversion. As a result of the analysis, a total of 19 patient were determined inv16 positivity includes 12 adult (% 4) and 7 children (9%).Conclusion: Inv16 of quantitative analysis is supported to be an e ffective method for clinical evaluation of patients with AML, regarding remission of disease and implimentation of the treatment protocols toward achieving remission. More studies with AML patients are required for confirmation of the results.

To investigate the association between the 3 most frequently observed single nucleotide polymorphisms of the NAT2 gene and the risk of developing prostate cancer in the Turkish population. Materials and methods: A total of 110 unrelated patients with prostate cancer were included in this case-control association study and constituted the study group. The control group also consisted of 150 unrelated but healthy men. Genomic DNA was isolated from peripheral blood leukocytes of all patients and analyzed with a sensitive real-time PCR method. After melting curve analysis genotypes were identified for the NAT2*5A, NAT2*6A, and NAT2*7A/B polymorphisms. Results: Prostate cancer patients had a higher frequency of the mutant NAT2*6A (13.6% versus 4.0%, P = 0.009) and heterozygote NAT2*7A/B (20.9% versus 9.3%, P = 0.008) genotypes when compared with the controls. Conclusion: The NAT2*6A and NAT2*7A/B gene polymorphisms were significantly associated with prostate cancer in the Turkish population. Real-time PCR analysis of the NAT2 acetylator phenotype can therefore be used to recognize individuals with a high risk of developing prostate cancer.

Effective molecular mechanisms of resistance to cisplatinCisplatin is a platinating chemotherapeutic agent widely used in the treatment of many solid tumors. It wasdiscovered for the first time as a growth inhibitor of Escherichia coli in 1970, later years its anticancer effectsrediscovered and began to use in the treatment of testicular, ovarian, cervical, head and neck, non-small celllung cancers and lymphoma. Although a good response to cisplatin can be achieved in the treatment oftesticular cancer, therapeutic efficacy is limited due to toxicity or drug resistance in other solid tumors. Inthis review the molecular mechanisms trough the development of cisplatin resistance and the new treatmentstrategies were evaluated. Understanding the molecular mechanisms of cisplatin resistancewill providesimportant insights for designingmore efficient platinum-based chemotherapeutics, Cisplatin birçok solid tümörün tedavisinde sıklıkla kullanılan platin bileşiği bir kemoterapotik ajandır. İlk defa 1970 yılında Escherichia coli' nin büyüme inhibitörü olarak keşfedilmiş, daha sonraki yıllarda anti kanser etkisi fark edilerek testis, over, serviks, baş ve boyun, küçük hücreli dışı akciğer ve lenfoma tedavisinde kullanılmaya başlanmıştır. Testis kanserlerinde cisplatine genellikle iyi yanıt alınabilmesine karşılık diğer solid tümörlerde toksisite veya ilaç direnci nedeniyle terapötik etkinlik sınırlı kalmaktadır. Bu derlemede cisplatin yanıtının oluşmasını sağlayan biyokimyasal yolaklar, direnç gelişiminde etkili moleküler mekanizmalar ve yeni tedavi stratejileri değerlendirilmiştir. Cisplatin direnci gelişimindeki moleküler mekanizmaların anlaşılması daha efektif platin bazlı kemoterapotiklerin geliştirilmesi için önemli bilgiler sağlayacaktır.

Amaç: Bu çalışmanın amacı azospermik ve oligozoospermik infertil Türk erkeklerinde ve sağlıklı kontrol olgularında Y kromozom mikrodelesyonlarının sıklıklarının belirlenmesidir. Gereç ve yöntemler: Bu çalışmaya 64 azospermik ve 51 oligozoospermik infertilite olgusu ve herhangi bir yardımcı üreme tekniği olmadan doğal yollardan çocuk sahibi olduğu bilinen 70 sağlıklı erkek dahil edildi. Olguların periferik kan örnekleri toplanarak DNA izolasyonu yapıldı. On beş farklı primer dizisi kullanıla- rak gerçekleştirilen multipleks PCR işleminden sonra agaroz jel elektroforezi ile Y kromozomunun AZFa, AZFb, AZFc ve AZFd bölge mikrodelesyonları belirlendi. Bulgular: Azospermi grubundaki 8 (%12,5) olguda ve oligozoospermi grubundaki 3 (%5,9) olguda Y kro- mozom mikrodelesyonu belirlendi. İnfertil olguların tümü değerlendirildiğinde ise mikrodelesyon oranı %9,6 olarak bulundu. İnfertilite olgularının 4’ünde (%3,48) AZFa, 2’sinde (%1,74) AZFb, 3’ünde (%2,61) AZFc ve 7’sinde (%6,09) AZFd bölge delesyonu saptandı. Kontrol grubundaki sağlıklı olgularda ise Y kro- mozom mikrodelesyonu belirlenmedi. Sonuç: Azospermik ve oligozoospermik infertil erkeklerde Y kromozom mikrodelesyonlarının bulunması nedeniyle yardımcı üreme teknikleri öncesinde rutin genetik testler yapılması ve genetik danışmanlık ve- rilmesi gerektiğini önermekteyiz., Objective: The aim of this study was to determine the frequencies of Y chromosome microdeletions in infertile azoospermic and oligozoospermic Turkish men and in healthy control subjects. Material and methods: Sixty-four azoospermic and 51 oligozoospermic patients with infertility and 70 healthy men who had a child naturally without assisted reproductive technology were included in this study. DNA was extracted from peripheral blood samples collected from the patients. Following multiplex PCR performed with 15 different primer sequences, Y chromosome AZFa, AZFb, AZFc and AZFd region mi- crodeletions were determined by agarose gel electrophoresis. Results: Y chromosome microdeletions were detected in 8 (12.5%) patients in the azoospermia group and 3 (5.9%) patients in the oligozoospermia group. The overall prevalence of Y chromosome microdeletions in infertile men was 9.6%. Y chromosome microdeletions were not found in the healthy control group. Among the infertile cases, there were 4 (3.48%) AZFa, 2 (1.74%) AZFb, 3 (2.61%) AZFc and 7 (6.09%) AZFd region microdeletions. Conclusion: The presence of Y chromosome microdeletions among azoospermic and oligozoospermic in- fertile males suggests that routine genetic testing and genetic counseling prior to the use of assisted repro- duction techniques are necessary.

Tuberculosis is treated with a group of drugs that need to be used over a long period of time and isoniazid is the major drug in this group. Antituberculosis drug-induced hepatitis is the most serious problem in tuberculosis treatment. The enzyme N-acetyltransferase-2 (NAT-2) metabolizes isoniazid in the liver so it is considered to cause hepatotoxicity. The association of polymorphic NAT acetylator status and antituberculosis drug-induced hepatitis is discussed. To determine whether acetylator status is a risk factor for antituberculosis drug-induced hepatitis, we genotyped NAT2*5A, NAT2*6A, NAT2*7A/B and NAT2*14A polymorphisms in 100 patients diagnosed with tuberculosis. 70 patients who did not develop hepatotoxicity were classified as the control group, and 30 patients who were diagnosed with antituberculosis drug-induced hepatitis were classified as the study group. NAT2 polymorphisms were divided into three phenotypic groups according to the analytical results obtained. Among the 70 patients constituting the control group; 14 (20%), 37 (52.9%), 19 (27.10%) patients were rapid, intermediate and slow acetylators respectively. In contrast, among the patients constituting the study group; 3 (10%), 4 (13.3%), 23 (76.7%) patients were rapid, intermediate and slow acetylators. The difference was statistically significant when the control and study groups were compared for their acetylator status. The proportion of slow acetylators was much higher in the study group. In conclusion, NAT2 acetylator phenotype analysis by molecular biology methods prior to medical treatment for tuberculosis, can be used both for determining the high-risk group of patients who may develop hepatotoxicity and for closer follow-up during treatment period.

The arginine- to-glutamine change at codon 3500 of the apolipoprotein B-100 (apo B) is a well- known genetic cause of hypercholesterolemia. Since increased cholesterol levels lead to atherosclerosis, identification of the carries of the apo B-3500 mutation is an important step in the risk stratification of individuals and families with hypercholesterolemia. The prevalence of this mutation in Turkish population is not well known. We aimed to investigate the frequency of apo B-100 mutations (codon 3500) C9774T (Arg 3500--Trp) and G9775A (Arg 3500--Gln) in patients with atherosclerosis in comparison with healthy subjects.This cross-sectional study included 442 unrelated subjects living on the West coast of Turkey. Subgroups consisted of 165 patients with coronary artery disease, 163 patients with ischemic stroke, and 114 healthy control subjects.We did not find any apo B-100 mutation both in the patient and control groups.As it is hypothesized that this mutation arose within the Central European region from a common ancestor approximately 7000 years ago and spread across Europe, our result of the absence of the R3500Q mutation in Turkish patients give an important information about the geographical distribution of the apo B-R3500Q, that the mutation has not reached to Anatolia.

Torsades de pointes (TdP) during bradyarrhythmias have been reported to be associated with gender, degree of QT prolongation and duration of bradyarrhythmia. We sought to investigate the repolarization characteristics on 12-lead electrocardiogram (ECG) and the incidence of TdP in patients with acquired complete atrioventricular block (CAVB).Fifty consecutive patients with acquired CAVB were included in the study. Patients with coronary artery disease, systolic dysfunction and previous cardiac surgery were excluded. Patients were monitored during hospitalization for ventricular arrhythmias (VA). Serum potassium, magnesium, calcium levels and thyroid-stimulating hormone were measured. Heart rate, QRS duration, QT/QTc, JT/JTc and Tpeak-Tend intervals were measured. Pathologic U waves, T-U complex, and QT morphologies were remarked.Patients presented with presyncope (n=39, 78%), syncope (n=12, 24%), and palpitations (n=8, 16%). All patients were in sinus rhythm. Duration of CAVB was 8.5 days (median). Patients were divided into two groups based on JT interval. Group 1 (JT=or500 ms, n=13) tended to have more female patients and more VAs in comparison to Group 2 (JT500 ms, n=37). Group 1 patients had more pathologic U waves and T-U complexes, longer Tpeak-Tend intervals, and more long QT2 syndrome (LQT2)-like QT morphology in comparison to Group 2 patients. Group 2 patients had more often syncope. One patient in Group 2 developed ventricular fibrillation in the presence of hypokalemia and hypomagnesemia.Torsades de Pointes during CAVB was rare among our patient population. The predictors of VA during CAVB were presence of prolonged QTc/JTc intervals, pathologic U wave and T-U complex, prolonged Tpeak-Tend interval, and LQT2-like QT morphology.

Karaciğer transplantasyonu, son evredeki karaciğer hastalıklarında ve akut karaciğer yetmezliğinde rutin bir tedavi haline gelmiştir. Koagülasyon faktörlerinden biri olan Faktör V karaciğer tarafından sentezlenmektedir. Koagülasyon faktör genlerindeki defektler, anormal veya derin ven tromboz gelişimi için risk faktörü oluşturabilecek yetersiz proteinlerin oluşumuna sebep olmaktadır. Aktif protein C ye karşı kazanılmış olan direnç (APCr) en sık karşılaşılan koagülasyon anomalisidir ve % 90 - 95 oranında faktör V Leiden mutasyonu sonucu gelişmektedir. Bu mutasyon, G1691A genotiplemesi şeklinde karakterize edilmektedir. Trombofilik anomalilerde canlı donörlerin genotiplemesi mutlaka gerekmektedir. Karaciğer transplantasyonu sonrası olguların yaklaşık % 2.7' sinde derin ven trombozu gözlenmektedir. Bu amaçla, organ alıcı ve donörlerinde FV Leiden gen mutasyonu analizi yapılmalıdır. Çalışmamızda alıcı (191 kişi) ve donör (693 kişi) olarak Anabilim Dalı' miza yollanan toplam 884 olgunun kan örneklerinden genomik DNA izole edilmiş ve Faktör V Leiden mutasyonları Real-time Online PCR ile saptanmıştır. Toplam 884 olgunun dahil edildiği çalışmamızda FV Leiden gen mutasyonu için 80 olgu heterozigot, üç olgu homozigot, 801 olgu normal genotipte olduğu belirlenmiştir. Karaciğer transplantasyonu sonrasında, derin-ven trombozisi geliştiren hastaların, donör karaciğerinde homozigot Faktör V Leiden gen mutasyonu sonucu ortaya çıkan sonradan kazanılmış aktive olmuş Protein C (APC) rezistansı temeline dayandığı belirtilmiştir. FV Leiden gen mutasyonunun rutin taraması pre ve post operatif koagülasyonların kullanımı transplantasyon olgularında tromboembolik komplikasyonların gelişimini önlemede uygun bir stratejidir. Sonuç olarak bu genotipleme, hem alıcıyı hem de donörün trombozdan korunmasını sağlamaktadır., Liver transplantation has become a routine procedure for the treatment of end stage of chronic liver disease and acute liver failure. Factor V which is one of the coagulation factors is synthesized by the liver. Defects in coagulation factor genes may lead to the production of abnormal or deficient proteins that constellate risk factors for the development of deep-vein thrombosis. Acquired Active protein C resistance is the most encountered coagulation abnormality and in 90-95 % of cases and it is caused by a point mutation in the factor V gene. The factor V Leiden mutation is genotyped for a single base-pair change (G1691A). Patients who develop venous thrombosis after liver transplantation should be screened for thrombophilic abnormalities. Deep-vein thrombosis occurs approximately 2.7% of patients after liver transplantation. For this reason, FV Leiden mutation has determined in organ donors and recipients. In our study; recipient (191 individuals),-donors (693 individuals) totally 884 cases appealed to our department. Genomic DNA of the cases isolated and FV Leiden mutation was determined by Real Time Online PCR. Totally 884 individual, included to our study. The prevalence of the heterozygous mutation was 80 and homozygous mutation was 3. Number of normal genotype was determined 801 individuals. Routine screening for factor V Leiden mutation by polymerase chain reaction, and appropriate pre-operative and post-operative anticoagulation after transplantation might be valuable strategy to prevent tromboembolic complications in transplant recipients.

Bu çalışma 15-16 Mayıs 2017 tarihlerinde düzenlenen “Literature as a Marplot: Seeing the Discursive Formations of Modernity in Turkey through The Time Regulation Institute” Kongresi‘nde bildiri olarak sunulmuştur. Ahmet Hamdi Tanpınar tarafından yazılan Saatleri Ayarlama Enstitüsü, Türkiye’nin modernleşme sürecini Osmanlı’nın son döneminden başlatıp romanın yazıldığı 1954’lere kadar getirerek ele alır. Bu süreçte Modernite’nin söylemsel oluşumlarla insanlara nasıl nüfuz ettiği, Modernite için önemli bir kavram olan “zaman”ın, yine Modernite’nin önemli kurumlarından olan enstitü aracılığıyla somutlaştırılması yolu ile işlenir. Bir ihtiyaçtan kaynaklanmadan kurulan Saatleri Ayarlama Enstitüsü, kuruluşunu önce söylemsel bir zemine oturtarak meşruiyet kazanır. Böylece topluma nüfuz etmeye başlar. Çalışma süresi boyunca söylemsel oluşumların bir parçası olarak işleyen kurum, nihayet mevcut söylemin değişmesiyle lağvedilir. Romanın ironik dili bu noktada son derece önemlidir. Roman, insanın yaşarken idrak edemediği hâl ve durumları ironik bir dille ortaya koyarak resmettiği absürt tablo ile insana içinde yaşadığı gerçekliğe dışarıdan bakabilmesini sağlar. Written by Ahmet Hamdi Tanpınar, Saatleri Ayarlama Enstitüsü (The Time Regulation Institute) deals with Turkey’s modernization process, starting from the last period of the Ottoman Empire and bringing it to 1954, when the novel was written. In this process, how Modernity penetrates people with discursive formations is handled through the concretization of “time”, which is an important concept for Modernity, through the institute, which is also one of the important institutions of Modernity. The Time Regulation Institute, which was founded out of a need, gains legitimacy through discursive formations. Thus, it begins to permeate society. The institution, functioning as a part of discursive formations, is finally dissolved with the change of discourse. The ironic language of the novel is extremely important at this point. With the absurd picture it paints, the novel ironically reveals the situations that people cannot comprehend while living, thus enabling people to look at the reality they live in from an outside perspective.

Türk edebiyatında popüler romanlar, genellikle edebî değerden yoksun olmaları nedeniyle hor görülmüş, çok okunan romanlar olmalarına rağmen akademik çalışmalarda ihmal edilmiştir. Bugün bu önyargı yavaş yavaş kırılmaya, popüler romanlar da çeşitli açılardan incelenmeye başlanmıştır. Okuma yazma oranının düşük olduğu Tanzimat ve Meşrutiyet yıllarında popüler romanlar, okuma-yazma oranının artmasını sağlamış, tarihî popüler romanlar ayrıca tarihteki değerleri hatırlatarak ve tarihten hareketle aktüel mesajlar vererek millî bilincin uyanmasında önemli rol üstlenmişlerdir. Toplumda görülen millî temayüller, Cumhuriyet rejiminin kurulmasıyla birlikte yeni devletin temel ideolojisi hâline gelmiştir. Popüler tarihî romanlar da bu dönemden sonra milli değerlerin yerleşmesine hizmet eder. Popüler tarihî roman yazarlarından Murat Sertoğlu da romanlarında bu amacı taşır. Bu çalışmada 56 yıl gibi uzun bir süre yazdıklarıyla Türk halkını peşinden sürükleyen Murat Sertoğlu’nun popüler edebiyat içindeki yeri üzerinde durulacaktır. Popular novels in Turkish literature were often despised because of their lack of literary value, and although they were widely read novels, they were ignored in academia. Today this prejudice is gradually being broken, and popular novels have also been studied from various perspectives. In Tanzimat and Meşrutiyet periods when literacy rate is low, popular novels have increased the literacy rate. In addition, evoking the historical values and hereby giving actual messages, historical popular novels played an important role in the awakening of national consciousness. National tendency in society has become the basic ideology of the Republic of Turkey. Popular historical novels serve to establish national values hereupon. Murat Sertoğlu, one of the popular historical novelists, has this aim in his novels. In this study, the place of Murat Sertoğlu in popular literature will be examined.

Ayna, insanın kendini bütünüyle görebilmesini sağlayan, böylece insanın kendisini keşfetmesine aracılık eden bir nesnedir. Ayna, öznenin kendini kurmasıyla yakından ilişkili olduğu için bireyin hikâyesini anlatan romanlarda gerçek birer nesne ya da metafor olarak çokça kullanılmıştır. Halid Ziya Uşaklıgil’in romanlarında da aynalar, özellikle romanın ana karakteri konumunda olan kadınların kişiliklerini açığa çıkarma noktasında son derece önemli rol oynar. Kadın karakterler aynada daha önce farkında olmadıkları ya da yadsıdıkları benliklerini keşfeder ve kabul eder; böylece kendilerini bu görüntü aracılığıyla yeniden tanımlamaya başlar. Dolayısıyla aynada oluşan görüntünün tasviri aynaya bakan kişinin kendini nasıl gördüğüyle veya görmek istediğiyle ilgili ipuçları verir. Bu çalışmada, Halid Ziya’nın ilk romanından başlayarak Halid Ziya külliyatında ayna sahnesinin zirvesi kabul edilebilecek olan Aşk-ı Memnu’ya kadar olan romanlar kadın-ayna ilişkisi açısından incelenmiştir. Mirrors are such objects that enable people to see themselves fully, thus mediating their self-discovery. Since the mirror is closely related to the selfestablishment, it is often used as a real object or as a metaphor in novels that tell the story of an individual. The mirrors in Halid Ziya novels play a very crucial role to reveal the personalities of female protagonists. The female characters discover their identities that they have never realized or that they denied before. After then, they redefine themselves through their images in the mirrors. Therefore, the description of the image in the mirror gives some clues as to how the character looking in the mirrors sees herself or how she imagines herself. In this study, the novels starting from Halid Ziya’s first novel Sefile to Aşk-ı Memnu that is the summit of mirror stage in his corpus are examined in terms of women-mirror relation.
