A highly sensitive lateral flow immunoassay (LFIA) for imidacloprid, a widely used neonicotinoid insecticide, has been developed. The LFIA realizes the indirect coupling of anti-imidacloprid antibodies and gold nanoparticle (GNP) labels directly in the course of the assay. For this purpose, the common GNPs conjugate with anti-imidacloprid antibodies and are changed into a combination of non-modified, anti-imidacloprid antibodies, and the GNPs conjugate with anti-species antibodies. The given approach provides the possibility of selecting independent concentrations of GNPs and anti-imidacloprid antibodies to obtain the influence of minimal imidacloprid concentrations in the samples on the formation of detected, labeled immune complexes. A comparative study of imidacloprid LFIAs with common and indirect antibody–label coupling was implemented. The second variant reduced the limit of detection (LOD) of imidacloprid 20 times, reaching 0.2 ng/mL and 0.002 ng/mL for visual and instrumental detection, respectively, thus surpassing the existing LFIAs for imidacloprid. The developed highly sensitive LFIA was tested for imidacloprid detection in freshly squeezed fruits and berries without any additional sample preparation. The imidacloprids revealed were in the range of 75–97% for grape, 75–85% for orange, and 86–97% for apple samples. The time of the testing was 15 min. [ABSTRACT FROM AUTHOR]

An immunochromatographic test system based on the indirect competitive format has been developed for the rapid control of cyanotoxin microcystin-LR (MC-LR) in drinking water. To increase the assay sensitivity, a hydroquinone-based reduction of silver ions on the surface of gold nanoparticles as part of the immune complexes formed on the immunochromatographic membrane has been carried out. This stage allows for the enhancement of the colouration in test strip zones. The developed test system is characterised by an instrumental limit of detection of 0.05 ng/mL, a cut-off of 0.5 ng/mL, and a working range of the detectable concentrations of 0.08–0.42 ng/mL. The assay duration was 17 min. The applicability of the test system for MC-LR detection in drinking water is demonstrated. The assay recoveries vary from 90% to 120%. [ABSTRACT FROM AUTHOR]

Gatifloxacin (GAT), an antibiotic belonging to the fluoroquinolone (FQ) class, is a toxicant that may contaminate food products. In this study, a method of ultrasensitive immunochromatographic detection of GAT was developed for the first time. An indirect format of the lateral flow immunoassay (LFIA) was performed. GAT-specific monoclonal antibodies and labeled anti-species antibodies were used in the LFIA. Bimetallic core@shell Au@Ag nanoparticles (Au@Ag NPs) were synthesized as a new label. Peroxidase-mimic properties of Au@Ag NPs allowed for the catalytic enhancement of the signal on test strips, increasing the assay sensitivity. A mechanism of Au@Ag NPs-mediated catalysis was deduced. Signal amplification was achieved through the oxidative etching of Au@Ag NPs by hydrogen peroxide. This resulted in the formation of gold nanoparticles and Ag+ ions, which catalyzed the oxidation of the peroxidase substrate. Such "chemical enhancement" allowed for reaching the instrumental limit of detection (LOD, calculated by Three Sigma approach) and cutoff of 0.8 and 20 pg/mL, respectively. The enhanced assay procedure can be completed in 21 min. The enhanced LFIA was tested for GAT detection in raw meat samples, and the recoveries from meat were 78.1–114.8%. Aim: Long‐standing theory predicts that the intensity of biotic interactions increases from high to low latitudes. Studies addressing geographic variation in predation on insect prey have often relied on prey models, which lack many characteristics of live prey. Our goals were to explore global latitudinal patterns of predator attack rates on standardised live insect prey and to compare the patterns in predation on live insects with those on plasticine prey models. Location: Global forested areas. Time Period: 2021–2023. Major Taxa: Arthropods, birds. Methods: We measured predation rates in 43 forested locations distributed across five continents from 34.1° S to 69.5° N latitude. At each location, we exposed 20 sets of three bait types, one set per tree. Each set included three live fly larvae (maggots), three live fly puparia and three plasticine models of the puparia. We used glue rings to isolate half of the sets from non‐flying predators. Results: Arthropod attack rates on plasticine prey decreased linearly from low to high latitudes, whereas attack rates on maggots had a U shaped distribution, with the lowest predation rates at temperate latitudes and the highest rates at tropical and boreal latitudes. This difference emerged from intensive predator attacks on live maggots, but not on plasticine models, in boreal sites. Site‐specific attack rates of arthropod predators on live and plasticine prey were not correlated. In contrast, bird attack rates on live maggots and plasticine models were positively correlated, but did not show significant latitudinal changes. Main Conclusions: Latitudinal patterns in predation differ between major groups of predators and between types of prey. Poleward decreases in both arthropod and combined arthropod and bird predation on plasticine models do not mirror patterns of predation on our live prey, the latter likely reflecting real patterns of predation risk better than do patterns of attack on artificial prey. Tylosin (TYL) is a macrolide antibiotic widely used in animal husbandry. Due to associated health risks, there is a demand for sensitive methods for mass screening of TYL in products of animal origin. This article describes the development of lateral flow immunoassays (LFIAs) for TYL detection using direct (anti-TYL antibodies conjugated with nanoparticles) and indirect antibody labeling (anti-species antibodies conjugated with nanoparticles and combined with native anti-TYL antibodies). The choice of LFIA conditions, such as concentrations of hapten–protein conjugates, specific antibodies, and gold nanoparticle (GNP) conjugates with antibodies, as well as incubation time of reagents and the concentration of detergent in the sample buffer, is presented. The achieved limits of TYL detection using LFIAs with indirect labeling were 0.8 ng/mL (visual) and 0.07 ng/mL (instrumental), compared to 4 ng/mL (visual) and 0.4 ng/mL (instrumental) for the case of direct labeling. The sensitivity of the LFIA using the indirect format was up to seven times higher, allowing the determination of the target analyte at low concentrations. Study of the issues of identification, analysis, and comparative assessment of the risks of violations of the safety of the main railway transport infrastructure. The aim of the study: is to develop a methodology for the identification of economic risks of investment projects of construction of mainline infrastructure of railway transport. The object of the study: railway transport enterprises involved in all stages of the life cycle of the mainline infrastructure of railroad transport. The subject of the study: tools and procedures for the identification of economic risks of violations of the security of trunk railway transport infrastructure. In the turbulent global economy, the effectiveness of project management is based on the perception of the human factor as the main in the formation of aggregate uncertainty. Large infrastructure companies should pay special attention to the identification, analysis, and assessment of project risks. One of the key figures in Russia's transport communication, influencing not only domestic traffic flows, but also international logistics chains, is JSC "Russian Railways". Disruptions of the transportation process due to the realization of risks at the main infrastructure facilities of JSC "Russian Railways" predetermine economic consequences both for consumers of services, operators, and infrastructure owners.
