UNLABELLED: The bacterial chemotaxis system is a well-understood signaling pathway that promotes bacterial success. Chemotaxis systems comprise chemoreceptors and the CheA kinase, linked by CheW or CheV scaffold proteins. Scaffold proteins provide connections between chemoreceptors and CheA and also between chemoreceptors to create macromolecular arrays. Chemotaxis is required for host colonization by many microbes, including the stomach pathogen Helicobacter pylori. This bacterium builds chemoreceptor-CheA contacts with two distinct scaffold proteins, CheW and CheV1. H. pylori cheW or cheV1 deletion mutants both lose chemoreceptor array formation, but show differing semisolid agar chemotaxis assay behaviors: ∆cheW mutants exhibit total migration failure, whereas ∆cheV1::cat mutants display a 50% reduction. On investigating these varied responses, we found that both mutants initially struggle with migration. However, over time, ∆cheV1::cat mutants develop a stable, enhanced migration capability, termed migration-able (Mig+). Whole-genome sequencing analysis of four distinct ∆cheV1::cat Mig+ strains identified single-nucleotide polymorphisms (SNPs) in hpg27_252 (hp0273) that were predicted to truncate the encoded protein. Computational analysis of the hpg27_252-encoded protein revealed it encoded a hypothetical protein that was a remote homolog of the PilO Type IV filament membrane alignment complex protein. Although H. pylori lacks Type IV filaments, our analysis showed it retains an operon of genes for homologs of PilO, PilN, and PilM. Deleting hpg27_252 in the ∆cheV1::cat or wild type strain resulted in enhanced migration in semisolid agar. Our study thus reveals that while cheV1 mutants initially have significant migration defects, they can recover the migration ability through genetic suppressors, highlighting a complex regulatory mechanism in bacterial migration. IMPORTANCE: Chemotactic motility, present in over half of bacteria, depends on chemotaxis signaling systems comprising receptors, kinases, and scaffold proteins. In Helicobacter pylori, a stomach pathogen, chemotaxis is crucial for colonization, with CheV1 and CheW as key scaffold proteins. While both scaffolds are essential for building chemoreceptor complexes, their roles vary in other assays. Our research reexamines cheV1 mutants behavior in semisolid agar, a standard chemotaxis test. Initially, cheV1 mutants exhibited defects similar to those of cheW mutants, but they evolved genetic suppressors that enhanced migration. These suppressors involve mutations in a previously uncharacterized gene, unknown in motility behavior. Our findings highlight the significant chemotaxis defects in cheV1 mutants and identify new elements influencing bacterial motility.

Essential oils (EOs) from plants have attracted the attention of scientists due to their multiple biological activities. Antimicrobial activity of essential oils is the main reason for their widespread use in industries, which is determined through minimum inhibitory concentration (MIC) by using in vitro models. Therefore, the MIC is a crucial parameter to evaluate the antimicrobial potential of EOs. 492 papers were screened through the critical review of the related literatures in the past 50 years using four databases and read deeply for an outline. This review first listed common MIC determination methods of EOs, including agar diffusion method, agar dilution method, broth dilution method, E-Test, thin-layer chromatography-bioautography, etc. and their merits and demerits were introduced in detail. Agar and broth dilution methods are the gold standards in a more exact approach for MIC determination. Then, their functions and factors influencing the accuracy were listed and analyzed deeply. The application of the MIC determination in food and pharmaceuticals was finally discussed. Introduction: Cow's urine has been described as a highly effective animal-derived substance. Its therapeutic and pharmacological actions make it effective in the treatment of a variety of disorders. One of its notable properties is its antibacterial action, where the drug's ability to target and inhibit the growth of bacteria. However, due to the pungent odour and taste of cow's urine, it is difficult to consume in its natural form. Additionally, the collection of fresh cow's urine can be challenging, especially in urban areas where access to cows may be limited. So, it is important to modify it into a dosage form, like Gomutra Ghana, which improves palatability and shelf life. Aim: To study the pharmaceutical development, quality control assessment, and in-vitro antibacterial activity evaluation of Gomutra Ghana. Materials and Methods: The pharmaceutical and analytical studies were carried out in Mahatma Gandhi Ayurved College Hospital and Research Centre and Dattatraya Ayurved Pharmacy, Salod (H), Wardha, Maharashtra, India. The in-vitro (antibacterial) study was carried out in JNMC, Sawangi (M), Wardha, Maharashtra, India, from April 2022 to August 2023. The in-vitro antibacterial study of cow's urine and Gomutra Ghana was performed on five bacterial strains. Fresh cow's urine was collected from a goshala (cowshed), and Gomutra Ghana was prepared in three batches by boiling the cow's urine to a thicker consistency, then sun-drying it, followed by drying in an electric dryer. The final product was stored in a clean, sealed glass jars. Analytical parameters like pH, loss on drying at 105ºC, total ash, water-soluble ash, acid-insoluble ash, water-soluble extractives, alcohol-soluble extractives, and microbial contamination were studied. The in-vitro antibacterial activity was evaluated using the agar disc diffusion method at concentrations of 5 μL, 10 μL, and 15 μL, with results compared to those of a broad-spectrum and commonly used antibiotic, ciprofloxacin (5 μg). Statistical analysis was performed using Statistical Package for the Social Sciences (SPSS) version 27.0, employing Analysis of Variance (ANOVA) to compare three groups (Ciprofloxacin, cow's urine, and Gomutra Ghana) at all three concentrations. A significance level was set with a p-value of <0.05 for the antibacterial study. Results: High-Performance Thin-Layer Chromatography (HPTLC) showed a peak from cow's urine and Gomutra Ghana, with maximum heights of 500.1 and 450.4, which were nearly the same, so they may contain similar constituents. An extra peak was observed in Gomutra Ghana, suggesting the presence of additional compounds formed during its preparation. Gomutra Ghana extract showed the highest zone of inhibition against Escherichia coli. Conclusion: Gomutra Ghana showed antibacterial properties, which warrant further exploration in humans for safety and efficacy in humans. [ABSTRACT FROM AUTHOR]

Bio-artificial photosynthetic systems can reduce CO2 into multicarbon compounds by simulating natural photosynthesis. Here, inspired by organic photovoltaic structures, we demonstrate a bio-artificial photosynthetic system based on the hybridization of polymer semiconductor films and bacteria. The study suggests that the polymer-based semiconductor film can efficiently drive the non-photosynthetic bacteria to convert CO2 to acetate. By systematically characterizing the charge transport behavior of the bio-artificial photosynthetic system, the bulk-heterojunction structure and charge transport layers are proven to enhance the system performance markedly. The scalable floating artificial bio-leaf system can produce acetate to gram scale in a week. Notably, the semiconductor film is easy to recycle and maintains stable performance, showing good sustainable production capability of the system. A quasi-solid-state artificial bio-leaf is successfully prepared using agar to simulate the morphology and function of natural leaves. Last, the acetate production converted from CO2 was used to grow yeast for food production, thus achieving a complete simulation of natural photosynthesis. [ABSTRACT FROM AUTHOR]

Background There are limited treatment options for prosthetic joint infections (PJI) due to multidrug-resistant Staphylococcus epidermidis (MDRSE). Fosfomycin (FOF) has gained attention as a potential therapy, but there is a paucity of information on the phenotypic and genotypic susceptibility amongst S. epidermidis , including MDRSE. Objectives To investigate phenotypical and genotypical susceptibility to FOF in S. epidermidis isolates prospectively collected from PJIs in Sweden. Methods MIC determination was performed using in-house agar dilution (AD) and a commercial AD panel. Genes and gene variants associated with FOF resistance were analysed. Results Multidrug resistance was common [74/89 (83%) isolates were MDRSE]. FOF inhibited all isolates except one, which had an MIC > 256 mg/L. The commercial AD panel demonstrated good overall performance but tended to overestimate the MIC, resulting in 84% essential agreement with the gold standard. Genomic analysis with publically available tools for whole-genome sequencing (WGS) data suggested genotypic FOF resistance in all isolates, but in-depth analysis revealed that fosB , associated with FOF resistance, was only present in the phenotypically resistant isolate. No other genes or gene variants associated with FOF resistance were detected. Conclusions Phenotypic resistance to FOF and presence of fosB were rare in this collection, indicating FOF's potential as a treatment option for S. epidermidis. The commercial AD panel demonstrated high reproducibility, but EA with the reference method was less than optimal. Findings of genotypic FOF resistance using common tools for WGS data should be critically evaluated and appropriately verified with relevant fosB references for S. epidermidis. Objectives Neisseria gonorrhoeae strains associated with the high-level ceftriaxone-resistant FC428 clone or containing its main resistance determinant, penA allele 60.001, have shown global transmission. In Hangzhou, China, 10% of the isolates were associated with the FC428 clone in 2019. Here, we investigated ceftriaxone resistance and the prevalence of FC428-associated strains in Hangzhou in 2020–22. Methods A total of 209 gonococcal isolates were investigated for antimicrobial susceptibility to ceftriaxone and other antibiotics by agar dilution method. Sequence types and penA alleles were determined by PCR and sequence analysis. Results Resistance to ceftriaxone (MIC > 0.125 mg/L) was observed for 16% (33/209) of the isolates, whereas 6.7% (14/209) of the isolates displayed high-level ceftriaxone resistance (MIC = 1 mg/L). These 14 high-level ceftriaxone-resistant isolates and another isolate displaying an MIC = 0.25 mg/L contained penA allele 60.001, with eight of these isolates, all from 2020 to 2021 belonging to MLST ST1903, the sequence type commonly associated with the original FC428 clone. Importantly, the six penA allele 60.001-containing isolates from 2022 belonged to MLST ST8123, ST7365 and ST7367, which are among the most frequently encountered sequence types found in China. Therefore, these results indicate that endemic lineages in China have acquired penA allele 60.001. Conclusions Here, we report continued transmission of gonococcal strains associated with the FC428 clone or containing penA allele 60.001 in Hangzhou. A major concern for public health is the acquisition of penA allele 60.001 by successful endemic lineages, which might enhance the transmission of this high-level ceftriaxone resistance trait. Objective: This research aimed to analyze the prevalence, molecular characteristics, toxinotyping, alpha toxin production potential, and antibiotic resistance pattern of Clostridium perfringens (C. perfringens) isolates in meat samples collected from various sources. Methods: Sixty meat samples were screened for alpha toxin using enzymelinked immunosorbent assay, revealing a positivity rate of 13.3%, predominantly in raw poultry meat. Subsequent culturing on Perfringens agar identified nine samples harboring characteristic C. perfringens colonies, primarily isolated from raw poultry meat. Molecular confirmation through 16S rRNA gene amplification and sequencing authenticated twelve isolates as C. perfringens, with nine strains exhibiting genetic resemblance to locally isolated strains. Toxinotyping assays targeting alpha toxinspecific genes confirmed all nine isolates as type A C. perfringens, with no detection of beta or epsilon toxin genes. Hemolytic assays demonstrated varying alpha toxin production potentials among isolates, with accession number OQ721004.1 displaying the highest production capacity. Moreover, antibiotic resistance profiling revealed multidrug resistance patterns among the isolates. Results: The study identified distinct clusters within C. perfringens strains, indicating variations. Phylogenetic analysis delineated genetic relatedness among strains, elucidating potential evolutionary paths and divergences. Conclusion: The findings underscore the need for robust surveillance and control measures to mitigate the risk of C. perfringens contamination in meat products, particularly in raw poultry meat. Enhanced monitoring and prudent antimicrobial stewardship practices are warranted in both veterinary and clinical settings to address the observed antibiotic resistance profiles and prevent foodborne outbreaks. Locomotion is essential to most of the living organisms. The nematode Teratorhabditis palmarum (Gerber & Giblin-Davis, 1990), must be able to accommodate its locomotory gait to its physical environment. Particularly, when crawl on a solid surface, it shows forward movement as the contraction waves propagates from anterior to the posterior region. In the present study, we analyzed the locomotory behavior of T. palmarum on agar by using computational worm-tracking system and mathematical tools. Our tracking system was made to assist worms that travel a vast area with recurrent turns and reversals at high speeds. The locomotory parameters studied were velocity, wavelength, amplitude, angle of bending, curvature map, forward, reversal, omega and pause that was extracted from a two min video recording. Here, we examined the locomotory behaviour (movement and wave characteristic) of two, six and ten days old nematodes. In movement characteristic, except pause which increases; forward wave, backward wave and omega wave decrease significantly. In wave characteristic, wavelength and amplitude increase while average velocity and bending angle decrease significantly. Locomotion was observed between various age groups and it was concluded that as the nematodes aged, mild locomotory deficits appeared that were enough to reduce behavioral activity. Overall, our result provides the detailed analysis of locomotion and its dynamics in enabling better understand the mechanism of muscle ageing. [ABSTRACT FROM AUTHOR]

In this research, biomass of the red algae Phycocalidia vietnamensis (Bangiales) was investigated for agar production. Agar was prepared by the hot water extraction method with 0, 4, 6, 8, 10% (w/v) NaOH pre-treatments. Samples without NaOH pre-treatment did not form gel. The agar yield of the NaOH pre-treated samples ranged from 7.13 to 12.42% dry weights. The highest gel strength of 596 ± 35 g/cm2 was recorded for agar treated with 10% NaOH, followed by 350 ± 17 g/cm2 with 8% and 150 ± 23 g/cm2 with 6%. In addition, the extracted agar was tested for bacteriological application, and total bacterial counts ranged from 1.2 × 103 to 2.4 × 103 CFU mL− 1. This study suggests that the agar extracted with 10% NaOH pre-treatment is suitable for bacteriological applications, while agar obtained with 6% and 8% NaoH pre-treatment could be used in the food industry. Agar, a phycocolloid naturally extracted from Gracilariopsis heteroclada is strongly influenced by salinity. Wild stocks of G. heteroclada were exposed to varying salinities (20, 30, 40, 50, 60) for 6 days, and subsequently processed for agar extraction using 5 % NaOH. The extracted agar was evaluated regarding yield, gel rheology, colour composition, chemical properties, and infrared spectra. Results highlighted that hypersaline conditions (salinity 50) could produce high agar yield (4.77 %) and viscosity (10.67 mPa s), while agar samples at salinity 40 exhibited gels with high cohesiveness (6.35 mm), gel breaking strength (3.01 N), and gel strength (390.61 g cm−2) while having a rather high 3,6-anhydrogalactose content (7.49 %). All samples exhibited FTIR signature peaks at 930 cm−1, confirming the identity of extracted agar from G. heteroclada. Exposure at increasing hypersalinity increased the sulphate levels of agar from G. heteroclada, which implies synthesis of sulphated polysaccharides. Moreover, high 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activities were obtained in acid hydrolysed agars at salinities of 40 (74.09 %) and 50 (75.57 %), suggesting that G. heteroclada agars from hypersaline conditions potentially offer antioxidative roles beyond its traditional food use. This article explores the role of LRPPRC in pancreatic cancer progression. The study found that LRPPRC is upregulated in pancreatic cancer tissues and cell lines and is negatively associated with overall survival rate. Knockdown of LRPPRC led to increased reactive oxidative species accumulation, decreased mitochondrial membrane potential, and induced autophagy/mitophagy and mitochondrial dysfunction. The study suggests that LRPPRC may act as an oncogene by maintaining mitochondrial homeostasis and could be used as a predictive marker for patient prognosis in pancreatic cancer. Using agar and quaternized chitosan with good water solubility as starting materials, quaternized chitosan-agar hydrogel particles were prepared by non-covalent bonding and the sustained-release property of quaternized chitosan-agar hydrogel particles on urea was studied. The structure of the product was characterized by Fourier transform infrared spectroscopy, X-ray diffraction and scanning electron microscopy・ The results showed that quaternized chitosan-agar hydrogel particles with porous structure have been synthesized and successfully loaded with urea・ The prepared hydrogel particles had good swelling rate (397・ 53%) and the encapsulation rate of urea (51. 83%)・ The cumulative release rate of urea was 81. 37% after 72 h at room temperature, showing a good slow-release effect. Food texture is one of the most important factors for assessing the quality and acceptability of food. However, the study of food texture has been delayed compared with other factors, such as flavor and taste, due to the difficulty of quantitative analysis related to real physiological senses. Furthermore, the numerical and systematic evaluation of the texture property of dispersion systems, in which solid particles are dispersed in a liquid medium, is very difficult, despite most foods being in a solid–liquid dispersion state during mastication in the human mouth. In this study, the texture property of a solid–liquid dispersion system with spherical and cubic gel particles of agar and konjac was examined to evaluate the physical behavior of food during mastication using the back extrusion method. The yield stress of the system strongly depended on the size and shape of the particles, the mixing ratio of particles of different sizes and shapes, and the concentration of components in the particles. The proposed index, reflecting the size, shape, and number of particles and the yield stress of a single particle, expressed well the measured yield stress of the entire dispersion system. However, the adhesiveness and recoverability showed relatively little dependence on particle size. The findings obtained in this study will contribute to elucidating the texture property of various foods and to the development of new and novel food products and cuisines, thereby benefiting food science and industry. Calcium alginate spheres with a volume of about 5 mL can be used for important purposes. Those that incorporate oolong tea give, in addition to the recreational aspect, the possibility of drinking small quantities of this tea, because oolong tea can be used as a compound with antioxidant properties. This incorporation can be achieved by reverse spherification (5 mL). Six types of spheres have been made, all of them with calcium alginate and the presence or absence of agar-agar and xanthan gum—XG—in two concentrations. The weight loss of the spheres, the release of bioactive compounds over time (a total of 48 h), the surface (internal and external) of the membranes, and the physical characteristics of these membranes have been analyzed. The data obtained indicate that the presence of XG prevents the formation of precipitates inside the spheres and slows down weight loss. It also provides opacity to the spheres. However, the incorporation of agar-agar does not have a significant influence on the different parameters analyzed. The release of catechins reaches a maximum of 80% of what could be achieved under ideal conditions, and it reaches 90% in the first 3 h. The incorporation of XG increases the shelf life of the spheres, slows down the release of chelate, and decreases weight loss over time, allowing for a new perspective on the spherification process. Gelling agents are necessary for the preparation of solid or semisolid media. For more than a hundred years, agar has been the primary gelling agent. However, a substantial body of evidence has accumulated suggesting that agar-based media inhibit the growth of many microbial species through the generation of reactive oxygen species (ROS), toxic organic contaminants, or competitive exclusion effects. In this review we have compiled the largest amount of data to date on the use of various gelling agents in microbial isolation and cultivation, with the particular emphasis on rare microbe isolation cases. Our analysis suggested that microbial-derived compounds (especially gellan gum), as gelling agents, are superior to agar in their ability to isolate and maintain either new or known microbial species. We analyzed the reasons behind this success and concluded that there are phylum-level differences in microbial responses to the changes in conditions from natural to the laboratory conditions (with respect to gelling agent usage). Consequently, we hypothesize that at least partial success of microbial-derived gelling agents lies in the recreation of the natural microenvironment conditions (which we address as the “familiarity of conditions” hypothesis). Red algae, abundant along the Moroccan coast, have fostered an agar-agar industry. However, industrial processing of red algae produces significant fibrous waste, posing environmental challenges. A new challenge is to find sustainable methods for repurposing this fibrous waste. Valorizing red-algae fibers in building materials promotes sustainable development and eco-construction. This study aims to valorize these red-algae residues as fiber reinforcements in compressed earth bricks (CEB). Various mass ratios of red-algae fibers (0.5%–3%) were incorporated into CEB. The addition of raw red-algae fibers to mixtures slightly increased the Atterberg limit values of the earth mixture. A slight adjustment in manufacturing water content was also observed with fiber addition. The incorporation of red-algae fibers did not improve the mechanical strengths of the CEB. Nevertheless, at a 1.5% addition ratio, CEB exhibited compressive and flexural strengths of 4.07 and 0.77 MPa respectively. To enhance mechanical properties, fibers were pre-saturated and subjected to alkaline and double coating treatments. Untreated and treated fibers were added to CEB at an optimal mass addition rate of 1.5%. Pre-saturated fibers showed a slight improvement of 5% in mechanical strength. However, alkali-treated and double-coated fibers offer a significant improvement of 20% in the mechanical strengths of CEB, demonstrating the efficacy of treatments. [ABSTRACT FROM AUTHOR]

Tissue-mimicking materials (TMMs) are now essential reference objects for quality control, development and training in all medical imaging modalities. This review aims to provide a comprehensive synthesis of materials used in the fabrication of TMMs for MRI phantoms, focusing on their composition, fabrication methods, and relaxation properties (T1 and T2). A systematic review was conducted, covering articles published between 1980 and 2023. Inclusion criteria encompassed studies involving physical MRI phantoms with measured T1 and T2 relaxation times. Exclusion criteria filtered out non-MRI studies, and digital/computational models. The review identifies and categorizes TMMs based on their primary gelling agents: agar, carrageenan, gelatin, polyvinyl alcohol (PVA), and other less common gels. Agar emerged as the most frequently used gelling agent due to its versatility and favorable MRI signal properties. Carrageenans, noted for their strength and minimal impact on T2 values, are often used in combination with agar. Gelatin, PVA, and other materials like Polyvinyl chloride (PVC) and PolyvinylPyrrolidone (PVP) also demonstrate unique advantages for specific applications. The review also highlights the challenges in phantom stability and the impact of various additives on the relaxation properties. This synthesis provides a valuable guide for the fabrication of MRI phantoms tailored to desired T1 and T2 relaxation times, facilitating the development of more accurate and reliable imaging tools. Understanding the detailed properties of TMMs is fundamental to improve the quality control and educational applications of MRI technologies, especially with the advent of new magnetic field strengths and parametric imaging techniques. As experts in MRI systems, radiographers, educators, and researchers need to understand TMM compositions and methods of fabrications to develop MRI phantoms for educational tools and research purposes. Essential oils (EOs) have been gaining popularity in the past decades among researchers due to their potential to replace conventional chemicals used in the fight against pests, pathogenic and spoilage microbes, and oxidation processes. EOs are complex mixtures with many chemical components, the content of which depends on many factors—not just the plant genus, species, or subspecies, but also chemotype, locality, climatic conditions, phase of vegetation, method of extraction, and others. Due to this fact, there is still much to study, with antimicrobial effect being one of the key properties of EOs. There are many methods that have been frequently used by researchers for in vitro evaluation; however, although the research has been going on for decades, an internationally accepted standard is still missing. Most of methods are based on time-proven standards used for the testing of antibiotics. Due to the specific properties of EOs and their components, such as volatility and hydrophobicity, many modifications of these standard procedures have been adopted. The aim of this review is to describe the most common methods and their modifications for the testing of antimicrobial properties of EOs and to point out the most controversial variables which can potentially affect results of the assays. Background. Since the ants play a key role in transmitting microorganisms, it is important to identify the fungal species isolated from the external surfaces of ants caught from hospitals. Therefore, the present study aimed to identify the fungal species isolated from the external surfaces of ants caught from Babol hospitals. Methods. Ants were collected by trapping inside sterile tubes and were washed with normal saline for 20 seconds. Then, 100 μL of the resulting solution was cultured in Sabouraud Dextrose Agar (SDA) medium and incubated for one week at 35C°. Macroscopic, microscopic, and RFLP-PCR molecular techniques were employed to confirm the grown isolates. Results. A total of 79 fungal isolates including 47 isolates of hyaline fungi (59.49%) and 32 isolates of black fungi (40.51%) were obtained from 63 captured ants. Out of 47 cultured fungi, 15 fungi (31.91%) were Aspergillus niger and 15 (31.91%) were A. flavus, 12 (25.53%) were A. fumigatus and 5 isolates were Penicillium (10.63%). Conclusion. Taking into account the increasing number of immunocompromised patients, outbreak of COVID-19 in recent years, and numerous reports about the invasive fungal diseases among COVID-19 patients, it was found necessary to control the ants as carriers of these organisms. Practical Implications. It was necessary to control these insects effectively in order to prevent the increasing prevalence of the fungal diseases among the hospitalized people. Precision in dosing is crucial for optimizing therapeutic outcomes and preventing overdosing, especially in preterm infants. Traditional manual adjustments to adapt the dose often lead to inaccuracies, contamination risks, and reduced precision. To overcome these challenges, semi-solid extrusion 3D printing was used to create personalised gel-based caffeine dosage forms. The hydrogels, made from agar and hydroxypropyl methylcellulose, demonstrated excellent rheological properties, ensuring uniform extrusion and accurate shape retention during and after printing. This gel formulation allowed for precise adjustments of caffeine volume and content tailored to a neonate weighing 1.36 kg, achieving a recovery of 103.46%, well within acceptable limits. Additionally, three production batches confirmed the process's reproducibility with minimal variability. Forced degradation studies showed that both pure caffeine and caffeine in the gel matrix exhibited similar stability profiles, confirming the drug's chemical integrity. The printed gel dosage forms also displayed immediate-release characteristics, with over 80% of caffeine released within 45 min, highlighting their suitability for rapid therapeutic action. These findings emphasise the potential of SSE 3DP and gel-based formulations to produce personalised drug delivery systems with high precision, reproducibility, and reliability. Acne is a common skin condition that affects people of all ages and can lead to significant physical and psychological distress. The first line of action against acne is topical products, though the most effective are topical antibiotics. In such a scenario, the development of effective and safe herbal formulations for the treatment of acne is of great importance. Rubia cordifolia, Aloe barbadensis, and Allium cepa extracts are rich sources of bioactive metabolites and are safe compared with antibiotics, in addition to being cost effective, sustainable, and eco-friendly. Also, their combination has not been studied for treating acne, and their potential benefits need to be investigated. The present study aimed to develop an effective polyherbal gel formulation of R. cordifolia, A. barbadensis, and A. cepa combined extract for treating acne and validate its effect with reference to conventional antibiotics. Plant materials were extracted in water by the reflux method, and phytochemical analysis was done for flavonoid, anthraquinone, and phenolic contents. The combined extract (R. cordifolia, A. barbadensis, and A. cepa extracts) was formulated in gel. The selected polyherbal gel was evaluated for in vitro antibacterial activity using agar well diffusion against Cutibacterium acnes (P. acnes) culture. Phytochemical analysis of the composite extract revealed the rich presence of flavonoids, phenolics, and anthraquinones. The polyherbal gels showed good physicochemical properties; however, FCEG-4 was selected for further studies. It was found to be effective against C. acnes (MTCC 1951) in agar well diffusion, as it showed a similar zone of inhibition as that of standard. Also, during in vivo studies, FCEG-4 showed comparable efficacy with clindamycin gel. It was concluded from the study that composite extracts incorporated in an aqueous-based gel system were effective in topical therapy of mild acne vulgaris, showing similar efficacy to that of clindamycin cream. Biomaterials engineering approaches for treating bone defects involve utilizing a combination of potent bioactive molecules to stimulate cell proliferation, fostering a conducive environment and scaffold for the regeneration process. Due to the aging global population, there is an urgent need for research in bone regeneration and wound healing. Hydroxyapatite (HAP) is a major mineral component of bone tissue with high biocompatibility and bioactivity. Agar and carboxymethyl cellulose (CMC) both exhibit the essential characteristics of biomaterials, either separately or in combination. Hence, this present study aimed to prepare HAP nanoparticles loaded Agar/CMC composite film for enhanced bone regenerative applications. The crystal structure, morphology, phase composition, thermal stability, and chemical state of the film composites were characterized using XRD, SEM, TGA, and FTIR. Cytotoxicity evaluation on rat fibroblasts cells indicated over 90% biocompatibility for the film composites. Moreover, in wound healing assays, the nanocomposite film-treated group (98.14 ± 0.15%) exhibited a 35% higher wound closure rate compared to the negative control group (62.08 ± 1.87%). Alizarin Red Staining assay revealed a 20.89 ± 6.9% increase in calcium deposition in treated MC3T3-E1 cells compared to the negative control, affirming their osteogenic potential. An Aceclofenac jelly is a just emerging formulation which is used to relieve menstrual cramps. The main aim of study is to create and evaluate the oral medicated jellies of Aceclofenac. Aceclofenac has the ability to inhibit synthesis of prostaglandins by inhibiting the cyclo-oxygenase enzyme, these prostaglandins are responsible for pain. Heating and congealing method were used to produce the jelly using sugar, gelatin and china grass (agar agar). The formulated jellies were evaluated for physical appearance, dissolution, viscosity, pH, drug content, weight variation and stability studies. Formulation F6 was optimized based on the evaluation tests. The formulation F6 was further subjected to stability studies for 3 months according to ICH guidelines and it was found that nothing is altered that was able to be seen. [ABSTRACT FROM AUTHOR]

Antifungal resistance remains a significant public health threat, yet poorly investigated. This study for the first time investigated the antifungal profile of moulds isolated from groundwaters in a rural community of Osun State, Southwest Nigeria. Groundwater samples were collected during dry and rainy seasons, and processed for mould isolation using standard methods. The isolates were presumptively identified and screened for in vitro susceptibility using 4 antifungals conventionally employed in clinical cases. Multiple antifungal resistance phenotypes and indices were equally evaluated. Representative of each putatively identified species were confirmed using molecular techniques. A total of 29 and 27 moulds were obtained during the dry and rainy seasons, respectively. Genus Aspergillus was the most prevalent (30.4%). Of the 56 isolates subjected to antifungal susceptibility testing, 55(98%) were resistant to fluconazole, followed by flucytosine 49(88%), ketoconazole 45(80%) and amphotericin B 31(55%). Exactly 46 isolates were multidrug-resistant, 23(50%) each, to 3 and 4 drugs, respectively. The multiple antifungal resistance index (MARI) evaluated was 1 in all the samples A-Q, except C (0.75), transcending the threshold limit of 0.2, indicating the isolates to be of high antifungal usage origin. The molecularly identified species had 99–100% similarity with reference strains from GenBank and evolutionary relationship of taxa revealed 10 distinct clades. The outcome of this study reveals a rise in the frequency of antifungal resistance in moulds towards standard drugs, thus, advocating for adequate investigations targeted at monitoring antifungal resistance in groundwater milieus. Microalgae and seaweed have been consumed as food for several decades to combat starvation and food shortages worldwide. The most famous edible microalgae species are Nostoc, Spirulina, and Aphanizomenon, in addition to seaweeds, which are used in traditional medicine and food, such as Nori, which is one of the most popular foods containing Pyropia alga as a major ingredient. Recently, many applications use algae-derived polysaccharides such as agar, alginate, carrageenan, cellulose, fucoidan, mannan, laminarin, ulvan, and xylan as gelling agents in food, pharmaceuticals, and cosmetics industries. Moreover, pigments (carotenoids particularly astaxanthins, chlorophylls, and phycobilins), minerals, vitamins, polyunsaturated fatty acids, peptides, proteins, polyphenols, and diterpenes compounds are accumulated under specific cultivation and stress conditions in the algal cells to be harvested and their biomass used as a feedstock for the relevant industries and applications. No less critical is the use of algae in bioremediation, thus contributing significantly to environmental sustainability. This review will explore and discuss the various applications of microalgae and seaweeds, emphasising their role in bioremediation, recent products with algal added-value compounds that are now on the market, and novel under-developing applications such as bioplastics and nanoparticle production. The transition between yeast and hyphae is crucial for regulating the commensalism and pathogenicity in Candida albicans. The mechanisms that affect the invasion of hyphae in solid media, whose deficiency is more related to the pathogenicity of C. albicans, have not been elucidated. Here, we found that the disruption of VAM6 or VPS41 which are components of the homotypic vacuolar fusion and protein sorting (HOPS) complex, or the Rab GTPase YPT72, all responsible for vacuole fusion, led to defects in hyphal growth in both liquid and solid media, but more pronounced on solid agar. The phenotypes of vac8Δ/Δ and GTR1OE-vam6Δ/Δ mutants indicated that these deficiencies are mainly caused by the reduced mechanical forces that drive agar and organs penetration, and confirmed that large vacuoles are required for hyphal mechanical penetration. In summary, our study revealed that large vacuoles generated by vacuolar fusion support hyphal penetration and provided a perspective to refocus attention on the role of solid agar in evaluating C. albicans invasion. The present study was designed to evaluate the cardiotoxicity effect of formulated agar (FA) from Turbinaria conoides in zebrafish embryos (ZFE). The agar was formulated by aqueous extract method. The physio-chemical and structural characterization was analyzed by texture analysis, Fourier transformed infrared spectroscopy (FTIR), scanning electron microscope (SEM), and thermogravimetric analysis. Spreadability and in vitro sample diffusion assays were determined. The agar isolated from T. conoides had a yield of 84.2% and moisture content of 38%. The FTIR analysis indicated the presence of alkanes and aliphatic amines while SEM analysis showed densely packed structure. The FA showed a water loss of 115.2, spreadability of 72.12%, and in vitro release of 98% in 3 h. At a concentration of 0.02 μg/mL in ZFE, FA's cardiotoxicity and teratogenic effects did not change significantly (P>0.05). [ABSTRACT FROM AUTHOR]

Filamentous fungi and fungal-like organisms contribute to a wide range of important ecosystem functions. Evidence has shown the movement of liquid across mycelial networks in unsaturated environments, such as soil. However, tools to investigate liquid movement along hyphae at the level of the single cell are still lacking. Microfluidic devices permit the study of fungal and fungal-like organisms with cellular resolution as they can confine hyphae to a single optical plane, which is compatible with microscopy imaging over longer timescales and allows for precise control of the microchannel environment. The aim of this study was to develop a method that enables the visualization and quantification of liquid movement on hyphae of fungal and fungal-like microorganisms. For this, the fungal–fungal interaction microfluidic device was modified to allow for the maintenance of unsaturated microchannel conditions. Fluorescein-containing growth medium solidified with agar was used to track liquid transported by hyphae via fluorescence microscopy. Our key findings highlight the suitability of this novel methodology for the visualization of liquid movement by hyphae over varying time scales and the ability to quantify the movement of liquid along hyphae. Furthermore, we showed that at the cellular level, extracellular movement of liquid along hyphae can be bidirectional and highly dynamic, uncovering a possible link between liquid movement and hyphal growth characteristics. We envisage that this method can be applied to facilitate future research probing the parameters contributing to hyphal liquid movement and is an essential step for studying the phenomenon of fungal highways. The aim of this experiment was to study the biological characteristics and inhibitory mechanism of a novel Lactobacillus bacteriocin against Salmonella typhimurium. Whole genome sequencing was used to analyze the genomic information of bacteriocin-producing lactic acid bacteria, agar diffusion and dialysis methods were used to determine the bacteriocin inhibitory activity and molecular weight range, organic acid, hydrogen peroxide exclusion test and protease sensitivity test were used to determine the nature of bacteriocin, and the biological properties of bacteriocin were investigated by setting up pH value and temperature gradients, determining the growth curves and bacteriocin inhibitory curves of lactic acid bacteria, and the preliminary exploration of bacteriocin inhibitory mechanism was carried out by scanning electron microscope observation. Scanning electron microscope observation was used to explore the mechanism of bacteriocin inhibition. The results showed that a strain of Lactobacillus PG-9 with bacteriocin inhibitory activity was identified as Limosilactobacillus fermentum by molecular biology, with a full genome size of 1 985 177 bp, containing 1 947 coding genes. The bacteriocin PG-9B it produced had good bacteriostatic activity against Salmonella typhimurium, with an estimated molecular weight of less than 1 kDa. After excluding the effects of organic acids and hydrogen peroxide, it was found to be sensitive to various proteases, in the range of pH 3.0~7.0 and -20~121 °C, high antibacterial activity was retained. Scanning electron microscopy showed that the Limosilactobacillus fermentum citriodora bacteriocin PG-9B inhibits and kills cells by disrupting the cell surface morphology of Salmonella typhimurium. The study indicates that the favorable biological properties of Limosilactobacillus fermentum bacteriocin PG-9B can be used as a potential alternative antioxidant additive for the control of Salmonella typhimurium in the feed industry. Lactobacilli are considered important probiotics for the prevention of some infections. In this study, the antifungal effect of both cells and cell-free supernatants of twenty-three strains of lactobacilli were investigated against Candida albicans by co-aggregation, agar diffusion assay, agar spot assay and co-culture assay. In all cases, a fungistatic effect was recorded. In the agar diffusion assay and agar spot assay, an effect was established primarily for heterofermentative species via the production of lactic acid. The anti-Candida effect was higher with microbial suspension than with cultural supernatants in the co-culture assay. A strain-specific reduction in the yeast growth up to 28.9% in MRS broth and up to 17.1% in BHI broth was observed. Cells of Limosilactobacillus fermentum LLF-01 and Limosilactobacillus reuteri LLR-K67 showed the highest activity in both model systems. For all strains, a lower reduction up to 9.7% was recorded with cultural supernatants. L. fermentum LLF-01 showed the highest ability of co-aggregation (64.8%) with C. albicans, followed by Lactobacillus acidophilus LLA-01, Lactobacillus gasseri LLG-V74, Lactobacillus delbrueckii subsp. bulgaricus LLB-02 and two strains of Lactobacillus delbrueckii subsp. lactis LLL-14 and LLL-F18. The present study showed the potential of several strains of lactobacilli to affect the population of C. albicans in vitro. The combination of cultures with proved anti-Candida and co-aggregation activity in a probiotic formula may have a positive effect for the prevention of yeast overgrowth in the gut and hence for the suppression of candidiasis. This study involved the synthesis of the UiO-67 metal-organic framework; UiO-67 is a well-known type of MOF obtained by coordinating the Zr6O4(OH)4 metal unit with the 4,42-biphenyldicarboxylate organic linker, using the hydrothermal technique. The novelty of the current work is to synthesize UiO-67 MOFs, and their application as biological agents for antibacterial and cancer cells. Subsequently, the composite material UiO-67 was subjected to a comprehensive characterization process involving Fourier-transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), thermal gravimetric analyses (TGA) and surface area analysis, and the results showed the successful synthesis of the UiO-67 MOFs, with a high specific surface area of 1415 m2/g. Thesynthesized UiO-67 for its antibacterial properties tested against five pathogenic bacterial strains, which include three gram-positive and mcillin-resistant pairs including MRSA, S. aureus and Enterococcus faecalis, and Two gram-negative bacteria E. colli and S. typhimurium using the agar well diffusion method. These findings have shown enhanced, strong antibacterial activity against all the five used gram-positive and gram-negative bacterial strains. Furthermore, the anticancer efficacy of UiO-67 was evaluated on two distinct types of cancer cells: We are using MCF-7 (human breast cancer cell line) and HepG2 (human liver cancer cells). Objectives The novel dual-target triazaacenaphthylene, gepotidacin, recently showed promising results in its Phase III randomized controlled trial for the treatment of gonorrhoea. We investigated alterations in the gepotidacin GyrA and ParC targets in gonococci by in silico mining of publicly available global genomes (n  = 33 213) and determined gepotidacin MICs in isolates with GyrA A92 alterations combined with other GyrA and/or ParC alterations. Methods We examined gonococcal gyrA and parC alleles available at the European Nucleotide Archive. MICs were determined using the agar dilution method (gepotidacin) or Etest (four antimicrobials). Models of DNA gyrase and topoisomerase IV were obtained from AlphaFold and used to model gepotidacin in the binding site. Results GyrA A92 alterations were identified in 0.24% of genomes: GyrA A92P/S/V + S91F + D95Y/A/N (0.208%), A92P + S91F (0.024%) and A92P (0.003%), but no A92T (previously associated with gepotidacin resistance) was found. ParC D86 alterations were found in 10.6% of genomes: ParC D86N/G (10.5%), D86N + S87I (0.051%), D86N + S88P (0.012%) and D86G + E91G (0.003%). One isolate had GyrA A92P + ParC D86N alterations, but remained susceptible to gepotidacin (MIC = 0.125 mg/L). No GyrA plus ParC alterations resulted in a gepotidacin MIC > 4 mg/L. Modelling of gepotidacin binding to GyrA A92/A92T/A92P suggested that gepotidacin resistance due to GyrA A92T might be linked to the formation of a new polar contact with DNA. Conclusions In silico mining of 33 213 global gonococcal genomes (isolates from 1928 to 2023) showed that A92 is highly conserved in GyrA, while alterations in D86 of ParC are common. Background: Plants have long been recognized as having a wide range of biological properties, including antibacterial, analgesic, anticancer, antipyretic, and antihypertensive action. They are also a significant source of several chemicals with biological activity. The Rutaceae family consists of a shrub Atalantia monophylla. It is available all over India. The extracts from its leaves have been reported to have antimicrobial and antiviral properties. Aims and Objectives: The objectives of the study were to evaluate the extraction protein content and antimicrobial activity of A. monophyla protein extract. The objective of the present study is to isolate protein extract from the aqueous extract of A. monophyla and to screen for antimicrobial activity. Materials and Methods: The A. monophylla protein isolate was prepared from aqueous extract. The protein isolate was done using the ammonium sulfate method and quantified using Lowry's method. Agar disc diffusion method and minimum inhibitory concentration were used to evaluate the antimicrobial activity of the extract. Results: The Lowry's method revealed 5.73 mg/mL protein isolate. With advantages including high capacity, intrinsic safety and low cost, aqueous zinc‐ion batteries (AZIBs) are ideal electrochemical energy storage devices for large‐scale and portable energy storage. However, the development of AZIBs suffers from tricky challenges, such as the notorious Zn dendrite growth and severe parasitic reactions. Herein, as a low‐cost and nontoxic biomass, agar is adopted to construct an interface layer on Zn foil to mitigate side reactions and induce uniform Zn deposition on Zn anodes. The interaction between Zn2+ and polar functional groups of agar can regulate Zn2+ distribution and promote Zn2+ desolvation, thus simultaneously achieving homogenous Zn deposition and suppressed hydrogen evolution reaction. Meanwhile, SO42− anions are blocked from contacting Zn surface due to electrostatic repulsion, greatly restraining corrosion and passivation. Consequently, Zn||A‐Cu asymmetric cell operates normally for 590 cycles with an average coulombic efficiency of 99.5 %, suggesting good reversibility of Zn plating/stripping. Notably, A‐Zn symmetric cell exhibits a long lifespan of 1100 h at 2 mA cm−2. Furthermore, the A‐Zn||NVO full cell displays a superb capacity retention of 94.8 % after 3600 cycles at 5 A g−1. Objective: This study focuses on Candida albicans, a significant fungal pathogen in humans, particularly affecting immunocompromised individuals and developing resistance to commonly used antifungal drugs. To address this challenge, the research assessed the in-vitro anti-candida properties of cold-pressed coconut oil. Additionally, the study conducted an in-silico evaluation of the oil's bioactive compounds against candidapepsin-2, an enzyme crucial in the virulence and pathogenesis of Candida infections. Materials and Methods: Extracted coconut oil was tested for its sterility and evaluated for its antifungal activity in-vitro using the agar well-diffusion methods with fluconazole as a positive control. Growth kinetics assay and synergism activity with fluconazole were also assessed. The coconut oil was quantitatively screened for its bioactive compounds using gas chromatography coupled to a mass spectrophotometer (GC-MS) and the resulting bioactive compounds were assessed for absorption, distribution, metabolism, excretion and toxicity (ADMET) properties using the SWISSADME tool. Compounds that met Lipinski's rule of five (ROF) were subjected to molecular docking against candidapepsin-2 using the Biovia (Discovery) docking tool. Results: The test isolates exhibited zones of inhibition ranging from 47–76 mm to the extract (100%) compared to 12–42 mm exhibited against fluconazole (400 mg) and 2–3 mm in plates containing only agar with MICs and MFCs ranging from 1.57–6.25% and 3.13 −12.5% against the extract compared to the control drug where MICs and MFCs of 6.25–12.5% and 12.5–25.0% were observed, respectively. The coconut extract exerted a concentration-dependent effect on the test isolates over time as higher extract concentrations decreased the optical density of cells to 0.001–0.06 at 72 h of incubation. Equal proportions of the extract + fluconazole exerted greater inhibitory potentials on the test isolates compared with those of low extract proportions. The synergistic-antagonistic antifungal assay showed enhanced sensitivity of the resistant isolates. Of the eleven (11) bioactive compounds quantitatively screened, only nine (9) met Lipinski's ROF and also returned favourable pharmacokinetics comparable to fluconazole. Docking scores for the bioactive compounds ranged from −2.0 to −3.0 kcal/mol. The most recurring amino acid residues following molecular docking were ILE and THR. Conclusion: The bioactive compounds showed desirable activities in-vitro, especially in synergy with fluconazole against the drug-resistant Candida species and in-silico and thus, require further studies to validate their potential use in the management of Candida -related infections. Nano-biocomposite hydrogel samples were produced using graphene oxide (GO) and agar and applied as adsorbents of organic components in water. The hydrogels were prepared by varying the wt% of Agar and GO. The samples were characterized, and batch adsorption experiments evaluated the effect of initial pH, equilibrium isotherms, and kinetics for the adsorption of the anionic dye Acid Orange 7 (AO) and the cationic dyes Nile Blue A (NB) and methylene blue (MB) in an aqueous medium. Overall, both hydrogel samples exhibited satisfactory results for removing NB and MB; however, there was no effective removal for the anionic dye AO. Adsorption equilibrium isotherms were obtained, and Freundlich, Langmuir, and Sips models were fitted to the experimental equilibrium data; moreover, kinetic data were adjusted to driving force models and particle mass balance. The maximum experimental adsorption capacities, 141.48 mg·g−1 (MB) and 284.69 mg·g−1 (NB), were obtained, on a dry basis, for the sample produced with 70 wt% of agar and 30 wt% of GO. Both hydrogels exhibited remarkable regenerative potential for NB and MB, with the adsorption capacity remaining constant, even after five adsorption/desorption cycles. [ABSTRACT FROM AUTHOR]

Biodegradable edible biopolymers derived from renewable agricultural sources offer a promising alternative to synthetic plastics. This study focused on preparing agar-carboxymethyl cellulose films containing glycerol (20-60%) and potassium sorbate (0-20%). Film properties such as thickness, water vapor permeability, water solubility, moisture absorption, water drop contact angle, and color characteristics were evaluated. The films were also tested for antibacterial and anti-mold properties against three bacterial species and two mold species. Results indicated that the composition of glycerol and potassium sorbate significantly influenced film solubility and moisture absorption (p< 0.05). The films exhibited high water drop contact angles, indicating low hydrophilicity. Potassium sorbate and glycerol notably affected the color parameters (L, a, b, and yellow index) of the films. Antimicrobial tests revealed effective antibacterial and antifungal properties, with the level of potassium sorbate showing a significant impact (p < 0.05) on these properties. In conclusion, utilizing cost-effective biopolymers and incorporating antimicrobial agents like potassium sorbate not only supports environmental sustainability but also enhances film properties and inhibits microbial growth.

Aim: Agar-based phantoms are popular in high intensity focused ultrasound (HIFU) studies, with magnetic resonance imaging (MRI) preferred for guidance since it provides temperature monitoring by proton resonance frequency (PRF) shift magnetic resonance (MR) thermometry. MR thermometry monitoring depends on several factors, thus, herein, the PRF coefficient of agar phantoms was estimated. Materials and Methods: Seven phantoms were developed with varied agar (2, 4, or 6% w/v) or constant agar (6% w/v) and varied silica concentrations (2, 4, 6, or 8% w/v) to assess the effect of the concentration on the PRF coefficient. Each phantom was sonicated using varied acoustical power for a 30 s duration in both a laboratory setting and inside a 3T MRI scanner. PRF coefficients were estimated through linear trends between phase shift acquired using gradient sequences and thermocouple-based temperatures changes. Results: Linear regression (R2 = 0.9707–0.9991) demonstrated a proportional dependency of phase shift with temperature change, resulting in PRF coefficients between −0.00336 ± 0.00029 and −0.00934 ± 0.00050 ppm/°C for the various phantom recipes. Weak negative linear correlations of the PRF coefficient were observed with increased agar. With silica concentrations, the negative linear correlation was strong. For all phantoms, calibrated PRF coefficients resulted in 1.01–3.01-fold higher temperature changes compared to the values calculated using a literature PRF coefficient. Conclusions: Phantoms developed with a 6% w/v agar concentration and doped with 0%–8% w/v silica best resemble tissue PRF coefficients and should be preferred in HIFU studies. The estimated PRF coefficients can result in enhanced MR thermometry monitoring and evaluation of HIFU protocols.

Bioplastik merupakan material kemasan berbasis biomassa atau bahan terbarukan yang dapat digunakan untuk menanggulangi sampah plastik. Agar dan kitosan diketahui dapat disintesis menjadi bioplastik. Namun, karakteristik mekanik campuran agar dan kitosan masih belum sesuai dengan ekspektasi yakni memiliki nilai elongasi yang rendah. Plasticizer sangat dibutuhkan untuk memperbaiki karakteristik bioplastik. Penggunaan sorbitol sebagai plasticizer dalam pembuatan bioplastik berbasis agar dan kitosan hingga saat ini belum dilaporkan. Tujuan penelitian ini adalah menentukan konsentrasi sorbitol terbaik terhadap peningkatan karakteristik bioplastik berbasis agar dan kitosan. Metode yang dilakukan adalah mencampurkan sorbitol dengan konsentasi 0%, 1%, 1,25%, dan 1,5% (b/v) pada larutan utama bioplastik yaitu larutan agar 1% dan larutan kitosan 2%. Karakteristik bioplastik diketahui dengan melakukan uji ketebalan, kuat tarik, elongasi, daya serap air, gugus fungsi dengan FTIR, dan uji degradasi. Hasil penelitian ini menunjukkan bahwa konsentrasi sorbitol mempengaruhi karakteristik bioplastik. Formulasi bioplastik terpilih yaitu perlakuan penambahan sorbitol 1,25% dengan nilai ketebalan, daya serap air, kuat tarik, dan elongasi masing-masing sebesar 0,096±0,004 mm, 18,80±0,94%, 18,83±2,93 MPa, 95,89±12,62%. Bioplastik dengan 1,25% sorbitol membutuhkan waktu untuk terdegradasi sempurna selama 20 hari. Simpulan penelitian ini adalah karakteristik mekanis dari bioplastik berbasis kitosan dan agar dapat ditingkatkan dengan penambahan sorbitol 1,25% Abstract Bioplastic is a packaging material based on biomass or renewable materials that can be used to address plastic waste. Agar and chitosan are known to be able to be synthesized into bioplastic. However, the mechanical characteristics of agar and chitosan mixture are still not in line with expectations, particularly having low elongation values. Plasticizers are highly needed to improve the characteristics of bioplastics. The use of sorbitol as a plasticizer in the production of agar and chitosan-based bioplastics has not been reported to date. The aim of this research is to determine the optimal concentration of sorbitol for improving the characteristics of agar and chitosan-based bioplastics. The method involved mixing sorbitol at concentrations of 0%, 1%, 1.25%, and 1.5% (w/v) in the main bioplastic solution, which consists of 1% agar solution and 2% chitosan solution. The characteristics of the bioplastic were determined by conducting tests on thickness, tensile strength, elongation, water absorption, functional groups using FTIR, and degradation tests. The results of this study showed that the concentration of sorbitol affects the characteristics of bioplastics. The selected bioplastic formulation was the treatment with the addition of 1.25% sorbitol, with thickness, water absorption, tensile strength, and elongation values of 0.096±0.004 mm, 18.80±0.94%, 18.83±2.93 MPa, and 95.89±12.62%, respectively. Bioplastic with 1.25% sorbitol required 20 days to degrade completely. In conclusion, the mechanical characteristics of chitosan and agar-based bioplastics can be improved by the addition of 1.25% sorbitol.

The purpose of this in vitro study was to develop calcium sulfate (CS)-based disks infused with an antimicrobial drug, which can be used as a post-surgical treatment modality for osteomyelitis. CS powder was embedded with 10% antibiotic, amoxicillin (AMX) or moxifloxacin (MFX), to form composite disks 11 mm in diameter that were tested for their degradation and antibiotic release profiles. For the disk degradation study portion, the single drug-loaded disks were placed in individual meshes, subsequently submerged in phosphate-buffered saline (PBS), and incubated at 37 °C. The disks were weighed once every seven days and analyzed via Fourier-transform infrared spectroscopy, X-ray diffraction, energy dispersive X-ray spectroscopy, and scanning electron microscopy. During the antibiotic release analysis, composite disks were placed in PBS solution, which was changed every 3 days, and analyzed for antibiotic activity and efficacy. The antibacterial effects of these sustained-release composites were tested by agar diffusion assay using Streptococcus mutans (S. mutans) UA 159 as an indicator strain. The degradation data showed significant increases in the degradation of all disks with the addition of antibiotics. Following PBS incubation, there were significant increases in the amount of phosphate and decreases in the amount of sulfate. The agar diffusion assay demonstrated that the released concentrations of the respective antibiotics from the disks were significantly higher than the minimum inhibitory concentration exhibited against S. mutans over a 2–3-week period. In conclusion, CS-antibiotic composite disks can potentially serve as a resorbable, osteoconductive, and antibacterial therapy in the treatment of bone defects and osteomyelitis.
