The WD40 domain (WDD) of ATG16L1 plays a pivotal role in non-canonical autophagy. This study examined the role of recently identified LAP-like non-canonical autophagy (LNCA) in acute pancreatitis. LNCA involves rapid single-membrane LC3 conjugation to endocytic vacuoles in pancreatic acinar cells. The rationale for this study was the previously observed presence of trypsin in the organelles undergoing LNCA; aberrant trypsin formation is an important factor in pancreatitis development. Here we report that the deletion of WDD (attained in ATG16L1[E230] mice) eliminated LNCA, aggravated caerulein-induced acute pancreatitis and suppressed the fast trypsin degradation observed in both a rapid caerulein-induced disease model and in caerulein-treated isolated pancreatic acinar cells. These experiments indicate that LNCA is a WDD-dependent mechanism and suggest that it plays not an activating but a protective role in acute pancreatitis. Objective Despite evidence-based guidelines stating that lipase alone should be used in the diagnosis of suspected acute pancreatitis, health care providers continue to order amylase or amylase and lipase together. The purpose of this study was to assess the utilization of appropriate laboratory testing related to the diagnosis of acute pancreatitis. Methods The study used a retrospective cross-sectional design. The timeframe was from January 1, 2020, to December 31, 2020. A retrospective chart review was used to collect data for the following: patient-provider encounter notes, patient demographics, provider demographics, differential and final diagnosis, and laboratory test results. Data analysis include stratification of categorical variables and calculation of cost savings. Results For the 12-month period, this study found 2567 (9.3%) of all amylase and lipase tests to be unnecessary. Amylase tests (1881; 73.2%) made up the most unnecessary tests followed by lipase tests (686; 26.7%). An analysis of test-ordering behavior by providers revealed that 81.5% of all unnecessary tests were ordered by MDs. Finally, this study estimated a total cost savings of $128,350 if all unnecessary tests were eliminated. The current investigation was conducted with the objective of identifying yeasts that possess the ability to secrete a range of hydrolytic enzymes. Yeast isolates were collected from various samples, and the morphological characteristics of both the colony and yeast cells were examined. The capacity for enzyme production was assessed in multiple isolates by inoculating them in different media and quantifying their activities in terms of pectinase, amylase, urease, lipase, β-galactosidase, and protease. Out of a total of 42 yeast isolates, 30 displayed enzymatic activity. It is plausible that the yeast isolates obtained in this study hold potential for the production of industrial enzymes. Molecular identification techniques were employed to identify two isolates that exhibited multi-enzyme production and the highest enzyme activities. It was determined that these isolates belonged to two genera of yeasts, namely Pichia and Hanseniaspora. Specifically, isolate Y 16 was identified as Pichia kudriavizvii OL621856, while isolate Y 26 was identified as Hanseniaspora guillermondii OL621857 strains. [ABSTRACT FROM AUTHOR]

This study presents a comprehensive investigation into the production of amylase, a crucial enzyme with wide-ranging industrial applications, using locally sourced substrates from Kachchh, Gujarat. The research employed the Bacillus licheniformis strain and substrates such as coconut, rice husk, wheat bran, paddy straw, and maize straw. The study found paddy straw to be the most promising substrate for amylase production. The research also systematically optimized various process parameters for amylase production in Solid-State Fermentation (SSF) using the One Variable at a Time (OVAT) method. These parameters included incubation period, temperature, inoculum level, additional carbon sources, starch concentrations, additional nitrogen sources, initial pH, different mineral salt ions, initial moisture level, and surfactants. The results showed that the optimal conditions for maximum amylase yield were an incubation period of 48 hours, an incubation temperature of 35°C, an inoculum level of 10%, starch as the additional carbon source, a starch concentration of 2.5%, yeast extract as the additional nitrogen source, an initial pH of 7, NaCl as the mineral salt, an initial moisture level of 75%, and Tween 80 as the surfactant. This research provides a reliable and sustainable approach to enzyme production, offering valuable insights for the optimization of the solid-state fermentation process for maximum amylase production. Urine amylase levels are usually used to diagnose acute pancreatitis. However, there are reported cases where urine amylase levels are slightly increased in individuals without pancreatitis. Herein, we report the case of a young lady who presented with acute abdominal pain for 3 days. Her urine amylase level was 1717 U/L upon admission, and her condition was initially treated as acute pancreatitis. Unfortunately, the patient demonstrated abdominal guarding after 24 h; thus, urgent computed tomography (CT) was performed. CT revealed the presence of a dilated small bowel. She underwent emergency laparotomy, wherein a gangrenous small bowel with no evidence of saponification at the lesser sac was noted. Due to the non-specific nature of hyperamylasaemia, an alternative diagnosis other than acute pancreatitis should be considered if the clinical symptoms are not suggestive of pancreatitis or the condition worsens despite conservative management. Amylases represent a versatile group of catalysts that are used for the saccharification of starch because they can hydrolyze the glycosidic bonds of starch molecules to release glucose, maltose, and short-chain oligosaccharides. The amylolytic complex of the thermophilic filamentous fungus Humicola brevis var. thermoidea (AmyHb) was produced, biochemically characterized, and compared with the commercial amylase Termamyl. In addition, the biotechnological application of AmyHb in starch saccharification was investigated. The highest production was achieved using a wheat bran medium at 50 °C for 5–6 days in solid-state fermentation (849.6 ± 18.2 U·g−1) without the addition of inducers. Optimum amylolytic activity occurred at pH 5.0 at 60 °C, and stability was maintained between pH 5.0 and 6.0, with thermal stability at 50–60 °C, especially in the presence of Ca2+. These results were superior to those found with Termamyl. Both enzymes were strongly inhibited by Hg2+, Cu2+, and Ag+; however, AmyHb displayed increased activity in the presence of Mn2+ and Na+. In addition, AmyHb showed greater tolerance to a wide range of ethanol concentrations. AmyHb appears to be a complex consisting of glucoamylase and α-amylase, based on its substrate specificity and TLC. The hydrolysis tests on cornstarch flour showed that the cocktail of AmyHb50% + Termamyl50% significantly increased the release of glucose and total reducing sugars (36.6%) when compared to the enzymes alone. AmyHb exhibited promising physicochemical properties and good performance with commercial amylase; therefore, this complex is a biotechnological alternative candidate for the bioprocessing of starch sources. Purpose: We investigated the relationship between bile amylase (AMY) levels and biliary epithelial changes in pancreaticobiliary maljunction (PBM), a congenital anomaly characterized by pancreaticobiliary reflux due to duct fusion outside the duodenal wall. Methods: We enrolled 43 children with congenital biliary dilatation (CBD) of Todani types Ia, Ic, and IVa who underwent surgery at the Hokkaido Medical Center for Child Health and Rehabilitation between November 2007 and June 2023. We defined total AMY exposure in bile as bile AMY levels multiplied by the patient's age (months), representing amount of estimated AMY exposure until surgery. We retrospectively investigated the relationships between bile AMY levels and clinicopathological findings. Results: All patients exhibited hyperplasia in the gallbladder and bile duct epithelium, with dysplasia observed in 13 cases, but no carcinoma. Exposure to bile AMY ≥ 662,400 IU/L × months was an independent risk factor for dysplasia. Conclusion: The amount of estimated AMY exposure in bile rather than AMY levels in the bile is an independent risk factor for dysplasia in the biliary mucosa. [ABSTRACT FROM AUTHOR]

Dicerothamnus rhinocerotis (L.f.) Koekemoer, also known as rhinoceros bush and previously called Elytropappus rhinocerotis (L.f.) Less., is from the Asteraceae plant family. The plant is traditionally used to treat indigestion, stomach ulcers, influenza, and diarrhea. This study was aimed at investigating the phytochemistry, anti-glucosidase, anti-amylase, and anti-tyrosinase effects of D. rhinocerotis as research in this area is limited. The air-dried plant materials were macerated in 80% methanol (MeOH) and fractionated between hexane, dichloromethane (DCM), ethyl acetate (EtOAc), and butanol (BuOH). Column chromatography on silica gel was employed for the isolation of the compounds. A total of six compounds (1–6) were isolated from the fractions viz. acacetin (1), 15-hydroxy-cis-clerodan-3-ene-18-oic-acid (2), acacetin-7-glucoside (3), pinitol (4), apigenin (5), and β-sitosterol-3-O-glycoside (6). Compounds 2–4 and 6 are reported for the first time from this plant. Among the different fractions, the BuOH and EtOAc fractions had strong tyrosinase inhibitory activities with IC50 values of 13.7 ± 1.71 and 11.6 ± 2.68 µg/mL, respectively, while among the isolated compounds, apigenin (5) had the strongest inhibitory activity, with an IC50 of 14.58 µM, which competes favorably with Kojic acid (17.26 µM). The anti-glucosidase assay showed good activity in three of the fractions and compound 5, while the anti-amylase assays did not show significant inhibition activity. [ABSTRACT FROM AUTHOR]

To better cope with unfavorable climatic conditions and perform optimal physiological functions, the silkworm Bombyx mori secretes some of its defensive enzymes in the hemolymph. To understand the extent of enzyme secretion in the hemolymph, we studied the profiles of defense enzymes in different silkworm breeds. We collected and analyzed the hemolymph of 5th instar larvae of 21 different bivoltine silkworm breeds with different characteristics for protein content and activities of enzymes such as catalase, peroxidase, polyphenol oxidase, amylase, and protease. The results revealed significant differences between the breeds in terms of protein and defensive enzyme activities. The activity of catalase, peroxidase, polyphenol oxidase, amylase, and protease varied between silkworm breeds, with an average activity of 8.791 U mg−1, 0.203 U mg−1, 0.42 U mg−1, 0.264 mg ml−1 min−1 mg−1 and 0.193 µg ml−1 min−1, respectively. To understand the genetic relationship between the silkworm breeds in terms of the activity of defensive enzymes, we constructed a dendrogram using Euclidean distance. The 21 silkworm breeds were grouped into five different clusters with different enzyme activities, revealing sufficient diversity between the silkworm breeds that can be used for parental selection in silkworm breeding programs. This study is highly informative in enhancing knowledge on the role of defensive enzymes in the development of new silkworm breeds that can adapt to adverse environmental conditions. Thermophilic actinomycetes are commonly found in extreme environments and can thrive and adapt to extreme conditions. These organisms exhibit substantial variation and garnered significant interest due to their remarkable enzymatic activities. This study evaluated the potential of Streptomyces griseorubens NBR14 and Nocardiopsis synnemataformans NBRM9 strains to produce thermo-stable amylase via submerged fermentation using wheat and bean straw. The Box-Behnken design was utilized to determine the optimum parameters for amylase biosynthesis. Subsequently, amylase underwent partial purification and characterization. Furthermore, the obtained hydrolysate was applied for ethanol fermentation using Saccharomyces cerevisiae. The optimal parameters for obtaining the highest amylase activity by NBR14 (7.72 U/mL) and NBRM9 (26.54 U/mL) strains were found to be 40 and 30 °C, pH values of 7, incubation time of 7 days, and substrate concentration (3 and 2 g/100 mL), respectively. The NBR14 and NBRM9 amylase were partially purified, resulting in specific activities of 251.15 and 144.84 U/mg, as well as purification factors of 3.91 and 2.69-fold, respectively. After partial purification, the amylase extracted from NBR14 and NBRM9 showed the highest activity level at pH values of 9 and 7 and temperatures of 50 and 60 °C, respectively. The findings also indicated that the maximum velocity (Vmax) for NBR14 and NBRM9 amylase were 57.80 and 59.88 U/mL, respectively, with Km constants of 1.39 and 1.479 mM. After 48 h, bioethanol was produced at concentrations of 5.95 mg/mL and 9.29 mg/mL from hydrolyzed wheat and bean straw, respectively, through fermentation with S. cerevisiae. The increasing need for new treatments for obesity and diabetes has led to the development of new drugs and food supplements that could reduce carbohydrate absorption. Many starch blockers, based on common bean proteinaceous inhibitors against α-amylase (α-AI), are already present on the market. The extraction and purification of α-amylase inhibitor from a promising common bean cultivar from Sardinia (Nieddone) is described, highlighting the unique value of the Nieddone cultivar, particularly for its inhibitory activity on digestive enzymes and its complete lack of a hemagglutination effect on human red blood cells. The purification of α-AI involved two chromatographic steps (IEC and SEC) and was essential for revealing certain properties of the inhibitor. The purified inhibitor has a tetrameric structure (α2β2) and a molecular weight of approximately 42 kDa, as determined by SEC and SDS-PAGE, confirming it as a lectin-like inhibitor. The identification of the α-AI sequence was obtained by bottom-up high-resolution mass spectrometry, which allowed us to identify a unique peptide from the α chain and six unique peptides from the β chains. α-AI exhibited an optimum temperature of around 40 °C and two pH optima at 5 and 6.5, respectively. Its remarkable stability at high temperatures was measured (approximately 25% of activity retained even after 5 h at 100 °C), whereas the raw extract lost its activity entirely after just 10 min at 90 °C. Thus, the purification process significantly enhances the thermal stability of α-AI. The demonstrated effectiveness of the purified α-AI against the α-amylase enzyme in pigs, humans and insects underscores the protein's potential for treating obesity and diabetes, as well as for managing insect pests. Starch is a promising polymer for creating novel microparticulate systems with superior biocompatibility and controlled drug delivery capabilities. In this study, we synthesized polyethylene glycol (PEG)-modified starch microparticles and encapsulated folic acid using a solvent-mediated acid-base precipitation method with magnetic stirring, which is a simple and effective method. To evaluate particle degradation, we simulated physiological conditions by employing an enzymatic degradation approach. Our results with FTIR and SEM confirmed the successful synthesis of starch–PEG microparticles encapsulating folic acid. The average size of starch microparticles encapsulating folic acid was 4.97 μm and increased to 6.01 μm upon modification with PEG. The microparticles were first exposed to amylase at pH 6.7 and pepsin at pH 1.5 at different incubation times at physiological temperature with shaking. Post-degradation analysis revealed changes in particle size and morphology, indicating effective enzymatic degradation. FTIR spectroscopy was used to assess the chemical composition before and after degradation. The initial FTIR spectra displayed characteristic peaks of starch, PEG, and folic acid, which showed decreased intensities after enzymatic degradation, suggesting alterations in chemical composition. These findings demonstrate the ongoing development of starch–PEG microparticles for controlled drug delivery and other biomedical applications and provide the basis for further exploration of PEG–starch as a versatile biomaterial for encapsulating bioactive compounds. Latent associations between low serum amylase and reduced plasma insulin levels and increased adiposity have been described previously in a small study of asymptomatic middle‐aged humans. In the present study, we sought to determine the nature of such changes during the longitudinal progression from metabolically normal to overt type 2 diabetes mellitus (T2DM) in nonhuman primates (NHPs), a disease that appears to be the same in both pathophysiology and underlying mechanisms as that which most commonly develops in middle‐aged adult humans. Amylase and lipase levels were characterized in 157 unrelated adult rhesus monkeys (Macaca mulatta); 38% developed T2DM while under study. In all monkeys, multivariable linear regression analysis revealed that amylase could be negatively predicted by % body fat (β −0.29; p = 0.002), age (β −0.27; p = 0.005), and HbA1c (β −0.18; p = 0.037). Amylase levels were positively predicted by lipase levels (β = 0.19; p = −0.024) in all NHPs included in the study. Amylase was significantly lower in NHPs with metabolic syndrome (p < 0.001), prediabetes (PreDM) (p < 0.001), and T2DM (p < 0.001) compared to metabolically normal adult NHPs. Lipase increased in NHPs with PreDM (p = 0.005) and T2DM (p = 0.04) compared to normal NHPs. This is the first longitudinal study of any species, including humans, to show the dynamics of amylase and lipase during the metabolic progression from normal to metabolic syndrome, to PreDM and then to overt T2DM. The extraordinary similarity between humans and monkeys in T2DM, in pancreatic pathophysiology and in metabolic functions give these findings high translational value. [ABSTRACT FROM AUTHOR]

The present study aims to isolate amylolytic bacteria and later the formation of a bacterial consortium for alphaamylase production. The potent amylolytic strains in a selected consortium were identified as B. subtilis and B. licheniformis on the basis of 16S rRNA sequencing. Initially cultural conditions for alpha amylase were optimized in Plackett–Burman design, followed by the central composite design. Analysis of Variance (ANOVA) of the optimization study indicated pH 7, 0.05 %CaCl2 and 24 h incubation time were the significant variables that mainly effect the alpha amylase production. After the process of statistical optimization, the enzyme was then partially purified by means of ammonium sulfate precipitation at a 60% saturation level, resulting in a 2-fold purification with 1.4 U/mg specific activity. Enzyme kinetics studies showed that the maximum velocity of the reaction (Vmax) and Michaelis constant (Km) were 47.4 mg/ml and 13.2 mM, respectively. Amylase enzyme is used in various industries due to its diverse applications. In this study, bacteria from soil sample were primarily screened on starch agar medium to identify amylase producer through the detection of prominent clear zone. Total of five soil samples namely bakery points (A-1), sugar cane juice point (A-2), Lichi chinesis garden soil (A-3), rice field (A-4) and sugar industrial waste (A-5) were used in this study. Among the 17 strains isolated from three samples A-1, A-2 and A-3 were found positive for amylase production. The strains were further screened on the production medium. The N-1 bacterial strain revealed higher enzyme activity (92.21 ± 17 IU/ml) compared to the other strain and was thus selected for further work. The strain was identified as Bacillus lichenoformis from the 16S rRNA analysis. Enzyme production was enhanced by optimizing various parameters by one factor at a time technique. The agro industrial waste rice polish was used as substrate. The optimum temperature of the enzyme was 35°C, pH 5.5 and 2% (w/v) of substrate concentration. Qualitative detection by using sodium dodecyl polyacrylamide gel electrophoresis showed that molecular weight of enzyme was 35 kDa. Impact of diamides on structure and functional activities of soil microflora and soil health has not been well studied. Effect of chlorantraniliprole and cyantraniliprole on the activity of dehydrogenase, acidic, alkaline phosphatase, amylase, invertase and urease enzymes in chilli planted soil was assessed. Two applications of chlorantraniliprole @ 30, 37.5 and 60 g a.i. ha−1 and cyantraniliprole @ 60, 75 and 120 g a.i. ha−1resulted in adverse effect. Dehydrogenase activity was reduced by 21.34 and 22.07 per cent on 7thday, in double dose chlorantraniliprole and cyantraniliprole respectively. Activity of both acidic and alkaline phosphatase enzyme reduced by 4.98 and1.06 per cent at 5th day after application of chlorantraniliprole and cyantraniliprole @30 and 60 g a.i. ha−1 respectively. On the other hand, amylase and invertase activity showed non-significant reduction and urease enzyme activity was slightly reduced (0.02–3.44%). The recommended doses of these two diamides had short term adverse effect on these enzymes, while higher doses led to 20 -30 per cent reduction in enzyme activity). As a rule, an experiment carried out at school or in undergraduate study courses is rather simple and not very informative. However, when the experiments are to be performed using modern methods, they are often abstract and difficult to understand. Here, we describe a quick and simple experiment, namely the enzymatic characterization of ptyalin (human salivary amylase) using a starch degradation assay. With the experimental setup presented here, enzyme parameters, such as pH optimum, temperature optimum, chloride dependence, and sensitivity to certain chemicals can be easily determined. This experiment can serve as a good model for enzyme characterization in general, as modern methods usually follow the same principle: determination of the activity of the enzyme under different conditions. As different alleles occur in humans, a random selection of test subjects will be quite different with regard to ptyalin activities. Therefore, when the students measure their own ptyalin activity, significant differences will emerge, and this will give them an idea of the genetic diversity in human populations. To investigate the germination physiological mechanism of Paris polyphylla seeds, different stages of the germination process were divided based on the seed embryo morphology and the changing rules in endogenous hormone content and related enzyme activities were analyzed at different stages. The results were as follows: (1) The germination process of the seeds could be divided into eight stages based on seed embryo morphology: non-germinating embryo stage (S1), heart-shaped embryo stage (S2), embryo swelling stage (S3), radicle not breaking through seed coat stage (S4), cotyledon petiole elongation and radicle breaking through seed coat stages (S5), lower hypocotyl breaking through seed coat stage (S6), upper hypocotyl elongation stage (S7), and radicle elongation stage (S8). (2) The α-amylase activity in the seeds was significantly higher than β-amylase activity at all germination stages. (3) Superoxide dismutase (SOD) activity was the highest at stage S5 and the lowest at stage S1; peroxidase (POD) and catalase (CAT) activities generally increased with seed germination progress; soluble protein content initially decreased and then increased with seed germination progress. (4) The contents of hormones such as indoleacetic acid (IAA), gibberellin (GA3), abscisic acid (ABA), and salicylic acid (SA) decreased overall, while 1-aminocyclopropane-1-carboxylic acid (ACC), jasmonic acid (JA), brassinolides (BRs) increased overall during seed germination. Cytokinins (CTKs) content showed no significant change. The ratios of IAA/ABA and GA3/ABA decreased initially and then increased, while CTKs/ABA continuously increased with seed germination progress. In conclusion, the germination process of the seeds can be subdivided into eight stages based on seed embryo morphology, with varying endogenous hormone content and related enzyme activities at different stages. The activities of α-amylase and POD may be related to radicle elongation, while GA3 may affect the embryo formation, and ABA may inhibit the growth and development of the embryo.

This research consisted of studying the ability of new strains of microorganisms isolated from water of the "La izvor" lake system to synthesize enzymes: amylase, catalase, cellulase, lipase and selecting those with significant enzymatic potential. Express tests were performed to determine the enzymatic capacity: amylase, catalase, cellulase and lipase of 25 strains of actinobacteria, 25 strains of bacteria, 8 strains of microalgae and cyanobacteria, 35 strains of fungi. When determining the enzymatic capacity, specific indicators were used for each enzyme: amylase - Lugol's solution, catalase - H2O2 (3%), cellulase – carboxymethyl cellulose and Congo red, lipase – Tween 80. As a result of this study, 3 strains of actinobacteria, 9 strains of bacteria, 4 strains of fungi, and 5 strains of microalgae and cyanobacteria with significant enzymatic potential as prospects for biotechnology were selected.

Abstract Satureja is an aromatic plant that is used for flavoring, perfume, and food manufacturing due to its pleasant essential oil. Modern medicine research revealed several biological activities of Satureja essential oil, including antifungal, antibacterial, antiviral, antioxidant, anticancer, and anti-inflammatory. However, the functional properties of Satureja fatty acid have not been explored. This study examined the fatty acid profile, lipid nutritional quality, antioxidant, anti-amylase, and anti-lipase capacities of Satureja. The efficiency of Satureja fatty acid on the anti-oxidative and anti-inflammatory parameters in LPS-induced macrophage through the Nrf2/NF-kB/NADH oxidase pathway was examined. The whole lipid extract was prepared with chloroform/methanol/water solution. Fatty acids methyl ester from whole lipid extract were prepared with methanol/sulfuric acid reagent. The fatty acid profile was analyzed using gas chromatography-mass spectrometry. Total antioxidant was determined by ABTS decolorization. Lipase and amylase activities were determined by monitoring the decomposition of p-nitrophenyl butyrate and starch. The macrophage cell line was grown in DMEM media in the presence of fatty acid. The hydrogen peroxide production in treated cells was monitored using the FOX reagent. NADH oxidase activity was measured by monitoring NADH breakdown. The expression of NOX, NF-kB, and NRF2, were tested in the treated cells by real-time PCR. The main components of the Satureja fatty acid were linolenic acid (24.67–37.32%), palmitic acid (10.65–20.29%), linoleic acid (8.31–13.39%), oleic acid (4.42–14.35%), stearic acid (2.76–8.77%) and palmitoleic acid (1.77–4.95%). Given the nutritional quality, omega-3 PUFA (23.58–37.32%), SFA (21.53–26.70%), omega-6 PUFA (10.86–16.14%), omega-9 MUFA (4.42–14.35%), and omega-7 MUFA (1.77–4.95%) comprise the majority of fatty acids. Satureja fatty acid has a promising unsaturation index (120.77–164.27), PUFA/MUFA (2.07–6.41), hypocholesterolemic index (2.44–3.47), health-promoting index (2.03–2.42), PUFA/SFA (1.37–1.94), nutritive value index (0.53–1.71), MUFA/SFA (0.30–0.80) omega-6/omega-3 (0.34–0.65), atherogenicity index (0.41–0.49), and thrombogenicity index (0.17–0.27). Satureja fatty acid displayed strong antioxidant capacity (with IC50 ranging from 354 to 428 µg/mL), anti-lipase capacity (with IC50 ranging from 354 to 428 µg/mL), and anti-amylase capacity (with IC50 ranging from 370 to 390 µg/mL). LPS induced the expression of NOX, NRF2, and NF-kB and the synthesis of hydrogen peroxide in macrophage cells. In LPS-stimulated macrophages, Satureja fatty acid reduced NOX expression, hydrogen peroxide, and NF-kB expression and increased NRF2 at 0.04 mg/mL. In conclusion, Satureja fatty acids have potent antioxidant, anti-amylase, anti-lipase, and anti-inflammatory activities. The mechanisms in lowering oxidative stress markers depended on down-regulating superoxide-producing enzymes at gene and protein levels. Satureja polyunsaturated omega-3 fatty acids could be recommended for healthy products combined with dietary therapy to treat obesity, diabetes, and oxidative stress.

Two experiments were conducted to evaluate the effects of a genetically modified corn hybrid with α-amylase expressed in the kernel (AMY) on fermentation profile, aerobic stability, nutrient composition, and starch disappearance of whole-plant corn silage (WPCS) and earlage. Both hybrids, AMY and an isogenic corn hybrid (ISO), were grown in 10 replicated plots (5 for WPCS and 5 for earlage). Samples of each plot were collected at harvest, homogenized, and divided into 5 subsamples which were randomly assigned to 5 storage lengths (0, 30, 60, 90, and 120 d). Both datasets (WPCS and earlage), were analyzed separately as a completely randomized block design in a factorial arrangement of treatments, with a model including the fixed effects of hybrid, storage length, and their interaction, and the random effect of block. Minor differences on fermentation profile were observed between AMY and ISO for WPCS and earlage. An interaction between hybrid and storage length was observed for DM losses in WPCS, where losses were similar at 30, 60 and 90 d, but lower for AMY compared with ISO at 120 d. No effect of hybrid was observed on yeast and mold counts for WPCS or earlage. The aerobic stability of WPCS was greater for AMY than ISO. For earlage, AMY had greater DM losses and aerobic stability than ISO. An interaction between hybrid and storage length was observed for ammonia-N in both WPCS and earlage, where ammonia-N was similar at 0 d but greater for AMY than ISO throughout later storage lengths. A similar interaction was observed for water-soluble carbohydrates (WSC) concentrations in WPCS, where ISO had greater WSC than AMY at 0 d but was similar throughout later storage lengths. However, AMY earlage had a greater WSC concentration throughout storage length, but a lesser magnitude after ensiling. Starch concentration was greater for AMY than ISO in WPCS and earlage. Greater starch disappearances at 0 h and 6 h were observed for ISO in WPCS and earlage. Minor effects on fermentation profile, microbial counts, aerobic stability and nutrient composition suggests that AMY can be ensiled for prolonged periods with no concerns for undesirable fermentation or nutrient losses. Negros Oriental in the Philippines provides a rich diversity of geothermal habitats. Despite the abundance, there are only a few published thermophile studies in the area. This study aimed to isolate thermophilic cyanobacteria from a hot spring in Negros Oriental and use morphological and molecular features for characterization and identification. Six filamentous, homocystous cyanobacteria designated as CV1, CV2, CV3, CV4, CV5, and CV6 were successfully isolated and cultured. Morphological characterization showed light green to blue-green sheathed trichomes that form an interwoven pattern similar to the recently identified cyanobacterium, Alkalinema. Partial 16S rRNA gene sequences revealed 98.58% identity of the strains with Alkalinema pantanalense DCHY213, and phylogenetic analysis placed them under the family Leptolyngbyaceae -- hence forming a separate cluster from other A. pantanalense strains. The differences in ecotype and morphotype, as well as the distinct clustering in the phylogenetic tree of the Negros Oriental strains, imply the possibility of belonging to a different species. Amylase activity was detected in all strains except CV4, hence implying their potential for bioactive metabolite studies. These results highlight the importance of untapped extreme environments in the country for the discovery of novel or recently identified microorganisms that have taxonomical, ecological, and biotechnological applications. Objectives: We aimed to clarify the clinical utility of measuring serum pancreatic enzymes after endoscopic retrograde cholangiopancreatography (ERCP) for the purpose of predicting post‐ERCP pancreatitis (PEP) by a meta‐analysis of diagnostic test accuracy studies. Methods: Studies on the prediction accuracy of PEP by serum amylase or lipase measured at 2, 3, and 4 h after ERCP were collected. A literature search was performed in PubMed and the Cochrane Library database for studies published between January 1980 and March 2023. The quality of individual studies was evaluated using the Quality Assessment of Diagnostic Accuracy Studies‐2. Data were analyzed using Meta‐DiSc 2.0 software. Results: We searched the databases and identified 20 observational studies (12,313 participants). PEPs were defined according to criteria by Cotton or modified Cotton, revised Atlanta criteria, or the Japanese criteria. Meta‐analysis of eight studies (4389 participants) showed a pooled sensitivity of 71.1% (95% confidence interval [CI] 56.1–82.5) and pooled specificity of 91.2% (95% CI 85.9–94.6) for the serum amylase cut‐off value at 3 times the upper limit of normal (ULN). Another meta‐analysis of five studies (1970 participants) showed a pooled sensitivity of 85.8% (95% CI 61.9–95.7) and pooled specificity of 85.3% (95% CI 81.9–88.1) for the serum lipase cut‐off value at 3 times ULN. Conclusion: Despite a high risk of bias due to various reference standards, this updated meta‐analysis and the utility assessment by a decision tree showed the utility of serum amylase or lipase levels more than 3 times ULN measured 2–4 h after ERCP for predicting PEP. Leptospirosis is a zoonotic disease. We present a case of acute pancreatitis associated with leptospirosis. An 88-year-old woman was admitted to the hospital with high fever and severe myalgia of the lower extremities. Based on the clinical presentation, hepatic dysfunction with a mild increase in bilirubin, renal dysfunction, and life history, the possibility of leptospirosis was considered. Plain computed tomography of the trunk on admission revealed no special findings. Appropriate antimicrobial therapy was administered at an early stage. After treatment initiation, the clinical symptoms and blood test abnormalities began to improve, and the patient appeared to be doing well. Although no abdominal or back pain was consistently noted during hospitalization, the serum amylase level increased over time; therefore, the patient underwent another computed tomography scan on the ninth day. Acute pancreatitis, which was absent upon admission, was noted. Appropriate treatment for pancreatitis was administered, and the patient was discharged. A subsequent serum antibody test confirmed the diagnosis of leptospirosis. Herein, we also summarized previous cases of acute pancreatitis associated with leptospirosis. The time of onset for pancreatitis was inconsistent, and there were a few cases of pancreatitis without abdominal or back pain. In contrast, serum amylase or lipase levels were elevated in all patients, which could be an important trigger for suspected complications of pancreatitis. When leptospirosis is suspected, complications of pancreatitis should always be considered, even in the absence of apparent abdominal pain. This study aimed to examine the effect of acute exogenous melatonin administration on salivary cortisol and alpha‐amylase (sCort and sAA) as representatives of the HPA axis and the sympathetic nervous system, respectively. A single‐dose prolonged‐release melatonin (2 mg) or a placebo tablet was given to healthy volunteers (n = 64) at 20:00 h in a crossover design. The saliva was collected at six time points (20:00, 21:00, awakening, 30 min after awakening, 10:00, and 12:00 h) and was measured for sCort, sAA, and salivary melatonin (sMT) levels. Pulse rates and sleep parameters were also collected. Melatonin was effective in improving sleep onset latency by 7:04 min (p =.037) and increasing total sleep time by 24 min (p =.006). Participants with poor baseline sleep quality responded more strongly to melatonin than participants with normal baseline sleep quality as they reported more satisfaction in having adequate sleep (p =.017). Melatonin administration resulted in higher sCort levels at awakening time point (p =.023) and a tendency of lower sAA levels but these were not significant. Melatonin ingestion at 20:00 h resulted in a marked increase in sMT levels at 21:00 h and remained higher than baseline up to at least 10:00 h (p <.001). Melatonin increases sCort levels at certain time point with a tendency to lower sAA levels. These opposing effects of melatonin suggested a complex interplay between melatonin and these biomarkers. To investigate the germination physiological mechanism of Paris polyphylla seeds, different stages of the germination process were divided based on the seed embryo morphology and the changing rules in endogenous hormone content and related enzyme activities were analyzed at different stages. This study aimed to explore the fermentative performance of nine lactic acid bacterial strains with probiotic potential during sorghum fermentation. The strain's attributes including proliferation counts, pH levels, production of organic acid antibacterial activity, and their ability to break down starch were evaluated during the fermentation period in the presence and absence of glucose as a carbon source. In addition, the inhibitory activity of these potential probiotic strains against pathogenic bacteria (Salmonella typhimurium, Escherichia coli, and Staphylococcus aureus) was examined through a co-culturing technique. The results demonstrated that all 4 Lactobacillus strains exhibited robust growth in both glucose and glucose-free fermentation experiments. Glucose supplementation significantly enhanced lactic acid yield which ranged from 0.19 to 0.44% compared to fermentation without glucose which ranged from 0.04 to 0.29%. The selected Lactobacillus strains effectively lowered the media pH below 4.0 after 24 h, producing substantial lactic acid. Notably, in the absence of glucose, only Lb. helveticus D7 and Lb. amylolyticus D12 achieved pH levels below 4 after 8 h, producing the highest lactic acid amounts of 0.27 and 0.29% after 24 h, respectively. Amylase activity was detected on two strains, D7 and D12. Furthermore, most of the tested Lactobacillus strains demonstrated complete inhibition (6 log to 0 Log CFU/mL) of pathogen growth after 24 h of co-culturing, suggesting their potential for enhancing the safety quality of sorghum-based fermented products. Symplocos paniculata are reported to exhibit seed dormancy, which impedes its cultivation and widespread adoption. In this study, a comprehensive method was established to overcome seed dormancy by subjecting seeds to scarification in 98% H2SO4 for 10 min, followed by 1000 mg·L−1 GA3 soaking for 48 h and stratification at 4 °C for 100 days. The seed germination percentage has increased significantly, to a peak of 42.67%, though the seeds could not germinate timely by NaOH scarification. Additionally, the dynamic changes of key stored substances (proteins, soluble sugars, starches, and fats), associated enzyme activities (amylases, peroxidase, and catalase), and endogenous hormones (abscisic acid, gibberellic acid, and indole-3-acetic acid) in seeds were investigated. The results demonstrated a continuous degradation of starch and fat in S. paniculata seeds, while the levels of protein and soluble sugar exhibited fluctuations, which probably facilitated seed dormancy breaking through energy supply and transformation. The enzymatic activities underwent rapid changes, accompanied by a gradual decrease in ABA content within the seeds with increasing stratification time. Notably, GA3, GA3/ABA, and (GA3 + IAA)/ABA showed significant increases, indicating their positive regulatory roles in seed germination. Species belonging to the genus Bacillus produce many advantageous extracellular enzymes that have tremendous applications on a commercial scale for the textile, detergent, feed, food, and beverage industries. This study aimed to isolate potent thermo-tolerant amylolytic and cellulolytic bacterium from the local environment. Using the Box–Behnken design of response surface methodology, we further optimized the amylase and cellulase activity. The isolate was identified by 16S rRNA gene sequencing as Bacillus subtilis QY4. This study utilized potato peel waste (PPW) as the biomaterial, which is excessively being dumped in an open environment. Nutritional status of the dried PPW was determined by proximate analysis. All experimental runs were carried out in 250 mL Erlenmeyer flasks containing acid treated PPW as a substrate by the thermos-tolerant Bacillus subtilis QY4 incubated at 37 °C for 72 h of submerged fermentation. Results revealed that the dilute H2SO4 assisted autoclaved treatment favored more amylase production (0.601 IU/mL/min) compared to the acid treatment whereas high cellulase production (1.269 IU/mL/min) was observed in the dilute acid treatment and was found to be very effective compared to the acid assisted autoclaved treatment. The p-value, F-value, and coefficient of determination proved the significance of the model. These results suggest that PPW could be sustainably used to produce enzymes, which offer tremendous applications in various industrial arrays, particularly in biofuel production. The objective: To delay the digestion of carbohydrates by inhibiting the activity of carbohydrate digestive enzymes, thereby preventing and controlling the effects of diabetes has become a research hotspot across multiple fields. j8 Conglycinin is the most abundant functional protein in soy protein, but it is unknown whether the peptides produced after gastrointestinal digestion possess the activity to inhibit carbohydrate digestive enzymes. Method: Conglycinin was subjected to simulated gastrointestinal digestion, and glucosidase inhibitory peptides and amylase inhibitory peptides were selected through virtual screening and ADMET prediction, and the inhibitory effects of the peptides on " --glucosi-dase and " --amylase were tested. Results: ! --Conglycinin was virtually digested into 95 small molecular peptides by pepsin, trypsin, and chymotrypsin; eight "--glucosidase inhibitory peptides and "--amylase inhibitory peptides were selected; it was found that hydrogen bonds and electrostatic interactions play an important role in the binding of peptides with glucosidase and amylase; in vitro verification found that the tetrapeptide EASY has a strong inhibitory effect on glucosidase, with an IC0 value of 208.6 g/mL, and no obvious inhibitory effect on amylase was observed. Conclusion: The glucosidase inhibitory peptide EASY possesses the activity to inhibit carbohydrate digestive enzymes and may become a potential inhibitor for controlling diabetes. Objectives: The contribution of pancreatic secretions in iron metabolism has been elucidated, but the clinical outcomes of iron deficiency on pancreatic function are debatable. This study aimed to investigate the modulation of euglycemic endocrine and exocrine pancreatic excretions in response to variations in iron availability. Subjects and Methods: Serum levels of insulin, glucagon, insulin-to-glucagon ratio (IGR), and amylase were determined in 170 adult subjects with variable levels of serum iron. Results: Control (n = 46) and iron-deficient (n = 124) subjects had significant differences (p < 0.001) in their average levels of insulin (68.7 ± 0.5 vs. 100.0 ± 2.0 pmol/dL), glucagon (17.9 ± 0.6 vs. 10.8 ± 0.8 pmol/dL), IGR (4.0 ± 0.1 vs. 19.5 ± 2.1), and amylase (29.7 ± 0.9 vs. 17.5 ± 0.2). The upregulation of serum insulin levels increases proportionally and gradually to the extent of iron deficiency as compared to an abrupt downregulation of serum levels of glucagon and amylase. A significant association was observed between serum iron and IGR (r = −0.645, p < 0.001) and amylase levels (r = 0.653, p < 0.001). The receiver operating characteristic curve analysis defines an excellent predictivity of the reduced serum iron level to discriminate subjects with upregulated IGR and amylase levels with area under curves of 0.938 and 0.905, respectively. Conclusion: Iron deficiency is associated with an adaptive modulation of euglycemic endocrine and exocrine secretions that is consistent with a status of insulin resistance. Highlights of the Study: Iron deficiency (ID) exacerbates serum insulin levels proportional to the extent of deficiency. ID decreases the serum levels of glucagon and amylase with abrupt downregulation patterns. ID provides excellent predictivity for dysregulation in serum levels of insulin, glucagon, and amylase. This current article was dedicated to the determination of the composition of phenolic compounds in extracts of four species of the genus Filipendula in order to establish a connection between the composition of polyphenols and biological effects. A chemical analysis revealed that the composition of the extracts studied depended both on the plant species and its part (leaf or flower) and on the extractant used. All four species of Filipendula were rich sources of phenolic compounds and contained hydrolyzable tannins, condensed tannins, phenolic acids and their derivatives, and flavonoids. The activities included data on those that are most important for creating functional foods with Filipendula plant components: the influence on blood coagulation measured by prothrombin and activated partial thromboplastin time, and on the activity of the digestive enzymes (pancreatic amylase and lipase). It was established that plant species, their parts, and extraction methods contribute meaningfully to biological activity. The most prominent result is as follows: the plant organ determines the selective inhibition of either amylase or lipase; thus, the anticoagulant activities of F. camtschatica and F. stepposa hold promise for health-promoting food formulations associated with general metabolic disorders. The food enzyme α‐amylase (4‐α‐d‐glucan glucanohydrolase; EC 3.2.1.1) is produced with the non‐genetically modified microorganism Bacillus licheniformis strain AE‐TA by Amano Enzyme Inc. The food enzyme is intended to be used in eight food manufacturing processes. Since residual amounts of food enzyme‐total organic solids (TOS) are removed in two food manufacturing processes, dietary exposure was calculated only for the remaining six processes. It was estimated to be up to 0.056 mg TOS/kg body weight per day in European populations. The production strain of the food enzyme fulfils the requirements for the qualified presumption of safety approach to safety assessment. Consequently, in the absence of other concerns, the Panel considered that toxicological studies were not needed for the safety assessment of this food enzyme. A search for the similarity of the amino acid sequence of the food enzyme to known allergens was made and two matches with respiratory allergens were found. The Panel considered that the risk of allergic reactions upon dietary exposure to this food enzyme cannot be excluded (except for the production of distilled alcohol), but the likelihood is low. Betel (Piper betle L.), a vine growing primarily in Southeast Asian countries, are widely used in folk medicine to prevent and treat indigestion, cough, bronchitis and asthma. The aims of the study were to determine volatile constituents and pigments as well as to compare phenolic composition, antioxidant activity and enzyme inhibitory effects of betel leaf extracts obtained with different organic solvents. The results showed that the sample was composed of various major volatile organic compounds, of which eugenol, γ-muurolene and δ-cadinene accounted for more than 64%. It was also a rich source of pigments, including lutein (400.68 μg/g) and zeaxanthin (101.68 μg/g). The ethyl acetate extract had the highest total phenolic content (71.72 ± 0.54 mg GAE/g) despite containing low levels of the phenolic compounds examined in the study. The acetonic extract generally showed higher antioxidant activity compared to the others. The methanolic extract exhibited the strongest inhibitory effects on α-amylase (597.23 ± 59.06 μg/mL) and xanthine oxidase (43.07 ± 1.69 μg/mL). Enzymatic reactions can be modulated by the incorporation of organic solvents, leading to alterations in enzyme stability, activity, and reaction rates. These solvents create a favorable microenvironment that enables hydrophobic reactions, facilities enzyme-substrate complex formation, and reduces undesirable water-dependent side reactions. However, it is crucial to understand the impact of organic solvents on enzymatic activity, as they can also induce enzyme inactivation. In this study, the enzymatic performance of Aspergillus oryzae α-amylase (Taka-amylase) in various organic solvents both experimentally and computationally was investigated. The results demonstrated that ethanol and ether sustain Taka-amylase activity up to 20% to 25% of the organic solvents, with ether providing twice the stability of ethanol. Molecular dynamics simulations further revealed that Taka-amylase has a more stable structure in ether and ethanol relative to other organic solvents. In addition, the analysis showed that the loop located near the active site in the AB-domain is a vulnerable site for enzyme destabilization when exposed to organic solvents. The ability of Taka-amylase to preserve the secondary loop structure in ether and ethanol contributed to the enzyme's activity. In addition, the solvent accessibility surface area of Taka-amylase is distributed throughout all enzyme structures, thereby contributing to the instability of Taka-amylase in the presence of most organic solvents. Introduction: Serum amylase and lipase elevations are classically related to pancreatic diseases. Hyperamylasemia is noticed to various common non-pancreatic clinical conditions, however hyperlipasemia is considered the specific marker of pancreatic diseases. Methodology and Results: This paper set forth 3 cases of Diabetic Keto Acidosis that presented with abdominal pain which are initially misapprehended to be Acute Pancreatitis due to markedly raised serum amylase and lipase levels. Summary: This work presents the impact of Satureja essential oil (SEO) on the mRNA generation of NFR2, NF‐kB, and NOX (NADH oxidase) in the macrophage stimulated with hyperglycaemia. Furthermore, SEO incorporated into gelatin particles and physico‐chemical properties were investigated. The major variation between different constituents of SEO was found in carvacrol (35.9%–87.6%) and γ‐terpinene (0.43%–33.9%). Different SEO with different constituents exhibited comparable antioxidant capacity (IC50 = 0.170–0.215 mg mL−1), anti‐linoleic acid oxidation (IC50 = 0.137–0.195 mg mL−1), anti‐lecithin oxidation (IC50 = 0.126–0.134 mg mL−1), anti‐starch oxidation (IC50 = 0.155–0.199 mg mL−1), anti‐protein oxidation (IC50 = 0.166–0.211 mg mL−1), anti‐amylase capacity (IC50 = 0.202–0.230 mg mL−1), and anti‐lipase capacity (IC50 = 0.24–0.260 mg mL−1). Hyperglycaemia stimulates NOX, NFR2, NF‐kB, and hydrogen peroxide in macrophages. SEO (at 0.03 and 0.06 mg mL−1) decreased NOX and NF‐kB activity and hydrogen peroxide and increased NFR2 in hyperglycaemia‐stimulated macrophages. The physio‐chemical parameters of gelatin dispersion were electrical conductivity (1095 microsiemens cm−1), osmolarity (176 milliosmol kg−1), surface tension (42 mN m−1), zeta potential (−40 mV), particle size (123 nm), particle size distribution (0.18), and viscosity (3.47 mPa.s). The techno‐functional parameters of gelatin particles were water content (10.4%), water solubility (83%), water swelling (138%), hygroscopicity (35%), hydrophobicity (6.8 μg g−1), emulsification activity (62%), emulsification stability (52%), foam activity (49%), foam stability (42%), water‐holding capacity (4.5 g g−1), and oil‐holding capacity (2.4 g g−1). SEO causes minor changes in the techno‐functional and physico‐chemical properties of gelatin. The current investigation introduces gelatin/SEO as a possible functional ingredient for bioactive food products for the treatment of diabetes, oxidative stress, and inflammation. The studies of bacterial communities from extreme econiches are presently aimed mainly at analyzing the biodiversity of microorganisms using molecular biology methods. Cultivated bacteria from karst caves represent a unique group of microorganisms, the biochemical potential of which has been poorly studied. In the present work, bacteria from biofilms on the walls of the Kapova Cave (Shulgan-Tash Nature Reserve, Bashkortostan) were isolated and characterized in order to assess the ability of identified isolates to produce extracellular hydrolytic enzymes. Most of the isolates (89%) were members of the phylum Proteobacteria, with the remaining ones belonging to the phyla Actinobacteria, Firmicutes, and Bacteroidetes, which accounted for 5, 4, and 2% of the isolates, respectively. Strains with high levels of secreted protease, RNase, and amylase activity were identified as Stenotropomonas rhizophila, Lysinibacillus fusiformis, and Pseudomonas stutzeri, respectively. Ramie leaves are rich in antioxidant polyphenols and amylase enzyme, which could be a good source for food applications. This study investigated the impact of combined ultrasound-enzyme treatment (U-EAT), including ultrasound intensity (10-30%), content of Viscozyme L. enzyme addition (0.5-1%) and extraction time (5-15 min) on polyphenol recovery, antioxidant activity, and amylase activity. The result revealed that U-EAT could enhance the efficiency of extraction, with a significantly higher yield of bioactive compounds and antioxidant capacity, and amylase enzyme activity compared to an untreated sample. Multiple regression analysis was applied to predict the efficacy of extraction through the values of total phenolic content, antioxidant activity and enzyme activity. The established models shown high accuracy in predicting the effect of the extracted conditions on the yield of the extraction of antioxidant and enzyme amylase. It was found that the maximum yield of extraction could be obtained when the operation conditions were ultrasound intensity of 26.5%, enzyme concentration of 0.87% and extraction time of 10.88 min. Under these optimal conditions, the highest values were found (TPC of 99.047 mgGAE/100 g, antioxidant activity of 65.02% and amylase activity of 45.36 U/mL). In summary, the findings indicate that the utilization of ultrasound combined with the enzymatic extraction process of polyphenol and amylase enzyme from ramie leaves holds potential for enhancing both the extraction yield and bioactivity of the extract. Soil is a vital ecosystem for various life forms, including bacteria. Bacteria are the main group of soil microbes, mostly because of their ability to produce various extracellular enzymes. Amylases are the enzymes that break down starch, which are essential in biotechnology industries such as food, fermentation, textiles, and paper manufacturing. The demand for amylase is often high since many enterprises do not have access to local supplies of the enzyme. The objective of the current investigation is to isolate and identify bacterial isolates that can produce the enzyme amylase from soil samples. Soil samples were collected from the unexplored Biligiriranga hills (11.9988° N, 77.1398° E) in south-western Karnataka Bacterial isolates were isolated from soil samples and were primarily screened on starch agar medium to determine the amylase production. Three out of fifteen bacterial isolates that produced amylase were selected based on the highest clear zone around the bacterial colony. These three bacterial isolates were identified both microscopically and biochemically. Gram staining demonstrated that the isolates are rod-shaped and gram-positive organisms. Biochemical studies indicated that the isolates were positive for the catalase and starch hydrolysis tests, whereas negative for the potassium hydroxide and methyl red tests. Besides, 16S rRNA gene amplification and sequencing confirmed the isolate as Bacillus subtilis. The 16S rRNA sequence of B. subtilis strain B has been deposited in the NCBI GenBank. The purpose of the current study was to evaluate the functional activity and storage viability (at 4 °C and 35 °C) of an immobilized as well as lyophilized multienzyme, viz., pectinase, cellulase, and amylase (PCA) that was produced by Bacillus subtilis NG105 under solid state fermentation (SSF) at 35 ℃ for 10 days using mosambi peel as a substrate. After SSF, the culture media was divided into two aliquots. From the first aliquot, the produced ME was extracted, precipitated, and further immobilized on calcium alginate beads (MEICA). In order to immobilize on mosambi peel matrix, the second aliquot was mixed with acetone and subsequently lyophilized (MELMP). Thus, ready MEICA and MELMP extracted 87.5 and 91.5% juice from mango pulp, respectively. In the reusability study, after 5 cycles, MEICA exhibited 23.8%, 24.4%, and 36.5% PCA activity, respectively. The PCA activity of MEICA and MELMP was examined after 60 days of storage at 4 ℃. The result revealed that the PCA for MEICA declined from 100 to 66%, 58.2%, and 64.5%, respectively, while for MELMP, it dropped from 100 to 84.2%, 82.1%, and 69.7%, respectively. Further, after 60 days of storage, the reduction of total protein content (TPC) in free multienzyme (FME), MEICA, and MELMP was 92.2%, 91.5%, and 36.3% observed, respectively. In the localization study, the maximum levels of multienzyme activity were found in cell exudates. This study demonstrated that immobilizing of multienzyme through lyophilization on waste substrates like mosambi peel boosted its stability and shelf-life along with greatly reducing the cost of products.
