The aim of the study was to develop a molecular diagnosis protocol of monocytic ehrlichiosis and granulocytic anaplasmosis in dogs by conventional polymerase chain reaction (PCR) and to compare the results from PCR and a rapid serological test. One hundred and six blood samples from dogs were tested by the rapid serological test SNAP 4Dx Plus (IDEXX Laboratories, Westbrook, ME) and by conventional PCR. Sixty-two of them (58.49%) were positive for antibodies to Ehrlichia canis/Ehrlichia ewingii and 14 (13.21%) for antibodies to Anaplasma phagocytophilum/Anaplasma platys. In 24 (22.64%) blood samples, antibodies against both pathogens were simultaneously de-tected. Six (5.66%) samples were seronegative. Fourty-six of the 106 tested samples (43.4%) were positive for a 345 bp segment of the ribosomal gene of family Anaplasmataceae. In 28 of them the presence of a 444 bp fragment of the ankA gene of A. phagocytophilum was detected, and in 26: a 409 bp fragment of the gene of E. canis. Nine samples were simultaneously positive for genetic se-quences of E. canis and A. phagocytophilum. The target DNA fragments specific for the two studied pathogens were not detected in one of the Anaplasmataceae-positive samples. In the remaining 60 cases (56.6%), the presence of a 345 bp segment of the ribosomal gene was not detected. In the present study, the DNA of E. canis and of A. phagocytophilum was detected for the first time in Bulgarian dogs by the conventional PCR.

Abstract Background Anaplasma phagocytophilum is a Gram-negative obligate intracellular bacterium that replicates in neutrophil granulocytes. It is transmitted by ticks of the Ixodes ricinus complex and causes febrile illness called granulocytic anaplasmosis primarily in humans, horses, dogs, sheep, cattle and goats. In comparison, clinically apparent disease has been described rarely in cats especially compared to dogs and horses. It is currently unknown whether cats are less susceptible to A. phagocytophilum or whether granulocytic anaplasmosis might be underdiagnosed in cats. Methods To address this question, we examined clinical signs and laboratory findings in seven A. phagocytophilum infected cats from Germany and Switzerland. We then genetically characterized feline A. phagocytophilum strains and compared them to those from other hosts showing clinically apparent disease. For this purpose, ankA-based, groEL-based and multilocus sequence typing (MLST) were applied. Furthermore, the concordance between these typing methods was assessed. Results Fever, lethargy and anorexia were the most common clinical signs in cats suffering from granulocytic anaplasmosis. The most frequent laboratory finding was thrombocytopenia. All three typing methods consistently indicated that the A. phagocytophilum strains found infecting cats are the same as those that cause disease in humans, dogs and horses. In general, the three typing methods applied exhibited high concordance. Conclusions The genetic characterization of the feline A. phagocytophilum strains indicates that strain divergence is not the explanation for the fact that granulocytic anaplasmosis is much less frequently diagnosed in cats than in dogs and horses. Otherwise, it may be possible that cats are less susceptible to the same strains than dogs and horse are. However, due to the unspecific clinical signs, it should be considered that granulocytic anaplasmosis may be under-diagnosed in cats. Graphical Abstract

Simple Summary: Bacteria continuously circulate in the natural environment and frequently cross into the human environment. While some of these bacteria can have a positive effect on human health, they are often accompanied by pathogenic forms. The aim of the present study was to more closely examine the zoonotic, tick-borne bacterium Anaplasma phagocytophilum, the causative agent of human and animal anaplasmosis. The study examined tissue samples from roe deer and red deer. Anaplasma phagocytophilum samples were genotyped based on DNA amplification, sequencing and phylogenetic analysis. Our results showed that detected bacteria from red deer belong to ecotype I and cluster I, which is why those bacteria are potentially pathogenic to humans. Detected bacteria from roe deer were mainly in ecotype II and clusters II and III, which means those bacteria are not pathogenic for humans. Background: The Gram-negative bacterium Anaplasma phagocytophilum is an intracellular pathogen and an etiological agent of human and animal anaplasmosis. Its natural reservoir comprises free-ranging ungulates, including roe deer (Capreolus capreolus) and red deer (Cervus elaphus). These two species of deer also constitute the largest group of game animals in Poland. The aim of the study was to genotype and perform a phylogenetic analysis of A. phagocytophilum strains from roe deer and red deer. Methods: Samples were subjected to PCR amplification, sequencing, and phylogenetic analysis of strain-specific genetic markers (groEL, ankA). Results: Five haplotypes of the groEL gene from A. phagocytophilum and seven haplotypes of ankA were obtained. The phylogenetic analysis classified the groEL into ecotypes I and II. Sequences of the ankA gene were classified into clusters I, II, and III. Conclusions: Strains of A. phagocytophilum from red deer were in the same ecotype and cluster as strains isolated from humans. Strains of A. phagocytophilum from roe deer represented ecotypes (I, II) and clusters (II, III) that were different from those isolated from red deer, and these strains did not show similarity to bacteria from humans. However, roe deer can harbor nonspecific strains of A. phagocytophilum more characteristic to red deer. It appears that the genetic variants from red deer can be pathogenic to humans, but the significance of the variants from roe deer requires more study. [ABSTRACT FROM AUTHOR]

ABSTRACT The tick-borne obligatory intracellular bacterium Anaplasma phagocytophilum infects granulocytes of humans as well as domesticated and wild animals, causing a febrile disease collectively called granulocytic anaplasmosis. The host species specificity and zoonotic potential of A. phagocytophilum strains remain unclear. In the USA, human granulocytic anaplasmosis (HGA) is primarily reported in Wisconsin and Minnesota, the northeastern coastal states, and northern California. In horses, A. phagocytophilum causes equine granulocytic anaplasmosis (EGA). Although EGA has been reported in the same US regions as HGA, horses with a definitive diagnosis of EGA have not been reported in Ohio, where the incidence of HGA is low. In this study, samples from archived blood smears or whole blood from three horses from three different counties in Ohio that presented in 2018, 2020, and 2021 with clinical signs and laboratory findings consistent with EGA were evaluated by PCR and sequencing. Sequences of ankA (encoding ankyrin A) of the three A. phagocytophilum strains were identical and related to strains isolated from humans in the northeastern USA yet distinct from those in California or Europe based on phylogenetic analysis. Each Ohio strain contains unique A. phagocytophilum p44 genes encoding the immunodominant major outer membrane proteins, underscoring the antigenic diversity of A. phagocytophilum strains that cause EGA in Ohio. From 2018 to 2023, 14 additional cases of EGA were identified by serodiagnosis in Ohio and nearby locations in Pennsylvania, pointing to a heretofore unrecognized prevalence of EGA and the risk of transmission of anaplasmosis to humans and animals in this region. IMPORTANCE The tick-borne obligatory intracellular bacterium Anaplasma phagocytophilum infects humans as well as domesticated and wild animals, causing a febrile disease collectively called granulocytic anaplasmosis. The epidemiology and the host species specificity and zoonotic potential of A. phagocytophilum strains remain unclear. In this study, ankA (encoding ankyrin A) and p44 gene sequences of A. phagocytophilum were determined in clinical specimens from horses in Ohio and compared with those found in A. phagocytophilum strains from various hosts and geographic regions. With increasing numbers of seropositive horses, the study points out the unrecognized prevalence and uncharacterized strains of A. phagocytophilum infection in horses and the importance of A. phagocytophilum molecular testing for the prevention of equine and human granulocytic anaplasmosis.

This study evaluated the health risks associated with nitrate and nitrite in water of Anka artisanal mining area. 104 water samples were obtained in both the dry and rainy seasons using standard methods. Inductively Coupled Plasma-Optical Emission Spectrometer (ICPOES) was used to estimate the concentrations of NO3 and NO2. The single pollution index (SPI) and the Nemerow Pollution Index (NPI) were used to evaluate the extent of contamination. The approach given by the USEPA was used to calculate non-carcinogenic health risks. During the dry season, the average concentrations of NO3 and NO2 in groundwater in the area were 298.70 mg/L and 0.87 mg/L, respectively, whereas during the rainy season, they were 64.34 mg/L and 9.16 mg/L. During the dry season, the mean values of NO3 and NO2 in surface water were 71.99 mg/L and 8.13 mg/L respectively, while during the wet season, they were 71.79 mg/L and 8.24 mg/L. Infants are more susceptible to non-carcinogenic health concerns linked with NO3 and NO2 in water from this area, according to a health risk assessment. To avoid health hazards connected with nitrate and nitrite, it is advised that mining activities in the vicinity be closely monitored. [ABSTRACT FROM AUTHOR]

Simple Summary: The populations of bison and moose, the largest wild ruminants in Poland, are growing every year. These animals return to Polish forests after many years of risk of extinction or, as in the case of European bison, are reintroduced step by step. Unfortunately, they are still rare and require close health surveillance and monitoring. One serious threat to their health and life is the bacterial parasite Anaplasma phagocytophilum. Moose and bison can also be sources of pathogenic bacteria for humans, which can be transferred through tick bites. In line with the World Health Organization (WHO) "OneHealth" program, indicating that animal health affects human health and vice versa, the following study examines the occurrence of Anaplasma phagocytophilum bacteria in European bison and Eurasian moose populations using molecular biology tools. Our results provide useful data regarding Anaplasma phagocytophilum that could be used in future strategies for the diagnosis and treatment of people and animals. Wild large ungulates, like European bison (Bison bonasus) and Eurasian moose (Alces alces), form an important part of the circulation of Anaplasma phagocytophilum, a Gram-negative, intracellular, tick-transmitted bacterium, in the natural environment. Bison and moose tissue samples were subjected to 16S rDNA, groEL and ankA partial gene marker amplification with specific primers using various variants of PCR. Out of 42 examined individuals, Anaplasma sp. were detected in 4/13 Eurasian moose (31%) and 7/29 European bison (24%). In addition, 12 groEL and 5 ankA partial gene positive samples were obtained from the examined animals. The phylogenetic analysis of the groEL partial gene classified samples from European bison to ecotype I, and samples from Eurasian moose to ecotype I and II; the analysis of the ankA partial gene assigned the samples to clusters I and IV. This study extends knowledge about A. phagocytophilum in wild large ungulates in Poland. This is the first report about the occurrence of Anaplasma sp. in one of the largest populations of free living European bison in the world. Our findings confirm that strains of A. phagocytophilum from Bison bonasus and Alces alces may constitute a natural reservoir of pathogenic HGA Anaplasma strains. [ABSTRACT FROM AUTHOR]

Abstract Background Anaplasma phagocytophilum is a Gram-negative obligate intracellular bacterium that replicates in neutrophil granulocytes. It is transmitted by ticks of the Ixodes ricinus complex and causes febrile illness in humans and animals. The geographical distribution of A. phagocytophilum spans the Americas, Europe, Africa and Asia. However, human disease predominantly occurs in North America but is infrequently reported from Europe and Asia. In North American strains, the absence of the drhm gene has been proposed as marker for pathogenicity in humans whereas no information on the presence or absence of the drhm gene was available for A. phagocytophilum strains circulating in Europe. Therefore, we tested 511 European and 21 North American strains for the presence of drhm and compared the results to two other typing methods: multilocus sequence typing (MLST) and ankA-based typing. Results Altogether, 99% (478/484) of the analyzable European and 19% (4/21) of the North American samples from different hosts were drhm-positive. Regarding the strains from human granulocytic anaplasmosis cases, 100% (35/35) of European origin were drhm-positive and 100% (14/14) of North American origin were drhm-negative. Human strains from North America and Europe were both part of MLST cluster 1. North American strains from humans belonged to ankA gene clusters 11 and 12 whereas European strains from humans were found in ankA gene cluster 1. However, the North American ankA gene clusters 11 and 12 were highly identical at the nucleotide level to the European cluster 1 with 97.4% and 95.2% of identity, respectively. Conclusions The absence of the drhm gene in A. phagocytophilum does not seem to be associated with pathogenicity for humans per se, because all 35 European strains of human origin were drhm-positive. The epidemiological differences between North America and Europe concerning the incidence of human A. phagocytophilum infection are not explained by strain divergence based on MLST and ankA gene-based typing.

Anaplasma phagocytophilum, a tick-borne obligately intracellular bacterium of neutrophils, causes human granulocytic anaplasmosis. Ankyrin A (AnkA), an effector protein with multiple ankyrin repeats (AR) is injected via type IV-secretion into the host neutrophil to gain access to the nucleus where it modifies the epigenome to promote microbial fitness and propagation. AR proteins transported into the host cell nucleus must use at least one of two known eukaryotic pathways, the classical importin β-dependent pathway, and/or the RanGDP- and AR (ankyrin-repeat)-dependent importin β-independent (RaDAR) pathway. Truncation of the first four AnkA N-terminal ARs (AR1-4), but not other regions, prevents AnkA nuclear accumulation. To investigate the mechanism of nuclear import, we created point mutations of AnkA N-terminal ARs, predicted to interfere with RaDAR protein import, and used importazole, a specific inhibitor of the importin α/β, RanGTP-dependent pathway. Nuclear colocalization analysis shows that nuclear localization of AnkA is unaffected by single AR1-4 mutations but is significantly reduced by single mutations in consecutive ARs suggesting RaDAR protein nuclear import. However, AnkA nuclear localization was also decreased with importazole, and with GTPγS. Furthermore, A. phagocytophilum growth in HL-60 cells was completely suppressed with importazole, indicating that A. phagocytophilum propagation requires a β-importin-dependent pathway. A typical classical NLS overlapping AR4 was subsequently identified suggesting the primacy of the importin-α/β system in AnkA nuclear localization. Whether the mutational studies of putative key residues support RaDAR NLS function or simply reflect structural changes that diminish engagement of an AR-NLS-importin pathway needs to be resolved through careful structure-function studies.

Antineutrophil cytoplasmic autoantibody (ANCA)-associated vasculitis (AAV) primarily affects small- and medium-sized arteries, including kidney vessels, thus causing rapidly progressive glomerulonephritis. The pathogenesis of AAV is intricate and several factors, including infections, are known to possibly trigger the autoimmune process. Numerous studies have reported that SARS-CoV-2 might cause acute kidney injury (AKI). To date, a modest number of AAV with COVID-19 cases has been reported. Abstract Background Anaplasma phagocytophilum is a Gram-negative obligate intracellular bacterium that replicates in neutrophil granulocytes. It is transmitted by ticks of the Ixodes ricinus complex and causes febrile illness in humans and animals. We used multilocus sequence typing (MLST) and ankA gene-based typing to study the molecular epidemiology of the A. phagocytophilum strains circulating in a German cattle herd over one pasture season. The aim was to investigate whether co-infection with two distinct variants, reinfection with the same and/or superinfection by a different strain occurred during one pasture season. Eight genetic loci were sequenced in 47 PCR-positive samples from 15 animals. Results Five different sequence types (ST) and four ankA alleles were detected in the cattle herd. Three different ST caused clinically overt tick-borne fever in primary infected animals. The concordance between ST and ankA allele was 100%. Therefore, the housekeeping genes used for MLST and the highly variable ankA gene were concatenated to increase resolution. Co-infection could be proven because samples of chronologically close collection dates were included. Co-infecting A. phagocytophilum strains differed by 14 to 18 single nucleotide polymorphisms (SNPs). Most superinfecting variants varied by 14 SNPs from the previous strain and appeared in median after a free interval of 31 days. Thus, it is unlikely that superinfecting strains arose by in-animal evolution. Immunity against re- or superinfection was assumed because the cattle developed clinical signs only during primary infection. Conclusions The tick-pathogen-vertebrate host interaction is probably much more complex than previously thought taking into account the frequently occurring events of co-infection, reinfection and superinfection. This complex situation could not be easily simulated in an experimental infection and underlines the value of field studies.

Purpose: The Anaplasma genus includes a Gram-negative bacterium infecting the blood cells of wild and domestic mammals, causing tick-borne fever. Infection with pathogenic Anaplasma phagocytophilum strains may cause Human Granulocytic Anaplasmosis. Wild boars (Sus scrofa) may act as natural wild reservoir hosts for potentially zoonotic A. phagocytophilum strains; however, there is still little data to confirm this statement. The aim of this study was to verify whether wild boars can be classified as natural reservoirs of Anaplasma spp. and to compare the suitability of spleen and liver samples for such analysis. Methods: Liver and spleen samples were collected from 59 wild boars (2017–2019). The organs were tested for Anaplasma phagocytophilum using short (partial) fragments of three markers: 16S rRNA, groEL, ankA. Results: Anaplasma spp. DNA was detected in 12 out of 59 samples, with a prevalence of 20.34%. The presence of A. phagocytophilum was confirmed by sequencing of the partial 16S rRNA gene. Positive individuals were tested for the characteristic markers: groEL and ankA. The analysis of the nucleotide sequences of 16S rRNA, groEL and ankA, indicated that the strains of A. phagocytophilum detected in these studies are potentially zoonotic for humans. Conclusion: Wild boars from Poland can be classified as a natural reservoir of the zoonotic strain of Anaplasma phagocytophilum. Artisanal mining of precious metals has become a lucrative venture in Anka, Zamfara State, North-West Nigeria. The use of crude methods of mining and mineral processing and the apparent ignorance of the hazards involved have been linked to some health and environmental consequences. Hence, this study focuses on the assessment of radioactivity levels in rock, soil and sediment samples as well as the radiological hazards of the artisanal mining in Anka. The gamma-ray spectrometry method was employed in the sample analysis. The results show that the estimated mean activity concentrations of 226Ra, 238U, 232Th and 40K in soil were 37.94 ± 6.01 Bqkg−1, 41.60 ± 11.06 Bqkg−1,151.15 ± 21.09 Bqkg−1 and 380.34 ± 116.41 Bqkg−1, respectively. The activity concentrations in rocks and sediments were also analyzed. The radioactivity levels were highest in rock samples. ; For rock samples, mean radium equivalent was 499.18 Bqkg−1, gamma dose rate was 221 ± 35 nGyh−1, internal and external hazard indices were 1.35 and 1.53, respectively and the radioactivity level index was 3.24. The outdoor and indoor annual effective dose equivalents were 271.03 µSvy−1 and 985.39 µSvy−1, respectively. The mean indoor and outdoor excess lifetime cancer risk for soil were 0.55 × 10−3 and 2.18 × 10−3. The evaluation of radiological health risks shows that the values obtained in this study were generally higher than admissible recommended limits and the world averages. The results obtained in this study show that the risk of exposure to ionizing radiation is linked to the artisanal mining and mineral processing in Anka.

Toxic metals are deleterious to the environment. In this study, risks associated with metals in water from Anka area, Nigeria were unraveled. A total of 104 water samples were collected. The concentrations of metals were determined using high performance liquid chromatography–inductively coupled plasma-mass spectrometry (HPLC–ICP-MS), while cations and anions were analyzed using inductively coupled plasma optical emission spectrometry (ICP-OES). Results revealed that the mean concentration of heavy metals in groundwater are Cu: 4.16 mg/l; Zn: 4.23 mg/l; Pb: 0.05 mg/l; Fe: 0.52 mg/l. For surface water, the mean concentration of Cr, Ni, Cu, As, Pb, and Zn is 0.06 mg/l, 6.53 mg/l, 5.92 mg/l, 0.06 mg/l, and 6.45 mg/l, respectively. Average concentrations of Cr, Co, Ni, Cu, Pb, and Zn in mine water are 0.12 mg/l, 2.11 mg/l, 0.66 mg/l, 4.44 mg/l, 2.72 mg/l, and 11.82 mg/l, respectively. Generally, metals in the samples are above recommended drinking water quality standards. Samples were extremely contaminated and released by geogenic and aggravated by mining. The metals pose great ecological risk but do not pose non-carcinogenic health risk while they pose carcinogenic health risk in both through oral and dermal contact with contaminated water. Anaplasma phagocytophilum (AP) has vast geographical and host ranges and causes disease in humans and domesticated animals. We investigated the role of northward migratory birds in the dispersal of tick-borne AP in the African-Western Palearctic. Ticks were collected from northward migratory birds trapped during spring migration of 2010 at two localities in the central Mediterranean Sea. AP DNA was detected by PCR (gltA and 16S rRNA) and variant determination was performed using ankA sequences. In total, 358 ticks were collected. One of 19 ticks determined as Ixodes was confirmed positive for AP DNA. The tick was collected from a woodchat shrike (Lanius senator senator) trapped in Greece, and molecularly determined to belong to the I. ricinus complex and sharing highest (95%) 16S RNA sequence identity to I. gibbosus. The ankA AP sequence exhibited highest similarity to sequences from rodents and shrews (82%) and ruminants (80%). Phylogenetic analyses placed it convincingly outside other clades, suggesting that it represents a novel AP variant. The divergent Ixodes species harboring a novel AP variant could either indicate an enzootic cycle involving co-evolution with birds, or dissemination from other regions by avian migration. None of the 331 Hyalomma marginatum sensu lato ticks, all immature stages, were positive for AP DNA, lending no evidence for the involvement of Hyalomma ticks transported by birds in the ecology of AP. In recent times, there had been reported cases of Pb poisoning in Anka gold mining area, Northwest Nigeria. Therefore, this study was carried out to determine the extent of bioaccumulation of heavy metals in the hairs and nails of children in the area. Forty samples (twenty nails and twenty hairs) samples were collected from ten boys and ten girls of ages 5–9 residing in the area. To ascertain the sources of heavy metals in children, 15 soils samples, 15 groundwater samples, 5 samples of mine tailings, and 5 plants samples were collected. Hair and nails of the subjects were collected using internationally acceptable techniques. All samples were kept in uncontaminated ziplock bags prior to laboratory preparation and analysis. The samples were cleaned using nonionic detergent (triton X-100) and deionized water. The hairs and nails were digested with 10 mL of 6:1 mixture of nitric acid and perchloric acid. The soils, mine tailings, and plants were air-dried at room temperature, sieved, and chemically digested using the aqua regia method. The concentrations of metals in all the samples were determined using high-performance liquid chromatography–inductively coupled plasma–mass spectrometry. Statistical analysis was employed to unravel potential sources of metals in the media. Results showed that heavy metals in children of the area are above results from similar studies and pathological ranges for heavy metals in hairs and nails. Also, heavy metals in environmental media are above the recommended standards. Multivariate analysis showed that the metals are mainly from mining and other anthropogenic sources. Results of correlation between heavy metals in hairs and nails with those in geological samples revealed that heavy metal that bioaccumulates in the children of this area are mostly from contaminated environmental media. Background: Anaplasma phagocytophilum is a Gram-negative obligate intracellular bacterium that replicates in neutrophil granulocytes. It is transmitted by ticks of the Ixodes ricinus complex and causes febrile illness in humans and animals. The geographical distribution of A. phagocytophilum spans the Americas, Europe, Africa and Asia. However, human disease predominantly occurs in North America but is infrequently reported from Europe and Asia. In North American strains, the absence of the drhm gene has been proposed as marker for pathogenicity in humans whereas no information on the presence or absence of the drhm gene was available for A. phagocytophilum strains circulating in Europe. Therefore, we tested 511 European and 21 North American strains for the presence of drhm and compared the results to two other typing methods: multilocus sequence typing (MLST) and ankA-based typing. Results: Altogether, 99% (478/484) of the analyzable European and 19% (4/21) of the North American samples from different hosts were drhm-positive. Regarding the strains from human granulocytic anaplasmosis cases, 100% (35/35) of European origin were drhm-positive and 100% (14/14) of North American origin were drhm-negative. Human strains from North America and Europe were both part of MLST cluster 1. North American strains from humans belonged to ankA gene clusters 11 and 12 whereas European strains from humans were found in ankA gene cluster 1. However, the North American ankA gene clusters 11 and 12 were highly identical at the nucleotide level to the European cluster 1 with 97.4% and 95.2% of identity, respectively. Conclusions: The absence of the drhm gene in A. phagocytophilum does not seem to be associated with pathogenicity for humans per se, because all 35 European strains of human origin were drhm-positive. The epidemiological differences between North America and Europe concerning the incidence of human A. phagocytophilum infection are not explained by strain divergence based on MLST and ankA gene-based typing. Background: Anaplasma phagocytophilum is a Gram-negative obligate intracellular bacterium that replicates in neutrophil granulocytes. It is transmitted by ticks of the Ixodes ricinus complex and causes febrile illness in humans and animals. We used multilocus sequence typing (MLST) and ankA gene-based typing to study the molecular epidemiology of the A. phagocytophilum strains circulating in a German cattle herd over one pasture season. The aim was to investigate whether co-infection with two distinct variants, reinfection with the same and/or superinfection by a different strain occurred during one pasture season. Eight genetic loci were sequenced in 47 PCR-positive samples from 15 animals. Results: Five different sequence types (ST) and four ankA alleles were detected in the cattle herd. Three different ST caused clinically overt tick-borne fever in primary infected animals. The concordance between ST and ankA allele was 100%. Therefore, the housekeeping genes used for MLST and the highly variable ankA gene were concatenated to increase resolution. Co-infection could be proven because samples of chronologically close collection dates were included. Co-infecting A. phagocytophilum strains differed by 14 to 18 single nucleotide polymorphisms (SNPs). Most superinfecting variants varied by 14 SNPs from the previous strain and appeared in median after a free interval of 31 days. Thus, it is unlikely that superinfecting strains arose by in-animal evolution. Immunity against re- or superinfection was assumed because the cattle developed clinical signs only during primary infection. Conclusions: The tick-pathogen-vertebrate host interaction is probably much more complex than previously thought taking into account the frequently occurring events of co-infection, reinfection and superinfection. Anaplasma phagocytophilum is a tick-transmitted Gram-negative obligate intracellular bacterium that replicates in neutrophil granulocytes. It causes tick-borne fever in cattle and sheep. We report here the case of a 5-year-old cow from Germany with clinically overt granulocytic anaplasmosis presenting with fever, lower limb oedema and drop in milk-yield. The herd encompassed 10 animals, 8 other animals showed subclinical infection. The strains from the 9 A. phagocytophilum positive cows were molecularly characterized using ankA gene-based and multilocus sequence typing (MLST). Seven of 9 (78%) animals were infected simultaneously with different ankA variants belonging to ankA clusters I and IV. MLST analysis also revealed the presence of multiple strain types. This could be due to co-transmission or superinfection. The search for new antimalarial agents is at the forefront for the global fight against antimalarial drug resistance as malaria treatments take longer than the three days plan. These changes pose a threat to the progress made thus far and search for newer and more rapid antimalarial agents is needed to maintain that success. The potentials of Magnesium Oxide Nanoparticles (MgO NPs) enhanced Chloroquine were assessed on Plasmodium berghei in vivo. Magnesium Oxide Nanoparticles were synthesized using sol-gel method and characterized using TEM (transmission electron microscope) and EDS (energy dispersive x-ray spectroscope), SEM, UV-VIS and FTIR. Concentration of 10 mg/ml of MgO NPs were prepared and combined with graded doses of chloroquine (25mg, 12.5mg and 6.25mg per kg) and assayed on Plasmodium berghei. Mice for this research were cared for at the Pharmacology Department Aminu Kano Teaching Hospital (AKTH) and weighed before the start of the experiment. Plasmodium berghei ANKA (PbA) infection was successfully inoculated in all the mice from a donor. The mice were grouped into five A, B, C, D and E consisting of 6 mice each. Group A- no treatment, Group B-50mg of chloroquine Group C were treated with 10mg/ml MgO NPs and 25mg chloroquine and Group D were treated with 10mg/ml MgO NPs and 12.5mg chloroquine and group E- 10mg/ml MgO NPs and 6.25mg of chloroquine once a day for 4 days. TEM shows that the particles are all under 100nm cylindrical and spherical in shape. EDS shows that the sample contains magnesium and oxygen in the ratio of 2:1. In-vivo antiplasmodial activity shows that all the groups that were given NPs combined with chloroquine had better parasite clearance rate (F(3, 20) =47.39, p< 0.00 at 0.05 level of significance). By all counts chloroquine enhanced with MgO NPs is considered to be more effective. The T-HSD shows that there is statistically significant difference between groups. It is hoped this study will inform malaria agencies of the potentials of this promising agent in the treatment of Plasmodium infection and further studies be carried out on the said compound. AnkA binds to gene promoters to dampen cis-transcription and also has features of matrix attachment region (MAR)-binding proteins that regulate three-dimensional chromatin architecture and coordinate transcriptional programs encoded in topologically-associated chromatin domains. We hypothesize that identification of additional AnkA binding sites will better delineate how A. phagocytophilum infection results in reprogramming of the neutrophil genome. Using AnkA-binding ChIP-seq, we showed that AnkA binds broadly throughout all chromosomes in a reproducible pattern, especially at: i) intergenic regions predicted to be matrix attachment regions (MARs); ii) within predicted lamina-associated domains; and iii) at promoters ≤3,000 bp upstream of transcriptional start sites. These findings provide genome-wide support for AnkA as a regulator of cis-gene transcription. Moreover, the dominant mark of AnkA in distal intergenic regions known to be AT-enriched, coupled with frequent enrichment in the nuclear lamina, provides strong support for its role as a MAR-binding protein and genome re-organizer. AnkA must be considered a prime candidate to promote neutrophil reprogramming and subsequent functional changes that belie improved microbial fitness and pathogenicity.

Tick-borne infections such as granulocytic anaplasmosis number among emerging infectious diseases. Anaplasma phagocytophilum is an obligate intracellular bacterial parasite infecting the granulocytes of vertebrates. This bacterium is the aetiologic agent of HGA (human granulocytic anaplasmosis). Molecular methods allow quick and accurate detection of pathogens in ticks, humans, or animals. Monitoring of the environment for A. phagocytophilum involves both classical and nested PCR, since these simple methods are most efficient. As markers, parts of the 16S rRNA, ankA, groESL, msp2, or msp4 genes are used for analyses. Molecular methods have enabled analysis of the genomes of pathogens, differentiation between strains and thus, in association with ecological studies, they facilitate an understanding of their biology, pathogenicity and mode of diffusion in the natural environment.

Roman wall fragments dating from 2nd and 3rd centuries AD, from two sites of the Roman province of Germania Superior, near the present day towns of Wössingen (Baden-Württemberg) and Mülheim-Kärlich (Rheinland-Pfalz), were analyzed with synchrotron-based scanning macro-X-ray fluorescence (SR-MA-XRF) at the FLUO beamline (ANKA Synchrotron Radiation Facility, Karlsruhe Institute of Technology). Lead, iron, copper, calcium, potassium, among other elements, were detected and are correlated with red, blue, yellow and green pigments. In the fragments from Mülheim-Kärlich, red has high correlation with Pb (red lead pigment), Fe and Ca were detected in 2 different hues of yellow respectively, and Cu is correlated with the blue pigment. The green pigment investigated in the Wössingen's fragments has high correlation with Fe and K, indicating the use of green earth, and the red pigment is correlated with iron, indicating the use of the red ochre. Synchrotron-based scanning macro-X-ray fluorescence applied in fragments of wall paintings has shown to be a fast, non-destructive and effective technique in the identification and discrimination of the pigments, plaster, different layers and impurities in paintings through the 2D elemental distribution. The use of mythology in Contemporary Arabic Poetry first appeared as a result of the attempts to develop the contents of the Arabic Poetry towards the second half of the 19th century. The socio-politic developments in the Arabic world following the Second World War constitute a turning point in the Arabic Literature. The anger caused by the lost of some Arabic lands is especially reflected in the young poets. These poets consider the condition of Arabic world as a "death" state; however, they compare it to the death state of the nature which is always reborn. Consequently, a new movement called "Tammoz Poets" that connects the belief that the Arabic world will be reborn and the myths symbolizing the rebirth of nature in the Sumerian, Egyptian and Greek Mythologies is born and takes its name from the Sumerian Myth of Tammoz. This Poetry movement uses various myths in the poems and assigns new meanings to these old myths in order to affect people and to pass their intended messages, and uses various figures from the Arabic culture and folklore to create different symbols. Mining is a major human activity that has contributed significantly to high degree of environmental and human health degradation. This study was done to uncover the degree of contamination and risks associated with metals in environmental media collected from Anka area, Northwest Nigeria. A total of eighty-two (82) samples which include 42 soils, 22 stream sediments, 13 tailings and 5 plants were collected. Media were air-dried, pulverized and sieved to collect fine particles. They were digested and analyzed for toxic metal contents using High Performance Liquid Chromatography-Inductively Coupled Plasma Mass Spectrometer (HPLC-ICPMS). Metals analyzed include As, Cd, Co, Cr, Cu, Hg, Ni, Pb, Sb, Zn and Fe. From the results, degree of contamination in samples were obtained using pollution indices, while ecological and health risks were calculated using acceptable risk indices. Average concentrations of metals in the soils were: As: 0.64 μg/g; Cr: 42.55 μg/g; Pb: 131.76 μg/g; Hg: 0.85 μg/g while in sediments, the average concentration were As: 15.46 μg/g; Cr: 111.82 μg/g; Pb: 2234.02 μg/g; Hg: 2.12 μg/g. In tailings, the mean concentration were As: 4.79 μg/g; Cr: 96.95 μg/g; Pb: 2802.56 μg/g; Hg: 1.25 μg/g. Metals in plants are above acceptable limits. Metals in the media are both from geogenic and mining activities, and they pose high ecological risks. High carcinogenic (>10−4) and non-carcinogenic (>1) health risks are associated with metals in media from this area. Further epidemiological studies should be carried to know the extent of occurrence of diseases associated with mining in the area. Anaplasma phagocytophilum is a Gram-negative obligate intracellular bacterium that replicates in neutrophil granulocytes. It is transmitted by ticks and causes tick-borne fever in domestic ruminants such as sheep, cattle and goats. However, in contrast to sheep and cattle little is known about the clinical course of infection in goats. We report here on three cases of symptomatic infection with A. phagocytophilum in two goats (Capra aegagrus hircus) and one water buffalo (Bubalus bubalis). The animals showed symptoms and laboratory findings similar to sheep and cattle. To our knowledge, this is the first report on the symptomatic infection of water buffalos with A. phagocytophilum. The infecting strains were genetically characterized by 16S rRNA gene, ankA gene and multilocus sequence typing (MLST). Four other strains from asymptomatically infected goats were also included. The ankA sequences from five goats were part of the formerly described ankA gene clusters I and IV that are known to contain A. phagocytophilum strains from sheep and cattle. However, the sequences from one goat and from the water buffalo belonged to ankA gene cluster II that was formerly described to be restricted to roe deer. A similar observation was made for MLST as three goats clustered with sequences from sheep and cattle, whereas three other goats and the water buffalo were found to be part of the roe deer cluster. However, since most of the strains from sheep and cattle were distinct from the roe deer strains, roe deer might not represent major reservoir hosts for tick-borne fever in domestic ruminants. When differing parts of the 16S rRNA gene were used for typing the results were conflicting. Vascular pathology is central to malaria pathogenesis and associated with severity of disease. We have previously documented shedding of the cerebral endothelial glycocalyx in experimental malaria and hypothesized that this action is implicated in the pathogenesis of cerebral malaria (CM). Quantification and characterization of the intraluminal vascular glycocalyx are technically challenging. Here, we used ferritin labeling, computerized image analysis, and biochemical characterization by using in vivo biotinylation and pull down. Image analysis divided mice with CM and uncomplicated malaria and uninfected control mice into 3 non-overlapping groups. Biochemical assessment of the luminal surface revealed malaria-induced alterations in all components of the glycocalyx in CM. This loss was mirrored in increases of the same components in peripheral blood samples. Corticosteroid treatment protected against CM, reduced inflammation, and prevented glycocalyx loss. Adjunctive antithrombin-3 also prevented glycocalyx loss and significantly reduced CM-associated mortality, as well as reduced local inflammation and prevented blood-brain barrier leakage. In contrast, inhibition of matrix metalloproteases with batimastat had limited effects on the glycocalyx and disease progression. Thus, glycocalyx loss may be associated with malaria pathogenesis and could be targeted by adjunctive treatment.-Hempel, C., Sporring, J., Kurtzhals, J. A. L.
