Introduction: Avian coccidiosis presents a significant challenge to the poultry industry in Egypt, highlighting the urgent need for validating new drug targets offering promising prospects for the development of advanced anticoccidials. Although numerous reports highlight the activity of lactoferrin (LF) against various microorganisms, its potential against Eimeria has not been explored. The present study evaluated the potential anticoccidial effect of LF and diclazuril in broiler chickens experimentally infected with Eimeria tenella. Methods: A total of 100 one-day-old broiler chicks were divided into five equal groups (20 each) as follows: Group 1 (G1) served as the normal healthy control group, Group 2 (G2) consisted of chickens infected with 1 × 105 sporulated E. tenella oocysts at 14 days of age, Group 3 (G3) comprised infected chickens treated with diclazuril (0.5 mL/L in drinking water) for 3 days successively, Group 4 (G4) included infected chickens treated with LF (at a dose of 250 mg/kg of diet) from one day of age until the end of the study, and Group 5 (G5) comprised infected chickens treated with both LF and diclazuril. Results: The positive control group (G2) experienced significant reductions in body weight (BW), BW gain, serum glucose, lipase, amylase, total antioxidant capacity, several hematological indices, and total proteins, along with alterations in various antioxidant enzymes. Conversely, serum levels of aspartate aminotransferase (AST), Alanine aminotransferase (ALT), Alkaline phosphatases (ALP), urea, creatinine, nitric oxide, mean corpuscular volume (MCV), White blood cells (WBCs), heterophils, alpha 2, beta 1, and liver contents of malondialdehyde were elevated in this group. Moreover, higher oocyst counts and lesion scores, along with histopathological alterations, were observed in G2. Remarkably, treatment with diclazuril and/or LF demonstrated potent antioxidant and anticoccidial effects, resulting in reduced shedding of oocysts, lesion scores, and lymphocytic infiltrates in the cecum. Additionally, these treatments improved the antioxidant and immune systems in chickens and restored all histopathological changes reported in the infected non-treated group (G2). Conclusion: This study offers novel perspectives on the potential anticoccidial effects of the combination of LF and diclazuril in broiler chickens infected with E. tenella, highlighting the potential synergistic actions of LF in treating poultry coccidiosis. Quails are an important alternative to chicken production for protein sources, offering many advantages over other poultry species. However, raising quail faces certain challenges, such as a shortage of specified hatcheries and the lack of local markets for quail eggs and meat, particularly in Egypt. In addition, there is less interest in quail's medication and vaccine production. A significant disease affecting the health and productivity of quails is coccidiosis, which is associated with poor feed conversion ratio, lower growth performance, heightened mortality, and high cost of vaccination and treatment. Attention to quail coccidiosis and its clinical forms, diagnosis, morphological characterization, control, and prevention is very critical for improving quail meat and egg production. The efficacy of Mahonia bealei (Fort.) leaves extract (MBLE) against coccidiosis in chickens was studied in vivo. For this purpose, a total of 120 Hyland brown chickens (one-day-old) were randomly divided into six groups, each comprising 20 chickens, viz. MBLE-L, MBLE-M, and MBLE-H; drug control group; infected-untreated group; and uninfected-untreated group. Except for the uninfected/untreated group, the chickens in all other groups received an oral inoculation of 1.0 × 104 pieces/feather sporulated oocysts of E. tenella on day 15. Then, for 7 days in a row, the drug control group was given sulfachloropyrazine sodium soluble powder (1.0 g/day) in drinking water, while the three treatment groups MBLE-L, MBLE-M and MBLE-H were given the MBLE @0.25, 0.5, and 1.0 g/day, respectively. The anticoccidial effects were evaluated by lesion score, body weight gain, oocyst output, and histopathological changes in the liver, kidney, and cecum of the chickens in each group. The results showed that no chickens died in all groups except 2 chickens died in the untreated group. The MBLE groups were able to decrease coccidia oocyst output, mitigate the impact of coccidian infection on chicken weight increase, and lessen pathological alterations in the liver, kidney, and cecum of infected chickens. Among these, the MBLE-H group demonstrated the greatest efficacy with an anticoccidial index of 159.17. Coccidia is an ubiquitous intestinal protozoan of poultry that invade the mucosa and induce epithelial cell necrosis and inflammation. To assess the pathogenicity of two commercial anticoccidial vaccines against a local isolate of Eimeria tenella, ninety (90) day-old dominant black chicks were assigned to 9 groups of 10 birds each. Immunisation was done at 5 days of age by oral gavage. Infection was done with 1.7×104 E. tenella of either Houghton strain (H-strain) or local isolate (L-isolate) at 4 weeks of age. Increased pathogenicity of the local isolate was observed, as it produced a more severe gross pathologic lesion score than the Houghton strain in unvaccinated but infected groups of chick-ens 6 days post-infection (dpi). The high virulence and pathogenicity of the local E. tenella was re-sponsible for the differences in the severity of clinical signs, gross and microscopic lesions observed between the H-strain and L-isolate infected groups. The histopathology showed that the vaccinated groups, infected with the H-strain of E. tenella, did not show presence of oocysts or schizonts by 6 dpi. The successful immunogenicity and effectiveness of these two vaccines as measured by the histopathologic lesions and the presence of oocysts in the enterocytes of the caecal crypts of these chickens were reduced in vaccinated and L-isolate infected groups. The findings in the L-isolate groups were setbacks to the use of live attenuated and non-attenuated anticoccidial vaccines for control of coccidiosis and in particular, caecal coccidiosis in chickens reared in environments dominated by virulent E. tenella.

ABSTRACT: Nanomedicine is a critical therapeutic approach for treating most poultry illnesses, particularly parasitic infections. Coccidiosis is a severe protozoan infection affecting poultry; the emergence of drug-resistant Eimeria strains demands the development of new, safe therapies. Consequently, the objective of this work was to investigate the efficacy of the biosynthesized selenium nanoparticles (SeNPs) by Paenibacillus polymyxa (P. polymyxa) against Eimeria tenella (E. tenella) experimental infection in broiler chickens. The prepared SeNPs absorbed the UV at 270 nm were spherical with a size of 26 nm, and had a surface negative charge of −25 mV. One hundred and fifty, 1-day-old male broiler chicks were randomly allocated into 5 groups (30 birds/group with triplicates each) as follows: T1: negative control (noninfected and nontreated with SeNPs); T2: delivered SeNPs (500 µg/kg diet) for 35 successive days, T3: E. tenella-infected (positive control birds), T4: E. tenella-infected and treated with SeNPs (500 µg/kg diet) and T5: E. tenella-infected chicks and treated with anticoccidial agent (sulfadimidine, 16% solution 8 mL/L of drinking water) for 5 successive days. At 14 d of age, each bird in infected groups was orally treated with 3 × 103 sporulated oocyst of E. tenella. SeNPs considerably decreased the number of oocysts in broiler feces compared to positive control and anticoccidial drug, followed by a substantial reduction of parasite phase count in the cecum (15, 10, and 8 for meronts, gamonts, and developing oocysts) when compared with positive control birds. The Eimeria experimental infection lowered the activity of antioxidant enzymes, superoxide dismutase (SOD), glutathione peroxidase (GPx), and reduced glutathione (GSH) while increasing the stress parameters nitric oxide (NO) and malonaldehyde (MDA). Moreover, the production of proinflammatory (TNF-α and IL-6) and apoptotic genes (BcL2 and Cas-3) were significantly elevated. Administrating SeNPs to chicks significantly decreased oxidative stress, inflammation, and apoptotic markers in the cecum tissue. Therefore, growth performance, carcass weights, antioxidant enzymes, and blood properties of infected chicks were enhanced. The findings compared the protecting role of Se-nanoparticles against cecum damages in E. tenella-infected broilers.

Abstract Background Avian coccidiosis is an important parasitic disease that has serious adverse effects on the global poultry industry. The extensive use of anticoccidial drugs has resulted in an increase in drug resistance. Ethanamizuril (EZL) is a novel triazine with high anticoccidial activity. Methods We compared oocyst production and sporulation between EZL-sensitive (S) and EZL-resistant Eimeria tenella strains (R10 and R200) and used label-free quantitative proteomics to identify differentially expressed proteins (DEPs) between these strains. Results We generated two EZL-resistant E. tenella strains: strain R10, which was induced using a constant dose of 10 mg EZL/kg poultry feed, and strain R200, which was generated by gradually increasing the EZL dosage to 200 mg EZL/kg poultry feed. With an increase in resistance, the total oocyst output decreased, but the percentage of sporulation did not change significantly. We identified a total of 7511 peptides and 1282 proteins, and found 152 DEPs in the R10 strain versus the S strain, 426 DEPs in the R200 strain versus the S strain and 494 DEPs in the R200 strain versus the R10 strain. When compared with the S strain, 86 DEPs were found to have consistent trends in both resistant strains. The DEPs were primarily involved in ATP and GTP binding, invasion, and membrane components. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway analyses of the DEPs suggested that they are involved in transcription and translation processes. Protein–protein interaction network analysis of the 86 DEPs showed that 10 proteins were hubs in the functional interaction network (≥ 8 edges) and five of them were ribosomal proteins. Conclusions The results of the present study indicate that the resistance mechanisms of E. tenella against EZL might be related to the transcriptional and translational processes, especially in the factors that inhibit the growth of parasites. The DEPs found in this study provide new insights into the resistance mechanisms of E. tenella against EZL. Further research on these potential targets holds promise for new chemotherapeutic approaches for controlling E. tenella infections. Graphical Abstract

This study was conducted in order to compare well established used chemical anticoccidial medication (diclazuril) against natural prepared safe alternative products of garlic extract (GE), Moringa oleifera (MO) leaves extract, onion extract (OE), in order to control experimentally infected with Eimeria tenella species in chickens. Performance parameter in form of average body weight (ABW) and feed conversion rate (FCR) were studied together with biochemical parameters (malondialdehyde (MDA), superoxide dismutase (SOD) and catalase (CAT), mortality rate, oocyst count in addition to total white blood cell (WBCs), lymphocytes and heterophils counts. Histopathological examination of intestinal tract in all test groups was studied. Results revealed that the lowest mortality rate was found in group treated with MO leaves extract. All challenged herbal extract treated groups revealed ABW and FCR lower than diclazuril treated infected group. All treated groups were lower in both average lesion score and average oocyst count two weeks post challenge when compared with control positive group indicate positive impact of all studied therapies either chemical or herbal products but with variable degrees as best effect was diclazuril followed by MO group, followed by GE group and finally group treated with OE. Experimental infection of chickens with E. tenella oocysts significantly increased MDA concentration when compared with control negative non-treated group (P < 0.01). However, infected birds fed with OE, GE, MO leaves extracts and diclazuril administration for a week pre-infection had significantly declined MDA concentrations compared with infected non-treated (P < 0.01). Control positive birds showed significant decrease in SOD and CAT activities vs. the healthy birds either at week pre-infection or at two days' post-infection (P < 0.01). However, SOD activities in birds fed with OE, MO leaves extract and diclazuril for a week pre-infection significantly higher (P < 0.01) than control positive. Histopathological finding revealed that best was group treated with diclazuril followed by group received MO, followed by group received GE and finally group received OE. It could be concluded that herbal extract may be representing a good alternative anticoccidial medications specially that the later may developed resistance for many Eimeria species in continuous use in veterinary field. Increase in drug resistance as well as ineffective immunization efforts against various pathogens (viruses, bacteria and fungi) pose a significant threat to the poultry industry. Spirulina is one of the most widely used natural ingredients which is becoming popular as a nutritional supplement in humans, animals, poultry and aquaculture. It contains protein, vitamins, minerals, fatty acids, pigments, and essential amino acids. Moreover, it also has considerable quantities of unique natural antioxidants including polyphenols, carotenoids, and phycocyanin. Dietary supplementation of Spirulina can beneficially affect gut microbial population, serum biochemical parameters, and growth performance of chicken. Additionally, it contains polyphenolic contents having antibacterial effects. Spirulina extracts might inhibit bacterial motility, invasion, biofilm formation, and quorum sensing in addition to acting directly on the bacterium by weakening and making the bacterial cell walls more porous, subsequently resulting in cytoplasmic content leakage. Additionally, Spirulina has shown antiviral activities against certain common human or animal viruses and this capability can be considered to exhibit potential benefits against avian viruses also. Spirulan, a calcium-rich internal polysaccharide of Spirulina, is potentially responsible for its antiviral effect through inhibiting the entry of several viruses into the host cells, boosting the production of nitric oxide in macrophages, and stimulating the generation of cytokines. Comparatively a greater emphasis has been given to the immune modulatory effects of Spirulina as a feed additive in chicken which might boost disease resistance and improve survival and growth rates, particularly under stress conditions. This manuscript reviews biological activities and immune-stimulating properties of Spirulina and its potential use as a dietary supplement in poultry to enhance growth, gut health and disease resistance. SUMMARY: In the United States, anticoccidials belonging to the ionophore class are considered antibiotics and cannot be used in marketing programs such as “No Antibiotics Ever” (NAE) or “Raised Without Antibiotics” (RWA). Therefore, as a growing percentage of broiler production becomes NAE/RWA, producers must learn to effectively prevent coccidiosis without the ionophore anticoccidials that have been the backbone of coccidiosis prevention programs worldwide for many decades. This requires that veterinarians and producers learn to effectively prevent and control coccidiosis with live coccidiosis vaccines, chemically-synthetized anticoccidials, and innovative techniques. For NAE/RWA production, synthetic anticoccidials and live coccidiosis vaccines can be used to prevent coccidiosis. For organic production, only live coccidiosis vaccines can be used. To make the vaccine programs work, all factors critical to achieving the development of active immunity, from vaccine storage, administration, and management in the field, must be optimized. Producers should utilize all of the tools at their disposal by using carefully planned programs and rotations along with improved brooding practices. Experiences show that coccidiosis can be effectively prevented in NAE/RWA poultry production through attention to planning, dedication and management.

Publications are an important measure of scientific and technological progress. The quantitative examination of the number of publications in a certain research topic is known as bibliometrics. Bibliographic studies are widely used to analyse the condition of research, future potential, and current growth patterns in a certain topic. It can serve as a basis for making decisions and implementing strategies to achieve long-term development goals. To our knowledge, no research has been conducted in these domains; so, this work aims to employ bibliometric analysis to provide comprehensive data on publications related to anticoccidial drugs. As a result, the current study uses bibliometric analysis to track the evolution of anticoccidial drugs and its consequences in the academic and public worlds via a survey of relevant scientific and popular publications. The Dimensions database was used to retrieve the bibliographical statistics, which were then cleaned and analyzed. The data was also loaded into the VOS viewer, which generated a network visualization of the authors with the most joint articles. The investigation discovered three stages of publications and citations since the first article on anticoccidial drugs in 1949. The first stage, which ran from 1920 to 1968, was characterized by a scarcity of research articles on anticoccidial drugs. From 1969 to 2000, the second stage was marked by a stable and marginally increased number of articles. The scientific field was characterized by an increasing trend in the number of publications and their citations from 2002 to 2021. The study gave a complete list of the top anticoccidial drugs funding agents, countries, research institutes, most cited publications, and important co-authorship and partnerships. The outcomes of the study will help veterinary practitioners and researchers understand the trends and best sources of knowledge in the field of anticoccidial medications.

SUMMARY: Avian coccidiosis is an intestinal infection caused by a parasitic intracellular protozoan of Eimeria species. The effect of coccidian intestinal infection in birds renders significant economic losses in production, and therefore, it is crucial for industry players and researchers to find solutions to its occurrence. The use of synthetic additives and anticoccidial supplements has been established to be effective in preventing and controlling coccidiosis. However, concerns about anticoccidial drug resistance in Eimeria species due to frequent use have been reported. Alternatively, the use of attenuated live vaccines has been successful but it comes with challenges because of poor farm management. Garlic and its bioactive compounds have been found to exhibit anti-inflammatory and antioxidant properties. The anticoccidial potential of garlic is linked to its immunostimulatory activity. Garlic and its derivatives inhibit the sporulation of oocysts in vitro. Supplementation of garlic in coccidiosis-infected broilers improves weight gain and feed efficiency, and reduces fecal oocysts output, lesion score, and clinical signs postinfection. Thus, this review aims to highlight the clinical signs of coccidiosis, the techniques involved in oocysts determination, and the anticoccidial potential of garlic in improving the health of broiler chickens. Future studies can bridge the knowledge gap on the mode of action of the bioactive compounds of garlic against coccidiosis and the Eimeria species-specific gene and immune-protective antigens that they inhibit and improve in infected birds.

In this study, chitosan nanoparticles (CsNPs) were used as nanocarrier for ultrasonicated ethanolic extract of Rosmarinus officinalis (UEERO) as a new nanoformulation against Eimeria tenella. Herein, CsNPs have been synthesized by ionic gelation method at pH 3 (CsNPs3) and pH 5 (CsNPs5), followed by characterization of morphology, size, polydispersity index (PDI), surface charge, and loading efficiency of UEERO. An in vitro sporulation inhibition assay (10, 5, 2.5, 1.25, 0.62, 0.31, 0.15, 0.07, 0.04, 0.02, and 0.01 mg/ml normal saline solution) against E. tenella was conducted. Results showed that free CsNPs and UEERO-CsNPs3/5 were cubic- and spherical-shaped with positive charge and average size of ~ 150.8 nm (314.4 nm) and 151.7 nm (321.1 nm), respectively. The total loading efficiency using UV–vis spectrophotometer, was 80.05 at pH 5 and 64.39% at pH 3. The in vitro sporulation inhibition assay revealed that UEERO, CsNPs3/5, and UEERO-CsNPs3/5 showed a potential inhibitory effect on sporulation (%), distortion in wall (%), and sporocyst abnormality (%) in a dose-dependent manner. Accordingly, the concentration (10 mg/ml) showed the best efficacy after 24 h in UEERO, free CsNPs, and UEERO-CsNPs. Moreover, UEERO-CsNPs3 and UEERO-CsNPs5 had stopped the sporulation (%) after 72 h. Taken all together, UEERO-CsNPs3 and UEERO-CsNPs5 are best effective against E. tenella in a dose-dependent manner in terms of sporulation (%), distortion in wall (%), and sporocysts abnormality. Due to the emergence of drug resistance against synthetic chemicals, effective alternatives like essential oils are required for the control of coccidiosis in poultry. Hence, the present in-vivo experiment was conducted on Citrus (C.) sinensis essential oil for investigation of its anticoccidial efficacy. For this purpose, 72 dayold chicks were procured from local market and divided randomly into six equal groups: A, B, C, D, E and F. On day 14, all the groups except F were administered with 50,000 sporulated oocysts (mixed Eimeria species). On the same day, groups A, B and C were given 1, 2 and 3% of C. sinensis essential oil, respectively, in feed, group D with Toltrazuril® while groups E and F were left unmedicated. The results revealed C. sinensis essential oil to have positive effect on FCR in broilers. Moreover, there was a significant improvement in oocysts per gram of faeces, lesion score, oocyst score and faecal score in oil treated broilers at the maximum inclusion level of 3% comparable to the standard control. However, apart from significant effect on WBCs count and serum levels of ALT and LDH, C. sinensis essential oil had very limited non-significant effect on hematological and serum biochemical parameters, and organs' weight. ABSTRACT: Yellow-feathered broiler chickens generally have a longer growth cycle than white broilers and therefore face different coccidiosis challenges. General single vaccine and drug regimens struggle to prevent coccidiosis for yellow broilers. In this study, a single vaccine, and a combination of coccidiostat regimens was employed to explore the preventative and control effects of different pilot regimens on coccidiosis in yellow-feathered broilers. A total of 2,000 one day old Chinese Huang Youma female broilers were allocated into 4 experimental groups, each with 5 replicates. All birds were fed the same starter feed from Days 1 to 25, and all groups were inoculated with a vaccine on Day 4. After Day 26, the groups were then fed as follows: (1) Negative control group: basal diet + vaccine (NC); (2) NC + maduramycin (NCMD); (3) NC + narasin (NCNR); and (4) NC + salinomycin (NCSL). From Days 26 to 75, the NCNR group had a lower FCR than the other groups. The 75-d BW was higher in the NCNR group than in the NCSL group but was not significantly higher than that in the NC and NCMD groups. The growth performance followed the same trend during the whole experiment (Days 1–75). Compared to the NC group, the NCNR and NCSL groups had higher intestinal mucosa SIgA concentrations at Day 40 and Day 60 (P < 0.001); however the NCMD group had lower IgG levels at Day 40 and Day 60 (P = 0.036, P = 0.006 respectively). The combination groups had significantly reduced AKP levels and urine acid concentrations at Day 60 in comparison to those of the NC group (P = 0.004). The NCNR and NCMD groups had less severe intestinal coccidiosis lesion scores than the NC and NCSL groups in older birds. Thus, a single vaccine and/or combinations with different coccidiostats had different effects on broilers. The NCNR group showed comparatively better growth performance, blood biochemical indices, immune response, and coccidiosis lesion scores.

Background: Avian coccidiosis is an important parasitic disease that has serious adverse effects on the global poultry industry. The extensive use of anticoccidial drugs has resulted in an increase in drug resistance. Ethanamizuril (EZL) is a novel triazine with high anticoccidial activity. Methods: We compared oocyst production and sporulation between EZL-sensitive (S) and EZL-resistant Eimeria tenella strains (R10 and R200) and used label-free quantitative proteomics to identify differentially expressed proteins (DEPs) between these strains. Results: We generated two EZL-resistant E. tenella strains: strain R10, which was induced using a constant dose of 10 mg EZL/kg poultry feed, and strain R200, which was generated by gradually increasing the EZL dosage to 200 mg EZL/kg poultry feed. With an increase in resistance, the total oocyst output decreased, but the percentage of sporulation did not change significantly. We identified a total of 7511 peptides and 1282 proteins, and found 152 DEPs in the R10 strain versus the S strain, 426 DEPs in the R200 strain versus the S strain and 494 DEPs in the R200 strain versus the R10 strain. When compared with the S strain, 86 DEPs were found to have consistent trends in both resistant strains. The DEPs were primarily involved in ATP and GTP binding, invasion, and membrane components. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway analyses of the DEPs suggested that they are involved in transcription and translation processes. Protein–protein interaction network analysis of the 86 DEPs showed that 10 proteins were hubs in the functional interaction network (≥ 8 edges) and five of them were ribosomal proteins. Conclusions: The results of the present study indicate that the resistance mechanisms of E. tenella against EZL might be related to the transcriptional and translational processes, especially in the factors that inhibit the growth of parasites. The DEPs found in this study provide new insights into the resistance mechanisms of E. tenella against EZL. This study was carried out to examine the anticoccidial effect of Citrus aurantium L ethanol leaf extract against the oocysts of Eimeria tenella isolated from broiler chickens. The fresh leaves of C. aurantium were collected from Emirate Garden, Katsina, authenticated, air-dried at room temperature, pulverised by milling and subjected to extraction. Sporulation inhibition bioassay was employed to examine the activity of C. aurantium ethanol extract on the sporulation of E. tenella oocysts. In this assay, deep well petri dishes containing 100 unsporulated oocysts were subjected to 2 ml of five different concentrations of the extract (2.5, 5, 10, 20 and 30 mg/ml) in triplicates while oocysts sporulated in 2.5% potassium dichromate solution (K2Cr2O7) and phenol served as control groups. The content of the Petri dishes was stirred to ensure adequate oxygenation. The experimental set-up was incubated at room temperature and examined after 24 and 48 hours for sporulation inhibition. The sporulated and unsporulated oocysts were determined by counting using the Mcmaster apparatus. Phytochemical screening of C. aurantium revealed the presence of alkaloids, saponins, carbohydrates, steroids and tannins. The result showed that ethanolic leaf extract of C. aurantium to possess anticoccidial activity against unsporulated oocysts of E. tenella in a concentration-dependent manner. There was significant difference (p < 0.05) in the sporulation inhibition activity, with the highest (97 ± 0.8%) at 30 mg/ml and the lowest activity (8 ± 1.0%) at 2.5 mg/ml concentration of the extract after 48 hours of incubation. There was a general trend of sporulation inhibition with an increase in the concentration of the plant extract. The findings from this study showed ethanol leaf extract of C. aurantium possesses a remarkable In vitro anticoccidial effect that may be further scientifically explicated. [ABSTRACT FROM AUTHOR]

Background: Coccidiosis is a debilitating poultry disease that costs the world more than £10.4 billion annually. Citrus aurantium L, also known as bitter orange, belongs to the family Rutaceae, it's been used in traditional Chinese medicine to treat nausea, constipation, cancer, cardiovascular effect, and as antioxidant. Anticoccidial activity of the ethanolic leaf extract of Citrus aurantium L. against experimental Eimeria tenella infection was studied in broiler chickens. Methodology: C. aurantium leaves were collected, authenticated, pulverized and extracted using ethanol. Thirty-six broiler chicks were obtained and thirty chickens were experimentally infected with 20,000 sporulated Eimeria tenella oocysts and assigned randomly into six groups of six chickens each. Group A was uninfected and untreated (negative control), group B served as infected-untreated control, group C were infected and treated with a standard drug; amprolium 1g/L of drinking water (positive control), groups D, E, and F were infected and treated with ethanolic leaf extract of C. aurantium at 125, 250 and 500 mg/kg orally, respectively, for seven consecutive days. Weight gain was calculated and the oocysts per gram of faeces were counted using the McMaster counting apparatus. Blood samples from each experimental group were collected on days 7 and 14 post-infection for haematological examination. Results: Results revealed that treatment with ethanolic leaf extract of C. aurantium dose-dependently decreased the E. tenella oocysts shedding, increase body weight, improved caecal lesion and significantly (p < 0.05) increased the PCV, TRBC and Hb concentration with significantly decreased (p < 0.05) TWBC of the infected chickens. Conclusion: C. aurantium provided beneficial effects against E. tenella-induced coccidiosis in broiler chickens. The plant should be further validated to investigate the essential compounds responsible for the anticoccidial effect.

Coccidiosis is an enteric disease caused by protozoa, especially Cystoisospora suis which can lead to large losses in production. Its transmission occurs through different routes, mainly affecting piglets in the first weeks of life and normally progressing with diarrhea unresponsive to antibiotic therapy. The objective of this work was to evaluate, in two seasons of the year, the occurrence of C. suis in piglets and sows, the transmission routes and source of infection and the influence of the preventive use of toltrazuril on these epidemiological parameters. Two experiments were carried out, the first being in winter; under the influence of prophylactic use of toltrazuril; and the second in the summer, six months after the suspension of the prophylactic use of toltrazuril. The study included 36 sows (18 controls, 18 treated toltrazuril). The following samples were collected: rest of the sows, water from the individual drinkers in each cage, organic swabs present in the keepers' shoes, environmental swabs pre and housing, present in the cages in the rooms and accumulations of the maternity cage. As were disclosed to coproparaological and PCR exams. The presence of C. bebe ocysts is the winter and summer control following samples: sow feces (2.84%) of the winter and summer control groups (7.60%) of the winter and summer treated groups and the treated group winter and summer, rectal swabs from litters (9.03%) in the control and treated groups in winter, collected from the environment (20.22%) from all groups. Like others, they were negative for the presence of the parasite. There was no difference between the control and toltrazuril groups and no positive samples were observed in the summer. The possible route of transmission of the agent in the present work was the water from the douros. Preventive therapy with toltrazuril in farms with low infection pressure is not necessary to control C. suis.

Avian coccidiosis is one of the major parasitic diseases in the poultry industry. The infection is caused by Eimeria species, and its treatment relies mainly on the administration of anticoccidial drugs, which can result in drug resistance and side effects. The recent trends in avian coccidiosis treatment is directed to the development of a new therapy using herbal compounds. S-Methylcysteine (SMC) is considered one of the organosulfur compounds in garlic that showed promising activity in the treatment of different pathological conditions via a wide range of anti-inflammatory and antioxidant mechanisms. In this study, the anticoccidial activity of SMC was investigated in Eimeria tenella-infected chickens compared to diclazuril as a widely used anticoccidial drug. In this regard, 14-day-old broilers were divided into six groups (n = 18). The first group (G1) was the healthy control group, while the second group (G2) was the non-infected SMC group treated at a dose of 50 mg/kg b.w. (high dose). Moreover, the third group (G3) was the positive control group (infected and non-treated). The fourth group (G4) was the infected group treated with SMC of 25 mg/kg b.w. (low dose), while the fifth group (G5) was the infected group treated with SMC of 50 mg/kg b.w. (high dose). Conversely, the sixth group (G6) was the diclazuril-treated group. The anticoccidial effects of SMC and diclazuril were evaluated by counting oocysts and recording the body weight gain, feed conversion ratio, clinical signs, lesions, and mortality rate. Interestingly, SMC showed potent anticoccidial activity, which was exemplified by reduction of oocyst count. Furthermore, the biochemical, antioxidant, and anti-inflammatory parameters in the cecal tissues were restored toward their control levels in G4, G5, and G6. Histopathological observation of cecal tissues was consistent with the aforementioned results revealing the ameliorative effect of SMC against E. tenella infection. This study concluded novel findings in relation to the anticoccidial role of SMC as a plant-based compound against the E. tenella-induced coccidiosis in broiler chickens combined with its antioxidative and anti-inflammatory properties. Further studies for exploring the mechanistic pathways involved in this activity and the potential benefits from its use in association with conventional anticoccidial drugs are warranted.

This study aims to assess the ability of vegetable oil of Pistacia lentiscus L. (lentiscus oil) in stimulating growth performance of broiler chickens and protecting them against coccidiosis. For this purpose, an in vitro test was first carried out to evaluate the destructive effect of this oil on Eimeria spp oocysts. On the other hand, an in vivo study was carried out to evaluate, once again, the capacity of the vegetable oil of Pistacia lentiscus L. in stimulating broilers growth performance and reducing the coccidiosis clinical signs. Thus, day old chicks were randomly divided into four equal groups: (1) uninfected and not-supplemented control (NI NS); (2) uninfected and supplemented (NI S); (3) infected and not supplemented (I NS); (4) infected and supplemented (IS). Each group was divided into three replicates containing each of them two subjects. The experimental groups (2 and 4) are supplemented with lentiscus oil by force-feeding at the rate of 1 mL per day from the 18th day until the end of the experiment. The chicks of the third and the fourth group are inoculated orally with sporulated oocysts (6.5 × 105 oocysts of Eimeria spp) on the day 20 of age. The results showed that lentiscus oil has an anticoccidial dose-dependent effect as shown by oocysts counting and released substances measurement at 273 nm. The growth performance of the (NI S) group was found better with an improvement percentage of 9.14% compared to the control (p < 0.05). Likewise, the weight gain of the (I S) group seems slightly higher than that of the control one (1316 g and 1235 g, respectively) (p < 0.05). In conclusion, the vegetable oil of Pistacia lentiscus L. has shown, not only, a promising growth effect in broiler chickens, but also, it seems to have a protective effect against coccidiosis sequels caused by Eimeria acervulina infection. There is a need to advance commercial poultry production to cater to the essential protein needs of an ever-increasing population, however, the rampant occurrence of coccidiosis infection poses a threat to this achievement. This study evaluated the in vivo anticoccidial activities of the extracts and fractions of Garcinia kola against experimental Eimeria tenella infection using broiler chickens as experimental subjects. A total of 40 broiler chicks were experimentally infected with E. tenella and assigned randomly into five groups consisting of eight chicks each. Three days post experimental infection groups I and II were administered orally with tween 80 (0.8%) and Amprolium (30 mg/kg) and served as untreated and treated control groups, respectively whereas Groups III, IV, and V were administered orally with crude methanol extract (CME) at doses of 200, 400 and 600 mg/kg, respectively, for five consecutive days. Daily weight gains were recorded and faecal oocysts per gram (OPG) counts were made by the McMaster Egg counting technique. Blood samples from each experimental group were collected on days 0, 3, 6, and 8 for haematological examination. In the acute toxicity studies, the CME of G. kola did not produce any toxic effect or mortality at doses between 10 and 5000 mg/kg. The CME G. kola was then considered safe and the LD50 was assumed to be > 5000 mg/kg. Graded doses of CME of G. kola considerably (P < 0.05) improved body weight gain and decreased OPG in a dose-depended manner. There was also significant improvement in the Packed Cell Volume (PCV), Red Blood Cell (RBC) and White Blood Cell (WBC) counts upon treatment with the graded doses of CME of G. kola. Besides, G. kola significantly decreased histopathological lesions in the caecum. The results of this study indicates that G. kola may provide beneficial effects against E. tenella-induced coccidiosis in broiler chickens. [ABSTRACT FROM AUTHOR]

Abstract Background Ponazuril is used for the treatment of equine protozoal myeloencephalitis (EPM). Coadministration of ponazuril with oil could result in higher serum and cerebrospinal fluid (CSF) concentrations of ponazuril. Hypothesis Coadministration of corn oil will result in higher serum and CSF concentrations of ponazuril than when ponazuril is administered alone. Animals Ten resident university‐owned adult horses of either sex and >2 years of age. Methods Cohort study. Ponazuril oral paste (5 mg/kg BW; ponazuril treatment group (PON); n = 5), or ponazuril oral paste (5 mg/kg BW; ponazuril and oil treatment group (PONOIL; n = 5) coadministered with 2 oz of corn oil q24h for 21 days. Horses were treated once daily, for 21 days. Blood was collected on days 0, 7, 14, and 21 before dosing. In addition, CSF was collected on days 1, 7, 14, and 21. The concentration of ponazuril was determined in serum and CSF and results compared using repeated measures ANOVA. Results Coadministration of ponazuril with 2 oz of corn oil resulted in higher concentrations of ponazuril in serum (at steady state) than that found in horses given ponazuril alone (6.2 ± 0.9 mg/L versus 4.5 ± 1.0 mg/L; P = .004) (mean ± 1 SD). Cerebrospinal fluid concentrations of ponazuril were also greater in horses that received ponazuril and oil (0.213 mg/L ± 0.04 versus 0.162 ± 0.04 mg/L) (P = .03). Conclusions and Clinical Importance Results suggest that coadministration of corn oil with ponazuril might enhance the effectiveness of treatment with ponazuril.

SUMMARY: Coccidia have typically been controlled using anticoccidial drugs, while the use of vaccines has increased due to concerns over the development of drug resistance in Eimeria spp. Because they contain drug-sensitive Eimeria, vaccination has received interest as a potential intervention for managing drug resistance. Vaccines can also be used in conjunction with anticoccidial drugs as part of a bio-shuttle program in which vaccination of broilers on day of hatch is followed by the administration of an ionophore in-feed starting at approximately 16 to 18 d post-hatch. Although they have been demonstrated to mitigate Eimeria vaccine-related performance losses, very little research on the effects of bio-shuttle programs on growth performance, pathology, and Eimeria populations has been published. In this study, we developed a longitudinal profile of the response of commercial broiler chickens before, during, and after bio-shuttle application as part of the broader drug and vaccine program of a commercial integrator over a 12-month period. Evaluation of production data collected from a commercial integrator found that, overall, growth performance and intestinal Eimeria pathology to be worst during the bio-shuttle administration period and best during the period after bio-shuttle administration. The correspondence between growth performance and pathology suggests that the development of drug-resistant Eimeria populations may have been reduced or delayed by bio-shuttle application. The results of our study are expected to inform future research evaluating the effects of bio-shuttle administration in poultry production.

Origanum vulgare subsp. hirtum, Thymus vulgaris, and Salvia fructicosa are aromatic plants commonly found in Mediterranean countries and are traditionally used in Greece as a remedy for humans, since they are well known as potent antibacterial, antioxidant, and anti-inflammatory agents. Essential oils (EOs) derived from plants cultivated in the mountainous region of Epirus, Greece, were investigated for their inhibitory activity against key microorganisms with relevance to avian health, while also assessing their antioxidant and anti-inflammatory activity. The total phenolic content (TPC) of the EOs was estimated according to the Folin–Ciocalteu method, while the antioxidant capacity was tested through the EOs’ ability to scavenge free radicals by means of the DPPH, ABTS, and FRAP assays. Antibacterial and anti-inflammatory effects were examined by the agar disc diffusion method and the lipoxygenase (LOX) inhibition test, respectively. Furthermore, the EOs’ ability to inhibit the invasion of sporozoites of Eimeria tenella (Wisconsin strain) along with any toxic effects were assayed in Madin–Darby bovine kidney (MDBK) cells. The antioxidant activity of the EOs was observed in descending order: oregano > thyme > sage. The antimicrobial effects of thyme and oregano were equivalent and higher than that of sage, while the anti-inflammatory effect of thyme was higher compared to both sage and oregano. The intracellular invasion of sporozoites was evaluated by the detection of E. tenella DNA by qPCR from cell monolayers harvested at 2 and 24 h post-infection. Parasite invasion was inhibited by the addition of oregano essential oil at the concentration of 100 μg/mL by 83% or 93% after 2 or 24 h, respectively, and was higher compared to the addition of thyme and sage, which had similar effects, but at a less intensive level. The cytotoxic assessment of all three essential oils revealed that they had no effect on MDBK cells compared to dimethyl sulfoxide (DMSO), used as the control substance. The supplementation of oregano, thyme, and sage essential oils had a potent antioxidant, anti-inflammatory, antimicrobial, and anticoccidial in vitro effect that is comparable to synthetic substances or approved drugs, justifying the need for further evaluation by in vivo studies in broilers reared in the absence of antimicrobial and anticoccidial drugs or synthetic antioxidant and/or anti-inflammatory compounds.

The impact of zinc oxide nanoparticles (ZnO-NPs) on the pathogenesis of coccidiosis in broiler chickens was tested. A total of 160 1-day-old broiler chicks (Ross 308) were randomly allocated into 4 groups (n = 40). Group 1: unchallenged, unmedicated; Group 2: challenged, unmedicated; Group 3: challenged, supplemented with diclazuril (1 ppm); Group 4: challenged, supplemented with ZnO-NPs (20 ppm). Mixed Eimeria species (E. maxima, E. acervulina, E. mivati, and E. tenella) of a commercial coccidial vaccine (FORTEGRA®) were used to perform the coccidial challenge by 15× of its vaccinal dose on the 14th day of age. Diclazuril and ZnO-NPs supplementation in Group 3 and 4, respectively, reduced the mortality rate due to coccidial challenge to 5.8% compared to 11.9% in Group 2. The growth performance was improved by ZnO-NPs in coccidiosis-infected group (p ≤ 0.05) compared to Group 2 and was comparable to that of Group 3 (p ≥ 0.05). The average oocyst count was lower in Groups 3 and 4 (7.8 × 103 and 14.3 × 103, respectively) than in Group 2 (67 × 103 oocysts). Group 3 had a decreased gross lesion score in duodenum and caecum (p ≤ 0.05) as well as jujenum and ileum (p ≥ 0.05) compared to Group 2; while the average lesion scores of all intestinal parts in Group 4 were significantly decreased (p ≤ 0.05). However, diclazuril was superior to ZnO-NPs in reducing caecal lesion score (p ≤ 0.05). Plasma carotenoids levels were increased by diclazuril (p ≥ 0.05) and ZnO-NPs (p ≤ 0.05) supplementation compared to Group 2. Oxidative stress appeared on the fourth week post-challenge (pc) in Group 2 (p ≤ 0.05) compared to Group 1, while the dietary supplementation with either diclazuril or ZnO-NPs numerically decreased Malondialdhyde (p ≥ 0.05) and statistically increased antioxidant activity (p ≤ 0.05). Both medications significantly improved the PCV%, Hb% and RBCs count on the 6th-day and 4th-week pc (p ≤ 0.05) compared to Group 2, though this improvement was higher significantly in Group 4 than Group 3 on the 6th day pc (p ≤ 0.05). Neither coccidial challenge nor medications had an impact on the total WBCs count as well as organ index, except Bursa of fabricious index that significantly improved by ZnO-NPs on the 4th-week pc compared to Group 2. Coccidial challenge reduced total protein and globulin levels and increased the serum alanine aminotransferase, serum cholesterol, and low-density lipoprotein levels (p ≤ 0.05) compared to Group 1, while those of both medicated groups (Group 3 and 4) were comparable to Group 1 (p ≥ 0.05). In conclusion, ZnO-NPs were found to be as effective as diclazuril against coccidiosis. However, further research is needed to fully comprehend its anticoccidial mechanisms.

Coccidiosis is a crucial parasitic disease of the poultry industry. As a result of the enormous global economic losses and the increased resistance to the conventional anticoccidial agents, there is a continuous need to find new anticoccidials. Here, the anticoccidial effect of the fluoroquinolone lomefloxacin versus diclazuril in experimentally infected broilers was tested for the treatment of Eimeria tenella infection. Ninety 14-day-old Cobb strain broiler chickens were allocated into five groups, each with 18 chicks. Group 1 (G1) was separated as an uninfected negative control and received no treatment; group 2 (G2), infected untreated (positive control); group 3 (G3), infected and treated with lomefloxacin at a dose rate of 100 ppm in drinking water; group 4 (G4), infected and treated with diclazuril at a dose rate of 2.5 ppm in drinking water; group 5 (G5), infected and treated with lomefloxacin at a dose rate of 100 ppm plus diclazuril at dose rate of 2.5 ppm in drinking water. Clinical signs, mortality rates, number of oocysts per gram of faeces (OPG), growth performance parameters (weight gain: WG and feed conversion ratio: FCR), lesion scoring, haematological and serum biochemical analyses, antioxidant biomarkers and histopathologic inspection of the caeca were used as evaluation criteria for the anticoccidial efficacy of both lomefloxacin and diclazuril. The findings herein showed that administration of lomefloxacin and/or diclazuril improved growth performance parameters (WG, FCR) and significantly (P ≤ 0.05) reduced OPG, and diminished the severity of bloody diarrhoea and mortalities. Additionally, haematological indices and serum biochemical parameters such as ALT, AST, ALP, creatinine, uric acid, total proteins, albumin and globulin were improved. Finally, a significant elevation in the levels of the antioxidant biomarkers was observed in the chicks of G3, G4 and G5 as compared with those of G2. [ABSTRACT FROM AUTHOR]

Coccidiosis, a parasitic infection caused by one or more of the numerous species of Eimeria is recognised as the disease that has severe economic impact on poultry production. In an experiment to determine the effect of vaccination on the infection with a Local isolate and Houghton strain of Eimeria tenella (E. tenella). Ninety (90) day-old Dominant black cockerel chicks of D109 strain were randomly divided into nine groups (A–I). Two commercially available anti-coccidial vaccines—Livacox® (Biopharm, Czech Republic) and Immucox® (Vetech, Ontario, Canada) were used to immunize the chicks. Immunization was done at 5 days-old by oral gavage and infection was carried out with 1.7 × 104E. tenella of either Houghton strain or a field (Local) isolate at 4-weeks old. Six (6) days post-challenge, generalized pallor, moderate bloody diarrhea, slight reduction in feed intake and weight gain characterized the infected groups. There is a significant difference (p < 0.05) in packed cell volume of group B, unvaccinated and infected with Houghton strain of E. tenella and group E vaccinated with Livacox® and infected with Houghton strain. The Immunogenicity of Livacox® and Immucox® was determined primarily on the ability of the vaccinated chickens to overcome the effect of the virulent challenge by E. tenella including blood loss, reduction in feed intake and feed conversion and weight loss. Background: Ponazuril is used for the treatment of equine protozoal myeloencephalitis (EPM). Coadministration of ponazuril with oil could result in higher serum and cerebrospinal fluid (CSF) concentrations of ponazuril. Hypothesis: Coadministration of corn oil will result in higher serum and CSF concentrations of ponazuril than when ponazuril is administered alone. Animals Ten resident university‐owned adult horses of either sex and >2 years of age. Methods: Cohort study. Ponazuril oral paste (5 mg/kg BW; ponazuril treatment group (PON); n = 5), or ponazuril oral paste (5 mg/kg BW; ponazuril and oil treatment group (PONOIL; n = 5) coadministered with 2 oz of corn oil q24h for 21 days. Horses were treated once daily, for 21 days. Blood was collected on days 0, 7, 14, and 21 before dosing. In addition, CSF was collected on days 1, 7, 14, and 21. The concentration of ponazuril was determined in serum and CSF and results compared using repeated measures ANOVA. Results: Coadministration of ponazuril with 2 oz of corn oil resulted in higher concentrations of ponazuril in serum (at steady state) than that found in horses given ponazuril alone (6.2 ± 0.9 mg/L versus 4.5 ± 1.0 mg/L; P =.004) (mean ± 1 SD). Cerebrospinal fluid concentrations of ponazuril were also greater in horses that received ponazuril and oil (0.213 mg/L ± 0.04 versus 0.162 ± 0.04 mg/L) (P =.03). Graded concentrations (200, 400 and 800 mg/kg) of the aqueous stem bark extract of Khaya senegalensis was evaluated for its therapeutic efficacy against experimentally induced coccidiosis in broiler chicken. The phytochemical analysis shows the presence of tannins, saponins, cardiac glycosides and steroids. There was significant reduction in oocyst count across the groups in a graded dose manner with 800 mg/kg being the most efficacious dose. There was also weight gain across the treatment groups with immuno-modulatory and erythropoetic activities observed. Also, a significant (p < 0.05) graded dose-dependent reduction in the oocyst count in the treatment groups. A significant (p < 0.05) increase in mean weight gain was also recorded across the experimental groups except the negative control. The haematology also showed a dose-dependent increase in red blood cells, haemoglobin and packed cell volume of the treatment groups. The extract had no significant difference (p > 0.05) on the white blood cells, but a slight decrease in the white blood cells and heterophil counts was observed at 400 mg/kg. Furthermore, the aspartate amino transaminase level showed a significant difference (p < 0.05). Fluctuating levels of other serum biochemical parameters such as total protein, albumin and potassium were observed. No significant difference (p > 0.05) in the sodium concentration was observed. In addition, oxidative stress biomarkers such as catalase significantly increased (p < 0.05) in all the experimental groups in addition to the concomitant increase in reduced gluthathione (GSH) and superoxide dismutase (SOD) levels. Coccidia have typically been controlled using anticoccidial drugs, while the use of vaccines has increased due to concerns over the development of drug resistance in Eimeria spp. Because they contain drug-sensitive Eimeria , vaccination has received interest as a potential intervention for managing drug resistance. Vaccines can also be used in conjunction with anticoccidial drugs as part of a bio-shuttle program in which vaccination of broilers on day of hatch is followed by the administration of an ionophore in-feed starting at approximately 16 to 18 d post-hatch. Although they have been demonstrated to mitigate Eimeria vaccine-related performance losses, very little research on the effects of bio-shuttle programs on growth performance, pathology, and Eimeria populations has been published. In this study, we developed a longitudinal profile of the response of commercial broiler chickens before, during, and after bio-shuttle application as part of the broader drug and vaccine program of a commercial integrator over a 12-month period. Evaluation of production data collected from a commercial integrator found that, overall, growth performance and intestinal Eimeria pathology to be worst during the bio-shuttle administration period and best during the period after bio-shuttle administration. The correspondence between growth performance and pathology suggests that the development of drug-resistant Eimeria populations may have been reduced or delayed by bio-shuttle application. Nicarbazin is an anticoccidial used in poultry production. Its residue of concern left in chicken meat is 4,4′-dinitrocarbanilide (DNC) whose content can be reduced through meat thermal processing. Thermal analysis techniques can be considered to elucidate how DNC degradation proceeds. This study aimed to investigate the thermal behavior of DNC using TG–DSC–MS and Py–GC–MS. TG–DSC–MS data showed that DNC decomposition occurs in a single step (252–280 °C) as an endothermic event (ΔH of 670 J g−1). Fragments detected at m/z 43, 44 and 65 indicated the release of isocyanic acid, carbon dioxide and 4-nitroaniline as breakdown products. In addition, Py–GC–MS analysis confirmed that DNC is thermally degraded above 250 °C generating 4-nitrophenyl isocyanate and 4-nitroaniline as the major pyrolysis products. Findings obtained from thermal analysis herein are useful to better comprehend the DNC disappearance after submitting chicken meat to heat processing. [ABSTRACT FROM AUTHOR]

Nanomedicine is a critical therapeutic approach for treating most poultry illnesses, particularly parasitic infections. Coccidiosis is a severe protozoan infection affecting poultry; the emergence of drug-resistant Eimeria strains demands the development of new, safe therapies. Consequently, the objective of this work was to investigate the efficacy of the biosynthesized selenium nanoparticles (SeNPs) by Paenibacillus polymyxa ( P. polymyxa ) against Eimeria tenella ( E. tenella ) experimental infection in broiler chickens. The prepared SeNPs absorbed the UV at 270 nm were spherical with a size of 26 nm, and had a surface negative charge of −25 mV. One hundred and fifty, 1-day-old male broiler chicks were randomly allocated into 5 groups (30 birds/group with triplicates each) as follows: T1: negative control (noninfected and nontreated with SeNPs); T2: delivered SeNPs (500 µg/kg diet) for 35 successive days, T3: E. tenella -infected (positive control birds), T4: E. tenella -infected and treated with SeNPs (500 µg/kg diet) and T5: E. tenella -infected chicks and treated with anticoccidial agent (sulfadimidine, 16% solution 8 mL/L of drinking water) for 5 successive days. At 14 d of age, each bird in infected groups was orally treated with 3 × 103 sporulated oocyst of E. tenella. SeNPs considerably decreased the number of oocysts in broiler feces compared to positive control and anticoccidial drug, followed by a substantial reduction of parasite phase count in the cecum (15, 10, and 8 for meronts, gamonts, and developing oocysts) when compared with positive control birds. The Eimeria experimental infection lowered the activity of antioxidant enzymes, superoxide dismutase (SOD), glutathione peroxidase (GPx), and reduced glutathione (GSH) while increasing the stress parameters nitric oxide (NO) and malonaldehyde (MDA). Moreover, the production of proinflammatory (TNF-α and IL-6) and apoptotic genes (BcL2 and Cas-3) were significantly elevated. Administrating SeNPs to chicks significantly decreased oxidative stress, inflammation, and apoptotic markers in the cecum tissue. Therefore, growth performance, carcass weights, antioxidant enzymes, and blood properties of infected chicks were enhanced. For decades antibiotics have been used in poultry rearing to support high levels of production. Nevertheless, several problems have arisen because of the misuse of antibiotics (i.e., antibiotic resistance, residues in animal products, environmental pollution). Thus, the European Union (EU) as well as the European Food Safety Authority (EFSA) promote action plans to diminish the use of antibiotics in animal production. Alternatives to antibiotics have been studied. Polyphenols (PPs) or organic acids (OAs) seem to be two accredited solutions. Phenolic compounds, such as phenols, flavonoids, and tannins exert their antimicrobial effect with specific mechanisms. In contrast, short chain fatty acids (SCFAs) and medium chain fatty acids (MCFAs), the OAs mainly used as antibiotics alternative, act on the pathogens depending on the pKa value. This review aims to collect the literature reporting the effects of these substances applied as antimicrobial molecules or growth promoter in poultry feeding (both for broilers and laying hens). Organic acids and PPs can be used individually or in blends, exploiting the properties of each component. Collected data highlighted that further research needs to focus on OAs in laying hens’ feeding and also determine the right combination in blends with PPs.

The present study was conducted on broiler chickens ( at day 21 age ) which were infected by oral administration with Eimeria tenella oocyst suspension containing (1 × 10 5) oocyst / chick . Broiler chickens were divided Randomly into nine groups (10 chickens per each group) and were watched during the first and second weeks after infection The results revealed that the infected chickens clinically showed dullness and Anorexia . Microscopically , There were haemorrhagic inflammation , haemorrhagic spots on the intestine and in addition to congestion and enlargement of the caecal tonsils . The study was also included the supplementation of broiler chicken by methionine , choline and anticoccidials (Amprolium and Diclazuril) at day 9 post infection and it continues for 12 days .The results revealed that no significant differences in plasma glucose , cholesterol , triglyceride and high density lipoprotein level were observed whereas , significant decrease in the level of uric acid , aspartate aminotransferase (AST) activity and alanine aminotransferase (ALT) activity and also significant increase in the level of plasma protein and albumin were evident in treating groups as compared with control . This may be due to high total protein concentration in the blood serum as there is evidence to suggest the existence of inverse proportion between the total protein concentration and activity of enzymes GOT and GPT in serum and may result in adding amino acids to the diet to reduce the body's need to create energy from sources protein and then reduce the activity of enzymes (ALT) and (AST).

Coccidiosis is a deadly disease that hampers chicken's productivity and welfare. Thus, the disease is a major menace to the global poultry industry. Coccidiosis which is caused by the apicomplexan parasite of the genus Eimeria has seven known species which affect the different parts of the intestinal tract of chickens. The disease which occurs by ingestion of sporulated oocyst has been associated with poor poultry management system. Mixed infection among the species of this parasite contributes to both pathogenicity and misdiagnosis of the disease. A progress in identification and diagnosis approach which cuts across pathological, morphological and molecular has been reported for this parasite. Control measures which include anticoccidial drugs, vaccines and natural products have dominated literature for this disease. However, the emergence of genetic and antigenic diversity with implication on resistance to anticoccidials among different strains of Eimeria parasite has generated concerns on the effectiveness of the current anticoccidial vaccines. A new look on the control strategy therefore becomes imperative. Coccidiosis is a self-limiting disease that is universally present in poultry operations, causing extensive damage to the intestinal lining of the bird. Global economic losses from coccidiosis are estimated to be $3 billion per year. In-feed anticoccidial use has been the predominant form of coccidiosis control. However, due to widespread emergence of antimicrobial resistance, concerns have been raised regarding the safety of anticoccidials and the potential impact on human, animal, and environmental health. To investigate the benefits, risks, and alternatives to anticoccidial use, a comprehensive review of recent literature was conducted. Several live vaccines are available, which, when used in combination with anticoccidials, have been shown to help restore sensitivity of infective parasites. However, their use has been limited because of increased cost; increased susceptibility to bacterial enteritis; challenges with consistent application; and slow development of immunity. Various alternative feed products are available, but do not have a direct anticoccidial effect, and few studies have demonstrated consistent field efficacy of these products. Consumer and environmental safety of anticoccidials is monitored and assessed by governing bodies. Furthermore, there is a lack of current evidence to indicate that bacterial resistance poses a public health concern. The findings from this review indicate that in the absence of alternatives, poultry production is optimized by using anticoccidials, benefiting all three pillars of sustainability, including social (bird health, welfare, and food safety), economic (production efficiency), and environmental aspects. Eimeria species parasites can cause the disease coccidiosis, most notably in chickens. The occurrence of coccidiosis is currently controlled through a combination of good husbandry, chemoprophylaxis and/or live parasite vaccination; however, scalable, cost-effective subunit or recombinant vaccines are required. Many antigens have been proposed for use in novel anticoccidial vaccines, supported by the capacity to reduce disease severity or parasite replication, increase body weight gain in the face of challenge or improve feed conversion under experimental conditions, but none has reached commercial development. Nonetheless, the protection against challenge induced by some antigens has been within the lower range described for the ionophores against susceptible isolates or current live vaccines prior to oocyst recycling. With such levels of efficacy it may be that combinations of anticoccidial antigens already described are sufficient for development as novel multi-valent vaccines, pending identification of optimal delivery systems. Selection of the best antigens to be included in such vaccines can be informed by knowledge defining the natural occurrence of specific antigenic diversity, with relevance to the risk of immediate vaccine breakthrough, and the rate at which parasite genomes can evolve new diversity. This study evaluated the efficacy and the economic viability of two anticoccidial treatment regimens tested in lambs naturally exposed to Eimeria spp. re-infections in a grazing system during a 140-day period. Twenty-four suckling lambs were distributed into three groups based on the individual count of oocysts per gram of feces (OPG) and body weight. Animals were treated with toltrazuril 5% (20 mg/kg) at 14- (GI) or 21-day (GII) intervals, and GIII was kept as untreated control. A cost-benefit analysis of each treatment regimen was calculated. Additionally, economic analysis was performed on four hypothetical scenarios, in which lambs could be having 10, 25, 50, or 85% decrease in their expected body weight gain due to clinical. Efficacy of toltrazuril against Eimeria spp. was 96.9-99.9% (GI) and 74.2-99.9% (GII). E. ovinoidalis was most frequently identified, but no clinical signs of coccidiosis were observed in lambs. There were no differences in weight gain among the groups. The cost of treatment per lamb was $13.09 (GI) and $7.83 (GII). The estimation model showed that the cost-benefit ratio favored treatment with toltrazuril when lambs fail to gain weight. In the studied flock, the break-even point for toltrazuril administered at 14-day intervals was reached with 85% decrease in mean weight gain. In conclusion, toltrazuril can be used at 14-day intervals to control Eimeria spp. (re)-infection in lambs raised on pasture. Recently, use of botanicals as an alternative to anticoccidial drugs has been appealing approach for controlling avian coccidiosis. Therefore, this study was conducted to evaluate the anticoccidial activity of aqueous methanolic extract (100, 200 and 300 mg/kg of body weight) of Beta vulgaris (roots) in broiler chicks. A total of 315 day old broiler chicks were divided into seven equal groups (A, B, C, D, E, F and G). At 14th day of age, all groups except group G, which served as non infected non medicated control, were infected orally with 60,000 sporulated oocysts of mixed Eimeria species. At the same day, groups A, B and C were treated with graded oral doses of B. vulgaris aqueous methanolic extract (100, 200 and 300 mg/kg of body weight, respectively). Group D was treated with Vitamin-E, group E served as infected medicated control group (Baycox ® treated) and group F served as infected non medicated control group (PBS treated). Treatment with extract, reference drug Baycox ® , Vitamin E and PBS was continued for three consecutive days (14–16 days of age). Though, not at par with reference drug (Baycox ® ), B. vulgaris demonstrated good anticoccidial activity adjudged based on considered criteria, i.e., feed conversion ratio, lesion score, oocyst score and oocysts per gram of feces. In the United States, anticoccidials belonging to the ionophore class are considered antibiotics and cannot be used in marketing programs such as "No Antibiotics Ever" (NAE) or "Raised Without Antibiotics" (RWA). Therefore, as a growing percentage of broiler production becomes NAE/RWA, producers must learn to effectively prevent coccidiosis without the ionophore anticoccidials that have been the backbone of coccidiosis prevention programs worldwide for many decades. This requires that veterinarians and producers learn to effectively prevent and control coccidiosis with live coccidiosis vaccines, chemically-synthetized anticoccidials, and innovative techniques. For NAE/RWA production, synthetic anticoccidials and live coccidiosis vaccines can be used to prevent coccidiosis. For organic production, only live coccidiosis vaccines can be used. To make the vaccine programs work, all factors critical to achieving the development of active immunity, from vaccine storage, administration, and management in the field, must be optimized. Producers should utilize all of the tools at their disposal by using carefully planned programs and rotations along with improved brooding practices. The effect of Artemisia annua ethanolic extract (AE) as a potential source of herbal anticoccidial activity was investigated on experimental coccidiosis in chicken. One hundred ninety-two one-day-old chicks were divided in to 8 groups ( n = 24) including AE prevention group, AE-treated group, simultaneously challenged AE-medicated group, challenged-untreated group (positive control), unchallenged-untreated group (negative control), salinomycine prevention group, salinomycine-treated group, and simultaneously challenged salinomycine-medicated group, in a completely randomized design. Oral challenge carried out by a suspension containing a mixture of 200,000 oocysts Eimeria acervulina, 30,000 oocysts Eimeria necatrix, and 20,000 oocysts Eimeria tenella on day 21 of age. Weight gain in AE prevention group significantly increased compared to positive control group ( p < 0.05). Unlike salinomycine prevention group, the food conversion ratio (FCR) of AE prevention group was not significantly higher than negative control. Oocyst per gram (OPG) in simultaneously challenged AE-medicated group had no significant difference, while for 38% of the days, in simultaneously challenged salinomycine-medicated group significantly decreased ( p < 0.05). The food intake of AE-treated group had no significant difference with salinomycine-treated group ( p > 0.05). In half of the days of OPGs sampling, AE-treated group was reduced significantly compared to positive control group ( p < 0.05). Collectively, the in vivo study of anticoccidial effects of AE in the prevention section was more effective than the treatment section, while the treatment section was more effective than the simultaneous section. Anticoccidial effects of Aloe secundiflora crude leaf extract was tested in broiler chickens following oral infection with Eimeria tenella. Sixty 22-day-old birds were divided into six groups of ten birds each. Three treatment groups A, B, and C were fed with the extract (100, 250, and 500 mg/day, respectively) mixed in feed for 10 days, and three control groups: group D (drug control) administered 300 mg/l of sulfachloropyrazine sodium soluble powder in drinking water for 5 days, group E (infected/non-medicated positive control), and group F (uninfected/non-medicated negative control). Except for group F, all groups were orally inoculated with 75,000 sporulated oocysts of E. tenella. The effects of the extract on E. tenella infection were evaluated by severity of bloody diarrhea, body weight (BW) gain, oocyst output, and lesion score. No bird in the treated groups died of coccidiosis, and severity of bloody diarrhea was milder than in the positive control group. BW gains in the treated groups were significantly higher than in group E ( p < 0.05). The lesion scores of the treated groups were significantly lower than that of group E. Oocyst output in groups A, B, and C were 11.23, 8.24, and 6.82 × 10, respectively. As compared with the negative control group (12.84 × 10), the reductions in oocyst production were 12.54, 35.83, and 46.88%, respectively. Oocyst output significantly reduced with an increase in Aloe dosage. The findings of this study suggest that Aloe secundiflora extract presents an alternative anticoccidial agent for the control of avian coccidiosis. [ABSTRACT FROM AUTHOR]
