The maize female gametophyte contains four cell types: two synergids, an egg cell, a central cell, and a variable number of antipodal cells. In maize, these cells are produced after three rounds of free-nuclear divisions followed by cellularization, differentiation, and proliferation of the antipodal cells. Cellularization of the eight-nucleate syncytium produces seven cells with two polar nuclei in the central cell. Nuclear localization is tightly controlled in the embryo sac. This leads to precise allocation of the nuclei into the cells upon cellularization. Nuclear positioning within the syncytium is highly correlated with their identity after cellularization. Two mutants are described with extra polar nuclei, abnormal antipodal cell morphology, and reduced antipodal cell number, as well as frequent loss of antipodal cell marker expression. Mutations in one of these genes, indeterminate gametophyte2 encoding a MICROTUBULE ASSOCIATED PROTEIN65-3 homolog, shows a requirement for MAP65-3 in cellularization of the syncytial embryo sac as well as for normal seed development. Simple Summary: The present work is devoted to the features of antipodal cells at the early stages of wheat seed development, ensuring the formation and protection of a full-fledged grain. Using the methods of cell and molecular biology, changes in the structure of these cells and their components during active functioning and death are shown. For the first time, data on the expression of some genes confirming the functions of antipodal cells were obtained. The characteristic features of cells at the stages of death were revealed. The data obtained indicate the key role of antipodal cells in the process of grain maturation and subsequently allow us to identify substances that ensure the formation of tissue, which is the food of the whole world. The ultrastructure of antipodal cells of the Triticum aestivum embryo sac was studied at different stages of differentiation and programmed cell death. We studied in detail the nuclei of cells of the antipodal complex of wheat embryo sac (Triticum aestivum L.) during programmed cell death (PCD). The antipodal complex has been reported to be formed before double fertilisation of the embryo sac. Polyploidisation leads to the formation of giant polytene chromosomes in the nuclei of antipodal cells. These chromosomes are involved in secretory functions and are important for the development of cellular endosperm. Terminal deoxynucleotidyl transferase dUTP nick end labelling assay and immunodetection revealed DNA breaks in the nuclei and release of cytochrome c from mitochondria into the cytoplasm of antipodal cells during PCD. We used transmission electron microscopy, immunodetection and histochemistry to analyse the characteristic structural changes in the nuclei of antipodal cells during PCD. These included sequential structural changes in the nuclei containing polytene chromosomes, segregation of some components of the nucleolus into the bodies of polytene chromosomes, extrusion of nucleolar components and parts of chromosomes into the cytoplasm of antipodal cells and then into the endosperm coenocyte. This article describes the development of female gametophyte in Sedum rupestre L. New embryological information about the processes of megasporogenesis and megagametogenesis provided in this paper expand the current knowledge about the embryology of the studied species. S. rupestre is characterized by monosporic megasporogenesis and the formation of Polygonum–type embryo sac. The process of megasporogenesis is initiated by one megaspore mother cell, resulting in the formation of a triad of cells after meiosis and cytokinesis. The functional megaspore, which is located chalazally, is a mononuclear cell present next to the megaspore in the centre of the triad. Only one of the two non-functional cells of the triad is binucleate, which occur at the micropylar pole. In this paper, we explain the functional ultrastructure of the female gametophytic cells in S. rupestre. Initially, the cytoplasm of the gametophytic cells does not differ from each other; however, during differentiation, the cells reveal different morphologies. The antipodals and the synergids gradually become organelle-rich and metabolically active. The antipodal cells participate in the absorption and transport of nutrients from the nucellar cells towards the megagametophyte. Their ultrastructure shows the presence of plasmodesmata with electron-dense material, which is characteristic of Crassulaceae, and wall ingrowths in the outer walls. The ultrastructure of synergid cells is characterized by the presence of filiform apparatus and cytoplasm with active dictyosomes, abundant profiles of endoplasmic reticulum and numerous vesicles, which agrees with their main function—the secretion of pollen tube attractants. Bisporic embryo sacs contain nuclei derived from two members of a megaspore tetrad whereas tetrasporic embryo sacs contain nuclei derived from all four members of a tetrad. Megaspores from the same tetrad are less genetically similar than are megaspores from different tetrads. Therefore, cells derived from different megaspores in bisporic and tetrasporic embryo sacs are expected to compete with each other rather than cooperate. The tacit assumption of the plant embryological literature that bisporic and tetrasporic embryo sacs function as integrated organisms is demonstrably false, both theoretically and empirically. This competition within embryo sacs can be expressed as the formation of eggs by descendants of more than one megaspore or by the suppression of the descendants of all but one megaspore. Both phenomena have evolved multiple times. In contrast, all nuclei of monosporic female gametophytes are genetically identical. Consequently, monosporic development is predicted to be more evolutionarily stable than bisporic or tetrasporic development. The triploid endosperm produced by most monosporic gametophytes was probably derived from an ancestrally diploid endosperm and has been considered a key adaptation of the most successful lineages of flowering plants. The greater weight given to the doubled maternal genome in triploid endosperm may have facilitated a more efficient distribution of resources among a mother’s seeds because it reduced the influence of competition for maternal resources among unrelated paternal genomes of endosperm.

Key message: The central cell withstands heat stress better than the egg and antipodal cells. Insilco analysis of transcriptomic data identified several heat responsive genes which are central cell specific. Crop damage due to heat stress (HS) is a major cause of yield lost. Plants are particularly susceptible to negative effects of HS during gametophyte development and fertilization. Extensive studies have been performed on the male gametophyte under HS, but how the female gametophyte copes with HS is largely unknown. To learn how the different cell types of the female gametophyte reacts to HS, we studied unfertilized CDC123::H2B:YFP ovules. We found that the YFP-specific florescent signal persisted in the central cell during HS significantly more than the egg cell. We also found that the fluorescent signal persistence was the lowest in the antipodal cells. This finding suggests that the reaction of the female gametophyte to HS is rather unique and differentially mediated according to the cell's identity. In addition, mining through published transcriptomic datasets we found that several important heat stress responsive genes which are extremely upregulated during HS (more than 64-fold) are specifically expressed in the CC but not in the EC. Further research such as comparative transcriptomics and cell biology will likely shed more light on the phenomena reported here and increase our basic understandings about the ways sexual reproduction processes are affected by heat stress. A detailed study into the structural features of the multilevel antipodal complex of wheat Triticum aestivum L. embryo sac was performed at different stages of the complex's differentiation after double fertilization. The heterogeneity of nuclei ploidy in individual antipodal complexes caused by the asynchrony of the endoreduplication rounds of the nuclear DNA was revealed. The nuclei ploidy of basal, middle, and apical layers of the complexes was measured at the early, middle, and late stages of differentiation. At the early stage of differentiation, the nuclei ploidy of the antipodal complex's basal layer adjacent to the chalasal region of the nucellus of the embryo sac reaches 13 C, the nuclei of the apical layer cells that contact the endosperm syncytium reaches 63 C, and the nuclei of the middle layer located between the basal and apical layers reach 30 C. At the middle stage of differentiation, the nuclei ploidy in the basal layer increases to 17 C. The nuclei ploidy of the apical layer cells increases to 95 C, and nuclei ploidy of the middle layer increases to 45 C. At the stage of late differentiation, the nuclei ploidy in the basal layer increases to 24 C; the apical layer ploidy increases to 215 C; the middle layer ploidy increases to 63 C. Changes in the shape and structure of the nuclei during differentiation were revealed. They manifest themselves in heterogeneity in shape, size and structure of chromatin; the formation of individual polytene chromosomes; nuclear membrane invaginations; and the variation in the number of nucleoli. Data on the distribution and structure of cytoplasmic organelles of the antipodal cells, endoplasmic reticulum, dictyosomes, mitochondria, and microtubules at different stages of differentiation of the antipodal complexes are fundamentally new. The increased number of cytoplasmic organelles was revealed. During the differentiation, prolong cisterns of the granular reticulum are replaced by concentric rings, mitochondria and plastids of extended and cupped shape appear, and the microtubule network is rebuilt. The features of the antipodal complex's cell structure may reflect changes in the functions of the antipodal complex during the differentiation. At the early stage, all cells of the complex perform an osmoregulatory function, and cells of different layers of the complex specialize at the middle stage of differentiation. The ploidy level of cell nuclei with polytene chromosomes reflects their functional significance in the formation of endosperm at the nuclear stage of development, and, subsequently, of normal full-fledged grain. Available documentation about the development of the female gametophyte of Crassulaceae is very limited. The aim of this study was to extend the embryological knowledge of Crassulaceae by analysing the development of the embryo sac in Sedum sediforme. Transmission electron microscopy and light microscopy including Nomarski optics (DIC) were used to observe individual stages of female gametophyte development. Cytochemical staining enabled detection of lipids, insoluble polysaccharides and proteins in gametophyte cells during their formation. Their increased accumulation was observed during nucellar cell and unfunctional cell degeneration in the embryo sac at the coenocytic and cellular stages (megagametogenesis). The female gametophyte develops in anatropous, bitegmic and crassinucellate ovules. The mature embryo sac is built of seven cells but after antipodes degeneration it is formed by the egg apparatus and a central cell. The monosporic Polygonum type was observed. One megaspore mother cell (MMC) formed three cells after meiosis. A triad was formed from a functional megaspore (placed chalazally), one uninucleate megaspore and a binucleate cell located at the micropylar end. Plasmodesmata with adhering electron-dense dome were noticed in walls of the coenocytic embryo sac and in the outer walls of ephemeral antipodes. Moreover, similar to synergids, antipodes form wall ingrowths. Here, we report new structural features of the antipodal cells (the presence of plasmodesmata with an electron-dense dome) which have not been described before. Fertilization is accompanied by the targeted degradation of gamete-adjoining accessory cells, and here recent advances in our understanding of the molecular mechanisms underlying their demise are reviewed.Plant development and growth is sustained by the constant generation of tremendous amounts of cells, which become integrated into various types of tissues and organs. What is all too often overlooked is that this thriving life also requires the targeted degeneration of selected cells, which undergo cell death according to genetically encoded programmes or environmental stimuli. The side-by-side existence of generation and demise is particularly evident in the haploid phase of the flowering plants cycle. Here, the lifespan of terminally differentiated accessory cells contrasts with that of germ cells, which by definition live on to form the next generation. In fact, with research in recent years it is becoming increasingly clear that the gametophytes of flowering plants constitute an attractive and powerful system for investigating the molecular mechanisms underlying selective cell death. Morphological changes in the nuclear degeneration of the synergid (mainly the synergid that receives the pollen tube) and antipodal cells inTriticum aestivumwere studied. Although located in the same embryo sac, and derived from the same megaspore, nuclear degeneration of the synergid and antipodal cells differs greatly. Nuclear degeneration in the synergid is characterized by pycnosis, i.e., total chromatin condensation, nuclear deformation and distinct shrinkage in volume, followed by the formation of an irregular and densely stained mass-the degenerated nucleus-while the nucleolus disappears prior to the degradation of chromatin. In contrast, in the nuclear degeneration of antipodal cells, chromatin is only partly condensed and the nuclear volume changes only slightly after the distinct chromatin condensation. Chromatolysis then occurs, i.e., stainable contents disappear while the nuclear envelope is retained. The nucleoli persist after the disappearance of the chromatin. The possible functions of nuclear degeneration of synergid and antipodal cells are discussed, especially with respect to the guidance of pollen tube growth and the proliferation of free-nuclear endosperm. The antipodal cells have been the stepchildren in most investigations of the female gametophyte. In Hordeum vulgare cv. Bomi, three antipodal cells are originally developed chalazally but because of differential growth of the embryo sac they soon become laterally situated and their number increases to 35-50 cells and the shape, size and structure of the cells change in the time before as well as after fertilization. The cells persist until about 60-70 h after pollination. At that time, the embryo consists of about 12-15 cells and a cellularization of the nuclear endosperm has started peripherally. The size of nuclei, and especially nucleoli, in the antipodal cells increases tremendously in the investigated period and the amounts of organelles also change. The walls of antipodals are diversified depending on which cells they are separating, and wall invaginations are developed especially between antipodal cells and surrounding nucellar cells in the placental region. The mitochondria appear in various shapes in section view, very often as cups or dumbbells with a rim in the ends containing cristae and a thin cristae-free base. These bases are sometimes stretched out as thin parts and at last a simple parting occurs. Binary fissions of the plastids happen especially in the hours before and just after egg fertilization. ER is extraordinarily well developed in the whole period of investigation and many ribosomes are attached to the membranes. Dictyosomes form numerous vesicles, especially in the antipodals near the nucellar cells in the placental region.
