The use of low-cost sensors, with open-source code, facilitates greater spatial resolution and flexibility of environmental monitoring, thus generating more information and overcoming limitations of traditional commercial sensors. Measurement of water turbidity using submerged sensors can be problematic in that rapid biofouling requires frequent site visits to remove, clean, calibrate and replace the sensor. We therefore designed an automated system using low-cost commercially-available sensors that pumps water from the stream, samples it for turbidity and purges remaining water, leaving the turbidity sensor dry between measurements, thus greatly reducing the biofouling problem and minimizing operation costs. Our station was able to estimate suspended sediment concentrations between 0 and 6 g/L with a root mean square error (RMSE) around 5 % of the total range, which meets typical research and operational study requirements. The results showed that the monitoring station is capable of monitoring water level and turbidity for long periods without the need of cleaning the turbidity sensor, due to its purge function. We demonstrated that spatially intense measurement of turbidity within catchments and drainage networks can be achieved at a relatively low cost, which allows a better understanding of the main sources of suspended sediments and their spatial and temporal variability.

Wastewater-based surveillance is emerging as an important tool for the COVID-19 pandemic trending. Current methods of wastewater collection, such as grab and auto-composite sampling, have drawbacks that impede effective surveillance, especially from small catchments with limited accessibility. Passive samplers, which are more cost-effective and require fewer resources to process, are promising candidates for monitoring wastewater for SARS-CoV-2. Here, we compared traditional auto sampling with passive sampling for SARS-CoV-2 detection in wastewater. A torpedo-style 3D-printed passive sampler device containing both cotton swabs and electronegative filter membranes was used. Between April and June 2021, fifteen passive samplers were placed at a local hospital's wastewater outflow alongside an autosampler. Reverse transcription and quantitative polymerase chain reaction (RT-qPCR) was used to detect SARS-CoV-2 in the samples after processing and RNA extraction. The swab and membrane of the passive sampler showed similar detection rates and cycle threshold (Ct) values for SARS-CoV-2 RNA for the N1 and N2 gene targets. The passive method performed as well as the grab/auto sampling, with no significant differences between N1 and N2 Ct values. There were discrepant results on two days with negative grab/auto samples and positive passive samples, which might be related to the longer duration of passive sampling in the study. Overall, the passive sampler was rapid, reliable, and cost-effective, and could be used as an alternative sampling method for the detection of SARS-CoV-2 in wastewater. [ABSTRACT FROM AUTHOR]

Microfluidic autosamplers for electrospray ionization mass spectrometry (ESI-MS) are of major importance when using ESI-MS as a high-throughput and low sample consumption analytical method. In this article, microfluidic ESI-MS autosampler designs are overviewed and a group-owned prototype is discussed. The socalled gap sampler is a pin-based sampler for miniaturized flow injection (FI) analysis. To date, it has been used in various applications. Following proof of concept applications with FI of small molecules, pin modifications were implemented for unspecific and specific extraction for the analysis of complex samples. Most recently, further optimization allowed the study of non-covalent protein-ligand interactions for bioaffinity screenings, which constitutes a major milestone in the development of this novel high-throughput autosampler.

Spectroscopic instruments are becoming increasingly popular for measuring the isotopic composition and fluxes of a wide variety of gases in both field and laboratory experiments. The popularity of these instruments has created a need for automated multiplexers compatible with the equipment. While there are several such peripherals commercially available, they are currently limited to only a small number of samples (≤16), which is insufficient for some studies. To support researchers in constructing custom, larger-scale systems, we present our design for a scalable gas sampling peripheral that can be programmed to autonomously sample up to 56 vessels – the “multiplexer”. While originally designed to be used with a Picarro cavity ring-down spectroscopy (CRDS) system, the multiplexer design and data processing approach implemented can be easily adapted to serve as a gas sampling/delivery platform for a wide variety of instruments including other cavity ring-down systems and infra-red gas analyzers. We demonstrate the basic capabilities of the multiplexer by using it to autonomously sample head-space CO2 from 14 laboratory-incubated soils amended with 13C-enriched pyrogenic organic matter for analysis in a Picarro G2201-i cavity ring-down spectroscopy system.

Benchtop nuclear magnetic resonance (NMR) spectrometers are versatile analytic instruments with low acquisition and operation cost. However, in the basic version, samples must be manually measured one after the other. We herein describe the open-source autosampler RotoMate that allows the automated operation of such instruments. The hardware is easily assembled from 3D-printed and inexpensive off-the-shelf parts, and is controlled by an Arduino Uno. A software package interlinks the operation of the autosampler with the software of the NMR spectrometer and the software for the processing of the spectra. Experiments for up to 30 samples can be inserted into an interactive sample list. The autosampler automatically inserts and ejects the samples, initiates measurements on the spectrometer according to parameters specified in the sample list, and interacts with a common NMR software in the processing and visualization of the obtained spectroscopic raw data. If an internal standard is present, conversions and yields of chemical reactions are automatically calculated, enabling e.g. the monitoring of reactions. The device was fitted to a Magritek Spinsolve instrument and can interact with a free academic version of ACD NMR software to process the spectra, but can likely be adapted to similar instruments and spectroscopy software packages.

Water sampling is an essential undertaking for water utilities and agencies to protect and enhance our natural resources. The high variability in water quality, however, often necessitates a spatially distributed sampling program which is impeded by high-cost and large sampling devices. This paper presents the BoSL FAL Pump - a low-cost, easily constructed, 3D-printed peristaltic pump which can be made from commonly available components and is sized to suit even the most space constrained installations. The pump is 38 mm in height and 28 mm in diameter, its components cost $19 AUD and the construction time is just 12 min (excluding 3D printing times). The pump is driven by a direct current motor which is commonly available, cheap and allows for flexibility in the energy supply (5–12 V). Optionally, the pump has a Hall effect sensor and magnet to detect rotation rates and pumping volumes to improve the accuracy of pumping rates/volumes. The pump can be easily controlled by commonly available microcontrollers, as demonstrated by this paper which implements the ATmega328P on the Arduino Uno R3. This paper validates the pump for long-term deployments at flow rates of up to 13 mL per minute in 0.14 mL volume increments at accuracy levels of greater than 99%. The pump itself is scalable, allowing for a wider range of pumping rates when, for example, large volume samples are required for pathogen and micropollutant detection.

Robotic positioning systems are used in a variety of chemical instruments, primarily for liquid handling purposes, such as autosamplers from vials or well plates. Here, two approaches to the design of open-source autosampler positioning systems for use with 96-well plates are described and compared. The first system, a 3-axis design similar to many low-cost 3D printers that are available on the market, is constructed using an aluminum frame and stepper motors. The other system relies upon a series of 3D printed parts to achieve movement with a series of linker arms based on Selective Compliance Assembly Robot Arm (SCARA) design principles. Full printer design files, assembly instructions, software, and user directions are included for both samplers. The positioning precision of the 3-axis system is better than the SCARA mechanism due to finer motor control, albeit with a slightly higher cost of materials. Based on the improved precision of this approach, the 3-axis autosampler system was used to demonstrate the generation of a segmented flow droplet stream from adjacent wells within a 96-well plate.

The drift of electronic nose (e-nose) sensors is the most challenging problem. An effective calibration method for e-nose is proposed in this article, which is based on projection on to convex sets (POCS) and extreme learning machine (ELM) method, including an automatic sampling platform and an e-nose sensor calibration model. It realizes a unified and universal calibration for both recognition and regression application to solve the electronic nose drift problem under long-term working conditions. Isopropanol and acetone gases that respond to most sensors were chosen as training gases for the model. Experiments show the effectiveness of the proposed e-nose calibration method. Automation of fluid manipulation is needed to increase throughput and reproducibility of experiments while reducing the time spent by researchers on performing repetitive tasks. Current solutions are, however, bulky and expensive, and hence not suited to enable automation in academic laboratories. This work, therefore, reports a low-cost, modular and programmable analytical platform comprising 3-D printed autosampler and peristaltic pump with footprints of 248 mm by 243 mm and 104 mm by 112 mm, respectively. The autosampler consisted of four sample probes/needles that can be driven independently to any of the available solution vials placed along the circumference of a circle. The 3-D printed autosampler developed in this work can access higher number of solution vials independently and is lower cost, but can accommodate fewer solution vials, than commercial devices. The autosampler and peristaltic pump were applied to determine: 1) refractive index sensitivity; 2) porosity; and 3) sensing capability of leaky waveguides (LWs) where the read-out instrument was also 3-D printed. We demonstrated that the automation of fluids enabled by the analytical platform improved the accuracy of results by separating the effect of temperature drifts from analyte solutions from the output of LWs. This work shows that 3-D printed instrumentation can provide results comparable with standard laboratory versions at much lower cost, making it ideal for use in resource-limited settings, and with the possibility of customization as experimental needs change. Elemental and isotopic analyses are performed using elemental analyzers, and are widely employed for diverse scientific fields. An elemental analyzer is typically equipped with an autosampler. Here we present an open-source autosampler for elemental and isotopic analysis of solid samples. The autosampler consists of 1) a sampling table, on which a carousel pushes samples inside an orifice, and 2) a purging pipe, placed directly beneath the orifice, where the sample is purged off surrounding air, and then delivered to the reaction tube. The action of the purging pipe ensured that air contamination, an issue for the analysis of some elements like nitrogen and oxygen, was negligible, and results for elemental and isotopic composition of nitrogen and carbon were inside specs. Compared to commercial alternatives, the autosampler presented here has the advantages of lower cost to build and maintain, universal compatibility with instruments from different manufacturers, capacity do deal with bulky samples, capacity for a larger number of samples in a single run, and no necessity for time-consuming purging in-between sample loading. The autosampler could potentially also be employed for the analyses of other elements (e.g. oxygen, hydrogen and sulfur) because they are performed using similar equipment.

Automated water sampling can be very useful, but open-source choices are limited. Here I present an autosampler which consists of a gantry robot that delivers water from a syringe pump to 24 capped 40 ml vials. The autosampler is controlled using an Android tablet automatized using Macrodroid. Three rinsing cycles ensure negligible carryover between consecutive samples. Hourly sampling from a creek under rainy conditions suggested that total organic carbon in water was diluted by the rain. Some important limitations: 1) the autosampler must be on a steady, flat, horizontal surface; 2) unattended sampling can only last as long as the batteries powering the tablet and the motors; 3) distance from the syringe pump to water cannot exceed ~2 m in height and ~4 m in length for 3 mm tubing; 4) sampling frequency does not exceed one sample every eleven minutes. However, because of its open design, the autosampler can be modified and improved to not only overcome these limitations, but also potentially expand its scope to more demanding sampling if necessary.

Historically, the dissolution testing has been used primarily as a quality control (QC) test for solid oral drug products performance. Dissolution testing is a basic technique used as a qualitative test to provide the measurement of the bioavailability of a drug as well as to demonstrate bioequivalence from batch-tobatch. The bioavailability and bioequivalence data obtained as a result of dissolution testing can be used in the development of a new formulation and product development processes toward product optimization, as well as to ensure continuing product quality and performance of the manufacturing process. In addition, dissolution is a requirement for regulatory approval for product marketing and is a vital component of the overall quality control program. Dissolution testing is conducted using a dissolution apparatus that conforms to the specifications outlined in the United States Pharmacopeia. In order to have a high degree of assurance that the dissolution apparatus is consistent and accurate in its performance, validation is required. Validation is defined as documented evidence that provides a high degree of assurance that a specific instrument performs consistently according to manufacturer’s specifications, user requirements meeting Good manufacturing practices (GMP) and Good laboratory practices (GLP). Validation is achieved by performing a series of validation activities; for a newly installed dissolution apparatus, validation is obtained through installation qualification (IQ), operational qualification (OQ) and performance qualification (PQ) through respective stages protocols. During different stages of qualification, it is ensured that dissolution tester was effectively installed, operated as per user manual & performed according to given programmed operation as per feeding instructions. During performance qualification the calibration results of installed dissolution tester were obtained within limit. Various physical parameters were tested like Spirit level test, Rotation per minute test, Temperature of water bath & each jar, Timer, Wobbling test & in Chemical test performance verification test with Prednisone tablet. Antarctica is considered the most pristine environment on Earth but is also characterized by its unique conditions such as the strong polar vortex and extreme cold. A detailed understanding of volatile organic compounds (VOCs) and the atmospheric oxidation reactions they undergo is essential to document biogeochemical cycles and to better understand their impact on radiative forcing. This research aims to provide a unique dataset of oxygenated (O)VOCs occurring in the Antarctic troposphere and provide insights into their temporal behavior. A home-made sequential sorbent tube auto sampler was deployed at the atmospheric observatory of the Princess Elisabeth station (71.95° S, 23.35° E, 1390 m asl) to collect 20 samples during the period from December 2019 to October 2020. The samples were analyzed consecutively by TD-GC-MS followed by direct thermal desorption of samples in a high-resolution PTR-Qi-TOFMS. Concentrations of 70 VOCs allocated to 4 different chemical groups (halogenated compounds, non-aromatic hydrocarbons, sulfur-containing compounds, and oxygenated aromatic and non-aromatic compounds) were determined. The results show temporal patterns for compounds such as bromoform (14 ± 6 ng/m3) and OVOCs such as furaldehyde (24 ± 9 ng/m³), amongst others, which are attributed to the seasonality of atmospheric conditions. Products of the atmospheric oxidation process show linear correlation indicating their mutual relationship and association with a common parent compound. The usage of an autonomous autosampler in the extreme conditions of Antarctica was demonstrated and proved to be a powerful tool in the sampling of air in such a remote location. Abstract The application of a fully automated autosampler/HPLC-ESI–MS/MS system for spatially resolved quantitative droplet-based liquid extraction surface sampling/profiling of stapled α–helical peptide ATSP-7041 in mouse whole-body thin tissue sections is reported. 20-μm-thick serial tissue sections of an ATSP-7041 dosed mouse were prepared and the absolute concentration of the targeted peptide was first determined in different organs using 2.3-mm diameter tissue punches, standard bulk tissue extraction protocols, and subsequent HPLC separation and tandem mass spectrometric analysis. The same organs/locations were then analyzed in neighboring tissue sections using the droplet-based surface sampling approach. The observed ATSP-7041 concentration using this method was always significantly lower than that measured by the tissue punch workflow at the same tissue location of a serial section. Calculated extraction efficiencies were 10.7 ± 0.5% (brain), 11.0 ± 3.2% (liver spot 1), 10.7 ± 2.6% (liver spot 2), 15.0 ± 0.6% (lung) and 12.9 ± 0.7% (blood). While these extraction efficiency values were low, they were reproducible within a given organ. This suggests that once the extraction efficiency is established for a given tissue type and drug, the reproducibility of the droplet-based approach could provide a non-labor intensive and high-throughput means to acquire spatially resolved quantitative analysis of multiple samples of the same type. In-house built capillary electrophoresis (CE) systems represent a significant share of laboratory instrumentation. In most of these instruments, sample injection is effected manually with low to moderate precision and requires skilled operators. Although few automated samplers have been previously developed, typically only one sample at a time can be injected. If a series of samples is to be analyzed, manual intervention is required. In the present work, we developed and constructed a fully automated, open source, CE autosampler, able to handle up to 14 different samples that can be used as a modular component of any in-house built CE instrument. An inexpensive, 3D printed, open source, autosampler for CE was developed. The autosampler consists of two parts: an injection unit with carousel containing sample and electrolyte vials and a flushing unit, containing a miniature pressure/vacuum pump. The autosampler is operated by an Arduino Mega microcontroller and an Arduino code written in the laboratory. The injection sequence is entered through a keypad and LCD display by the user. The instrument can operate autonomously for extended periods of time. It was used for fully automated analysis and/or calibration of up to 14 samples with excellent injection repeatability reaching less than 2.7% RSD for peak areas. The sampler performance was tested with two independently built CE instruments, a CE system with contactless conductivity detection (C4D) and a CE system with laser induced fluorescence (LIF) detector. A novel, 3D printed, Arduino-based autosampler for CE was developed. The autosampler allows autonomous hydrodynamic injection of up to 14 different samples with fully programmable injection sequence, including capillary flushing and high voltage and data acquisition control. It provides the missing instrumental sampling setup for laboratory made CE instruments. It can be simply constructed based on the open-source blueprints in any laboratory and be a useful and time-saving add-on to any modular CE instrument. Outer membrane vesicles (OMVs) are spherical membrane nanoparticles released by Gram-negative bacteria. OMVs can be quantified in complex matrices by nanoparticle tracking analysis (NTA). NTA can be performed in static mode or with continuous sample flow that results in analysis of more particles in a smaller time-frame. Flow measurements must be performed manually despite the availability of a sample changer on the NanoSight system. Here we present a method for automated measurements in flow mode. OMV quantification in flow mode results in lower variance in particle quantification (coefficient of variation (CV) of 6%, CV static measurements of 14%). Sizing of OMVs was expected to be less favorable in flow mode due to the increased movement of the particles. However, we observed a CV of 3% in flow mode and a CV of 8% in static measurements. Flow rates of up to 5 µL/min displayed correct size and particle measurements, however, particle concentration was slightly lower than in static measurements. The automated method was used to assess OMV release of batch cultures of Neisseria meningitidis. The bacteria released more OMVs in stationary growth phase, while the size of the vesicles remained constant throughout the culture. Taken together, this study shows that automated measurements in flow mode can be established with advanced scripting to reduce the workload for the user.

Understanding the abundance and fate of human viral pathogens in wastewater is essential when assessing the public health risks associated with wastewater discharge to the environment. Typically, however, the microbiological monitoring of wastewater is undertaken on an infrequent basis and peak discharge events may be missed leading to the misrepresentation of risk levels. To evaluate diurnal patterns in wastewater viral loading, we undertook 3-day sampling campaigns with bi-hourly sample collection over three seasons at three wastewater treatment plants. Untreated influent was collected at Ganol and secondary-treated effluent was sampled at Llanrwst and Betws-y-Coed (North Wales, UK). Our results confirmed the presence of human adenovirus (AdV), norovirus genotypes I and II (NoVGI and NoVGII) in both influent and effluent samples while sapovirus GI (SaVGI) was only detected in influent water. The AdV titre was high and relatively constant in all samples, whereas the NoVGI, NoVGII and SaVGI showed high concentrations during autumn and winter and low counts during the summer. Diurnal patterns were detected in pH and turbidity for some sampling periods; however, no such changes in viral titres were observed apart from slight fluctuations in the influent samples. Our findings suggest that viral particle number in wastewater is not affected by daily chemical fluctuations. Hence, a grab sample taken at any point during the day may be sufficient to enumerate the viral load of wastewater effluent within an order of magnitude while four samples a day are recommended for testing wastewater influent samples. Herein, quantitation aspects of a fully automated autosampler/HPLC-MS/MS system applied for unattended droplet-based surface sampling of repaglinide dosed thin tissue sections with subsequent HPLC separation and mass spectrometric analysis of parent drug and various drug metabolites were studied. Major organs (brain, lung, liver, kidney and muscle) from whole-body thin tissue sections and corresponding organ homogenates prepared from repaglinide dosed mice were sampled by surface sampling and by bulk extraction, respectively, and analyzed by HPLC-MS/MS. A semi-quantitative agreement between data obtained by surface sampling and that by employing organ homogenate extraction was observed. Drug concentrations obtained by the two methods followed the same patterns for post-dose time points (0.25, 0.5, 1 and 2 h). Drug amounts determined in the specific tissues was typically higher when analyzing extracts from the organ homogenates. In addition, relative comparison of the levels of individual metabolites between the two analytical methods also revealed good semi-quantitative agreement. [ABSTRACT FROM AUTHOR]

Described here are the results from the profiling of the proteins arginine vasopressin (AVP) and adrenocorticotropic hormone (ACTH) from normal human pituitary gland and pituitary adenoma tissue sections, using a fully automated droplet-based liquid-microjunction surface-sampling-HPLC-ESI-MS-MS system for spatially resolved sampling, HPLC separation, and mass spectrometric detection. Excellent correlation was found between the protein distribution data obtained with this method and data obtained with matrix-assisted laser desorption/ionization (MALDI) chemical imaging analyses of serial sections of the same tissue. The protein distributions correlated with the visible anatomic pattern of the pituitary gland. AVP was most abundant in the posterior pituitary gland region (neurohypophysis), and ATCH was dominant in the anterior pituitary gland region (adenohypophysis). The relative amounts of AVP and ACTH sampled from a series of ACTH-secreting and non-secreting pituitary adenomas correlated with histopathological evaluation. ACTH was readily detected at significantly higher levels in regions of ACTH-secreting adenomas and in normal anterior adenohypophysis compared with non-secreting adenoma and neurohypophysis. AVP was mostly detected in normal neurohypophysis, as expected. This work reveals that a fully automated droplet-based liquid-microjunction surface-sampling system coupled to HPLC-ESI-MS-MS can be readily used for spatially resolved sampling, separation, detection, and semi-quantitation of physiologically-relevant peptide and protein hormones, including AVP and ACTH, directly from human tissue. Droplet-based liquid microjunction surface sampling coupled with high-performance liquid chromatography (HPLC)-electrospray ionization (ESI)-tandem mass spectrometry (MS/MS) for spatially resolved analysis provides the possibility of effective analysis of complex matrix samples and can provide a greater degree of chemical information from a single spot sample than is typically possible with a direct analysis of an extract. Described here is the setup and enhanced capabilities of a discrete droplet liquid microjunction surface sampling system employing a commercially available CTC PAL autosampler. The system enhancements include incorporation of a laser distance sensor enabling unattended analysis of samples and sample locations of dramatically disparate height as well as reliably dispensing just 0.5 μL of extraction solvent to make the liquid junction to the surface, wherein the extraction spot size was confined to an area about 0.7 mm in diameter; software modifications improving the spatial resolution of sampling spot selection from 1.0 to 0.1 mm; use of an open bed tray system to accommodate samples as large as whole-body rat thin tissue sections; and custom sample/solvent holders that shorten sampling time to approximately 1 min per sample. The merit of these new features was demonstrated by spatially resolved sampling, HPLC separation, and mass spectral detection of pharmaceuticals and metabolites from whole-body rat thin tissue sections and razor blade ('crude') cut mouse tissue. Monitoring concentrations of bacterial pathogens and indicators of fecal contamination in coastal and estuarine ecosystems is critical to reduce adverse effects to public health. During storm events, particularly hurricanes, floods, Nor'easters, and tropical cyclones, sampling of coastal and estuarine waters is not generally possible due to safety concerns. It is particularly important to monitor waters during these periods as it is at precisely these times that pathogenic bacteria such as Vibrio spp. and fecal indicator bacteria concentrations fluctuate, potentially posing significant risks to public health. Automated samplers, such as the Isco sampler, are commonly used to conduct remote sample collection. Remote sampling is employed during severe storm periods, thereby reducing risk to researchers. Water samples are then stored until conditions are safe enough to retrieve them, typically in less than 21 h, to collect the samples. Concerns exist regarding potential "bottle effects", whereby containment of sample might result in altered results. While these effects are well documented in samples being held for 24 h or more, there is little data on bottle effects occurring during the first 24 h of containment, and less still on the specific effects related to this type of sampling regime. Estuarine water samples were collected in the fall of 2013, placed into an Isco autosampler and subsampled over time to determine the effects of storage within this type of autosampling device. Vibrio spp. and fecal indicator bacteria were quantified using replicated culture-based methods, including Enterolert™ and membrane filtration. The experiments demonstrated no significant impact of storage time when comparing concentrations of total Vibrio spp., Vibrio vulnificus, Vibrio parahaemolyticus, or Enterococcus spp. after storage compared to original concentrations. However, the findings also suggested that increased variability and growth can occur during the middle of the day. Therefore, if at all possible, analysis schedules should be modified to account for this variability, e.g. collection of samples after overnight storage should occur as early in the morning as practicable.
