Infections at the placement site of biomaterial-based devices and subsequent scar formation results in morbidity, which may require revision surgery. Biomaterials intended for permanent implantation in the body need to be biologically inert to avoid excessive foreign body response and to reduce bacterial attachment. In this study, we show that polymeric materials commonly used in medical devices, including polyetheretherketone (PEEK) and polypropylene, treated by gas cluster ion beam (GCIB) or by accelerated neutral atom beam (ANAB) result in a nanoscale-modified surface topography that changes the ability of extracellular proteins to bind. This leads to decreased bacterial attachment and an attenuated inflammatory response using both in vitro and in vivo assays. Differential adsorption of extracellular proteins to the polymeric surface improved the competitive attachment of osteoblasts over bacteria, without resorting to growth factor of antibiotic use.

Tissue engineering for spinal cord injury (SCI) remains a complex and challenging task. Biomaterial scaffolds have been suggested as a potential solution for supporting cell survival and differentiation at the injury site. However, different biomaterials display multiple properties that significantly impact neural tissue at a cellular level. Here, we evaluated the behavior of different cell lines seeded on chitosan (CHI), poly (ε-caprolactone) (PCL), and poly (L-lactic acid) (PLLA) scaffolds. We demonstrated that the surface properties of a material play a crucial role in cell morphology and differentiation. While the direct contact of a polymer with the cells did not cause cytotoxicity or inhibit the spread of neural progenitor cells derived from neurospheres (NPCdn), neonatal rat spinal cord cells (SCC) and NPCdn only attached and matured on PCL and PLLA surfaces. Scanning electron microscopy and computational analysis suggested that cells attached to the material's surface emerged into distinct morphological populations. Flow cytometry revealed a higher differentiation of neural progenitor cells derived from human induced pluripotent stem cells (hiPSC-NPC) into glial cells on all biomaterials. Immunofluorescence assays demonstrated that PCL and PLLA guided neuronal differentiation and network development in SCC. Biomaterial surfaces tend to adhere only under dry conditions, preventing them from being used in underwater environments such as body fluids. Therefore, biomaterial surfaces with underwater adhesion ability have received increasing attention. In nature, some organisms can adapt to different environments by evolving appendages with underwater adhesion capability and unique functions. Most of these outstanding functions are derived from special micro/nanostructures and/or chemical compositions of these organisms. Thus, modification of biomaterials surfaces with similar microstructures and chemical groups would help promote underwater adhesion. In this review, we discuss (1) the mechanisms of underwater adhesion; (2) surface modification strategies for underwater adhesion, including mussel-inspired, interlocking, bio-structured surface, and specific adhesion strategies; and (3) the challenges and potential of developing surface-modified, multiscale biomaterials with underwater adhesion.

Abstract: To explore molecular mechanism of mediation of adsorbed proteins to cell adhesion and growth on biomaterials, this study examined endothelial cell adhesion, morphology and viability on bare and titanium nitride (TiN) coated nickel titanium (NiTi) alloys and chitosan film firstly, and then identified the type and amount of serum proteins adsorbed on the three surfaces by proteomic technology. Subsequently, the mediation role of the identified proteins to cell adhesion and growth was investigated with bioinformatics analyses, and further confirmed by a series of cellular and molecular biological experiments. Results showed that the type and amount of adsorbed serum proteins associated with cell adhesion and growth was obviously higher on the alloys than on the chitosan film, and these proteins mediated endothelial cell adhesion and growth on the alloys via four ways. First, proteins such as adiponectin in the adsorbed protein layer bound with cell surface receptors to generate signal transduction, which activated cell surface integrins through increasing intracellular calcium level. Another way, thrombospondin 1 in the adsorbed protein layer promoted TGF-β signaling pathway activation and enhanced integrins expression. The third, RGD sequence containing proteins such as fibronectin 1, vitronectin and thrombospondin 1 in the adsorbed protein layer bound with activated integrins to activate focal adhesion pathway, increased focal adhesion formation and actin cytoskeleton organization and mediated cell adhesion and spreading. In addition, the activated focal adhesion pathway promoted the expression of cell growth related genes and resulted in cell proliferation. The fourth route, coagulation factor II (F2) and fibronectin 1 in the adsorbed protein layer bound with cell surface F2 receptor and integrin, activated regulation of actin cytoskeleton pathway and regulated actin cytoskeleton organization. Sufficient induction of blood vessel ingrowth decisively influence transplant functionality. In this study, microvascular response to transplants of surface modified bone substitutes were assessed in vivo. The surface modification of allogenic bone substitutes (dehydrated human femoral head) was achieved in a double-conductive low-pressure gasplasma reactor (Ar/O, 13.65 MHz, 1,000 W, 5 Pa). The modified bone substitutes ( n = 10) as well as untreated bone substitutes serving as controls ( n = 10) were placed into the dorsal skinfold chamber of female balb/c mice ( n = 10). Dynamic assessment of microcirculatory parameters was performed using intravital fluorescence microscopy during an implantation period of 10 days. The angiogenic response was found markedly accelerated in gasplasma-treated bone. Compared to untreated implants, the gasplasma-activated bone substitutes showed significantly higher microvascular density on days 5 and 10. The quantification of the microvascular diameters, red blood cell velocity, and microvascular permeability displayed stable perfusion and vascular integrity of the newly developed blood vessels throughout the 10-day observation period. Carbon materials are generally well tolerated by animal cells, so biomaterials of this type have come into routine use for constructing medical implants. The formation of surface layer makes them attractive for coating implants, e.g. in orthopaedic and dental surgery. The system biocompatibility is controlled mainly by its surface properties. The chemical state of the surface is given mainly by the material and by the method of surface preparation. On the other hand, biocompatibility can be improved by a suitable change of the surface morphology, e.g. by influencing the surface roughness. We prepared a surface coating of carbon fibre reinforced carbon composite using a layer of pyrolytic graphite and we used grinding and polishing of the layer for roughness control. The purpose of this article is to define the surface and surface roughness of carbon fibre reinforced carbon composite with and without a pyrolytic graphite layer and after defined surface treatment, and to correlate the results with several parameters of tissue cell growth. The main result of this work shows that, using the proper characterisation of the surface parameters, there is a statistically significant correlation with biocompatibility, especially, with adhesion and proliferation of cells. Integrins represent the primary mechanism of cell-extracellular matrix interactions and control cell morphology, proliferation, and differentiation. We have previously shown that substrate-dependent modulation of adsorbed fibronectin (Fn) conformation alters α5β1 integrin binding to Fn and directs C2C12 myoblast proliferation and differentiation (Mol. Biol. Cell 10 (1999) 785). The model substrates used in these experiments were bacteriological (untreated) polystyrene (B), tissue culture polystyrene (T), and type-I collagen-coated T (C). In the present study, we examined MC3T3-E1 osteoblast-like cell differentiation on Fn-coated B, T, and C substrates. Immunofluorescence staining revealed substrate-dependent differences in integrin α5β1 binding and clustering into focal adhesions (C>T>B), consistent with our previous integrin binding analysis. Alkaline phosphatase activity and matrix mineralization showed substrate-dependent differences (C>T>B, p<0.05). Similar trends were observed for alkaline phosphatase, osteocalcin, and bone sialoprotein gene expression. Blocking experiments with antibodies directed against Fn completely inhibited matrix mineralization on Fn-coated C, indicating that Fn is critical to expression of the osteoblastic phenotype on this extracellular matrix component. These substrate-dependent differences in osteoblast differentiation correlated with differences in α5β1 binding, suggesting that these differences arise from substrate modulation of integrin-matrix interactions. Substrate-dependent modulation of cell function may provide a versatile mechanism to control cell responses in numerous biomedical applications. [Copyright &y& Elsevier]

The use of natural supplies is a resource to mimic an original extracellular matrix that allows for migration, proliferation, and cellular organization. Chitosan from Brazilian Atlantic Ocean had low protein, minerals percentage and excellent antibacterial activity. The aim of this study was to evaluate and to compare the effectiveness of different types of acids as solvents with Brazilian chitosan-membrane in the healing process of skin lesions. Experimental full-thickness 2 × 2 cm wounds were created on the dorsum skin of Wistar rats. The applied different treatments were saline, collagenase®, microcrystalline chitosan salt membrane (MCSM), microcrystalline chitosan acetic acid membrane (MCAAM), and microcrystalline chitosan hydrochloric acid membrane (MCHAM). The wound repairs were measured morphologically and histologically on days 0, 3, 7, 10, and 14. The exudate formation and the final wound contractions were similar in all of the groups. There were mild exudations in the groups with chitosan-membranes, despite the formation of crust under the membrane. This configured a serum hematic aspect, but there was no impact on the healing process. The MCHAM group had more favorable aspects that histologically showed the healing phases. A significant migration of neutrophils and macrophages seen by myeloperoxidade and Beta-N-Acetylglucosaminidase activities was evident in the chitosan groups, MCHAM and MCSM, respectively.
