Appropriate abiotic stress response is pivotal for plant survival and makes use of multiple signaling molecules and phytohormones to achieve specific and fast molecular adjustments. A multitude of studies has highlighted the role of alternative splicing in response to abiotic stress, including temperature, emphasizing the role of transcriptional regulation for stress response. Here we investigated the role of the core-splicing factor PORCUPINE (PCP) on temperature-dependent root development. We used marker lines and transcriptomic analyses to study the expression profiles of meristematic regulators and mitotic markers, and chemical treatments, as well as root hormone profiling to assess the effect of auxin signaling. The loss of PCP significantly alters RAM architecture in a temperature-dependent manner. Our results indicate that PCP modulates the expression of central meristematic regulators and is required to maintain appropriate levels of auxin in the RAM. We conclude that alternative pre-mRNA splicing is sensitive to moderate temperature fluctuations and contributes to root meristem maintenance, possibly through the regulation of phytohormone homeostasis and meristematic activity.

Natural populations are subject to selection caused by a range of biotic and abiotic factors in their native habitats. Identifying these agents of selection and quantifying their effects is key to understanding how populations adapt to local conditions. We performed a factorial reciprocal-transplant experiment using locally adapted ecotypes of Arabidopsis thaliana at their native sites to distinguish the contributions of adaptation to soil type and climate. Overall adaptive differentiation was strong at both sites. However, we found only very small differences in the strength of selection on local and non-local soil, and adaptation to soil type at most constituted only a few per cent of overall adaptive differentiation. These results indicate that local climatic conditions rather than soil type are the primary driver of adaptive differentiation between these ecotypes.

The Violaceae family is rich in metal-tolerant species and species producing cyclic peptides (cyclotides) that are linked to the resistance to biotic factors. Plants that inhabit areas polluted with heavy metals have developed various mechanisms of tolerance. To test the role of cyclotides in protection against abiotic factors, including heavy metals, cell suspension cultures of Viola species/genotypes (V. lutea ssp. westfalica, V. tricolor, V. arvensis, and V. uliginosa), representing different levels of tolerance to heavy metals (from the most tolerant-MET to the least tolerant populations/species-NMET), were used. The relative abundances of the cyclotides in the control, untreated cell suspensions of all the selected species/genotypes, and cells treated with Zn or Pb (200 mu M or 2000 mu M) for 24 h or 72 h were determined via MALDI-MS. Transmission electron microscopy with X-ray microanalysis was used to detect putative co-localization of the cyclotides with Zn or Pb in the cells of V. tricolor treated with the highest concentration of heavy metals for 72 h. Cyclotide biosynthesis was dependent on the type of heavy metal and its concentration, time of treatment, plant species, and population type (MET vs. NMET). It was positively correlated with the level of tolerance of particular Viola species. The increased production of cyclotides was observed in the cells of metallophyte species, mostly in Zn-treated cells. The nonmetallophyte-V. uliginosa presented a decrease in the production of cyclotides independent of the dose and duration of the metal treatment. Cyclotides co-localized with Pb more evidently than with Zn, suggesting that cyclotides have heavy metal affinity. V. lutea ssp. westfalica transcriptome mining yielded 100 cyclotide sequences, 16 known and 84 novel named viwe 1-84. These findings support the hypothesis that cyclotides are involved in certain mechanisms of plant tolerance to heavy metals.

Pollinator behavior is vital to plant-pollinator interactions, affecting the acquisition of floral rewards, patterns of pollen transfer, and plant reproductive success. During buzz pollination, bees produce vibrations with their indirect flight muscles to extract pollen from tube-like flowers. Vibrations can be transmitted to the flower via the mandibles, abdomen, legs, or thorax directly. Vibration amplitude at the flower determines the rate of pollen release and should vary with the coupling of bee and flower. This coupling often occurs through anther biting, but no studies have quantified how biting affects flower vibration. Here, we used high-speed filmography to investigate how flower vibration amplitude changes during biting in Bombus terrestris visiting two species of buzz-pollinated flowering plants: Solanum dulcamara and Solanum rostratum (Solanaceae). We found that floral buzzing drives head vibrations up to 3 times greater than those of the thorax, which doubles the vibration amplitude of the anther during biting compared with indirect vibration transmission when not biting. However, the efficiency of this vibration transmission depends on the angle at which the bee bites the anther. Variation in transmission mechanisms, combined with the diversity of vibrations across bee species, yields a rich assortment of potential strategies that bees could employ to access rewards from buzz-pollinated flowers.

Global warming is one of the biggest threats to global biodiversity causing not only changes in the patterns of precipitation and temperature but also disturbing ecological interactions. The aim of our study was to forecast the effect of climate change on the distribution of food-deceptive orchid species whose pollination strategy relies on a strict association with pollinators and co-occurring rewarding Faboideae plants. We used the ecological niche modeling approach to evaluate future overlap of the suitable niches of studied orchid species with the predicted distribution of their ecological partners. Models were made based on two different global circulation models (FIO, CNRM). CNRM projections predict expansion of orchids' geographical range. In contrast, FIO prediction is less optimistic, forecasting species range contraction. The studied Faboideae species showed different responses to predicted global warming with no consistent patterns in how their suitable niches might change. Most climate change projections and scenarios of the future modifications of temperature and precipitation patterns do not predict significant loss of suitable niches of Trichocolletes bees (Colletidae) pollinating Diuris species. However, global warming has the potential to disrupt interactions between the studied orchids and their co-occurring pea plants by altering the overlap of their geographical ranges which can further disturb pollination success. CNRM projections predict an overall loss of Faboideae within the potential geographical range of Diuris brumalis. Conversely, FIO projections suggest a less extensive predicted divergence. Our simulations offer suggestions for conservation strategies of orchids and potentially for other species that have a similar pollination strategy. We present a robust, five-locus phylogeny of the Megasporaceae and, based on this, propose several taxonomic innovations. The new genus Antidea is erected for Aspicilia brucei, which occupies a position near the base of the phylogeny, and the new species Aspicilia indeterminata and A. suavis are described from Montana. We also show that all North American (and some European) records of Aspilidea myrinii are misidentifications with many representing a second species in the genus, differing from A. myrinii by having elevated apothecia and narrower ascospores and for which we make the new combination Aspilidea subadunans. Finally, we make the new combinations Lobothallia determinata and L. peltastictoides, and report the lichenicolous fungus Sagediopsis aspiciliae (on A. subadunans) as new to North America.

The widespread use of plant grafting enables eudicots and gymnosperms to join with closely related species and grow as one. Gymnosperms have dominated forests for over 200 million years, and despite their economic and ecological relevance, we know little about how they graft. Here we developed a micrografting method in conifers using young tissues that allowed efficient grafting with closely related species and between distantly related genera. Conifer graft junctions rapidly connected vasculature and differentially expressed thousands of genes including auxin and cell-wall-related genes. By comparing these genes to those induced during Arabidopsis thaliana graft formation, we found a common activation of cambium, cell division, phloem and xylem-related genes. A gene regulatory network analysis in Norway spruce (Picea abies) predicted that PHYTOCHROME A SIGNAL TRANSDUCTION 1 (PAT1) acted as a core regulator of graft healing. This gene was strongly up-regulated during both spruce and Arabidopsis grafting, and Arabidopsis mutants lacking PAT genes failed to attach tissues or successfully graft. Complementing Arabidopsis PAT mutants with the spruce PAT1 homolog rescued tissue attachment and enhanced callus formation. Together, our data show an ability for young tissues to graft with distantly related species and identifies the PAT gene family as conserved regulators of graft healing and tissue regeneration.

Biased sex ratios among reproductive individuals are common in plants, but the underlying mechanisms, as well as the evolutionary consequences, are not well understood. The classical theory of Düsing and Fisher predicts an equal primary sex ratio at seed production, based on the selective advantage of the rare sex. Biased sex ratios among reproductive plants can arise from sexual dimorphism in survival and flowering. Sex ratio biases can also be present from the seed stage; in these cases, assumptions of Düsing's and Fisher's theory, for example, random mating or demographic equilibrium, are thought to be violated. We investigated mechanisms leading to female-biased sex ratios in the arctic-alpine dwarf willow Salix herbacea L. We studied sex ratios in three natural populations over 3 years as well as in 29 crosses (full-sib families) under controlled conditions over four growth periods. We tested whether sex ratio was associated with habitat parameters (elevation and snowmelt time), or with germination, survival or flowering, and whether females and males differed in size or flowering that may cause observation bias. We detected a strong and consistent female bias, both in natural populations (sex ratio [proportion of females]: 0.71–0.82) and in our controlled experiment (overall sex ratio: 0.70–0-72). Female bias became more pronounced with increasing elevation. Our data did not support sexual dimorphism in size or flowering. Family sex ratios varied largely (from 0.25 to 1), including many female-biased families, unbiased families and two male-biased families. Families with lower germination, seedling establishment, survival or flowering did not have stronger female bias, indicating that intrinsically higher survival or flowering in females does not explain overall female bias. Heritage forests are gaining greater prominence and importance as places with high cultural and ecological value, and ideal for biodiversity conservation worldwide. However, there are few studies documenting these forests in Mozambique. This thesis presents 88 different cultural heritage sites in Inhambane Province, of which 52 are heritage forests. In this thesis I focus on three case study areas, Luido, Chitanga, and Mapoka showing with satellite images analyses that forest boundaries have remained relatively stable over the last three decades. Heritage forests are connected to discourse and discursive practices of heritage. Chiefs are the main representatives of the local institutions and its associated social codes and system of laws for protection of heritage forests. In post-colonial Mozambique, such local heritage practices were constrained and controlled, however, they were still maintained. Botanical inventories of the forests show that local heritage practices taking place in and around have a fundamental role to the existence of plant diversity. A total of 14 plant species with great conservation value was identified. Most of these locally protected heritage sites are not included in formally protected areas, and they are currently experiencing various types of pressures from within and outside the community, mostly from logging activities. Formal recognition of communities as collaborators and partners to the continuity of these areas is essential and feasible, but such need to be implemented cautiously and driven by communities. In mountainous areas, wild edible plants are an important part of the local diet. Climate change and anthropogenic activities have profound effects on wild edible plants in these areas. Ethnobotanical studies are important for understanding the use patterns and harvest impacts on these plants. In this regard, the Shahrood region, with its diverse historical/ethnic background, is an appropriate starting point to investigate exploitation patterns and impacts of harvest of wild edible plants. During 2021-2022, ethnobotanical surveys were conducted in 12 villages in the region using semi-structured interviews and participatory observations. Data collection included assessing wild edible plant species diversity, their habitats, collection time and quantities, plant parts used and methods of consumption and preparation. The data were analyzed using use report indicators, relative frequency of citations, and use value (UV). A total of 1086 use reports were documented from a total of 44 interviews. A total of 67 wild species (66 plants and 1 fungus) belonging to 54 genera and 24 families were used as edible in the study area. Rosaceae (9 species), Apiaceae (8), Lamiaceae (8), Asteraceae (7), and Amaryllidaceae (5) were the families with the most reported species. Allium (5 species) was the most diverse genus in terms of species diversity. Herbs were the most commonly used life form (79.1%), followed by shrubs (13.4%) and trees (6%). The most consumed plant parts were young leaves (25%) and young aerial parts (21.4%). There were 13 use categories identified. The majority of edible plants were collected in April-May, mainly from areas nearby villages (37.7%) and rangelands (33.7%). Premise: We assessed changes in traits associated with water economy across climatic gradients in the ecologically similar peat mosses Sphagnum cuspidatum and Sphagnum lindbergii. These species have parapatric distributions in Europe and have similar niches in bogs. Sphagnum species of bogs are closely related, with a large degree of microhabitat niche overlap between many species that can be functionally very similar. Despite this, ecologically similar species do have different distributional ranges along climatic gradients that partly overlap. These gradients may favor particular Sphagnum traits, especially in relation to water economy, which can be hypothesized to drive species divergence by character displacement. Methods: We investigated traits relevant for water economy of two parapatric bryophytes (Sphagnum cuspidatum and S. lindbergii) across the border of their distributional limits. We included both shoot traits and canopy traits, i.e., collective traits of the moss surface, quantified by photogrammetry. Results: The two species are ecologically similar and occur at similar positions along the hydrological gradient in bogs. The biggest differences between the species were expressed in the variations of their canopy surfaces, particularly surface roughness and in the responses of important traits such as capitulum mass to climate. We did not find support for character displacement, because traits were not more dissimilar in sympatric than in allopatric populations. Conclusions: Our results suggest that parapatry within Sphagnum can be understood from just a few climatic variables and that climatic factors are stronger drivers than competition behind trait variation within these species of Sphagnum.

The shift from outcrossing to self-fertilization is one of the main evolutionary transitions in plants and has broad effects on evolutionary trajectories. In Brassicaceae, the ability to inhibit self-fertilization is controlled by 2 genes, SCR and SRK, tightly linked within the S-locus. A series of small non-coding RNAs also encoded within the S-locus regulates the transcriptional activity of SCR alleles, resulting in a linear dominance hierarchy between them. In Brassicaceae, natural allopolyploid species are often self-compatible (SC) even when one of the progenitor species is self-incompatible, but the reason why polyploid lineages tend to lose self-incompatibility (SI) and the timing of the loss of SI (immediately after ancestral hybridization between the progenitor species, or at a later stage after the formation of allopolyploid lineages) have generally remained elusive. We used a series of synthetic diploid and tetraploid hybrids obtained between self-fertilizing Capsella orientalis and outcrossing Capsella grandiflora to test whether the breakdown of SI could be observed immediately after hybridization, and whether the occurrence of SC phenotypes could be explained by the dominance interactions between S-haplotypes inherited from the parental lineages. We used RNA-sequencing data from young inflorescences to measure allele-specific expression of the SCR gene and infer dominance interactions in the synthetic hybrids. We then evaluated the seed set from autonomous self-pollination in the synthetic hybrids. Our results demonstrate that self-compatibility of the hybrids depends on the relative dominance between S-alleles inherited from the parental species, confirming that SI can be lost instantaneously upon formation of the ancestral allopolyploid lineage. They also confirm that the epigenetic regulation that controls dominance interactions between S-alleles can function between subgenomes in allopolyploids. Boreal conifers – the ‘Christmas trees’ – maintain their green needles over the winter by retaining their chlorophyll. These conifers face the toughest challenge in February and March, when subzero temperatures coincide with high solar radiation. To balance the light energy they harvest with the light energy they utilise, conifers deploy various mechanisms in parallel. These include, thylakoid destacking, which facilitates direct energy transfer from Photosystem II (PSII) to Photosystem I (PSI), and excess energy dissipation through sustained nonphotochemical quenching (NPQ). Additionally, they upregulate alternative electron transport pathways to safely reroute excess electrons while maintaining ATP production. From an evolutionary and ecological perspective, we consider these mechanisms as part of a comprehensive photosynthetic alteration, which enhances our understanding of winter acclimation in conifers and their dominance in the boreal forests.

Urbanization impacts plant-herbivore interactions, which are crucial for ecosystem functions such as carbon sequestration and nutrient cycling. While some studies have reported reductions in insect herbivory in urban areas (relative to rural or natural forests), this trend is not consistent and the underlying causes for such variation remain unclear. We conducted a continental-scale study on insect herbivory along urbanization gradients for three European tree species: Quercus robur, Tilia cordata, and Fraxinus excelsior, and further investigated their biotic and abiotic correlates to get at mechanisms. To this end, we quantified insect leaf herbivory and foliar secondary metabolites (phenolics, terpenoids, alkaloids) for 176 trees across eight European cities. Additionally, we collected data on microclimate (air temperature) and soil characteristics (pH, carbon, nutrients) to test for abiotic correlates of urbanization effects directly or indirectly (through changes in plant secondary chemistry) linked to herbivory. Our results showed that urbanization was negatively associated with herbivory for Q. robur and F. excelsior, , but not for T. cordata. . In addition, urbanization was positively associated with secondary metabolite concentrations, but only for Q. robur. . Urbanization was positively associated with air temperature for Q. robur and F. excelsior, , and negatively with soil nutrients (magnesium) in the case of F. excelsior, , but these abiotic variables were not associated with herbivory. Contrary to expectations, we found no evidence for indirect effects of abiotic factors via plant defences on herbivory for either Q. robur or F. excelsior. . Introduction: Phenotypic complexity in species complexes and recently radiated lineages has resulted in a diversity of forms that have historically been classified into separate taxa. Increasingly, with the proliferation of high-throughput sequencing methods, additional layers of complexity have been recognized, such as frequent hybridization and reticulation, which may call into question the previous morphological groupings of closely related organisms. Methods: We investigated Northern European, Asian, and Beringian populations of Ranunculus auricomus agg. with phylogenomic analysis of 736 genes and 27,586 SNPs in order to deduce the interrelatedness and hybrid origin of this phenotypically and taxonomically complicated group from Europe characterized by a history of hybridization, polyploidy, apomixis, and recent radiation. The ploidy levels and the reproductive mode of the Northern European populations were assessed via flow cytometric seed screening. In addition, in order to examine the phenotypic plasticity of the dwarf forms previously described as species and summarized as the Ranunculus monophyllus group, we conducted climate chamber experiments under cold (northern) and warm (temperate) conditions. Results: The Northern European populations are tetra- to hexaploid and propagate primarily through apomixis. The complex is characterized by highly reticulate relationships. Genetic differentiation of the main clusters has occurred between the above-mentioned geographical regions. We find evidence for the hybrid origin of the taxa in these areas with differing genomic contributions from the geographically nearest European sexual progenitor species. Furthermore, polyphyly in the taxa of the R. monophyllus group is supported. Experiments show low lability in the traits associated with the R. monophyllus group. Environmental cues, such as temperature and light, are central developmental signals for plants. Due to their sessile lifestyle, they must constantly surveil the environment and adapt accordingly. Ambient temperature fluctuations and light quality can cause abiotic stress and significantly influence plant physiological processes. Consequently, an appropriate response to the environment is pivotal for plant survival. A central mechanism for environmental adaptation, and the appropriate response to abiotic stresses, is based on transcriptomic adjustments, through the regulation of RNA polymerase II and co-transcriptional alternative splicing. One mechanism for the regulation of transcription is the dynamic phosphorylation of the RNA polymerase II C-terminal domain by Cyclin-Dependent Kinases. Co-transcriptional alternative splicing, which describes the dynamic processing of primary RNA transcripts into multiple messenger RNAs, is tightly linked to the regulation of RNA polymerase II. Intriguingly, both processes have been shown to play essential roles in plants’ environmental responses. However, mechanistic insights for many of these processes are still lacking, and it remains unclear how environmental signalling is translated into transcriptomic changes. Additionally, many studies focus on the temperature response of above-ground tissue, while root-specific responses are only poorly studied. By assessing the transcriptomic response, as well as phenotypic alterations, of multiple splicing mutants across a temperature range from 16 °C to 27 °C, we show that alternative splicing reacts to moderated changes in ambient temperature, resulting in extensive transcriptomic changes. Among these, we find many adjustments related to light signalling, regulation of the circadian clock, and temperature signalling. Chlorophyll is essential in photosynthesis, converting sunlight into chemical energy in plants, algae, and certain bacteria. Its structure, featuring a porphyrin ring enclosing a central magnesium ion, varies in forms like chlorophyll a, b, c, d, and f, allowing light absorption at a broader spectrum. With a 20-carbon phytyl tail (except for chlorophyll c), chlorophyll is anchored to proteins. Previous findings suggested the presence of chlorophyll with a modified farnesyl tail in thermophilic cyanobacteria Thermosynechoccocus vestitus. In our Arabidopsis thaliana PSII cryo-EM map, specific chlorophylls showed incomplete phytyl tails, suggesting potential farnesyl modifications. However, further high-resolution mass spectrometry (HRMS) analysis in A. thaliana and T. vestitus did not confirm the presence of any farnesyl tails. Instead, we propose the truncated tails in PSII models may result from binding pocket flexibility rather than actual modifications.

The present study evaluated the repercussions of magnetopriming on the root system architecture of soybean plants subjected to arsenic toxicity using synchrotron radiation source based micro-computed tomography (SR-µCT). This will be used evey where as abbreviation for the technique for three-dimensional imaging. Seeds of soybean were exposed to the static magnetic field (SMF) of strength (200 mT) for 1h prior to sowing. Magnetoprimed and non-primed seeds were grown for 1 month in a soil-sand mixture containing four different levels of sodium arsenate (0, 5, 10, and 50 mg As kg−1 soil). The results showed that arsenic adversely affects the root growth in non-primed plants by reducing their root length, root biomass, root hair, size and number of root nodules, where the damaging effect of As was observed maximum at higher concentrations (10 and 50 mg As kg−1 soil). However, a significant improvement in root morphology was detected in magnetoprimed plants where SMF pretreatment enhanced the root length, root biomass, pore diameter of cortical cells, root hair formation, lateral roots branching, and size of root nodules and girth of primary roots. Qualitative analysis of x-ray micro-CT images showed that arsenic toxicity damaged the epidermal and cortical layers of the root as well as reduced the pore diameter of the cortical cells. However, the diameter of cortical cells pores in magnetoprimed plants was observed higher as compared to plants emerged from non-primed seeds at all level of As toxicity. Thus, the study suggested that magnetopriming has the potential to attenuate the toxic effect of As and could be employed as a pre-sowing treatment to reduce the phytotoxic effects of metal ions in plants by improving root architecture and root tolerance index. This study is the very first exploration of the potential benefits of magnetopriming in mitigating the toxicity of metals (As) in plant roots utilizing the micro-CT technique.

Metacaspases are cysteine proteases present in plants, fungi and protists. While the association of metacaspases with cell death is studied in a range of organisms, their native substrates are largely unknown. Here, we explored the in vivo proteolytic landscape of the two metacaspases, CrMCA-I and CrMCA-II, present in the green freshwater alga Chlamydomonas reinhardtii, using mass spectrometry-based degradomics approach, during control conditions and salt stress. Comparison between the cleavage events of CrMCA-I and CrMCA-II in metacaspase mutants revealed unique cleavage preferences and substrate specificity. Degradome analysis demonstrated the relevance of the predicted metacaspase substrates to the physiology of C. reinhardtii cells and its adaptation during salt stress. Functional enrichment analysis indicated an involvement of CrMCA-I in the catabolism of carboxylic acids, while CrMCA-II plays an important role in photosynthesis and translation. Altogether, our findings suggest distinct cellular functions of the two metacaspases in C. reinhardtii during salt stress response.

BBXs are B-Box zinc finger proteins that can act as transcription factors and regulators of protein complexes. Several BBX proteins play important roles in plant development. Two Arabidopsis thaliana microProteins belonging to the BBX family, named miP1a and miP1b, homotypically interact with and modulate the activity of other BBX proteins, including CONSTANS, which transcriptionally activates the florigen, FLOWERING LOCUS T. Arabidopsis plants overexpressing miP1a and miP1b showed delayed flowering. In tomato, the closest homologs of miP1a and miP1b are the microProteins SlBBX16 and SlBBX17. This study was aimed at investigating whether the constitutive expression of SlBBX16/17 in Arabidopsis and tomato impacted reproductive development. The heterologous expression of the two tomato microProteins in Arabidopsis caused a delay in the flowering transition; however, the effect was weaker than that observed when the native miP1a/b were overexpressed. In tomato, overexpression of SlBBX17 prolonged the flowering period; this effect was accompanied by downregulation of the flowering inhibitors Self Pruning (SP) and SP5G. SlBBX16 and SlBBX17 can hetero-oligomerize with TCMP-2, a cystine-knot peptide involved in flowering pattern regulation and early fruit development in tomato. The increased expression of both microProteins also caused alterations in tomato fruit development: we observed in the case of SlBBX17 a decrease in the number and size of ripe fruits as compared to WT plants, while for SlBBX16, a delay in fruit production up to the breaker stage. These effects were associated with changes in the expression of GA-responsive genes.

The plant endoplasmic reticulum (ER) contacts heterotypic membranes at membrane contact sites (MCSs) through largely undefined mechanisms. For instance, despite the well-established and essential role of the plant ER-chloroplast interactions for lipid biosynthesis, and the reported existence of physical contacts between these organelles, almost nothing is known about the ER-chloroplast MCS identity. Here we show that the Arabidopsis ER membrane-associated VAP27 proteins and the lipid-binding protein ORP2A define a functional complex at the ER-chloroplast MCSs. Specifically, through in vivo and in vitro association assays, we found that VAP27 proteins interact with the outer envelope membrane (OEM) of chloroplasts, where they bind to ORP2A. Through lipidomic analyses, we established that VAP27 proteins and ORP2A directly interact with the chloroplast OEM monogalactosyldiacylglycerol (MGDG), and we demonstrated that the loss of the VAP27-ORP2A complex is accompanied by subtle changes in the acyl composition of MGDG and PG. We also found that ORP2A interacts with phytosterols and established that the loss of the VAP27-ORP2A complex alters sterol levels in chloroplasts. We propose that, by interacting directly with OEM lipids, the VAP27-ORP2A complex defines plant-unique MCSs that bridge ER and chloroplasts and are involved in chloroplast lipid homeostasis.

The boreal forest ecosystems of the northern hemisphere are dominated by conifers, of which Norway spruce (Picea abies [L.] H. Karst.) is one of the most common species. Due to its economic interest to the agroforestry industry, as well as its ecological significance, it is important to understand seasonal growth and biomass production in Norway spruce. Solid evidence that the circadian clock regulates growth in conifers has proved elusive, however, resulting in significant gaps in our knowledge of clock function in these trees. Here, we reassess the impact of the circadian clock on growth in Norway spruce. Using a combination of approaches monitoring the physiology of vegetative growth, transcriptomics and bioinformatics, we determined that the clock could be playing a decisive role in enabling growth, acting in specific developmental processes influenced by season and geographical location to guide bud burst and growth. Thus, the evidence indicates that there is time for spruce.

Kenneth (Ken) Sauer was a mainstay of research in photosynthesis at the University of California, Berkeley and the Lawrence Berkeley National Laboratory (LBNL) for more than 50 years. Ken will be remembered by his colleagues, and other workers in the field of photosynthesis as well, for his pioneering work that introduced the physical techniques whose application have enriched our understanding of the basic reactions of oxygenic photosynthesis. His laboratory was a training ground for many students and postdocs who went on to success in the field of photosynthesis and many others. Trained as a physical chemist, he always brought that quantitative approach to research questions and used several spectroscopic methods in his research. His broad scientific interests concerned the role of manganese in oxygen evolution, electronic properties of chlorophylls, energy transport in antenna complexes, and electron transport reactions. He was also an enthusiastic teacher, an enormously successful mentor who leaves behind a legion of scientists as his abiding legacy, a lover of music and the outdoors with many interests beyond science, and a dedicated family man with a great sense of humility. In this tribute, we summarize some aspects of Ken Sauer’s life and career, illustrated with selected research achievements, and describe his approach to research and life as we perceived it, which is complemented by reminiscences of several current researchers in photosynthesis and other fields. The supporting material includes Ken Sauers’s CV and publication list, as well as a list of the graduate students and postdocs he trained and of researchers that spent a sabbatical in his lab.

Prokaryotic maintenance respiration and associated metabolic activities constitute a considerable proportion of the total respiration of carbon to CO2 in the ocean's mixed layer. However, seasonal influences on prokaryotic maintenance activities in terms of morphological and metabolic adaptations at low (winter) and high productivity (summer) are still unclear. To address this, we examined the natural prokaryotic communities at the mesocosm scale to analyse the differences in their morphological features and gene expression at low and high maintenance respiration, experimentally manipulated with the specific growth rate. Here, we showed that morphological features including membrane blebbing, membrane vesicles, and cell-cell connections occurred under high productivity. Metabolic adaptations associated with maintenance activities were observed under low productivity. Several Kyoto Encyclopedia of Genes and Genomes categories related to signal transduction, energy metabolism, and translational machinery supported maintenance activities under simulated winter conditions. Differential abundances of genes related to transporters, osmoregulation, nitrogen metabolism, ribosome biogenesis, and cold stress were observed. Our results demonstrate how specific growth rate in different seasons can influence resource allocation at the levels of morphological features and metabolic adaptations. This motivates further study of morphological features and their ecological role during high productivity, while investigations of metabolic adaptations during low productivity can advance our knowledge about maintenance activities.

Plants have developed various strategies to deal with abiotic stresses throughout their lifetimes. However, environmental stresses can have long-lasting effects, positively modifying plant physiological responses to subsequent stress episodes, a phenomenon known as preconditioning or stress memory. Intriguingly, this memory can even be transmitted to offspring, referred to as "inter- or transgenerational memory". Chenopodium quinoa is a pseudocereal that can withstand several abiotic stresses, including nitrogen (N) limitation. This research highlights the critical role of maternal N conditions in shaping the physiological and metabolic responses of their offspring. Mother quinoa plants (F0) were grown under High N (HN) or Low N (LN) conditions. LNF0 plants exhibited lower panicle biomass, net photosynthesis, and yield compared to HNF0 plants. Seeds from LNF0 retained proteins, reduced amino acids' levels, and increased lipids (such as PI 34:2), especially phosphatidylcholines, and their unsaturation level, which was associated with faster germination compared to HNF0 seeds. Offsprings seedlings (F1) grown under either HN or LN had similar proteins and amino acid proportions of their seeds. However, LNF0LNF1 seedlings displayed significantly higher biomass and number of root tips. These changes were significantly correlated with transpiration, net photosynthesis, and stomatal conductance, as well as with starch content, suggesting higher CO2 fixation at the whole plant level in LNF0LNF1 plants. Our findings suggest that quinoa transmits maternal environmental stress information to its offspring, modulating their resilience. This work underscores the potential of utilizing maternal environmental conditions as a natural priming tool to enhance crop resilience against nutritional stress.

Somatic embryogenesis (SE) is a powerful model system for studying embryo development and an important method for scaling up availability of elite and climate-adapted genetic material of Norway spruce (Picea abies L. Karst). However, there are several steps during the development of the somatic embryo (Sem) that are suboptimal compared to zygotic embryo (Zem) development. These differences are poorly understood and result in substantial yield losses during plant production, which limits cost-effective large-scale production of SE plants. This study presents a comprehensive data resource profiling gene expression during zygotic and somatic embryo development to support studies aiming to advance understanding of gene regulatory programmes controlling embryo development. Transcriptome expression patterns were analysed during zygotic embryogenesis (ZE) in Norway spruce, including separated samples of the female gametophytes and Zem, and at multiple stages during SE. Expression data from eight developmental stages of SE, starting with pro-embryogenic masses (PEMs) up until germination, revealed extensive modulation of the transcriptome between the early and mid-stage maturing embryos and at the transition of desiccated embryos to germination. Comparative analysis of gene expression changes during ZE and SE identified differences in the pattern of gene expression changes and functional enrichment of these provided insight into the associated biological processes. Orthologs of transcription factors known to regulate embryo development in angiosperms were differentially regulated during Zem and Sem development and in the different zygotic embryo tissues, providing clues to the differences in development observed between Zem and Sem. This resource represents the most comprehensive dataset available for exploring embryo development in conifers.

Pollen, spores, and microscopic charcoal from the sediments of Grosssee (1,619 m a.s.l.), a small lake in the lower subalpine vegetation zone of the Glarus Alps, Switzerland, were analysed to reconstruct vegetation patterns and land use over the past ca. 12,300 calibrated C-14 years bp (cal bp). Pollen data revealed an open landscape covered with grasses and herbs such as Artemisia during the Late Glacial Period. The catchment was likely initially afforested with Betula and Pinus cembra or Pinus sylvestris during the Early Holocene. Thermophilous taxa such as Ulmus, Tilia, and Alnus glutinosa-type expanded from ca. 11,000-9,200 cal bp, and mesophyllic Picea abies and Fagus sylvatica followed, and expanded beginning from ca. 8,000-7,600 cal bp. Interestingly, Alnus viridis (synonym: A. alnobetula) expanded about 2,000 years earlier than at comparable sites in the northern Swiss Alps. Its expansion was profound and persistent, and percentages > 15% were already achieved by ca. 7,000 cal bp. Local erosion events that followed are well explained by vegetation changes and inferred human land use activities at Grosssee. In particular, this led to a more open landscape, and land uses (e.g. grazing) intensified from the Mid- to Late Holocene. Indicators of environmental disturbance including persistently high levels of A. viridis, monolete fern spores, and microscopic charcoal were pronounced after ca. 4,000 cal bp. At that time, high influxes of spores from coprophilous fungi and the consistent presence of cultural indicators (Cerealia-type, Plantago lanceolata) indicate increased grazing and high levels of human impact. Land use and grazing activities seemed to have been particularly pronounced and to have promoted diversity in the vegetation over the past 1,000 years.

In modern agriculture, nano-priming represents an innovative approach, harnessing the power of nanotechnology to enhance crop yields and nutrition. However, to effectively harness the potential of nanoparticles (NPs) for agriculture applications, understanding their mode of action and optimal application rates for positive effects on microgreen growth and physiology is critical. In this interdisciplinary study, we investigated the priming of sunflower seeds with a range of concentrations (25, 50, and 100 mg/L) of ferric oxide (Fe2 O3) nanoparticles (FeNPs) and compared them with control samples. Our findings revealed a significant increase in plant biomass, leaf size, and photosynthetic activity in treated samples. The activities of photosystems I and II increased with higher FeNPs concentration. The treated samples exhibited elevated levels of total phenolics, anthocyanin, and antioxidant enzyme activity, along with increased macronutrients and micronutrients. These findings highlight the potential of FeNPs as a promising tool for enhancing plant growth and physiology in sunflower microgreens.

The genetic control underlying natural variation in lignin content and composition in trees is not fully understood. We performed a systems genetic analysis to uncover the genetic regulation of lignin biosynthesis in a natural ‘SwAsp’ population of aspen (Populus tremula) trees. We analyzed gene expression by RNA sequencing (RNA-seq) in differentiating xylem tissues, and lignin content and composition using Pyrolysis-GC-MS in mature wood of 268 trees from 99 genotypes. Abundant variation was observed for lignin content and composition, and genome-wide association study identified proteins in the pentose phosphate pathway and arabinogalactan protein glycosylation among the top-ranked genes that are associated with these traits. Variation in gene expression and the associated genetic polymorphism was revealed through the identification of 312 705 local and 292 003 distant expression quantitative trait loci (eQTL). A co-expression network analysis suggested modularization of lignin biosynthesis and novel functions for the lignin-biosynthetic CINNAMYL ALCOHOL DEHYDROGENASE 2 and CAFFEOYL-CoA O-METHYLTRANSFERASE 3. PHENYLALANINE AMMONIA LYASE 3 was co-expressed with HOMEOBOX PROTEIN 5 (HB5), and the role of HB5 in stimulating lignification was demonstrated in transgenic trees. The systems genetic approach allowed linking natural variation in lignin biosynthesis to trees´ responses to external cues such as mechanical stimulus and nutrient availability.

Premise: Density-dependent pollinator visitation can lead to density-dependent mating patterns and within-population genetic structure. In Gymnadenia conopsea, individuals in low-density patches receive more self pollen than individuals in high-density patches, suggesting higher relatedness at low density. Ongoing fragmentation is also expected to cause more local matings, potentially leading to biparental inbreeding depression. Methods: To evaluate whether relatedness decreases with local density, we analyzed 1315 SNP loci in 113 individuals within two large populations. We quantified within-population genetic structure in one of the populations, recorded potential habitat barriers, and visualized gene flow using estimated effective migration surfaces (EEMS). We further estimated the magnitude of biparental inbreeding depression that would result from matings restricted to within 5 m. Results: There was no significant relationship between local density and relatedness in any population. We detected significant fine-scale genetic structure consistent with isolation by distance, with positive kinship coefficients at distances below 10 m. Kinship coefficients were low, and predicted biparental inbreeding depression resulting from matings within the closest 5 m was a modest 1–3%. The EEMS suggested that rocks and bushes may act as barriers to gene flow within a population. Conclusions: The results suggest that increased self-pollen deposition in sparse patches does not necessarily cause higher selfing rates or that inbreeding depression results in low establishment success of inbred individuals. The modest relatedness suggests that biparental inbreeding depression is unlikely to be an immediate problem following fragmentation of large populations. The results further indicate that habitat structure may contribute to governing fine-scale genetic structure in G. conopsea.

The nomenclature and taxonomy of Acarospora molybdina is revised using morphological and molecular data. The new combination Myriospora molybdina is proposed and Acarospora hysgina is recognized as a distinct species while A. brunneola is reduced to synonymy with A. hysgina. In its new circumscription M. molybdina is an arctic species, in Scandinavia only occurring in northernmost Norway. Further localities are reported from Greenland, Russia, Svalbard and the USA (Alaska). Acarospora hysgina is the correct name for a species distributed along the west coast of Sweden and Norway, formerly thought to belong to A. molybdina. Localities are also reported from Canada (New Brunswick), Greenland and the USA (Maine). The following names are lectotypified: Acarospora brunneola, A. molybdina var. confusa, Lecanora ereutica beta microcyclos, Parmelia ereutica, P. hysgina and P. molybdina.

Subarctic regions are warming faster than other parts of the globe, and warming is expected to impact carbon (C) assimilation and its allocation into plant biomass and soluble sugars in plant tissues. We analyzed the concentration of soluble sugars (fructose, glucose, and sucrose) in fine roots and rhizomes for three dominant species: Anthoxanthum odoratum, Equisetum spp., and Ranunculus acris. We also examined the concentration and pool of soluble sugars at the plant community level with the aim to investigate the impact of soil warming duration [medium-term (11 years, MTW) vs. long-term (> 60 years, LTW)] and magnitude on soluble sugars in geothermally warmed subarctic grasslands. Among three species, R. acris exhibited the highest concentration of soluble sugars in both fine roots and rhizomes. Comparing total soluble sugar (TSS) between fine roots and rhizomes, rhizomes exhibited a higher concentration in A. odoratum and Equisetum. spp., whereas fine roots had a higher concentration in R. acris. Soil warming did not affect TSS in E. spp. and R. acris, while in A. odoratum, it increased TSS in fine roots and rhizomes in MTW and only in fine roots in LTW. At the plant community level in MTW, soil warming did not affect the soluble sugar concentration in fine roots. However, it increased the TSS and sucrose concentration in rhizomes, which positively correlated with the abundance of grasses. The TSS pool in fine roots decreased with soil warming in MTW, mainly due to a decline in fine root biomass that described 70 % of the decline in the TSS pool. Also, in LTW, soil warming decreased the TSS pool in fine roots, but 74 % of the decline was mainly driven by decreased soluble sugar concentration, specifically that of sucrose, and not by the change in fine root biomass. Seasonal dynamics of root growth play an important role in large-scale ecosystem processes; they are largely governed by growth regulatory compounds and influenced by environmental conditions. Yet, our knowledge about physiological drivers of root growth is mostly limited to laboratory-based studies on model plant species. We sampled root tips of Eriophorum vaginatum and analyzed their auxin concentrations and meristem lengths biweekly over a growing season in situ in a subarctic peatland, both in surface soil and at the permafrost thawfront. Auxin concentrations were almost five times higher in surface than in thawfront soils and increased over the season, especially at the thawfront. Surprisingly, meristem length showed an opposite pattern and was almost double in thawfront compared with surface soils. Meristem length increased from peak to late season in the surface soils but decreased at the thawfront. Our study of in situ seasonal dynamics in root physiological parameters illustrates the potential for physiological methods to be applied in ecological studies and emphasizes the importance of in situ measurements. The strong effect of root location and the unexpected opposite patterns of meristem length and auxin concentrations likely show that auxin actively governs root growth to ensure a high potential for nutrient uptake at the thawfront.

Most animal pollination results from plant–insect interactions, but how we perceive these interactions may differ with the sampling method adopted. The two most common methods are observations of visits by pollinators to plants and observations of pollen loads carried by insects. Each method could favour the detection of different species and interactions, and pollen load observations typically reveal more interactions per individual insect than visit observations. Moreover, while observations concern plant and insect individuals, networks are frequently analysed at the level of species. Although networks constructed using visitation and pollen-load data have occasionally been compared in relatively specialised, bee-dominated systems, it is not known how sampling methodology will affect our perception of how species (and individuals within species) interact in a more generalist system. Here we use a Diptera-dominated high-Arctic plant–insect community to explore how sampling approach shapes several measures of species' interactions (focusing on specialisation), and what we can learn about how the interactions of individuals relate to those of species. We found that species degrees, interaction strengths, and species motif roles were significantly correlated across the two method-specific versions of the network. However, absolute differences in degrees and motif roles were greater than could be explained by the greater number of interactions per individual provided by the pollen-load data. Thus, despite the correlations between species roles in networks built using visitation and pollen-load data, we infer that these two perspectives yield fundamentally different summaries of the ways species fit into their communities. Further, individuals' roles generally predicted the species' overall role, but high variability among individuals means that species' roles cannot be used to predict those of particular individuals. Nitrogen can be taken up by trees in the form of nitrate, ammonium and amino acids, but the influence of the different forms on tree growth and development is poorly understood in angiosperm species like Populus. We studied the effects of both organic and inorganic forms of nitrogen on growth and wood formation of hybrid aspen trees in experimental conditions that allowed growth under four distinct steady-state nitrogen levels. Increased nitrogen availability had a positive influence on biomass accumulation and the radial dimensions of both xylem vessels and fibers, and a negative influence on wood density. An optimal level of nitrogen availability was identified where increases in biomass accumulation outweighed decreases in wood density. None of these responses depended on the source of nitrogen except for shoot biomass accumulation, which was stimulated more by treatments complemented with nitrate than by ammonium alone or the organic source arginine. The most striking difference between the nitrogen sources was the effect on lignin composition, whereby the abundance of H-type lignin increased only in the presence of nitrate. The differential effect of nitrate is possibly related to the well-known role of nitrate as a signaling compound. RNA-sequencing revealed that while the lignin-biosynthetic genes did not significantly (FDR <0.01) respond to added NO3- , the expression of several laccases, catalysing lignin polymerization, was dependent on N-availability. These results reveal a unique role of nitrate in wood formation and contribute to the knowledge basis for decision-making in utilizing hybrid aspen as a bioresource.

Plants tightly control growth of their lateral organs, which led to the concept of apical dominance. However, outgrowth of the dormant lateral primordia is sensitive to the plant's nutritional status, resulting in an immense plasticity in plant architecture. While the impact of hormonal regulation on apical dominance is well characterized, the prime importance of sugar signaling to unleash lateral organ formation has just recently emerged. Here, we aimed to identify transcriptional regulators, which control the trade-off between growth of apical versus lateral organs. Making use of locally inducible gain-of-function as well as single and higher-order loss-of-function approaches of the sugar-responsive S1-basic-leucine-zipper (S1-bZIP) transcription factors, we disclosed their largely redundant function in establishing apical growth dominance. Consistently, comprehensive phenotypical and analytical studies of S1-bZIP mutants show a clear shift of sugar and organic nitrogen (N) allocation from apical to lateral organs, coinciding with strong lateral organ outgrowth. Tissue-specific transcriptomics reveal specific clade III SWEET sugar transporters, crucial for long-distance sugar transport to apical sinks and the glutaminase GLUTAMINE AMIDO-TRANSFERASE 1_2.1, involved in N homeostasis, as direct S1-bZIP targets, linking the architectural and metabolic mutant phenotypes to downstream gene regulation. Based on these results, we propose that S1-bZIPs control carbohydrate (C) partitioning from source leaves to apical organs and tune systemic N supply to restrict lateral organ formation by C/N depletion. Knowledge of the underlying mechanisms controlling plant C/N partitioning is of pivotal importance for breeding strategies to generate plants with desired architectural and nutritional characteristics.

Quinoa is an agriculturally important crop species originally domesticated in the Andes of central South America. One of its most important phenotypic traits is seed colour. Seed colour variation is determined by contrasting abundance of betalains, a class of strong antioxidant and free radicals scavenging colour pigments only found in plants of the order Caryophyllales. However, the genetic basis for these pigments in seeds remains to be identified. Here we demonstrate the application of machine learning (extreme gradient boosting) to identify genetic variants predictive of seed colour. We show that extreme gradient boosting outperforms the classical genome-wide association approach. We provide re-sequencing and phenotypic data for 156 South American quinoa accessions and identify candidate genes potentially controlling betalain content in quinoa seeds. Genes identified include novel cytochrome P450 genes and known members of the betalain synthesis pathway, as well as genes annotated as being involved in seed development. Our work showcases the power of modern machine learning methods to extract biologically meaningful information from large sequencing data sets.
