Low iron (Fe) bioavailability can limit the biosynthesis of Fe-containing proteins, which are especially abundant in photosynthetic organisms, thus negatively affecting global primary productivity. Understanding cellular coping mechanisms under Fe limitation is therefore of great interest. We surveyed the temporal responses of Chlamydomonas (Chlamydomonas reinhardtii) cells transitioning from an Fe-rich to an Fe-free medium to document their short and long-term adjustments. While slower growth, chlorosis and lower photosynthetic parameters are evident only after one or more days in Fe-free medium, the abundance of some transcripts, such as those for genes encoding transporters and enzymes involved in Fe assimilation, change within minutes, before changes in intracellular Fe content are noticeable, suggestive of a sensitive mechanism for sensing Fe. Promoter reporter constructs indicate a transcriptional component to this immediate primary response. With acetate provided as a source of reduced carbon, transcripts encoding respiratory components are maintained relative to transcripts encoding components of photosynthesis and tetrapyrrole biosynthesis, indicating metabolic prioritization of respiration over photosynthesis. In contrast to the loss of chlorophyll, carotenoid content is maintained under Fe limitation despite a decrease in the transcripts for carotenoid biosynthesis genes, indicating carotenoid stability. These changes occur more slowly, only after the intracellular Fe quota responds, indicating a phased response in Chlamydomonas, involving both primary and secondary responses during acclimation to poor Fe nutrition.

The nonequilibrium Fermi's golden rule (NE-FGR) approach is developed to simulate the electronic transitions between multiple excited states in complex condensed-phase systems described by the recently proposed multi-state harmonic (MSH) model Hamiltonian. The MSH models were constructed to faithfully capture the photoinduced charge transfer dynamics in a prototypical organic photovoltaic carotenoid-porphyrin-C60 molecular triad dissolved in tetrahydrofuran. A general expression of the fully quantum-mechanical NE-FGR rate coefficients for transitions between all pairs of states in the MSH model is obtained. Besides, the linearized semiclassical NE-FGR formula and a series of semiclassical approximations featuring Wigner and classical nuclear sampling choices and different dynamics during the quantum coherence period for the MSH model are derived. The current approach enables all the possible population transfer pathways between the excited states of the triad, in contrast to the previous applications that only addressed the donor-to-acceptor transition. Our simulations for two triad conformations serve as a demonstration for benchmarking different NE-FGR approximations and show that the difference between all levels of approximation is small for the current system, especially at room temperature. By comparing with nonadiabatic semiclassical dynamics, we observe similar timescales for the electronic population transfer predicted by NE-FGR. The retinal light response in animals originates from the photoisomerization of an opsin-coupled 11-cis-retinaldehyde chromophore. This visual chromophore is enzymatically produced through the action of carotenoid cleavage dioxygenases. Vertebrates require two carotenoid cleavage dioxygenases, β-carotene oxygenase 1 and retinal pigment epithelium 65 (RPE65), to form 11-cis-retinaldehyde from carotenoid substrates, whereas invertebrates such as insects use a single enzyme known as Neither Inactivation Nor Afterpotential B (NinaB). RPE65 and NinaB couple trans-cis isomerization with hydrolysis and oxygenation, respectively, but the mechanistic relationship of their isomerase activities remains unknown. Here we report the structure of NinaB, revealing details of its active site architecture and mode of membrane binding. Structure-guided mutagenesis studies identify a residue cluster deep within the NinaB substrate-binding cleft that controls its isomerization activity. Our data demonstrate that isomerization activity is mediated by distinct active site regions in NinaB and RPE65-an evolutionary convergence that deepens our understanding of visual system diversity.

Vitamin A deficiency remains prevalent on a global scale, including in regions where maize constitutes a high percentage of human diets. One solution for alleviating this deficiency has been to increase grain concentrations of provitamin A carotenoids in maize (Zea mays ssp. mays L.)-an example of biofortification. The International Maize and Wheat Improvement Center (CIMMYT) developed a Carotenoid Association Mapping panel of 380 inbred lines adapted to tropical and subtropical environments that have varying grain concentrations of provitamin A and other health-beneficial carotenoids. Several major genes have been identified for these traits, 2 of which have particularly been leveraged in marker-assisted selection. This project assesses the predictive ability of several genomic prediction strategies for maize grain carotenoid traits within and between 4 environments in Mexico. Ridge Regression-Best Linear Unbiased Prediction, Elastic Net, and Reproducing Kernel Hilbert Spaces had high predictive abilities for all tested traits (β-carotene, β-cryptoxanthin, provitamin A, lutein, and zeaxanthin) and outperformed Least Absolute Shrinkage and Selection Operator. Furthermore, predictive abilities were higher when using genome-wide markers rather than only the markers proximal to 2 or 13 genes. These findings suggest that genomic prediction models using genome-wide markers (and assuming equal variance of marker effects) are worthwhile for these traits even though key genes have already been identified, especially if breeding for additional grain carotenoid traits alongside β-carotene. Predictive ability was maintained for all traits except lutein in between-environment prediction. The TASSEL (Trait Analysis by aSSociation, Evolution, and Linkage) Genomic Selection plugin performed as well as other more computationally intensive methods for within-environment prediction. The findings observed herein indicate the utility of genomic prediction methods for these traits and could inform their resource-efficient implementation in biofortification breeding programs.

Cyanobacteria uniquely contain a primitive water-soluble carotenoprotein, the orange carotenoid protein (OCP). Nearly all extant cyanobacterial genomes contain genes for the OCP or its homologs, implying an evolutionary constraint for cyanobacteria to conserve its function. Genes encoding the OCP and its two constituent structural domains, the N-terminal domain, helical carotenoid proteins (HCPs), and its C-terminal domain, are found in the most basal lineages of extant cyanobacteria. These three carotenoproteins exemplify the importance of the protein for carotenoid properties, including protein dynamics, in response to environmental changes in facilitating a photoresponse and energy quenching. Here, we review new structural insights for these carotenoproteins and situate the role of the protein in what is currently understood about their functions.

Mutants lacking functional HYD2 homoeologs showed improved seedling growth, but comparable or increased susceptibility to salt stress in tillering plants, suggesting a developmentally restricted role of HYD2 in salt response. Salinity stress threatens global food security by reducing the yield of staple crops such as wheat (Triticum ssp.). Understanding how wheat responds to salinity stress is crucial for developing climate resilient varieties. In this study, we examined the interplay between carotenoid metabolism and the response to salt (NaCl) stress, a specific form of salinity stress, in tetraploid wheat plants with mutations in carotenoid β-hydroxylase 1 (HYD1) and HYD2. Our investigation encompassed both the vulnerable seedling stage and the more developed tillering stage of wheat plant growth. Mutant combinations lacking functional HYD2 homoeologs, including hyd-A2 hyd-B2, hyd-A1 hyd-A2 hyd-B2, hyd-B1 hyd-A2 hyd-B2, and hyd-A1 hyd-B1 hyd-A2 hyd-B2, had longer first true leaves and slightly enhanced root growth during germination under salt stress compared to the segregate wild-type (control) plants. Interestingly, these mutant seedlings also showed decreased levels of neoxanthin and violaxanthin (xanthophylls derived from β-carotene) and an increase in β-carotene in roots. However, tillering hyd mutant and segregate wild-type plants generally did not differ in their height, tiller count, and biomass production under acute or prolonged salt stress, except for decreases in these parameters observed in the hyd-A1 hyd-B1 hyd-A2 hyd-B2 mutant that indicate its heightened susceptibility to salt stress. Taken together, these findings suggest a significant, yet developmentally restricted role of HYD2 homoeologs in salt-stress response in tetraploid wheat. They also show that hyd-A2 hyd-B2 mutant plants, previously demonstrated for possessing enriched nutritional (β-carotene) content, maintain an unimpaired ability to withstand salt stress.

High-density lipoproteins (HDLs) are key regulators of cellular cholesterol homeostasis but are functionally altered in many chronic diseases. The factors that cause HDL functional loss in chronic disease are not fully understood. It is also unknown what roles antioxidant carotenoids play in protecting HDL against functional loss. The aim of this study was to measure how various disease-associated chemical factors including exposure to (1) Cu2+ ions, (2) hypochlorous acid (HOCL), (3) hydrogen peroxide (H2O2), (4) sialidase, (5) glycosidase, (6) high glucose, (7) high fructose, and (8) acidic pH, and the carotenoid antioxidants (9) lutein and (10) zeaxanthin affect HDL functionality. We hypothesized that some of the modifications would have stronger impacts on HDL particle structure and function than others and that lutein and zeaxanthin would improve HDL function. HDL samples were isolated from generally healthy human plasma and incubated with the corresponding treatments listed above. Cholesterol efflux capacity (CEC), lecithin-cholesterol acyl transferase (LCAT) activity, and paraoxonase-1 (PON1) activity were measured in order to determine changes in HDL functionality. Median HDL particle diameter was increased by acidic pH treatment and reduced by HOCl, high glucose, high fructose, N-glycosidase, and lutein treatments. Acidic pH, oxidation, and fructosylation all reduced HDL CEC, whereas lutein, zeaxanthin, and sialidase treatment improved HDL CEC. LCAT activity was reduced by acidic pH, oxidation, high fructose treatments, and lutein. PON1 activity was reduced by sialidase, glycosidase, H2O2, and fructose and improved by zeaxanthin and lutein treatment. These results show that exposure to oxidizing agents, high fructose, and low pH directly impairs HDL functionality related to cholesterol efflux and particle maturation, whereas deglycosylation impairs HDL antioxidant capacity. On the other hand, the antioxidants lutein and zeaxanthin improve or preserve both HDL cholesterol efflux and antioxidant activity but have no effect on particle maturation.

We develop a theory of singlet fission in carotenoid dimers. Following photoexcitation of the "bright" state (i.e., a singlet electron–hole pair) in a single carotenoid, the first step in the singlet fission process is ultrafast intramolecular conversion into the highly correlated "dark" (or 2Ag) state. This state has both entangled singlet triplet-pair and charge-transfer character. Our theory is predicated on the assumption that it is the singlet triplet-pair component of the "dark" state that undergoes bimolecular singlet fission. We use valence bond theory to develop a minimal two-chain model of the triplet-pair states. The single and double chain triplet-pair spectra are described, as this helps explain the dynamics and the equilibrated populations. We simulate the dynamics of the initial entangled pair state using the quantum Liouville equation, including both spin-conserving and spin-nonconserving dephasing processes. By computing the intrachain and interchain singlet, triplet, and quintet triplet-pair populations, we show that singlet fission critically depends on the interchain coupling and the driving potential (that determines endothermic vs exothermic fission). In this study, the effects of combined stress factors involving light intensity and salinity (NaCl, MgCl2, CaCl2, and their combinations) on the two-stage cultivation of Chlorella vulgaris for simultaneous production of biomass and high-value products, were investigated. The two-stage strategies comprised a 25-day vegetative stage in BG11 medium, followed by a 15-day combined stress stage. During salt stress conditions, the addition of 15 g L−1 CaCl2 or 7.5 g L−1 MgCl2 / 7.5 g L−1 CaCl2 mixture with 140 µmol m−2 s−1 light intensity significantly promoted the growth of C. vulgaris achieving maximum biomass productivity of 50.50 ± 0.50 and 50.25 ± 3.25 mg L−1 d−1, respectively. Cultivation of C. vulgaris in a medium containing 7.5 g L−1 NaCl/ 7.5 g L−1 CaCl2 had remarkably increased the lipid content (31.15 ± 1.18%) and lipid productivity (14.55 ± 1.48 mg L−1 d−1). The saturated fatty acids (SFAs) at 39.52–59.29%, monounsaturated fatty acids (MUFAs) at 27.16–35.47%, and polyunsaturated fatty acids (PUFAs) at 7.18–29.97%, were obtained with palmitic (C16:0), oleic (C18:1), stearic (C18:0), and linolenic (C18:3) acids as predominant fatty acids. Cultures supplemented with 5 g L−1 NaCl / 5 g L−1 MgCl2 / 5 g L−1 CaCl2 and high light intensity exposure attained consistently high carbohydrate content (52.71 ± 2.50%). The combination of 7.5 g L−1 NaCl / 7.5 g L−1 MgCl2 also resulted in a marked increase in the protein content (35.32 ± 2.20%) and total carotenoids (0.31 ± 0.03 μg mL−1) as compared to the Controls. The highest antioxidant activity (86.16%) was achieved with a 7.5 g L−1 NaCl / 7.5 g L−1 CaCl2 combination in the growth stage. The antioxidant activities were attributed to the presence of phenolics, flavonoids, and tannins due to the stressed conditions. Summary Development of health‐promoting natural products has received importance, and fermented pumpkin can be a viable option, mainly regarding its carotenoid contents. This study aimed to investigate the quality of fermented pumpkin beverages (FPB) under various fermentation conditions (control, T1 = 30 °C, 12 h; T2 = 30 °C, 18 h; T3 = 30 °C, 24 h; T4 = 40 °C, 12 h; T5 = 40 °C, 18 h; T6 = 40 °C, 24 h) by Lactobacillus plantarum. Cell count of lactic acid bacteria, pH, chemical quality (soluble solid, acidity, total sugar, total phenolic content, total flavonoid content, and antioxidant activity), carotenoid contents, and colour attributes (h*, chroma, L*, and yellowing index) of FPB samples were studied. As a result, LAB fermentation significantly affected acidity and total sugar of FPB samples, while it did not change pH and soluble solids significantly. Besides, carotenoid contents and colour attributes of FPB were not different during fermentation, indicating that L. plantarum fermentation retained carotenoids and maintained physical appearances. Regarding sugars and organic acids, their quantity changed due to fermentation and lactic acids increased greatly up to three times higher than control after 24 h of fermentation at 30 °C. Moreover, fermentation by L. plantarum improved total phenolic content, total flavonoid content, and antioxidant activity, and these may recommend FPB as a potential functional product. The human body can easily convert β‐carotene into retinol (a precursor of vitamin A), a significant ingredient in dietary supplements. The need for commercial natural pigment production, including β‐carotene, has led to intensive research in this area. Processing after biosynthesis is a vital stage that involves the use of nontoxic solvents, efficient mechanical disruption of cells, and the isolation of the required compounds. With the growing popularity of green extraction methods and the increasing market demand for carotenoids due to their health benefits, the use of clean, nontoxic, flash‐hydrolyzed biomass for carotenoid production presents a significant advantage. Furthermore, the remaining biomass can serve as a protein source for animal feed, thereby aligning with the principles of the biorefinery concept. In this study, our aim was to optimize the disruption of cells in two microalgal species using flash hydrolysis (FH). We applied two distinct levels of pressure and four separate temperature conditions over a brief residence time. Following this, we employed an ethanol extraction method to assess the efficiency of carotenoid pigment extraction. Flash hydrolysis is a chemical‐free subcritical water‐based continuous‐flow process, injected with wet biomass slurry at a high temperature (150–250 °C) for a very short time (8–12 s) to obtain bioproducts. In this study, we discovered that the pigment β‐carotene constituted 0.04 to 0.3% of the dry biomass in the case of Chlorella vulgaris. Bands of lutein and zeaxanthin were also observed in the thin layer chromatography (TLC) analysis of the treated slurry. This was achieved under conditions of an effective temperature of 200 °C and a pressure of 1700 psi. The Arthrospira platensis (1Tex), β‐carotene yield was 0.014 to 0.021% of dry biomass. Some lutein and zeaxanthin bands were also found in the treated slurry with an effective temperature of 150 °C and 1000 psi pressure. This study reports pigment extraction by FH for the first time. It is a viable process to scale up high‐value natural products like carotenoids from microalgae at the industrial level without adverse environmental impacts. Due to low residence time, the temperature does not have a negative effect on pigments; cell rupture was accomplished effectively using water as a solvent. A considerable amount of pigment could be recovered from microalgae at specific combinations of temperature and pressure depending on the type of cell wall that microalgae possess. Context Although the nutritional composition of organic food has been thoroughly researched, there is a dearth of published data relating to its impact on human health. Objective This systematic review aimed to examine the association between organic food intake and health effects, including changes in in vivo biomarkers, disease prevalence, and functional changes. Data Sources PubMed, EMBASE, Web of Science, the Cochrane Library, and ClinicalTrials.gov were searched from inception through Nov 13, 2022. Data Extraction Both observational and interventional studies conducted in human populations were included, and association between level of organic food intake and each outcome was quantified as "no association," "inconsistent," "beneficial correlation/harmful correlation," or "insufficient". For outcomes with sufficient data reported by at least 3 studies, meta-analyses were conducted, using random-effects models to calculate standardized mean differences. Data Analysis Based on the included 23 observational and 27 interventional studies, the association between levels of organic food intake and (i) pesticide exposure biomarker was assessed as "beneficial correlation," (ii) toxic metals and carotenoids in the plasma was assessed as "no association," (iii) fatty acids in human milk was assessed as "insufficient," (iv) phenolics was assessed as "beneficial", and serum parameters and antioxidant status was assessed as "inconsistent". For diseases and functional changes, there was an overall "beneficial" association with organic food intake, and there were similar findings for obesity and body mass index. However, evidence for association of organic food intake with other single diseases was assessed as "insufficient" due to the limited number and extent of studies. Conclusion Organic food intake was found to have a beneficial impact in terms of reducing pesticide exposure, and the general effect on disease and functional changes (body mass index, male sperm quality) was appreciable. Anoxygenic phototrophic bacteria (green and purple sulfur bacteria) thrive in anoxic environments where light penetrates a sulfide-containing (euxinic) water column. Genomic data and photosynthetic bacterial carotenoid pigments should provide complementary information on the spatio-temporal dynamics of anoxygenic phototrophs in modern euxinic environments. In turn, these contemporary depositional settings often serve as analogues for ancient counterparts. However, in some modern environments, DNA-informed patterns of phototrophic sulfur bacteria occurrence do not match distributions of their carotenoid inventories. One possible explanation for these seemingly incompatible observations is that the rapid sulfurization of carotenoids and incorporation into macromolecules via multiple carbon–sulfur bonds prevents or confounds their detection by conventional means. Here, to evaluate this conundrum, we revisit some representative contemporary euxinic environments where anoxygenic phototrophic bacteria have mostly been detected based on genomic analyses. Although free intact carotenoids are sporadically detected in surface sediments, their distributions do not reveal a complete picture. Exogenously sourced fossil carotenoids (e.g., paleorenieratane) is an additional complication. Carotenoid inventories obtained by desulfurization with Raney nickel, on the other hand, stand in stark contrast to those present as free lipids. In particular, sulfur-linked carotenoids present in euxinic lake sediments provide a more complete picture of compositions of anoxygenic sulfur bacterial communities and account for discrepancies reported in previous studies. We observe a closer alignment between genomic data and patterns of sulfurized carotenoids and, importantly, our results highlight how sulfurization serves as a pathway for the rapid modification of highly functionalised lipids and their sequestration into the macromolecular component of sediment extracts. Bacterial pigments stand out as exceptional natural bioactive compounds with versatile functionalities. The pigments represent molecules from distinct chemical categories including terpenes, terpenoids, carotenoids, pyridine, pyrrole, indole, and phenazines, which are synthesized by diverse groups of bacteria. Their spectrum of physiological activities encompasses bioactive potentials that often confer fitness advantages to facilitate the survival of bacteria amid challenging environmental conditions. A large proportion of such pigments are produced by bacterial pathogens mostly as secondary metabolites. Their multifaceted properties augment potential applications in biomedical, food, pharmaceutical, textile, paint industries, bioremediation, and in biosensor development. Apart from possessing a less detrimental impact on health with environmentally beneficial attributes, tractable and scalable production strategies render bacterial pigments a sustainable option for novel biotechnological exploration for untapped discoveries. The review offers a comprehensive account of physiological role of pigments from bacterial pathogens, production strategies, and potential applications in various biomedical and biotechnological fields. Genetically engineered cyanobacterial strains that have improved growth rate, biomass productivity, and metabolite productivity could be a better option for sustainable bio-metabolite production. The global demand for biobased metabolites with nutraceuticals and health benefits has increased due to their safety and plausible therapeutic and nutritional utility. Cyanobacteria are solar-powered green cellular factories that can be genetically tuned to produce metabolites with nutraceutical and pharmaceutical benefits. The present review discusses biotechnological endeavors for producing bioprospective compounds from genetically engineered cyanobacteria and discusses the challenges and troubleshooting faced during metabolite production. This review explores the cyanobacterial versatility, the use of engineered strains, and the techno-economic challenges associated with scaling up metabolite production from cyanobacteria. Challenges to produce cyanobacterial bioactive compounds with remarkable nutraceutical values have been discussed. Microalgae are a source of a wide variety of commodities, including particularly valuable pigments. The typical pigments present in microalgae are the chlorophylls, carotenoids, and phycobiliproteins. However, other types of pigments, of the family of water-soluble polyphenols, usually encountered in terrestrial plants, have been recently reported in microalgae. Among such microalgal polyphenols, many flavonoids have a yellowish hue, and are used as natural textile dyes. Besides being used as natural colorants, for example in the food or cosmetic industry, microalgal pigments also possess many bioactive properties, making them functional as nutraceutical or pharmaceutical agents. Each type of pigment, with its own chemical structure, fulfills particular biological functions. Considering both eukaryotes and prokaryotes, some species within the four most promising microalgae groups (Cyanobacteria, Rhodophyta, Chlorophyta and Heterokontophyta) are distinguished by their high contents of specific added-value pigments. To further enhance microalgae pigment contents during autotrophic cultivation, a review is made of the main related strategies adopted during the last decade, including light adjustments (quantity and quality, and the duration of the photoperiod cycle), and regard to mineral medium characteristics (salinity, nutrients concentrations, presence of inductive chemicals). In contrast to what is usually observed for growth-related pigments, accumulation of non-photosynthetic pigments (polyphenols and secondary carotenoids) requires particularly stressful conditions. The effect of the storage conditions of light (presence; absence) and relative humidity (50%; 75% RH) on the water content and colour of Porphyridium cruentum microalga powder was studied, then two packaging materials, low-density polyethylene (PE-LD) and metallised polyethylene terephthalate with polyethylene (PETmet/PE), were used to study their effect on the water content, colour, total carotenoids and chlorophylls contents, total phenolic content (TPC), and antioxidant activity (ABTS, DPPH, ORAC) of P. cruentum during storage at 23 °C and 50% RH. An atmosphere with 50% RH is recommended to minimise the water vapour absorption and lightness loss. In addition to the expected light protection, PETmet/PE material protected P. cruentum powder also against water absorption, contrary to PE material. There were no relevant colour changes of the microalga during storage in PETmet/PE. Regarding the total content of carotenoids and the antioxidant activity of the microalga, no significant differences were found between the biomasses stored in either packaging material. The TPC and DPPH values were practically constant during two months of storage, while ABTS and ORAC decreased more than 50% and 20%, respectively, during this time. [ABSTRACT FROM AUTHOR]

Vitamin A deficiency and its associated disorders are pervasive in sub‐Saharan Africa (SSA) including many middle‐ and low‐income countries across the world. Provitamin A‐enriched maize (Zea mays L.) inbred lines with desirable agronomic and adaptive traits have been developed and used to generate and commercialize maize varieties with medium to high levels of provitamin A in a few countries to curb vitamin A deficiency. Nonetheless, these inbred lines have not been made widely available to the public and private sector breeders in many countries. The main purpose for releasing the 21 provitamin A‐enriched tropical maize inbred lines (PI 705424–PI 705444, Reg. nos. GP‐624–GP‐644) is to supply maize breeders with elite source germplasm for increasing provitamin A and other carotenoids to much higher levels to offset losses during storage, natural degradation, and processing. These inbred lines were developed at the International Institute of Tropical Agriculture (IITA) from backcrosses of high β‐carotene temperate lines as donors and elite tropical lines as recipients. These inbred lines were developed through repeated self‐pollination with rigorous visual selection among and within lines for plant vigor, synchronous silk emergence and pollen shedding, low ear placement, and resistance to lodging and major tropical diseases, followed by selection for bright yellow to orange kernel color with semi flint to flint kernel texture after harvest. The released maize inbred lines will be diverse sources of favorable alleles to accelerate genetic gain in provitamin A and other beneficial carotenoid enrichment for human health. To understand the effects and related potential mechanism of H2O2 on pigment metabolism in postharvest broccoli, an integrated analysis of transcriptome and metabolome was performed. Results suggested that 65 differentially expressed genes and 26 differentially accumulated metabolites involved in chlorophyll, carotenoid, and flavonoid metabolism were identified. H2O2 treatment delayed the decrease of chlorophyll content by upregulating the expressions of chlorophyll synthetic genes, thylakoid synthetic genes, and 15 light‐harvesting complex genes compared with the control and diphenylene iodonium treatments. H2O2 treatment decreased the accumulation of 11 flavonoids and 5 flavonols by downregulating the flavonoid synthetic genes. In addition, H2O2 treatment promoted carotenoid biosynthesis to eliminate reactive oxygen species in thylakoids, thereby protecting chlorophyll molecules from degradation. The inhibition of flavonoids and flavonols accumulation and chlorophyll decrease was the crucial reason for the delayed yellowing in H2O2 treatment. Microgreens and baby leaves are increasingly appreciated and consumed due to the increased concentrations of antioxidant compounds, vitamins, nutrients, and minerals than adult plants. The objective was to evaluate the effect of different substrates and protected environments on the growth of microgreen and baby leaf kale (Brassica oleracea var. sabellica L.) and their contents of carotenoids and chlorophylls. The experiments were carried out in a completely randomized design in a factorial arrangement of 4 × 2 (four environments × two substrates) for the microgreens and 4 × 5 (four environments × five substrates) for the baby leaf, both with six replications. For microgreens, the Carolina Soil® commercial substrate promoted greater height, fresh matter, dry matter, productivity, chlorophyll a, chlorophyll b, total chlorophyll, and carotenoids in all evaluated environments. In the greenhouse with a screen of 35% shading under the film, greater height, fresh matter, dry matter, and productivity were obtained, and in the greenhouse with a screen of 42/50% shading under the film, higher contents of microgreen chlorophylls and carotenoids were obtained. For baby leaves, the greenhouse with 42/50% shading screen under the film promoted plants with greater height, fresh matter, dry matter, and productivity in all substrates. The baby leaf grown in the greenhouse with a screen of 35% shading under the film showed higher chlorophyll and carotenoid contents in the pure Carolina Soil® substrate. For greater productivity, with higher contents of pigments, greenhouses with screens under the film and the use of organomineral commercial substrate are the most suitable for microgreen and baby leaf kale. Background Aim Methods Results Conclusion The investigations of snail slime and its possible biological activities have been performed recently.This study aimed to evaluate the properties of Helix aspersa snail slime (HAS), and carotenoids‐fortified slime (HASC).Snails were fed with carrots for 10 days. Slime was collected and extracted using water. Saponins identification, antioxidant, collagenase, and tyrosinase enzyme inhibition activities, carotenoids, total phenols, and flavonoids content, were determined. UHPLC–MS/MS analysis was performed for phytochemical characterization.Saponins were detected in the HAS extract only. However, HASC was shown to contain a higher carotenoid content than HAS (29.51 ± 0.4 vs. 18.11 ± 0.2 μg/g). similarly, total phenolic and flavonoid content were higher for the the HASC extract compared to the HAS (182.3 ± 5.2 vs. 150.28 ± 3.3 mg/g equivalent to gallic acid), and (77.62 ± 1.2 vs. 14.19 ± 0.9 mg/g equivalent to quercetin). As expected, the HASC extract exhibited higher antioxidant activity compared to the HAS, using DPPH and the ABTS assays (IC50 = 7.75 ± 0.14 vs. 20.1 ± 0.4 μg/mL), and (IC50 = 7.6 ± 0.26 vs. 19.57 ± 1.4 μg/mL). UHPLC–MS/MS analysis revealed the presence of several phytocomponents of which, hexadecanoic acid and ascorbic acid, were observed in the HASC extract. Furthermore, HASC extract exhibited superior inhibitory activity compared to HAS against collagenase and tyrosinase enzymes (IC50 = 8.4 ± 1.19 vs. 15.3 ± 1.12 μg/mL) and (IC50 = 30.1 ± 0.91 vs. 35 ± 1.3 μg/mL).These findings highlight the potential of HASC as a valuable ingredient in various pharmaceutical applications, due to their content of various phenolic, antioxidants, carotenoids, hexadecanoic, and ascorbic acids. Alzheimer's disease (AD) is characterized by, among other things, dementia and a decline in cognitive performance. In AD, dementia has neurodegenerative features and starts with mild cognitive impairment (MCI). Research indicates that apoptosis and neuronal loss occur in AD, in which oxidative stress plays an important role. Therefore, reducing oxidative stress with antioxidants is a natural strategy to prevent and slow down the progression of AD. Carotenoids are natural pigments commonly found in fruits and vegetables. They include lipophilic carotenes, such as lycopene, α- and β-carotenes, and more polar xanthophylls, for example, lutein, zeaxanthin, canthaxanthin, and β-cryptoxanthin. Carotenoids can cross the blood–brain barrier (BBB) and scavenge free radicals, especially singlet oxygen, which helps prevent the peroxidation of lipids abundant in the brain. As a result, carotenoids have neuroprotective potential. Numerous in vivo and in vitro studies, as well as randomized controlled trials, have mostly confirmed that carotenoids can help prevent neurodegeneration and alleviate cognitive impairment in AD. While carotenoids have not been officially approved as an AD therapy, they are indicated in the diet recommended for AD, including the consumption of products rich in carotenoids. This review summarizes the latest research findings supporting the potential use of carotenoids in preventing and alleviating AD symptoms. A literature review suggests that a diet rich in carotenoids should be promoted to avoid cognitive decline in AD. One of the goals of the food industry should be to encourage the enrichment of food products with functional substances, such as carotenoids, which may reduce the risk of neurodegenerative diseases. This in vivo study performed in rat adjuvant arthritis aims to advance the understanding of astaxanthin's therapeutic properties for the possible treatment of rheumatoid arthritis (RA) in monotherapy and along with the standard RA treatment, methotrexate (MTX), in combination therapy. The main goal was to elucidate astaxanthin's full therapeutic potential, evaluate its dose dependency, and compare its effects in monotherapy with other carotenoids such as β-carotene and β-cryptoxanthin (KXAN). Moreover, potential differences in therapeutic activity caused by using different sources of astaxanthin, synthetic (ASYN) versus isolated from Blakeslea trispora (ASTAP), were evaluated using one-way ANOVA (Tukey-Kramer post hoc test). KXAN was the most effective in reducing plasma MMP-9 levels in monotherapy, significantly better than MTX, and in reducing hind paw swelling. The differences in the action of ASTAP and ASYN have been observed across various biometric, anti-inflammatory, and antioxidative parameters. In combined therapy with MTX, the ASYN + MTX combination proved to be better. Summary Co‐digestion of cantaloupe (C) melon fresh cuts (FC) coated with probiotics [Lactiplantibacillus plantarum 75/xanthan gum with Natal plum (NP)] or beetroot leaves (BL) was evaluated to determine their bioaccessibility of individual anthocyanins (cyanidin‐3‐glucoside, cyanidin‐3‐O‐sambubioside) and carotenoids (lutein, trans‐β‐carotene and cis‐β‐carotene). Co‐digestion of probiotic‐coated C‐melon‐FC with NP increased the percentage (%) bioaccessibility of cyanidin‐3‐glucoside (11.18%) and cyanidin‐3‐O‐sambubioside (8.89%) compared to single digestion of Natal plum. Bioaccessibility of lutein (18.2%), trans‐β‐carotene (36.92%) and cis‐β‐carotene (16.74%) increased when co‐digestion of probiotic‐coated C‐melon‐FC with BL compared to the single digestion of uncoated C‐melon. Co‐digestion of C‐melon‐FC‐L75 with BL showed the highest antioxidant activity. L. plantarum had the highest LAB survival at the intestinal phase (6.01–6.26 Log CFU mL−1) during single and co‐digestion showing probiotic potential. Using stress elicitors in fruits and vegetables is considered a good strategy to increase the content of bioactive compounds in plant foods. However, bioactive compounds can affect the sensory characteristics of food products, and little is known about their shelf-life stability in fresh produce treated with elicitors. In the present work, carotenoids and polyphenols were quantified by spectrophotometric methods in red and green butterhead lettuce treated with elicitors that had previously been demonstrated to increase bioactive compounds: arachidonic acid (AA), methyl jasmonate (MJ), and Harpin protein (HP). The bioactive compounds were determined immediately and during three weeks after harvest. A descriptive sensory analysis was carried out, which included odor, taste, tactile, and visual attributes of control and elicitor-treated lettuce. Carotenoids showed greater shelf-life stability than polyphenols, and both were more stable in red than in green lettuce during the first two weeks of storage. The best elicitor was MJ, which increased phenolic compounds (red and green lettuce), anthocyanins, and carotenoids (red lettuce) through the storage period. Color intensity, crispness, wettability, and bitter taste were some of the primary sensory attributes in butterhead lettuce and were not affected by any treatment. Other organoleptic properties were also not affected by the elicitors. These results suggest that elicitation could improve the content of bioactive compounds, which is stable through the shelf-life of butterhead lettuce, without any adverse effect on the sensory properties. This study evaluated the impact of the encapsulation of sea buckthorn and grape pomace extracts in liposomal formulations on the retention and release of bioactive compounds and their antioxidant activity. The profile and composition of lipophilic extracts of sea buckthorn and hydrophilic extracts of grape pomace were analyzed. Encapsulation efficiency, retention rate, and the content of bioactive compounds encapsulated in liposomal formulations prepared in two media—water and ethanol—were evaluated. The encapsulation efficiency varied between 84 and 90%, indicating the superior encapsulation of the bioactive compounds. The retention rate varied between 79 and 86%, which indicated the stability of the liposome-encapsulated compounds over time. The antioxidant activity of the encapsulated samples was determined in vitro, under the conditions of gastric (pH 1.8) and intestinal (pH 8.2) digestion, in relation to the non-encapsulated extracts. The antioxidant activity of both liposomal formulations was higher than that of the nonencapsulated extracts during gastric digestion. Moreover, an increase over time in the antioxidant activity, expressed as % DPPH inhibition, was observed for all samples, with around 90% DPPH inhibition for non-encapsulated extracts and 92% for the encapsulated extracts, demonstrating the stability of bioactive compounds in acidic pH. Oppositely, when exposed to intestinal simulated digestion (alkaline pH), the antioxidant activity decreased over time to around 24% DPPH inhibition for both encapsulated and nonencapsulated extracts. Brazilian soils are predominantly rich in aluminum, which becomes mobile at pH < 5, affecting sensitive plants; however, some species have developed aluminum tolerance mechanisms. The purpose of this study was to compare the physiological responses of Crotalaria genus species, family Fabaceae, which have the ability to associate with nitrogen-fixing bacteria under the influence of Al3+ in the soil. The soil used was Oxisol; the experimental design was in randomized blocks in a factorial scheme (2 × 3): soil factor (available toxic aluminum content; correction of dolomitic limestone—MgCO3) and species factor (C. juncea; C. spectabilis; C. ochroleuca); cultivated within 43, 53, and 53 days, respectively, with five replications; 30 experimental samples. Mass and length, pigments, gas exchange, and changes in nitrogen metabolism were evaluated. C. juncea showed a higher concentration of amino acids in the leaves, internal carbon, and stomatal conductance in soil with Al3+, as well as higher production of ureides, allantoinic acid, allantoic acid, proteins, and amino acids in the nodules, with 78% of the Al3+ accumulation occurring in the roots. C. ochroleuca demonstrated greater shoot length and nodule number production in limed soil; in soil with Al3+, it showed a 91% increase in chlorophyll a content and 93% in carotenoids. The aim of this study was to investigate an ultrasound-assisted method for the extraction of bioactive compounds from tomato pomace. This method was based on a simple ultrasound treatment using different solvents (e.g., acetone, ethanol, ethyl acetate) as extraction media. Three different variables were chosen to determine their influence on extraction efficiency, such as solvent/liquid ratio (0.1:25 g/mL, 0.2:25 g/mL, 0.3:25 g/mL), temperature (30 °C, 40 °C, 50 °C), and time (2 h, 6 h, 10 h); the variables were selected through Box–Behnken design experiments. These parameters were optimized to achieve the highest yield, and the results showed that acetone was the best extraction solvent for lycopene (3.36 mg/100 g), total phenolic content (TPC) (31.15 QUE mg/L), and total flavone content (TFC) (66.80 GAE mg/L), while DPPH activity (82.94%) was the highest when ethanol was used as the solvent. Regarding the solvent/liquid ratio, the best results were obtained at a ratio of 0.1:25. Temperature influenced the extraction, with the best results observed at 50 °C for lycopene, DPPH, and polyphenols, while the flavonoids were better extracted at 30 °C. Time played an important role as well, with the best extractions observed at 10 h. Given the health benefits of carotenoids, it is crucial to evaluate their levels in patients undergoing malabsorptive procedures like one anastomosis gastric bypass (OAGB). This study aimed to assess serum carotenoid levels before and 6 months following OAGB. Prospectively collected data from patients who underwent primary OAGB were analyzed. Data included anthropometrics, dietary intake assessments, and biochemical tests. Serum samples were analyzed for lipid profile and serum carotenoids, including lutein, zeaxanthin, α-carotene, β-carotene, phytofluene, ζ-carotene, and lycopene. Data from 27 patients (median age 47.0 years and 55.6% female) were available before and 6 months post-OAGB. The median pre-surgical BMI was 39.5 kg/m2, and the median excess weight loss at 6 months post-surgery was 63.9%. Significant decreases in all carotenoid levels were observed over time (p < 0.001 for all). A median relative decline of 65.1% in absolute total carotenoid levels and 12.7% in total cholesterol levels were found. No associations were observed between changes in clinical outcomes and carotenoid levels during the study period. This study reveals significant decreases in carotenoid levels within the first 6 months following OAGB. Nutritional intervention studies are needed to explore how incorporating carotenoid-rich foods affects post-surgery carotenoid levels and clinical outcomes. [ABSTRACT FROM AUTHOR]

SUMMARY Biofortification of green leafy vegetables with pro‐vitamin A carotenoids, such as β‐carotene, has remained challenging to date. Here, we combined two strategies to achieve this goal. One of them involves producing β‐carotene in the cytosol of leaf cells to avoid the negative impacts on photosynthesis derived from changing the balance of carotenoids and chlorophylls in chloroplasts. The second approach involves the conversion of chloroplasts into non‐photosynthetic, carotenoid‐overaccumulating chromoplasts in leaves agroinfiltrated or infected with constructs encoding the bacterial phytoene synthase crtB, leaving other non‐engineered leaves of the plant to sustain normal growth. A combination of these two strategies, referred to as strategy C (for cytosolic production) and strategy P (for plastid conversion mediated by crtB), resulted in a 5‐fold increase in the amount of β‐carotene in Nicotiana benthamiana leaves. Following several attempts to further improve β‐carotene leaf contents by metabolic engineering, hormone treatments and genetic screenings, it was found that promoting the proliferation of plastoglobules with increased light‐intensity treatments not only improved β‐carotene accumulation but it also resulted in a much higher bioaccessibility. The combination of strategies C and P together with a more intense light treatment increased the levels of accessible β‐carotene 30‐fold compared to controls. We further demonstrated that stimulating plastoglobule proliferation with strategy P, but also with a higher‐light treatment alone, also improved β‐carotene contents and bioaccessibility in edible lettuce (Lactuca sativa) leaves. Atlantic salmon (Salmo salar) is an important source of food globally; however, fillet color can significantly affect consumer purchasing, leading to potential food waste. Fish diets can be supplemented with astaxanthin to increase the organic pigment, carotenoid, responsible for flesh coloration; however, there is variation in the amount of overall fillet coloration in response to feeding astaxanthin. The uptake of this pigment is influenced by the environment and genetics and has been shown to be heritable. Therefore, we set out to determine the genomic associations of two separate year classes of farmed North American Atlantic salmon with measured Minolta Chroma Meter (lightness, redness, and yellowness) and SalmoFan phenotypic traits. Using ASReml-R genome-wide association, two genetic markers on chromosome 26 were significantly associated with almost all color traits, and these two markers explained between 6.0% and 12.5% of the variances. The genomic region on chromosome 26 was importantly found to be associated with the beta-carotene oxygenase 1 (bco1) gene, which is essential in the conversion of beta-carotenoids to vitamin A, implying that this gene may also play an important role in flesh coloration in North American Atlantic salmon. Additionally, there were several genomic regions significantly associated with color traits, in which the accompanying genes had functions in line with thermogenesis, immune function, and pathogenic responses. Background: Vitamin A is essential for physiological processes like vision and immunity. Vitamin A's effect on gut microbiome composition, which affects absorption and metabolism of other vitamins, is still unknown. Here we examined the relationship between gut metagenome composition and six vitamin A-related metabolites (two retinoid: -retinol, 4 oxoretinoic acid (oxoRA) and four carotenoid metabolites, including beta-cryptoxanthin and three carotene diols). Methods: We included 1053 individuals from the TwinsUK cohort with vitamin A-related metabolites measured in serum and faeces, diet history, and gut microbiome composition assessed by shotgun metagenome sequencing. Results were replicated in 327 women from the ZOE PREDICT-1 study. Results: Five vitamin A-related serum metabolites were positively correlated with microbiome alpha diversity (r = 0.15 to r = 0.20, p < 4 × 10−6). Carotenoid compounds were positively correlated with the short-chain fatty-acid-producing bacteria Faecalibacterium prausnitzii and Coprococcus eutactus. Retinol was not associated with any microbial species. We found that gut microbiome composition could predict circulating levels of carotenoids and oxoretinoic acid with AUCs ranging from 0.66 to 0.74 using random forest models, but not retinol (AUC = 0.52). The healthy eating index (HEI) was strongly associated with gut microbiome diversity and with all carotenoid compounds, but not retinoids. We investigated the mediating role of carotenoid compounds on the effect of a healthy diet (HEI) on gut microbiome diversity, finding that carotenoids significantly mediated between 18 and 25% of the effect of HEI on gut microbiome alpha diversity. The article outlines the physiological and biochemical parameters of 10 exotic accessions of two local cultivars of grass pea (Lathyrus sativus L.). By analyzing the plant leaf spectrophotometrically at the bud formation phase, exotic sample Bio-520 x 1330 showed chlorophyll а of 21.47 mg/ml, Bio (520 x Bio) x 273 with chlorophyll b of 12.79 mg/ml, and Ratan x IG 135481 had carotenoids at 5.57 mg/ml. Based on total pigments, Ratan x 1307 gave the highest value (30.24 mg/ml), while in the flowering phase, Ratan x IG 135481 showed chlorophyll а of 21.37 mg/ml, Ratan x 2125 with chlorophyll b of 16.51 mg/ml, and Prateek x IG 140034 with carotenoid of 6.02 mg/ml. For Ratan x 2125, the total pigments were 37.14 mg/ml, with the highest values for all traits. In the ripening phase, Bio 520 x 1330 with chlorophyll а (11.92 mg/ml), Ratan x 2125 with chlorophyll b (16.63 mg/ml), 1330 x 2125 with carotenoid (4.04 mg/ml), and Ratan x 2125 with total pigments (25.62 mg/ml). Higher protein content resulted in seeds of Prateek x IG 140035 (26.3%), Bio (520 x Bio) x 273 (26.1%), and Ratan x 1307 (26.0). Besides, 1330 x 2125 has biochemical elements (Li, B, Na, Cu, and As), Ratan x 2125 (Mg, K, Ca, Ti, Sr, and Ba), Ratan x IG 135481 (Rb, Mo, and Cd), and Prateek x IG 140034 with chemical elements (Al, Fe, and Ag). Promoting more floret primordia within a spike to acquire fertile potential during the differentiation and pre-dimorphism phases is critical for increasing the number of fertile florets per spike (NFFs). However, it is yet unknown the physiological mechanism regulating the complex and dynamic process. This study aimed to clarify how intra-spike hormones, pigments, and assimilates coordinate with each other to regulate spike morphology and then floret primordia development. A two-year field experiment was conducted with two winter wheat genotypes: N50 (big-spike with greater NFFs) and SM22 (mediumspike with fewer NFFs). We monitored high temporal and spatial-resolution changes in the number and morphology of floret primordia within a spike, as well as in intra-spike hormones, pigments, and assimilates. Our results revealed that the big-spike genotype had more NFFs than the medium-spike genotype, not only because they had more spikelets, but also because they had greater NFFs mainly at central spikelets. More floret primordia at central spikelets had sufficient time to develop and acquire fertile potential during the differentiation phase (167-176 d after sowing, DAS) and the pre-dimorphism phase (179 DAS) for the big-spike genotype than the medium-spike genotype. Floret primordia with fertile morphology during the pre-dimorphism phase always developed into fertile florets during the dimorphism phase. Those early-developed floret primordia most proximal and intermediate to the rachis in the big-spike genotype developed faster than the medium-spike genotype. Correspondingly, the spike dry matter and pigments (chlorophyll a, chlorophyll b, carotene, and carotenoids) content during 170-182 DAS, auxin (IAA) and cytokinin (CTK) content on 167 DAS were significantly higher in the big-spike genotype than in the medium-spike genotype, while jasmonic acid (JA) content was significantly lower in the big-spike genotype compared to the medium-spike genotype during 167-182 DAS. Since the significant differences in intra-spike hormone content of the two genotypes appear earlier than those in dry matter and pigments, we propose a possible model that helped the N50 genotype (big-spike) to form more fertile florets, taking the intra-spike hormone content as a signaling molecule induced assimilates and pigments synthesis, which accelerated the development of more floret primordia during the differentiation phase and then acquired fertile potential during the pre-dimorphism phase, finally improved the NFFs. Our high temporal and spatial-resolution analysis provides an accurate time window for precision cultivation and effective physiological breeding to improve the number of fertile florets in wheat. Neoporphyra haitanensis, a red alga harvested for food, thrives in the intertidal zone amid dynamic and harsh environments. High irradiance represents a major stressor in this habitat, posing a threat to the alga's photosynthetic apparatus. Interestingly, N. haitanensis has adapted to excessive light despite the absence of a crucial xanthophyll cycle‐dependent photoprotection pathway. Thus, it is valuable to investigate the mechanisms by which N. haitanensis copes with excessive light and to understand the photoprotective roles of carotenoids. Under high light intensities and prolonged irradiation time, N. haitanensis displayed reduction in photosynthetic efficiency and phycobiliproteins levels, as well as different responses in carotenoids. The decreased carotene contents suggested their involvement in the synthesis of xanthophylls, as evidenced by the up‐regulation of lycopene‐β‐cyclase (lcyb) and zeaxanthin epoxidase (zep) genes. Downstream xanthophylls such as lutein, zeaxanthin, and antheraxanthin increased proportionally to light stress, potentially participating in scavenging reactive oxygen species (ROS). When accompanied by the enhanced activity of ascorbate peroxidase (APX), these factors resulted in a reduction in ROS production. The responses of intermediates α‐cryptoxanthin and β‐cryptoxanthin were felt somewhere between carotenes and zeaxanthin/lutein. Furthermore, these changes were ameliorated when the organism was placed in darkness. In summary, down‐regulation of the organism's photosynthetic capacity, coupled with heightened xanthophylls and APX activity, activates photoinhibition quenching (qI) and antioxidant activity, helping N. haitanensis to protect the organism from the damaging effects of excessive light exposure. Simple Summary: To investigate the impact of tomato consumption on reducing metabolic syndrome risk factors in overweight postmenopausal women, we conducted a randomized controlled trial with an 8-week open-label dietary intervention. Overweight postmenopausal women aged 45–70 were randomly divided into two groups: a control diet and a tomato diet. The tomato diet group showed significantly lower body fat mass, body fat percentage, waist circumference, and hip circumference compared with the control group. They also had significantly lower serum total cholesterol, triglycerides, systolic blood pressure, and blood sugar levels, as well as higher high-density lipoprotein cholesterol. Additionally, antioxidant biomarkers such as FRAP, beta-carotenoids, and lycopene were significantly higher in the tomato diet group. These findings suggest that fresh tomato consumption can enhance antioxidant biomarkers and reduce metabolic syndrome risks in postmenopausal women. (1) Background: According to the 2005~2008 Nutrition and Health Survey in Taiwan (NAHSIT), more than half of Taiwanese women (57.3%) had metabolic syndrome during menopause. Metabolic syndrome is a set of risk factors for cardiovascular disease (CVD) that increase the risk of cardiovascular disease, diabetes, and mortality. Epidemiological studies suggest that the consumption of tomato-based foods might reduce the risk factors for CVD. The aim of this study is to examine the effects of tomato consumption on lowering the metabolic syndrome risk factors among overweight postmenopausal women. (2) Methods: We conducted a randomized controlled trial using 8-week open-label dietary intervention. Overweight postmenopausal women aged 45–70 years old were recruited from Taipei Medical University in October 2013. They were randomly assigned into two groups (a control diet vs. a tomato diet). Blood samples were collected at the baseline and at the 4th and 8th weeks. The lipid profile, blood sugar, and antioxidant biomarkers, i.e., the ferric-reducing ability of plasma (FRAP) and serum carotenoids, were analyzed. Blood pressure, body weight, and body fat were also measured every week. (3) Results: After the 8-week dietary intervention, body weight, body mass index, waist circumference, and hip circumference were significantly lower in both groups (p < 0.05). Body fat mass, body fat percentage, waist circumference, and hip circumference were significantly lower in the tomato diet group than in the control diet group. The tomato diet group had significantly lower serum total cholesterol, triglyceride, systolic blood pressure and blood sugar, and higher high-density lipoprotein cholesterol than the control diet group. The antioxidant biomarkers, FRAP, beta-carotenoids, and lycopene were significantly higher in the tomato diet group than in the control diet group. • Infant diet assessment validity varies by dietary component and feeding stage. • Carotenoid intake estimates differ by milk carotenoid adjustment and approach. • Carotenoid intake estimates from 3 days of food diaries correlate with 7 days. Validated carotenoid assessment methods are needed to study infant carotenoid nutrition. This is a secondary analysis of repeated diet assessments of healthy participants collected at 4- (n = 21), 6- (n = 12), and 8- (n = 9) months of age in Houston, TX between April 2019 and June 2020. Intake was assessed with 3 assessment tools, analyzed with 3 nutrient databases, and underwent 3 adjustments to account for milk composition variability. We hypothesized that manual adjustment of milk carotenoid intake based on laboratory measurements would improve the validity of all assessment approaches and that using a database with greater coverage of infant food carotenoid compositions would improve accuracy. Generalized linear mixed models assessed associations between tool, nutrient database, age, and milk carotenoid adjustment variables with carotenoid, energy, fruit, and vegetable intakes. The effect of the number of food diary days on intake estimate precision was evaluated by testing the correlation between intake estimates derived from 1, 3, or 5, vs. 7 days. Visit age influenced energy intake estimates (p =.029), along with assessment tool (p =.020). Estimates of vegetable intake were influenced by tool (p =.009). Combined fruit and vegetable intake differed by nutrient database (p =.007). Carotenoid intake differed by age (p =<.0001), tool (p =.002), and nutrient database (p =.004). A minimum of 3 food diary days strongly correlated (rho = 0.79-1) with reference estimates across ages. Milk carotenoid adjustment was most influential in estimating 4-month olds' carotenoid intake, while nutrient database and tool were important for 6- and 8-month-olds', highlighting the dynamic nature of infant diet assessment validity across feeding stages. Summary: Eugenia pyriformis Cambess (uvaia), a native species of the Atlantic Forest ecosystem, has received limited attention in scientific investigations. Renowned for its distinctive organoleptic characteristics, this fruit is suitable for immediate consumption and industrial processing. Additionally, it is rich in vitamins and bioactive compounds, which offer potential health benefits. These bioactive compounds, such as ascorbic acid, carotenoids, and phenolic compounds, have demonstrated antioxidant and antimicrobial properties in both in vitro and in vivo studies. This review consolidates recent research on uvaia, focusing on its chemical composition, biological activities, and potential applications within the food industry. Pigments serve a multitude of functions in biology including light harvesting for photosynthesis, radiation protection, membrane support, and defense. The ubiquity of pigments—especially within extremophiles found in high-radiation, high-salinity, and dry environments—and their detectability via mission-ready techniques have elevated these molecules as promising targets in the search for evidence of life elsewhere. Moreover, the detection of pigments has been proposed as a "smoking gun" for extraterrestrial life as it has been suggested that these molecules cannot be generated abiotically. However, while pigments may hold promise as a biosignature, current understanding of their possible prebiotic origins remains understudied and uncertain. Better understanding of the abiotic synthesis of pigments is critical for evaluating the biogenicity of any pigment detected during missions, including by the Mars Perseverance rover or from returned samples. Compounding this uncertainty is the broad definition of pigment as it includes any compound capable of absorbing visible light and by itself does not specify a particular chemical motif. While not experimentally verified, there are promising prebiotic routes for generating pigments including hemes, chlorophylls, and carotenoids. Herein, we review the biochemistry of pigments, the inherent assumptions made when searching for these molecules in the field, their abiotic synthesis in industry and prebiotic reactions, prebiotically relevant molecules that can mimic their spectral signatures, and implications/recommendations for future work. Summary: Carotenoids are indispensable to plants and critical components of the human diet. The carotenoid metabolic pathway is conserved across plant species, but our understanding of the genetic basis of carotenoid variation remains limited for the seeds of most cereal crops. To address this issue, we systematically performed linkage and association mapping for eight carotenoid traits using six recombinant inbred line (RIL) populations. Single linkage mapping (SLM) and joint linkage mapping (JLM) identified 77 unique additive QTLs and 104 pairs of epistatic QTLs. Among these QTLs, we identified 22 overlapping hotspots of additive and epistatic loci, highlighting the important contributions of some QTLs to carotenoid levels through additive or epistatic mechanisms. A genome‐wide association study based on all RILs detected 244 candidate genes significantly associated with carotenoid traits, 23 of which were annotated as carotenoid pathway genes. Effect comparisons suggested that a small number of loci linked to pathway genes have substantial effects on carotenoid variation in our tested populations, but many loci not associated with pathway genes also make important contributions to carotenoid variation. We identified ZmPTOX as the causal gene for a QTL hotspot (Q10/JLM10/GWAS019); this gene encodes a putative plastid terminal oxidase that produces plastoquinone‐9 used by two enzymes in the carotenoid pathway. Natural variants in the promoter and second exon of ZmPTOX were found to alter carotenoid levels. This comprehensive assessment of the genetic mechanisms underlying carotenoid variation establishes a foundation for rewiring carotenoid metabolism and accumulation for efficient carotenoid biofortification. This study was conducted to explore the effect of vermicompost on the growth and physiological parameters of the tomato plant (Solanum lycopersicum L.). The experiment was carried out in a complete random block design. In the initial nursery phase, different percentages (20, 30, and 40%) of vermicompost/compost were used, and the mixture containing 15% of each was evaluated in the cell plug trays. In the transplanting phase, seedlings randomly selected based on their superior growth performance from treatments involving 20% compost/vermicompost, as well as the mixture were subsequently transplanted into pots filled with organic soil. Plants were monitored until the flowering stage, and measurements were taken at different growth stages of the plant, such as shoot length, stem diameter, number of leaves, root length, fresh and dry weight of the plant. Moreover, some physiological parameters, such as chlorophyll and carotenoid contents, were assessed. An ANOVA using SPSS was used to process the data collected from the experiment. The results showed that vermicompost treatments induced a significant increase in shoot length, stem diameter, leaves number, root length, contents of chlorophyll a and carotenoid of tomato plants compared to the control, with an improvement of 54, 38, 38, 70, 44, and 69.5%, respectively. BACKGROUND: Microgreens constitute dietary sources of bioactive compounds imparting numerous health benefits and enhancing sensory experience. They can be successfully cultivated in soilless systems where biostimulants can be easily integrated as seed‐priming and post‐germination agents improving the sustainability of a crop's final production. Compared to an untreated control, three priming agents (a commercial legume‐derived protein hydrolysate (A250), a novel protein hydrolysate derived from peanut biomass (H250) and hydropriming (H2O)) were applied to Komatsuna and Mibuna seeds grown as microgreens and compared for their effects on yield parameters, mineral composition, ABTS and FRAP antioxidant capacity, carotenoid concentration and phenolic compounds. RESULTS: Significant effects of the main experimental factors and their interactions were identified on antioxidant capacity. Compared to the control and hydropriming, the highest ABTS and FRAP values were observed in Mibuna with the A250 and H250 treatments, respectively. Additionally, the H250 treatment increased the total concentrations of phenolic acid derivatives and flavonoid derivatives in Mibuna and Komatsuna, in tune with the levels of total flavonoids. Concerning mineral composition, the highest concentrations in both species were those of phosphorus and nitrate. CONCLUSION: These results highlight the potential of select plant‐based biostimulants as priming agents to enhance the antioxidant capacity, nutrient content and bioactive compound content, thus further increasing their functional and nutritive quality. In the light of this, the possibility of reducing the application of fertilizers by promoting a green transition for the intensive production of microgreens could subsequently be evaluated. © 2024 Society of Chemical Industry. Salinity is a common abiotic stress that limits crop productivity. Although there is a wealth of evidence suggesting that miRNA and lncRNA play important roles in the response to salinity in rice seedlings and reproductive stages, the mechanism by which competing endogenous RNAs (ceRNAs) influence salt tolerance and yield in rice has been rarely reported. In this study, we conducted full whole-transcriptome sequencing of rice panicles during the reproductive period to clarify the role of ceRNAs in the salt stress response and yield. A total of 214 lncRNAs, 79 miRNAs, and 584 mRNAs were identified as differentially expressed RNAs under salt stress. Functional analysis indicates that they play important roles in GO terms such as response to stress, biosynthesis processes, abiotic stimuli, endogenous stimulus, and response to stimulus, as well as in KEGG pathways such as secondary metabolite biosynthesis, carotenoid biosynthesis, metabolic pathways, and phenylpropanoid biosynthesis. A ceRNA network comprising 95 lncRNA–miRNA–mRNA triplets was constructed. Two lncRNAs, MSTRG.51634.2 and MSTRG.48576.1, were predicted to bind to osa-miR172d-5p to regulate the expression of OsMYB2 and OsMADS63, which have been reported to affect salt tolerance and yield, respectively. Three lncRNAs, MSTRG.30876.1, MSTRG.44567.1, and MSTRG.49308.1, may bind to osa-miR5487 to further regulate the expression of a stress protein (LOC_Os07g48460) and an aquaporin protein (LOC_Os02g51110) to regulate the salt stress response. This study is helpful for understanding the underlying molecular mechanisms of ceRNA that drive the response of rice to salt stress and provide new genetic resources for salt-resistant rice breeding.
