In France, it was reported for the first time in 2012 on seed carrots. The CaLisoproject validated a Lso detection method and thus made the health quality of carrot seeds reliable andguaranteed the same reliability for potato plants. In France, our large-scale surveys show that the LsoDand LsoE haplotypes are present in the majority of Apiaceae production areas, and that Bactericeratrigonica is the main psyllid species present on these crops. This psyllid is probably the main vector ofLso in France. In addition, Lso was not detected on potatoes, especially those produced near Apiaceaecrops, which underlines the very low risk of Lso transmission from carrots to potatoes. The studies carried out improved our knowledge of this pathosystem. Lso does not seem to be emerging on Apiaceae inFrance. The major threat for Solanaceae would probably be an introduction of the potato psyllid, B.cockerelli, in Europe.; ‘Candidatus Liberibacter solanacearum’ (Lso) est la bactérie du phloème responsable de la maladie du« Zebra Chip » aux USA. En France,les prospections à grande échelle montrent que les haplotypes LsoD et LsoE sont présents dans lamajorité des zones de productions d’apiacées et que Bactericera trigonica est la principale espèce depsylles présente sur ces cultures. Ce psylle constitue probablement le principal vecteur de Lso en France.De plus, Lso n’a pas été détectée sur pommes de terre, notamment celles produites à proximité decultures d’apiacées, ce qui souligne le très faible risque de transmission de Lso de la carotte vers lapomme de terre. Les études menées ont permis de mieux connaitre ce pathosystème. Lso ne semblepas émergente sur apiacées en France mais pourrait l’être sur solanacées (haplotypes A et B) en casd’introduction du psylle de la pomme de terre, B. cockerelli, en Europe.

This study aims at investigating p-menthane, a novel bio-based solvent resulting from the hydrogenation of d-limonene, as a green alternative to n-hexane or toluene for the extraction and solubilization of natural substances. First, conductor-like combination of quantum chemistry (COSMO) coupled with statistical thermodynamics (RS) calculations show a comparable solubilization profile of p-menthane and n-hexane for carotene, volatile monoterpenes such as carvone and limonene, and model triglycerides. Other data obtained experimentally in solid/liquid extraction conditions further indicate that p-menthane showed similar performances to n-hexane for extracting carotenes from carrots, aromas from caraway seeds, and oils from rapeseeds, as these products showed a comparable composition. p-Menthane was also tested using common analytical extraction procedures such as Soxhlet for determination of oil content via multiple extraction stages, and Dean&ndash, Stark for determination of water content via azeotropic distillation. For both systems, yields were comparable, but for Dean&ndash, Stark, the distillation curve slope was higher when using p-menthane, and the time needed to attain 100% water recovery was 55% shorter than for toluene. Taken together, these results reveal the potential of p-menthane as a green replacer for petroleum-based solvents such as n-hexane or toluene.

These different plans have set the goal of a 50% reduction in pesticide use. It is for this purpose that the producers of the two main French carrot production areas for the fresh market (New Aquitaine and Normandy) have built a project to construct and evaluate carrot- dominated vegetable systems that reduce the use of pesticides by at least 50%. The project relies on a network of eight innovative cropping systems, located on three production sites, and conducted from 2013 to 2018. The levers and combinations of levers studied have, at least partially, given satisfaction on the environmental aspect and the management of pests. However, the economic balance of cropping systems is currently difficult to achieve and some levers proposed do not meet the demand of markets (production niches, visual quality of certain products) and the current organization of the sector. Today, in order to begin the transfer to producers, new levers and combinations of levers must be tested, and decision rules need to be refined. This work will be the subject of a second project DEPHY Expé: AlterCarot which will be conducted from 2019 to 2024.; L’évolution des attentes sociétales, depuis le début des années 2000, a conduit les politiques européennes et nationales à adopter un arsenal législatif visant à réduire l’utilisation des produits phytosanitaires afin d’en limiter les impacts. Le présent rapport fait suite au livrable Traitement et analyse des radargrammes (Action II.2). Il en constitue la suite logique, opératoire et scientifique, ce livrable ne reprend pas les descriptions sédimentaires et les analyses sédimentologiques du livrable de l’action II.2 ; il en constitue la synthèse complétée par la présentation des résultats d’analyses chimiques (laboratoire EDYTEM et Laboratoire LEHNA-IPE), des analyses de radionucléides (Laboratoire LSCE de Gif-sur-Yvette) et des analyses de PCBi (Laboratoire LABERCA). Une première interprétation des résultats est présentée.

International audience; The study comprises a systematic and quantitative evaluation of potential intrinsic and extrinsic factors that impact vitamin C degradation in a real food matrix. The supernatant of centrifuged apple purée was fortified in vitamin C, and degradation was followed without stirring. Model discrimination indicated better fit for the zero order model than the first order model which was hence chosen for determination of rate constants. pH influenced strongly vitamin C degradation in citrate-phosphate buffer but not in the apple purée serum. To get an idea of the impact of the food matrix, stability in apple purée serum was compared with that in carrot purée. In the latter, stability was slightly higher. Vitamin C degradation rates were not influenced by its initial concentration. The temperature effect was only marked in the temperature range 40–60 °C. In the range 60–80 °C, filling height of tubes had the greatest impact.

Oxygen availability in different media during heat treatment (8 h at 80 °C) and the related vitamin C loss was assessed. Dissolved oxygen in water containing 3 mmol kg−1 of ascorbic acid decreased initially and seemed to be replaced by oxygen from the headspace in the course of time, as oxygen values increased again. In apple puree and carrot puree in contrast, oxygen was depleted within 60 min. Vitamin C in ultrapure water was stable even in the presence of oxygen. A trigger seemed to be crucial to launch vitamin C degradation. Fe3+ ions added to water, but also the media Mc Ilvaine citrate-phosphate buffer (pH 3.5) or apple puree, initiated degradation. Adding Fe3+ ions to apple puree did not accelerate vitamin C degradation but shifted the equilibrium between ascorbic acid and dehydroascorbic acid to the latter. Oxygen deprivation stabilized completely vitamin C, independently of the medium tested. A temperature decrease to 70 or 60 °C, in contrast, had no effect on the degradation extent of vitamin C in water containing 20 μmol kg−1 Fe3+ ions but led to complete stability in apple puree.

This study was designed to evaluate the applicability of a new liquefied gas, HFO-1234ze, as a safe alternative for the substitution of usual solvents such as n-hexane, acetone or ethanol, for the extraction of natural products. The performance of this new solvent was assessed using several plant materials: rapeseeds, carrots, olive leaves and rosemary leaves. An in-silico evaluation of the relative performance of each solvent was performed using Hansen solubility parameters and COSMO-RS model. Extraction experiments were also performed at laboratory scale on several plant materials, in order to determine qualitative and quantitative characteristics of the extracts. Finally, safety considerations, as well as energetic and environmental impacts using this new liquefied gas solvent were evaluated in a scale-up perspective.

International audience; Recent trends in food extraction techniques have largely focused on finding solutions that minimize the use of toxic solvents or to find alternatives for n-hexane. As an original solution, a new process for extraction of natural product using liquefied gases has been developed at laboratory scale. The applicability of this new process was demonstrated using n-butane as alternative solvent to n-hexane. Extraction of lipophilic natural products such as carrot oleoresin, caraway aromas and sunflower oil was assessed using liquefied n-butane. COSMO-RS simulations allowed determination of the relative solubility of the lipophilic compounds in liquefied n-butane and n-hexane. This theoretical approach was validated with extraction experiments on carrots, caraway seeds and sunflower seeds. Compositions of the extracts were comparable but the extract yields were lower using n-butane. For example the extraction yields were, with n-butane and with n-hexane respectively, 36.9% and 53.4% for sunflower seeds oil, 10.9% and 15.0% for caraway seed essential oil, 25.7% and 55.8% for carrots oleoresin. However the extracts obtained by using n-butane are directly free of solvents and also food grade, as it is an authorized solvent for foodstuff production.

Two methods that are used at people's home to warm-up food namely a microwave and an Actifry® device, and a system that is usually employed in canteen kitchens to keep food warm, were examined on their impact on the stability of vitamin C and 5-methyltetrahydrofolate. The puree was heated until ebullition with the microwave, which lasted 1.5 min. Puree which was warmed-up by the Actifry® device needed around 20 min to attain 80 °C. When the puree was kept warm by a water bath, the vitamin stability was pursued for 180 min at 80 °C. During the latter, stability was examined at the surface and bottom of recipients that were filled with 600 g or 200 g of puree corresponding to a filling height of 5 cm and 1.5 cm respectively. Although vitamin C and 5-methyltetrahydrofolate are usually referred to as heat- and oxygen-susceptible molecules, none of the methods led to major vitamin losses. In terms of the warm holding method, no difference was observed between withdrawals at the surface and the bottom of vessels for neither of the two filling levels. Vitamins were also fairly stable at the surface of recipients where oxygen exposition is high.

UMR BFP - Equipe Virologie; First Report of [i]Carrot torradovirus 1[/i] (CaTV1), a Member of the [i]Torradovirus[/i] Genus, Infecting Carrots in France

Le présent rapport constitue le livrable correspondant à l’action II.3 de l’OSR4 2015-2017 : « Détermination de la dynamique sédimentaire spatiale et temporelle de la plaine alluviale du secteur de Pierre-Bénite (PBN) : Rapport d'exécution, traitement et analyse des carottes (ouverture, description, granulométrie). Il fait suite à une caractérisation électromagnétique de la zone de Pierre-Bénite par radar géologique qui avait permis d’identifier des zones préférentielles de carottage peu profond (anciens chenaux, remplissages postérieurs au XIXème siècle, etc.).

Elderlies are often victims of undernutrition which can lead to multiple health damages. Prevention of this state of undernutrition is one of the main concerns of health authorities. With this objective, the OPTIFEL project designed new optimized foodstuffs, to improve foods for elderlies, by taking into account their needs and wishes, while preserving their safety. Bacillus cereus is a spore-forming bacterial pathogen able to grow in various conditions as low temperature, absence of oxygen, low pH and in different types of food. It is one of the main causes of foodborne outbreaks, particularly in socio-medical institutions. The aim of this work was to determine the behavior of B. cereus at different steps of the life of the food product. Cold storage can allow multiplication of psychrotolerant B. cereus strains that can represent a risk of infection for consumers. Anaerobic conditions reduce the adaptation capacity of B. cereus at low temperature and low pH. The composition and the preparation of the food product also influence the behavior of B. cereus, as we showed with carrot broth in which oxygen created a lethal effect for the bacterium. Certain psychrotolerant B. cereus strains are able to produce an emetic toxin named cereulide. We showed that this production starts at 8 °C and can reach at 10 °C concentrations sufficient to cause intoxications. If this toxin is produced in food, it would not be destroyed during the food preparation. In contrast, vegetative cells of B. cereus are very heat-sensitive and those who would have been able to develop could be destroyed by a reheating of the food product before its consumption. This work allowed the identification of important parameters to limit the risk of foodborne disease by B. cereus.; Les personnes âgées sont souvent victimes de dénutrition pouvant conduire à de multiples dégradations de la santé. La prévention de cet état de dénutrition est une des principales préoccupations des autorités de santé. C’est dans cet objectif que le projet OPTIFEL a conçu de nouveaux produits alimentaires optimisés, afin d’améliorer l’alimentation des personnes âgées, en tenant compte de leurs besoins et souhaits, tout en préservant leur sécurité sanitaire. Bacillus cereus est une bactérie pathogène sporulée capable de se développer dans des conditions variables comme le froid, l’absence d’oxygène, des bas pH ainsi que des matrices alimentaires diverses. Elle est l’une des principales causes de Toxi-Infection Alimentaire Collective (TIAC) surtout en Institut Médico-Sociaux (IMS). Ce travail de thèse avait donc pour objectif de déterminer le comportement de B. cereus à différentes étapes de la vie d’un produit alimentaire. La conservation au froid peut permettre la multiplication de souches psychrotolérantes de B. cereus qui peuvent alors représenter un risque d’infection pour le consommateur. Les conditions anaérobies réduisent la capacité d’adaptation de B. cereus au froid et aux bas pH. La composition de l’aliment et sa préparation influence également le comportement de B. cereus, comme nous l’avons constaté avec un bouillon de carotte dans lequel la présence d’oxygène crée un effet létal pour la bactérie. Certaines souches psychrotolérantes de B. cereus sont capables de produire de la toxine émétique appelée céréulide. Nous avons montré que cette production peut se faire à partir de 8°C et atteindre des concentrations pouvant causer des intoxications à partir de 10°C. S’il est produit dans l’aliment, le cereulide ne serait pas détruit lors de la préparation de l’aliment. Par contre les cellules végétatives de B. cereus sont très thermosensibles et celles qui auraient pu se développer pourront être détruites par un réchauffage du produit avant sa consommation. Ce travail a permis de définir les paramètres importants pour limiter le risque d’infection alimentaire par B. cereus.

The color of aqueous solutions obtained by heating carrot (Daucus carota L.) roots in water ("stocks") is different when the thermal treatment is applied with or without exposure to light. CIE L*, a*, and b* scale values of stocks processed for different times were recoreded and 4 patterns were initially observed. To explain the 1st part of this evolution (patterns 1 and 2), pectin extraction and beta-elimination in stocks were studied. Light dependence was investigated to explain patterns 3 and 4. A model with 2 compounds is proposed to explain all the color variations.

The present study was designed to evaluate five green solvents, i.e. 2-methyltetrahydrofuran (2-MeTHF), dimethyl carbonate (DMC), cyclopentyl methyl ether (CPME), isopropyl alcohol (IPA) and ethyl acetate, for the substitution of n-hexane in the extraction of carotenoids from carrots. Initially, solvent selection was made through the theoretical physicochemical solvent properties and solubility results obtained using two simulation programs, Hansen Solubility Parameters (HSPs) and Conductor-like Screening Model for Realistic Solvation (COSMO-RS) which use a statistical thermodynamics approach based on the result of quantum chemical calculation, for comprehension of the dissolving mechanism. On the basis of the HSPs analysis, non-polar or slightly polar solvents were the most suitable solvents for extraction of carotenoids. COSMO-RS analysis showed a higher probability of solubility for all the carotenoids from carrot in CPME, 2-MeTHF and ethyl acetate compared with n-hexane. The experimental results using a conventional solid–liquid extraction by maceration showed that the best green solvents were CPME, 2-MeTHF and ethyl acetate in accordance with the predictive results from COSMO-RS. The highest carotenoid content (78.4 mg 100 g−1 DM) was observed in CPME where 66% was represented by β-carotene and 34% was α-carotene. These results support the potential of CPME and 2-MeTHF as alternative green solvents for extraction of carotenoids. Great research works support the sector of drying of fruits and vegetables in a number of ways. Each year, experimental results and mathematical modelling try through conventional and innovative technological processes to protect agricultural products from seasonality and variability in the market prices. Nonetheless, there still exist considerable issues to improve and control this strategic agricultural sector. Swell-drying is very relevant drying process coupling hot air dehydration with Instant Controlled Pressure Drop (DIC). In the present work, carrot and apple were used as a fruit and vegetable model. Scientific literature on drying is unanimous that the drying time decreases with increasing air temperature, decreasing air relative humidity, and reducing thickness. However, the effect of airflow velocity leads to contradictory conclusions. Some authors have claimed that it does not trigger any modification of drying; while some articles tried to establish empirical models of the effective diffusivity versus airflow velocity; what is fundamentally erroneous. This thesis aims at analyzing external and internal transfer phenomena during drying. So, it was possible to recognize the Critical Airflow Velocity CAV depending on the effective diffusivity and the product size, to have the internal water transfer as the limiting phenomenon. From experimental results, an empirical model was established between the effective moisture diffusivity and airflow temperature, water content, and absolute expansion ratio., Dans le séchage des fruits et légumes, beaucoup de questions non résolues subsistent. Le «Swell-drying» est un procédé de séchage couplant une étape de déshydratation par air chaud à la texturation par Détente Instantanée Contrôlée (DIC). Dans le cadre de ce travail de recherche, la carotte et la pomme sont utilisées comme aliment modèle pour le traitement de la plupart des végétaux. La littérature scientifique sur le séchage des agromatériaux est unanime sur l’effet positif de la température de l'air, l’effet négatif de l'humidité relative de l'air et de l'épaisseur vis-à-vis du temps de séchage. Cependant, malgré un nombre important d’études, des conclusions contradictoires sont notées en termes de corrélations entre la cinétique de séchage et la vitesse de l'air qui, selon certains auteurs, n’entraîne aucune modification du séchage; alors que certains travaux ont tenté d'établir des modèles empiriques entre la diffusivité effective et la vitesse de l'air ; ce qui est fondamentalement erroné. L’objectif de cette thèse est d’analyser les phénomènes de transfert pendant le processus du séchage et d'identifier le phénomène limitant entre les transferts internes et externes. Ce travail aboutir à l’identification de la vitesse critique de l'air CAV capable de rendre la diffusion interne de l’eau comme phénomène limitant. La CAV doit donc dépendre de la diffusivité effective et de la taille du produit. À partir de résultats expérimentaux qui couvrent de larges plages de température d’air, de teneur en eau, et de taux d'expansion absolu, un modèle empirique a été établi.

Core description is used extensively by sedimentologists to yield critical information from sediments. It consists of a naturalistic description of the visual and textural characteristics of the sediment. It is generally a manual procedure. However, digital core descriptions are now required re manage data processing. Hand-written description can be keyed into a computer, but it is tedious and time consuming. Moreover, recording and typing errors can occur. A system is presented for digital description of sediment cores using a bar code principle. It can be used in the laboratory as well as on board vessels. It comprises a box inside which the half-core can be placed. A ruler and several sets of bar codes are printed on the inside box top. Bar codes are read by a waterproof sensor pencil that is interfaced with a PSION 3A Docket computer. The software mimicks the traditional method of cope description, the geologist answering questions using the optical pencil. It offers versatility and is designed with the geoscientist's need for flexibility and ease of use in mind. It allows computerised core data management and is highly upgradable. It can help to harmonise core description and to compare it with core data provided by the new multiparameter measurement systems. Detailed log plots no longer require tedious coding. Logmeter data can be used to automatically create customized, full-featured log graphs displaying lithology, descriptions, curves, histograms, graphic symbols and attractive legends, using commercial or dedicated softwares., Le logmètre est un outil de saisie de données basé sur l'utilisation de codes-barres. Il peut être utilisé indifféremment à bord des navires ou en laboratoire pour la description des carottes de sédiments. Le système se présente comme un boîtier à l'intérieur duquel on place la demi-carotte à décrire. Le couvercle intérieur du boîtier comporte une règle graduée et différents ensembles de codes-barres qui sont lus au moyen d'un stylo laser interfacé avec un ordinateur de poche de type Psion 3A. Changes in cell wall integrity upon thermal treatment were assessed in carrot cells using novel microscopic approaches using Congo red and different cell wall polysaccharide specific probes (JIM7, LM10, LM11, LM15, LM21, LM22 and CBM3a). Strong thermal processing induced an increased accessibility of cellulose and hemicelluloses by Congo red and the specific probes, except galactomannan, which detection was not affected by the thermal processing. Detection of pectin by JIM7 disappeared upon thermal processing, pointing at the leaching out effect of pectin from cell wall due to β-elimination. Changes observed after thermal processing were moreover similar to changes observed after enzymatic degradation of pectin, and a combination of thermal and pectinases treatments did not cause additional effects. These observations indicated that the presence of native pectin is the main factor governing cell wall polysaccharides accessibility and overall cell wall integrity in carrot, which can be modulated through thermal processing. Industrial relevance This work provides new evidences on the specific role of pectin in carrot cell wall integrity, more specifically on how it can be modulated by thermal processing. New light microscopy approaches to assess changes in cell wall integrity are presented. This information is important for food industry since plant cell wall acts as a structural barrier for the release of carotenoids and other micronutrients in plant-based food products.

Article de revue (Article scientifique dans une revue à comité de lecture); International audience; Although different mechanisms have been proposed in the recent years, plant pathogen partial resistance is still poorly understood. Components of the chemical warfare, including the production of plant defense compounds and plant resistance to pathogen-produced toxins, are likely to play a role. Toxins are indeed recognized as important determinants of pathogenicity in necrotrophic fungi. Partial resistance based on quantitative resistance loci and linked to a pathogen-produced toxin has never been fully described. We tested this hypothesis using the Alternaria dauci – carrot pathosystem. Alternaria dauci, causing carrot leaf blight, is a necrotrophic fungus known to produce zinniol, a compound described as a non-host selective toxin. Embryogenic cellular cultures from carrot genotypes varying in resistance against A. dauci were confronted with zinniol at different concentrations or to fungal exudates (raw, organic or aqueous extracts). The plant response was analyzed through the measurement of cytoplasmic esterase activity, as a marker of cell viability, and the differentiation of somatic embryos in cellular cultures. A differential response to toxicity was demonstrated between susceptible and partially resistant genotypes, with a good correlation noted between the resistance to the fungus at the whole plant level and resistance at the cellular level to fungal exudates from raw and organic extracts. No toxic reaction of embryogenic cultures was observed after treatment with the aqueous extract or zinniol used at physiological concentration. Moreover, we did not detect zinniol in toxic fungal extracts by UHPLC analysis. These results suggest that strong phytotoxic compounds are present in the organic extract and remain to be characterized. Our results clearly show that carrot tolerance to A. dauci toxins is one component of its partial resistance.

Carrot is one of the major vegetables, and is a basic product highly valued by regular buyers. In spite of the natural resources of the species, diversity for consumers is poor. The aim of this work was to identify consumer preferences based on appreciation of batches representative of the carrot sensory space. Ten batches were described by a trained panel using a list of 10 attributes. The same products were given to a panel of 299 consumers from 3 French cities (Avignon, Angers, and Paris). They gave a score of general appreciation and answered a questionnaire about usage and attitude. The results show that carrots are mainly characterized by the perception of bitterness, pungency and chemical flavor in opposition to the perception of sweetness. Some batches had a higher level for global aroma. Other batches were characterized by juiciness as opposed to a high level of firmness and retention in the mouth. The highest levels of consumer satisfaction were achieved with flavored, juicy and sweet carrots. Main rejected characteristics were too much bitterness and too intense chemical flavor. The segmentation study identified three groups of consumers, differing mainly by their attitude towards the product. A group representing 20% of the panel was characterized by very marked preferences for sweet and flavored products and also for interest in a high quality product.

International audience; An experimental approach, allowing us to understand the effect of natural structural barriers (cell walls, chromoplast substructures) on carotenoid bioaccessibility, was developed. Different fractions with different levels of carotenoid bio-encapsulation (carotenoid-enriched oil, chromoplasts, small cell clusters, and large cell clusters) were isolated from different types of carrots and tomatoes. An in vitro method was used to determine carotenoid bioaccessjbility. In the present work, a significant decrease in carotenoid in vitro bioaccessibility could be observed with an increasing level of bio-encapsulation. Differences in cell wall material and chromoplast substructure between matrices influenced carotenoid release and inclusion in micelles. For carrots, cell walls and chromoplast substructure were important barriers for carotenoid bioaccessibility while, in tomatoes, the chromoplast substructure represented the most important barrier governing bioaccessibility. The highest increase in carotenoid bioaccessibility, for all matrices, was obtained after transferring carotenoids into the oil phase, a system lacking cell walls and chromoplast substructures that could hamper carotenoid release.

Diplôme : Master Recherche; Alternaria dauci est un champignon nécrotrophe phytopathogène qui s'attaque à la carotte (Daucus carota). Jusqu'ici, aucun cas de résistance complète n'a été mis en évidence, mais certaines variétés, bien que sensibles à l'infection, sont capables de limiter le développement du champignon. De précédentes études ont pu mettre en évidence que les réactions de défense des plantes contre les agents phytopathogènes nécrotrophes impliquaient souvent la voie de l'acide jasmonique. Le travail présenté dans ce rapport intervient dans le cadre de l'étude des facteurs génétiques de la résistance de la carotte à A. dauci. Afin de mieux caractériser cette interaction, des portions de gènes de la voie de signalisation de l'acide jasmonique tels que COI1 et JAR1 ont été clonées. Des amorces dégénérées ont été conçues pour amplifier des fragments de ces gènes à partir des séquences protéiques de différentes espèces. Les réactions PCR effectuées avec ces amorces ont abouti au clonage partiel de JAR1 et de deux fragments identifiés comme homologues à COI1, mais différents entre eux. Une fois la séquence complète de ces gènes obtenue, ils seront utilisés dans un premier temps pour l'identification de polymorphismes de substitution (SNP) potentiellement liés à des loci de traits quantitatifs (QTL) liés à la résistance à A. dauci. Ces gènes seront également utilisés pour étudier leur niveau d'expression au cours de l'attaque fongique dans différents cultivars de carotte.

Trois champignons dont le pouvoir pathogène en culture artificielle ne se maintient pas de façon satisfaisante, ont été expérimentés : Pyrenochaeta lycopersici, Phytophthora infestans et Rhizoctonia violacea. Sauf pour P lycopersici, la conservation sous azote liquide est impossible ou très aléatoire; les techniques de maintien en condition anaérobie ne conviennent guère (et encore avec des réserves) qu’à P infestans. Pour les 3 parasites, nous avons utilisé avec succès la culture associée du champignon et de racines transformées. Cette technique permet, dans la majorité des cas, un léger gain d’agressivité. Plusieurs cultures de racines transformées ont été expérimentées : tomate, pomme de terre, carotte, betterave à sucre et endive. Après passages répétés sur ces cultures, nous n’avons observé ni préférence, ni adaptation parasitaire. L’intérêt des cultures associées en phytopathologie est discuté., Alternative means were sought to preserve 3 fungi, Pyrenochaeta lycopersici (tab I), Phytophthora infestans (tab II) and Rhizoctonia violacea (tab III) the pathogenicity of which is not maintained satisfactorily in artificial culture. With the exception of P lycopersici, preservation in liquid nitrogen is either not possible or very uncertain; maintenance in anaerobic conditions may suit P infestans only if at all. We successfully maintained the 3 pathogens in culture on transformed roots; with this method, the aggressiveness of the strains is maintained, and nearly always slightly increased, and it is not necessary to inoculate and reisolate from host plants in the greenhouse. Several cultures of transformed roots have been tested : tomato, potato, carrot, sugar-beet and endive. After repeated transfers on these cultures, no parasitic specialization was observed. The interest of mixed cultures is discussed.

version n°4, février 2009imprimable en recto verso; The identification of pathways that induce the development of epidemics is a crucial step in the conception and the optimisation of control methods against plant diseases. The development of soil-borne plant diseases is due to epidemiological mechanisms closely dependent on the pathogen life history traits. Such mechanisms can be best investigated in vulnerable agrosystems, such as vegetable field crops, where short rotations and high inputs favour the emergence of severe disease problems. In carrots, one of the major sanitary problems is cavity spot, a root disease caused by a complex of Pythium spp. which affects root quality in several production basins in France and worldwide. The objectives of the present research were to identify, understand and hierarchise the processes that induce the spatio-temporal kinetics of carrot cavity spot epidemics. Interactions between species during the infectious stage can be neglected and disease progress curves were supposed to be not affected by changes in the diversity of the complex of Pythium species. The occurrence of primary and secondary infections was demonstrated and quantified experimentally for the first time in this pathosystem. The analysis of pathometric relationships between disease assessment variables (disease incidence, lesion density, symptom intensity, and total disease area) and the fitting of simulation models to observed epidemiological data confirmed the occurrence of both types of infections. An epidemiological model was elaborated, based on the processes quantified experimentally. The effect of different factors, such as soil moisture, seed density and timing of fungicide applications, on these epidemiological processes was tested. Finally, several scenarios combining control methods for integrated pest management of carrot cavity spot were designed and their optimisation discussed.; Le développement de maladies d'origine tellurique relève de mécanismes épidémiologiques particuliers, propres au cycle biologique et aux traits d'histoire de vie des agents pathogènes incriminés. La caractérisation des processus qui déterminent le développement des épidémies est une étape clé dans la conception et l'optimisation de méthodes de protection contre ces maladies. Les cultures légumières de plein champ constituent des agrosystèmes particulièrement sensibles aux attaques parasitaires. Les Pythium sont responsables du cavity spot de la carotte, une maladie racinaire qui affecte la qualité des récoltes dans plusieurs bassins de production en France et dans le monde. L'objectif des recherches présentées ici est d'identifier, comprendre et hiérarchiser les processus déterminant la dynamique spatio-temporelle de cette maladie, essentiellement due à Pythium violae, en association avec d'autres espèces du complexe parasitaire. L'analyse de la composition d'un de ces complexes, complétée par la caractérisation biologique des principales espèces pathogènes, suggère que par souci de simplification, il est raisonnable de négliger les interactions entre espèces au cours de la phase infectieuse. L'existence d'infections secondaires (auto- et allo-infections) chez P. violae, et donc la nature polycyclique d'une épidémie de cavity spot, sont démontrées expérimentalement. Cette hypothèse est initialement étayée par deux étapes de l'analyse, l'une portant sur l'examen de relations pathométriques, et l'autre sur les ajustements de modèles aux données illustrant des cinétiques de la maladie. L'ensemble des résultats obtenus permet de concevoir un modèle épidémiologique exploratoire, basé sur l'occurrence des infections primaires et secondaires. Les effets de différents facteurs sur ces processus, comme l'application d'un fongicide, l'humidité du sol et la densité de semis, sont testés expérimentalement et discutés. Ils permettent d'envisager l'association de moyens de lutte complémentaires dans le cadre de stratégies de protection intégrée.

The aim of this study is to establish correlations between different descriptors and pathological potential of soils towards plants, in order to identify indicators of soil biological quality. Three cultural practices were applied in an experimental field before carrot culture (conventional, organic amendment with cattle manure and biofumigation with fodder radish). Twenty-one soil samples were collected in each plot at 3 key points during the cropping sequence (before treatment, before and after carrot culture). Physicochemical (carbon, nitrogen and organic matter contents, C/N ratio, pH, EC, ions and trace elements), biological (densities of cultivable bacteria and fungi, microbial biomass and basal respiration, densities of plant-parasitic nematode populations), microbial diversity (bacterial and fungal community structure by T-RFLP) and plant-pathological (receptivity to Rhizoctonia solani damping-off) properties were measured and correspondences were analysed by Principal Component Analysis. Before application of the cultural practices, soil characteristics were very similar in the 3 plots. Biofumigation resulted in a significant increase of the microbial densities, biomass and basal respiration. Both organic amendment and biofumigation modified the microbial community structures. Intra and inter-class PCA enabled to distinguish any effect between treatments and dates. The soil biological characteristics showed resilience to the situation that occurred before cultural practices. Relations between physicochemical, biological and molecular data sets, provided by co-inertia analysis, will be discussed and dealt with the receptivity of the soil samples to R. solani damping-off. This strategy will enable us to propose a bunch of soil biological quality indicators.

Because of the strong tendency of mercury (Hg) to bioaccumulate in the food chain, one of the greatest challenges faced by environmental mercury research in the Arctic is to quantify the relative contribution of anthropogenic sources of mercury to the contamination of this environment, as anthropogenically elevated mercury deposition over the past 150 years in Arctic ecosystems is potentially a serious environmental problem. To determine the magnitude of this concern, it is necessary to quantify the natural “background” of atmospheric mercury deposition and its variation over a millennial-scale period of time. A second problem is to understand the role which is played by the unique climate of the Arctic on the deposition of atmospheric mercury.Total mercury concentrations were determined in peat cores from the High Arctic of Canada, the Faroe Islands and southern Ontario. The cores were dated using 210Pb and 14C. The mercury concentrations were used to calculate rates of atmospheric mercury accumulation in the peat in order to quantify rates of atmospheric deposition of mercury in the Arctic. In addition, rates of atmospheric mercury accumulation in the Arctic were compared with peat cores from the temperate zone of North America (southern Ontario).Mercury concentration measurements and age dating of two peat hummocks from Bathurst Island, Nunavut indicate rather constant natural “background” mercury flux of ca. 1 µg m-2 per year from 5900 to 800 calibrated year BP. The values are well within the range of the Hg fluxes reported from other Arctic locations (Greenland and the Faroe Islands) but also by peat cores from southern Canada (Ontario) which provide a record of atmospheric Hg accumulation extending back to eight thousand years. Thus, pre-anthropogenic Hg fluxes in the Arctic were not significantly different from atmospheric Hg fluxes in the temperate zone. In pre-industrial times, therefore, the High Arctic was not more important as a sink for global atmospheric mercury than the temperate zone. Therefore, other processes have to be invoked as chief mechanism for transferring atmospheric Hg to the Arctic environment, possibly made more efficient in recent years by environmental changes, resulting in the mercury contamination of the Arctic food chain.The beginning of mercury contamination from anthropogenic sources dates from AD 1475 at the Luther Bog in southern Ontario, corresponding to biomass burning for agricultural activities by Native North Americans. During the late 17th and 18th centuries, deposition of anthropogenic mercury was at least equal to that of mercury from natural sources. Anthropogenic inputs of mercury to the bogs have dominated continuously since the beginning of the 19th century.The records from southern Ontario and the Faroe Islands show similar chronologies to one another, with mercury accumulation rates peaking in the 1950`s with 54 to 141 and 34 µg m-2 per year respectively. At both locations, in these (modern) samples, the Hg concentration profiles resemble those of lead (Pb), an element which is known to be immobile in peat bogs. The correlation between these two metals suggests that the predominant anthropogenic source of Hg (and Pb) was coal burning. While Hg accumulation rates have gone into strong decline since the late 1950`s, Hg deposition rates today still exceed the average natural background values by 7 to 13 times in southern Ontario. Aucune donnée n'a pu être calculée pour les 800 dernières années, faute d'archive naturelle intacte. Cette valeur de flux naturel de mercure est comparable aux flux de mercure calculés à partir de carottes de tourbes provenant pour d'autres localités dans l'Arctique (Groenland et les Iles Féroé) mais également provenant de carottes de tourbes prélevées dans le sud de l'Ontario (enregistrement des taux d'accumulation du mercure atmosphérique remontant à plus de huit milles ans). Ce constat permet de conclure que la retombée de mercure atmosphérique en Arctique au cours de la période pré anthropique n'est pas significativement différent de ces mêmes flux dans la zone tempéré. Ainsi au cours de l'époque pré industriel, l'Arctique n'a donc pas été un puit plus important pour le mercure atmosphérique global que la zone tempéré, discréditant l'hypothèse de la condensation naturelle du mercure atmosphérique dans les zones polaires. D'autres processus sont à invoquer comme mécanismes clés du transfert le mercure atmosphérique à l'environnement Arctique, mécanismes pouvant avoir été rendus plus efficaces dans un passé récent par des changements environnementaux, résultant d'une contamination au mercure de la chaîne alimentaire Arctique.Pour la zone tempérée, le début de la contamination au mercure d'origine anthropique dans le sud de l'Ontario est daté à AD 1475 (Luther Bog), correspond probablement à la combustion de biomasse liée à l'activité agricole des peuples natifs d'Amérique du Nord. Durant la fin du 17ème siècle et au cours du 18ème siècle, la retombée de mercure d'origine anthropique à été au moins égal au dépôt de mercure provenant de sources naturelles. Depuis le début du 19ème siècle, les retombées de mercure atmosphérique sont largement dominées par le mercure anthropique.Les enregistrements tourbeux du sud de l'Ontario et des Iles Féroé présentent des chronologies des taux accumulations de mercure très semblable avec un maximum dans les années 1950 avec de 54 à 141 et 34 µg m-2 par an respectivement. Pour ces deux sites, dans les échantillons modernes, les profiles de concentrations en mercure ressemble ceux de plomb (Pb), un élément qui est connu comme étant immobile dans les tourbières. La corrélation de ces deux métaux suggère que la source prédominante de mercure (et de plomb) était la combustion de charbon. Ceci est confirmé par les mesures des rapports isotopiques du plomb qui identifient clairement la combustion de charbon comme source principale du mercure et du plomb dans les tourbières aux cours des 150 dernières années. Alors que les taux d'accumulations du mercure dans les tourbières sont en net déclin depuis la fin des années 1950, les taux de retombée de mercure excède pourtant toujours les taux naturels d'un ordre de grandeur dans le sud de l'Ontario. L'augmentation des taux de retombée de mercure atmosphérique depuis le début de l'ère industrielle est démontrée, par cette approche, comme étant largement supérieure à celle prédite par les modèles actuels de cycle du mercure.

Dans le cadre de l'étude "Commercialisation et consommation des produits vivriers, horticoles et fruitiers en Polynésie Française", le Cirad, en collaboration avec le SDR, a réalisé une enquête sur les comportements de consommation et la perception du marché des fruits et légumes. A l'occasion de la Foire Agricole de Papeete qui rassemble des visiteurs venant des différentes îles du territoire, un questionnaire a été administré auprès de 325 personnes. Les objectifs de cette enquête étaient: - de mesurer l'importance relative de la consommation des fruits et légumes; - d'identifier les facteurs de différenciation de ces niveaux de consommation liés aux caractéristiques des consommateurs; - de mesurer l'importance relative des différents mode d'approvisionnement en ces produits; - d'identifier la perception, par les consommateurs du marché de ces produits, avec un approfondissement pour certains d'entre eux (orange, banane, mangue, pommes de terre, carotte, choux): perception des lieux d'approvisionnement, des prix, de la qualité; - d'identifier les souhaits des consommateurs pour une amélioration de l'offre en fruits et légumes; - d'identifier la perception, par les consommateurs, de la diffusion des prix par les médias.
