Cancer is a major public health concern because it is one of the main causes of morbidity and mortality worldwide. As a result, numerous studies have reported the development of new therapeutic compounds with the aim of selectively treating cancer while having little negative influence on healthy cells. In this context, earthworm coelomic fluid has been acknowledged as a rich source of several bioactive substances that may exhibit promising anticancer activity. Therefore, the objective of the present review is to evaluate the findings of the reported studies exploring the antitumor effects of coelomic fluid in the context of its possible utilization as a natural therapeutic agent to cure different types of cancer. The possible mechanisms underlying the coelomic fluid's anticancerous potential as well as the possibility for future development of cutting-edge therapeutic agents utilizing coelomic fluid-derived natural bioactive compounds to treat cancer disorders have been discussed along with future challenges. In addition, the feasibility of encapsulation of bioactive compounds derived from coelomic fluid with nanomaterials that could be further explored to attain more effective anticancer competence is discussed. Abstract In contrast to most fishes, salmonids exhibit the unique ability to hold their eggs for several days after ovulation without significant loss of viability. During this period, eggs are held in the body cavity in a biological fluid, the coelomic fluid (CF) that is responsible for preserving egg viability. To identify CF proteins responsible for preserving egg viability, a proteomic comparison was performed using 3 salmonid species and 3 non-salmonid species to identify salmonid-specific highly abundant proteins. In parallel, rainbow trout CF fractions were purified and used in a biological test to estimate their egg viability preservation potential. The most biologically active CF fractions were then subjected to mass spectrometry analysis. We identified 50 proteins overabundant in salmonids and present in analytical fractions with high egg viability preservation potential. The identity of these proteins illuminates the biological processes participating in egg viability preservation. Among identified proteins of interest, the ovarian-specific expression and abundance in CF at ovulation of N-acetylneuraminic acid synthase a (Nansa) suggest a previously unsuspected role. We show that salmonid CF is a complex biological fluid containing a diversity of proteins related to immunity, calcium binding, lipid metabolism, proteolysis, extracellular matrix and sialic acid metabolic pathway that are collectively responsible for preserving egg viability.

Abstract Fetal bovine serum (FBS) plays a pivotal role in animal cell culture. Due to ethical and scientific issues, searching for an alternative, comprising the three R’s (Refinement, Reduction and Replacement) gained global attention. In this context, we have identified the heat inactivated coelomic fluid (HI-CF) of the earthworm, Perionyx excavatus as a potential alternative for FBS. Briefly, we formulated HI-CF (f-HICF) containing serum free medium which can aid the growth, attachment, and proliferation of adherent cells, similar to FBS. In this study, we investigated the biochemical characterization, sterility, stability, formulation, and functional analysis of HI-CF as a supplement in culturing animal cells. Notably, vitamins, micronutrients, proteins, lipids, and trace elements are identified and compared with FBS for effective normalization of the serum free media. HI-CF is tested to be devoid of endotoxin and mycoplasma contamination thus can qualify the cell culture grade. The f-HICF serum free media was prepared, optimised, and tested with A549, HeLa, 3T3, Vero and C2C12 cell lines. Our results conclude that f-HICF is a potential alternative to FBS, in accordance with ethical concern; compliance with 3R's; lack of unintended antibody interactions; presence of macro and micronutrients; simple extraction; cost-effectiveness and availability.

Earthworms’ coelomic fluid (CF) has been discovered to possess properties that promote plant development. In particular, the earthworm’s coelomic fluid-associated bacteria (CFB) are the primary factor influencing the plants’ response. To investigate this, we used bacteria isolated from the CF and selected based on different plant growth-promoting traits, in a mesocosm ecosystem that includes plants. This experiment aimed to assess their impact on the metabolism of plants growing under abiotic stress environments (alkaline soil and nitrogen (N), phosphate (P), and potassium (K) deficit) and compare the lipid profiles of plants under the various treatments. We used seven different bacterial species isolated from the CF of Aporrectodea molleri and as a plant model Zea mays L. For the metabolomic analysis method, we used gas chromatography-mass spectrometry lipidomic. After observing the metabolomic profiles, we found that a few molecular pathways are involved in how plants react to bacterial biostimulants. The bacterial isolates belonging to Pantoea vagans, Pseudomonas aeruginosa, Bacillus paramycoides, and Bacillus thuringiensis have led to a significant increase in synthesizing several metabolites belonging to various chemical categories. Contrary to predictions, abiotic stress did not cause a drop in the composition and concentration of lipids in plants treated with the CFB, demonstrating the rigidity of the protective mechanisms. The statistical analysis based on the Pearson method revealed a positive significant correlation between plant growth parameters (length of the aerial part, surface of the leaves, and biomass) and some metabolites belonging to fatty acids, carboxylic acids, benzene derivatives, and alkanes. Moreover, the standard metabolic components of all treatments in much higher concentrations during bacterial treatments than the control treatment suggests that the bacteria have stimulated the overexpression of these metabolic components. According to these results, we could assume that plants treated with CFB exhibit an adaptability of abiotic stress defense mechanisms, which may be attributed to the upregulation of genes involved in lipid biosynthesis pathways.

The goal of the study was to develop necessary skills for B. Tech students through an undergraduate project in biotechnology. Understanding of biotechnology involves keen observation of results and asking systematic and logical questions to find out the causes of key observation. This paper introduces teaching and attainment of essential skills during execution of their final year project. The project was focused on identifying the significance of coelomic fluid (CF) of earthworm in plant tissue culture medium by using the model plant tobacco. Project outcomes are: understanding the purpose of an experiment, conducting experiments in basic and applied areas of biotechnology, interpretation of results and communicate clearly during viva-voce examination and documenting the results in a scientific way. Essential skills viz: reading and understanding the literature, how to design an experiment with proper controls, logical explanation of results and communication skills were evaluated every month by rubrics designed by investigators. Assessment was further done by feedback system and surveys from the students. The project outcomes were matched with essential skills and their attainment was measured. Based on the evaluation it is concluded that this research project-based learning helped students to develop essential skills such as designing of experiments, logical explanation of results, communication and documentation of their observations with a conclusion in a scientific way. The cavity of earthworm is filled with transparent fluid called coelomic fluid which is derived from the mesenchymal lining, exhibits a wide variety of biological activities. The coleomic fluid consists of various cells and many biomolecular components involved in innate immunity. This study aims to highlight the use of various standard methods for the extraction of coelomic fluid and also to calculate the differential coelomocytes count of three epigeic earthworm species such as Eudrilus eugeniae (EE), Eisenia fetida (EF) and Perioynx excavatus (PE). In present study, cold shock method was used for extraction of coelomic fluid from different epigeic earthworm species as it is more advantages than the other methods, especially for earthworms. During the study, four different kinds of coelomocytes were witnessed such as amoebocytes, chloragogen cells, mucocytes and circular cells. The total coelomocytes count was somewhat equal in all three species with no significant difference, whereas a significant difference was noticed in differential count of coelomocytes i.e. Among all the coelomocytes amoebocytes, chloragogen cell and mucocytes were more in EE (56.33±0.33, 37.00±0.57 and 3.66±0.33) followed by PE (31.00±0.57, 16.33±0.33 and 2.00±0.57) and less in EF (24.00±0.32, 8.00±0.21 and 2.00±0.33) whereas circular cells were more in EF ( 66.00±0.57) followed by PE (50.66±0.33) and less in EE (2.66±0.33). Abstract Background Coelomic fluid, a pharmacologically active compound in earthworms, exhibits a range of biological activities, including antioxidant, anti-inflammatory, and anticancer. However, the biological activities exerted by the coelomic fluid can be restrained by its low bioavailability and stability. Liposomes are progressively utilized as an entrapment system for natural bioactive compounds with poor bioavailability and stability, which could be appropriate for coelomic fluid. Thus, the present study was designed to fabricate, characterize, and evaluate the stability of liposomal formulation for Allolobophora caliginosa coelomic fluid (ACCF) as a natural antioxidant compound. Methods The ACCF-liposomes were developed with a subsequent characterization of their physicochemical attributes. The physical stability, ACCF release behavior, and gastrointestinal stability were evaluated in vitro. The biological activities of ACCF and its liposomal formulation were also determined. Results The liposomal formulation of ACCF had a steady characteristic absorption band at 201 nm and a transmittance of 99.20 ± 0.10%. Its average hydrodynamic particle size was 98 nm, with a PDI of 0.29 ± 0.04 and a negative zeta potential (-38.66 ± 0.33mV). TEM further confirmed the formation of vesicular, spherical nano-liposomes with unilamellar configuration. Additionally, a remarkable entrapment efficiency percent (77.58 ± 0.82%) with a permeability rate equal to 3.20 ± 0.31% and a high retention rate (54.16 ± 2.20%) for ACCF-liposomes were observed. The Fourier transform infrared spectroscopy (FTIR) result demonstrated that ACCF successfully entrapped inside liposomes. The ACCF-liposomes exhibited a slow and controlled ACCF release in vitro. Regarding stability studies, the liposomal formulation enhanced the stability of ACCF during storage and at different pH. Furthermore, ACCF-liposomes are highly stable in intestinal digestion conditions comparable to gastric digestion. The current study disclosed that liposomal formulation potentiates the biological activities of ACCF, especially antioxidant, anti-inflammatory, and thrombolytic activities. Conclusion These promising results offer a novel approach to increasing the bioaccessibility of ACCF, which may be crucial for the development of pharmaceuticals and nutraceutical-enriched functional foods.

The functioning of the immune and reproductive systems is crucial for the fitness and survival of species and is strongly influenced by the environment. To evaluate the effects of short-term heat stress (HS) on these systems, confirming and deepening previous studies, female sea urchin Paracentrotus lividus were exposed for 7 days to 17 °C, 23 and 28 °C. Several biomarkers were detected such as the ferric reducing power (FRAP), ABTS-based total antioxidant capacity (TAC-ABTS), nitric oxide metabolites (NOx), total thiol levels (TTL), myeloperoxidase (MPO) and protease (PA) activities in the coelomic fluid (CF) and mitochondrial membrane potential (MMP), H2O2 content and intracellular pH (pHi) in eggs and coelomocytes, in which TAC-ABTS and reactive nitrogen species (RNS) were also analyzed. In the sea urchins exposed to HS, CF analysis showed a decrease in FRAP levels and an increase in TAC-ABTS, TTL, MPO and PA levels; in coelomocytes, RNS, MMP and H2O2 content increased, whereas pHi decreased; in eggs, increases in MMP, H2O2 content and pHi were found. In conclusion, short-term HS leads to changes in five out of the six CF biomarkers analyzed and functional alterations in the cells involved in either reproductive or immune activities. The coelomic fluid (haemocoel) and coelomocytes of echinoderms were scarcely studied in Egypt. Remarkably, the most popular species is the fivearms starfish. While, in this work, the microscopical and chemical characteristics of the 7-arms giant starfish "Luidia maculata" were addressed. The starfish were gathered at 7 meters' depth in the seawater of Ras Sedr, Gulf of Suez, Red Sea, Egypt. As far as we know, no other microscopical data have been published on this species in Egypt. The extracted coelomic fluid of this giant starfish "Luidia maculata" was microscopically and chemically analyzed using TEM and GC/MS, respectively. The exhibited data showed the presence of glycogen synthase kinase3-β protein by protein docking, which is considered a promising constituent for wound healing. Finally, this work spotted the light on novel documented data on the microscopical and chemical characteristics of barely studied species of starfish "Luidia maculata", in which the studied coelomocytes showed an amoeboid state resembling the human white blood cells. The examined flagella displayed a typical microtubule arrangement "9+2 array". The coelomic fluid extracted from "Luidia maculata" exhibits glycogen synthase kinase3-β (GSK3-β) protein, which could be considered a promising biovital constituent for many medicinal applications including wound healing in diabetics. Simple Summary: The immune response to pathogens is one of the organic functions at risk due to climatic change. The effects of a short-term increase in temperature on immune cell functions have been evaluated using a simple animal model, such as the sea urchin, exposed to temperatures above its comfort zone (17 °C), causing moderate and severe heat stress. After 3 and 7 days of exposure to the three temperatures tested, the coelomic fluid of the animals was collected. The coelomocytes were isolated and typed and the mitochondrial activity, the amount of lipid peroxidation, and the production of hydrogen peroxide were evaluated. These cells were also stimulated with phorbol 12-myristate 13-acetate (PMA) to evoke respiratory burst, a marker of immune response. Heat stress altered both the distribution of the various types of coelomocytes and their mitochondrial activity; it also increased lipid peroxidation and reduced the production of hydrogen peroxide. The respiratory burst occurred in the coelomocytes of all three experimental groups; however, it was greater in the group kept at 17 °C. In conclusion, thermal increase alters immune cell functions with possible repercussions on defense mechanisms against pathogens. Due to global warming, animals are experiencing heat stress (HS), affecting many organic functions and species' survival. In this line, some characteristics of immune cells in sea urchins subjected to short-term HS were evaluated. Paracentrotus lividus adult females were randomly divided into three groups and housed in tanks at 17 °C. In two of these tanks, the temperatures were gradually increased up to 23 and 28 °C. Celomatic fluid was collected after 3 and 7 days. The coelomocytes were morphologically typed and evaluated for their mitochondrial membrane potential (MMP), lipoperoxidation extent (LPO), and hydrogen peroxide content (H2O2). Respiratory burst was induced by treatment with phorbol 12-myristate 13-acetate (PMA). HS caused a significant change in the coelomocytes' type distribution. MMP increased in the 23 °C-group and decreased in the 28 °C-group at both 3 and 7 days. LPO only increased in the 28 °C-group at 7 days. H2O2 progressively decreased together with the temperature increase. Respiratory burst was detected in all groups, but it was higher in the 17 °C group. Antibiotic drug resistance is a global public health issue that demands new and novel therapeutic molecules. To develop new agents, animal secretions or products are used as an alternative agent to overcome this problem. In this study, earthworm (Pheretima posthuma) coelomic fluid (PCF), and body paste (PBP) were used to analyze their effects as antibiofilm agents against four bacterial isolates MH1 (Pseudomonas aeruginosa MT448672), MH2 (Escherichia coli MT448673), MH3 (Staphylococcus aureus MT448675), and MH4 (Klebsiella pneumoniae MT448676). Coelomic fluid extraction and body paste formation were followed by minimum inhibitory concentrations (MICs), biofilm formation time kinetics, and an antibiofilm assay, using heat and cold shock, sunlight exposure auto-digestion, and test tube methods. The results showed that the MIC values of PCF and PBP against S. aureus, P. aeruginosa, K. pneumoniae, and E. coli bacterial isolates ranged from 50 to 100 μg/mL, while, the results related to biofilm formation for P. aeruginosa, S. aureus, and K. pneumoniae strains were observed to be highly significantly increased (p < 0.005) after 72 h. E. coli produced a significant (p < 0.004) amount of biofilm after 48 h. Following time kinetics, the antibiofilm activity of PCF and PBP was tested at different concentrations (i.e., 25–200 μg/mL) against the aforementioned four strains (MH1–MH4). The findings of this study revealed that both PBP (5.61 ± 1.0%) and PCF (5.23 ± 1.5%) at the lowest concentration (25 μg/mL) showed non-significant (p > 0.05) antibiofilm activity against all the selected strains (MH1-MH4). At 50 μg/mL concentration, both PCF and PBP showed significant (p < 0.05) biofilm inhibition (<40%) for all isolates. Further, the biofilm inhibitory potential was also found to be more significant (p < 0.01) at 100 μg/mL of PCF and PBP, while it showed highly significant (p < 0.001) biofilm inhibition at 150 and 200 μg/mL concentrations. Moreover, more than 90% biofilm inhibition was observed at 200 μg/mL of PCF, while in the case of the PBP, <96% biofilm reduction (i.e., 100%) was also observed by all selected strains at 200 μg/mL. In conclusion, earthworm body fluid and paste have biologically active components that inhibit biofilm formation by various pathogenic bacterial strains. For future investigations, there is a need for further study to explore the potential bioactive components and investigate in depth their molecular mechanisms from a pharmaceutical perspective for effective clinical utilization. Earthworms’ celomic fluid has long attracted scientists’ interest due to their toxic properties. It has been shown that the elimination of coelomic fluid cytotoxicity to normal human cells was crucial for the generation of the non-toxic Venetin-1 protein-polysaccharide complex, which exhibits selective activity against Candida albicans cells as well as A549 non-small cell lung cancer cells. To find the molecular mechanisms behind the anti-cancer properties of the preparation, this research investigated the proteome response of A549 cells to the presence of Venetin-1. The sequential window acquisition of all theoretical mass spectra (SWATH-MS) methodology was used for the analysis, which allows for a relative quantitative analysis to be carried out without radiolabelling. The results showed that the formulation did not induce significant proteome responses in normal BEAS-2B cells. In the case of the tumour line, 31 proteins were up regulated, and 18 proteins down regulated. Proteins with increased expression in neoplastic cells are mainly associated with the mitochondrion, membrane transport and the endoplasmic reticulum. In the case of altered proteins, Venetin-1 interferes with proteins that stabilise the structures, i.e., keratin, glycolysis/gluconeogenesis and metabolic processes.

Objective: In this paper, the inhibition of protein degradation in autolysis of white shrimp （Penaeus vannamei） surimi by extracts from the coelomic fluid of sea cucumber （Stichopus japonicus）, a by-product of sea cucumber processing, was studied. Methods: The protein degradation in autolysis of white shrimp was studied by SDS-PAGE assay. Furthermore, the effect of sea cucumber coelomic fluid and its extracts on white shrimp protein degradation was determined by SDS-PAGE and TCA-soluble oligopeptide assay. Results: The optimal conditions of white shrimp autolysis were incubation at pH 3.0, 50 ℃ for 2 h. The bond intensities of myosin heavy chain around （200 kDa） and actin around （44.3 kDa） decreased suggesting the protein degradation of white shrimp. The sea cucumber coelomic fluid showed inhibitory effect on protein degradation in autolysis of white shrimp. When the concentration of coelomic fluid extract of sea cucumber was in the range of 6.25%~25.00%, the protein degradation was significantly inhibited. The release of TCA-soluble oligopeptide was also significantly inhibited by coelomic fluid extract at 12.50%~25.00%. Conclusion: Sea cucumber coelomic fluid extracts possess inhibitory effect on protein degradation of white shrimp, indicating suppression of white shrimp surimi autolysis.

This study evaluated the impact of Aporrectodea molleri's coelomic fluid-associated bacteria (CFB) on Zea mays L. growth and soil biochemical characteristics under abiotic stress conditions, including alkaline soil (pH = 8) and nitrogen (N), phosphate (P), and potassium (K) deficit. Compared to maize cultivated in uninoculated soil, the effect of CFB on boosting plant growth under abiotic stress was notably exceptional. Different CFB treatments increased significantly root and shoot length by 50% and 21%, respectively. Furthermore, the presence of isolates in soil resulted in a significant increase in plant fresh and dry weights (of up to 113% and 91% for roots, and up to 173% and 44% for shoots), leaf surface (78%), and steam diameter (107%). Overall, soil inoculation with CFB significantly (P < 0.05) enhanced chlorophyll and water content in the plant compared to the untreated soil. Despite the soil's alkaline condition, CFB drastically boosted soil quality by increasing nutrient availability (up to 30 ppm for N, 2 ppm for P, and 60 ppm for K) and enzyme activity (up to 1.14 μg p-NP h−1 g−1 for acide phosphatase, 9 μg p-NP h−1 g−1 for alkaline phosphatase and 40 μg NH4-N 2 h−1 g−1 for urease), throughout the early stages of the growth period. Interestingly, alkaline phosphatase concentrations were substantially greater in treatments with different isolates than acid phosphatase. Furthermore, the principal component analysis showed that the inoculation with bacteria strains CFB1 Buttiauxella gaviniae and CFB3 Aeromonas hydrophila had a significantly better stimulatory stimulatory and direct influence on maize growth than the other isolates had a substantial effect on soil's biochemical features. In this study, activities and carbohydrate specificities of agglutinins in body fluids of invertebrates from waters of the Russian coast of the Sea of Japan were assessed. Samples from 30 animal species belonging to four phyla, seven classes, and the order Sipuncula (Annelida: Polychaeta, Sipuncula; Arthropoda: Thecostraca, Malacostraca; Mollusca: Bivalvia, Gastropoda; Echinodermata: Holothuroidea, Asteroidea) were analyzed. For samples of 12 species, a range of carbohydrate specificities was determined, some of which are characteristic of corresponding systematic groups, and the other are unique and have not previously been mentioned in the literature. In addition, for the studied representatives of four classes (Polychaeta, Thecostraca, Malacostraca, and Bivalvia) and the order Sipuncula, patterns of similarities and differences in the carbohydrate specificities of their agglutinins were identified. For example, the specificity to acidic and neutral carbohydrates of pathogen-associated molecular patterns was clearly pronounced for polychaetes and, to a lesser extent, for malacostracans. In the case of bivalves and barnacles, another most evident specificity was that to N-acetylated amines. Sipunculans, which are one of the least studied groups as regards the carbohydrate specificity of agglutinins, showed a unique pattern of specificity with the highest inhibitory effect to pectin. Acute saline stress can cause death of Phasolosma esculenta, an important aquaculture species in China. Hence, investigating the sensitivity and physiological responses of P. esculenta to saline stress is important. The molecular, physiological, and immune responses of the intestine and body wall of P. esculenta upon saline stress were investigated in this study. The mortality rate of P. esculenta increased with the decreasing salinity. The Na+/K+-ATPase and acid phosphatase activity peaked at 6 h and 12 h, respectively, in the intestine and body wall, and subsequently stabilized at a higher level than control group. Further, superoxide dismutase activity significantly decreased within 12 h and then increased slowly, while its genes expression decreased and reached its lowest level at 6 h. In the intestine and body wall of P. esculenta, saline stress induced the transcription of Heat Shock Protein-70, Heat Shock Protein-90, and ferritin, along with the enlargement and loosening of connective tissue cells. Moreover, saline stress induced a more prominent adaptive response in the intestine than body wall. This research indicated that the enzymatic system, nonenzymatic system, and gene expressions of P. esculenta changed under low saline stress and provided references for the further research on the low saline stress response in P. esculenta. Due to their tissue structure similar to mammalian skin and their close evolutionary relationship with chordates, holothurians (Echinodermata: Holothuroidea) are particularly interesting for studies on wound healing. However, previous studies dealing with holothuroid wound healing have had limited approaches, being restricted to tissue repair or perivisceral immune response. In this study, we combined tissue, cellular and humoral parameters to study the wound healing process of Holothuria grisea. The immune responses of the perivisceral coelom were assessed by analyzing the number, proportion and viability of coelomocytes and the volume and protein concentration of the coelomic fluid. Additionally, the morphology of the healing tissue and number of coelomocytes in the connective tissue of different body wall layers were examined over 30 days. Our results showed that perivisceral reactions started 3 h after injury and decreased to baseline levels within 24 h. In contrast, tissue responses were delayed, beginning after 12 h and returning to baseline levels only after day 10. The number of coelomocytes in the connective tissue suggests a potential cooperation between these cells during wound healing: phagocytes and acidophilic spherulocytes act together in tissue clearance/homeostasis, whereas fibroblast-like and morula cells cooperate in tissue remodeling. Finally, our results indicate that the major phases observed in mammalian wound healing are also observed in H. grisea , despite occurring at a different timing, which might provide insights for future studies. Based on these data, we propose a model that explains the entire healing process in H. grisea. The functioning of the immune and reproductive systems is crucial for the fitness and survival of species and is strongly influenced by the environment. To evaluate the effects of short-term heat stress (HS) on these systems, confirming and deepening previous studies, female sea urchin Paracentrotus lividus were exposed for 7 days to 17 °C, 23 and 28 °C. Several biomarkers were detected such as the ferric reducing power (FRAP), ABTS-based total antioxidant capacity (TAC-ABTS), nitric oxide metabolites (NOx), total thiol levels (TTL), myeloperoxidase (MPO) and protease (PA) activities in the coelomic fluid (CF) and mitochondrial membrane potential (MMP), H2O2 content and intracellular pH (pHi) in eggs and coelomocytes, in which TAC-ABTS and reactive nitrogen species (RNS) were also analyzed. In the sea urchins exposed to HS, CF analysis showed a decrease in FRAP levels and an increase in TAC-ABTS, TTL, MPO and PA levels; in coelomocytes, RNS, MMP and H2O2 content increased, whereas pHi decreased; in eggs, increases in MMP, H2O2 content and pHi were found. In conclusion, short-term HS leads to changes in five out of the six CF biomarkers analyzed and functional alterations in the cells involved in either reproductive or immune activities.

The peanut worm (Sipunculus nudus) is an important economic and ecological fishery resource in China. To determine how S. nudus interacts with its culture environment, in this study, the diversity, composition, and interrelationships of bacterial communities in the intestine, coelomic fluid, and culture environment of S. nudus through high-throughput sequencing using the Illumina HiSeq sequencing platform. Alpha diversity analysis showed that the highest bacterial community richness and diversity were found in the sediments. Species annotation revealed that Proteobacteria dominated in all samples (48.92%–58.36%), and the other dominant phyla were highly variable, indicating a certain independence between the environment and the composition of the bacterial community of S. nudus. The coelomic fluid of S. nudus contained high concentrations of Bacillus and could be a source of potential probiotic bacteria for isolation and culture. Cluster and operational taxonomic unit Venn diagram analyses showed that the bacterial community composition in the intestine was more similar to that in the sediments compared with water. The predicted functional analysis of bacterial communities indicated that the functions of bacterial communities in the different surrounding environment were highly similar and involved in various aspects, such as membrane transport, amino acid metabolism, carbohydrate metabolism, and replication and repair. Our findings have important implications on the establishment of sustainable microecological regulation and management strategies for S. nudus culture and provide a reference for the development and utilization of probiotic bacteria.

Echinoderms are an acknowledged source of bioactive compounds exerting various beneficial effects on human health. Here, we examined the potential in vitro anti-hepatocarcinoma effects of aqueous extracts of the cell-free coelomic fluid obtained from the sea urchin Arbacia lixula using the HepG2 cell line as a model system. This was accomplished by employing a combination of colorimetric, microscopic and flow cytometric assays to determine cell viability, cell cycle distribution, the possible onset of apoptosis, the accumulation rate of acidic vesicular organelles, mitochondrial polarization, cell redox state and cell locomotory ability. The obtained data show that exposed HepG2 cells underwent inhibition of cell viability with impairment of cell cycle progress coupled to the onset of apoptotic death, the induction of mitochondrial depolarization, the inhibition of reactive oxygen species production and acidic vesicular organelle accumulation, and the block of cell motile attitude. We also performed a proteomic analysis of the coelomic fluid extract identifying a number of proteins that are plausibly responsible for anti-cancer effects. Therefore, the anti-hepatocarcinoma potentiality of A. lixula's preparation can be taken into consideration for further studies aimed at the characterization of the molecular mechanism of cytotoxicity and the development of novel prevention and/or treatment agents. The electrospun mats consisting of integrated coelomic fluid (CF) and chitosan (Chs) into polyvinyl alcohol (PVA) nanofibers were produced and evaluated for use as wound dressings. CF was obtained from earthworms (Eisenia andrei (Fetida)) using an electric shock method, while Chs was chemically produced from shrimp chitin and then characterized using titration, Fourier transform infrared (FT-IR) spectroscopy, and viscometry. The wound dressings with different CF contents were evaluated for their antibacterial, antioxidant, and cell viability properties. The dressings infused with CF showed significantly higher antibacterial and antioxidant activity, as well as improved cell viability compared to the control without CF. In vivo studies using adult Wistar albino rats showed that the Chs/PVA/CF wound dressings promoted wound healing and re-epithelialization. Moreover, histological examinations of the injuries coated with Chs/PVA/CF displayed improved re-epithelialization. Due to global warming, animals are experiencing heat stress (HS), affecting many organic functions and species’ survival. In this line, some characteristics of immune cells in sea urchins subjected to short-term HS were evaluated. Paracentrotus lividus adult females were randomly divided into three groups and housed in tanks at 17 °C. In two of these tanks, the temperatures were gradually increased up to 23 and 28 °C. Celomatic fluid was collected after 3 and 7 days. The coelomocytes were morphologically typed and evaluated for their mitochondrial membrane potential (MMP), lipoperoxidation extent (LPO), and hydrogen peroxide content (H2O2). Respiratory burst was induced by treatment with phorbol 12-myristate 13-acetate (PMA). HS caused a significant change in the coelomocytes’ type distribution. MMP increased in the 23 °C-group and decreased in the 28 °C-group at both 3 and 7 days. LPO only increased in the 28 °C-group at 7 days. H2O2 progressively decreased together with the temperature increase. Respiratory burst was detected in all groups, but it was higher in the 17 °C group. In conclusion, the increase in temperature above the comfort zone for this animal species affects their immune cells with possible impairment of their functions.

Earthworms are tube-shaped, segmental worms and have a double transport system consisting of earthworm coelomic fluid (ECF) moving within the fluid-filled colon. As one of the naturally available extracts (NAEs), ECF exerts different biological functions, such as inhibiting the proliferation of cancer cells. Pharmacokinetic limitations in prostate cancer (PCa) treatment and undesirable side effects of chemotherapeutic drugs necessitate the discovery of new anticancer agents. Recent studies have focused on the antitumor activity of different regions of earthworms as a new potential anticancer agent. However, there are limited findings on the potential effects of ECF on PCa cells in vitro. Therefore, we aimed to evaluate the potential anticancer effects of ECF (obtained from Eisenia foetida) on two different types of PCa cells. In this study, the protein content of ECF, the most effective dose and incubation time and potentially genotoxic and apoptotic effects were determined in PC-3 and LNCaP cells for the first time. Bax and Bcl-2 gene and protein expression levels were determined by real-time polymerase chain reaction (RT-PCR) and western blotting, respectively. ECF increased apoptotic cell death; however, it did not have a significant genotoxic effect on PCa cells. The apoptotic effect of ECF in PCa cells varies according to the AR status and is more effective in AR-insensitive cells (PC-3). Therefore, our results will specifically support the pharmacological development of ECF as a potential anticancer molecule in the treatment of PCa in the future. [ABSTRACT FROM AUTHOR]

Cockayne's syndrome (CS) is a rare autosomal recessive multisystem disease characterised by early severe progression of symptoms. This study reports the feasibility of earlier prenatal diagnosis of CS by coelocentesis at 8 weeks of gestation respect to amniocentesis or villocentesis. Three couples at risk for CS asked to perform prenatal diagnosis by coelocentesis. Coelomic fluid was aspired from coelomic cavity in four singleton pregnancy at 8 weeks of gestation and 40 foetal cells were recovered by micromanipulator. Maternal DNA contamination was evaluated by quantitative fluorescent PCR (QF-PCR) and target regions of foetal DNA containing parental mutations of ERCC6 gene were amplified and sequenced. In all these cases, molecular analysis was possible. One foetus resulted affected of CS and the diagnosis was confirmed on placental tissue after voluntary abortion. In three cases, foetuses resulted carrier of a parental mutation and the results were confirmed after the birth. This study suggests that reliable prenatal diagnosis of CS could be performed using foetal cells present in coelomatic fluid in earlier pregnancy. Coelocentesis could be applied in prenatal diagnosis of CSs as well as for other monogenic diseases, at very early stage of pregnancy, if parental mutations are already known. What is already know on this subject? Previous studies utilising coelocentesis for prenatal determination of foetal sex reported variable success ranging from 58% to 95%, because of low total DNA content and presence of maternal cell contamination. This procedure has never been reported for early prenatal diagnosis at 8 weeks of gestation for rare genetically transmitted diseases such as Cockayne's syndrome. What do the results of this study add? This study demonstrates that coelomic fluid sampling combined with well-standardised laboratory procedures can be applied for prenatal diagnosis at eight weeks of gestation for any rare monogenic disease if molecular defects are known. What are the implications of these findings for clinical practice and/or further research? The findings of this study in at risk couples for monogenic diseases investigated by coelocentesis demonstrate that embryo-foetal cell selection from CF allows reliable and early prenatal diagnosis of diseases. This technique is attractive to parents because it provides prenatal diagnosis of genetic disease at least 4 weeks earlier than what can be achieved by the traditional procedures reducing anxiety of parents and provides the option for medical termination of affected cases at 8–10 weeks' gestation, which is less traumatic and safer than second-trimester surgical termination. Earthworm metabolism is recognized as a useful tool for monitoring environmental insults and measuring ecotoxicity, yet extensive earthworm metabolic profiling using 1H nuclear magnetic resonance (NMR) spectroscopy has been limited in scope. This study aims to expand the embedded metabolic material in earthworm coelomic fluid, coelomocytes, and tissue to aid systems toxicology research. Fifty-nine metabolites within Eisenia fetida were identified, with 47 detected in coelomic fluid, 41 in coelomocytes, and 54 in whole-worm samples and tissue extracts. The newly detected but known metabolites 2-aminobutyrate, nicotinurate, Nδ,Nδ,Nδ-trimethylornithine, and trigonelline are reported along with a novel compound, malylglutamate, elucidated using 2D NMR and high-resolution MS/MS. We postulate that malylglutamate acts as a glutamate/malate store, chelator, and anionic osmolyte and helps to provide electrolyte balance.

Synthetic chemicals, such as fertilizers and pesticides, are abundantly used in agriculture to enhance soil fertility and prevent the occurrence of diseases, respectively. Many studies have reported a negative influence of these chemicals on the soil environment. Natural sources from earthworms and their products, as a result of vermicomposting, may be considered better alternatives. The aim of this review was to reveal the source of antifungal efficiency of vermicompost and its derivatives, such as vermiwash, coelomic fluid, skin secretion of earthworms, and metabolites from decomposer bacteria in vermicompost, in order to highlight their application in agriculture. The synergistic activity of bioactive compounds present in coelomic fluid, mucus, skin secretion, and metabolites from associated bacteria (decomposer) assisted crop plants for effective action against various soil pathogenic fungi, such as Rhizoctoniasolani, Alternaria solani, Aspergillus niger, A.flavus, Fusariumoxysporum, and F. graminearum. Thus, these bioactive metabolites can be recommended to suppress plant fungal diseases. Earthworms' coelomic fluid (CF) has been discovered to possess properties that promote plant development. In particular, the earthworm's coelomic fluid-associated bacteria (CFB) are the primary factor influencing the plants' response. To investigate this, we used bacteria isolated from the CF and selected based on different plant growth-promoting traits, in a mesocosm ecosystem that includes plants. This experiment aimed to assess their impact on the metabolism of plants growing under abiotic stress environments (alkaline soil and nitrogen (N), phosphate (P), and potassium (K) deficit) and compare the lipid profiles of plants under the various treatments. We used seven different bacterial species isolated from the CF of Aporrectodea molleri and as a plant model Zea mays L. For the metabolomic analysis method, we used gas chromatography-mass spectrometry lipidomic. After observing the metabolomic profiles, we found that a few molecular pathways are involved in how plants react to bacterial biostimulants. The bacterial isolates belonging to Pantoea vagans , Pseudomonas aeruginosa , Bacillus paramycoides , and Bacillus thuringiensis have led to a significant increase in synthesizing several metabolites belonging to various chemical categories. Contrary to predictions, abiotic stress did not cause a drop in the composition and concentration of lipids in plants treated with the CFB, demonstrating the rigidity of the protective mechanisms. The statistical analysis based on the Pearson method revealed a positive significant correlation between plant growth parameters (length of the aerial part, surface of the leaves, and biomass) and some metabolites belonging to fatty acids, carboxylic acids, benzene derivatives, and alkanes. Moreover, the standard metabolic components of all treatments in much higher concentrations during bacterial treatments than the control treatment suggests that the bacteria have stimulated the overexpression of these metabolic components. According to these results, we could assume that plants treated with CFB exhibit an adaptability of abiotic stress defense mechanisms, which may be attributed to the upregulation of genes involved in lipid biosynthesis pathways.

Objective: To explore the efficacy of earthworm’s coelomic fluid against gentamicin-induced hepatic and renal toxicity in rats. Methods: The animals were divided randomly into three groups (n = 6 per group): control, gentamicin, and Allolobophora caliginosa coelomic fluid-treated groups. Toxicity was established after injection of gentamicin daily for 8 days at a dose of 100 mg/kg. Aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, total proteins, albumin, creatinine, urea, uric acid, malondialdehyde, glutathione, catalase and histopathology of tissues were investigated in the study. Results: Allolobophora caliginosa coelomic fluid significantly decreased urea, creatinine, uric acid, alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, and malondialdehyde levels while significantly increasing levels of total proteins, albumin, glutathione and catalase. The histopathological investigation showed partial restoration of renal and hepatic architecture. Conclusions: This study shows the potency of Allolobophora caliginosa coelomic fluid in improving the biochemical and histopathological changes induced by gentamicin in the liver and kidney of the rats.

Antibiotic drug resistance is a global public health issue that demands new and novel therapeutic molecules. To develop new agents, animal secretions or products are used as an alternative agent to overcome this problem. In this study, earthworm (Pheretima posthuma) coelomic fluid (PCF), and body paste (PBP) were used to analyze their effects as antibiofilm agents against four bacterial isolates MH1 (Pseudomonas aeruginosa MT448672), MH2 (Escherichia coli MT448673), MH3 (Staphylococcus aureus MT448675), and MH4 (Klebsiella pneumoniae MT448676). Coelomic fluid extraction and body paste formation were followed by minimum inhibitory concentrations (MICs), biofilm formation time kinetics, and an antibiofilm assay, using heat and cold shock, sunlight exposure auto-digestion, and test tube methods. The results showed that the MIC values of PCF and PBP against S. aureus, P. aeruginosa, K. pneumoniae, and E. coli bacterial isolates ranged from 50 to 100 μg/mL, while, the results related to biofilm formation for P. aeruginosa, S. aureus, and K. pneumoniae strains were observed to be highly significantly increased (p < 0.005) after 72 h. E. coli produced a significant (p < 0.004) amount of biofilm after 48 h. Following time kinetics, the antibiofilm activity of PCF and PBP was tested at different concentrations (i.e., 25–200 μg/mL) against the aforementioned four strains (MH1–MH4). The findings of this study revealed that both PBP (5.61 ± 1.0%) and PCF (5.23 ± 1.5%) at the lowest concentration (25 μg/mL) showed non-significant (p > 0.05) antibiofilm activity against all the selected strains (MH1-MH4). At 50 μg/mL concentration, both PCF and PBP showed significant (p < 0.05) biofilm inhibition (p < 0.01) at 100 μg/mL of PCF and PBP, while it showed highly significant (p < 0.001) biofilm inhibition at 150 and 200 μg/mL concentrations. Cancer is a major public health concern because it is one of the main causes of morbidity and mortality worldwide. As a result, numerous studies have reported the development of new therapeutic compounds with the aim of selectively treating cancer while having little negative influence on healthy cells. In this context, earthworm coelomic fluid has been acknowledged as a rich source of several bioactive substances that may exhibit promising anticancer activity. Therefore, the objective of the present review is to evaluate the findings of the reported studies exploring the antitumor effects of coelomic fluid in the context of its possible utilization as a natural therapeutic agent to cure different types of cancer. The possible mechanisms underlying the coelomic fluid's anticancerous potential as well as the possibility for future development of cutting-edge therapeutic agents utilizing coelomic fluid-derived natural bioactive compounds to treat cancer disorders have been discussed along with future challenges. In this study, the effect of coelomic fluid isolates (CFIs) from sea cucumbers (Stichopus japonicus) on autolysis of minced muscle tissue of S. japonicus was investigated. The coelomic fluid of S. japonicus at the concentration of 90% showed suppression effects on activities of papain and trypsin with inhibitory rate of 26.6% and 40.4%, respectively. Moreover, the effect of CFIs on autolysis of minced muscle tissue from S. japonicus was analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and trichloroacetic acid (TCA)-soluble oligopeptide assay. CFIs exhibited significantly inhibitory effect on autolytic muscle mince samples with inhibitory rate at 56.2–87.2%. Both heat and proteinase treatments did not influence the inhibitory effect of CFIs on autolysis of minced muscle tissue from S. japonicus. These results suggest that the non-protein and thermally stable substances contribute to the inhibitory effect of CFIs, which can be used to improve sea cucumber quality. [ABSTRACT FROM AUTHOR]

Fetal bovine serum (FBS) is a widely used growth supplement in the in vitro culturing of animal and human cells, tissues and organs, notably due to the occurrence of abundant micro- and macronutrients, along with growth factors. Over the years, increasing demand, high price, batch-to-batch variability in quality and composition, increasing ethical concerns lead to the search for an alternative to FBS. Several approaches have been suggested and employed in the past, but none is implemented as widely as FBS, and each supplement has its own disadvantages. In this review, we described the importance of FBS in cell culture, discussed the issues associated with FBS use and presented the efforts made in the recent past to reduce or replace FBS. The potential of four different alternative sources to FBS, namely, bovine ocular fluid, sericin protein, human platelet lysate and earthworm heat inactivated coelomic fluid was evaluated. In the end, we present the conceptual perspective using the Human Platelet Lysate (HPL) and earthworm Heat Inactivated Coelomic Fluid (HI-CF) combination to alternate FBS and its context in scientific and economic impacts.

Therefore, the anti-hepatocarcinoma potentiality of A. lixula’s preparation can be taken into consideration for further studies aimed at the characterization of the molecular mechanism of cytotoxicity and the development of novel prevention and/or treatment agents.

Summary: In this study, we tried to maintain the vitality of Chinese sturgeon (Acipenser sinensis) ova before fertilization with several treatments in vitro. The ovulated eggs were allocated to groups with different incubation medium (coelomic fluid and artificial media), temperature (4°C and 16°C) and storage duration (2 h and 6 h). The maximum fertilization and hatching rate were observed for the control group in which the ova were fertilized immediately after spawning, with the values of 82.45% and 84.73%, respectively. Compared with the control group, the fertilization and hatching rate of all the treatment groups stored at 4°C or in coelomic fluid decreased significantly (P < 0.05). The fertilization rate of the treatment group stored in artificial medium at 16°C did not change obviously in the first 2 h (P > 0.05), but declined dramatically (P < 0.05) after 6 h. In comparison with the control group, no significant (P > 0.05) reduction was shown in hatching rate of the treatment group stored in artificial medium at 16°C for 6 h. The results showed that the ova of Chinese sturgeon can be stored for at least 6 h at 16°C in artificial medium without weakening; this provides a practical application method for the routine hatchery practice of Chinese sturgeon, as well as certain relevant research. Sea cucumbers have been emerged as important models to study organ regeneration and development owing to the capacity to regenerate its organs quickly after evisceration. Evisceration is a special defense mechanism for sea cucumber to eject all of internal organs when they encounter predators or adverse environmental conditions. However, little was known about the dynamics of bacterial community in coelomic fluid after evisceration. In the present study, evisceration was induced by intracelomic injection of 0.35 M KCl, and the significantly alternation of bacterial community in coelomic fluid of sea cucumber Apostichopus japonicus was observed with lower diversity and total bacterial load at 7 dpe (days post evisceration) and 14 dpe. The bacterial community was tended to restore at 28 dpe. In particular, relative abundances of Bacteroidetes and Rubritaleaceae, which involved in degradation of polysaccharides and lipid, increased significantly at 7 dpe (p < 0.05), and returned to the original level at 28 dpe. In addition, the predicted functions of bacterial community indicated that the bacteria associated with metabolism pathways of amino acid, lipid and carbohydrate also increased significantly at 7 dpe. These results suggested that the bacterial community in coelomic fluid of A. japonicus was highly dynamic and could rebuild a stable community structure after evisceration. It was suggested that the enriched metabolic related beneficial bacteria at early stage played a role after evisceration in terms of decomposing polysaccharides and lipid to provide energy.

In order to make clear the protein compositions of Sipunculus nudus and investigate its immune-related proteins, proteomic analysis was performed on body wall and coelomic fluid of Sipunculus nudus. A total of 1659 proteins were identified, and 539 proteins were differentially expressed in the coelomic fluid compared to those in the body wall, of which 415 proteins were up-regulated while 124 proteins were down-regulated. Gene Ontology (GO) analysis showed that the GO terms involved in the two parts of Sipunculus nudus were similar, with metabolic processes, catalytic activity and cell occupying the top categories of biological process, molecular function and cellular component, respectively. KEGG pathway analysis showed that 49 pathways in body wall and 48 in coelomic fluid were mapped respectively, and these pathways were mainly related to cellular processes, environmental information processing, genetic information processing and metabolism. The COG analysis showed that 757 proteins from body wall and 889 from coelomic fluid were classified into 26 COG categories, respectively. Pfam annotation revealed the mainly immune-related proteins contained in Sipunculus nudus , such as insulin-like growth factor binding protein, catalase, basement membrane proteoglycan, titin. Our research provides the first proteomic information of Sipunculus nudus , which contributes to the study of functional proteins in Sipunculus nudus and is of great significance for the application of Sipunculus nudus in functional foods and medicines. Data concerning the toxicity of the transformation products of some pesticides considered emerging contaminants are still incipient. This study aimed to evaluate acute (filter paper contact and avoidance test) and chronic (assays carried out in Red yellow Ultisoil) effects of the transformation products of the fungicide imazalil (IMZ) by heterogeneous photocatalysis (TiO2/UV) in Eisenia andrei. Some endpoints, i.e. biomass variation, disorder in reproduction and avoidance behavior, cytotoxicity (cell density and feasibility), and coelomic fluid (eleocytes and amoebocytes, immune cell) and antioxidant system (catalase (CAT) and glutathione S-transferase (GST)) changes were assessed. The studied degradation times (6, 18, 35 and 90 min) represented 70; 35; 10 and 0% of the initial IMZ concentration (5 mg L−1). No lethality and no significant difference in biomass variations were noted in the acute contact test. Decreased GST and increased CAT activities, as well as cell typing alterations and decreased cell viability and density detected at the 90-min degradation timepoint (PDT-90) indicate direct cytotoxic IMZ transformation product effects. In the avoidance test, no significant earthworm repulsion or attraction was observed. PDT-90 transformation products were responsible for losses in biomass and a reduction in the number of earthworm cocoons and juveniles present in soil. No cytotoxic effects were observed in the long term, although increased CAT and decreased GST activities were observed. Investigations on the relative toxicity of IMZ transformation products are relevant for future discussions regarding the environmental treatment, control and destination of this compound and its derivatives. Synthetic chemicals, such as fertilizers and pesticides, are abundantly used in agriculture to enhance soil fertility and prevent the occurrence of diseases, respectively. Many studies have reported a negative influence of these chemicals on the soil environment. Natural sources from earthworms and their products, as a result of vermicomposting, may be considered better alternatives. The aim of this review was to reveal the source of antifungal efficiency of vermicompost and its derivatives, such as vermiwash, coelomic fluid, skin secretion of earthworms, and metabolites from decomposer bacteria in vermicompost, in order to highlight their application in agriculture. The synergistic activity of bioactive compounds present in coelomic fluid, mucus, skin secretion, and metabolites from associated bacteria (decomposer) assisted crop plants for effective action against various soil pathogenic fungi, such as Rhizoctoniasolani, Alternaria solani, Aspergillus niger, A.flavus, Fusariumoxysporum, and F. graminearum. Thus, these bioactive metabolites can be recommended to suppress plant fungal diseases. Vermicompost and its derivatives should be considered for use in agricultural fields to control harmful soil fungi and increase crop productivity.

Biofluorescence in echinoderms is largely unexplored, and even though the green sea urchin Strongylocentrotus droebachiensis is a well-studied species, the presence and/or function of fluorescence remains very poorly understood. Hyperspectral imaging was conducted on adult sea urchins (N = 380) while fluorospectrometric analysis was conducted on sea urchin coelomic fluid (N = 30). Fluorescence was documented in both the spines and coelomic fluid of S. droebachiensis . Intact spines exhibited a low intensity green emission (∼550-600 nm), while broken spines averaged a high emission peak in the green spectrum (∼580 nm). Sea urchins produce a red exudate with a pronounced emission peak (∼680 nm) with a shoulder peak (∼730 nm). The sampled coelomic fluid exhibited high variability, with a majority exhibiting a low-level green fluorescence while pronounced emission peaks (N = 5) were found in the red spectrum (∼680 nm). The complex fluorescence produced by S. droebachiensis warrants further investigation on its applicability for monitoring welfare of sea urchins in aquaculture facilities., (Creative Commons Attribution license.)

Earthworm coelomic fluid (CF) is known as a rich source of various bioactive compounds with promising anticancer features. However, it has been demonstrated that CF affects functionality of both, cancer and normal cells. This non-selective activity causes a major problem for medical application of CF.In this study, we present the anticancer activity of the active protein-carbohydrate fraction (AF) isolated from thermally treated CF of earthworm Dendrobaena veneta. The in vitro effect of the AF was examined in human colon model including normal human colon epithelium (CCD 841 CoTr) and human colon adenocarcinoma (HT-29 and LS180) cell lines. We investigated the impact of AF on cell viability neutral red and lactate dehydrogenase assays, morphology May-Grünwald-Giemsa staining assay proliferation MTT tetrazolium salt and BrdU incorporation assays as well as cell cycle progression propidium iodide/RNase staining and the activity of human 20S proteasome the hydrolysis of AMC from a Suc-LLVY-AMC peptide substrate. Additionally, the influence of AF on apoptosis was examined in HT-29 cells by Annexin V/PI, Hoechst 33342 staining and active caspase-3 assays.Our investigation demonstrated that AF at the tested concentration range does not affect the viability and morphology of CCD 841 CoTr cells. Simultaneously, AF inhibits human 20S proteasome activity as well as significantly decreases mitochondrial metabolism, disturbs cell cycle and induces apoptosis via activation of procaspase-3 in HT-29 cancer cells. Obtained results demonstrate the antiproliferative and proapoptotic activity of AF that can be useful in developing therapeutic strategies to treat human colon cancer.
