Cognitive dysfunction is a frequent complication of type 2 diabetes mellitus (T2DM), usually accompanied by metabolic disorders. However, the metabolic changes in diabetic cognitive dysfunction (DCD) patients, especially compared to T2DM groups, are not fully understood. Due to the subtle differences in metabolic alterations between DCD groups and T2DM groups, the comprehensive detection of the untargeted metabolic profiles of hippocampus and urine samples of rats was conducted by LC–MS, considering the different ionization modes and polarities of the examined compounds, and feature-based molecular networking (FBMN) was performed to help identify differential metabolites from a comprehensive perspective in this study. In addition, an association analysis of the differential metabolites in hippocampus and urine was conducted by the O2PLS model. Finally, a total of 71 hippocampal tissue differential metabolites and 179 urine differential metabolites were identified. The pathway enrichment results showed that glutamine and glutamate metabolism, alanine, aspartate, and glutamate metabolism, glycerol phospholipid metabolism, TCA cycle, and arginine biosynthesis in the hippocampus of DCD animals were changed. Seven metabolites (AUC > 0.9) in urine appeared as key differential metabolites that might reflect metabolic changes in the target tissue of DCD rats. This study showed that FBMN facilitated the comprehensive identification of differential metabolites in DCD rats. The differential metabolites may suggest an underlying DCD and be considered as potential biomarkers for DCD. Large samples and clinical experiments are needed for the subsequent elucidation of the possible mechanisms leading to these alterations and the verification of potential biomarkers. In order to improve the identification ability of air targets, a comprehensive identification method of air target based on intuitionistic fuzzy set and evidence theory is proposed. This method adopts the idea of multi-attribute decision-making to solve the problem of air target comprehensive recognition. Firstly, the problem of air target comprehensive identification is described by referring to the advanced practice of the U.S. military. Then, the multi-attribute decision-making modeling is carried out for air target comprehensive identification, the attribute weight and expert weight are calculated by combining AIFS cross entropy, conflict coefficient and evidence distance, and the evidence discount method is used to modify and fuse the evidence in aggregation process of attribute information and expert information.Finally, an example is given to verify the feasibility of the proposed method.

Natural gases in China are mainly coal-derived gas, with assistance from oil-typed gas. At present, many genetic identification methods, from hydrocarbon composition and isotope to light hydrocarbon and biomarker indexes, have been formed, but combined methods from non-hydrocarbon gases are lacking. Based on compositions and isotopes geochemical characteristics and the differences of non-hydrocarbon nitrogen gas in coal-derived gas and oil-typed gas, and combining the isotopic geochemical characteristics of non-hydrocarbon helium, the comprehensive identification methods of coal-derived gas and oil-typed gas for hydrocarbon gases according to the associated non-hydrocarbon gases of nitrogen and helium are established and the preliminary applications have been engaged. The main recognitions are as follows:1)Coal-derived gas generally has relatively lower nitrogen abundance, mainly distributed from 0 to 31.2% with main frequency from 0 to 2%. Oil-typed gas, on the other hand, usually has relatively higher nitrogen abundance, mainly distributed from 1.1 to 57.1% with main frequency from 2 to 16%. 2) Coal-derived gas generally has relatively heavier nitrogen isotope values, mainly distributed from -8 to 19.3‰ with main frequency from -8 to 8‰.Oil-typed gas usually has relatively lower nitrogen isotope values, mainly distributed from -10.6 to 4.6‰ with main frequency from -8 to 4‰.3)The geochemical characteristic differences of coal-derived gas and oil-typed gas are mainly due to the fact that the sapropel parent material is relatively rich in nitrogen element and rich in light δ14N, while the humic parent material is relatively poor in nitrogen element and rich in heavy δ15N. The differences on thermal maturity of source rocks, the redox conditions of source rock sedimentary environment, and the salinity of water body are also important effective factors.4)Differences on nitrogen abundances and isotopes in coal-derived gas and oil-typed gas have great significance in genetic identification. The genetic identification chart of R/Ra-δ15N for organic and inorganic nitrogen in natural gas and the comprehensive joint identification charts of R/Ra-δ15N-N2 of nitrogen and helium for coal-derived gas and oil-typed gas have been established, and are of great reference in investigating the origins and sources of natural gas and guiding natural gas exploration in China.

Clp1, a polyribonucleotide 5′-hydroxyl kinase in eukaryotes, is involved in pretRNA splicing and mRNA 3′-end formation. Enzymes similar in amino acid sequence to Clp1, Nol9, and Grc3, are present in some eukaryotes and are involved in prerRNA processing. However, our knowledge of how these Clp1 family proteins evolved and diversified is limited. We conducted a large-scale molecular evolutionary analysis of the Clp1 family proteins in all living organisms for which protein sequences are available in public databases. The phylogenetic distribution and frequencies of the Clp1 family proteins were investigated in complete genomes of Bacteria, Archaea and Eukarya. In total, 3,557 Clp1 family proteins were detected in the three domains of life, Bacteria, Archaea, and Eukarya. Many were from Archaea and Eukarya, but a few were found in restricted, phylogenetically diverse bacterial species. The domain structures of the Clp1 family proteins also differed among the three domains of life. Although the proteins were, on average, 555 amino acids long (range, 196–2,728), 122 large proteins with >1,000 amino acids were detected in eukaryotes. These novel proteins contain the conserved Clp1 polynucleotide kinase domain and various other functional domains. Of these proteins, >80% were from Fungi or Protostomia. The polyribonucleotide kinase activity of Thermus scotoductus Clp1 (Ts -Clp1) was characterized experimentally. Ts -Clp1 preferentially phosphorylates single-stranded RNA oligonucleotides (K m value for ATP, 2.5 µM), or single-stranded DNA at higher enzyme concentrations. Objective: To identify the biomedical frontiers by combining quantitative analysis with qualitative analysis. Methods:This study adopted multi-dimension identification methods, from three aspects as follows: analysis of the key areas of typical national medical science and technology strategic planning, tracking biomedical frontier dynamics and expert review, bibliometrics of biomedical pubilcations and patents. Then, those meeting at least two dimensions were finally defined as biomedical frontiers, based on the identifing results of the three dimensions. Results: A total of 34 biomedical frontiers were identified, including precision medicine, tumor immunity,targeted therapy, medical artificial intelligence, medical robots, etc. Conclusion: The identification of the biomedical frontiers by the multi-dimensional comprehensive methods could provide decision support for the development and layout of biomedical frontiers, and also provide direction and guidance for biomedical innovation. Valine-glutamine motif-containing (VQ) proteins are transcriptional cofactors widely involved in plant growth, development, and response to various stresses. Although the VQ family has been genome-wide identified in some species, but the knowledge regarding duplication-driven functionalization of VQ genes among evolutionarily related species is still lacking. Here, 952 VQ genes have been identified from 16 species, emphasizing seven Triticeae species including the bread wheat. Comprehensive phylogenetic and syntenic analyses allow us to establish the orthologous relationship of VQ genes from rice (Oryza sativa) to bread wheat (Triticum aestivum). The evolutionary analysis revealed that whole-genome duplication (WGD) drives the expansion of OsVQ s, while TaVQ s expansion is associated with a recent burst of gene duplication (RBGD). We also analyzed the motif composition and molecular properties of TaVQ proteins, enriched biological functions, and expression patterns of TaVQ s. We demonstrate that WGD-derived TaVQ s have become divergent in both protein motif composition and expression pattern, while RBGD-derived TaVQ s tend to adopt specific expression patterns, suggesting their functionalization in certain biological processes or in response to specific stresses. Furthermore, some RBGD-derived TaVQ s are found to be associated with salt tolerance. Several of the identified salt-related TaVQ proteins were located in the cytoplasm and nucleus and their salt-responsive expression patterns were validated by qPCR analysis. Yeast-based functional experiments confirmed that TaVQ27 may be a new regulator to salt response and regulation. Overall, this study lays the foundation for further functional validation of VQ family members within the Triticeae species. • With our integrative framework, we genome-wide identified VQ proteins from 16 species. • TaVQ s are expanded by recent duplication events by comparing VQ genes among the Triticeae species. • RBGD-derived TaVQ genes likely become functionally divergent to the OsVQ orthologs. • Homoeolog expression bias in wheat contributes to the regulation and divergence of TaVQ s. • Several TaVQ s linked with salt tolerance may be regulated via the ABRE element. High-resolution mass spectrometry has been a powerful tool for the research of chemical constituents in traditional Chinese medicine (TCM) formulas. However, the chromatographic peaks were difficult to discriminate clearly in data collection or analysis because of the complexity and the greatly different content of the constituents in TCM formula, which increased the difficulty of identification. In this study, a high-performance liquid chromatography coupled with linear ion trap-Orbitrap mass spectrometry based strategy focused on the comprehensive identification of TCM formula constituents was developed. Identification was carried out from a high dose of medicinal materials to equivalent dose of formula. Meanwhile, combined with mass spectrometry data, chromatographic behaviors, reference standards and previous reports, the identification of constituents in Xiang-Sha-Liu-Jun-Zi-Jia-Jian granules was described. 169 compounds were unambiguously or tentatively characterized, mainly including flavonoids, alkaloids, triterpenic acids, triterpene saponins, lactones, sesquiterpenoids and some other compounds. Among them, 11 compounds were unambiguously confirmed by comparing with reference standards. These results demonstrated that the method was effective and reliable for comprehensive identification of constituents of Xiang-Sha-Liu-Jun-Zi-Jia-Jian granules extracts and reveal the material basis of its therapeutic effects. Faults and fractures of multiple scales are frequently induced and generated in compressional structural system. Comprehensive identification of these potential faults and fractures that cannot be distinguished directly from seismic profile of the complex structures is still an unanswered problem. Based on the compressional structural geometry and kinematics theories as well as the structural interpretation from seismic data, a set of techniques is established for the identification of potential faults and fractures in compressional structures. Firstly, three-dimensional (3D) patterns and characteristics of the faults directly interpreted from seismic profile were illustrated by 3D structural model. Then, the unfolding index maps, the principal structural curvature maps, and tectonic stress field maps were obtained from structural restoration. Moreover, potential faults and fractures in compressional structures were quantitatively identified relying on comprehensive analysis of these three maps. Successful identification of the potential faults and fractures in Mishrif limestone formation and in Asmari dolomite formation of Buzurgan anticline in Iraq demonstrates the applicability and reliability of these techniques.
