A number of baby wipe formulations contain 2-phenoxyethanol (PE) as a preservative and cetylpyridinium chloride (CPC) as a surfactant with antimicrobial activity. Previously, we reported the skin absorption of PE in porcine skin and human skin in vitro. In the present work, the permeation of PE from preparations with CPC and without CPC was investigated in human skin in vivo. The studies were conducted using Confocal Raman Spectroscopy (CRS) and tape stripping (TS) methods. The CRS studies showed that the area under the curve (AUC) of PE for the formulation with and without CPC were not significantly different (p > 0.05). The TS data indicated no significant difference in the amounts of PE recovered from tapes 1–6 for the preparation with and without CPC (p > 0.05). When comparing the in vitro and in vivo data, a correlation was observed between the cumulative amount of PE permeated through human skin in vitro at 24 h and the AUC as measured by CRS (r 2 = 0.97). In addition, the cumulative amount of PE permeated through human skin in vitro at 24 h was found to correlate with the amount of PE recovered from tape 1 to 6 in vivo (r 2 = 0.95). Both CRS and TS techniques demonstrated limitations in assessing the distribution of PE and CPC in the skin in vivo , primarily attributed to the Raman signal intensities of compounds under investigation and the variability in the amount of SC collected by TS. Despite the limitations of CRS and TS, the results from the present study add further insights to the in vitro permeation data. Additionally, the findings of the present study encourage the further development and application of CRS for non-invasive evaluation of topical skin formulations in vivo. Alkanediols are widely used as multifunctional ingredients in dermal formulations. In addition to their preservative effect, considering their possible impact on drug penetration is also essential for their use. In the present study, the influence of 2-methyl-2,4-pentanediol, 1,2-pentanediol, 1,2-hexanediol and 1,2-octanediol on the skin penetration of triamcinolone acetonide from four different semisolid formulations was investigated. Furthermore, confocal Raman spectroscopy measurements were performed to examine the influence of the alkanediols on stratum corneum lipid content and order. Alkanediols were found to increase the penetration of triamcinolone acetonide. However, the extent depends strongly on the formulation used. In certain formulations, 1,2-pentanediol showed the highest effect, while in others the penetration-enhancing effect increased with the alkyl chain length of the alkanediol used. None of the tested alkanediols extracted lipids from the stratum corneum nor reduced its thickness. Notwithstanding the above, the longer-chained alkanediols cause the lipids to be converted to a more disordered state, which favors drug penetration. This behavior could not be detected for the shorter-chained alkanediols. Therefore, their penetration-enhancing effect is supposed to be related to an interaction with the hydrophilic regions of the stratum corneum.

Non-ionic emulsifiers are commonly found in existing pharmaceutical and cosmetic formulations and have been widely employed to enhance the penetration and permeation of active ingredients into the skin. With the potential of disrupting skin barrier function and increasing fluidity of stratum corneum (SC) lipids, we herein examined the effects of two kinds of non-ionic emulsifiers on intercellular lipids of skin, using confocal Raman spectroscopy (CRS) with lipid signals on skin CRS spectrum. Non-ionic emulsifiers of polyethylene glycol alkyl ethers and sorbitan fatty acid esters were studied to obtain a deep understanding of the mechanism between non-ionic emulsifiers and SC lipids. Emulsifier solutions and dispersions were prepared and applied onto excised porcine skin. Water and sodium laureth sulfate solution (SLS) served as controls. SC lipid signals were analysed by CRS regarding lipid content, conformation and lateral packing order. Polyethylene glycol (PEG) sorbitan esters revealed no alteration of intercellular lipid properties while PEG-20 ethers appeared to have the most significant effects on reducing lipid content and interrupting lipid organization. In general, the polyoxyethylene chain and alkyl chain of PEG derivative emulsifiers might indicate their ability of interaction with SC components. HLB values remained critical for complete explanation of emulsifier effects on skin lipids. With this study, it is possible to characterize the molecular effects of non-ionic emulsifiers on skin lipids and further deepen the understanding of enhancing substance penetration with reduced skin barrier properties and increased lipid fluidity.

Abstract: The Tekkedağ volcanic complex, which extends as a ridge in the direction of NW–SE, is one of the poorly known volcanic centers and is exposed to the southwest of Kayseri located within the Central Anatolian Volcanic Province (CAVP) of Turkey. The mineralogical composition of Tekkedağ volcanics reveals an assemblage of plagioclase (labradorite, bytownite)+pyroxene (augite, diopside and enstatite)+Fe–Ti oxide (magnetite, rutile)±olivine (forsterite) mineral composition having hypocrystaline porphryric, hypohyaline porphryric, gleomeroporphryric and seriate textures under the microscope. Confocal Raman Spectroscopy (CRS) has been used to define the mineral types. Tekkedağ volcanics have medium K2O contents and are calc-alkaline in character. Geochemically, Tekkedağ volcanics show a narrow range of major element compositions and are classified as augite andesite/basaltic andesite. On the variation diagrams based on MgO versus major and trace elements, they show good positive and negative correlations, indicating fractional crystallization of plagioclase, clinopyroxene and Fe–Ti oxide. Tekkedağ volcanics display enrichment in large-ion lithophile elements (LILEs) relative to high field-strength elements (HFSEs) in chondrite, MORB, E-MORB and lower crust normalized multi-element diagrams. In all normalized multi-element diagrams, the trace element patterns of all samples are similar in shape and exhibit depletions in Ba, Nb, P and Ti as characteristics of subduction-related magmas. Rare earth element (REE) patterns for Tekkedağ volcanics show REE enrichment with respect to chondrite values. They exhibit marked enrichment in light rare earth elements (LREEs) ((La/Sm) N =4.13–4.62) relative to heavy rare earth elements (HREEs) ((Sm/Lu) N =1.34–1.92). Furthermore, all samples have negative Eu anomalies ((Eu/Eu*) N =0.77–0.90), indicating the significant role of plagioclase in the fractional crystallization. Elemental ratios such as K/P (15.46–21.69), La/Nb (2.01–4.26), Rb/Nb (8.74–10.59), Ba/Nb (38.54–75.77), Nb/Ta (1.16–2.14), Ce/P (2.13–4.32) and Th/U (1.77–3.97) propose that the magma was subjected to conceivable crustal contamination during the evolution of these Tekkedağ volcanics. This statement is supported by the AFC modeling based on the trace elements. As a result, despite the lack of isotopic data, the petrographic and geochemical results suggest a significant role of plagioclase, clinopyroxene and Fe–Ti oxide fractionation during the evolution of the Volcanic Arc Basalts (VAB) nature of the Tekkedağ volcanics. Furthermore, these results reveal that the volcanics of Tekkedağ were produced from a parental magma derived from an enriched source of mixed subduction and/or crustal products.
