1. Utopian Visions, dystopic realities. Multidisciplinary Reflections on Contemporary Challenges

Abstract

The crisis of globalisation, the rise of autocratic competitors to the liberal-democratic model, the return of symmetric warfare on European soil, the problem of energy resources and global disparities in accessing them, the consequences of the Anthropocene on the lives of millions of people around the World, are just some of the problematic issues that new generations of scholars and academics are called to answer in the next decades. This impressive conjuncture of critical phenomena reveals the exhaustion of an entire mindset, which belongs to the Twentieth Century and, in particular, to the illusion that the History was over, together with some of the most harmful Nineteenth Century’s legacies: industrial pollution, economic and social inequalities, radical nationalism and imperialism. In other words, it looks like the Utopian visions of the past – and especially the ones related with the Western liberal thought – have come to terms with their limitis which, in their turns, have produced a series of dystopic effects in the real World. Given this general picture, it is not so surprising to note a revival of dystopian narratives in literature, tv series, film-making and a variety of other cultural products. Placed within a broader interdisciplinary research project focused on the penetration of dystopian narratives in the Twenty-First century’s collective imagery (as well as the analysis of the political role played by dystopias in present times) this collective work brings together the thoughts of seven young doctoral students, who read some of the most crucial issues of our times through the lenses of utopia and dystopia, as well as by different disciplinary perspectives. From the challenges related to the natural resources management and the energy transition to the rise of alterative political models to the Western liberal-democracy, each essay accounts for the long term impact of utopian theories and ideas over time and their eventual dystopic effects

Abstract

The paper records the results of an extensive investigation conducted in Italy regarding the setting up of testing procedures for the assessment of the failure mode behaviour of polymeric housed distribution type surge attesters. Different sample preparation methods and short circuit test procedures are compared and discussed. The evaluation of the performance takes into consideration all the major hazard issues, such as the ejection of fragments, the detachment of the connections and fire ignition. Several aspects to be taken into account for the standardization of the failure mode test are also addressed.

Abstract

A national decommissioning programme of offshore hydrocarbon platforms, regardless of the total or partial removal hypothesis for further alternative uses, should be based on sustainability and safety principles. In order to carry out this sensitive and complex activity, it is necessary that the competent authorities and operators have, in addition to a relevant regulatory framework, criteria and methodologies that make the assessment of technological options from a safety, social, economic and environmental viewpoints possible. This paper presents a first overview of the study performed on decision support system methodologies with focus on Multi-Criteria Analysis (MCA), carried out within the framework of the "Safe and Sustainable Decommissioning (SSD)" project whose goal is to develop a decommissioning program based on the definition of indicators and of objective criteria for removing or converting offshore hydrocarbon platforms

Dopo un lungo e tortuoso iter la legge 27 maggio 2015, n. 69, contenente disposizioni in materia di delitti contro la pubblica amministrazione, di associazione di tipo mafioso e falso in bilancio, è stata pubblicata nella Gazzetta Ufficiale n. 124 del 30 maggio 2015. Il nuovo testo si compone di dodici articoli divisi in due Capi: il primo, reca disposizioni in materia di delitti contro la pubblica amministrazione e di associazione di stampo mafioso, nonché ulteriori modifiche al codice di procedura penale, alle relative norme di attuazione e alla c.d. Legge Severino (n. 190/2012); il secondo, interviene sulla disciplina del c.d. falso in bilancio. Abstract

The causes of Late Pleistocene megafaunal extinctions (60,000 to 11,650 years ago, hereafter 60 to 11.65 ka) remain contentious, with major phases coinciding with both human arrival and climate change around the world. The Americas provide a unique opportunity to disentangle these factors as human colonization took place over a narrow time frame (~15 to 14.6 ka) but during contrasting temperature trends across each continent. Unfortunately, limited data sets in South America have so far precluded detailed comparison. We analyze genetic and radiocarbon data from 89 and 71 Patagonian megafaunal bones, respectively, more than doubling the high-quality Pleistocene megafaunal radiocarbon data sets from the region. We identify a narrow megafaunal extinction phase 12,280 ± 110 years ago, some 1 to 3 thousand years after initial human presence in the area. Although humans arrived immediately prior to a cold phase, the Antarctic Cold Reversal stadial, megafaunal extinctions did not occur until the stadial finished and the subsequent warming phase commenced some 1 to 3 thousand years later. The increased resolution provided by the Patagonian material reveals that the sequence of climate and extinction events in North and South America were temporally inverted, but in both cases, megafaunal extinctions did not occur until human presence and climate warming coincided. Overall, metapopulation processes involving subpopulation connectivity on a continental scale appear to have been critical for megafaunal species survival of both climate change and human impacts.

Abstract

A new system aimed at performing partial discharge measurements and condition assessment on electrical apparatus is presented. It is able to separate the conventional phase resolved partial discharge pattern into a series of sub-patterns, each of them containing partial discharge data generated by a single kind of defect, which makes easier identification of the source generating partial discharges. Application of the system to partial discharge data recorded on an hydrogenerator is reported to support its effectiveness.

Abstract

A synthetic cDNA coding for human pancreatic RNase, equipped with a secretion signal sequence, was cloned and stably expressed in Chinese hamster ovary cells. The recombinant RNase, secreted into the culture medium, was purified and characterized. It was found to be indistinguishable, by structural and catalytic parameters, from the enzyme isolated from human pancreas. Furthermore, the glycosylated forms were separated from the non-glycosylated form. Up until now, human RNases have been isolated only in small amounts from autoptic specimens. This has hindered the exploitation of a human RNase for the constrnction of immunotolerated immunotoxins. On the other hand, the availability of an effective system for the expression of a human RNase may render feasible the transfer, by protein engineering, of the interesting pharmacological actions of non-human RNase to an immunotolerated, human RNase.

Abstract

Selective deamidation of proteins and peptides is a reaction of great interest, whether it has physiological significance as in protein aging, or occurs as a disturbing event in the preparation of natural or recombinant proteins. Deamidation of bovine pancreatic ribonuclease A, RNase A, a classical model protein, has been reported to occur only after denaturation of the protein, or under harsh conditions. In this paper convenient procedures are described for selective deamidation of Asna7 in native RNase A under mild conditions. Furthermore, for the first time, both products of deamidation were isolated: the aspartyl and the isoaspartyl containing protein derivatives. Replacement of Asns7 with either residue lowers the catalytic activity of the enzyme, on RNA and on model substrates, except when a dinucleotide with a purine on the 5’ side is the substrate. In the latter case an intriguing increase in the specificity constant is observed. The Aspe7 derivative was found to refold, after full denaturation and reduction, at the same rate as the fully amidated protein, whereas the iso-Asp“ derivative refolded at half that rate. It is hypothesized that this effect is due to a delayed formation of disulfide 65-72 for the presence of the abnormal isopeptide bond between residues 67 and 68.

Abstract

The state of health of an electrical cable may be difficult to know, without destructive or very expensive tests. To overcome this, partial discharge (PD) measurements have been proposed as a relatively economic and simple-to-apply experimental technique for retrieving information on the state of health of an electrical cable. The retrieval is based on a relationship between PD measurements and the health of the cable. Given the difficulties in capturing such relationship by analytical models, empirical modeling techniques based on experimental data have been propounded. In this view, a set of PD measurements have been collected by Enea Ricerca sul Sistema Elettrico-ERSE during past campaigns, for building a diagnostic system of electrical cable health state. These experimental data may contain contradictory information which remarkably reduces the performance of the state classifier, if not a priori identified and possibly corrected. In the present paper, a novel technique based on the Adaboost algorithm is proposed for identifying contradictory PD patterns within an a priori analysis aimed at improving the diagnostic performance. Adaboost is a bootstrap-inspired, ensemble-based algorithm which has been effectively used for addressing classification problems. [ABSTRACT FROM AUTHOR]

Abstract

Bovine seminal RNAase (BS-RNAase) is a singular ribonuclease of the pancreatic RNAase superfamily, with unusual structural, catalytic, and biological properties [1]. It is the only dimeric protein in the superfamily, with intersubunit disulfide bridges [2]. In native BS-RNAase the majority of the subunits interchange their N-terminal segments, while in the remaining dimers each N-terminal segment folds onto its respective subunit [3]. It is homotropically regulated in the second, rate determininig step of the catalytic reaction, with negative and positive cooperativity [4]. It is selectively deamidated; this produces a characteristic isoenzymic composition, made up of α2, αs and s2 isoenzymes, produced by the progressive transformation of β subunits into α subunits through selective deamidation at Asn67 [5]. It is a RISBASE, i.e. a Ribonuclease with Special Biological Actions, including its antitumor, anti-spermatogenic and immunosuppressive actions [1]. All these properties are lost when the enzyme is monomerized, although the monomeric derivative is fully and catalytically active [1,4]. A definition at the molecular level of the structural determinants underlying these diverse biological properties may help to shed light on the molecular mechanisms of such activities. To reach this goal, protein engineering can be a very effective approach, as it should allow to verify the effects of appropriate mutations on the catalytic and the “special” biological activities of the enzyme.
