No. of bitstreams: 0 Previous issue date: 2021-07-01 Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq) Lithobates catesbeianus tadpoles were exposed for 96 h to water from two sites of the Sorocaba River (summer and winter), Ibiúna (PI) and Itupararanga reservoir (PIR) that contained metals. In the liver, in PI, the glutathione peroxidase (GPx) decreased, and the glutathione S-transferase (GST) and carbonyl proteins (PCO) increased. In PIR, the glutathione reduced (GSH) increased, while there was a decrease in catalase (CAT), GPx, GST, PCO, and superoxide dismutase (SOD). In winter, GPx and GST increased in both points. Regarding the kidneys, lipoperoxidation (LPO) levels and GST decreased, while GSH increased in the summer. In the winter, LPO increased in PI. In the muscle, in the summer, there was an increase in GSH and GST and change in PCO. In the winter, the levels of PCO increased and CAT decreased in PIR. The area and volume of the hepatocyte and nucleus area increased in the summer and decreased in the winter. Hepatic melanin decreased in the summer after exposure to PIR water. There were the systemic effects of Sorocaba River water exposure at different times of the year with alterations in biomarkers at different levels, in which kidney shows highest Integrated Response of Biomarkers (IBR) value followed by liver and muscle. Biochemical biomarkers were more sensitive than morphological ones. The more sensitive biochemical markers were MT, PCO, GST and LPO. These effects confirm the hypothesis of metabolic alteration in bullfrog tadpoles by the Sorocaba River water. Acute exposure to water from the Sorocaba River induced oxidative stress, mainly in the liver and kidneys, and caused liver damage in bullfrog tadpoles even in low concentrations of metals. Abstract

The liver is a well‐defined organ regarding its functions; however, it may present morphological variations among species. Since anurans may be exposed to toxic substances in their environment, which can disrupt liver's morphology and function, descriptive studies of organs are important to be carried out under natural condition. Thus, this study aimed to describe morphological aspects of liver in three Neotropical anuran species commonly used as experimental models: Physalaemus cuvieri (Leptodactylidae), Leptodactylus fuscus (Leptodactylidae) and Rhinella diptycha (Bufonidae). Anatomical, histological and ultrastructural analyses of six male samples of each species were carried out. A restricted analysis of the three species showed that livers' anatomy and ultrastructure are quite similar—they present three lobes, rounded nucleus polyhedral hepatocytes and some glycogen contents; however, histologically, there was a difference in the structural arrangements. In R. diptycha the parenchyma was arranged in a single cord, whereas in P. cuvieri and L. fuscus, there were double ones. This parenchyma structuring is highly variable considering Amphibia class. Knowledge about hepatic morphology and ultrastructure in amphibians is a key measure to detect changes in experimental studies and to infer about possible physiological and / or metabolic changes, when these animals are exposed to adverse conditions. 27. Hematological parameters of a Neotropical wild frog population, with a phylogenetic perspective on blood cell composition in Anura

Abstract

Hematological parameters can provide key information to an animal health status. However, this information is usually hard to obtain. Here, we described the hematological parameters of Leptodactylus podicipinusin the Brazilian Pantanal. We measured red blood cell morphometrics, erythrogram, and leukogram. We also tested for phylogenetic signal in the erythrogram and leukogram of 48 frog species from 15 families, testing if body size explains their variation. Lymphocytes were the most abundant leukocytes (>60%) in L. podicipinus, followed by neutrophils (∼10%). Given that L. podicipinusis an abundant and widely distributed species in central Brazil, knowing its hematological pattern can help establish a baseline and improve its use as a bioindicator of environmental degradation. Mean corpuscular hemoglobin and value contributed more to the phylomorphospace of erythrogram, in which Leptodactylusspp. and Hypsiboas ranicepshad lower values of these variables, whereas Bufotes viridisand Hyla arboreahad high values. The phylogenetic signal was spread throughout the dimensions of the leukogram phylomorphospace. The variables that most contributed to it were total leukocytes counts, lymphocytes, and neutrophils. We also found a moderate phylogenetic signal for both the erythrogram and leukogram. Accordingly, body size accounted for a low proportion of variation in both the leukogram (4.7%) and erythrogram (0.57%). By applying phylogenetic comparative methods to hematological parameters, our results add a new perspective on the evolution of blood cell physiology in frogs.

Abstract

Amphibians have pigmented cells in organs beyond just skin. Their functions involve free radical neutralisation, early innate response, and a relationship with environmental temperature and UV light. In gonads, pigment containing-cells seem to be restricted mainly to the testes and related to sperm production. However, we report for the first time ovarian melanisation in Pseudis minuta and its ontogenetic changes in larval and postmetamorphic stages. Melanin containing-cells on the ovarian surface initially appear at early premetamorphic stages whereas in the cortex they occur later. In consecutive stages, melanin containing-cells were more evident among oocytes but without a clear pattern, being located randomly within the germinal epithelium or in the stroma. Although their function is unclear, a relationship with the fast acquisition of sexual maturity must be further explored. [ABSTRACT FROM AUTHOR]

Abstract

The greatest diversity of anurans in the world is in Brazil and one of the major challenges is to reconcile the accelerated economic development with strategies that aim to maintain this diversity in forest fragments, often representing ESUs of some biomes. This study aimed to obtain data that will support conservation projects through the pioneering use of ISSR analysis in Neotropical anurans, estimating the intra- and interpopulation genetic diversity of four populations of P. cuvieri (Paraná and São Paulo regions). Of the 65 loci scored 58 were polymorphic, with 0.797 intrapopulation variation and 0.203 interpopulation variation. The index of interpopulation genetic differentiation (Fst) proved to be high among the population of Marmeleiro-PR and the three populations of SP (Fst > 0.288); genetic dissimilarity was related to the geographical distance. The ISSR proved to be efficient and useful molecular markers in comparison with other markers most widely used for preliminary diagnosis of genetic diversity in populations of amphibians, and could be applied as a tool for future conservation projects, since they could identify potential ESUs and influence decisions on the preservation of fragments.
