Abstract

Introduction. Sphingolipids are a group of lipid molecules, the focus on which has been gradually increasing during recent years. This review presents sphingolipids, as valuable compounds with a high potential for industry. Literature. Structures of sphingolipids are described and their natural sources are presented. Different methods for extraction, purification and structural characterization of sphingolipids are evaluated. Activity of sphingolipids towards various microorganisms is discussed and methods for chemical modifications of natural sphingolipids to obtain novel properties are depicted. Finally, applications for implementing sphingolipid molecules in food, cosmetic, pharmaceutical or medical industry are proposed. Conclusions. Sphingolipids are molecules of high impact and their importance will inevitably increase in the future.

Abstract

Introduction. Over the centuries, bio-sourced compounds isolated from plants, insects and microorganisms have been a potent source of drugs for the treatment of human diseases. Literature. Bio-sourced extracts offer a wide diversity of compounds with a large number of potentially beneficial effects in humans. Serendipity has frequently played a key role in the discovery of new medicines. The canonical discovery of penicillin required both chance and a prepared mind to understand and exploit its potential for the treatment of human infections. Nowadays, most anti-cancer drugs currently in clinical use were at least partly discovered by a "fortunate happenstance". Conclusions. In this review, we recapitulate the story of one of these compounds, 2-propylpentanoic acid, derived from the Valeriana officinalis flowering plant and its path to validation as a cancer treatment.

Abstract

Description of the subject. Current trends suggest an increasing future demand for conventional meats, which indicates a strong need to shift this dependency to other alternative protein sources such as insects. Literature. From a nutritional point of view, of all the insects consumed globally, grasshoppers are particularly important as a human food. Data from the literature regarding the nutrient composition, amino acid profile, fatty acid profile, mineral composition and vitamin content of grasshoppers as reviewed in this paper, suggest that a number of grasshopper species are a good source of nutrients. It also highlights some of the health related aspects that might arise from the consumption of grasshoppers, mostly linked to agricultural practices and the allergic response of sensitive individuals. The paper also summarizes some religious, social and economic factors that are associated with grasshopper consumption. Conclusions. The success of introducing grasshoppers as a novel food in western countries depends on changes in consumer attitudes. It would be interesting to develop food products derived from grasshoppers in a form acceptable to consumers. Furthermore, it is important to explore the food potential of some grasshopper species native to western countries and to develop their rearing methodologies to enhance availability.

Abstract

Isolation of an amylolytic chrysophyte, Poterioochromonas sp., from the digestive tract of the termite Reticulitermes santonensis. The aim of this work was the isolation and cultivation of amylolytic protists living in the digestive tract of the termite Reticulitermes santonensis (Feytaud). A chrysophyte identified as Poterioochromonas sp. was isolated in a special medium containing rice grains as a source of carbon and nitrogen. Then, the protist was grown in a medium containing starch as a carbon source, tryptone, and a phosphate buffer at different pH values (5, 6 and 7). Yeast extract was added or not. Ciprofloxacin was used to avoid the bacterial development. Other antibiotics were also tested but showed an inhibitive effect on the growth of Poterioochromonas sp. Yeast extract allowed reaching 1.9 (pH 5), 2.3 (pH 6) and 2.2 (pH 7) times higher final cell concentrations, and 2.8 (pH 5), 2.8 (pH 6) and 2.2 (pH 7) times higher biomass yields. The starch concentration did not decrease in the medium until 3 and 4 days of culture, with and without yeast extract, respectively. Eight days of culture were necessary for hydrolyzing the starch completely, with and without yeast extract. Maltose and maltotriose were detected in the culture media and were hydrolyzed progressively. Maximal maltose concentrations were 0.68, 0.66 and 0.51 g·l-1 in the medium containing yeast extract. Maltotriose concentrations were only 0.17, 0.14 and 0.12 g·l-1. Other glucose oligomers were also detected but in lower quantities. It was determined that the protist developed a weak amylase activity, particularly at a weakly acidic pH (5-6). Such a pH also allowed a better growth of the protist. A maximal amylase activity of 112 nkat·l-1 was measured with yeast extract at pH 5. No other enzymatic activity (protease, cellulase or xylanase) was detected except amylase. The degradation products of starch, which were obtained by enzymatic hydrolysis, allow the identification of α-amylase, amyloglucosidase and possibly β-amylase activities.

Abstract

Introduction: The human liver is a complex organ composed of highly specialized parenchymal and non-parenchymal cells assembled in collaborative sinusoidal units within an extracellular matrix (ECM) network. Recent advances in ‘omics’ technologies allow the study of the whole transcriptome at single cell resolution, as well as the ECM composition of a whole organ. In this study, we aimed at providing a high-resolution transcriptomic and ECM proteomic map of the adult human liver. Methods: 80,000 liver cells from 2 human livers (Hepatic Biobank, Cliniques universitaires Saint-Luc) were loaded in a 10X Genomics instrument to generate 8 independent libraries, sequenced with the Illumina technology for an average of 65,000 reads per cells. Human liver fragments from 4 donors were decellularized by a combination of thermic and osmotic shocks, enzymatic digestion and use of detergents. Peptidic and proteomic composition of the resulting ECM was determined by LC/MS. Results: Distinct parenchymal and non-parenchymal cell populations and subpopulations were identified. Based on the expression of known metabolic markers, hepatocytes have been efficiently zonated along the portal-central axis, whereas newly identified, non-metabolic markers are currently evaluated for the zonation of non-parenchymal cells. Proteomic and peptidomic analysis reveals abundance of elastin and type I, II and VI collagens. Conclusions: Our study provides a unique dataset that paves the way for a thorough analysis of the microenvironment of human liver cells, and could prove to be of interest to liver stem cells researchers.

Abstract

Background and Aims: The study of human primary liver cell biology is largely hampered by the lack of adequate in vitro culture systems that allow the maintenance of the cells’ viability and physiologic phenotype for prolonged periods of time. In this context, a growing interest for liver decellularization and for the use of liver extracellular matrix (ECM) in cell culture and tissue engineering applications emerged over the past five years. Whereas several publications describe detailed procedures for animal liver decellularization and ECM-based hydrogel production, few is known about the composition and properties of isolated human liver ECM. In this study, we analyzed the composition of human liver ECM and derived hydrogels, and we further addressed the question of the potential benefits of a liver ECM protein-coated surface for the culture of primary human liver cells. Method: A combination of thermic and osmotic shock, enzymatic digestion and detergents has been used to decellularize human liver fragments unsuitable for transplantation. Liver ECM was partially digested with pepsin for coating purposes and characterized by histology and mass spectrometry. Primary human liver cells, including hepatocytes, hepatic stellate cells (HSC) and endothelial cells (EC) were seeded on liver-coated culture dishes. Adhesion (2h), viability (24h), proliferation (7 days) and phenotypic properties were evaluated and compared to a surface of reference (CellBIND or rat tail collagen coating). Results: Our protocol led to a residual amount dsDNA below decellularization standards. Coating derived from liver ECM showed no toxicity for primary human liver cells. Mass spectrometry revealed the abundance of type I and type III fibrillar collagen components (COL1A1, COL1A2, COL3A1) as well as elastin in liver ECM hydrogels used for coating. Preliminary results further suggest the benefits of human liver ECM on human primary liver cell properties in 2D culture as compared to surfaces of reference. Conclusion: Our study provides a first detailed characterization of human liver ECM and derived hydrogels, easily and reproducibly isolated from small human liver fragments. Our results further open the way for the use of human liver ECM as a coating for the culture of primary human liver cells in 2D. In a near future, 3D applications will be developed.
