Cell-non-autonomous signals dictate the functional state of cellular quality control systems, remodeling the ability of cells to cope with stress and maintain protein homeostasis (proteostasis). One highly regulated cell-non-autonomous switch controls proteostatic capacity in Caenorhabditis elegans adulthood. Signals from the reproductive system down-regulate cyto-protective pathways, unless countered by signals reporting on germline proliferation disruption. Here, we utilized dihomo-γ-linolenic acid (DGLA) that depletes the C. elegans germline to ask when cell-non-autonomous signals from the reproductive system determine somatic proteostasis and whether such regulation is reversible. We found that diet supplementation of DGLA resulted in the maintenance of somatic proteostasis after the onset of reproduction. DGLA-dependent proteostasis remodeling was only effective if animals were exposed to DGLA during larval development. A short exposure of 16 h during the second to fourth larval stages was sufficient and required to maintain somatic proteostasis in adulthood but not to extend lifespan. The reproductive system was required for DGLA-dependent remodeling of proteostasis in adulthood, likely via DGLA-dependent disruption of germline stem cells. However, arachidonic acid (AA), a somatic regulator of this pathway that does not require the reproductive system, presented similar regulatory timing. Finally, we showed that DGLA- and AA-supplementation led to activation of the gonadal longevity pathway but presented differential regulatory timing. Proteostasis and stress response regulators, including hsf-1 and daf-16, were only activated if exposed to DGLA and AA during development, while other gonadal longevity factors did not show this regulatory timing. We propose that C. elegans determines its proteostatic fate during development and is committed to either reproduction, and thus present restricted proteostasis, or survival, and thus present robust proteostasis. Given the critical role of proteostatic networks in the onset and progression of many aging-related diseases, such a choice could impact susceptibility to protein misfolding diseases later in life.

Summary: Background & aims: The study examined the value of n-3 LC-PUFA-enriched yogurt as means of improving cardiovascular health. Design: Fifty three mildly hypertriacylglycerolemic subjects (TAG ≥ 1.7 mmol/L) participated in a randomized, placebo-controlled, double-blind, parallel designed study. The subjects consumed 1) control yoghurt; 2) yoghurt enriched with 0.8 g n-3 LC-PUFA/d; or 3) yoghurt enriched with 3 g n-3 LC-PUFA/d for a period of 10 wks. Blood samples were taken at the beginning and the end of the study period. Results: Following daily intake of 3 g n-3 LC-PUFA for 10 weeks, n-3 LC-PUFA levels increased significantly in plasma and red blood cells (RBC) with concomitant increase in the EPA-derived mediators (PGE3, 12-, 15-, 18-HEPE) in plasma whilst cardiovascular risk factors such as HDL, TAG, AA/EPA ratio, and n-3 index were improved (P < 0.05); the decrease of TAG and increase in HDL were associated with the CD36 genotype. Conclusion: The observed increase of n-3 LC-PUFA in RBC and plasma lipids due to intake of n-3 LC-PUFA enriched yoghurt resulted in a reduction of cardiovascular risk factors and inflammatory mediators showing that daily consumption of n-3 PUFA enriched yoghurt can be an effective way of supplementing the daily diet and improving cardiovascular health. Abstract: Introduction: Past research has reported that single nucleotide polymorphisms (SNPs) in fatty acid desaturase 1 and 2 (FADS1/2) can influence plasma fatty acid (FA) profiles. Changes in FA profiles are known to influence inflammatory processes; therefore both FA and SNPs in FADS1/2 may affect inflammation. The goals of this study were to (i) examine the relationships between individual n−6 FA and estimates of FA desaturation with circulating high sensitivity C-reactive protein (hsCRP) levels, and (ii) determine whether SNPs in FADS1/2 are associated with changes in hsCRP. Methods: FA and hsCRP were measured in fasted plasma samples from 878 healthy young adults (20–29yrs). Circulating levels of plasma linoleic (LA), γ-linolenic (GLA), dihomo-γ-linolenic (DGLA) and arachidonic (AA) acids were measured by gas chromatography and used to calculate desaturase indices for FADS1/2. Nineteen SNPs in FADS1/2 were genotyped in all subjects and six (rs174579, rs174593, rs174626, rs526126, rs968567 and rs17831757) were further analyzed. Results: Significant inverse associations were found between LA and hsCRP (p=8.55×10−9) and the FADS1 desaturase index and hsCRP (p=4.41×10−6). A significant positive association was found between DGLA and hsCRP (p=9.10×10−11). Several SNPs were associated with circulating levels of individual FA and desaturase indices, with minor allele carriers having lower AA levels and reduced desaturase indices. A single SNP in FADS2 (rs526126) was weakly associated with hsCRP (p=0.05). Conclusions: This study highlights the relationships between FA and hsCRP, and confirms that FA are strongly influenced by SNPs in FADS1/2. Furthermore, we found weak evidence that SNPs in FADS1/2 may influence hsCRP levels in young adults. [Copyright &y& Elsevier]

Abstract: Objective: We examined the association of cardiac hypertrophy or fibrosis with the phospholipid fatty acid (FA) composition of heart and kidney in hypertensive rats. Design and methods: Eight-week-old spontaneously hypertensive rats (SHRs) (n=8) and Wistar Kyoto rats (WKYs, n=8) as a normotensive control, were fed ad libitum for 6weeks with regular AIN-76 diet. Phospholipid FA compositions in the left ventricle and kidney were measured and histological analyses were performed. Results: Compared with WKYs, SHRs had lower proportions of γ-linolenic acid, α-linolenic acid, eicosadienoic acid, eicosatrienoic acid, dihomo-γ-linoleic acid, docosadienoic acid and nervonic acid in heart, and stearic acid (SA), γ-linolenic acid, and eicosapentaenoic acid (EPA) in kidney. After adjusting for food intake, SHRs still maintained higher proportions of SA, and total saturated FAs in the heart and a lower proportion of eicosapentaenoic acid in the kidney. Additionally, compared with WKYs, SHRs showed larger cardiomyocyte diameters in the left ventricles, indicating cardiac hypertrophy and interstitial fibrosis. Cardiomyocyte diameters also positively correlated with cardiac SA (r=0.550, p<0.05) and negatively with kidney EPA (r=−0.575, p<0.05). Conclusion: Tissue FA compositions were associated with cardiac hypertrophy in a hypertensive setting, implicating the pathogenic role of tissue FAs in hypertension and related complications. Abstract: We investigated effects of irradiance on the relationships between chlorophyll fluorescence transients (OJIP), carotenoid-to-chlorophyll ratio, and fatty acids in a nitrogen-deprived Parietochloris incisa (Chlorophyta, Trebouxiophyceae) Δ5 desaturase mutant accumulating valuable LC-PUFA dihomo-γ-linolenic acid (DGLA). High light (270μE·m−2·s−1 PAR) and nitrogen starvation brought about a decrease in maximum quantum yield of photosystem II ( ) and electron transport ( ) but enhanced the quantum yield of thermal dissipation ( ) and induced non-photochemical quenching (NPQ) in an irradiance-dependent manner. Under high irradiance a decline in the rate of total fatty acid accumulation and DGLA percentage in comparison with the cultures grown under 130μE·m−2·s−1 PAR was recorded. Increasing irradiance from 130 to 270μE·m−2·s−1 enhanced total fatty acid accumulation only within the first week of nitrogen starvation and negatively affected DGLA production. Regardless of irradiance, , , and exhibited tight (r 2 =0.8–0.9) relationships with the stress-induced changes of total fatty acid and DGLA content and the carotenoid-to-chlorophyll ratio. The applicability and limitations of OJIP and its derived parameters for on-line monitoring of physiological condition and accumulation of value-added products in microalgal cultures grown in photobioreactors are discussed. Abstract: Dihomo-γ-linolenic acid (DGLA) is one of the essential fatty acids, and has anti-inflammatory and anti-allergic effects. To assess the toxicity of a novel DGLA oil produced by the fungus Mortierella alpina, we examined it in the Ames test and in acute and subchronic oral toxicity tests in rats. In the Ames test, no mutagenicity was found up to 5000μg/plate. The acute toxicity test revealed no toxicity related to DGLA oil at 10g/kg. In the subchronic toxicity test, DGLA oil (500, 1000, and 2000mg/kg) was orally administered. Water and soybean oil (2000mg/kg) were used for the no-oil control and soybean oil control groups, respectively. There was no death in either sex. Because of administration of large amounts of oil, food consumption was low in the soybean oil control and the three test groups, which appeared to mildly decrease urinary excretion of Na, K, and Cl, as well as total serum protein, albumin, and blood urea nitrogen levels. There were no toxicological changes in body weight, food consumption, ophthalmological examination, urinalysis, hematological examination, blood biochemical examination, necropsy, organ weight, or histopathological examination. These findings show that the no-observed-adverse-effect level of the DGLA oil was 2000mg/kg. [Copyright &y& Elsevier]

Orthogonal experiments were employed to optimize the correlated parameters of reduction, sulfonation, substitution and hydrolysis. These reactions were used to convert γ-linolenic acids into dihomo-γ-linolenic acids (DGLA). For the reduction, the best reaction conditions were at 35 °C for 4.5 h with LiAlH4 and γ-linolenic acid (in the ratio of 40 g:100 g); for the sulfonation, reaction at 29 °C for 3.5 h with 150 g γ-linolenic alcohol and 65 mL mesyl chloride, then the water phase being extracted with dichloromethane (3 × 100 mL); for the substitution, the reaction at 80 °C for 2.5 h with metallic sodium and sulfonate (at a ratio of 8 g:100 g); and for the hydrolysis, reaction at 80 °C for 2.5 h with NaOH and dihomo dioate (at a ratio of 50 g:100 g). The four reactions gave yields that exceeded 90% for each step. Finally, crystallization and decarboxylation provided DGLA in an overall yield of 60% and >95% purity. [ABSTRACT FROM AUTHOR]

The article compares the dietary effects of dihomo-γ-linolenic acid (DGLA) contained in the DGLA oil produced by a fungus with GLA on the fatty acid composition. The DGLA concentrations in the liver, serum, and brain of the DGLA group were higher than those of the GLA oil group. The results suggest that the dietary effect of DGLA would be more dominant than GLA.

Abstract Considerable arguments remain regarding the diverse biological activities of polyunsaturated fatty acids (PUFA). One of the most interesting but controversial dietary approaches focused on the diverse function of dihomo-dietary γ-linolenic acid (DGLA) in anti-inflammation and anti-proliferation diseases, especially for cancers. This strategy is based on the ability of DGLA to interfere in cellular lipid metabolism and eicosanoid (cyclooxygenase and lipoxygenase) biosynthesis. Subsequently, DGLA can be further converted by inflammatory cells to 15-(S)-hydroxy-8,11,13-eicosatrienoic acid and prostaglandin E1 (PGE1). This is noteworthy because these compounds possess both anti-inflammatory and anti-proliferative properties. PGE1 could also induce growth inhibition and differentiation of cancer cells. Although the mechanism of DGLA has not yet been elucidated, it is significant to anticipate the antitumor potential benefits from DGLA.
