Background: Lactic whey, a significant agro-industrial byproduct, poses environmental risks due to its chemical composition. Despite various valorization efforts, effective utilization remains a challenge. This study explores the potential of Neochloris oleoabundans, a microalgae known for its metabolic versatility and resilience to adverse conditions, to produce exopolysaccharides (EPS) using lactic whey as a substrate. We compared EPS production from lactose, the primary sugar in whey, with whole lactic whey. Characterization of the EPS was performed using Fourier transform infrared spectroscopy (FT-IR) and gas chromatography–mass spectrometry (GC–MS), while morphological analysis was conducted via scanning electron microscopy (SEM). This research aims to assess the feasibility of converting lactic whey into valuable EPS, providing a sustainable approach to managing this agro-industrial waste. Results: Lactic whey has produced the highest EPS and the FT-IR spectra revealed structural variations in the monomers which compose these polymers. Galactose and glucose were shown to be the primary monomers, according to GC–MS EPS analysis. SEM revealed a homogenous matrix and N. oleoabundans's bioflocculant characteristics. Exopolysaccharide (EPS)-enriched extracts derived from microalgae exhibit a wide range of bioactive properties, including antibacterial and antifungal properties. However, these properties vary depending on the microalgae species, the antimicrobial assay used, and selected targeted microorganisms. This study offers to investigate the antimicrobial properties of exopolysaccharide-enriched extracts obtained from five microalgae species scarcely studied in this context. The targeted microorganisms selected for this study included Gram-positive (Bacillus subtilis) and Gram-negative bacteria (Pseudomonas aeruginosa), fungi (Cladosporium cladosporioides), and microalgae (Chlorella vulgaris). Well-diffusion assay, broth microdilution assay, and growth measurements using absorbance were used to compare the methods and fully estimate the antimicrobial properties. Using absorbance measurements, growth rate inhibitions of at least 80% were observed on all targeted species for at least one microalgal EPS-enriched extract. At a concentration of 500 mgGlcEq · L−1, most active extracts of B. subtilis were obtained from Chlamydomonas reinhardtii (87.1% of growth inhibition), Nostoc commune (53.7%), and Eustigmatos polyphem (46.4%). EPS-enriched extracts from C. reinhardtii (86.2%), N. commune (59.9%), and Porphyridium cruentum (31.1%) were found to be the most effective against P. aeruginosa. Antifungal activities were the highest for EPS extracts from Microchloropsis gaditana (86.0%), C. reinhardtii (16.6%), and E. polyphem (17.8%). The results indicated microalgae growth inhibition by EPS-enriched extracts from N. commune (99.3%), C. reinhardtii (84.8%), and M. gaditana (84.1%). To our knowledge, this study is the first to explore the algicidal properties of EPS-enriched extracts derived from microalgae, identifying promising candidates for future investigations into their potential applications. This study investigates the potential of released polysaccharides (RPS) from the halophilic cyanobacterium Cyanothece sp. CE4 as biosorbents for heavy metals, specifically copper (Cu), nickel (Ni), and zinc (Zn). By combining ICP-OES, SEM-EDX, FT-IR spectroscopy, and XAS techniques, this work provides a comprehensive chemical and spectroscopic analysis of the biosorption mechanisms driving metal removal. The results revealed a strong binding affinity for Cu, followed by Ni and Zn, with RPS functional groups playing a key role in metal coordination. The RPS efficiently removed metals from both monometallic and multimetallic solutions, emphasizing their adaptability in competitive environments. XAS analysis highlighted unique metal-specific coordination patterns. Ni preferentially binds to oxygen donors and Zn to chlorine, and Cu exhibits non-selective binding. Remarkably, the extracted RPS achieved a maximum Cu removal capacity of 67 mg per gram of RPS dry weight, surpassing previously reported biosorption capacities. This study not only advances the understanding of biosorption mechanisms by cyanobacterial RPS but also emphasizes their dual role in environmental remediation and circular resource management. Background and Objectives: It has been shown that strains of the genus Enterococcus isolated from donkey milk from different regions of the Republic of Armenia have antimicrobial activity, synthesize different polysaccharides and produce disaccharide polymers (glucose and galactose). The quantitative synthesis of polysaccharides (8-15%) depends on the composition of the nutrient medium, temperature and growing time. Materials and Methods: Species identification of LAB strains was confirmed by 16S rDNA gene sequencing method using universal primers LAB strains. The methods used for extraction, purification and detection of exopolysaccharides are based on the method of Sørensen et al. (2022). The antibacterial activity of EPS was investigated by agar diffusion assay. Determination of the immunostimulating property was carried out using the ELISA method. Results: The antimicrobial activity of the polysaccharide and antimicrobial protein-like fractions of the genus Enterococcus strains depends on its concentration, time of interaction with the test culture, and the species of the pathogenic bacteria. Conclusion: The obtained results were shown that strainsisolated from fermented donkey milk that are capable of synthesizing two substances of different nature with high antimicrobial properties during the growth process are promising for further research and application for their use as probiotics and biopreparations in pharmaceuticals. The availability of water plays an important role in plant growth. However, water availability depends on the climate and irrigation channels; therefore, there is little water available for plants during the dry season. Bacteria found in soil can produce exopolysaccharides to survive under extreme land conditions, namely, during drought conditions. The research objectives were to (1) isolate and select bacteria originating from dry land for use on chili plants and (2) determine the effect of water supply frequency and select soil bacteria on chili plants. The experiment consisted of two stages. (1) Bacterial selection and characteristics, which included exopolysaccharide bacteria selection on specific ATCC No. media. 14; pathogenicity test (hypersensitivity and hemolysis); characterization and biochemical testing including pH, temperature, salinity, oxidation, and catalase; bacterial functional tests (P and K solvents and nitrogen-fixing); and (2) tests of chili plants in the greenhouse. The treatment consisted of two factors: (a) application of selected bacteria, (b) frequency of water application (every 1, 2, 3, and 5 days), and (3) molecular identification of selected bacteria. The results showed that the best growth of chili plants was obtained by treating them with bacteria and watering them every other day. Extremophiles, microorganisms blooming in extreme environmental conditions, hold particular significance in the domain of microbial research. This review paper focuses on extremophilic microorganisms, emphasizing their adaptations and the diverse products they generate, with a particular emphasis on exopolysaccharides (EPSs). EPSs, high molecular weight carbohydrate biopolymers, stand out as valuable products with applications across various industries. The review explores EPS production by bacteria in extreme conditions, including thermophilic, halophilic, and psychrophilic environments. Noteworthy examples, such as B. thermantarcticus and H. smyrnensis AAD6T, highlight the vast potential of extremophiles in EPS production. Additionally, the paper explores the major synthesis pathways of EPSs, shedding light on the factors influencing biosynthesis. The commercial significance of EPSs, especially for extremophiles, is underlined by their applications in medicine, food, environmental protection, agriculture, cosmetics, and more. Furthermore, the review sheds light on the role of extremophiles in various ecosystems, such as acidophiles, alkaliphiles, halophiles, hyperthermophiles, oligotrophs, osmophiles, piezophiles, and radioresistant organisms. Bacillus is a ubiquitous soil bacterium for its plant growth-promoting properties, and it is widely used as a biofertilizer. This research aimed to isolate and determine the morphology and biochemical properties associated with plant-growth promotion ability characterizing Bacillus strains. The bacteria were isolated from the rhizosphere of various highland vegetables grown in Andisols soil in Indonesia. Bacillus isolation was conducted using the serial dilution plate method by means of tryptic soy agar. Characterization of the isolated bacteria included Gram staining, biochemical characteristics, pathogenicity testing, and the production of metabolites, including organic acids, phytohormones, and exopolysaccharides. Four bacterial isolates were identified as Bacillus based on colony and cell morphology, the presence of endospore, as well as biochemical properties and metabolite production. The species determination by PCR amplification and 16S rRNA gene sequence analysis revealed that the four Bacillus were identified as Bacillus safensis strain MDL5, Bacillus altitudinis strain RPW2, and Bacillus sp. strain SZ057. The study presents the occurrence of Bacillus within the rhizosphere of vegetables and the understanding of their properties being considered for biofertilizer development. [ABSTRACT FROM AUTHOR]

Background: Exopolysaccharides (EPS) derived from microbial sources hold great promise for various industrial applications due to their biodegradability and diverse biological activities. Methods and results: In this study, EPS was isolated and characterized from Bacillus amyloliquefaciens strain BDIFST240014, a Gram-positive bacterium known for its robust EPS production. Taxonomic classification was confirmed through 16 S rRNA gene sequencing. We optimized the growth conditions for EPS production, yielding a maximum of approximately 74.81 mg/L after 48 h of incubation at 37 °C. UV-vis and FTIR spectroscopy were used to characterize the EPS, revealing key functional groups indicative of its polysaccharide nature. The EPS contained total carbohydrate and protein contents of 841.234 µg/5 mg and 630.54 µg/5 mg, respectively, along with a potent water holding capacity (WHC) of 103.31% and oil holding capacity (OHC) of 106.78%, underscoring its potential as a multifunctional biopolymer. Antimicrobial assays demonstrated significant activity against both Gram positive and Gram negative pathogens, including S. aureus, B. subtilis, E. coli, and S. typhi. Antioxidant assays confirmed its potent DPPH radical scavenging activity (71.52%) and Fe³⁺ reducing capacity. Furthermore, emulsification studies showed its efficacy with various oils (coconut oil, soybean oil, sunflower oil, olive oil) and a hydrocarbon (n-hexane), with the highest emulsification indices (E1 and E24) of 60.78% and 60.39%, respectively, against coconut and sunflower oils. Notably, the EPS selectively promoted the growth of probiotic strains while not enhancing the growth of pathogenic bacteria, highlighting its prebiotic potential. Cyanobacteria are most abundant in aquatic systems and can grow in freshwater, saline or brackish water, and cold/hot springs. Cyanobacteria have attracted considerable research attention in the last decade as a potential source of numerous biological products in large quantities, such as biofuels, pigments, polyunsaturated fatty acids, nutraceuticals, enzymes, and polysaccharides. Unlike most plant and fungal polysaccharides, the chemical composition, immunomodulatory activity, and molecular mechanisms of action of Cyanobacterium sp. Rippka B-1200 polysaccharides have been studied much less. The complexity of their primary structure due to the high variability of monosaccharides, their diverse bonds, the presence of substituents and high viscosity made detailed structural studies of cyanobacterial polysaccharides rare, which determines the need for analysis of cyanobacteria biomass components to identify active metabolites with promising biological activity. The aim of this study was to investigate the immunomodulatory properties of polysaccharides from Cyanobacterium sp. Rippka B-1200. Pharmacological and nutraceutical value of Cyanobacterium sp. Rippka B-1200 has been set, defining our study's scientific novelty. As a result, the molecular weight of immunoactive polysaccharide (6.0–8.0 kDa) was determined. The analysis shows that endopolysaccharide samples at a concentration of 300 mg/kg showed no significant immunomodulatory effect (1.60 ± 0.15 mg/g), and the thymus mass index of animals in the experimental group was comparable to that of the control group in which animals were immunosuppressed with cyclophosphamide (1.15 ± 0.24 mg/g). When exopolysaccharide samples were used at a concentration of 600 mg/kg, the thymus mass index of animals in the experimental group (3.60 ± 0.32 mg/g) was statistically comparable to that of the control group (without polysaccharide) in which immunosuppression was not induced (thymus index was 3.70 ± 0.25 mg/g). It was found that endopolysaccharide samples at a concentration of 600 mg/kg also exhibited high immunomodulatory activity. When Cyanobacterium sp. Rippka B-1200 endopolysaccharide samples were used, no internal organ changes were observed in experimental animals after immunosuppression. This study presents the synthesis of metal nanoparticles (NPs) with antimicrobial properties from cyanobacterial biomass. Silver (AgNP), copper (CuNP), and zinc (ZnNP) nanoparticles were prepared from exopolysaccharides (EPSs) obtained from isolated cyanobacterial strains. The antimicrobial activity of AgNPs against Escherichia coli and Staphylococcus aureus was evaluated, and compared with CuNPs and ZnNPs, AgNPs were found to have a greater capacity to inhibit bacterial growth. The main factors influencing antimicrobial activity are the concentration and type of metal used. Using an optimized experimental design, specific conditions were established to maximize the antimicrobial efficacy of the synthesized NPs. The characterization of the nanoparticles included UV–VIS, FTIR, and EDX techniques, which confirmed the formation and purity of the AgNPs. This study highlights the effectiveness of cyanobacterial EPS as a reducing and stabilizing agent and provides a sustainable and efficient alternative for producing nanoparticles with biomedical applications. Due to their safety and health-promoting benefits, exopolysaccharides (EPS) secreted by lactic acid bacteria (LAB) have recently attracted considerable interest. Therefore, this study aims to investigate the in vitro antioxidant, anti-inflammatory and anti-diabetic activities, as well as the human compatibility of crude EPS produced by three novel strains of Lactiplantibacillus plantarum, specifically E1K2R2, E2AT5 and E1K1R2 isolated from infant faeces. The antioxidant activity was measured using various radical scavenging assays. The anti-inflammatory activity was evaluated through protein denaturation inhibition, antiproteinase and erythrocyte membrane stabilization assays. The anti-diabetic potential was assessed by measuring the α-amylase inhibitory activity and the biocompatibility of the produced EPS was investigated using the hemolysis assay. The results demonstrated that all crude EPS exhibited significant, concentration-dependent capacity (0.1 to 2 mg/mL) for antioxidant, anti-inflammatory and anti-diabetic activities. Among the strains, EPS from L. plantarum E1K2R2 followed by E2AT5, showed the highest efficacy against oxidative stress, inflammation and diabetes, as indicated by their lower IC50 values (0.78 to 7.65 mg/mL and 0.72 to 9.84 mg/mL, respectively) compared to EPS from L. plantarum strain E1K1R2 (0.79 to 11.59 mg/mL). This suggests a promising potential for health improvement. Additionally, all EPS were found to be biocompatible, displaying minimal hemolytic activity (<4%) at high concentrations. The use of microalgae as a source of food and pharmaceutical ingredients has garnered growing interest in recent years. Despite the rapid growth of the nutraceutical market, knowledge about the potential of bioactive molecules from microalgae remains insufficient. The present study aimed to investigate the biotechnological potential of the green microalga Desmodesmus armatus isolated from a semi-arid region of Brazil. The algal biomass was characterized in terms of gross biochemical composition, exopolysaccharide content, enzymatic inhibition capacity, and antioxidant, antibacterial, and hemolytic activities from solvents of different polarities (water, ethanol, acetone, and hexane). D armatus biomass had 40% of crude protein content, 25.94% of lipids, and 25.03% of carbohydrates. The prebiotic potential of exopolysaccharides from D armatus was demonstrated, which stimulated the growth of Lacticaseibacillus rhamnosus and Lactiplantibacillus plantarum bacteria strains. Moreover, the enzyme inhibition capacity for the proteases chymotrypsin (34.78%–45.8%) and pepsin (16.64%–27.27%), in addition to α-amylase (24.79%) and lipase (31.05%) was confirmed. The antioxidant potential varied between the different extracts, with 2,2-diphenyl-1-picrylhydrazyl sequestration values varying between 17.51% and 63.12%, and those of the 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) method between 6.82% and 22.89%. In the antibacterial activity test, only the ethanolic extract showed inhibition against Listeria sp. (at minimum inhibitory concentration [MIC] = 256 µg mL−1). This fraction also presented the highest significant levels of hemolysis (31.88%–52.45%). In summary, the data presented in the study suggest the presence of biocompounds with biotechnological and nutraceutical potential in the D armatus biomass. Abstract Iron (Fe) deficiency in calcareous soils is a significant agricultural challenge, affecting crop productivity and nutritional quality. This study aimed to isolate, characterize, and evaluate Fe solubilizing rhizobacterial isolates from maize rhizosphere in calcareous soils as potential biofertilizers. Forty bacterial isolates coded as SG1, SG2, …, SG40 were isolated and screened for siderophore production, with ten showing significant Fe solubilizing capabilities. These isolates were further assessed for phosphate solubilization and exopolysaccharides production. The selected bacterial isolates were also screened under axenic conditions for their ability to improve maize growth. The isolates SG8, SG13, SG24, SG30 and SG33 significantly enhanced growth parameters of maize. Notably, SG30 showed highest increment in shoot length (58%), root length (54%), root fresh and dry biomass (67% and 76%), SPAD value (67%), relative water contents (69%), root surface area (61%), and Fe concentration in shoots (79%) as compared to control. The biochemical characterization of these strains showed that all these strains have capability to solubilize insoluble phosphorus, produce indole-3-acetic acid (IAA), and ammonia with catalase, urease and protease activity. Molecular identification through 16s rRNA gene sequencing confirmed high similarity (99.7–100%) of the selected isolates to various Bacillus species, including B. pyramidoids, B. firmicutes, and B. cereus. The study provides a strong base for developing eco-friendly, cost-effective biofertilizers to address Fe deficiency in crops and promote sustainable agriculture.

The adsorption of nickel, cobalt and copper ions onto the surface of exopolysaccharides extracted from cyanobacterial mats has been studied to provide a sustainable and efficient approach for the remediation of heavy metals. The physical characterisation of the EPS surface, including SEM, EDX and FTIR, confirmed an irregular compact structure with multiple cracks, rough macropore structure and the existence of hydroxyl, amine, amide, polysulphide and carbonyl groups. The maximum adsorption capacity (qmax 47.17 , 47.02 and 46.95 mg/g for Cu(II), Co(II) and Ni(II), respectively) was achieved at pH = 6, 0.3 g/L EPS dose, 120 min duration time and 35°C. The remediation affinity of EPS decreased in the order of Ni(II) > Co(II) > Cu(II). The Langmuir model (R2 > 0.95) better fits the adsorption behaviour than the Freundlich model (R2 < 0.90) and the Dubinin – Radushkevich isotherm model, indicating chemical adsorption between the biosorbent and metal ions. Kinetically, the pseudo-second-order model better describes adsorption than does the pseudo-first-order model. Finally, thermodynamic studies have demonstrated that the adsorption process is spontaneously endothermic, with positive enthalpy values (ΔH°) and negative free energy (ΔG°). Bacillus licheniformis 24 (BL24) is an efficient, non-pathogenic producer of 2,3-butanediol (2,3-BD). However, during inulin fermentation, the strain produces large amounts of exopolysaccharides (EPS), which interfere with the process' performance. The present study aims to investigate the effect that inactivation of the sacB gene, encoding levansucrase in BL24, has on 2,3-BD production efficiency. Knockout of the sacB gene was accomplished via insertional inactivation. The sacB-knockout variant formed 0.57 g/L EPS from sucrose and 0.7–0.8 g/L EPS from glucose and fructose, a 15- and 2.5-fold reduction relative to the wild type, respectively. Likewise, during batch fermentation with soluble inulin Frutafit® CLR, the mutant BLΔsacB produced significantly less EPS than the wild type, allowing the maintenance of pH at values favoring 2,3-BD synthesis. At pH 6.50, BLΔsacB reached a record titer of 128.7 g/L 2,3-BD, with productivity of 1.65 g/L/h, and a yield of 85.8% of the theoretical maximum. The obtained concentration of 2,3-BD is two-fold higher compared to that of the wild type. Subsequent RT-qPCR assays confirmed a successful sacB knockout. Three of the genes involved in inulin hydrolysis (sacA, sacC, and fruA) maintained their expression levels compared to the wild type, while that of levB increased. Although total EPS accumulation could not be completely eliminated via sacB gene knockout alone, the overall reduction in EPS content has enabled the highest yield of 2,3-BD from inulin to date, a promising result for the industrial production from inulin-rich substrates. Exopolysaccharides (EPSs) are large-molecular-weight, complex carbohydrate molecules and extracellularly secreted bio-polymers released by many microorganisms, including lactic acid bacteria (LAB). LAB are well known for their ability to produce a wide range of EPSs, which has received major attention. LAB-EPSs have the potential to improve health, and their applications are in the food and pharmaceutical industries. Several methods have been developed and optimized in recent years for producing, extracting, purifying, and characterizing LAB-produced EPSs. The simplest method of evaluating the production of EPSs is to observe morphological features, such as ropy and mucoid appearances of colonies. Ethanol precipitation is widely used to extract the EPSs from the cell-free supernatant and is generally purified using dialysis. The most commonly used method to quantify the carbohydrate content is phenol–sulfuric acid. The structural characteristics of EPSs are identified via Fourier transform infrared, nuclear magnetic resonance, and X-ray diffraction spectroscopy. The molecular weight and composition of monosaccharides are determined through size-exclusion chromatography, thin-layer chromatography, gas chromatography, and high-performance liquid chromatography. The surface morphology of EPSs is observed via scanning electron microscopy and atomic force microscopy, whereas thermal characteristics are determined through thermogravimetry analysis, derivative thermogravimetry, and differential scanning calorimetry. In the present review, we discuss the different existing methods used for the detailed study of LAB-produced EPSs, which provide a comprehensive guide on LAB-EPS preparation, critically evaluating methods, addressing knowledge gaps and key challenges, and offering solutions to enhance reproducibility, scalability, and support for both research and industrial applications. In Italy, watermelon cultivation spans 9510 hectares, with production levels largely influenced by seasonal market demand. As a result, surplus watermelon left unsold by September often remain in the fields, where they decompose naturally and go to waste. A chemical analysis of the watermelon liquid fraction waste (WW) indicates a high carbohydrate concentration, highlighting the potential for biotechnological valorization of this waste stream, converting it into lipids or exopolysaccharides (EPSs). This study investigates the feasibility of utilizing WW as an alternative growth substrate for microalgae, aligning with circular economy principles and advancing sustainable agricultural practices. By repurposing agricultural byproducts, this research supports biorefinery objectives, aiming to convert biomass into high-value secondary products, including biofuels, pigments, and nutraceuticals. Scenedesmus and Chlorella strains demonstrated promising growth and adaptability in WW, achieving biomass yields of 0.95 ± 0.07 g L−1 and 0.37 ± 0.02 g L−1, respectively, with a significant EPS production observed as medium gelation. Although lipid accumulation was limited in this case by the WW substrate, the lipid profiles of both strains were distinctively altered, notably lacking linolenic acid. [ABSTRACT FROM AUTHOR]

In the study, lactic acid bacteria which could produce exopolysaccharids (EPS) were screened from pickled vegetables, the yield of the EPS was determined, and the strain PC715 which have the highest EPS producing ability was identified, at the mean time, the probiotic characteristics of PC715 were also evaluated, including its growth status, acid and bile salt tolerance, self-aggregation, co-aggregation, hydrophobicity, safety, antibacterial activity, and in vitro antioxidant capacities. The results showed that based on the 16S rDNA sequence, the strain with the highest EPS production yield (PC715) was identified as Lactobacillus plantarum. The yield of EPS715 was about 870 mg/L detected by the phenol-sulfuric method. The ultraviolet-visible spectrum indicated no proteins or nucleic acids were in EPS715. L. plantarum PC715 could grow well at both 28 °C and 37 °C, its survival rates at acid conditions (pH=2.5, 3.0, and 4.0) were 75.54%, 91.77%, and 93.69%, respectively, and its survival rates in bile salts (1, 2, and 3 mg/mL) were higher than 40%. The self-aggregation rate of the stain was about 27.64%, the co-aggregation towards Escherichia coli was at about 16%, and the hydrophobic ability were between 0.88%-16.97% towards different organic solvents. L.plantarum PC715 were sensitive to tetracycline, ampicillin, erythromycin, chloramphenicol, and clindamycin, had no hemolytic activity, and had inhibitory effects on E.coli, Bacillus cereus, Staphylococcus saprophyte, Hafnia alvei, Staphylococcus au-reus, and Acinetobacter sp. The scavenging capacity of DPPH, hydroxyl, and ABTS+ radicals of L.plantarum PC715 cell-free supernatant were (62.90±0.87)%, (93.39±2.20)%, and (60.19±1.07)%, respectively. This study showed that exopolysaccharides producing -L.plantarum PC715 might had probiotic potential and could be used as the starter culture in the fermentation industry. [ABSTRACT FROM AUTHOR]
