Male Idesia polycarpa, which display distinct morphological and physiological traits, exhibit greater adaptability to stressful environments than females. However, the connection between this adaptability and rhizosphere processes remains unclear. Here, we investigate the differences in root bacterial community structures between male and female plants at different developmental stages, identifying bacterial strains associated with plant sex through functional predictions. This study aims to inform the optimal allocation of male and female plants during cultivation and provide a theoretical basis for sex identification and breeding. Samples from seven-year-old male and female plants were collected during the flowering (May) and fruit ripening (October) stages. Rhizosphere nutrient content and bacterial diversity were analyzed using Illumina high-throughput sequencing technology. The results demonstrate that total nitrogen (TN), total carbon (TC), and available potassium (AK) varied between sexes at different times. No significant differences between male and female plants were observed in the Shannon, Simpson, and Chao1 indexes during the flowering period. However, the Chao1 and Shannon indexes were significantly higher at fruit maturity in male rather than female plants. The predominant phyla of rhizosphere bacteria were Pseudomonadota, Acidobacteriota, and Actinomycetes. Interestingly, from flowering to fruit ripening, the dominant phyla in both male and female plants shifted from Actinomycetes to Pseudomonadota. A significant correlation was observed between pH and AK and rhizosphere bacteria (p < 0.05), with metabolism being the main functional difference. This study provides preliminary insights into the functional predictions and analyses of bacteria associated with Idesia polycarpa. The above findings lay the groundwork for further investigation into the sex-specific differences in microbial flora across different developmental stages, elucidating the mechanisms underlying flora changes and offering theoretical support for the high-quality management of Idesia polycarpa. The NAC (NAM, ATAF and CUC) gene family plays an important role in plant development and abiotic stress response. However, up to now, the identification and research of the NAC (PeNAC) family members of passion fruit are still lacking. In this study, 25 PeNACs were identified from the passion fruit genome, and their functions under abiotic stress and at different fruit-ripening stages were analyzed. Furthermore, we analyzed the transcriptome sequencing results of PeNACs under four various abiotic stresses (drought, salt, cold and high temperature) and three different fruit-ripening stages, and verified the expression results of some genes by qRT-PCR. Additionally, tissue-specific analysis showed that most PeNACs were mainly expressed in flowers. In particular, PeNAC-19 was induced by four various abiotic stresses. At present, low temperatures have seriously endangered the development of passion fruit cultivation. Therefore, PeNAC-19 was transformed into tobacco, yeast and Arabidopsis to study their function of resisting low temperature. The results show that PeNAC-19 responded to cold stress significantly in tobacco and Arabidopsis, and could improve the low temperature tolerance of yeast. This study not only improved the understanding of the PeNAC gene family characteristics and evolution, but also provided new insights into the regulation of the PeNAC gene at different stages of fruit maturation and abiotic stresses. In light of the continuous increase in organic agriculture, the availability of organic seeds has gained a lot of importance. Especially since the new EU organic regulation came into force on 1 January 2022, proposing reducing the possibility of using untreated conventional seeds in the absence of organic seeds in the future. At the same time, the breeding of tolerant, resistant and adapted varieties is at the basis of organic production as is research to improve seed production and seed quality. In this study, we investigated seed production of 8 tomato genotypes. The aim was to see whether different fruit harvesting frequencies affect seed quality and germination rate. The hypotheses we tested were (i) whether regularly removing fruits from the field would affect total fruit and seed harvest, (ii) whether storage of fruits and (iii) their ripening stage at harvest, had an impact on seed germination. Our results show that while seed production differs between genotypes and extraction time-points, different harvesting procedures, and with that different fruit maturity levels, did not affect thousand-seed weight and seed germination; these findings benefit both small and larger-scale seed producers. Aquaporins (AQPs) in plants can transport water and small molecules, and they play an important role in plant development and abiotic stress response. However, to date, a comprehensive study on AQP family members is lacking. In this study, 27 AQP genes were identified from the passion fruit genome and classified into four groups (NIP, PIP, TIP, SIP) on the basis of their phylogenetic relationships. The prediction of protein interactions indicated that the AQPs of passion fruit were mainly associated with AQP family members and boron protein family genes. Promoter cis-acting elements showed that most PeAQPs contain light response elements, hormone response elements, and abiotic stress response elements. According to collinear analysis, passion fruit is more closely related to Arabidopsis than rice. Furthermore, three different fruit ripening stages and different tissues were analyzed on the basis of the transcriptome sequencing results for passion fruit AQPs under drought, high-salt, cold and high-temperature stress, and the results were confirmed by qRT-PCR. The results showed that the PeAQPs were able to respond to different abiotic stresses, and some members could be induced by and expressed in response to multiple abiotic stresses at the same time. Among the three different fruit ripening stages, 15 AQPs had the highest expression levels in the first stage. AQPs are expressed in all tissues of the passion fruit. One of the passion fruit aquaporin genes, PeTIP3-2, which was induced by drought stress, was selected and transformed into Arabidopsis. The survival rate of transgenic plants under drought stress treatment is higher than that of wild-type plants. The results indicated that PeTIP3-2 was able to improve the drought resistance of plants. Our discovery lays the foundation for the functional study of AQPs in passion fruit. [ABSTRACT FROM AUTHOR]

Seed oil production in Jatropha seeds through different maturity stages have been investigated. In order to meet the high demand of oil (feedstock) for large scale biodiesel production, increasing oil content or output in Jatropha seeds is required. Jatropha fruits were harvested at four different maturity stages and the seeds were analysed for oil content. The seed oil was analysed for fatty acid profile. Results from four different geographical locations investigated namely; Mmadinare, Thamaga, Maun and Shashe, have shown a similar trend in lipid accumulation in Jatropha seeds as the fruits mature from green to brown dry. However, maximum oil content in seeds varies with geographical location. Accumulation of oil in Jatropha seeds during maturation follows a parabolic trend and reaches its peak when fruits are yellow. Oil yield in Jatropha seed kernels ranges from 38.7% to 45.8% for the four maturity stages investigated. Overall results have revealed that harvesting Jatropha fruits when they are yellow increases seed oil output by 6-9% when compared to harvesting the fruits when they are brown dry. There is a relationship between the trend in fatty acid composition in Jatropha seed oil and seed oil content trend during fruit maturation. Based on the trend of unsaturated fatty acids in Jatropha seed oil, particularly linoleic and oleic acids, it can be deduced that reduction of seed oil content from yellow brown to brown dry stage is a result of breakdown of some of the unsaturated fatty acids., (© 2020 The Authors.)
