Aggregation of protein-based therapeutics can occur during development, production, or storage and can lead to loss of efficacy and potential toxicity. Native mass spectrometry of a covalently linked pentameric monoclonal antibody complex with a mass of ∼800 kDa reveals several distinct conformations, smaller complexes, and abundant higher-order aggregates of the pentameric species. Charge detection mass spectrometry (CDMS) reveals individual oligomers up to the pentamer mAb trimer (15 individual mAb molecules; ∼2.4 MDa) whereas intermediate aggregates composed of 6-9 mAb molecules and aggregates larger than the pentameric dimer (1.6 MDa) were not detected/resolved by standard mass spectrometry, size exclusion chromatography (SEC), capillary electrophoresis (CE-SDS), or by mass photometry. Conventional quadrupole time-of-flight mass spectrometry (QTOF MS), mass photometry, SEC, and CE-SDS did not resolve partially or more fully unfolded conformations of each oligomer that were readily identified using CDMS by their significantly higher extents of charging. Trends in the charge-state distributions of individual oligomers provides detailed insight into how the structures of compact and elongated mAb aggregates change as a function of aggregate size. These results demonstrate the advantages of CDMS for obtaining accurate masses and information about the conformations of large antibody aggregates despite extensive overlapping m/z values. These results open up the ability to investigate structural changes that occur in small, soluble oligomers during the earliest stages of aggregation for antibodies or other proteins.

Small extracellular vesicles (sEVs) are cell-released vesicles ranging from 30-150nm in size. They have garnered increasing attention because of their potential for both the diagnosis and treatment of disease. The diversity of sEVs derives from their biological composition and cargo content. Currently, the isolation of sEV subpopulations is primarily based on bio-physical and affinity-based approaches. Since a standardized definition for sEV subpopulations is yet to be fully established, it is important to further investigate the correlation between the biomolecular composition of sEVs and their physical properties. In this study, we employed a platform combining single-vesicle surface-enhanced Raman spectroscopy (SERS) and machine learning to examine individual sEVs isolated by size-exclusion chromatography (SEC). The biomolecular composition of each vesicle examined was reflected by its corresponding SERS spectral features (biomolecular "fingerprints"), with their roots in the composition of their collective Raman-active bonds. Origins of the SERS spectral features were validated through a comparative analysis between SERS and mass spectrometry (MS). SERS fingerprinting of individual vesicles was effective in overcoming the challenges posed by EV population averaging, allowing for the possibility of analyzing the variations in biomolecular composition between the vesicles of similar and/or different sizes. Using this approach, we uncovered that each of the size-based fractions of sEVs contained particles with predominantly similar SERS spectral features. Indeed, more than 84% of the vesicles residing within a particular group were clearly distinguishable from that of the other EV sub-populations, despite some spectral variations within each sub-population. Our results suggest the possibility that size-based EV fractionation methods produce samples where similarly eluted sEVs are correlated with their respective biochemical contents, as reflected by their SERS spectra. Our findings therefore highlight the possibility that the biogenesis and respective biological functionalities of the various sEV fractions may be inherently different.

The integration of mechanically interlocked molecules (MIMs) into polymeric materials has led to the development of mechanically interlocked polymers (MIPs). One class of MIPs that have gained attention in recent years are slide‐ring gels (SRGs), which are generally accessed by crosslinking rings on a main‐chain polyrotaxane. The mobility of the interlocked crosslinking moieties along the polymer backbone imparts enhanced properties onto these networks. An alternative synthetic approach to SRGs is to use a doubly threaded ring as the crosslinking moiety, yielding doubly threaded slide‐ring gel networks (dt‐SRGs). In this study, a photo‐curable ligand‐containing thread was used to assemble a series of metal‐templated pseudo[3]rotaxane crosslinkers that allow access to polymer networks that contain doubly threaded interlocked rings. The physicochemical and mechanical properties of these dt‐SRGs with varying size of the ring crosslinking moieties were investigated and compared to an entangled gel (EG) prepared by polymerizing the metal complex of the photo‐curable ligand‐containing thread, and a corresponding covalent gel (CG). Relative to the EG and CG, the dt‐SRGs exhibit enhanced swelling behavior, viscoelastic properties, and stress relaxation characteristics. In addition, the macroscopic properties of dt‐SRGs could be altered by "locking" ring mobility in the structure through remetalation, highlighting the impact of the mobility of the crosslinks. Background: No clinical trials have evaluated the antimicrobial activity and substantivity of gel formulations containing chlorhexidine (CHX) and cymenol. Objective: To compare the in situ antimicrobial effect and substantivity of a new 0.20% CHX + cymenol gel (test) with the current 0.20% CHX gel formulation (control) on salivary flora and dental plaque biofilm up to seven hours after a single application. Methods: A randomised-crossover clinical trial was conducted with 29 orally healthy volunteers participating in the development of Experiments 1 (saliva) and 2 (dental plaque biofilm). All subjects participated in both experiments and were randomly assigned to receive either the test or control gels. Samples were collected at baseline and five minutes and one, three, five, and seven hours after a single application of the products. The specimens were processed using confocal laser scanning microscopy after staining with the LIVE/DEAD® BacLight™ solution. Bacterial viability (BV) was quantified in the saliva and biofilm samples. The BV was calculated using the DenTiUS Biofilm software. Results: In Experiment 1, the mean baseline BV was significantly reduced five minutes after application in the test group (87.00% vs. 26.50%; p < 0.01). This effect was maintained throughout all sampling times and continued up to seven hours (40.40%, p < 0.01). The CHX control followed the same pattern. In Experiment 2, the mean baseline BV was also significantly lower five minutes after applying the test gel for: (1) the total thickness of biofilm (91.00% vs. 5.80%; p < 0.01); (2) the upper layer (91.29% vs. 3.94%; p < 0.01); and (3) the lower layer (86.29% vs. 3.83%; p < 0.01). The reduction of BV from baseline was observed for the full-thickness and by layers at all sampling moments and continued seven hours after application (21.30%, 24.13%, and 22.06%, respectively; p < 0.01). Again, the control group showed similar results. No significant differences between test and control gels were observed in either saliva or dental plaque biofilm at any sampling time. Conclusions: A 0.20% CHX + cymenol gel application demonstrates potent and immediate antimicrobial activity on salivary flora and de novo biofilm. Background: Restoring the function of the ovary is important for chemotherapy-induced ovarian failure (COF) patients. Stem cell and extracellular vesicles (EVs) therapy show promise but need further improvement. Methods: Human umbilical cord mesenchymal stem cells (hUC-MSCs) were primarily cultured and further three-dimensional (3D) cultured using an ultra-low attachment surface method. The expression levels of nutritional cytokines and immunomodulatory and stemness-related genes of 3D-cultured hUC-MSCs were analyzed. EVs were isolated by ultracentrifugation and characterized. Ovaries were decellularized with sodium dodecyl sulfate to obtain extracellular matrix (ECM). Lyophilized EVs from three-dimensional (2D) or 3D hUC-MSCs were mixed with ECM to prepare the 2D/3D-MSC-EVs-ECM gels. The therapeutic effect of the MSC-EVs-ECM gel on cyclophosphamide (CTX) -treated rats was analyzed through various tests. RNA sequencing was used to analyze the expression changes of genes before and after treatment. Results: After culturing in ultra-low attachment dishes, hUC-MSCs aggregated into spheroids and significantly upregulated the expression levels of immunomodulatory and stemness-related genes. The total EVs yield was also upregulated (5.6-fold) after 3D culture. The cell viability of CTX-treated ovarian granulosa cells (OGCs) was significantly rescued by coculture with the 3D-MSC-EVs-ECM gel. Hormones indicative of ovarian function, AMH, E2, and FSH, were recovered in both the CTX + 2D-MSC-EVs-ECM gel group and the CTX + 3D-MSC-EVs-ECM gel group, while the apoptosis-related protein Bax was significantly downregulated. The 3D-MSC-EVs-ECM gel was more effective than the 2D-MSC-EVs-ECM gel. Significantly differentially expressed genes, such as Hbb-b1, Gpd1, and Sirpa, were detected by RNA sequencing. Hbb-b1 was increased in the ovaries of CTX-treated rats, and this increase was attenuated by injecting the 2D/3D-MSC-EVs-ECM gel. Gpd1 was increased after CTX treatment, and this increase was reversed by the 3D-MSC-EVs-ECM gel. Sirpa was decreased in the ovaries of CTX-treated rats, and this decrease was attenuated by injecting the 3D-MSC-EVs-ECM gel. Poly(N‐isopropylacrylamide) (PNIPAM) gels change their volume and hydrophilicity in response to temperature stimulus in flexible actuators, flexible sensors, and energy storage management. However, traditional PNIPAM gels exhibit low mechanical properties and poor self‐recovery ability due to weak intermolecular (chain) interactions and necessary covalent crosslinking. Herein, a water/ionic liquid (H2O/IL) binary solvent system that provides additional hydrogen bonding and hydrophobic interactions as well as can significantly increase the solubility (up to 60 wt.%) of NIPAM is developed. The fully physically crosslinked PNIPAM ionogel is prepared by utilizing the H2O/IL binary solvent and exhibited excellent mechanical performances, rapid self‐recovery properties, and strong adhesion strength. In addition, the lower critical solution temperature of the PNIPAM ionogel can also be tuned over a wide range (37‐70 °C) by adjusting the IL ratio in the binary solvent. Furthermore, based on the temperature sensitivity and ionic conductivity of the PNIPAM ionogel, applications of PNIPAM ionogels in smart devices, temperature‐managed adhesion, and flexible sensing are demonstrated, respectively. This work improves various performances of PNIPAM gels by using the H2O/IL binary solvent to adjust non‐covalent interactions and provided new ideas for developing high‐performance temperature‐sensitive gels. PVA‐κcarrageenan (κCar) gel is widely used in the pharmaceutical field for transdermal drug delivery, enhancing drug stability and bioavailability. This study aims to modify PVA‐κCar gel by introducing tannic acid. Lidocaine was used as a model drug for incorporation into the novel gel. The PVA‐κCar matrix was mixed with tannic acid prior to freeze– thaw cycle to create a gel composite. The physicochemical features of this gel were evaluated using FTIR, XRD, thermal analysis, and SEM. The presence of tannic acid has reduced the thermal stability of PVA‐κCar from 84 to around 70 °C. The gel content of tannic acid‐modified PVA‐κCar was 34.6% (A) and 52.1% (B). The addition of tannic acid substantially increased gel viscosity. Cytotoxicity analysis showed that tannic acid‐modified PVA‐κCar gel did not affect the viability of HCT colon cancer cells. The encapsulation efficiency of lidocaine in tannic acid‐modified PVA‐κCar was 13 ± 0.2% (A) and 12 ± 0.5% (B), while the drug loading was 0.96 ± 0.01 (A) and 0.91 ± 0.02 (B). In vitro release of lidocaine from the modified PVA‐κCar exhibited a sustained release mechanism in phosphate‐buffered saline solution. Summary: In this study, temperature‐dependent changes in the molecular conformation of curdlan were investigated using differential scanning calorimetry (DSC), microscopic observation, particle size analysis and molecular dynamics (MD) simulation. The DSC results showed that the gel formation temperature of curdlan is related to the glass transition temperature (Tg). The results of microscopic observation and particle size analysis showed that the curdlan particles swelled with increasing temperature, and the original rough and irregular surface morphology gradually became relatively smooth. The MD results show that the weak gel is the result of hydrogen bonds and hydrophobic interaction, whereas the strong gel is less affected by hydrogen bonds, and hydrophobic interaction is the main driving force. In recent years, RG-I pectin isolated by low-temperature alkaline extraction methods has attracted the attention of a large number of researchers due to its huge health benefits. However, studies on other applications of RG-I pectin are still lacking. In this study, we summarized the sources (e.g. potato pulp, sugar beet pulp, okra, apple pomace, citrus peel, pumpkin, grapefruit, ginseng, etc.), extraction methods, fine structure and applications of RG-I pectin in physiological activities (e.g. anti-cancer, anti-inflammatory, anti-obesity, anti-oxidation, immune regulation, prebiotics, etc.), emulsions, gels, etc. These neutral sugar side chains not only endow RG-I pectin with various physiological activities but the entanglement and cross-linking of these side chains also endow RG-I pectin with excellent emulsifying and gelling properties. We believe that this review can not only provide a comprehensive reading for new workers interested in RG-I pectin, but also provide a valuable reference for future research directions of RG-I pectin. Cellulose is an attractive biomolecule that exhibits irreversible intermolecular interactions and forms assemblies with high stability and robustness. Nevertheless, its tendency to irregularly aggregate prevents the polysaccharide chains from assembling into a fine structure. Herein, we demonstrate the formation of gels with relatively homogeneous structures via the self-assembly of cello-oligosaccharides modulated by organic solvent additives or elevated temperatures. Cello-oligosaccharides in a dissolved state in an aqueous alkaline solution started to self-assemble upon the addition of an acid for neutralization at room temperature, forming hydrogels with heterogeneity at the sub-micrometer or larger scale. On the other hand, cello-oligosaccharide assembly in the presence of 20% (v/v) water-miscible organic solvents produced approximately ten times stiffer gels with more homogeneous structures. The highest Young's modulus of the gels in this study was ~ 1.5 kPa. Moreover, increasing the assembly temperature from 25 °C to 50 °C also increased the gel stiffness. It was suggested that organic solvent additives and elevated temperatures decreased the solubility of cello-oligosaccharides and thus increased the assembly kinetics for the formation of more homogeneous network structures before non-organized aggregation. The electrophysiological interface is essential for understanding electrical activities within biological organisms and human‐machine interaction. However, traditional preformed bioelectrodes show insufficient interface contact with hairy and irregular skin, causing signal attenuation and distortion. Here, an on‐skin‐formed silk protein bioelectrode is reported with a conformable and robust interface, achieving a high signal‐to‐noise ratio for electromyographic signals, showing up to a 38.9% increase compared to traditional ones. The fluid‐gel transition of silk protein is induced by a gelling agent composed of ethanol and sodium alginate, leading to a conformable and seamless interface with hairy and irregular human skin, thus resulting in a high signal‐to‐noise ratio. The developed bioelectrode can record electrophysiological signals stably, even underwater. Additionally, human‐machine interaction using electromyographic signals is designed to precisely control the model car's motion trajectory. Non‐conjugated luminescent polymers have received much attention due to their excellent flexibility, processability, and biocompatibility. However, the single emission color and low solid‐state luminescence efficiency restrict their further development. Herein, by introducing carboxyl‐terminated alkyl chains on the rigid and helical polymaleimide backbone, a series of non‐conjugated luminescent polymers are designed and synthesized. Due to the "spring lock" formed by the synergism of scalable helical main chains and flexible alkyl side chains, these polymers exhibit effective cluster luminescence with thermally activated delayed fluorescence (TADF) property, thereby significantly improving the solid‐state luminescence efficiency with the quantum yield up to 59.6%. The TADF emission wavelengths of the polymer powders can be tuned from 498 to 639 nm through changing reaction solvents, because the conformation of helical chains forming clusters is sensitive to solvent polarity. Interestingly, metal ions can induce the polymers to construct self‐repairing cluster luminescence gels, which show excellent potential applications in ammonia detection and information encryption. This work not only provides an effective design strategy for the colorful cluster luminescence with high‐efficiency TADF, but also further reveals the cluster luminescence mechanism and enlightens the promising applications of cluster luminescent gels. A full analysis was conducted to investigate the impact of germinated brown rice slurry on the gel properties of hairtail surimi. The potential benefits were also addressed for the dysphagia in the elderly. Some parameters were then measured to determine the gel strength, texture profile, water-holding capacity, dynamic rheology, whiteness, water state distribution, protein secondary structure, microstructure, in vitro digestibility, and International Dysphagia Diet Standardization Initiative (IDDSI) level. The results demonstrated that there was a significant impact of germinated brown rice slurry on the gel strength of hairtail surimi. Moreover, the gel strength of hairtail surimi was reduced from (649.91±64.35) to (480.93± 98.84) g·mm (P<0.05), indicating a substantial reduction. The gel strength was reduced to make the swallowing easier and safer, which was beneficial for individuals with dysphagia. Furthermore, the brown rice slurry shared a significant impact on various textural properties, such as hardness, cohesiveness, and chewiness. The brown rice group exhibited a significant decrease in hardness (from 585.63±61.35 to 399.87±51.23), cohesiveness (from 81.14±10.23 to 50.16±7.13), and chewiness (from 30.14±5.00 to 18.31±3.97) (P<0.05), compared with the control group. The germinated brown rice slurry was then incorporated into the hairtail surimi, resulting in a softer and more easily chewable product. Interestingly, there was no significant effect on the whiteness value, despite these changes observed in the texture properties. The visual appearance of hairtail surimi failed to alter using germinated brown rice slurry. Furthermore, an augmentation in the water-holding capacity was observed after the addition of germinated brown rice slurry to hairtail surimi. The water-holding capacity exhibited a significant increase from (77.92%±0.73) to (81.92%±1.40) (P<0.05). The brown rice slurry was incorporated to induce the distribution of water states, thereby enhancing the protein-water binding. As such, the moisture was retained within the commonly-used product during cooking or processing, in order to prepare the meals for individuals with dysphagia. A more porous network structure was found to form the surimi gel after the addition of brown rice, compared with the control group. This structure was attributed to the unique properties of brown rice, such as the high fiber content and water absorption. The high porosity greatly contributed to the open gel matrix for better water retention. Furthermore, there were outstanding changes in the protein structure within the surimi gel. The α-helix structure content of myofibrillar protein significantly increased from (47.12%±1.09) to (52.74%±1.65) after the incorporation of brown rice (P<0.05). The functionality of protein was enhanced for the gel-forming ability. There was a significant decrease in the β-sheet structure content from (25.67%±0.84) to (20.99%±0.65) after the addition of brown rice into the surimi gel (P<0.05). A softer texture then improved the tenderness, due to the decreased protein aggregation. Meanwhile, the IDDSI level was determined to evaluate these structural effects on the practical applications in both groups. According to the IDDSI standards, the brown rice group reached level 5 consistency, representing the texture of a mildly thick liquid diet. While the control group achieved level 6 consistency, indicating the texture of a moderately thick liquid diet. In addition to structural modifications, aroma compounds also exhibited notable differences between both groups after sensory analysis. The inclusion of germinated brown rice slurry led to significant increases in the various aroma compounds, including aldehydes, ketones, alcohols, and esters (P<0.05). There was a positive contribution towards flavor perception. Therefore, the germinated brown rice slurry was incorporated to enhance the textural attributes for the overall sensory, in order to enrich the aroma profiles. An optimal amount of germinated brown rice slurry can be expected to soften the gel characteristics of hairtail surimi. The objective of this study was to isolate chitosan from shrimp shells using physical–chemical treatments and then evaluate the impact of additives on the chitosan matrix. Two different additives, glycerol and gel, were incorporated into the chitosan polymer using the casting process, and film thickness was measured to investigate the effect of additives on film properties. Scanning electron microscopy (SEM)-EDS was used to determine the morphology and percentage of elements in the film, while contact angle measurements were taken to assess the effect of film wettability on surface properties. The CT-gel sample exhibited a contact angle of 79.15° on the right and 78.17° on the left, which was crucial for CT, CT-gly, and CT-gly-gel. The mechanical behavior of the films was determined using a tensile test, and it was found that chitosan-glycerol systems had high transparency and homogeneity, with their properties depending on the content and types of additives. On the other hand, chitosan-gel systems had low transparency and a greenish hue, but were still homogeneous. Pure films and films with additives (CT, CT-gel, CT-gly, and CT-gel-gly) were found to be highly flexible due to their mechanical properties. Gelatin hydrogel with excellent biodegradability and biocompatibility is widely used in drug delivery, electronic devices, and flexible sensors. However, it is prone to dehydration, which severely restricts its applications. The incorporation of hydrated ions, for example choline chloride (ChCl), can not only improve its water retention capacity but also keep its biocompatibility. Unfortunately, the improvement in water retention is realized at the expense of mechanical properties because the ChCl can suppress the crystallization of gelatin. Here, ChCl is introduced into gelatin hydrogel using a simple post‐treatment to simultaneously improve its water retention capacity and mechanical properties. As a result, the resultant gelatin gel possesses a maximum fracture strength of 5.4 MPa with a strain of 623% and can maintain its weight for at least 30 days. In addition, the gelatin gel exhibits intriguing properties, such as self‐healing, anti‐freezing, anti‐drying, recycling, conductivity, and moldability, which can be successfully served as wearable sensors to monitor human movements. Further, a sensitive touch sensor based on electrode potential is established for the first time using the obtained gel, which can transmit messages through the touch potential signal. Because of widespread cultural approval. Strong acceptance by human body, and reduced occurrence of side effects, about 75-80% of the worldwide population, especially in less developed areas, continues to favor herbal remedies as their main option for fundamental healthcare. Herbal treatments comprise botanical or plant-based components and serve to address injuries, infections, and ailments. They are also employed for preventive health measures and to facilitate the recovery process. This research explored the possible therapeutic advantages of Zingiber officinale, commonly recognized as ginger, a naturally occurring anti-inflammatory substance with extensive culinary use. Numerous investigations have highlighted the advantages of ginger in managing conditions such as morning sickness, chronic dyspepsia, hypoglycemia, risk factors for heart disease, chemotherapy-induced nausea, and menstrual discomfort. Ginger ethanolic extract preparations in the form of transdermal gel consist of different concentrations and combinations of Carbopol 934 and Carbopol 940. All gel formulations exhibited favorable characteristics such as Spreadability, uniformity, Viscosity, and extrusion. Among these compositions, formulation F3, which contains Carbopol 934, displayed the highest release of the active ingredient in vitro and the ethanolic extract demonstrated significant anti-inflammatory effects in vitro. The efficacy of pregabalin in pain treatment has led to the search for new formulations for its use through different routes of administration. This study aimed to prepare, characterize, and evaluate the cytotoxicity of pregabalin (PG) gels for topical application in the oral cavity. Solutions with three different concentrations of PG were prepared and added to a 1.0% carbopol gel base. Thermal analyses (TG and DSC) and FTIR were performed on the gel and pure pregabalin. Stability (preliminary and accelerated) and rheology studies were also conducted on the gels. Cytotoxicity was evaluated in human gingival fibroblasts in the following groups: WG (1.0% carbopol gel base), PG2G (2.0% pregabalin gel), PG5G (5.0% pregabalin gel), and PG10G (10% pregabalin gel). A transparent and homogeneous gel with a pH of 6 was obtained. The formulations showed stability, and the different drug concentrations did not influence the product's characteristics. None of the tested groups showed cytotoxicity for the analyzed cells. The pregabalin gels exhibited favorable and non-toxic characteristics for human gingival fibroblasts in vitro. Adding hydrocolloids into native starch is a secure and valid method of physical modification. However, few research focuses on the interactions between konjac gum (KGM) and purple potato starch (PPS). In this study, the effects of KGM on the physicochemical, rheological, textural properties, and in vitro digestibility of PPS are evaluated. The swelling power, solubility, transparency, zeta potential value, particle size, gel adhesiveness, and cohesiveness of PPS are enhanced with increasing KGM content. The results show the combination of PPS and KGM exhibits a higher adhesiveness and a denser structure than that of PPS alone. Static rheological analysis shows PPS/KGM composites exhibit pseudoplastic and shear thinning enhancement behaviors with the rise of the KGM content from 0% to 2.5%. Dynamic rheological analysis reveals that storage modulus (G') > loss modulus (G″), mainly showing elastic behavior, and G′, G″, and the loss tangent (tan δ) increase with the rise of the KGM content from 0% to 2.5%. Fourier transform infrared spectroscopy analysis shows stronger hydrogen bonding of PPS/KGM complexes compared to PPS only. Moreover, KGM increases the contents of resistant starch. The objective of the study was to develop a novel topical gel by mixing Potentilla tormentilla ethanolic extract, thermosensitive poloxamer 407, and carbomer 940 and evaluating its stability and rheological behavior. The irritation potential of the gel was evaluated in accordance with the Organization for Economic Cooperation and Development Guidelines 404. The potential anti-inflammatory effects of the developed gel were evaluated in vivo in rats using the carrageenan-induced paw edema test. Moreover, the in silico binding affinity for chlorogenic and ellagic acid, as dominant components in the extract, against cyclooxygenase (COX) 1 and 2 was also determined. Our findings suggest that the gel containing Potentilla tormentilla extract remained stable throughout the observation period, exhibited pseudoplastic behavior, and caused no irritation in rats, thus being considered safe for topical treatment. Additionally, the developed gel showed the capability to reduce rat paw edema, which highlights significant anti-inflammatory potential. In silico analysis revealed that chlorogenic and ellagic acid exhibited a reduced binding affinity against COX-1 but had a similar inhibitory effect on COX-2 as flurbiprofen, which was confirmed by molecular dynamics results. The research was focused on incorporating the polyphenols from Indian gooseberries (Phyllanthus emblica L.) in phospholipid nanocarriers, aiming to enhance skin penetration, bioavailability and stability. Design-Expert® aided in achieving the optimal liposomal formulation, characterized by particle size, zeta potential, and a polydispersity index. Liposomes were prepared by applying the film hydration method. The liposomes were further loaded into a hydrogel (Carbopol 934) for its controlled release and stabilizing effects on liposomes. Multiple antioxidant assessment methods DPPH, ABTS, FRAP, CUPRAC were performed. Additional evaluations encompassed FTIR, SEM, rheological studies, and in vitro/ex vivo diffusion comparisons between liposomal-loaded gel (GEL-LE) and pure extract with gel (GEL-E). Depending on the formulation and extract amount, the total antioxidant content per sample varies between 59.3-486.75 mg. When the optimal formulation (LE) contained 1.8 % soybean and 0.07 % cholesterol the mean particle size was 74.66 nm, zeta potential - -50.35 and polydispersity index-0.3. In vitro results exhibited 42.28 % cumulative release for GEL-LE and GEL-E by 23.44 %. Ex vivo findings showed a 6% discrepancy in cumulative release (21% for GEL-LE). Chinese herbs are a huge treasure trove of natural products and an important source of many active molecules. The theory of traditional Chinese medicine compatibility (TCMC) is widely applied in clinical practice, but its mechanism is still ambiguous. This study aims to open a new window for this predicament by studying the interaction between the main active ingredients from a drug pair. Carrier-free assembly of natural products improves the shortcomings of traditional nanodelivery systems and opens a new path for the development of new nanomaterials. The drug pair "Pueraria and Hedyotis diffusa" has been commonly used in clinical practice, with a predominant therapeutic effect. This study is devoted to the study of the binary small molecule co-assembly of the main active molecules from the drug pair. In this study, we introduce a carrier-free composite gel, formed by the co-assembly of puerarin (PUE) and deacetylasperulosidic acid (DAA) via non-covalent bonds including π–π packing, intermolecular hydrogen bonding, and C=O π interactions. With a strain point 7-fold higher than that of P gel, the P − D gel exhibited favorable rheological properties. The survival rate of SW1990 cells in the P − D group was only 21.39% when the concentration of administration reached 200 μM. It thus demonstrated activity in inhibiting SW1990 cells' survival, suggesting potential in combating pancreatic cancer. Furthermore, this research offers a valuable concept for enhancing the mechanical properties and bioactivity of hydrogel materials through the utilization of a multi-component natural small molecule co-assembly approach. Letrozole (LTZ) is used as first-line treatment for hormone-positive breast cancer (BC) in postmenopausal women. However, its poor aqueous solubility and permeability have reduced its clinical efficacy. Herein, we developed LTZ-nanotransferosomes (LTZ-NT) to address above mentioned issues. The LTZ-NT were optimized statistically using Design Expert® followed by their characterization via dynamic light scattering (DLS), Transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FTIR), and Differential scanning calorimetry (DSC). The optimized LTZ-NT was incorporated into 1% chitosan-gel to develop LTZ-NTG. Moreover, in vitro drug release and ex vivo permeation of LTZ-NTG were performed and compared with LTZ-dispersion and LTZ-NT. Additionally, skin irritability and histopathology of LTZ-NTG were investigated. Furthermore, in vitro antitumor study of LTZ-NTG was investigated in BC cell lines. The optimized LTZ-NT showed suitable zeta potential (30.4 mV), spherical size (162.5 nm), and excellent entrapment efficiency (88.04%). Moreover, LTZ-NT exhibited suitable thermal behavior and no interactions among its excipients. In addition, LTZ-NTG had an optimal pH (5.6) and a suitable viscosity. A meaningfully sustained release and improved permeation of LTZ was observed from LTZ-NTG. Additionally, LTZ-NTG showed significantly enhanced cell death of MCF-7 and MCC-7 cells. Androgenic alopecia (AGA) has a considerable impact on the physical and mental health of patients. Nano preparations have apparent advantages and high feasibility in the treatment of AGA. Cardamonin (CAR) has a wide range of pharmacological activities, but it has the problems of poor solubility in water and low bioavailability. There are few, if any, researches on the use of nano-loaded CAR to improve topical skin delivery of AGA. In this study, a CAR-loaded liposomal formulation (CAR@Lip and CAR@Lip Gel) was developed and characterized. The prepared CAR@Lip exhibited a uniform and rounded vesicle in size. CAR@Lip and CAR@Lip Gel can significantly improve the cumulative release of CAR. Additionally, CAR@Lip can obviously promote the proliferation and migration of human dermal papilla cells (hDPCs). Cell uptake revealed that the uptake of CAR@Lip significantly increased compared with the free drug. Furthermore, both CAR@Lip and CAR@Lip Gel groups could markedly improve the transdermal performance of CAR, and increase the topical content of the drug in the hair follicle compared with CAR. The ratchet effect of hair follicles could improve the skin penetration depth of nanoformulations. Notably, Anti-AGA tests in the mice showed that CAR@Lip and CAR@Lip Gel groups could promote hair growth, and accelerate the transition of hair follicles to the growth stage. The anti-androgen effect was revealed by regulating the expression of IGF-1, VEGF, KGF, and TGF-β, participating in SHH/Gli and Wnt/β-catenin pathways. Acne and hyperpigmentation are the most common problems in adolescents globally. Resveratrol and glabridin are proven to be effective against acne and hyperpigmentation. We developed a gel comprising these two actives for acne and hyperpigmentation treatment. The purpose was to establish a reliable UV method of analysis for quantification of resveratrol and glabridin, with high precision and accuracy. UV spectral analysis indicated 306nm and 281nm to be the λmax for resveratrol and glabridin, respectively, in methanol. The linearity studies conducted as per ICH guidelines Q2 R1 resulted in a correlation coefficient (r2) of 0.99. This method was used to assay the developed gel for resveratrol and glabridin content and demonstrated the suitability of this UV spectroscopic method for simultaneous quantification. Summary: The purpose of this experiment was to explore the improvement of tussah pupa powder on the deterioration of pork batter emulsion gel by using proteomics, distribution of moisture, rheological properties and surface hydrophobicity. The results showed that the myosin decomposed as the heating temperature increased; as the increase of the addition proportion of tussah pupa powder at each sterilisation temperature point, compared to the control group, the pH of a pork batter emulsified gel showed a significant increased trend (P < 0.05); the low molecular weight proteins or formed oligomers of tussah pupa powder could be used as fillers to participate in the formation process of gel network; the vitellogenin of tussah pupa powder could form a 'skeleton' in the pork batter system; as the sterilisation temperature at 100 °C (30 min) and 110 °C (30 min), the surface hydrophobicity was the largest as the addition amount 2%. In short, the tussah pupae powder could improve the deterioration characteristics of the pork batter emulsified gel by influencing the pH, surface hydrophobicity and the formation of gel network of pork batter system. The work reports a novel nanophytosomal gel encapsulating Alpinia galanga (L.) Willd leaf essential oil to treat periodontal infections. Alpinia oil-loaded nanophytosomes (ANPs) were formulated by lipid layer hydration technique and were evaluated by FESEM, cryo-TEM, loading efficiency, zeta potential, particle size, release profile etc. Selected ANPs-loaded gel (ANPsG) was evaluated by both in vitro and in vivo methods. Selected ANPs were spherical, unilamellar, 49.32 ± 2.1 nm size, 0.45 PDI, −46.7 ± 0.8 mV zeta potential, 9.8 ± 0.5% (w/w) loading, 86.4 ± 3.02% (w/w) loading efficiency with sustained release profile. ANPsG showed good spreadability (6.8 ± 0.3 gm.cm/sec), extrudability (79.33 ± 1.5%), viscosity (36522 ± 0.82 cps), mucoadhesive strength (44.56 ± 3.5 gf) with sustained ex vivo release tendency. Satisfied ZOI and MIC was observed for ANPsG against periodontal bacteria vs. standard/control. ANPsG efficiently treated infection in ligature induced periodontitis model. Plant-based hydrocolloid systems which are mixed with different carbohydrate types are becoming popular due to vegan concerns for both non-food and food industry. In this study, different sugar types (d-allulose and sucrose) at different concentrations (20% and 40%) were used to explore their effects on the gelation and physical properties of agar–agar. For characterization of these agar–agar-based gel systems, in addition to common physical methods (moisture content, water activity, color, hardness, viscosity), novel methods such as TD-NMR relaxometry, and FTIR were also used to get deep insight of the microstructures and water dynamics of the gel systems. Results clearly indicated that sugar type and concentration were vital factors affecting the agar–agar gel properties and dynamics of the system. Utilization of allulose in the gel systems led to formation of less moist and softer agar–agar matrix characterized with longer T2 spin–spin relaxation times. Man has used medicinal plants as remedies for several human diseases for centuries. This paper therefore evaluates the physicochemical and antibacterial properties of ethanolic and gel extracts of common wireweed (Sida acuta Burm.f.) using appropriate standard techninques. The filtrate was concentrated to a semi-solid mass (extract) and the antibacterial activity of the extract against Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, and Escherichia coli were assessed using the agar well diffusion technique. The gels have good homogeneity, and spreadability, and are easily washable. The pH ranges between 5.8 and 6.3 and is within the normal skin pH range (4.0-6.8). The extrudability ranges from 45 to 70%. The viscosity of the gels is between 6178.6 mPas and 59,343 mPas and they are shear thinning systems. The gels prepared using Sida acuta ethanolic extract have antibacterial activity that is comparable to that of the extract alone. Simple Summary: This study evaluated the use of a carboxymethylcellulose (CMC)/polyethylene oxide (PEO) composite material to prevent adhesions following orthopedic surgery in dogs. Ten purpose-bred laboratory Beagle dogs were allocated consecutively into two groups—one received the anti-adhesion material during an ulna osteotomy, while the other group did not. The results showed no significant differences between the groups in terms of surgical site size, lameness, or bone healing on imaging evaluation. The group that received the anti-adhesion material had significantly lower adhesion and better healing scores based on macroscopic and histopathologic evaluation. These findings suggest the CMC/PEO composite is a safe and potentially effective way to prevent post-surgical adhesions in canine orthopedic patients, without compromising bone healing. Further research is needed to fully characterize the clinical benefits of this approach. The formation of adhesions is a common complication following traumatic injuries and surgical procedures, often resulting in pain, stiffness, and loss of function. This study aimed to evaluate the feasibility and safety of using a composite material comprising of carboxymethylcellulose (CMC), polyethylene oxide (PEO), and calcium chloride, for preventing adhesions between muscle and bone during the healing stage, as well as its effect on the bone healing process. Ten healthy purpose-bred laboratory Beagle dogs were randomly subjected to two consecutive operations with a 6-month interval, alternating between left and right forelimbs. On the left forelimb an osteotomy at the ulna was performed, while on the right forelimb the same procedure was supplemented by the application of the anti-adhesion agent in the osteotomy site prior to closure. Clinical, diagnostic imaging, macroscopic, and histological evaluations were performed at various time points. The results showed no significant differences in surgical site perimeter (p = 0.558), lameness (p = 0.227), and radiographic bone healing (p = 0.379) between the two groups. However, the macroscopic (p = 0.006) and histological assessments revealed significantly lower adhesion scores (p = 0.0049) and better healing (p = 0.0102) in the group that received the anti-adhesion agent. These findings suggest that the CMC/PEO composite material is a safe and potentially effective intervention for preventing post-traumatic and post-surgical adhesions in canine patients without compromising bone healing. Background: Acne is the most common skin disorder which is known as a chronic inflammatory disease with psychological burden and reduced quality of life. Objective: The objective of this study is to determine, in a real-world effectiveness and clinical experience in treating acne scars which includes evaluating reduction in depth, diameter, erythema and pigmentation, with an acne scar gel, and to evaluate the effect of the product on quality of life. Methods: Overall, 435 patients were assessed for 90 days. The acne scars were counted and classified as mild, moderate or severe and the type of acne scar was registered. The study participants had gone through both oral and topical treatment for a time range of 1 week to over a month depending on the severity of the scars. They had also gone through medical procedures as per the requirement. The results were assessed at an interval of 30 days to 90 days and were graded between 1-5. Results: Overall facial appearance of atrophic acne scars improved after treatment with acne scar gel. Clinical experience with acne scar gel (in terms of reduction of scar depth, diameter, erythema, and pigmentation), and subject satisfaction (happiness and confidence) score along with the sensation and perception of the skin improved in most subjects. Conclusions: Acne scar gel was effective and safe for treatment of moderate-to-severe acne scars. When encountering heavy oil reservoirs during drilling, due to the change in pressure difference inside the well, heavy oil will invade the drilling fluid, and drilling fluid will spill into the reservoir along the formation fractures, affecting the drilling process. A supramolecular polymer gel-based temporary plugging agent was prepared using acrylamide (AM), butyl acrylate (BA), and styrene (ST) as reacting monomers, N, N-methylenebisacrylamide (MBA) as a crosslinking agent, ammonium persulfate (APS) as an initiator, and poly(vinyl alcohol) (PVA) as a non-covalent component. A supermolecular polymer gel with a temperature tolerance of 120 °C and acid solubility of 90% was developed. The experimental results demonstrated that a mechanically robust, thermally stable supramolecular polymer gel was successfully synthesized through the copolymerization of AM, BA, and ST, as well as the in situ formation hydrogen bonding between poly (AM-co-BA-co-ST) and PVA, leading to a three-dimensional entangled structure. The gel-forming solution possessed excellent gelling performance even in the presence of a high content of salt and heavy oil, demonstrating superior resistance to salt and heavy oil under harsh reservoir conditions. High-temperature and high-pressure plugging displacement experiments proved that the supramolecular polymer gel exhibited high pressure-bearing capacity, and the blocking strength reached 5.96 MPa in a wedge-shaped fracture with a length of 30 cm. Abstract Background No clinical trials have evaluated the antimicrobial activity and substantivity of gel formulations containing chlorhexidine (CHX) and cymenol. Objective To compare the in situ antimicrobial effect and substantivity of a new 0.20% CHX + cymenol gel (test) with the current 0.20% CHX gel formulation (control) on salivary flora and dental plaque biofilm up to seven hours after a single application. Methods A randomised-crossover clinical trial was conducted with 29 orally healthy volunteers participating in the development of Experiments 1 (saliva) and 2 (dental plaque biofilm). All subjects participated in both experiments and were randomly assigned to receive either the test or control gels. Abstract Background Restoring the function of the ovary is important for chemotherapy-induced ovarian failure (COF) patients. Stem cell and extracellular vesicles (EVs) therapy show promise but need further improvement. Methods Human umbilical cord mesenchymal stem cells (hUC-MSCs) were primarily cultured and further three-dimensional (3D) cultured using an ultra-low attachment surface method. The expression levels of nutritional cytokines and immunomodulatory and stemness-related genes of 3D-cultured hUC-MSCs were analyzed. EVs were isolated by ultracentrifugation and characterized. Ovaries were decellularized with sodium dodecyl sulfate to obtain extracellular matrix (ECM). Lyophilized EVs from three-dimensional (2D) or 3D hUC-MSCs were mixed with ECM to prepare the 2D/3D-MSC-EVs-ECM gels. The therapeutic effect of the MSC-EVs-ECM gel on cyclophosphamide (CTX) -treated rats was analyzed through various tests. RNA sequencing was used to analyze the expression changes of genes before and after treatment. Results After culturing in ultra-low attachment dishes, hUC-MSCs aggregated into spheroids and significantly upregulated the expression levels of immunomodulatory and stemness-related genes. The total EVs yield was also upregulated (5.6-fold) after 3D culture. The cell viability of CTX-treated ovarian granulosa cells (OGCs) was significantly rescued by coculture with the 3D-MSC-EVs-ECM gel. Hormones indicative of ovarian function, AMH, E2, and FSH, were recovered in both the CTX + 2D-MSC-EVs-ECM gel group and the CTX + 3D-MSC-EVs-ECM gel group, while the apoptosis-related protein Bax was significantly downregulated. The 3D-MSC-EVs-ECM gel was more effective than the 2D-MSC-EVs-ECM gel. Significantly differentially expressed genes, such as Hbb-b1, Gpd1, and Sirpa, were detected by RNA sequencing. Hbb-b1 was increased in the ovaries of CTX-treated rats, and this increase was attenuated by injecting the 2D/3D-MSC-EVs-ECM gel. Gpd1 was increased after CTX treatment, and this increase was reversed by the 3D-MSC-EVs-ECM gel. Sirpa was decreased in the ovaries of CTX-treated rats, and this decrease was attenuated by injecting the 3D-MSC-EVs-ECM gel. Conclusions Our study demonstrated that the 3D-MSC-EVs-ECM gel is an efficient strategy for the recovery of ovarian function in CTX-induced ovarian failure.

Jam, as a common processed fruit product, has a long history of consumption and is widely loved by consumers due to its diverse products and rich nutrition. The texture of traditional jam can only be formed under high sugar (55%–75%) and highly acidic (pH 2.0–3.5) conditions, which is a challenge for the production of low-sugar jam. In addition, the texture of jam is vulnerable to damage during transport and storage, leading to jam quality deterioration. Food thickeners, food additives that increase the viscosity, consistency and gel strength of liquid foods, are widely used for promoting the formation of the texture of low-sugar jam and maintaining its stability during transport and storage. In this article, the deterioration phenomenon and formation theory of jam texture are reviewed. The thickening or gelling mechanism of food thickeners (e.g., pectin) and their application in jam are described with a focus on the effects of food thickeners on the sensory attributes, texture, rheological properties, and microstructure of jam. Moreover, several possible future research directions are proposed, aiming to provide ideas for the development and application of thickeners in jam.

Background and objective: Salicylic acid (SA) has keratolytic activities and it is used topically to treat dandruff and seborrheic dermatitis. The hydrotropic phenomenon in pharmaceutical preparations is utilized to enhance the solubility of water-insoluble drug molecules that promoted to prepare the salicylic acid as a hair gel for local effect and it can extend its keratolytic action for a longer period compared to the salicylic acid solution. Methods: Preformulation study was performed to exclude any unwanted chemical interaction between SA and excipients using Fourier-transform infrared spectroscopy (FTIR). The solubility of SA was determined separately in sodium acetate and sodium citrate solutions at a concentration of 1, 3, and 5 %w/v, using distilled water as a solvent. An optimum gel formulation was developed and it was used to prepare the SA gel formulation. Characterizations were performed in terms of physical appearance, viscosity, pH, and spreadability, in-vitro studies were performed in physiological pH, and ex-vivo diffusion studies were performed utilizing rats’ skin. Results: FTIR did not show any chemical interaction between the drug and sodium citrate. The hydrotropic solution of sodium citrate with a concentration of 5% w/v increased the solubility of SA by 28 folds, while the sodium acetate solutions with a concentration of 5% w/v increased the solubility of SA by 19 folds. The optimum gel formula (F1) with a drug content of 97% showed a slow dissolution rate and minimum diffusion through the skin. Conclusion: The hydrotropic solubilization technique significantly influenced the solubilization of salicylic acid in the water and the highest solubility rate was achieved from 5% w/v sodium citrate solution. The formulated hydrogels using carbopol 971P as gelling agent decreased the diffusion rate of SA.

Hypothesis: Capillary suspensions offer a new approach to generate novel materials. They are ternary liquid-liquid-solid systems characterized by particles connected by liquid bridges of one fluid suspended in a second immiscible bulk fluid. The viscosity of the bulk liquid can be modulated to customize the structure and rheological properties of capillary suspensions. Experiments and simulations: Using experiments and numerical simulations, we investigated capillary suspensions in the pendular state, using silica particles and water as a bridging liquid. To modulate the viscosity of the bulk fluid, we use different ratios of either dodecane and diisononyl phthalate, or silicone oils with varying chain lengths as bulk liquids. The rheological behavior was characterized using the maximum storage and loss moduli and the yielding behavior. This was related to structural changes of the systems, which was visualized using confocal laser scanning microscopy. In addition, we used Molecular Dynamics (MD) simulations to gain more insights into the behavior of two particles connected by a liquid bridge for various bulk liquids. Findings: Experiments show that higher bulk liquid viscosity reduces strength, yield stress, and yield strain in capillary suspensions, which is partly attributed to a reduced inter-connectivity of the percolating network. In this study, the effects of four Hofmeister salts (KCl, NaCl, CaCl2 and MgCl2) at different concentrations (0.02, 0.04 and 0.06 mol/L) on gel properties and rheological properties of soy protein isolate (SPI) were systematically explored. The results showed that the addition of salt ions could improve the gel strength of SPI, but higher concentrations (0.04 and 0.06 mol/L) of CaCl2 and MgCl2 reduced the gel stickiness of the samples. In the dynamic rheological test, each salt ion increased the storage modulus (G’) and loss modulus (G”) of the samples to varying degrees. Higher concentrations of CaCl2 and MgCl2 caused excessive aggregation of SPI, whereas lower concentrations of CaCl2 and MgCl2 exerted an opposite effect. In the temperature sweep test, CaCl2 and MgCl2 changed the trend of G’ and G”, while KCl and NaCl simply increased G’ and G” without changing their trend. In the steady rheological test, higher concentrations of CaCl2 and MgCl2 reduced the apparent viscosity and shear force of the samples, while an opposite effect was observed under the other salt conditions. The decreasing order of the effects of higher concentrations of salt ions on the apparent viscosity and shear force was K+ > Na+ > Mg2+ > Ca2+. In summary, at specific salt ion concentrations (0.04 and 0.06 mol/L), the changes in gel and rheological properties of SPI were consistent with the Hofmeister effect.

Background/Aims Gastrointestinal bleeding is a significant and potentially lethal event. We aimed to review the efficiency and safety of self-assembling peptides for the treatment and prevention of gastrointestinal tract bleeding. Methods We conducted a systematic search for studies describing the endoscopic use of self-assembling peptides for treatment or prevention of bleeding in the gastrointestinal tract in a parallel, independent fashion. The primary outcomes were rates of successful initial hemostasis, delayed bleeding, and rebleeding. The secondary outcomes were adverse events and ease and volume of gel used. Results Seventeen studies were analyzed. Overall success rate of self-assembling peptides in gastrointestinal bleeding was 87.7% (38%–100%), regardless of etiology or associated treatments. Rebleeding rate ranged from 0% to 16.2%, with a mean of 4.7%, and overall delayed bleeding rate was 5% (range, 0%–15.9%). Only three adverse events were reported in a pooled number of 815 patients. The volume of gel used varied (0.43 to 3.7 mL) according to indication and type of bleeding. Conclusions The limited available data on the use of self-assembling peptides in gastrointestinal endoscopy suggest a high efficiency and good safety profile.

Background and purpose: A wound is a break in the skin. Oral wounds are among the most common reasons for patients to visit a dental clinic, manifesting as lesions with clear boundaries, often slightly sunken, accompanied by or with complete loss of epithelium, covered by a fibrinoleukocytic membrane. Various factors can cause developing wound lesions, including trauma, infection, disease, medication, malignancy, and non-specific causes. Zinc plays a role in cell proliferation, protein synthesis, and wound healing following injury. Given the contradictions and potentials of zinc, and the lack of a study on the effect of zinc on oral wounds, conducting such a study became necessary. This study aimed to investigate the effect of zinc sulfate gel on the healing process of intraoral wounds in rats and compare it with triamcinolone. Materials and methods: This experimental study was conducted in the animal laboratory of the Oral and Dental Diseases Research Center of Kerman University of Medical Sciences. In this study, rats with an approximately 2 x 2 mm wound created using a 2-mm punch in the hard palate were used. The animals were randomly divided into 3 groups (14 rats in each group). Group A (0.1% triamcinolone acetonide gel), Group B (5% zinc sulfate gel), and Group C (placebo) received the respective treatments. Each group was divided into two subgroups based on microscopic evaluation of wound healing: A1, B1, and C1 for one week, and A2, B2, and C2 for two weeks of treatment. The wound sizes were measured along the largest diameter on days 1, 3, 7, and 14 using a Williams periodontal probe. Histological samples were prepared on days 7 and 14 and sent to the pathology laboratory for evaluation. The histopathological criterion for the wounds was based on tissue inflammation. The wounds were also divided into grades 1-5 for re-epithelialization. Finally, the data were analyzed using SPSS 20 software. Results: Regarding the severity of inflammation in the three study groups on days 7 and 14, there was no significant difference (P=0.538). Evaluation of the extent of re-epithelialization in the three study groups on days 7 and 14 showed that the mean extent of re-epithelialization was not significantly different between the triamcinolone and zinc sulfate groups (P=0.18), but the mean extent of re-epithelialization in the triamcinolone group was significantly greater than the placebo group (P=0.001). Also, in terms of wound size on days 1, 3, 7, and 14, the trend of wound size change was the same in the groups, and the mean wound size in the triamcinolone and zinc sulfate groups was less than the placebo group, although the mean of the triamcinolone group was slightly lower, the difference was not significant (P=0.118), indicating that the zinc sulfate group performed as well as the triamcinolone group. Conclusion: The use of zinc sulfate gel resulted in a decrease in inflammation, an increase in re-epithelialization, and a decrease in wound size. Although the triamcinolone gel performed better in reducing inflammation, increasing re-epithelialization, and decreasing wound size, the difference was not significant. The prolonged use of triamcinolone can lead to adverse effects in some oral ulcers, such as candidiasis and hyperpigmentation. Additionally, zinc supplementation has fewer undesirable effects. Therefore, further research is needed to consider replacing triamcinolone with zinc sulfate.

Background: Gingival incision is a procedure widely used in dentistry that can potentially cause infection. Star fruit leaves (Averrhoa bilimbi) extract contains saponins, tannins, flavonoids, and phenols that can act as antibacterials. Shrimpshells chitosan have good biocompatibility, biodegradability, non-toxicity, and antimicrobial properties. Star fruit leaves (A. bilimbi) extract and shrimp shell chitosan are made in gel preparations to accelerate drug delivery to target cells. Purpose: To specify the effect of applying a gel extract of star fruit leaves (A. bilimbi) and chitosan shrimp shells on the formation of new blood vessels in the healing process of gingival incision wounds. Methods: 25 males Rattus norvegicus needed to make an incision in the labial gingiva and were divided into five groups, namely the K-group (without any application), K+ (aloe vera gel application), and three treatment groups PI, PII, and PIII (5%, 10%, and 15% gel extract of Star fruit leaves (A. bilimbi) and shrimp shell chitosan). Topical application is carried out twice a day for seven days. Microscopic observations with hematoxylin-eosin staining were used to count the number of new blood vessels. The analysis used the one-way analysis of variance and the post-hoc Tukey Honest Significant Different method. Results: The treatment in the PII group showed significantly (p < 0.05) higher results than other groups. Conclusion: The formulation of 10% gel Star fruit leaves (A. bilimbi) extract and shrimp shells chitosan has increased blood vessels in the healing process of gingival incision wounds.

Objective: The study aimed to investigate the feasibility of a newly available topical gel in improving the symptoms of various vulvovaginal skin conditions (NCT05396261). Methods: Fifty-two women with diagnosed lichen sclerosus, lichen simplex chronicus, or genitourinary syndrome of menopause participated in this prospective single-arm feasibility study. Consented patients applied the product daily internally and externally to the genital area for approximately 6 months (short-term) and optionally up to 2 years (long-term). Outcome measures included patient-rated symptoms, investigator-assessed clinical signs, and visual severity of pathology of these vulvovaginal conditions. Clinical outcomes, patient adherence to the treatment, and adverse events were assessed, and the statistical analysis was split according to short-term and long-term treatment. Results: The majority of patients enrolled in the study suffered from an uncontrolled disease (90.4%). All patients showed significant improvement in all patient-rated symptoms (p < 0.001), overall clinical signs (p < 0.001), and visual severity of pathology (p < 0.001) short-term. These favorable results were maintained from month 6 up to 2 years. Patient compliance was high, and no adverse events were reported with use of the investigational product. Conclusions: This topical medical device could be an effective symptomatic management option for women suffering from various vulvovaginal conditions.

Abstract Background Following publication of the 2009 World Health Organizations Guidelines for Hand Hygiene in Health Care, a debate has emerged regarding the relative antimicrobial efficacy of the different formats (rinse, gel, foam) of ABHRs and their ability to contribute to reduction of healthcare-associated infections (HAIs). Methods Data regarding the in-vivo antimicrobial efficacy of ABHRs and other factors that likely affect their effectiveness in reducing HAIs were reviewed, and a comprehensive review of studies that reported the effectiveness of each of the three ABHR formats to improve hand hygiene compliance and reduce HAIs was conducted. Results The amount of rubbing time it takes for hands to feel dry (dry time) is the major driver of ABHR antimicrobial efficacy. ABHR format is not a major factor, and several studies found that rinse, gel, and foam ABHRs have comparable in-vivo antimicrobial efficacy. Other factors that likely impact the ability of ABHRs to reduce transmission of healthcare-associated pathogens and HAIs include ABHR formulation, the volume applied to hands, aesthetic characteristics, skin tolerance, acceptance by healthcare personnel, and hand hygiene compliance rates. When accompanied by complementary strategies, promoting the use of each of the three ABHR formats has been associated with improvements in hand hygiene compliance rates. A review of 67 studies failed to identify an ABHR format that was significantly more effective in yielding statistically significant reductions in transmission of healthcare-associated pathogens or HAIs. Conclusions Current evidence is insufficient to definitively determine if one ABHR format is more effective in reducing transmission of healthcare-associated pathogens and HAIs. More rigorous studies such as multicenter randomized controlled trials comparing the different formats are needed to establish if one format is significantly more effective in reducing HAIs.

In order to improve the utilization rate of strawberry tail fruit and expand the product categories of strawberry processing, strawberry pulping was combined with xanthan gum and carrageenan to form a base material suitable for 3D printing. The optimal formula of strawberry mixed gel 3D printing was obtained by using single factor and response surface optimization experiments. The results showed that: The influence of different raw materials on the printing accuracy was in the order of K-carrageenan>strawberry pulp>xanthan gum, and the optimal process formula was strawberry pulp additive amount of 40 g, xanthan gum additive amount of 2 g, K-carrageenan additive amount of 3 g. Under this condition, the printing accuracy of strawberry 3D printing products was 98.39%, and strawberry 3D printing products were most consistent with the set model (cube: The upper side length was 15 mm, the lower side length was 15 mm, the height was 15 mm), at this time, the material could be well extruded and the support capacity was the best.

Tendon injuries, often leading to debilitating adhesions, pose significant challenges in clinical practice. Conventional treatments have limitations, necessitating novel strategies. Injectable hydrogels, known for their biocompatibility, have shown promise. To enhance their antiadhesive properties, researchers have started incorporating drugs. Existing drug delivery systems often peak in the initial days, falling short during the fibroblast proliferation phase which occurs ≈1 week after injury. In this research, CI1040 is selected as an antiadhesion drug, encapsulated within zeolitic imidazolate framework‐8(ZIF‐8), and incorporated into oxidized hyaluronic acid/N‐carboxyethyl chitosan(OHA/CEC) hydrogel(Gel), resulting in the synthesis of CI1040@ZIF@Gel. This unique pH‐responsive drug release system involves encapsulating CI1040 within ZIF‐8, a substance that degrades under acidic conditions while remaining stable in physiological environments. This system selectively releases the drug during the fibroblast proliferation phase, responding to the localized pH reduction post‐tendon injury. CI1040@ZIF@Gel exerted a 65% inhibition on extracellular signal‐regulated kinase (ERK) phosphorylation, reducing the production of collagen types III (Col III) in the adhesion area by 56%. These results indicate that CI1040@ZIF@Gel can effectively inhibit fibroblast proliferation and adhesion through the interleukin 9 receptor/mitogen‐activated protein kinase/extracellular signal‐regulated kinase(IL9r/MEK/ERK) pathway, while also acting as a physical barrier to prevent the formation of tendon adhesion. Objective: To evaluate the efficacy of recombinant bovine basic fibroblast growth factor (rb-bFGF) assisted non-ablative fractional laser therapy for acne scars and its impact on skin barrier function. Methods: 98 patients with acne scars admitted to our hospital from May 2021 to May 2023 were selected and divided into a control group and an observation group based on different treatment methods, with 49 patients in each group. Both groups were treated with non-ablative fractional laser, and the observation group was added with rb-bFGF gel. Evaluate and compare the clinical efficacy, skin barrier function, etc. of the two groups. Results: The edema time, pain time, erythema duration, scab formation time and healing time of the observation group were significantly lower than control group (P<0.05), and the ECCA and VSS scores were also significantly lower than control group (P<0.05). The markedly effective rate of the observation group was significantly higher than control group(79.59% vs. 57.14%, P<0.05). Compared with before treatment, the skin moisture content of the two groups significantly increased and the transepidermal water loss significantly decreased after treatment (P<0.05), while the increase/decrease amplitude of the observation group was significantly greater than control group(P<0.05). After treatment, the QOL Acne scores in the observation group were significantly higher than control group(P<0.05). Conclusion: The efficacy of rb-bFGF assisted non-ablative fractional laser treatment for acne scars is certain, which can accelerate postoperative symptom recovery, improve scar condition and skin barrier function, and improve quality of life. The kelp gel edible granules developed utilizing the gel properties of alginate are prone to quality deterioration if improperly stored during the storage process. This study comprehensively investigated the quality changes of kelp gel edible granules stored at 4 °C and 25 °C by evaluating indicators such as total bacterial count, coliform bacteria, pH, relaxation time, color difference, appearance, texture characteristics, gel strength, and sensory scoring. The results showed that during the storage at 4 °C, the total bacterial count remained within the national standard range, the hardness and chewiness increased, the gel strength first increased and then decreased, the partial exudation of the bound water in the product occurred, and the sensory score slightly decreased, with an overall minor change in quality. During the storage at 25 °C, significant quality changes were observed, with the total bacterial count exceeding the national standard on the 20th day; additionally, the hardness, chewiness, and gel strength all initially increased and then decreased, both the bound water and the restrained water in the product exuded, the moisture stability decreased, and the sensory score significantly decreased between 16 to 20 days. The spoilage of the product was characterized by a significant water loss, reduction in volume, color change from bright green to dark yellow-brown, and a distinct smell of decaying algae. No coliform bacteria was detected in all products during the storage period. In summary, the shelf life endpoint of the product stored at 25 °C is 16 days, and the shelf life of the product stored at 4 °C is greater than 20 days. Background: Gout is a chronic disease caused by the accumulation of uric acid crystals in joints, resulting in sudden pain, stiffness, and swelling. Febuxostat (FBX), a xanthine oxidase inhibitor, is commonly used to treat gout but has poor solubility, which affects its bioavailability. Materials and Methods: To address this issue, a skin-permeating emulgel was developed using Cinnamon (CN) oil as an oil solvent, cremophor RH 40 (Cr. RH 40) as a co-surfactant/solubilizer, and Poloxamer 407 as an emulsifier. A solubility study was performed followed by optimization by 32 factorial designs. The formulation was further evaluated for viscosity, globule size, drug release, and stability studies. Results: Solubility of FBX was found 1 in 10 parts in selected oil. The optimized emulgel containing 10% Cremophor RH 40 and 20% Poloxamer 407 shown improved drug release compared to the pure drug. All other parameters were found satisfactory. Conclusion: The formulated emulgel designed to penetrate the skin shown a good potential for topical formulations enhancing the effectiveness of drug. Moreover, creating a skin-permeating emulsion-based gel in a cost-effective and industrially viable manner could enhance drug penetration and absorption through the skin thereby faster cure of gout compared to the conventional product.
