The goal of the study was to characterize muscle activation/deactivation dynamics across the menstrual cycle in healthy young women. Twenty-two healthy eumenorrheic women (age: 27.0 ± 4.4 years; mean ± SD) were tested every other day for one menstrual cycle. Serum estradiol and progesterone were quantified at the time of testing. Peak torque (PT), time to peak torque (TPT), and half relaxation time (HRT) of soleus muscle twitch were measured. Muscle twitch was elicited by delivering 1ms width electrical pulses to the tibial nerve at an intensity that generated a maximum motor response (S-100) and at supramaximal intensity (S-120; 1.2 x S-100). The analyses were performed for each menstrual cycle phase: 1) the follicular phase to analyze the effect of estradiol while the progesterone concentrations remained at low concentrations; 2) the luteal phase to analyze the effect of progesterone with background estradiol concentrations. In the follicular phase, there was no association of estradiol for PT, TPT, and HRT. In the luteal phase, while estradiol had no association on PT, TPT, and HRT, progesterone expressed a significant association with HRT reduction but no association on PT or TPT. Also, there was a significant estradiol and progesterone interaction for HRT. However, the regression parameters are nearly zero, suggesting that the change in HRT may not have an impact on muscle performance across the menstrual cycle but implications on other women's health conditions with elevated sex hormone concentrations, such as pregnancy, may prove critical. Contractile properties of skeletal muscle are studied for various purposes and mainly by means of force or torque twitch responses. This study compared contractile properties estimated from isometric longitudinal and transversal vastus lateralis twitch mechanical actions using torque and tensiomyography (TMG) as assessment methods, respectively. We calculated delay, contraction, sustain, half relaxation time and peak amplitude from the maximal twitch response obtained by both methods in 19 healthy males (age 46.1±17.8 years). Results indicated a shorter delay (Δ=-23.4%; p<.001) and contraction time (Δ=-42.7%; p<.001) when calculated from the transversal tensiomyographic actions, and shorter half relaxation time (Δ=-26.2%; p=.025) when calculated from the longitudinal torque actions, while no difference in sustain time was found. Delay and contraction time did not correlate significantly when correlated between longitudinal and transversal actions; however, sustain time (r=.478; p=.038) and half relaxation time (r=.608; p=.006) did. In conclusion, the tensiomyography and torque gives different information reflecting that different mechanisms affect longitudinal and transversal twitch skeletal muscle deformations. The purpose of this study was to assess fatigue and recovery of isotonic power and isometric contractile properties after a series of maximal isotonic contractions. Using a Biodex dynamometer, 13 men [26 yr (SD3)] performed isotonic [50% of isometric maximal voluntary contraction (MVC) every 1.2 s through 75° range of motion] single-limb knee extensions at the fastest velocity they could achieve until velocity was reduced by 35%. Time to task failure was 38 s, and, compared with baseline, power declined by ∼42% [741.0 (SD 106.0) vs. 426.5 W (SD 60.3) at task failure], and MVC declined by ∼26% [267.3 (SD 42.5) vs. 198.4 N∼m (SD 45.7) at task failure]. Power recovered by 5 min, whereas MVC did not recover, and at 10 min was only ∼85% of baseline. Isometric MVC motor unit activation was ∼95% at rest and was unchanged at task failure (∼96%), but a small amount of failure was apparent between 1.5 and 10 min of recovery (∼87 to ∼91%). Half relaxation time measured from a 50-Hz isometric tetanus was significantly prolonged by ∼33% immediately after task failure but recovered by 1.5 min. A decline in the 10- to 50-Hz ratio of the evoked isometric contractions was observed at 5 and 10 min of recovery, which suggests excitation-contraction coupling impairment. Changes in velocity and half relaxation time during the protocol were strongly and negatively correlated (r = -0.85). Thus mainly peripheral mechanisms were implicated in the substantial depression but relatively fast recovery of isotonic power. The time courses of changes in parvalbumin (PA) content, isometric twitch tension, and half-relaxation time (1/2 RT) were studied in rabbit tibialis anterior muscle following chronic 10 Hz nerve stimulation of 1-21 days. Up to 5 days stimulation had no effect on PA content, but it induced a slight (10-15%) increase in the 1/2 RT. This change occurred together with the previously observed 50% decrease in Ca-uptake by the SR (Leberer et al. 1987). While prolonged stimulation produced no further decrease in the Ca-uptake by the SR, PA content declined after 5 days of stimulation. The reduction in PA content was accompanied by a progressive lengthening of the 1/2 RT. However, the increase in 1/2 RT was particularly pronounced after PA had fallen below 50% of its normal value. A 90% reduction in PA coincided with a 60% increase in the 1/2 RT. By this time the staircase phenomenon, normally observed in fast-twitch muscle, was completely abolished. Although the changes in PA content and 1/2 RT were not linearly related, these results suggest that PA plays an important role in the relaxation process of mammalian fast-twitch muscle.
