BACKGROUND: One-fifth of the global population is infected with soil-transmitted helminths (STH). Mass drug administration (MDA) with deworming medication is widely implemented to control morbidity associated with STH infections. However, surveillance of human infection prevalence by collecting individual stool samples is time-consuming, costly, often stigmatized, and logistically challenging. Current methods of STH detection are poorly sensitive, particularly in low-intensity and low-prevalence populations. METHODOLOGY/PRINCIPAL FINDINGS: We aimed to develop a sensitive and specific molecular method for detecting STH DNA in large volumes of soil (20 g) by conducting laboratory and proof of concept studies across field sites in Kenya, Benin, and India. We collected human stool (n = 669) and soil (n = 478) from 322 households across the three study sites. We developed protocols for DNA extraction from 20 g of soil and qPCR to detect Ascaris lumbricoides, Trichuris trichiura, Necator americanus, and Ancylostoma duodenale. Agreement between detection of STH via qPCR, digital droplet PCR (ddPCR), and microscopy-based methods was assessed using the Cohens Kappa statistic. Finally, we estimated associations between soil characteristics and detection of STH in soil by qPCR, as well as between STH detected in soil and STH detected in stool from matched households, adjusting for soil characteristics. The overall prevalence of STH in soil by qPCR was 31% for A. lumbricoides, 3% for T. trichiura, and 13% for any hookworm species. ddPCR and qPCR performed similarly. However, there was poor agreement between STH detected in soil by qPCR versus light microscopy. Microscopy underestimated the prevalence of A. lumbricoides and N. americanus and overestimated T. trichiura. Detection of an STH species in household soil was strongly associated with increased odds of a household member being infected with that same species. CONCLUSIONS/SIGNIFICANCE: Soil surveillance for STH has several benefits over stool-based surveillance, including lower cost and higher success rates for sample collection. Considering that delivery of MDA occurs at the community level, environmental surveillance using molecular methods could be a cost-effective alternate strategy for monitoring STH in these populations.

Helminths are considered a significant threat to the livestock industry, as they cause substantial economic losses in small and large ruminant farming. Their morbidity and mortality rates are also increasing day by day as they have zoonotic importance. Anthelmintic drugs have been used for controlling these parasites; unfortunately, due to the development of resistance of these drugs in helminths (parasites), especially in three major classes like benzimidazoles, nicotinic agonists, and macrocyclic lactones, their use is becoming very low. Although new anthelmintics are being developed, the process is time-consuming and costly. As a result, nanoparticles are being explored as an alternative to anthelmintics. Nanoparticles enhance drug effectiveness, drug delivery, and target specificity and have no resistance against parasites. Different types of nanoparticles are used, such as organic (chitosan) and inorganic (gold, silver, zinc oxide, iron oxide, and nickel oxide). One of them, silver nanoparticles (AgNPs), has unique properties in various fields, especially parasitology. AgNPs are synthesized from three primary methods: physical, chemical, and biological. Their primary mechanism of action is causing stress through the production of ROS that destroys cells, organs, proteins, and DNA parasites. Background: Trichuris trichiura is humans' second most prevalent soil-transmitted helminth (STH) infection after Ascaris lumbricoides, affecting approximately 4.6 million people worldwide. Despite its sub-optimal sensitivity, especially in low prevalence and infection intensity settings, the modified Kato-Katz (K-K) is still recommended as a diagnostic method by the World Health organization (WHO) guidelines. Methodology/principal findings: Within a randomized clinical trial (RCT) comprising four treatment arms with two different anthelmintics, the present study reports an important secondary research objective to determine the diagnostic agreement between K-K and real-time PCR evaluating treatment efficacy against T. trichiura. The parasitological results were analyzed, including cure rates (CR) of a subgroup of 94 participants positive at baseline for T. trichiura eggs for both techniques. The single-dose albendazole (ALB) arm resulted in significantly lower CRs than experimental arms of albendazole/ivermectin (ALB/IVM) combinations. The overall diagnostic agreement between both techniques was 88.7% [κ = 0.8 (P<0.001)]. Concordance between eggs per gram and Ct values was moderate, with the discordance source likely stemming from lighter infection intensities. Conclusions and significance: These findings indicate that real-time PCR is a suitable alternative for CR estimation in helminthiasis clinical trials. It also highlights the need to identify the most accurate diagnostic tools for RCTs, that would benefit from guiding principles to achieve harmonization across studies and are not necessarily the same as those used for epidemiological surveys. ABSTRACT Life history theory predicts that a trade‐off may occur when an increased investment in one fitness component causes a reduced investment in another. Parasites generate changes in the optimal life history traits of organisms, causing compromises for their hosts. The objective of this research was to determine whether trematodes Clinostomum sp. and Uvulifer sp. inflict measurable damage on the males of the viviparous fish Poeciliopsis infans, generating trade‐offs between somatic condition and reproductive traits (gonad mass and gonopodium length). We found (i) a negative relationship of large encysted metacercariae and non‐encysted metacercariae on somatic condition. Interestingly, individuals with a higher parasite load had a worse body condition, but bigger gonads and longer gonopodium; and, (ii) a bond between small encysted metacercariae and non‐encysted metacercariae with smaller gonopodia, only in fish with poorer somatic condition. The strongest correlation was given by the non‐encysted metacercariae, probably due to mechanical damage during migration through the body cavity. We did not find any trade‐off effects of Uvulifer sp. We consider that the statistical effects found on somatic condition can be attributed to a greater reproductive investment, generating energetic costs that compromise their defences against infection, allowing a greater parasite load. Background: Wild animals often suffer from infections with multiple species of parasites simultaneously. The exchange of parasites between different host species is common in nature and often involves intermediate hosts or sharing space such as pastures or watering holes. Supplementary feeding, leading to large aggregations of individuals, can have several adverse effects on wild ungulate populations, despite being a widespread management practice. One such effect is an increased risk of parasitic infections, particularly in social animals. We quantified the prevalence of selected helminths typically found in cervids, in samples of the European bison faeces, using molecular methods, and compared endoparasite species richness between supplementarily fed and nonfed European bison herds in the Białowieża Primeval Forest, NE Poland. Methods: Using the diagnostic PCR method, we analysed the faecal samples for molecular markers of nine parasite species which are typically found in cervids: moose, red deer, and roe deer. Results: All analysed samples tested positive for at least one parasite species, and the average number of parasite species per sample was 3.2. The most prevalent parasites were gastrointestinal nematodes: Ostertagia leptospicularis and Ostertagia antipini, found in 89.2% and 50.6% of the European bison faecal samples, respectively. We found significant differences in the prevalence of four parasite species between supplementarily fed and nonfed European bison herds. Co‐occurrence analysis showed that most of the associations between parasite species pairs were random. Conclusion: Management practices, such as supplementary feeding, can influence the spread of parasite infections in social mammals. This study also promotes the application of molecular methods for noninvasive parasitological monitoring of wildlife populations of endangered ruminant species sharing resources with other ungulates. Recent developments in microscopic and molecular tools have allowed the implementation of new approaches for assessing parasitic infections in wildlife populations. This is particularly important for the noninvasive detection and quantification of endoparasites in live animals. Here, we combined copromicroscopic (Mini‐FLOTAC) and molecular (qPCR) techniques to detect the infection of the macroparasite Ligula intestinalis (Cestoda, Pseudophyllidea) in fresh droppings of Gull‐billed Terns Gelochelidon nilotica (Charadriiformes, Laridae) breeding in southwestern Spain. Additionally, we sequenced the cytochrome b gene in parasite isolates from Gull‐billed Terns (definitive host) and Common Bleak Alburnus alburnus (second intermediate host) sampled around tern colonies to explore potential genetic differences between the isolates. The qPCR test showed a higher prevalence (18%; in 13/73 samples) than Mini‐FLOTAC (9%; in 8/88 samples), indicating that qPCR was more sensitive for diagnostic purposes than fecal flotation alone. Although the agreement between both techniques was substantial (84.2%) mainly due to the large number of uninfected samples, only Mini‐FLOTAC allowed us to quantify parasite shedding. When combining techniques, the prevalence of infection did not differ between adults and chicks, suggesting frequent trophic transmission from parents to their offspring via food provisioning. Phylogenetic analyses identified four haplotypes in the isolates from Gull‐billed Terns and Bleak, all of which were placed within a European clade composed of tapeworms recovered exclusively from phylogenetically derived cyprinid fish. This, combined with the short lifespan of mature tapeworms, suggests that Gull‐billed Terns became infected after consuming infected fish around their breeding colonies rather than on their West African wintering grounds. Altogether, our results represent the first record of L. intestinalis in Gull‐billed Terns and the first molecular characterization of the parasite in the Iberian Peninsula. This integrative coprodiagnostic protocol can be applied to other host–parasite systems, allowing researchers to study helminth infections in wild populations using a noninvasive approach. For decades, parasitic worms such as Trichuris muris have been maintained in laboratory animals, providing insights into host–parasite interactions and host immune responses. The most used T. muris isolate is the E isolate, established in the laboratory in 1954. However, one concern with these model systems is the potential for laboratory‐induced selection and therefore changes in host–parasite interactions. To address these concerns, we compare the E isolate with a recently isolated T. muris isolate (M isolate), established from wild house mice (Mus musculus domesticus, Isle of May, UK), in their capacity to infect laboratory mice. High dose infection of C57BL/6 mice revealed that significantly more parasites of the M isolate survived to the adult stage compared to the E isolate. Worm persistence was associated with heightened TNF‐α and IL‐10 secretion upon parasite‐specific re‐stimulation, and higher serum IgG1 and IgG2c levels, concomitant with an increase in T‐bet+ and ICOS+ CD4+ T effector‐memory cells. Differences in host response to the isolates were not as pronounced during low dose infection. Our study highlights the need for regular evaluation of lab‐maintained parasite isolates against freshly isolated parasites to understand whether the established lab strains remain relevant model systems for our understanding of parasitic infections. Pore-forming toxins (PFTs) are released by one cell to directly inflict damage on another cell. Hosts use PFTs, including members of the membrane attack complex/perforin protein family, to fight infections and cancer, while bacteria and parasites deploy PFTs to promote infection. Apicomplexan parasites secrete perforin-like proteins as PFTs to egress from infected cells and traverse tissue barriers. Other protozoa, along with helminth parasites, utilize saposin-like PFTs prospectively for nutrient acquisition during infection. This review discusses seminal and more recent advances in understanding how parasite PFTs promote infection and describes how they are regulated and fulfill their roles without causing parasite self-harm. Although exciting progress has been made in defining mechanisms of pore formation by PFTs, many open questions remain to be addressed to gain additional key insights into these remarkable determinants of parasitic infections. The synaptonemal complex (SC) is a meiotic interface that assembles between parental chromosomes and is essential for gamete formation. While the dimensions and ultrastructure of the SC are conserved across eukaryotes, its protein components are highly divergent. Recently, an unexpected component of the SC has been described in the nematode Caenorhabditis elegans : the Skp1-related proteins SKR-1/2, which are components of the Skp1, Cullin, F-box (SCF) ubiquitin ligase. Here, we find that the role of SKR-1 in the SC is conserved in Pristionchus pacificus. The P. pacificus Skp1 ortholog, Ppa-SKR-1 , colocalizes with other SC proteins throughout meiotic prophase, where it occupies the middle of the SC. Like in C. elegans , the dimerization interface of Ppa-SKR-1 is required for its SC function. A dimerization mutant, ppa-skr-1F105E , fails to assemble SC and produces almost no progeny. Interestingly, the evolutionary trajectory of SKR-1 contrasts with other SC proteins. Unlike most SC proteins, SKR-1 is highly conserved in nematodes. Our results suggest that the structural role of SKR-1 in the SC has been conserved since the common ancestor of C. elegans and P. pacificus , and that rapidly evolving SC proteins have maintained the ability to interact with SKR-1 for at least 100 million years. In Brazil, the wild boar (Sus scrofa) is the main invasive exotic species, responsible for causing various economic, environmental, and health damages to biodiversity. In view of the above, we aimed in our study to investigate the occurrence of endo and ectoparasites in free-ranging wild boars in the northwest region of the state of São Paulo, Brazil. In total, 45 animals of different sexes and ages were examined in this research. Fecal and urine samples were processed using coproparasitological techniques. Multiple incisions in muscles, liver, and pancreas were also made to investigate cestodes and trematodes, and the entire epidermal region of the animals was analyzed for ectoparasites search. In our study, we demonstrate an occurrence of 97.77% (44/45) and 43.33% (13/30) of endo and ectoparasites, respectively, in wild boars, with the identification of eggs of Metastrongylus spp., Strongyloides ransomi, Trichuris suis, Stephanurus dentatus, and nematodes of the order Strongylida, as well as oocysts of Eimeriidae protozoa. Ectoparasite infestations were mainly caused by ixodids of the genus Amblyomma spp. and by larvae of Cochliomyia hominivorax. [ABSTRACT FROM AUTHOR]

In tropical and subtropical regions, soil-transmitted helminth (STH) infections such as Ascaris lumbricoides , Trichuris trichiura , and hookworms have a significant impact on public health. Globally, A. lumbricoides infects approximately 0.8 billion people, while T. trichiura infects around 500 million. This study involved a comparison of three diagnostic methods, Kato-Katz and two flotation methods (concentration McMaster, and simple McMaster), for the detection of Ascaris and Trichuris in human faeces. We conducted a comparison of the number of eggs in faeces (or faecal egg counts (FECs)) obtained with these methods using freshly collected samples that were positive for T. trichiura and spiked with a known quantity of Ascaris sp. eggs. Additionally, for the concentration McMaster method we assessed FECs after storing the samples at 5 °C for up to 21 days. The concentration McMaster method demonstrated superiority over the simple McMaster method in terms of higher detection levels for both helminths, while the Kato-Katz method yielded FEC values very close to the 'true' spiking values. Importantly, the concentration McMaster method was considerably easier to read compared with the Kato-Katz method, and it allowed for sample storage at 5 °C for up to 14 days without affecting FEC results. Consequently, we conclude that the concentration McMaster method is an effective and user-friendly alternative for diagnosis of Ascaris and Trichuris infections in humans. ABSTRACT Histopathological studies of parasitic infections in fish from the natural environment of Brazilian Amazon, are quite scarce. In this study, we investigated the histopathological changes of the proximal intestine of specimens of the Amazonian fish Hoplias malabaricus infected by the hematophagous nematode Procamallanus (Spirocamallanus) paraensis. The prevalence of the infection was 60%, with an average abundance of 1.46 and an average intensity of 2.43 parasites/fish. No significant difference was observed in the prevalence of infection and fish size or sex, but larger fish showed greater infection intensity, which was also significantly higher in male hosts. Histological sections of the proximal intestine showed reduction and loss of the epithelial lining, exposure of the lamina propria where the nematode interacts with the intestine wall, through the insertion of the buccal capsule and fish cellular debris in the intestinal lumen. In addition, areas with bleeding and inflammatory infiltrate were observed, but no changes or presence of parasite structures were observed in the other tunics of the intestinal wall. In this manuscript, we explore the potential therapeutic use of helminths. After analyzing helminths' role in connection with human health from the perspective of their symbiotic and evolutionary relationship, we critically examine some studies on their therapeutic applications. In doing so, we focus on some prominent mechanisms of action and potential benefits, but also on the exaggerations and theoretical and methodological difficulties of such proposals. Simple Summary: This study investigates gastrointestinal helminth infections in synanthropic rodents (house mice and two rat species) from Italy's Emilia-Romagna region, in densely populated areas where these rodents live in close proximity to humans. Conducted between 2019 and 2021, the survey examined 111 rodents captured during pest control programs to identify parasitic worms in their gastrointestinal tracts. The findings revealed that 72.1% of the rodents were infected, with nematodes (roundworms) being the most common. Among the nematodes, Syphacia muris, Aspiculuris tetraptera, Nippostrongylus brasiliensis, and Heterakis spumosa were frequently detected, while tapeworms like Rodentolepis nana and Hymenolepis diminuta were found less often. This study also identified a rare trematode infection in a black rat. Infections with multiple parasite species were common, particularly in black and brown rats, though less frequent in mice. Notably, differences emerged between the detection of adult helminths and their eggs, suggesting that solely relying on necropsy may underestimate prevalence. Combining necropsy with microscopic sedimentation techniques provided a more accurate diagnosis. The presence of human-transmissible parasites emphasizes the importance of integrated rodent control and sanitation efforts to mitigate the risks of parasite transmission to humans, especially in areas where rodents are abundant and sanitation is poor. Synanthropic rodents are species well adapted to coexisting in anthropogenically influenced environments. This coexistence raises concerns about the potential risks of pathogen's transmission due to their close proximity to human habitats. This study presents an epidemiological survey of the gastrointestinal helminth fauna in synanthropic rodents (Mus musculus, Rattus rattus, and Rattus norvegicus) from the Emilia-Romagna Region (Italy), aiming to provide updated data on the endoparasitic populations in these species. A total of 111 rodents, sampled from 2019 to 2021 during pest control programs, were examined for parasitic infections. Helminths were extracted through necropsy and microscopic analysis of gastrointestinal tracts and sediment, with species identification based on morphological characteristics. Overall, 72.1% of the rodents were found to be parasitized, with nematodes being the most prevalent. Syphacia muris, Aspiculuris tetraptera, Nippostrongylus brasiliensis, and Heterakis spumosa were the most frequently identified nematodes. Tapeworms, including Rodentolepis nana and Hymenolepis diminuta, were also detected, albeit in lower frequencies. The trematode Brachylaima recurva was recovered only in one R. rattus. Co-infection was common, particularly among rats, with 51.8% of black rats and 22% of brown rats harboring multiple parasitic species. Mice exhibited lower levels of polyparasitism, with only two individuals showing mixed infections. Interestingly, disparities between the detection of adult helminths and parasitic eggs were noted, especially in cases where no adults were observed, but eggs were found through sediment analysis. These findings suggest that traditional necropsy, especially with poorly preserved carcasses, may underestimate parasite prevalence. This highlights the importance of combining necropsy with microscopic techniques, such as flotation and sedimentation, for a more thorough assessment. Using these methods, nematodes with direct life cycles, such as Syphacia spp., Nippostrongylus brasiliensis, and Heterakis spumosa, have been confirmed as widespread and cosmopolitan among rodent populations. The detection of zoonotic parasites raises concerns about potential transmission to humans, particularly in areas with poor sanitation and high rodent densities. Living organisms are exposed to multiple environmental factors that can affect their fitness. The negative effects of these simultaneous stressors can be additive or can interact in negative synergistic or antagonistic ways to affect the health of exposed individuals. Parasites can accumulate pollutants in their own tissues and have been shown to increase the tolerance of their hosts to different pollutants (antagonistic interaction between parasites and pollutants). Through an experimental approach, we tested the existence of combined antagonistic effects between intestinal parasites and lead exposure on urban feral pigeons (Columba livia) which are known to be exposed to trace metal pollution and harbour a wide variety of internal and external parasites. We experimentally exposed wild feral pigeons in captivity to two treatments: an anthelmintic treatment to eliminate intestinal nematode parasites; an exposure to lead for a period of 6 months. We tested the effects of these crossed treatments on several components of fitness: immunocompetence, reproduction, and body mass. Our findings suggest that the overall effects of lead exposure, either alone or in combination with the presence of intestinal parasites (without anthelmintic treatment) were negative, through either additive or synergistic means. Our results reveal the existence of negative combined effects between pollutant exposure and intestinal parasites, highlighting the importance of accounting for multiple stress factors when studying the effects of exposure to pollutants and/or other environmental stressors on the fitness of organisms. Bacillus thuringiensis crystal (Cry) proteins have been expressed in commercial transgenic crops for nearly 30 years, providing safe and effective control of insect pests and significantly reducing the application of hazardous chemical pesticides. B. thuringiensis crystal proteins have also been shown to target parasitic nematodes, including plant parasitic nematodes. Recently, transgenic soybean crops expressing Cry14Ab have been shown to provide control against the soybean cyst nematode Heterodera glycines, marking the first time a crystal protein is being commercialized in transgenic crops for control of a nematode pest. However, apart from H. glycines and the free-living nematode, Caenornabditis elegans, the breadth of nematode activity of Cry14Ab, e.g., against gastrointestinal parasitic nematodes (GINs), has not been reported. Here we study the efficacy of Cry14Ab against a wide range of gastrointestinal nematode parasites (GINs) in vitro and in vivo. We find that Cry14Ab is effective in vitro against the barber's pole worm Haemonchus contortus larvae, small strongyles cyathostomin larvae, the hookworm Ancylostoma ceylanicum adults, the roundworm Ascaris suum L4 larvae, and the whipworm Trichuris muris adults. In rodents infected with GIN parasites, Cry14Ab is effective as an in vivo anthelmintic against the hookworms A. ceylanicum and N. americanus, against the mouse parasite Heligmosomoides polygyrus bakeri, and against the roundworm A. suum. Cry14Ab also variably reduces the reproduction of the whipworm T. muris in vivo. Using optimized profile Markov Models, we looked for other putative anthelmintic Cry proteins and, within this list, identified a Bt crystal protein, GenBank accession no. MF893203, that we produced and demonstrated intoxicated GINs. This protein, with 90% amino acid identity to Cry14Ab, is active against C. elegans, A. ceylanicum adults, and A. suum L4 larvae in vitro. MF893203 was given the official designation of Cry14Ac. Cry14Ac is also an effective in vivo anthelmintic against A. ceylanicum hookworms in hamsters and intestinal A. suum in mice. Taken together, our results demonstrate that Cry14Ab and Cry14Ac have wide therapeutic utility against GINs. We are exploring a promising new approach using natural Crystal (Cry) proteins from Bacillus thuringiensis (Bt) bacteria, which are already widely used as safe and effective insecticides. To date, we have focused on the anthelmintic (deworming) properties of Bt Cry protein Cry5Ba. Here, our research demonstrates that a newly characterized and different Bt Cry protein, Cry14Ab, can also effectively target and kill various types of harmful parasitic nematodes in both laboratory and animal models. Moreover, we identify another related Bt protein, Cry14Ac, with similar activities against parasitic nematodes. These findings highlight the potential of Bt proteins as a novel class of deworming medications, offering hope for the development and clinical deployment of safer and more effective treatments against parasitic nematode infections. Neurodegenerative diseases such as Alzheimer's disease and polyglutamine diseases are characterized by abnormal accumulation of misfolded proteins, leading to neuronal dysfunction and subsequent neuron death. However, there is a lack of studies that integratemolecular, morphological, and functional analyses in neurodegenerative models to fully characterize these time-dependent processes. In this study, we used C. elegans models expressing Ab1-42 and polyglutamine to investigate early neuronal pathogenic features in olfactory neurons. Both models demonstrated significant reductions in odor sensitivity in AWB and AWC chemosensory neurons as early as day 1 of adulthood, while AWA chemosensory neurons showed no such decline, suggesting cell-type-specific early neuronal dysfunction. At the molecular level, Ab1-42 or Q40 expression caused age-dependent protein aggregation and morphological changes in neurons. By day 6, both models displayed prominent protein aggregates in neuronal cell bodies and neurites. Notably, AWB neurons in both models showed significantly shortened cilia and increased instances of enlarged cilia as early as day 1 of adulthood. Furthermore, AWC neurons expressing Ab1-42 displayed calcium signaling defects, with significantly reduced responses to odor stimuli on day 1, further supporting early behavioral dysfunction. In contrast, AWA neuron did not exhibit reduced calciumresponses, consistent with the absence of detectable decreases in olfactory sensitivity in these neurons. These findings suggest that decreased calcium signaling and dysfunction in specific sensory neuron subtypes are early indicators of neurodegeneration in C. elegans, occurring prior to the formation of visible protein aggregates. We found that the ER unfolded protein response (UPR) is significantly activated in worms expressing Ab1-42. Activation of the AMPK pathway alleviates olfactory defects and reduces fibrillar Ab in these worms. This study underscores the use of C. elegans olfactory neurons as a model to elucidate mechanisms of proteostasis in neurodegenerative diseases and highlights the importance of integrated approaches. Background: Quinic acid (QA), a cyclitol and cyclohexanecarboxylic acid, is a natural product that is present and can be isolated from edible herbals like tea, coffee and several fruits and vegetables. It was previously reported that QA exerted antioxidant and neuroprotective activity against dementia. However, it was not tested for its neuroprotective potential against Huntington's disease (HD). Since aging related disorders are greatly linked to oxidative stress conditions, we focused on testing the oxidative stress resistant activity and protective effect of QA against the development of HD by using the multicellular Caenorhabditis elegans (C. elegans) worm model. Methods: Firstly, QA was tested for its oxidative stress resistant properties. In survival assay, wild type and mutant skn-1 and daf-16 worms were exposed to oxidative stress conditions by using H2O2. Activation of SKN-1 pathway and expression of its downstream genes gcs-1 and gst-4 were also tested. Secondly, the effect of QA was evaluated on HD by testing its ability to decrease the formation of polyQ150 aggregates. Furthermore, its effect on the accumulation of polyglutamine (polyQ35 and polyQ40 aggregates) was tested. Results: Here we report that QA could improve the survival of C. elegans after exposure to oxidative stress caused by H2O2 while also exerting antioxidant effects through the activation of SKN-1/Nrf2 pathway. Moreover, QA could be a potential candidate to protect against HD due to its effects on decreasing the formation of polyQ150, polyQ35 and polyQ40 aggregates. Background: Soil-transmitted helminths (STHs) and mutualistic gut microbes coexist in the gastrointestinal tract. However, limited data exist regarding how STH infections are associated with gut microbiome profiles. Method: We conducted a cross-sectional analysis of baseline data collected in a longitudinal study to identify and explain differences in microbial communities between STH-infected and non-infected Ethiopian school children. We collected 138 stool samples and analyzed them for STH infection using standard direct wet mount and Kato Katz methods. The gut microbiome profiles were characterized using targeted amplicon sequencing of the 16S rRNA gene from the total DNA extracted from the stools. Results: Children infected with Trichuris trichiura showed significantly lower microbial diversity than those who were non-infected (p<0.05). We also observed significant difference in microbiome composition based on Trichuris trichiura infection status (PERMANOVA p< 0.01). A comparison of microbial taxa at the genus level among participants infected with different helminth species showed a significant increase in Agathobacter relative abundance among children infected with Trichuris trichiura compared to non-infected subjects (adjusted p = 0.001). Conclusions: Our results indicate that changes in the gut microbiome composition may vary depending on the species of helminth present. Further studies should investigate how Trichuris trichiura selectively alters microbiome composition compared to other STH species. Objective: Unusual clinical course. Background: Taeniasis is a helminthic infection caused by the Taenia species, specifically T. solium, T. saginata, and T. asiatica. Generally, the parasites infect the small intestine; however, instances of atypical migration have also been reported. Case Report: A 47-year-old Thai woman presented to Nan Hospital with epigastric pain, nausea, and vomiting. Physical examination revealed tenderness in the right upper quadrant, and laboratory analysis revealed mild direct hyperbilirubinemia. Abdominal ultrasonography revealed multiple gallstones and acute cholecystitis. During an elective cholecystectomy, a 70-cm-long tapeworm was found in the gallbladder. Histological examination confirmed the presence of parasitic infection suggestive of Taenia spp., with acute inflammation and focal mucosal necrosis of the gallbladder. Microscopic examination revealed 20 to 22 uterine branches in each of the 5 gravid proglottids, with 42 to 55 uterine twigs and an average ratio of uterine twigs to branches of 2.41, confirming that the parasite in question was possible for T. saginata or T. asiatica. The patient's symptoms resolved after surgery. She was prescribed an oral antibiotic and antiparasitic drug after the operation and experienced no post-surgical complications. Conclusions: Certain parasitic worms can migrate from the intestine to the biliary system. Although less common, T. solium, T. saginata, and T. asiatica can also be detected in the gallbladder and cause acute cholecystitis. This case stresses the potential for misdiagnosis in imaging studies and advises clinicians in endemic areas to consider Taenia spp. infections in the biliary system. [ABSTRACT FROM AUTHOR]

Schistosoma mansoni, an intravascular parasitic worm and the causative agent of schistosomiasis, relies on its tegument (outer layer) for survival and host interaction. This study explored the morphology and mechanical properties of S. mansoni tegument using Atomic Force Microscopy (AFM). Notably, we employed the PeakForce Quantitative Nanomechanical Mapping (PF-QNM) mode in air, enabling simultaneous acquisition of 3D topography and mechanical property contrasts (adhesion, elastic modulus). Additionally, nanoindentation (AFM contact mode) was performed on female worm tegument for elastic modulus measurement. Both techniques revealed an elastic modulus range of fractions or units of GPa for the tegument. Interestingly, mechanical property maps, particularly adhesion contrast, displayed a recurring pattern of light and dark bands. We also measured the depth of annular furrows on the female tegument, finding an average of 128 ± 10 nm. These findings establish AFM, particularly PF-QNM, as a valuable tool to characterize S. mansoni tegument properties, offering insights for future investigations into parasite biology and its response to immunological or pharmacological challenges. Dogs offer multiple benefits in their relationship with humans, but they can also be carriers of zoonotic parasites that affect human and animal health. Zoonoses account for about 58% of all human infectious diseases. The objective of this study was to assess intestinal parasitism in dogs with owners in the City of Barranquilla in the years 2016 to 2018. A retrospective descriptive study was carried out that included 3,279 reports of parasitological evaluation of feces from a clinical laboratory that serves a network of veterinary services in the city of Barranquilla. 49.2 % of the dogs had some type of intestinal parasite. The most frequent were helminths: Strongyloides sp. 9.6 %, Toxocara canis 7.7 % and Ancylostoma caninum 6.2 %; and the protozoa: Entamoeba spp. 10.0 %, Isospora spp. 6.9 % and Giardia spp. 5.7 %. The Principal Component Analysis of the parasite profiles by year showed significant differences. The presence of zoonotically transmitted intestinal parasites in dogs evidenced the need to establish corrective and preventive measures in the field of public health that allow their control, since they constitute a significant risk of disease in the community. This study investigated the role of host traits, habitat, and sampling season on the prevalence and intensity of parasites in three species of platycephalid fishes. Three host species sampled were dusky flathead (Platycephalus fuscus, n = 3), blue-spotted flathead (Platycephalus caeruleopunctatus, n = 38), and tiger flathead (Platycephalus richardsoni, n = 59). A total of 14 metazoan parasite species were collected over 15 months, between July 2020 and September 2021. The parasites found included a chondracanthid copepod, adult hemiuroidean trematodes, cestode plerocercoids, larval and adult acanthocephalans, larval and adult nematodes, and an unknown species of helminth. General linear models were used to assess the relationship between host traits and sampling season with parasite infection intensity. The infection intensity of an unidentified plerocercoid species in P. caeruleopunctatus was found to be significantly associated with both sampling season and the interaction of host mass with sex. In P. richardsoni the infection intensity of the acanthocephalan, Corynosoma sp. was found to correlate with sampling season. The highest richness of parasite taxa was recorded from P. richardsoni, which may be due to the wide depth range of P. richardsoni, coupled with its generalist diet. [ABSTRACT FROM AUTHOR]

This work presents the results of studying the molecular characteristics of parasitic tapeworms Echinococcus canadensis. The helminths were discovered during the autopsy of a wolf (Canis lupus Linnaeus, 1758) killed by hunters in the Kirov oblast in 2021. A molecular phylogenetic study was performed by analyzing the sequence of a fragment of the first subunit of the mitochondrial cytochrome oxidase gene (CoxI). It was found that the detected echinococci belong to the G10 genotype of E. canadensis, which is common in wolves in the northern territories of the Holarctic. We discovered four positions at which the substitutions characteristic only of this genotype are revealed. A substitution at one of the positions that is characteristic exclusively for the representatives of the G10 genotype found in Russia and Finland was also discovered. [ABSTRACT FROM AUTHOR]

This article explores the issue of anthelmintic resistance in Europe and the environmental concerns associated with its use. It suggests sustainable worm control practices, such as diagnostics, grazing management, and selective breeding, as alternatives to reduce the reliance on anthelmintics. The article emphasizes the need for a Community of Practice across Europe to promote sustainable worm control and involve all relevant stakeholders. It provides examples of different approaches to worm control in various European countries, including Ireland, Poland, the United Kingdom, and Greece. The article also discusses the need for improved deworming practices in the ruminant livestock population in Spain, highlighting the lack of registered anthelmintics for goats and the inefficient use of deworming due to the lack of diagnostics. It suggests the use of complementary control approaches and diagnostic tools for effective and sustainable control of helminths. The authors stress the importance of implementing sustainable worm control practices across Europe and propose the establishment of local and national networks, supported by an international stakeholder network, to ensure long-term sustainability. Simple Summary: Camels are one of the important economic animals in China; however, their health is often threatened by helminth. This study investigated the prevalence of helminth in camels by collecting 435 samples from five regions in Xinjiang using two methods. The results showed that 18.2% of the camels were infected with helminths, with the highest prevalence observed in Urumqi. Camels under three years old were found to be more susceptible. This study identified three major parasites (Trichostrongylus spp., Chabertia ovina, and Haemonchus contortus) in camels, and most camels were found to be simultaneously infected with multiple parasites. This research is crucial for formulating effective strategies to prevent and control camel parasites, and it contributes to improving the health and production efficiency of camels. Camels, vital to economies in Asia, Africa, and the Arabian Peninsula, have been domesticated for over 4000 years. They thrive in arid regions like Xinjiang, China, but face challenges from internal and external parasites, particularly gastrointestinal parasites, which impact health, meat and milk quality, and production efficiency. This study investigates the prevalence of gastrointestinal helminth infections in camels from five major regions in Xinjiang. We collected 435 fresh fecal samples and used the saturated saline flotation method and McMaster's method for detection. Molecular examination followed. The overall prevalence was 18.2% (95% confidence interval [CI]: 14.7–22.2%), with Urumqi showing the highest prevalence (29%, 95% CI: 23.4–35.1%) compared to other regions (odds ratio [OR]: 4.62, 95% CI: 2.63–8.41%). Younger camels (≤3 years old) were more likely infected by the parasites after adjusting for the region differences (OR: 10.53, 5.12–24.65%). However, we found no evidence that the prevalence was different between male and female camels. PCR analysis identified Trichostrongylus spp., Chabertia ovina, and Haemonchus contortus as predominant parasites, with observed co-infections indicating a complex parasitic landscape. Because of their host's energetic exhaustion, parasites are among the first species to be influenced by environmental changes. The change in habitats caused by human activities can negatively influence ecosystems, reducing the resistance of host species to parasite attacks. There is little information on the role of urban disturbance in altering diversity and parasitic interactions in aquatic snakes. In this study, the diversity and structure of the parasite community in the water snake Helicops angulatus were investigated. The snakes were collected in three habitat areas (urban, periurban, and forest) in three municipalities on the north coast of the state of Paraíba, Brazil. Forty of the 158 watersnakes analyzed (25.3%) were infected. In total, 137 helminths were collected, representing an overall mean infection intensity of 3.58 ± 2.0. Considering both habitats environments (urban, peri-urban and forest), six Nematoda taxa (Oswaldocruzia mazzai, Brevimulticaecum sp., Eustrongylides sp., Physaloptera sp., Filariidae larva, and an unidentified Nematoda larva), four trematodes (Infidum aff. similis, Travtrema aff. stenocotyle, Heterodiplostomum helicopsis, and encysted metacercariae), two Acanthocephala (Centrorhynchidae gen. sp. and Oligacanthorhynchidae gen. sp.), and one Pentastomida (Sebekia oxycephala) were found, all of which represent the first record for H. angulatus with the exception of S. oxycephala.. The species diversity of endoparasites in H. angulatus was greater in urbanized habitats. Regarding interaction networks, there was low modularity and no nesting. This result may be linked to anthropization, indicating changes in the parasite-host relationship due to urbanization. In the forest areas there was a significant difference in the number of individuals, which was due to a single species of parasite (Physaloptera sp., 21 individuals) where it infected a single female. Physaloptera was the most abundant endoparasite present in the three areas. There were no significant differences in the size and sex of the snakes with parasite infection. This study provides important information about the relationships between parasites, hosts, and the habitat. Background and Aim: Rodents are carriers or reservoirs of various bacteria, protozoa, viruses, and ectoparasites. Given the proximity of various rodent species and humans, there is a potential for the transmission of pathogens. Data on ecto- and endo-parasite prevalence in rodent populations in Gabon are limited. To fill this gap, we conducted a study in Libreville to investigate the occurrence of ecto- and endo-parasites in rodents. Materials and Methods: We captured and euthanized 68 rodents belonging to the genus Praomys and examined their ecto- and endo-parasite fauna, dissected their gastrointestinal tract for helminths, and prepared blood smears to examine blood-borne pathogens. Results: Our analyses identified three pathogen taxa: helminths (Protospirura spp., Trichuris spp., and Taenia spp.), protozoa (Babesia spp.), bacteria (Anaplasma spp.), and arthropods (Laelaps and Haemaphysalis). Overall, 91.2% of the rodents were infected with at least one pathogen and ectoparasite, with helminth and ectoparasite occurrence rate of 63.2% and ectoparasite occurrence at 44.1%. Protozoan infections (Babesia spp.) were found in 10.3% of the rodents, whereas bacteria (Anaplasma spp.) had an occurrence rate of 39.7%. Conclusion: Native rodents in Libreville harbor various infectious agents, ecto- and endo-parasites. These findings highlight the potential health risks associated with Praomys rodents for the transmission of various diseases to human population in Gabon and emphasize the need for investigation of rodents for their role as disease carriers.

Abstract Recent advancements in next-generation sequencing (NGS) technologies have created new opportunities for comprehensive screening of multiple parasite species. In this study, we cloned the 18 S rDNA V9 region of 11 species of intestinal parasites into plasmids. Equal amounts and concentrations of these 11 plasmids were pooled, and amplicon NGS targeting the 18 S rDNA V9 region was performed using the Illumina iSeq 100 platform. A total of 434,849 reads were identified, and all 11 parasite species were detected, although the number of output reads for each parasite varied. The read count ratio, in descending order, was as follows: Clonorchis sinensis, 17.2%; Entamoeba histolytica, 16.7%; Dibothriocephalus latus, 14.4%; Trichuris trichiura, 10.8%; Fasciola hepatica, 8.7%; Necator americanus, 8.5%; Paragonimus westermani, 8.5%; Taenia saginata, 7.1%; Giardia intestinalis, 5.0%; Ascaris lumbricoides, 1.7%; and Enterobius vermicularis, 0.9%. We found that the DNA secondary structures showed a negative association with the number of output reads. Additionally, variations in the amplicon PCR annealing temperature affected the relative abundance of output reads for each parasite. These findings can be applied to improve parasite detection methodologies and ultimately enhance efforts to control and prevent intestinal parasitic infections.

As part of an inventory of the parasitic helminths of rodents, two species of the class Oxyuroidea (Heteroxynema cucullatum Hall, 1916 and Rauschtineria eutamii (Tiner, 1948) Hugot, 1980) were collected from three Cliff Chipmunk specimens (Neotamias dorsalis (Baird, 1955)) in the Área de Protección de Flora y Fauna Cerro del Mohinora, Mexico. These nematodes have been recorded in different species of the genus Neotamias (Illiger, 1811), which can be found in sympatry mainly along the entire west coast of the USA. This is the first record of both species in Mexico.

Abstract Background Recent articles have explored the effect of worms on cancer cells. This review focused on various cell cultures employed to understand which cells are more commonly and less utilized. Methods The present review analyzed studies published between 2013 and 2023 to obtain information about different cell cultures used in cancer studies involving helminths. Databases such as PubMed, Google Scholar, HINARI, and the Cochrane Library were searched. Results This search yielded 130 records, but 97 papers were excluded because they were either irrelevant to the research topic (n = 72) or contradicted the research idea (n = 25).The remaining twenty-one articles focused on different types of worms, such as Echinococcus granulosus, Clonorchis sinensis, Opisthorchis felineus, Opisthorchis viverrini, Trichinella spiralis, Toxocara canis, and Heligmosomoides polygyrus. Conclusion Due to the presence of numerous antigens, parasites at different growth stages can impact various cells through unknown mechanisms. Given the high diversity of antigens and their effects, artificial intelligence can assist in predicting initial outcomes for future studies.

Abstract The subsistence economies of prehistoric pastoralists of the Eurasian steppes have long been viewed through an oversimplified model of reliance on domesticated animals. This conceptualization has begun to shift dramatically through the introduction of scientific analyses, pivoting towards an evidence-based interpretation of economic flexibility and adaptive heterogeneity. Here we provide insights into the dietary practices of Iron Age pastoralists in Siberia through an archaeoparasitological analysis. Soil samples from the Tunnug 1 site in southern Siberia reveal the presence of helminth eggs of Taenia sp. (likely), Trichuris sp., and Dibothriocephalus sp. This indicates that the diet of the analysed prehistoric population might have included beef and did include freshwater fish, occasionally consumed in undercooked or raw form. Despite the primary reliance on pastoralism and possibly small-scale millet agriculture, these populations engaged in diverse dietary practices, including fish consumption. Additionally, the presence of Trichuris sp. eggs points to poor sanitary conditions, possible consumption of contaminated plant foods, and the contamination of drinking water with feces. By providing direct evidence of dietary habits, archaeoparasitology complements isotopic analyses and contributes to a more nuanced understanding of the subsistence strategies.

Helminths have developed intricate mechanisms to survive and evade the host’s immune responses. Hence, understanding the excretory-secretory products (ESPs) by helminths is crucial for developing control tools, including drug targets, vaccines, and potential therapies for inflammatory and metabolic disorders caused by them. Proteomics, the large-scale analysis of proteins, offers a powerful approach to unravel the complex proteomes of helminths and gain insights into their biology. Proteomics, as a science that delves into the functions of proteins, has the potential to revolutionize clinical therapies against parasitic infections that have developed anthelminthic resistance. Proteomic technologies lay a framework for accompanying genomic, reverse genetics, and pharmacokinetic approaches to provide more profound or broader coverage of the cellular mechanisms that underlie the response to anthelmintics. With the development of vaccines against helminth infections, proteomics has brought a major change to parasitology. The proteome of helminths can be analyzed comprehensively, revealing the complex network of proteins that enable parasite survival and pathogenicity. Furthermore, it reveals how parasites interact with hosts’ immune systems. The current article reviews the latest advancements in helminth proteomics and highlights their valuable contributions to the search for anthelminthic vaccines.

The purpose of the research is to extend information across the distribution of zooanthroponotic helminth infections transmitted through freshwater fish in the Magadan Region.Materials and methods. The study area is located in the Upper Kolyma basin on the Magadan Region and the Republic of Sakha (Yakutia) border. The Omulevsky Park is also located here, one of the clusters of the Chersky National Park named after A. V. Andreev. The existing lake and river systems (Darpir, Momontai, Malyk, Urultun and Ui) are inhabited by 7 fish species: Arctic char (Salvelinus alpinus), east-Siberian grayling (Thymallus arcticus pallasi), round fish (Prosopium cylindraceum), Siberian sucker (Catostomus catostomus rostratus), burbot (Lota lota leptura), common minnow (Phoxinus phoxinus) and sculpin from the Kolyma (Cottus kolymensis). 685 specimens of fish were undergone parasitological survey using established procedures. The found helminths were fixed in 70% ethanol and clarified in glycerol.Results and discussion. In the study of the fish helminth fauna in the lake and river systems of the Upper Kolyma Highlands, three helminth species that posed a danger to humans and animals were identified, Dibothriocephalus dendriticus, D. ditremus (Diphyllobothriidae) and Eustrongylides excisus (Dioctophymatidae). Information was provided on fish infection with these parasites in the lake and river systems, and their distribution in the Magadan Region, and a brief description of these types of helminths was given. In domestic scientific and methodological publications, the problem of eustrongylidiasis as a zooanthroponotic disease is not sufficiently covered, therefore, emphasis is placed on nematodes of the genus Eustrongylides. The research results may attract the attention of sanitary and veterinary service specialists to this issue, especially given the creation of a national park in the Magadan Region. Data on the freshwater fish infection with Diphyllobothriidae in the Upper Kolyma Lake and river systems expand the diphyllobothriasis nosoarea in the Magadan Region and allows the Susumansky District to be included in its boundaries.

The purpose of the research is to study the parasite fauna in laboratory rats and mice in SPF-status and open-type vivariums and to compare the infection rate depending on the maintenance type within each type of samples.Materials and methods. The studies were conducted using life-time diagnosis (flotation method and Scotch-tape test), and the material were fecal samples from laboratory rodents and litter samples. Ten percent of the vivarium population and all newly arrived mice and rats in quarantine were examined. Fifty-four individual samples from pure line mice, and 24 litter samples were examined. A total of 234 samples were collected from the laboratory rats: 93 individual samples, 55 combined samples, and 17 sawdust samples; 69 samples were examined by Scotch-tape test.Results and discussion. Twenty six percent of the laboratory SPF vivarium mice were found to have protozoa: Giardia muris (11.1%) and Trichomonas sp. (20.4%). Nematodes Aspiculuris tetraptera (51.0%) and Syphacia obvelata (20.6%), and cestode Rodentolepis nana (12.0%) were recorded in the open-type vivarium mice. The laboratory open-type vivarium rats were found to have nematodes S. muris (up to 60.9%), A. tetraptera (5.4%), Trichosomoides crassicauda (1.8%), cestode R. nana (27.3%), protozoa Eimeria sp. (7.2%) and Giardia sp. (9.0%). The compared infection of individual with combined mouse fecal samples showed statistically significant differences for all parasites in general and for individual species. Pairwise comparisons of the infection showed that the combined sample detected R. nana statistically significantly more often versus the individual sample (27.3 vs. 5.4%, P < 0.001). The comparison of the infection depending on the type of rat maintenance within each sample type did not show statistically significant differences.

Abstract Background Tuberculosis (TB) and intestinal helminths are diseases that pose a dual burden on public health in low-income countries. Previous studies have shown that helminths can affect the shedding of bacteria or the bacterial load in the sputum of active TB patients. However, there is limited information on bacterial load in TB patients with helminth infections. Objective This study aimed to compare bacterial load in helminths-infected and non-infected pulmonary tuberculosis patients at selected public health facilities in Jimma zone, Oromia, Ethiopia. Methods The study was conducted in Jimma Zone, Oromia, Ethiopia. A facility-based comparative cross-sectional study was employed from August 01, 2020, to January 2021. A total of 124 (55 intestinal helminths-infected and 69 non-infected) newly diagnosed smear-positive pulmonary tuberculosis (PTB) patients were included in the study. A convenience sampling technique was employed to recruit study participants, and a semi-structured questionnaire was used to collect data regarding socio-demographic characteristics and possible risk factors for intestinal helminths co-infection. Stool examination was performed using both wet mount and Kato Katz technique. Additionally, weight and height measurements, sputum, and blood samples were taken to determine body mass index, bacilli load, and diabetic mellitus, respectively. Data were entered into Epi-Data software version 3.1 and analyzed using Statistical Packages for Social Sciences (SPSS) Version 25. A statistically significant difference was defined as a P-value of less than 0.05. Results Intestinal helminths reduced bacilli load 3 times more than intestinal helminths non-infected PTB (AOR = 3.44; 95% CI; 1.52, 7.79; P = 0.003) However, diabetes mellitus, HIV, drinking alcohol and cigarette smoking were not associated with bacilli load. The rate of co-infection TB with intestinal helminths was 44%. The three most prevalent parasites detected were Trichuris trichiura 29 (66%), hookworm 19 (43%), and Ascaris lumbricoides 11(25%)). In recent years, air sac parasitic helminths have been reported to cause severe disease in birds. In addition, various species appear to be expanding and infecting new avian hosts in various regions worldwide. In this context, an air sac nematode was initially detected in 2014 infecting the Eurasian scops owl, hospitalized in the local wildlife hospital in Mallorca (Balearic Islands, Spain). Years later, the parasite was detected in 2 other owl species. Air sac nematodes had never been reported in the Mallorcan Strigiformes before. A comprehensive molecular and morphological characterization analysis, including scanning electron microscopy, was required for species confirmation. The species was identified as Lissonema sicki, a parasite infrequently reported in South American owls. Since its first introduction to Mallorca, it has dramatically increased in prevalence in hospitalized birds, being highly prevalent in the Eurasian scops owl (41%), in the long-eared owl (11%) and in the barn owl (4%). The introduction pathway of this parasite to Europe remains unknown. This discovery underscores the expanding range and impact of L. sicki, emphasizing the importance of ongoing surveillance and research to comprehend and manage the implications of its emergence in new territories.

The aim of this study was to evaluate the isolated and associated use of the nematophagous fungi Pochonia chlamydosporia (isolate VC4) as an ovicide and Duddingtonia flagrans (isolate AC001) as a predator in the control of Taenia saginata eggs. Viable T. saginata eggs were obtained by dissecting mature proglottids from a specimen. For the experimental assay, four groups were formed in microtubes, as follows: (G1—T. saginata eggs/control); (G2—100 T. saginata eggs + VC4); (G3—100 T. saginata eggs + AC001); (G4—100 T. saginata eggs + VC4 + AC001). All the groups were stored in a B.O.D. incubation chamber at a temperature of 27 °C for 15 days and then the contents of the microtubes were analyzed using an optical microscope with a 40x objective. At the end of the assay the treated groups (G2 to G4) showed ovicidal activity (destruction of eggs) compared to the control group (G1). The highest ovicidal percentage was observed in group G2 (eggs + VC4), with 43.3%. In groups G3 and G4 (combination of fungal isolates), the ovicidal percentages were 25.7% and 25.6%, respectively. The results of this study shed light on a new possibility for the combined use of different species of nematophagous fungi, which could be used in the future for environmental biological control of T. saginata eggs.

Biological ageing refers to the gradual decrease in physiological functions, resulting in immune senescence, cellular damage and apoptosis. Telomere length is a biomarker of biological ageing. Limited studies have associated shorter telomere length with HIV and parasite single infections, with no studies reporting the association of HIV and parasite co-infection with telomere length. The study aimed to investigate whether telomere length shortening is accelerated in a South African population co-infected with HIV and helminths compared to participants singly infected with either HIV or helminths. Additionally, telomere length data were compared with participants’ biochemical and full blood count parameters. A total of 200 participants were in groups of uninfected control, HIV single infection, helminth single infection and HIV and helminth co-infection groups. Relative telomere length (RTL) was determined using Real-Time PCR and associated with biochemical and full blood count parameters using multivariate regression analysis models that were adjusted for confounders. The uninfected control group was used as a reference group. The uninfected control group had the highest mean RTL (1.21 ± 0.53) while the HIV-infected (0.96 ± 0.42) and co-infected (0.93 ± 0.41) groups had similar RTLs, and lastly, the helminth-infected group (0.83 ± 0.33) had the lowest RTL (p = 0.0002). When compared to the uninfected control group, a significant association between RTL and biochemical parameters, including blood iron (β = −0.48), ferritin (β = −0.48), transferrin saturation (β = −0.57), transferrin (β = −0.57), phosphate (β = −0.47), vitamin A (β = −0.49) and C-reactive protein (β = −0.52) were noted in the co-infected group (p < 0.05). In addition, a significant association between RTL and full blood count, including (β = −0.47), haematocrit (β = −0.46), mean corpuscular volume (β = −0.47), lymphocytes (β = −0.45), mean corpuscular haemoglobin concentration (β = −0.45), red cell distribution width (β = −0.47), monocytes (β = −0.45), eosinophils (β = −0.45), basophils (β = −0.44) and transferrin saturation (β = −0.57) were also noted in the co-infected group (p < 0.05). Accelerated biological ageing, as indicated by telomere length shortening, is associated with HIV and helminth co-infections.

Background: Trichinella spiralis (T. spiralis) is a parasitic helminth that causes a globally prevalent neglected zoonotic disease, and worms at different developmental stages (muscle larvae, adult worms, newborn larvae) induce immune attack at different infection sites, causing serious harm to host health. Several innate immune cells release extracellular traps (ETs) to entrap and kill most pathogens that invade the body. In response, some unicellular pathogens have evolved a strategy to escape capture by ETs through the secretion of nucleases, but few related studies have investigated multicellular helminths. Results: In the present study, we observed that ETs from neutrophils capture adult worms of T. spiralis, while ETs from macrophages trap muscle larvae and newborn larvae, and ETs had a killing effect on parasites in vitro. To defend against this immune attack, T. spiralis secretes plancitoxin-1, a DNase II-like protein, to degrade ETs and escape capture, which is essential for the survival of T. spiralis in the host. Conclusions: In summary, these findings demonstrate that T. spiralis escapes ET-mediated capture by secreting deoxyribonuclease as a potential conserved immune evasion mechanism, and plancitoxin-1 could be used as a potential vaccine candidate. Intestinal parasitic infections (IPIs) remain a significant contributor to morbidity and mortality globally, particularly in developing countries such as Ethiopia. Periodic assessments of IPI prevalence are essential prerequisite for effective control measures. Therefore, this cross‐sectional study is aimed at determining the prevalence of gastrointestinal parasitic infections and associated risk factors among schoolchildren at Wonji Shoa Secondary School, East Shoa Zone, Adama district, Oromia region, Ethiopia, between January and June 2022. A simple random stratified sampling technique was employed to select participants. Sociodemographic and risk factor data were collected using a structured questionnaire. Stool samples were examined to identify parasites. Data were analyzed using SPSS version 20. Descriptive statistics, chi‐square tests, and logistic regression were conducted to assess associations between variables and then the strength of the association. A p value < 0.05 was considered statistically significant. Of the 403 selected students, 330 completed the study that makes 81.89% response success. The overall IPI prevalence was 16.66% (55/330), with a higher prevalence among males (60%, 33/55) than females (40%, 22/55). Five parasite species were identified: two protozoa (Entamoeba histolytica and Giardia lamblia) with a combined prevalence of 9.70% (32/330) and three helminths (Ascaris lumbricoides, Hymenolepis nana, and Taenia spp.) with a combined prevalence of 6.97% (23/330). Cysts were detected in 62.5% of E. histolytica cases (15/24), and eggs were detected in 76.92% of A. lumbricoides cases (10/13). The study revealed a substantial IPI prevalence (16.66%) among the students. This finding underscores the need for effective prevention and control strategies. The predominance of parasitic infections among males is notable requiring further investigation of the factors. The identification of multiple parasite species indicates a complex epidemiological scenario. Recently a number of broad-range stool parasite PCR assays have been developed. However, there is ongoing disagreement regarding their diagnostic performance, as various studies have produced contradictory results. In this study, we compared the diagnostic accuracy of the Seegene Allplex GI-Parasite and Allplex GI-Helminth assays (SA) with the conventional methods used at the travel clinic of the Institute of Tropical Medicine (ITM) including microscopy, antigen testing, and molecular detection in order to provide insights into the strengths and limitations of this diagnostic tool which may be crucial to select the most appropriate diagnostic tools for the suspected pathogen. A total of 97 native stool samples from 95 patients with suspected gastrointestinal illness were analyzed, including 26 from a frozen collection and 71 prospectively collected samples. The diagnostic performance of SA was notably superior to the conventional workflow in detecting Dientamoeba fragilis (sensitivity 100% vs. 47.4%) and Blastocystis hominis (sensitivity 95% vs. 77.5%). SA had a comparable performance with the conventional workflow in detecting pathogenic protozoa (sensitivity 90% vs. 95%). In contrast, SA had a much lower diagnostic performance in detecting helminths (59.1%) compared to the conventional workflow (100%). We conclude that the Seegene Allplex GI-Parasite assay may be useful for protozoa screening in low-endemic industrialized countries. Simple Summary: Heterakis spumosa Schneider, 1886 is a parasite commonly found in rodents, primarily associated with the black rat Rattus rattus and the brown rat R. norvegicus, two global invasive rodent species, and the house mouse Mus musculus. This parasite originated probably from Asia, and has been reported in various rodent species, but molecular information from its putative native region is still lacking. Our sampling efforts across Southeast Asia allowed us to collect and analyze this species from two localities in Lao PDR and five localities in Thailand. Additionally, specimens from Europe were included in the analysis. All specimens were analyzed using mtCOI gene and nuclear ribosomal ITS1. Our findings revealed the presence of two distinct clades of H. spumosa, with no discernible association with hosts and geographical localities. Heterakis spumosa, a parasitic worm infecting rodents, is globally prevalent in black rats, brown rats, and house mice. It is hypothesized to originate from Asia due to its widespread presence in Southeast Asia in various Murinae. Previous molecular studies focused on European, African, and Japanese specimens, but none included samples from the putative native range. Rodents were collected between 2008 and 2015 across various localities in Southeast Asia and Europe, identified by morphology or genetic barcoding. Viscera were examined or preserved for later inspection. DNA was extracted from H. spumosa. PCR amplification targeting the mtCOI gene and ITS1 region was conducted in this study using newly designed primers (based on Heterakis reference sequences). PCR amplicons were subsequently sequenced and analyzed. In this study, the phylogenetic analysis using ITS1 sequences revealed that Heterakis samples from Thai and Laotian rodents belong to the species H. spumosa, exhibiting low genetic variation compared to samples from other regions. Genetic distance calculations using mtCOI sequences confirmed the marked distinction of H. spumosa from other Heterakis species. Our phylogenetic analyses using partial mtCOI and ITS1 sequences have significantly enhanced our comprehension of the genetic diversity and evolutionary history of the nematode H. spumosa. Background: Soil-transmitted helminths infect an estimated 18% of the world's population, causing a significant health burden. Microscopy has been the primary tool for diagnosing eggs from fecal samples, but its sensitivity drops in low-prevalence settings. Quantitative real-time polymerase chain reaction (qPCR) is slowly increasing in research and clinical settings. However, there is still no consensus on preferred qPCR targets. Methods: We aimed to compare soil-transmitted helminth (STH) DNA detection methods by testing naïve stool samples spiked with known quantities of STH eggs and larvae. DNA extracts from spiked samples were tested using independent quantitative realtime PCR (qPCR) assays targeting ribosomal or putative non-protein coding satellite sequences. Results: For Trichuris trichiura, there was a strong correlation between egg/larvae counts and qPCR results using either qPCR method (0.86 and 0.87, respectively). Strong correlations also existed for A. lumbricoides (0.60 and 0.63, respectively), but weaker correlations were found for Ancylostoma duodenale (0.41 for both assays) and Strongyloides stercoralis (0.48 and 0.65, respectively). No correlation for Necator americanus was observed when testing with either qPCR assay. Both assays had fair-to-moderate agreement across targets when using field-collected stool samples (0.28–0.45, for all STHs), except for S. stercoralis (0.12) with slight agreement. Conclusions: There is a strong correlation between qPCR results and egg/larvae counts. Our study confirms that qPCR is an effective diagnostic tool, even with low-intensity infections, regardless of the DNA-based diagnostic marker used. However, the moderate agreement between the two different qPCR assays when testing field samples highlights the need to understand the role of these targets in the genome so that the parasite burden can be quantified more accurately and consistently by qPCR. Background: Variations in vaccine responses have been observed between populations. A role for helminth infections has been proposed due to their immunomodulatory properties. In a secondary analysis of data from a randomised trial assessing effects of anthelminthic treatment on vaccine responses, we examined associations between helminth infections at baseline prior to vaccine administration, and vaccine responses among adolescents (9-17 years) in Koome Islands, Lake Victoria, Uganda. Methods: Participants received BCG [week 0], yellow fever (YF-17D), oral typhoid (Ty21a), HPV-prime [week 4], and HPV-boost, tetanus/diphtheria [week 28]. Outcomes were BCG-specific interferon-g ELISpot responses and antibody responses to yellow-fever-, typhoid-, HPV-, tetanus- and diphtheria-specific antigens measured at two time points post vaccination. S. mansoni infection was determined as positive if either the plasma Circulating Anodic Antigen (CAA) assay or stool PCR were positive. Hookworm and Strongyloides were determined by stool PCR. Linear mixed effects regression was used to assess associations. Results: Among 478 adolescents, 70% were Schistosoma mansoni (Sm) infected and 23% hookworm infected at baseline. Sm was associated with lower Salmonella Typhi O:LPS-specific IgG responses (adjusted geometric mean ratio (aGMR) 0.69 (0.57-0.83)), and hookworm with higher diphtheria-specific IgG (aGMR 1.16 (1.02, 1.31)) and lower HPV-16-specific IgG (aGMR 0.70 (0.55, 0.90)) post-vaccination. High Sm intensity was associated with lower BCG-specific interferon-g and S. Typhi O:LPS-specific IgG. Conclusions: We found inverse associations between Sm and responses to two live vaccines, whereas hookworm was positively associated with diphtheriaspecific IgG. These findings support the hypothesis that helminth infections can modulate vaccine responses, while also highlighting potential heterogeneity in the direction of these effects. The previously underestimated biodiversity of the Caatinga underwent a transformation with the emergence of new studies, intensifying interest in the biota of this complex dry forest. Studies on snakes from the Caatinga have increased recently, resulting in 112 recorded species. Contributing to this, we present a detailed list of 23 species rescued from the Itaparica reservoir implementation in the 1980s. Collected specimens are deposited in the Herpetological and Paleoherpetological Collection at the Universidade Federal Rural de Pernambuco. The recorded snake assemblage is typical of the dry Caatingas of the Sertaneja Depression. We discuss the impacts of flooding and specimen relocation without adequate planning or monitoring on snake diversity. As part of an inventory of the parasitic helminths of rodents, two species of the class Oxyuroidea (Heteroxynema cucullatum Hall, 1916 and Rauschtineria eutamii (Tiner, 1948) Hugot, 1980) were collected from three Cliff Chipmunk specimens (Neotamias dorsalis (Baird, 1955)) in the Área de Protección de Flora y Fauna Cerro del Mohinora, Mexico. These nematodes have been recorded in different species of the genus Neotamias (Illiger, 1811), which can be found in sympatry mainly along the entire west coast of the USA. Aims: Protozoan and helminth parasitic infections pose significant public health challenges, especially in developing countries with rural populations marked by suboptimal hygiene practices and socio‐economic constraints. The parasites are the etiological agents of these infections and have a notably elevated global prevalence. Therefore, this study focuses on estimating the frequency and transmission dynamics of several parasitic species, including Blastocystis, Giardia, Cryptosporidium spp., Entamoeba histolytica, Ascaris lumbricoides, Trichuris trichiura, Taenia spp. and hookworms, within a rural community in southwest Colombia with a particular emphasis on the One Health framework, considering environmental and zoonotic transmission potentials. Methods and Results: This study involved the analysis of 125 samples, encompassing human participants (n = 99), their domestic pets (dogs) (n = 24) and water sources (n = 2). Parasite detection was carried out utilizing a combination of microscopy and molecular techniques. Furthermore, the characterization of Blastocystis subtypes (STs) was achieved through Oxford Nanopore sequencing of the rRNA‐18S gene. The investigation also entailed the examination of potential associations between intestinal parasitism and various sociodemographic factors. Results revealed a high frequency of parasitic infections when employing molecular methods, with Blastocystis (n = 109/87%), Giardia (n = 20/16%), Ancylostoma duodenale (n = 28/22%), Ancylostoma ceylanicum (n = 7/5.6%), E. histolytica (n = 6/4.8%), Cryptosporidium spp. (n = 12/9.6%) and even Taenia (n = 1/0.8%) detected. Cryptosporidium spp. was also identified in water samples. Coinfections were prevalent, with 57% (n = 70) of samples exhibiting single‐parasite infections and 43% (n = 53) showing various degrees of polyparasitism, emphasizing the complexity of transmission dynamics. Blastocystis subtyping, conducted via Oxford Nanopore sequencing, revealed a diversity of subtypes and coexistence patterns, with ST2 being the most prevalent. Conclusions: This research underscores the importance of using molecular techniques for frequency estimation, particularly emphasizing the relevance of zoonotic transmission in parasitic infections. It highlights the significance of the One Health approach in comprehending the circulation of parasites among animals, humans and environmental sources, thereby directly impacting public health and epidemiological surveillance.
