The skin wound model in rats is a fundamental stage in preclinical trials, but there is a lack of standardization in these trials regarding the initial wound area, making analysis and comparison between studies difficult. Therefore, this study evaluates the healing progression of excisional skin lesions of varying diameters in Wistar rats, aiming to identify the optimal wound size for monitoring treatment effects on wound healing. Excisions of 0.8, 1.5, 2.0 and 3.0 cm in diameter were made on the back of the animals. Thirty animals were used per treatment and evaluated on days 3, 7, 10, 14 and 21 after surgery. The lesions were cleaned daily with saline solution until they were completely closed. The 0.8 cm group showed complete repair on D14, while in the other groups, the wounds persisted until day 21, with a reddened surface and no complete epidermal coverage, but with greater keratinization and presence of appendages in the 1.5 cm lesions. Therefore, as a standardization model for creating skin wounds, we suggest using 1.5 or 2.0 cm excisions, considering that 0.8 cm wounds close very early and 3.0 cm wounds, although behaving similarly to 2.0 cm wounds, are more invasive for the animals. The 1.5 cm model proved to be suitable for closure within 21 days. Simple Summary: Squilla mantis, also known as spot tail mantis shrimp, is a stomatopod crustacean widely distributed in the Mediterranean Sea and the adjacent Atlantic Ocean. In Italy, S. mantis is the main crustacean species targeted by commercial catches, and in particular, the Adriatic landings represented about 65% of the total national catches in 2021. Despite the economic, ecological, and nutritional importance of S. mantis, studies on reproductive biology are scarce or not updated in the Mediterranean Sea, particularly in the Adriatic Sea. In light of the above, the aim of this study was to describe the reproductive aspects of spot tail mantis shrimp, contributing to enriching the knowledge of S. mantis reproductive biology. Therefore, our findings could be considered an important tool for correct fisheries management and sustainable exploitation of this commercially fished resource in the Adriatic Sea. Fisheries management requires improvement in scientific knowledge to ensure sustainable exploitation of important commercial species and population renewal. Within this context, this study focused on the reproductive biology of spot tail mantis shrimp, Squilla mantis, in the Central Mediterranean Sea, aiming to understand the reproductive pattern of this species and validate the macroscopic maturity scale through histological analysis. A multi-year sampling was performed from 2016 to 2020 by a commercial fishing fleet in the Northern Central Adriatic Sea (GSA 17), and a total of 2206 individuals were collected. The monthly average value of the total sex ratio of S. mantis was 0.524 ± 0.044 (mean ± SEM) in favor of females, which dominated the population from September to April. The proposed 5 stage macroscopic maturity scale was validated histologically through histological analysis, confirming synchronous ovarian development. The somatic indexes (GSI and K Fulton) and monthly incidence of macroscopic ovarian maturity stages highlighted a protracted reproductive season from winter to spring (January–May). Although the length-weight relationship showed a similar growth trend between genders, males reached a bigger size in terms of carapace length (C.L.) and dominated the population from 32 mm (C.L.). Mechanical mismatch between native aortas and aortic grafts can induce graft failure. This study aims to compare the mechanical and microstructural properties of different graft materials used in aortic repair surgeries with those of normal and dissected human ascending aortas. Five types of materials including normal aorta (n = 10), dissected aorta (n = 6), human pericardium (n = 8), bovine pericardium (n = 8) and Dacron graft (n = 5) were collected to perform uniaxial tensile testing to determine their material stiffness, and ultimate strength/stretch. The elastin and collagen contents in four tissue groups except for Dacron were quantified by histological examinations, while the material ultrastructure of five material groups was visualized by scanning electron microscope. Statistical results showed that three graft materials including Dacron, human pericardium and bovine pericardium had significantly higher ultimate strength and stiffness than both normal and dissected aortas. Human and bovine pericardia had significantly lower ultimate stretch than native aortas. Histological examinations revealed that normal and diseased aortic tissues had a significantly higher content of elastic fiber than two pericardial tissues, but less collagen fiber content. All four tissue groups exhibited lamellar fiber ultrastructure, with aortic tissues possessing thinner lamella. Dacron was composed of densely coalesced polyethylene terephthalate fibers in thick bundles. Aortic graft materials with denser fiber ultrastructure and/or higher content of collagen fiber than native aortic tissues, exhibited higher ultimate strength and stiffness. The aim of this study was to verify the effectiveness of attenuated total reflectance‐fourier transform infrared (ATR‐FTIR) spectroscopy in the characterization of bone repair in mandibular osteotomy using erbium, chromium‐doped yttrium, scandium, gallium and garnet (Er,Cr:YSGG) laser and multilaminate drill on each side. Two mandible bone fragments were removed from 30 rabbits, and the process of bone repair was studied immediately, 3, 7, 15, 21, and 28 days after the surgery. The histological analysis allowed detecting differences in the early stages of tissue repair after bone cutting performed with the Er,Cr:YSGG laser or multilaminate drill. The science of dental tissue grafting is evolving, with an increased understanding of factors influencing graft behavior. Despite the widespread clinical use of soft tissue grafts, the histological characteristics of different gingival harvesting sites are still underexplored. This study aimed to fill this gap by analyzing 50 tissue samples harvested from 25 patients across three sites: the hard palate, maxillary tuberosity, and palatal rugae. Each sample underwent thorough histological and histomorphometric analysis. Conventional statistical analysis was performed using SPSS, while predictive modeling was conducted with RapidMiner Studio. The study identified significant histological differences among the graft sites, with notable variations in total graft height, epithelial height, and interdigitation perimeter. These findings underscore the importance of donor site selection in influencing graft success. Pair plots and principal component analysis (PCA) further highlighted the distinct histological features of each tissue type. The random forest classifier identified total graft height, epithelial height, and perimeter as the most influential factors in predicting graft site behavior. Understanding the reproductive ecology of crustaceans is important for developing appropriate resource management policies. Because the demand for sand crabs (Ovalipes punctatus) as a food resource has recently surged in Korea, their reproductive ecological characteristics should be comprehensively described. In this study, the reproductive ecological characteristics—sex ratio, female gonad maturity, abdominal width (AW), condition factor (K), and gonadosomatic index (GSI)—of individuals of O. punctatus collected from the Yellow Sea in Korea (35°15′00″N, 125°15′00″E) from January to December 2021 were examined. Based on the sex ratio (1:1.61, females:males), we determined that males were more abundant than females. The monthly K for both the males and females reached its maximum in April. The monthly GSI revealed that the males were most numerous in December, whereas the females were most abundant in May. The GSI graph indicates that the main spawning season of O. punctatus is from May to July. The AW and GSI of the females increased as their gonads matured; based on this correlation, the maturity of female individuals can be determined by examining the abdominal flap morphology. The results of histological analysis indicated that females of O. punctatus exhibit synchronous development, by which oocytes develop simultaneously. Furthermore, the size at maturity, at which more than half of the individuals participated in spawning, was 75.6 mm. Tauopathy is known to be a major pathognomonic finding in important neurodegenerative diseases such as progressive supranuclear palsy (PSP) and corticobasal degeneration. However, the mechanism by which tauopathy is triggered remains to be elucidated. We previously identified the point mutation c.11596C > G, p.Pro3866Ala in the Bassoon gene (BSN) in a Japanese family with PSP-like syndrome. We showed that mutated BSN may have been involved in its own insolubilization and tau accumulation. Furthermore, BSN mutations have also been related to various neurological diseases. In order to further investigate the pathophysiology of BSN mutation in detail, it is essential to study it in mouse models. We generated a mouse model with the mouse Bassoon p.P3882A mutation, which corresponds to the human BSN p.P3866A mutation, knock-in (KI) and we performed systematic behavioral and histological analyses. Behavioral analyses revealed impaired working memory in a Y-maze test at 3 months of age and decreased locomotor activity in the home cage at 3 and 12 months of age in KI mice compared to those in wild-type mice. Although no obvious structural abnormalities were observed at 3 months of age, immunohistochemical studies showed elevation of Bsn immunoreactivity in the hippocampus and neuronal loss without tau accumulation in the substantia nigra at 12 months of age in KI mice. Although our mice model did not show progressive cognitive dysfunction and locomotor disorder like PSP-like syndrome, dopaminergic neuronal loss was observed in the substantia nigra in 12-month-old KI mice. It is possible that BSN mutation may result in dopaminergic neuronal loss without locomotor symptoms due to the early disease stage. Background: There are many adhesion barrier materials, cross-linked or non-cross-linked hyaluronic acid (HA), used during surgeries. Purpose: This study investigates the efficacy of cross-linked and non-cross-linked HA in preventing Achilles tendon adhesions. We hypothesized that non-cross-linked HA may be more effective than cross-linked HA in preventing Achilles tendon adhesions following injury and repair. Methods: Twenty male Sprague Dawley rats, totaling 40 legs, underwent Achilles tendon transection and repair. Following the surgery, they were treated simultaneously with cross-linked and non-cross-linked HA formulations. The rats were divided into four groups: a positive control group, a group treated with BMC non-cross-linked HA gel, a group treated with DEFEHERE cross-linked HA gel, and a group treated with ANIKA cross-linked HA gel. Four weeks after surgery, macroscopic evaluation of peritendinous adhesion and histological analysis were conducted to assess the effectiveness of the treatments. Results: Non-cross-linked BMC HA demonstrated superior efficacy in preventing tendon adhesions compared to cross-linked HA and control groups. Histological analysis confirmed reduced adhesion severity in the non-cross-linked HA group (P < 0.05). The findings support the potential of non-cross-linked HA as a treatment to inhibit tendon adhesions. Further research, including clinical trials, is warranted to validate these results in human subjects. Conclusions: Non-cross-linked BMC HA had significantly lower tendon adhesions parameters and better healing scores in histological analysis than cross-linked HA and control group did. Inflammatory bowel diseases (IBD) are classified into two entities, namely Crohn's disease (CD) and ulcerative colitis (UC), which differ in disease trajectories, genetics, epidemiological, clinical, endoscopic, and histopathological aspects. As no single golden standard modality for diagnosing IBD exists, the differential diagnosis among UC, CD, and non-IBD involves a multidisciplinary approach, considering professional groups that include gastroenterologists, endoscopists, radiologists, and pathologists. In this context, histological examination of endoscopic or surgical specimens plays a fundamental role. Nevertheless, in differentiating IBD from non-IBD colitis, the histopathological evaluation of the morphological lesions is limited by sampling and subjective human judgment, leading to potential diagnostic discrepancies. To overcome these limitations, artificial intelligence (AI) techniques are emerging to enable automated analysis of medical images with advantages in accuracy, precision, and speed of investigation, increasing interest in the histological analysis of gastrointestinal inflammation. This review aims to provide an overview of the most recent knowledge and advances in AI methods, summarizing its applications in the histopathological analysis of endoscopic biopsies from IBD patients, and discussing its strengths and limitations in daily clinical practice.

Quercus pyrenaica Willd thrives in the intermediate zone between the Mediterranean sclerophyllous and the temperate deciduous forest. In December 2022, the presence of the bacteria Xylella fastidiosa (Xf) was confirmed in samples collected from a Quercus pyrenaica located in Sabrosa, Vila Real, Portugal. Following Xf infection, the transport of water and nutrients is hindered due to the occlusion of xylem vessels. This loss of hydraulic conductivity may lead to vessel blockage and subsequent embolism formation. The objective of this study was to investigate the interaction between Xf and Quercus pyrenaica tissues, as well as the mechanism by which the bacteria can spread through the plant's xylem vessels, ultimately resulting in the formation of vascular plugs. At the time of the sample collection (10 months post-detection), symptoms of Bacterial Leaf Scorch (BLS) began to appear. Examination of xylem vessels using both light and scanning electron microscopy (SEM) revealed the presence of various types of occlusions, predominantly tyloses. Additionally, fibrillar networks, gums, starch grains, and crystals were observed. The stem vessels exhibited significantly more occlusions compared to the leaves. Furthermore, individual bacterial cells were observed to be attached to the vessel wall. This implies that occlusions were primarily induced by tyloses and gums as a defensive response to the invasion of vascular pathogens, in addition to the pathogen itself. This study highlights the presence of starch grains in stems, which may function as a refilling mechanism, thereby preventing the loss of hydraulic conductivity in plants and potentially acting as a means to entrap the bacteria. These mechanisms exemplify the constitutive defense systems of the plant against Xf. Understanding the interaction between Xylella fastidiosa and Quercus pyrenaica is crucial, given that the latter species occupies nearly 95% of the natural distribution area of Portugal.

AbstractThis work investigated the biochemical disturbances and histological alteration in Psammomys obesus animal model fed different high calorie diets (HCDs) during three months. Four diets were used: a low-calorie natural diet, Chenopodiaceae halophyte plant used as control (LCD), a high standard carbohydrate diet rich in protein, HCD 0, a high carbohydrate diet rich in two concentrations of fat, HCD 1 and HCD 2. All animals having received HCDs developed dyslipidemia after one month of experiment with distinction of different sub-groups developing or not obesity and diabetes. HCDs induced a remarkable increasing in blood cholesterol, LDL-cholesterol and triglyceride levels indicating a fast induction of dyslipidemia and a significant increase of aminotransaminases activities revealing a pronounced hepatotoxicity. Animal developing diabetes showed a severe hepatic injury, a degeneration of the adipose tissue and a significant reduction of retinal thickness. Breast cancer (BC) cells characteristics have played a crucial role in clinical strategies decisionmaking and patient outcome prediction. Although there have been several studies examining this, there are still relatively few that analyze stem cell subpopulations or protein biomarkers. The conducted research sought to elucidate breast cancer subtype characteristics based on immunohistochemical analysis and cancer stem cell presence based on flow cytometry analysis in three breast cancer samples from Bandung, Indonesia. A flow cytometry analysis was conducted to determine how much CD44/CD24 was expressed as a marker of cancer stem cell in isolated BC cells using various cell references, such as cell lines of SKBR-3, MDA-MB-468, and Hs587T. Immunohistochemistry analysis was carried out to investigate the expression of p53, HER2, PGR, Ki67, and ER in sample tissue sections. According to histological analysis, 2 samples were solid mammary carcinoma, and 1 sample was mammary fibroadenoma. Immunohistochemistry and histological analysis showed that all BC tissues are classified as Luminal B. In addition, a large percentage of CD44+/CD24- subpopulation was found in isolated BC cells (> 90 % in patients 1 and 3; > 60 % in patient 2). All samples showed similarity to Hs587T cell line characteristic. This study has successfully characterized solid mammary tumors and mammary fibroadenoma with luminal B characteristics. In clinical practice, an innovative laser technology that provides contactless preparation of soft tissues with a wavelength of 445 nm has been introduced. This study aimed to investigate the morphological changes in the oral mucosa when exposed to laser radiation at a wavelength of 445 nm in the ablation mode. An experimental study was conducted to analyze the dynamics of reparative regeneration in the wound caused by that particular type of radiation, utilizing the procedure of lower lip frenuloplasty as an illustration. 48 sexually mature male laboratory rats were chosen as the research object. The procedure of preparing the oral vestibule was executed by employing a contactless laser beam with a wavelength of 445 nm and a power of 0.7 W in continuous mode (CW) and an uninitiated fiber. Histological examination showed that 25 min after the surgery, there were large areas of coagulation necrosis in the oral mucosa in the area affected by the blue laser. In 48 h, the area of necrosis decreased both in size and depth. By the 7th day after the surgery, the necrotic masses had grown into the connective tissue, while marginal regeneration of the epithelium was noted. By the 14th day, the wound surface was completely epithelialized, represented by fibrous scar tissue. Clinically, around the mandibular incisors, there was a wide area of attached keratinized gingiva. The findings of histological examination indicate a necrosis of coagulation type in the region of tissue ablation and also show the absence of phase II of the inflammatory response (the stage of exudation), which expedites the process of epithelialization of the oral mucosa wound. Aronia melanocarpa berries contain many compounds with potential benefits for human health. The food flavonoids quercetin and rutin, found in significant amounts in the fruits of A. melanocarpa, are known to have favourable effects on animal and human organisms. However, data on the effect of flavonols isolated from black chokeberry on immune functions during immunosuppression are not available in the literature. Thus, the aim of this study was to evaluate the effect of flavonol fraction isolated from A. melanocarpa fruits, in comparison with pure quercetin and rutin substances, on the dysfunctional state of rat thymus and spleen in immunodeficiency. The study was performed on Wistar rats. The animals were orally administered solutions of the investigated substances for 7 days: water, a mixture of quercetin and rutin and flavonol fraction of A. melanocarpa. For induction of immunosuppression, the animals were injected once intraperitoneally with cyclophosphamide. Substance administration was then continued for another 7 days. The results showed that under the influence of flavonols, there was a decrease in cyclophosphamide-mediated reaction of lipid peroxidation enhancement and stimulation of proliferation of lymphocytes of thymus and spleen in rats. The greater amberjack Seriola dumerili is economically important in both capture fisheries and aquaculture. It is a gonochoristic species with no noticeable sexual dimorphism, usually reaching maturity at 3–4 years. Spawning occurs in the Pacific from May to June but varies between regions. During a study on the maturity of greater amberjack, abnormal hermaphrodites (AH) with ovotestes were found. Of the 51 fish examined from June 2017 to April 2018, 2 individuals (3.9%) were classified as AH, with one each identified in February and April 2018. These AHs were larger than the gonochoristic (GC) individuals of the same age. In February, both AH and GCs had sperm-filled seminal lobules and ovaries at the primary yolk stage. By April, atresias were seen in both the AHs and GCs. While oocyte diameter increased in both AHs and GCs between February and April, the gonadosomatic index was lower in AHs. Plasma estradiol-17β and 11-ketotestosterone levels of AHs were similar to those of male GCs. These results suggest that AHs have accelerated growth with potential maturation as functional males. Objective: Ancient remains tell a lot to those who can solve their mysteries. Mummified remains of individuals have the potential to tell their life stories. Pathological conditions in mummies are identified, commonly, thorough macroscopic inspection and radiological techniques. Patients and methods: In 2017, a hypogeum tomb was excavated close to the courtyard of Djehuty's tomb (TT11) at the archaeological site of the Spanish Djehuty Project in Luxor (Egypt). In Sector 10, a set of at least 10 mummies were recovered. All showed clear signs of plundering and were found scattered at the site. Wrappings and associated goods indicated that the individuals were from the first half of the 20th dynasty (922 to 888 BCE). Results: All the individuals were male and from a high social status. Upon removing the individuals for study, two dried stomachs were found. There were clear macroscopic differences. A field histological analysis was performed which revealed that the stomach that showed no alterations, at first glance, presented a pathological pattern that was compatible with gastric sarcoma. Conclusions: Malignant diseases are seldom described in Paleopathology; to our knowledge, this is the oldest and most unique case of this type of pathology in mummies worldwide. The wide use of antioxidants is due to their involvement in free radical processes in foods and human body. Interest in the use of low-value raw materials providing products with functional properties and increasing their shelf life is rapidly increasing. However, any changes in the formulation and technology may affect the properties and composition of the finished product. During the work, the effect of replacing 34% (sample 1) or 17% (sample 2) beef broth with 70% water-ethanol extract of yellow onion peels in the formulation of the experimental meat pates was investigated. The control product contained only beef broth as liquid. The total antioxidant capacity by the DPPH radical method (TACDPPH), fatty acid composition and amino acid composition were determined; microelement content analysis, proteomic and microstructural studies of meat pate samples with and without the addition of extract were also carried out. Aims: Rotator cuff (RC) injuries are characterized by tendon rupture, muscle atrophy, retraction, and fatty infiltration, which increase injury severity and jeopardize adequate tendon repair. Epigenetic drugs, such as histone deacetylase inhibitors (HDACis), possess the capacity to redefine the molecular signature of cells, and they may have the potential to inhibit the transformation of the fibro-adipogenic progenitors (FAPs) within the skeletal muscle into adipocyte-like cells, concurrently enhancing the myogenic potential of the satellite cells. Methods: HDACis were added to FAPs and satellite cell cultures isolated from mice. The HDACi vorinostat was additionally administered into a RC injury animal model. Histological analysis was carried out on the isolated supra- and infraspinatus muscles to assess vorinostat anti-muscle degeneration potential. Results: Vorinostat, a HDACi compound, blocked the adipogenic transformation of muscle-associated FAPs in culture, promoting myogenic progression of the satellite cells. Furthermore, it protected muscle from degeneration after acute RC in mice in the earlier muscle degenerative stage after tenotomy. Drought is a major stress affecting rice yields. Combining partial root-zone drying (PRD) and different nitrogen fertilizers reduces the damage caused by water stress in rice. However, the underlying molecular mechanisms remain unclear. In this study, we combined treatments with PRD and ammonia:nitrate nitrogen at 0:100 (PRD0:100) and 50:50 (PRD50:50) ratios or PEG and nitrate nitrogen at 0:100 (PEG0:100) ratios in rice. Physiological, transcriptomic, and metabolomic analyses were performed on rice leaves to identify key genes involved in water stress tolerance under different nitrogen forms and PRD pretreatments. Our results indicated that, in contrast to PRD0:100, PRD50:50 elevated the superoxide dismutase activity in leaves to accelerate the scavenging of ROS accumulated by osmotic stress, attenuated the degree of membrane lipid peroxidation, stabilized photosynthesis, and elevated the relative water content of leaves to alleviate the drought-induced osmotic stress. Moreover, the alleviation ability was better under PRD50:50 treatment than under PRD0:100. Integrated transcriptome and metabolome analyses of PRD0:100 vs PRD50:50 revealed that the differences in PRD involvement in water stress tolerance under different nitrogen pretreatments were mainly in photosynthesis, oxidative stress, nitrogen metabolism process, phytohormone signaling, and biosynthesis of other secondary metabolites. Some key genes may play an important role in these pathways, including OsGRX4, OsNDPK2, OsGS1;1, OsNR1.2, OsSUS7, and YGL8. Thus, the osmotic stress tolerance mediated by PRD and nitrogen cotreatment is influenced by different nitrogen forms. Our results provide new insights into osmotic stress tolerance mediated by PRD and nitrogen cotreatment, demonstrate the essential role of nitrogen morphology in PRD-induced molecular regulation, and identify genes that contribute to further improving stress tolerance in rice. Aims: The aim of this randomized controlled clinical trial was to compare the gene expression, micro‐CT, histomorphometrical analysis between biphasic calcium phosphate (BCP) of 70/30 ratio and deproteinized bovine bone mineral (DBBM) in sinus augmentation. Materials and Methods: Twenty‐four patients in need for sinus floor augmentation through lateral approach were randomized into BCP 70/30 ratio or DBBM. After at least 6 months of healing, a total of 24 bone specimens were collected from the entire height of the augmented bone at the area of implant placement and underwent micro‐CT, histomorphometric and gene expression analysis. The 12 bone specimens of BCP 70/30 ratio were equally allocated to micro‐CT and histologic analysis (test group, n = 6) and gene expression analysis (test group, n = 6). Similarly, the 12 bone specimens of DBBM were also allocated to micro‐CT and histologic analysis (control group, n = 6) and gene expression analysis (control group, n = 6). The newly formed bone, remaining graft materials and relative change in gene expression of four target genes were assessed. Results: The micro‐CT results showed no statistically significant difference in the ratio of bone volume to total volume (BV/TV ratio) for the two groups (BCP 41.51% vs. DBBM 40.97%) and the same was true for residual graft material to total volume (GV/TV ratio, BCP 9.97% vs. DBBM 14.41%). Similarly, no significant difference was shown in the histological analysis in terms of bone formation, (BCP 31.43% vs. DBBM was 30.09%) and residual graft area (DBBM 40.76% vs. BCP 45.06%). With regards to gene expression, the level of ALP was lower in both groups of bone grafted specimens compared with the native bone. On the contrary, the level of OSX, IL‐1B and TRAP was higher in augmented bone of both groups compared with the native bone. However, the relative difference in all gene expressions between BCP and DBBM group was not significant. Conclusions: The BCP, HA/β‐TCP ratio of 70/30 presented similar histological and micro‐CT outcomes in terms of new bone formation and residual graft particles with DBBM. Quercus pyrenaica Willd thrives in the intermediate zone between the Mediterranean sclerophyllous and the temperate deciduous forest. In December 2022, the presence of the bacteria Xylella fastidiosa (Xf) was confirmed in samples collected from a Quercus pyrenaica located in Sabrosa, Vila Real, Portugal. Following Xf infection, the transport of water and nutrients is hindered due to the occlusion of xylem vessels. This loss of hydraulic conductivity may lead to vessel blockage and subsequent embolism formation. The objective of this study was to investigate the interaction between Xf and Quercus pyrenaica tissues, as well as the mechanism by which the bacteria can spread through the plant's xylem vessels, ultimately resulting in the formation of vascular plugs. At the time of the sample collection (10 months post-detection), symptoms of Bacterial Leaf Scorch (BLS) began to appear. Examination of xylem vessels using both light and scanning electron microscopy (SEM) revealed the presence of various types of occlusions, predominantly tyloses. Additionally, fibrillar networks, gums, starch grains, and crystals were observed. The stem vessels exhibited significantly more occlusions compared to the leaves. Furthermore, individual bacterial cells were observed to be attached to the vessel wall. This implies that occlusions were primarily induced by tyloses and gums as a defensive response to the invasion of vascular pathogens, in addition to the pathogen itself. This study highlights the presence of starch grains in stems, which may function as a refilling mechanism, thereby preventing the loss of hydraulic conductivity in plants and potentially acting as a means to entrap the bacteria. These mechanisms exemplify the constitutive defense systems of the plant against Xf. Understanding the interaction between Xylella fastidiosa and Quercus pyrenaica is crucial, given that the latter species occupies nearly 95% of the natural distribution area of Portugal. Polyoxotungstate nanoclusters have recently emerged as promising contrast agents for computed tomography (CT). In order to evaluate their clinical potential, in this study, we evaluated the in vitro CT imaging properties, potential toxic effects in vivo, and tissue distribution of monolacunary Wells–Dawson polyoxometalate, α2-K10P2W17O61.20H2O (mono-WD POM). Mono-WD POM showed superior X-ray attenuation compared to other tungsten-containing nanoclusters (its parent WD-POM and Keggin POM) and the standard iodine-based contrast agent (iohexol). The calculated X-ray attenuation linear slope for mono-WD POM was significantly higher compared to parent WD-POM, Keggin POM, and iohexol (5.97 ± 0.14 vs. 4.84 ± 0.05, 4.55 ± 0.16, and 4.30 ± 0.09, respectively). Acute oral (maximum-administered dose (MAD) = 960 mg/kg) and intravenous administration (1/10, 1/5, and 1/3 MAD) of mono-WD POM did not induce unexpected changes in rats' general habits or mortality. Results of blood gas analysis, CO-oximetry status, and the levels of electrolytes, glucose, lactate, creatinine, and BUN demonstrated a dose-dependent tendency 14 days after intravenous administration of mono-WD POM. The most significant differences compared to the control were observed for 1/3 MAD, being approximately seventy times higher than the typically used dose (0.015 mmol W/kg) of tungsten-based contrast agents. The highest tungsten deposition was found in the kidney (1/3 MAD—0.67 ± 0.12; 1/5 MAD—0.59 ± 0.07; 1/10 MAD—0.54 ± 0.05), which corresponded to detected morphological irregularities, electrolyte imbalance, and increased BUN levels. Simple Summary: We have developed a graph- and attention-based A.I. model (PathoGraph) that predicts survival and identifies tissue areas of special diagnostic interest in whole slide images (WSI) of glioblastoma. CD276, an important immune checkpoint molecule, was confirmed to be a marker of malignant glioma cells/putative glioma stem cells (GSCs). The previously developed PathoFusion framework was used to selectively detect these cells. The graphs and attention scores that were obtained on the basis of this selection provide information that is hidden from conventional microscopic observation. Computerized methods have been developed that allow quantitative morphological analyses of whole slide images (WSIs), e.g., of immunohistochemical stains. The latter are attractive because they can provide high-resolution data on the distribution of proteins in tissue. However, many immunohistochemical results are complex because the protein of interest occurs in multiple locations (in different cells and also extracellularly). We have recently established an artificial intelligence framework, PathoFusion which utilises a bifocal convolutional neural network (BCNN) model for detecting and counting arbitrarily definable morphological structures. We have now complemented this model by adding an attention-based graph neural network (abGCN) for the advanced analysis and automated interpretation of such data. Classical convolutional neural network (CNN) models suffer from limitations when handling global information. In contrast, our abGCN is capable of creating a graph representation of cellular detail from entire WSIs. This abGCN method combines attention learning with visualisation techniques that pinpoint the location of informative cells and highlight cell–cell interactions. We have analysed cellular labelling for CD276, a protein of great interest in cancer immunology and a potential marker of malignant glioma cells/putative glioma stem cells (GSCs). We are especially interested in the relationship between CD276 expression and prognosis. The graphs permit predicting individual patient survival on the basis of GSC community features. Our experiments lay a foundation for the use of the BCNN-abGCN tool chain in automated diagnostic prognostication using immunohistochemically labelled histological slides, but the method is essentially generic and potentially a widely usable tool in medical research and AI based healthcare applications. Rhododendron mucronulatum Turcz., a rare medicinal and ornamental deciduous multi-branched shrub native to northern parts of East Asia, is a valuable genetic resource for breeding and biotechnological producing bioactive metabolites. To create propagation system of R. mucronulatum, an efficient method of direct shoot regeneration from leaf explants based on thidiazuron (TDZ) and dark treatment was developed, and detailed histological analysis for revealing localization of cell competent to morphogenesis was carried out for the first time. The highest regeneration rate (75%) and number of shoots per explant (an average of 51.7) were achieved under 0.5 µM TDZ and dark treatment. The dark treatment was found to increase the regeneration rate more than two-fold compared to culturing under light conditions. The histological analysis showed direct organogenesis pathway. The competent to morphogenesis cells were located in epidermal tissue of leaf explants on adaxial side of petiole next to main vein. Two sites of beginning of TDZ-induced adventitious shoot formation were revealed: the shoot formation directly from epidermal tissues and shoot formation from de novo epidermis-derived protrusions. The formation of embryo-like structures was observed on the late morphogenesis stages. It was discovered that parenchymal and epidermal tissues of embryo-like structures were separated from explant tissue, but its vascular bundle was connected with vascular bundle of explant. These structures gave rise to development of adventitious shoots. The external fundamental system (EFS) is a band of slow-growing tissue, deposited in the periphery of the bone cortex of some tetrapod limb bones as a result of low bone growth rates. This structure has been commonly used as an indicator of maturity attainment, allowing researchers to infer life history traits (e.g. age at maturity, growth rates) in both fossil and extant taxa. However, some studies have recognised the absence of the EFS in the tibia of some vertebrate groups, thus raising concerns about the validity of this bone to reconstruct individual life history. This work analyses and describes for the first time the presence of the external fundamental system (EFS) in the tibia of red deer. Results show that the EFS seems to be strongly conditioned by local growth patterns, as its local deposition indicates. Finally, this study highlights the importance of tibia investigations to expand data of bone microstructure in Cervidae and illustrates that research on extant taxa are essential for deciphering growth dynamics and life history strategies of fossil organisms. Growth is an important trait in aquaculture that is influenced by various factors, among which genetic regulation plays a crucial role. Megalobrama amblycephala, one of the most important freshwater species in China, exhibits wide variations in body mass among individuals of the same age within the same pool. But the molecular mechanisms underlying wide variation in body mass remain unclear. Here, we performed muscle histological and transcriptome analysis of muscle tissues from Fast-Growing (FG) and Slow-Growing (SG) M. amblycephala at the age of 4 months old (4 mo) and 10 months old (10 mo) to elucidate its muscle development and growth mechanism. The muscle histological analysis showed smaller diameter and higher total number of muscle fibers in FG compared to SG at 4 mo, while larger diameter and total number of muscle fibers were detected in FG at 10 mo. The transcriptome analysis of muscle tissue detected 1171 differentially expressed genes (DEGs) between FG and SG at 4 mo, and 718 DEGs between FG and SG at 10 mo. Furthermore, 44 DEGs were consistently up-regulated in FG at both 4 mo and 10 mo. Up-regulated DEGs in FG at 4 mo were mainly enriched in the pathways related to cell proliferation, while down-regulated DEGs were significantly enriched in cell fusion and muscle contraction. Up-regulated DEGs in FG at 10 mo were mainly enriched in the pathways related to cell proliferation and protein synthesis. Therefore, these results provide novel insights into the molecular mechanism of M. amblycephala muscle growth at different stages, and will be of great guiding significance to promote the fast growth of M. amblycephala. Improving plant regeneration ability and shortening regeneration time can promote the development of genetic transformation breeding technology for horticultural crops. We optimized several culture conditions, including explant type, mother plant genotype, and medium, to improve shoot formation in winter squash (Cucurbita maxima Duch.). Histological analysis of the occurrence of shoots was also carried out. The results indicate that cotyledon was the most suitable explant for inducing the shoot regeneration of winter squash. We found that 'Jin-li' had a shorter shoot induction time and a higher average number of shoots. The highest induction rate of 95.23% among the five lines. The average shoot induction rate of five lines was the highest (84.85%) on Murashige and Skoog (MS) medium supplemented with 2.0 mg/L 6-benzylaminopurine (6-BA) and 0.2 mg/L indole-3-acetic acid (IAA). We also found that there was an interaction between genotypes and induction media, and their interaction had a greater impact on the shoot induction rate than individual effects. Histological observation revealed that the induced shoots of winter squash cotyledons originated from subepidermal cells. We also found that the optimal medium for de novo root regeneration was 1/2 MS. We acclimatized and cultivated regenerated plants and harvested their fruits, which maintained the characteristics of mother plants. Human placental explants (HPEs) culture has generated significant interest as a valuable in vitro model for studying tissue functions in response to adverse conditions, such as fluctuations in oxygen levels, nutrient availability, exposure to pathogenic microorganisms, and toxic compounds. HPEs offers the advantage of replicating the intricate microenvironment and cell-to-cell communication involved in this critical and transient organ. Although HPEs culture conditions have been extensively discussed, a protocol for assessing the viability and function of HPEs during short-term culture has not been previously outlined. In this study, we have developed a short-term HPEs culture protocol, specifically up to 72 h, and have employed quantitative, semi-quantitative, and qualitative analyses to evaluate tissue viability and function over time. Under our standardized conditions, placental villi explants began to regain their structural properties (the integrity of the trophoblast and villous stroma) and the functionality of the HPEs (production of angiogenic, endocrine, and immunological factors) starting from 48 h of culture. This restoration ensures a suitable environment for several applications. The data presented here can be highly valuable for laboratories aiming to implement an HPEs model, whether in the process of standardization or seeking to enhance and optimize working conditions and timing with placental tissue. Recent studies have attempted to characterize the layer-specific mechanical and microstructural properties of the aortic tissues in either normal or pathological state to understand its structural-mechanical property relationships. However, layer-specific tissue mechanics and compositions of normal and dissected ascending aortas have not been thoroughly compared with a statistical conclusion obtained. Eighteen ascending aortic specimens were harvested from 13 patients with type A aortic dissection and 5 donors without aortic diseases, with each specimen further excised to obtain three tissue samples including an intact wall, an intima-media layer and an adventitia layer. For each tissue sample, biaxial tensile testing was performed to obtain the experimental stress-stretch ratio data, which were further fed into the Fung-type model to quantify the tissue stiffness, and Elastin Van Gieson stain and Masson's trichrome stain were employed to quantify the elastic and collagen fiber densities. Statistical analyses were performed to determine whether any significant differences exist in mechanical properties and compositions between diseased and normal aortic tissues. The tissue stiffness of intima-media samples was significant higher in diseased group than that of normal group in longitudinal direction at the stretch ratio 1.30 (p = 0.0068), while no significant differences were found in the other direction or other tissue types. Even though there was no significant difference in elastic or collagen fiber densities between two groups, the diseased group generally had lower elastic fiber density, but higher collagen fiber density for all three tissue layers. Compared to normal aortic tissues, the elastic fiber density of the intima-media layer in the dissected aortic tissue was lower, while its tissue stiffness was significantly higher, indicating the tissue stiffness of the intima-media layer could be a potential indicator for aortic dissection.

Spices and herbs are widely used in the meat processing industry to improve the taste and flavor of the food products. They contain a wide range of essential oils and biologically active components possessing antioxidant potential. Surge of spices consumption leads to their adulteration; at the same time, species identification is complex and requires increased knowledge about the peculiarities of their structure. This study researched the antioxidant potential (AOP) of six spices and three fragrant herbs, defined their structure and histological parameters of their identification. To assess AOP, total antioxidant capacity (TAC) was defined using the methods of Oxygen Radical Absorbance Capacity (ORAC) and free radical DPPH, and the main classes of AO were identified with the help of qualitative reactions, microstructure was analyzed via three staining methods. Among six classes of AO the flavonoids were found in all extracts. All of four AO classes were found in basil and allspice. Allspice extract showed the highest TACDPPH (2,876.05 ± 19.83 µmol-eq.quercetin/l), the lowest value was found in parsley extract (157.97 ± 4.80 µmol-eq.quercetin/l). At the same time, the highest TACORAC was found in the extract of dill greens and basil greens — 9,789.51 ± 433.22 μmol-eq.quercetin/l and 9,692.91 ± 203.42 μmol-eq.quercetin/l, respectively, and its lowest content was found in ginger — 956.98 ± 241.79 µmol-eq. quercetin/l. The microstructural features of cells peculiar for each sample were defined: external protective tissues, seed hulls, storage tissue, secretory and formative tissues, and their ability to perceive staining with general and specific dyes. The results obtained make it possible to test the composition of dry spices and herbs, to reveal their presence in the ready-to-consume meat products and to exclude cases of their adulteration.

One of the most common complications of acute respiratory distress syndrome is acute kidney injury, the mechanisms of development of which remain not completely clarified. The purpose of this study is to examine morphofunctional changes in the kidneys of rats with induced acute respiratory distress syndrome at various time intervals after modelling the pathology. For the study, 56 healthy sexually mature male rats weighing 200-220 g were used, divided into 7 groups: control, 6, 24 hours, 3, 7, 14, and 28 days of the experiment. Respiratory distress in animals was caused by inhalation of lipopolysaccharide (5 mg/kg body weight). The kidneys of intact rats had a typical histological structure without specific features. Histological changes in the renal parenchyma of rats in the study groups included compaction of Malpighian bodies, damage and desquamation of epithelial cells of the nephron tubules, and the appearance of signs of disseminated intravascular coagulation. A month after the start of the experiment, both pathological changes in the nephrons and restored or preserved structural components of the kidney are observed, which indicates activation of intracellular reparative processes. The expression of TGF-β1 fibrosis marker as well as CD68 panmacrophage marker increased on days 3 and 7 of the experiment. The number of macrophages in the kidney samples remained consistently high until the end of the experiment, while the level of TGF-β1 decreased on day 28, indicating the start of the resolution phase. Biochemical analysis of renal markers showed an undulating course of inflammatory processes in the kidneys of experimental rats. The maximum concentration of creatinine, urea, and uric acid in the blood serum was observed at 24 hours of the experiment, which indicated the onset of acute kidney injury as a complication of respiratory distress. Preclinical examination of morphofunctional changes in the kidneys during acute respiratory distress syndrome will help choose an effective method for treating this pathological condition in humans in the future

The science of dental tissue grafting is evolving, with an increased understanding of factors influencing graft behavior. Despite the widespread clinical use of soft tissue grafts, the histological characteristics of different gingival harvesting sites are still underexplored. This study aimed to fill this gap by analyzing 50 tissue samples harvested from 25 patients across three sites: the hard palate, maxillary tuberosity, and palatal rugae. Each sample underwent thorough histological and histomorphometric analysis. Conventional statistical analysis was performed using SPSS, while predictive modeling was conducted with RapidMiner Studio. The study identified significant histological differences among the graft sites, with notable variations in total graft height, epithelial height, and interdigitation perimeter. These findings underscore the importance of donor site selection in influencing graft success. Pair plots and principal component analysis (PCA) further highlighted the distinct histological features of each tissue type. The random forest classifier identified total graft height, epithelial height, and perimeter as the most influential factors in predicting graft site behavior. This study offers valuable insights into the histological characteristics of soft tissue grafts, potentially leading to more predictable clinical outcomes.

Mechanical mismatch between native aortas and aortic grafts can induce graft failure. This study aims to compare the mechanical and microstructural properties of different graft materials used in aortic repair surgeries with those of normal and dissected human ascending aortas. Five types of materials including normal aorta (n = 10), dissected aorta (n = 6), human pericardium (n = 8), bovine pericardium (n = 8) and Dacron graft (n = 5) were collected to perform uniaxial tensile testing to determine their material stiffness, and ultimate strength/stretch. The elastin and collagen contents in four tissue groups except for Dacron were quantified by histological examinations, while the material ultrastructure of five material groups was visualized by scanning electron microscope. Statistical results showed that three graft materials including Dacron, human pericardium and bovine pericardium had significantly higher ultimate strength and stiffness than both normal and dissected aortas. Human and bovine pericardia had significantly lower ultimate stretch than native aortas. Histological examinations revealed that normal and diseased aortic tissues had a significantly higher content of elastic fiber than two pericardial tissues, but less collagen fiber content. All four tissue groups exhibited lamellar fiber ultrastructure, with aortic tissues possessing thinner lamella. Dacron was composed of densely coalesced polyethylene terephthalate fibers in thick bundles. Aortic graft materials with denser fiber ultrastructure and/or higher content of collagen fiber than native aortic tissues, exhibited higher ultimate strength and stiffness. This information provides a basis to understand the mechanical failure of aortic grafts, and inspire the design of biomimetic aortic grafts.

Fisheries management requires improvement in scientific knowledge to ensure sustainable exploitation of important commercial species and population renewal. Within this context, this study focused on the reproductive biology of spot tail mantis shrimp, Squilla mantis, in the Central Mediterranean Sea, aiming to understand the reproductive pattern of this species and validate the macroscopic maturity scale through histological analysis. A multi-year sampling was performed from 2016 to 2020 by a commercial fishing fleet in the Northern Central Adriatic Sea (GSA 17), and a total of 2206 individuals were collected. The monthly average value of the total sex ratio of S. mantis was 0.524 ± 0.044 (mean ± SEM) in favor of females, which dominated the population from September to April. The proposed 5 stage macroscopic maturity scale was validated histologically through histological analysis, confirming synchronous ovarian development. The somatic indexes (GSI and K Fulton) and monthly incidence of macroscopic ovarian maturity stages highlighted a protracted reproductive season from winter to spring (January–May). Although the length-weight relationship showed a similar growth trend between genders, males reached a bigger size in terms of carapace length (C.L.) and dominated the population from 32 mm (C.L.). The macroscopic L50 estimated was 25.94 mm (C.L.).

In this work, a four-week-old male C57Bl/6 mouse model of abdominal aortic aneurysm (AAA) was developed to examine the AAA rupture mechanism. Immunofluorescence staining was adopted for quantifying the degradation of elastin, and Picrosirius Red staining was adopted for evaluating the density of collagen. Atomic force microscopy with two probe tip sizes of 5 µm and 20 nm was adopted for mechanical characterization of the AAA. The microstructure changes and stiffness changes in both AAA samples and controlled samples were inspected. The degradation of elastin, wall thickening, formation of micro vessels, and increased density of collagen were observed in the AAA samples. The AAA samples also exhibited fragmented texture from AFM scanning. The histogram of stiffness measurements of the AAA samples with a 20 nm tip demonstrated two unique peak frequencies of stiffness intervals (0–10 kPa and 40–50 kPa). The stiffer regions were correlated with the increased density of collagen, as shown in the immunofluorescence images. The softer regions, combined with the fragmented texture, could be the key index contributing to the initiation and propagation of AAA rupture. Overall, the AAA group showed a higher stiffness than the control group (50.77 ± 62.4 kPa vs. 40.6 ± 51.86 kPa). The findings from this work may help in explaining ruptures in small AAA (<5.5 mm), which account for ten percent of all AAA ruptures. This study aimed to evaluate the acute and long-term exposure to different cupric sulfate pentahydrate (CuSO4.5H2O) concentrations in Nile tilapia (Oreochromis niloticus). Experiment 1 evaluated the LC50-96 h of juveniles at 12 different CuSO4.5H2O concentrations (0.00, 0.2, 0.4, 0.8, 1.6, 3.2, 6.4, 12.8, 25.6, 51.2, 102.4, and 204.8 mg L–1). In experiment 2, fish were exposed to four different sublethal cupric sulfate pentahydrate concentrations (0.0, 0.0069, 0.0696, and 0.6960 mg L–1) with evaluations on days 20 and 40. In the first experiment, the LC50 and safe concentration for 96 h were, respectively, 6.96 and 0.6960 mg CuSO4.5H2O L−1. In the second experiment, fish subjected to 0.6960 mg CuSO4.5H2O L–1 decreased growth performance and survival, altered hematological parameters, and had hepatic vacuolation and liver necrosis. In conclusion, we have found that to control the excessive algae growth, fish could tolerate a concentration of up to 0.0696 mg CuSO4.5H2O L−1 for 40 days. [ABSTRACT FROM AUTHOR]

Platelet rich fibrin (PRF) has gained recognition as a regenerative biomaterial with potential applications in soft tissue healing. The study was aimed to prepare bovine origin platelet rich fibrin gel and characterisation by histology and scanning electron microscopy. PRF gel was prepared from venous blood collected from cross-bred postpartum dairy cattle centrifuged at 3200 rpm for 12 minutes. Histology and scanning electron microscopy revealed a compact fibrin network and the cells were distributed within PRF gel. Vascular graft infection is still a life-threatening complication of reconstructive surgery. Among other options, application of cryopreserved homografts can eventuate favorable outcome, if graft replacement is necessary. The preparation and storage of these allografts need special infrastructure and deep subzero (− 80 °C) temperature. However, the longer storage time can lead to inferior results after implantation, based upon clinicians' experiences. The goal of our investigation was to circumscribe the optimal storage time interval with differential scanning calorimetry (DSC) and histological evaluation, using porcine aorta. All samples were deep-freezed using − 80 °C. Cryopreservated grafts were melted after 4, 6, 12, 16, 20, 24, 28 and 52 weeks; then, DSC and different types of histology were performed. Light microscopy analysis showed significant changes in the connective tissue fibers' structure from the 16th week; while, DSC measurements confirmed systematic decrease in the thermal stability from the same week during the follow-up period. Our investigation suggested that cryopreservation can lead to significant and increasing microstructural damage of the fibers following the 12th week; thus, the homograft implantation can result in higher success rate inside this timeframe. The hooded oyster, Saccostrea cucullata, is an economically important bivalve in Thailand with great potential for aquaculture production. Since its histological development remains unknown, we examined the organ development of field-collected S. cucullata associated with their shell size distributions from Libong Island, Thailand. The water quality parameters were not significantly different between the sampling areas except for the water temperature (p < 0.05). S. cucullata from the Stone Bridge (SB) site had smaller shell lengths and higher condition factors than those from Dugong Tourism by Drones (DT) except for the size of the organ. Histologically, the highest mean length of gill lamellae was 192.1±3.92 µm in 2.1-3 cm group. The thinnest of the mantle epithelium (73.78±3.08 µm in 4.1-5 cm group) of S. cucullata showed statistically significant difference between the sampled locations (Specific F value = 0.1783 and 0.8605 when p < 0.01 and p < 0.0001, respectively). The density of mucous-secreting cells (Msc) was more prominently distributed in the digestive gland than in other tissues. The 2.1-3 cm group or bigger had mature gonads with protandric characteristics, showing the rapid sexual differentiation in SB. Results obtained from this study improve the current knowledge of S. cucullata from natural environments, which potentially contributes to the establishment of its aquaculture. Iron oxide nanoparticles (IONP) are showing promise in many biomedical applications. One of these- magnetic hyperthermia- utilizes externally applied alternating magnetic fields and tumor-residing magnetic nanoparticles to generate localized therapeutic temperature elevations. Magnetic hyperthermia is approved in Europe to treat glioblastoma and is undergoing clinical assessment in the United States to treat prostate cancer. In this study, we performed biodistribution and histological analysis of a new IONP (RCL-01) in Wistar rats. These nanoparticles are currently undergoing clinical assessment in locally advanced pancreatic ductal adenocarcinoma to determine the feasibility of magnetic hyperthermia treatment in this disease. The study presented here aimed to determine the fate of these nanoparticles in vivo and whether this results in organ damage. Wistar rats were injected intravenously with relatively high doses of IONP (30 mgFe/kg, 45 mgFe/kg and 60 mgFe/kg) and compared to a vehicle control to determine the accumulation of iron in organs and whether this resulted in histological changes in these tissues. Dose-dependent increases of iron were observed in the liver, spleen and lungs of IONP-treated animals at 7 days postinjection; however, this did not result in significant histological changes in these tissues. Immunofluorescent imaging determined these nanoparticles are internalized by macrophages in tissue, suggesting they are readily phagocytosed by the reticuloendothelial system for eventual recycling. Notably, no changes in iron or dextran staining were found in the kidneys across all treatment groups, providing evidence for potential renal clearance. [ABSTRACT FROM AUTHOR]

The leaf is the most important organ for photosynthesis; a moderately curly leaf is beneficial for improving the light energy utilization rate. Male sterility is another key trait that can facilitate hybrid seed production. Previously we have developed a mutant line JQA showing wrinkled leaves and male sterility in sesame (Sesamum indicum L.), which is an important genetic stock for the development of super variety with ideal plant type. Before it can be fully utilized in breeding practice, a better understanding of the cytological basis for leaf and anther development in JQA is required. In this study, we investigated the developmental processes in JQA at three cytological levels. Histological observation showed that the wrinkled leaf trait was expressed during the whole life cycle. Compared with normal leaves, the JQA wrinkled leaves have much fewer chloroplasts in the differentiated palisade tissue, which affect the photosynthesis efficiency by reducing the number of chloroplasts and delaying their development, and finally leads to leaf wrinkle. In addition, transmission electron microscope observation showed that most of the microspore mother cells in JQA could not divide evenly into dyads and tetrads during meiosis and that the tapetal cells could not disintegrate in time to provide adequate nutrition for the microspores. Consequently, pollens and pollen sacs were impeded or degraded due to a severe lack of nutrition. Abstract The histological analysis of local and imported beef samples throughout storage at various intervals in 4 °C, before and after being frozen at − 18 °C, to detect the changes happened in the microstructure of muscle fibers to evaluate the meat nutritive properties in a step toward rapid evaluation of meat quality. The obtained results illustrated that freezing–thawing step of beef leads to the loss of its muscle fiber structure due to the high moisture content failure, highlighted the idea that imported beef show significant shrinkage in their muscle fibers from the beginning of its purchase to consumers as they seem to be imported as frozen and thawed just before exposed and sold as fresh. Through consumption survey, however consumers prefer local meat, it was detected that 67% of population is eating imported beef with 39.4% more than twice per week. Therefore, consumers’ minds should be changed to depend on locally slaughtered beef on facing their needs of the recommended daily intake of protein.

Abstract Background Colorectal cancer (CRC) is one of the most common malignancies in the world. This study proposes to reveal prognostic biomarkers for the prognosis and treatment of CRC patients. Methods Differential analysis of OSBPL3 was performed in pan-cancer, and the correlation between clinical stage and OSBPL3 was analyzed. Multiple omics analysis was used to compare the relationship between survival of patients and copy number variation, single nucleotide variant, and methylation status. Survival differences between high and low OSBPL3 expression groups were analyzed. Differentially expressed genes (DEGs) between high and low OSBPL3 expression groups were obtained, and functional enrichment analysis was implemented. Correlations between immune cells and OSBPL3 was analyzed. Drug sensitivity between the two OSBPL3 expression groups was compared. Moreover, the expression of OSBPL3 was verified by immunohistochemistry and real-time quantitative PCR. Results OSBPL3 was differentially expressed in 13 tumors and had some correlations with T and N stages. OSBPL3 expression was regulated by methylation and higher OSBPL3 expression was associated with poorer prognosis in CRC. 128 DEGs were obtained and they were mainly involved in signaling receptor activator activity, aspartate and glutamate metabolism. T cell gamma delta and T cell follicular helper were significantly different in the high and low OSBPL3 expression groups. Moreover, OSBPL3 showed negative correlations with multiple drugs. OSBPL3 was significantly upregulated in CRC samples compared to normal samples. Conclusions A comprehensive analysis demonstrated that OSBPL3 had potential prognostic value, and guiding significance for CRC chemotherapeutic.

Aronia melanocarpa berries contain many compounds with potential benefits for human health. The food flavonoids quercetin and rutin, found in significant amounts in the fruits of A. melanocarpa, are known to have favourable effects on animal and human organisms. However, data on the effect of flavonols isolated from black chokeberry on immune functions during immunosuppression are not available in the literature. Thus, the aim of this study was to evaluate the effect of flavonol fraction isolated from A. melanocarpa fruits, in comparison with pure quercetin and rutin substances, on the dysfunctional state of rat thymus and spleen in immunodeficiency. The study was performed on Wistar rats. The animals were orally administered solutions of the investigated substances for 7 days: water, a mixture of quercetin and rutin and flavonol fraction of A. melanocarpa. For induction of immunosuppression, the animals were injected once intraperitoneally with cyclophosphamide. Substance administration was then continued for another 7 days. The results showed that under the influence of flavonols, there was a decrease in cyclophosphamide-mediated reaction of lipid peroxidation enhancement and stimulation of proliferation of lymphocytes of thymus and spleen in rats. At that, the effect of the flavonol fraction of aronia was more pronounced.

The use of in vitro callus cultures as experimental model systems allows us to get closer to understanding the patterns and features of morphogenesis in intact plants. In this regard, the problem of realizing the morphogenetic potential of callus cells due to their pluri- and totipotency properties is of great interest. To solve this problem, it is important to use the histological approach, which involves studying the structures of developing tissues, organs and organisms in their interactions and relationships. This review article analyzes data devoted to the study of the histological features of formed primary morphogenic calli (formation of morphogenetic centers and superficial meristematic zones), as well as the in vitro morphogenesis pathways in calli that lead to the formation of regenerants (de novo organogenesis and in vitro somatic embryogenesis). The terminology used is considered. Some questions for discussion are raised. The opinion is expressed that histological (structural) studies should be considered as a methodologic basis for further investigation of various morphogenetic scenarios in in vitro callus cultures, especially in economically valuable plants and for biotechnological purposes.

Objective: The aim of this study was to present a mini review of oral Cementoblastoma and to report a particular case of this tumor. The Cementoblastoma is a rare benign lesion that represents less than 1-6% of all odontogenic tumors. Cementoblastoma, in the current WHO classification of odontogenic tumors, falls under the category of mesenchymal tumors (WHO 2017) and it is characterized by the proliferation of cementum-like tissue and, in all cases, tends to be associated with an erupting permanent tooth, most often the first molar. Case Presentation: A 15-year-old female presented a great Cementoblastoma with cortical expansion that affected the left mandibular body, extending from the canine to the first premolar (size 28x24 mm) and involving the mandibular canal. Surgery was performed under general anesthesia with total excision of the lesion, which was then sent for histological analysis. Due to the high risk of nerve injuries and the extreme fragility of the remaining bone after surgery, which required plates and intermaxillary blockage, the surgical site was regenerated with an iliac crest graft together with autologous and synthetic bone. Results: For the mini-review, 107 articles were found, but only 26 were selected. The patient was monitored for 12 months after surgery; a perfect healing was reported without complications, and she showed no signs of recurrence. Conclusions: This case report emphasizes the usefulness of clinical choices in a rare case of cementoblastoma of great size in a very young patient. In this condition, finding the most effective eradicative and reconstructive treatment, following the last published protocols, could achieve optimal clinical and psychological patient outcomes.
