A study of keracyanin chloride (KC) electrochemical behavior in an aqueous buffer solution using screen-printed carbon electrodes (SPCEs) and glassy carbon electrodes (GCEs) was performed. Cyclic voltammetry (CV) and square-wave voltammetry (SWV) were used to analyze the electrochemical response of KC under studied conditions. A clear redox wave was observed for KC, primarily due to the oxidation of the catechol 3′,4′-dihydroxyl group of its ring B, with a minor redox wave from oxidation of the hydroxyl groups in ring A. Compared to GCEs, using modified SPCEs resulted in two-fold amplification in the electrochemical oxidation signal of KC. Using SPCEs as a working electrode could provide high sensitivity in the quantification of KC and the ability to gauge KC quantification to significantly lower detection limits.

Environmental cues elicit anthocyanin synthesis in plant vegetative and reproductive tissues. Their accumulation in different organs accounts for their diverse biological functions, mainly related to their antioxidant properties, and it depends on a temporally and spatially regulated mechanism controlled by the action of a well-known multi-transcription factor complex. Despite the highly recognizable value of Cannabis sativa L. as a natural biorefinery of phytochemicals, very little information is known on anthocyanin pigmentation in this species. In this work, a targeted quantification of anthocyanins via HPLC-MS/MS, combined with the transcriptional profile via RT-qPCR of genes encoding for structural and decorating enzymes and regulatory transcription factors in different C. sativa tissues, help gain insights into the anthocyanin pathway in this species. To the best of our knowledge, this is the first report on the identification of cyanidin-3-rutinoside (keracyanin) as the major anthocyanin in C. sativa vegetative and floral tissues. Keracyanin amounts were higher than in small berries, suggesting that Cannabis biomass is a valuable source of colored antioxidants to be exploited in diverse applications. Furthermore, a gene putatively encoding for an anthocyanin DTX35 type transporter and CsTTG1 were identified in silico and their transcriptional levels were assessed via RT-qPCR. The results allow us to provide the first model of anthocyanin regulation in C. sativa, opening a new research scenario in this species for both breeding purposes and phytochemical exploitation. This work aimed to propose two analytical methods for the quantitative and qualitative analysis of major anthocyanins and non-anthocyanin phenolic compounds in jussara ( Euterpe edulis) extracts, using ultra performance liquid chromatography-mass spectrometry. These methods were evaluated for selectivity, precision, linearity, detection and quantification limits. The complete separation of 5 anthocyanins and 22 non-anthocyanins polyphenols was achieved in 4.5 and 7 min, respectively. Limits of detection ranged from 0.55 to 9.24 µg/L, with relative standard deviation for concentration up to 7.0%. In jussara extract, 13 of the 27 analytes were characterized. The dominant compound was cyanidin-3-O-rutinoside, representing about 73% of the total phenolic compounds content (approximately 23 mg/g of extract in dry weight). Other phenolic compounds found in the extract were: cyanidin-3-O-glucoside, pelargonidin-3-O-glucoside, quercetin, rutin, myricetin, kaempferol, kaempferol-3-O-rutinoside, luteolin, apigenin, catechin, ellagic acid and 4,5-dicaffeoylquinic acid. Fruit preparations (e.g., fruit juices and nectars) are rich in plant pigments and of great demand by the alimentary and pharmaceutical industry, basically not only due to their health-enhancing properties, but also due to their attractive colors and an overall high esthetic valor. Anthocyanins and anthocyanidins are an interesting group of plant pigments, and in this study, a thin-layer chromatographic detection was carried out of two anthocyanins (cyanin and keracyanin) and two anthocyanidins (pelargonidin and delphinidin) in a selection of the commercial and homemade fruit juices and the infusions prepared of dried plants. Moreover, the same preparations were evaluated for their antioxidant properties by means of two spectrophotometric methods (based on the 2,2′-azino-di-[3-ethylbenzthiazoline] sulfonate (ABTS) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) tests, respectively) and by the dot-blot test (based on DPPH). Finally, an effort was undertaken to correlate the chromatographically established occurrence of plant pigments in the investigated fruit preparations with their antioxidant properties. However, this correlation was not straightforward, and for at least two reasons. First, the four plant pigments considered are not the only secondary plant metabolites which exert an antioxidant activity. Second, different chemistries stand behind the two spectrophotometric antioxidant activity tests (ABTS vs. DPPH), and the different measuring techniques (spectrophotometry vs. dot blot) are assumed. Nevertheless, it was established that the juices and infusions with the highest detected numbers of plant pigments characterize with the high antioxidant activities also (with blueberries, chokeberries, and hibiscus flower on the top positions). In that way, confirmation of the antioxidant potential of the plant pigments was obtained.
