Our knowledge on the development and functions of CXCR5+ CD8 T cells is rudimentary when confronted to other extensively studied CD8 T cell subsets. A decade ago, it became apparent that CD8 T cells possess two additional and rather unexpected functional properties other than cytotoxicity, one involving what is known as B cell helper activity and the other involving suppression of self-reactive responses generally known as T cell regulation. Although these adaptive responses are well-known functions of CD4 T cells, they remain poorly understood in CD8 T cells. Thus far, three subsets of CXCR5+ CD8 T cells have been identified. The first subset of CXCR5+ CD8 T cells is present in chronic viral infections and is referred to as progenitors of exhausted T cells showing heightened proliferative and cytotoxic properties as compared to CXCR5- CD8 T cells. The second subset of CXCR5+ CD8 T cells functions as regulatory T cells that inhibit CD4 T follicular helper (Tfh) humoral responses and the development of autoantibodies. The third subset of CXCR5+ CD8 T cells was identified in mice with mutations in immunoregulatory genes (i.e. FOXP3 and IL-2-deficient mice) and involves CD8 T cells with Tfh-like properties that promote humoral autoimmunity through interaction with B cells. This review summarizes the phenotype, function, and differentiation of CXCR5+ CD8 T cells.

Benign terminal deoxynucleotidyl transferase (TdT)-positive cells have been documented in a variety of nonhematopoietic tissues. Scant data are, however, available on their presence in nonneoplastic lymph nodes. This study is aimed to (1) characterize the presence/distribution of benign TdT-positive cells in pediatric and adult reactive lymph nodes and (2) define the phenotype and nature of such elements. This retrospective study considered 141 reactive lymph nodes from pediatric and adult patients without history of neoplastic disease. TdT-positive cells were characterized by immunohistochemical and morphometric analyses, and their presence was correlated with the clinical-pathological features. The nature of TdT-positive cells was investigated by (1) double immunostaining for early lymphoid cell markers and (2) assessment of TdT expression in fetal lymph nodes. Sparse TdT-positive cells were documented in all pediatric cases and in most (76%) adult lymph nodes. TdT-positive cell density was higher in children than adults (15.9/mm2 versus 8.6/mm2; P < .05). TdT positivity did not correlate with any clinical or histological parameter, and double immunostaining disclosed a phenotype compatible with early lymphoid precursors (positivity for CD34 and CD10, and variable expression of CD7). A very high TdT-positive cell density (802.4/mm2) was reported in all fetal lymph nodes. In conclusion, TdT-positive cells are a common finding in pediatric and adult lymph nodes. The interstitial distribution and low number of such cells allow for the differential diagnosis with precursor lymphoid neoplasms. The high density in fetal lymph nodes and the phenotype of such cells suggest their belonging to an immature lymphoid subset gradually decreasing with age.

HMGA1 (high-mobility-group A1) proteins are architectural transcription factors that are found overexpressed in embryogenesis and malignant tumours. We have shown previously that they have a role in lymphopoiesis, since the loss of HMGA1 expression leads to an impairment of T-cell development and to an increase in B-cell population. Since RAGs (recombination activating genes) are key regulators of lymphoid differentiation, in the present study we investigate whether RAG2 expression is dependent on HMGA1 activity. We show that RAG2 gene expression is up-regulated in Hmga1−/− ES (embryonic stem) cells and EBs (embryoid bodies) as well as in yolk sacs and fibroblasts from Hmga1−/− mice, suggesting that HMGA1 proteins control RAG2 gene expression both in vitro and in vivo. We show that the effect of HMGA1 on RAG2 expression is direct, identify the responsible region in the RAG2 promoter and demonstrate binding to the promoter in vivo using chromatin immunoprecipitation. Since RAG2 is necessary for lymphoid cell development, our results suggest a novel mechanism by which HMGA1 might regulate lymphoid differentiation.

In this issue of the journal, we host a topic focused on the roles of human Forkhead-box (FOX) gene family members, including FOXN1, FOXP genes, and FOXO1, in the immune system. In the first review of the issue, Palamaro et al. focused their attention on the pivotal role of FOXN1 in T-cell development and the clinical implications of its mutation in humans. Dr Fleskens and Dr van Boxtel detailed the role of FOXP members in immune regulation. Concerning the roles of FOXP members in immune response, Vent-Schmidt et al. focused the attention on FOXP3. Passerini et al. highlighted the current knowledge on the involvement of FOXP3 in the development and function of Treg cells. Eventually, in the last review of this special issue, Szydłowski et al. focused on the role of FOXO1 in B-cell development and differentiation, paying particular attention to the role of PI3K-AKT signaling activation in development, differentiation, function, and homeostasis of B cells. Notably, studies on the role of FOX family members in immune response could be helpful to further investigate pathogenesis of immunological disease and to develop a novel therapeutics approach for human diseases.
