ABSTRACT: The eggshell is the outermost covering of an egg that provides physical and chemical protection. It is a major source of calcium and minerals for the growing embryo during incubation. The egg industry suffers from a considerable economic loss due to poor eggshell quality. Therefore, developing an accurate and precise method of determining eggshell quality is crucial in improving eggshells in subsequent generations of breeding stock. Hence, this study aimed to develop a method to accurately and precisely determine 1) eggshell thickness using a scanning electron microscope (SEM) and 2) eggshell mineral components using an SEM-Energy Dispersive Spectrometry (EDS) system. Four types of table eggs (N = 48; 12 eggs/group): Cage-Free Organic from the US Mainland (CFO-M) and Hawaii (CFO-H), Caged Non-Organic from the US Mainland (CNO-M), and Hawaii (CNO-H) were sourced from the grocery store. Approximately 0.5 mm2 pieces of eggshells from the equator region of the egg were taken and processed for visualization under the SEM. Three distinct layers of eggshell were identified under SEM: the outermost cuticle, the middle palisade, and the innermost mammillary region. The results showed that CFO-H eggs have a greater eggshell thickness (380.43 ± 2.69 µm) and effective thickness (306.28 ± 4.15 µm). Similarly, the mammillary knob count was denser in CNO-H eggs (186 ± 23.02 knobs/0.5 mm2). Calcium (97.36 ± 0.17%) was the highest among minerals in lower palisades (LP). The magnesium concentration was lowest in the LP region, whereas the phosphorus concentration was highest in the upper palisades. Our study established a scientific method to assess the eggshell quality and biochemical characteristics of eggs through SEM and EDS. This method can be used as a marker for selecting superior parent stock to improve eggshell quality in subsequent generations of breeding stock.

Post mortem examination of the hypothalamus of a 79-year-old woman, deceased in cardiac arrest without recorded neurological symptoms, revealed well-defined spherical protrusions located rostro-laterally to the mammillary bodies that appear to be regular size when compared to normal. Cytoarchitectonically, these accessory mammillary bodies are formed by the enlarged lateral mammillary nucleus that is normally a thin shell over the medial. The mammillary nuclei appear to function synergistically in memory formation in rats; however, the functional consequences of the present variation are difficult to interpret due to lack of human data. Most importantly, in addition to the possible functional consequences, lateral mammillary bodies can be falsely identified as various neuropathological processes of the basal diencephalon including gliomas; therefore, it is extremely important to disseminate this unique morphological variant among clinicians. Uranium-series analyses of water-table-type speleothems from Glenwood Cavern and “cavelets” near the town of Glenwood Springs, Colorado, USA, yield incision rates of the Colorado River in Glenwood Canyon for the last ~1.4 My. The incision rates, calculated from dating cave mammillary and cave folia calcite situated 65 and 90 m above the Colorado River, are 174 ± 30 m/My for the last 0.46 My and 144 ± 30 m/My for the last 0.62 My, respectively. These are consistent with incision rates determined from nearby volcanic deposits. In contrast, δ234U model ages (1.39 ± 0.25 My; 1.36 ± 0.25 My; and 1.72 ± 0.25 My) from three different samples of mammillary-like subaqueous crust collected from Glenwood Cavern, 375 m above the Colorado River, yield incision rates of 271 +58/-41 m/My, 277 +61/-42 m/ My, and 218 +36/-27 m/My. These data suggest a relatively fast incision rate between roughly 3 and 1 Ma. The onset of Pleistocene glaciation may have influenced this rate by increasing precipitation on the Colorado Plateau starting at 2.5 Ma. Slowing of incision just before 0.6 Ma could be related to the change in frequency of glacial cycles from 40 to 100 kyr in the middle Pleistocene. This interpretation would suggest that the cutting power of the Colorado River prior to 3 Ma was smaller. An alternative interpretation involving tectonic activity would invoke an episode of fast uplift in the Glenwood Canyon region from 3 to 1 Ma.

The hypothalamus is the higher neuroendocrine center of the brain and therefore possesses numerous intrinsic axonal connections and is connected by afferent and efferent fiber systems with other brain structures. These projection systems have been described in detail in the adult but data on their early development is sparse. Here I review studies of the time schedule and features of the development of the major hypothalamic axonal systems. In general, anterograde tracing experiments have been used to analyze short distance projections from the arcuate and anteroventral periventricular nuclei, while hypothalamic projections to the posterior and intermediate pituitary lobes and median eminence, mammillary body tracts and reciprocal septohypothalamic connections have been described with retrograde tracing. The available data demonstrate that hypothalamic connections develop with a high degree of spatial and temporal specificity, innervating each target with a unique developmental schedule which in many cases can be correlated with the functional maturity of the projection system.

The neural representation of directional heading is conveyed by head direction (HD) cells located in an ascending circuit that includes projections from the lateral mammillary nuclei (LMN) to the anterodorsal thalamus (ADN) to the postsubiculum (PoS). The PoS provides return projections to LMN and ADN and is responsible for the landmark control of HD cells in ADN. However, the functional role of the PoS projection to LMN has not been tested. The present study recorded HD cells from LMN after bilateral PoS lesions to determine whetherthe PoS provides landmark control to LMN HD cells. After the lesion and implantation of electrodes, HD cell activity was recorded while rats navigated within a cylindrical arena containing a single visual landmark or while they navigated between familiar and novel arenas of a dual-chamber apparatus. PoS lesions disrupted the landmark control of HD cells and also disrupted the stability of the preferred firing direction of the cells in darkness. Furthermore, PoS lesions impaired the stable HD cell representation maintained by path integration mechanisms when the rat walked between familiar and novel arenas. These results suggest that visual information first gains control of the HD cell signal in the LMN, presumably via the direct PoS ¡ LMN projection. This work addresses the problem of identifiability, that is, the question of whether parameters can be recovered from data, for linear compartmental models. Using standard differential algebra techniques, the question of whether a given model is generically locally identifiable is equivalent to asking whether the Jacobian matrix of a certain coefficient map, arising from input-output equations, is generically full rank. A natural next step is to study the set of parameter values where the Jacobian matrix drops in rank, which we refer to as the locus of non-identifiable parameter values , or, for short, the singular locus. In this work, we give a formula for coefficient maps in terms of acyclic subgraphs of the model's underlying directed graph and, then, study the case when the singular locus is defined by a single equation, the singular-locus equation. We prove that the singular-locus equation can be used to determine when submodels are generically locally identifiable. We also determine the singular-locus equation for two families of linear compartmental models, cycle and mammillary (star) models with input and output in a single compartment. We also state a conjecture for the corresponding equation for a third family: catenary (path) models. Finally, we introduce the identifiability degree , which is the number of parameter values that map to a generic input-output data vector. This degree was previously computed for mammillary and catenary models, and here we determine this degree for cycle models. [ABSTRACT FROM AUTHOR]

Transgenic mouse lines are instrumental in our attempt to understand brain function. Promoters driving transgenic expression of the gene encoding Cre recombinase are crucial to ensure selectivity in Cre-mediated targeting of floxed alleles using the Cre-Lox system. For the study of dopamine (DA) neurons, promoter sequences driving expression of the Dopamine transporter ( Dat ) gene are often implemented and several DAT-Cre transgenic mouse lines have been found to faithfully direct Cre activity to DA neurons. While evaluating an established DAT-Cre mouse line, reporter gene expression was unexpectedly identified in cell somas within the amygdala. To indiscriminately explore Cre activity in DAT-Cre transgenic lines, systematic whole-brain analysis of two DAT-Cre mouse lines was performed upon recombination with different types of floxed reporter alleles. Results were compared with data available from the Allen Institute for Brain Science. The results identified restricted DAT-Cre-driven reporter gene expression in cell clusters within several limbic areas, including amygdaloid and mammillary subnuclei, septum and habenula, areas classically associated with glutamatergic and GABAergic neurotransmission. While no Dat gene expression was detected, ample co-localization between DAT-Cre-driven reporter and markers for glutamatergic and GABAergic neurons was found. Upon viral injection of a fluorescent reporter into the amygdala and habenula, distinct projections from non-dopaminergic DAT-Cre neurons could be distinguished. The study demonstrates that DAT-Cre transgenic mice, beyond their usefulness in recombination of floxed alleles in DA neurons, could be implemented as tools to achieve selective targeting in restricted excitatory and inhibitory neuronal populations within the limbic neurocircuitry., (Copyright © 2019 Papathanou et al.)
