Aim: Marginal tree populations, either those located at the edges of the species' range or in suboptimal environments, are often a valuable genetic resource for biological conservation. However, there is a lack of knowledge about the genetic consequences of population marginality, estimated across entire species' ranges. Our study addresses this gap by providing information about several genetic indicators and their variability in marginal and core populations identified using quantitative marginality indices. Location: Southwestern Europe and North Africa. Methods: Using 10,185 SNPs across 82 populations of maritime pine (Pinus pinaster Ait.), a widespread conifer characterised by a fragmented range, we modelled the relationship of seven genetic indicators potentially related to population evolutionary resilience, namely genetic diversity (based on both all SNPs and outlier SNPs), inbreeding, genetic differentiation, recessive genetic load and genomic offset, with population geographical, demo‐historical and ecological marginality (as estimated by nine quantitative indices). Models were constructed for both regional (introducing gene pool as a random factor) and range‐wide spatial scales. Results: We showed a trend towards decreasing overall genetic diversity and increasing differentiation with geographic marginality, supporting the centre‐periphery hypothesis (CPH). However, we found no correlation between population inbreeding and marginality, while geographically marginal populations had a lower recessive genetic load (only models without the gene pool effect). Ecologically marginal populations had a higher genomic offset, suggesting higher maladaptation to future climate, albeit some of these populations also had high genetic diversity for climate outliers. Main Conclusions: Overall genetic diversity (but not outlier‐based estimates) and differentiation patterns support the CPH. Ecologically marginal populations and those at the southern edge could be more vulnerable to climate change due to higher climate maladaptation, as predicted by genomic offsets, and/or lower potentially adaptive genetic diversity. First, we studied the differences in climate and tree cover change in the two populations. Second, we analyzed the direct and indirect roles of local climate conditions and seed availability on breeding performance over 10 years in each population. Finally, we explored the influence of tree cover change on the occupancy dynamics in the two populations. Our results showed contrasting responses between populations: in Montseny, asynchronous seed production between oaks and beech precluded skip breeding, and breeding performance increased with seed availability. Furthermore, dormice in Montseny may use pollen production to anticipate the amount of beech nut resources and adjust their breeding effort. Boxes showed higher occupancy and colonization and fewer extinctions in Montseny than in Montnegre, where seed availability did not drive breeding performance. Results from Montnegre suggest that skip breeding was an adaptive response to a more pulsed, harsher environment. Here, females produced a similar number of pups than at Montseny. Long‐term studies dealing with population responses in marginal habitats can lead to a deeper understanding of the capacities of organisms to adapt to harsh environments. Chromosomal variability in peripheral populations of malaria mosquitoes of the Anopheles genus (Diptera, Culicidae) inhabiting the territory of Karelia has been studied. The modern northern limits of the ranges inhabited by sibling species of malarial mosquitoes An. beklemishevi, An. daciae, An. messeae s. s. and An. maculipennis were established. After 2010, the boundary of distribution of malaria mosquitoes shifted northward by 170 km, from the 65th parallel to the Arctic Circle. Inversion heterozygotes XL1, XL2, 2R2, 3R1, and 3R5 were found in peripheral populations of An. beklemishevi. Peripheral populations of An. messeae s. s. were homozygous for inversion of sex chromosome XL1 and differed in the frequencies of autosome inversions from the middle taiga populations. The frequency of heterozygotes for autosomal inversions 2R1, 3R1, and 3L1 increased in populations at the edge of the species range. Aim of study: Marine climatic transition zones are boundary areas of major climate zones, here the boundary between the subtropical and temperate zones. They present areas containing high abundance of organisms living at the limit of their physiological tolerance. These marginal populations are particularly sensitive to changes in their environment. As such, marine climatic transition zones are excellent natural playgrounds for climate change-related hypothesis testing, especially with respect to marine habitat response to ocean warming. The marginal biogenic habitats around Jeju Island, South Korea, which lies within the temperate transition zone, have gradually changed from macroalgal-dominated to hard coral-dominated habitats. Understanding the specific abiotic environmental factors that influence the distribution of the marginal populations in temperate transition zones (i.e., species at their occurrence limit) is crucial to predicting and managing temperate zone habitat changes caused by climate change. This study aims to identify the specific abiotic environmental factors that contribute to explaining the current spatial distribution of the declining temperate and expanding subtropical foundation species in Jeju waters. Methods: Coverage and composition of sessile benthic communities were determined by photo-quadrat analysis at two depths (10 m and 15 m) at three sites along the island's south, east, and north coasts in May and November 2022. Divergences in community composition between sites were characterized in light of ten quantitative environmental parameters. Results: Our results show that sessile foundation communities vary significantly at different sites around the island. While the south is defined by high-latitude hard corals, predominately Alveopora japonica, the east is defined by the temperate canopy-forming macroalga Ecklonia cava, and the north is characterized by coralline algae. Winter sea surface temperature, water transparency, nutrient concentration, and water movement were statistically the most impactful environmental factors determining which foundation species constitute each distinct benthic community. Context: Habitat fragmentation is expected to erode genetic diversity, which instead needs to be preserved for promoting species adaptation to a changing climate. As this expectation has found mixed support in forest trees, consistent results on the genetic consequences of fragmentation requires adequately replicated experimental designs, as well as an explicit assessment of which landscape features, if any, could mitigate its detrimental effects. Objective: Evaluating the role of several landscape attributes in buffering the detrimental effects of fragmentation in two metapopulations of silver fir. Methods: We genotyped 904 silver fir (Abies alba Mill.) trees from 18 local populations forming two metapopulations comparable for size and extension in the Apennines, a Mediterranean mountain range. We identified the signatures left by the fragmentation process on the genetic features of silver fir local populations. After removing potentially confounding effects due to different evolutionary histories, we used a multivariate approach for testing the relative effect of demographic, geographic, environmental and topographic factors on genetic features of both metapopulations. Results: We found comparable signals of the habitat fragmentation impact on the genetic diversity and structure of both investigated metapopulations. Fragmentation effects were less pronounced in the largest local populations (but not the least isolated), located on gentler slopes with higher soil water availability and lower heat exposure. Conclusions: Our results suggest the existence of a set of demographic and environmental factors that could have coherently buffered the detrimental genetic effects of fragmentation in both metapopulations. These findings could be useful to plan landscape restoration for the evolutionary rescue of mixed forests that once characterized Mediterranean mountain ecosystems. [ABSTRACT FROM AUTHOR]

Fundamental understanding of paleoecological proxies is necessary when attempting to compare, complement, or contrast two or more methods, and lack thereof may lead to erroneous conclusions. This comment addresses three such misunderstandings found in a recently published paper by Paillard et al. regarding soil macrofossil charcoal analysis (SMCA) and its relationship to lacustrine sediment analysis. The aim is to correct some misinformation associated with the following three assertions: (1) Broadleaved tree species produce fewer charcoal fragments than coniferous species. Although coniferous stands are more fire-prone, experimental burning shows that species with denser wood, that is, broadleaves, produce greater amounts of charcoal under similar fire conditions. (2) Preservation of charcoal particles is poor at the referenced study site. Once buried in the mineral soil compartment, charcoal particles remain quite stable. As such, SMCA has revealed Late Pleistocene marginal stands of broadleaved species. (3) Underestimating the importance of range-edge dynamics on the results of SMCA reconstructions. SMCA offers a stand-scale historical reconstruction that has proven well-suited to study peripheral stands and to reflect the heterogeneity of a landscape mosaic. By attempting to reconcile the SMCA (in situ) and lake sediments (ex situ) narratives, Paillard et al. missed one key aspect of comparing complementary proxies: they show different aspects of the past. Why species that in their core areas mainly reproduce sexually become enriched with clones in marginal populations ("geographic parthenogenesis") remains unclear. Earlier hypotheses have emphasized that selection might promote clonality because it protects locally adapted genotypes. On the other hand, it also hampers recombination and adaptation to changing conditions. The aim of the present study was to investigate the early stages of range expansion in a partially clonal species and what drives an increase in cloning during such expansion. We used genome‐wide sequencing to investigate the origin and evolution of large clones formed in a macroalgal species (Fucus vesiculosus) during a recent expansion into the postglacial Baltic Sea. We found low but persistent clonality in core populations, while at range margins, large dominant clonal lineages had evolved repeatedly from different sexual populations. A range expansion model showed that even when asexual recruitment is less favourable than sexual recruitment in core populations, repeated bottlenecks at the expansion front can establish a genetically eroded clonal wave that spreads ahead of a sexual wave into the new area. Genetic variation decreases by drift following repeated bottlenecks at the expansion front. This results in the emerging clones having low expected heterozygosity, which corroborated our empirical observations. We conclude that Baker's Law (clones being favoured by uniparental reproductive assurance in new areas) can play an important role during range expansion in partially clonal species, resulting in a complex spatiotemporal mosaic of clonal and sexual lineages that might persist during thousands of generations. In the context of global warming, a clear understanding of microrefugia—microsites enabling the survival of species populations outside their main range limits—is crucial. Several studies have identified forcing factors that are thought to favor the existence of microrefugia. However, there is a lack of evidence to conclude whether, and to what extent, the climate encountered within existing microrefugia differs from the surrounding climate. To investigate this, we adopt a “bottom-up” approach, linking marginal disconnected populations to microclimate. We used the southernmost disconnected and abyssal populations of the circumboreal herbaceous plant Oxalis acetosella in Southern France to study whether populations in sites matching the definition of “microrefugia” occur in particularly favorable climatic conditions compared to neighboring control plots located at distances of between 50 to 100 m. Temperatures were recorded in putative microrefugia and in neighboring plots for approximately 2 years to quantify their thermal offsets. Vascular plant inventories were carried out to test whether plant communities also reflect microclimatic offsets. We found that current microclimatic dynamics are genuinely at stake in microrefugia. Microrefugia climates are systematically colder compared to those found in neighboring control plots. This pattern was more noticeable during the summer months. Abyssal populations showed stronger offsets compared to neighboring plots than the putative microrefugia occurring at higher altitudes. Plant communities demonstrate this strong spatial climatic variability, even at such a microscale approach, as species compositions systematically differed between the two plots, with species more adapted to colder and moister conditions in microrefugia compared to the surrounding area. Premise: Understanding mechanisms fostering long‐term persistence of marginal populations should provide key insights about species resilience facing climate change. Cone serotiny is a key adaptive trait in Pinus banksiana (jack pine), which shows phenotypic variation according to the fire regime. Compared to range‐core populations within the fire‐prone boreal forest, low and variable serotiny in rear‐edge populations suggest local adaptation to uncommon and unpredictable wildfire regime. We assessed environmental/physiological factors that might modulate intraspecific variation in cone serotiny. Methods: We experimentally subjected closed cones to incrementing temperatures, then tested seed germination to determine whether and how various ecological factors (cone age, branch height, tree size, tree age) are related to cone dehiscence and seed viability in jack pines from rear‐edge and range‐core populations in eastern Canada. Results: Cones from rear‐edge populations dehisce at a lower opening temperature, which increases with cone age. Cones from range‐core stands open at a more constant, yet higher temperature. Cones from rear‐edge stands take between 13 and 27 years to reach the level of serotiny achieved at the range core. At the rear edge, seed viability is steady (51%), whereas it decreases from 70% to 30% in 20 years at the range core. Conclusions: We inferred the mechanisms of a bet‐hedging strategy in rear‐edge populations, which ensures steady recruitment during fire‐free intervals and successful postfire regeneration. This capacity to cope with infrequent and unpredictable fire regime should increase the resilience of jack pine populations as global changes alter fire dynamics of the boreal forest. The Brazilian coast presents diverse abiotic characteristics, such as temperature, and we know that surface seawater temperatures are rising as a result of climate change. Therefore, to better interpret the effects of such changes, we chose to investigate Gracilariopsis tenuifrons, a species of red algae widely occurring on the Brazilian coast. We specifically evaluated the effect of different temperatures (18–35 °C) on growth rates (GRs), photosynthetic parameters, and pigment contents of female gametophytes from five different populations distributed along 7450 km of coastline, including Maranhão State (MA, 2.4°S), Rio Grande do Norte State (RN, 5.4°S), Bahia State (BA, 14.48°S), and São Paulo State (SP1, 23.5°S and SP2, 24.2°S). All samples tolerated temperatures between 18 and 33 °C, but none of them could tolerate temperatures of 35 °C, irrespective of the populations. However, the highest GRs varied among populations (MA, 23–30 °C; RN, 28–33 °C; BA, 23–28 °C; SP1, 25–28 °C; SP2, 28 °C). MA and BA populations showed the highest maximal measurement (µmax) of the thermal performance curve when compared to SP1 and SP2 populations. The RN population showed the worst µmax values, 5.6% day−1; however, it was more resilient to stressful temperatures (33 °C) than the others. The BA population presented some physiological characteristics similar to populations from the northeastern and southeastern regions. Our data showed that Gp. tenuifrons presents latitudinal ecotypes associated with temperature. Northeastern populations could be more vulnerable in a drastic scenario of climate change (up to 4 °C); otherwise, this species could occupy regions further to the south of its current distribution. Opuntia is the most diverse and widespread genus in the Cactaceae; several species have broad distribution ranges and have successfully adapted to distinct habitats. One explanation for their success is the diversity of their reproductive systems. Opuntia macrocentra is one of the most common widespread species in North American deserts. A previous work on a northern population documented that this species has a mixed‐mating system and receives a low frequency of pollinators. In a southern population of O. macrocentra, we described its floral visitors, anthesis schedules and flower morphometry. Through controlled pollination experiments (selfing, outcrossing, supplementary pollen and natural pollination), we assessed the breeding and mating systems and measured pollen limitation. Eight different bee species visited the flowers. Based on their conduct, Diadasia rinconis and Lasioglossum sp. were the main pollinators. Flowers were homogamous and herkogamous. Floral traits and pollination experiments showed that O. macrocentra is xenogamous and self‐incompatible. Fruit set of the natural pollination, outcrossing and supplementary pollen treatments were not statistically different. Seed set was higher in natural pollination and the pollen limitation index for seed set was negative. Based on these results, we conclude that O. macrocentra is not pollen limited in the studied population. These results contrast with those of the previously studied population; therefore, we caution about making generalizations about the reproductive system based solely on studies in one population. The diversity of mating systems displayed by O. macrocentra may explain its broad distribution and adaptation to different environmental factors, one of which may be pollinator availability. Our results highlight the need to describe the reproductive ecology of populations instead of species, because reproductive systems and ecological factors such as plant–animal interactions may differ across their distribution range. Abstract Small, isolated populations present a challenge for conservation. The dueling effects of selection and drift in a limited pool of genetic diversity make the responses of small populations to environmental perturbations erratic and difficult to predict. This is particularly true at the edge of a species range, where populations often persist at the limits of their environmental tolerances. Populations of cisco, Coregonus artedi, in inland lakes have experienced numerous extirpations along the southern edge of their range in recent decades, which are thought to result from environmental degradation and loss of cold, well‐oxygenated habitat as lakes warm. Yet, cisco extirpations do not show a clear latitudinal pattern, suggesting that local environmental factors and potentially local adaptation may influence resilience. Here, we used genomic tools to investigate the nature of this pattern of resilience. We used restriction site‐associated DNA capture (Rapture) sequencing to survey genomic diversity and differentiation in southern inland lake cisco populations and compared the frequency of deleterious mutations that potentially influence fitness across lakes. We also examined haplotype diversity in a region of the major histocompatibility complex involved in stress and immune system response. We correlated these metrics to spatial and environmental factors including latitude, lake size, and measures of oxythermal habitat and found significant relationships between genetic metrics and broad and local factors. High levels of genetic differentiation among populations were punctuated by a phylogeographic break and residual patterns of isolation‐by‐distance. Although the prevalence of deleterious mutations and inbreeding coefficients was significantly correlated with latitude, neutral and non‐neutral genetic diversity were most strongly correlated with lake surface area. Notably, differences among lakes in the availability of estimated oxythermal habitat left no clear population genomic signature. Our results shed light on the complex dynamics influencing these isolated populations and provide valuable information for their conservation.

Marginal and peripheral populations are important for biodiversity conservation. Their original situation in a species’ geographic and ecological space often confers them genetic diversity and traits of high adaptive value. Yet theoretical hypotheses related to marginality are difficult to test because of confounding factors that influence marginality, namely environment, geography, and history. There is an urgent need to develop metrics to disentangle these confounding factors. We designed nine quantitative indices of marginality and peripherality that define where margins lie within species distributions, from a geographical, an environmental and a historical perspective. Using the distribution maps of eight European forest tree species, we assessed whether these indices were idiosyncratic or whether they conveyed redundant information. Using a database on marginal and peripheral populations based on expert knowledge, we assessed the capacity of the indices to predict the marginality status of a population. There was no consistent pattern of correlation between indices across species, confirming that the indices conveyed different information related to the specific geometry of the species distributions. Contrasting with this heterogeneity of correlation patterns across species, the relative importance of the indices to predict the marginality status of populations was consistent across species. However, there was still a significant country effect in the marginality status, showing a variation in expert opinion of marginality vis-á-vis the species distribution. The marginality indices that we developed are entirely based on distribution maps and can be used for any species. They pave the way for testing hypotheses related to marginality and peripherality, with important implications in quantitative ecology, genetics, and biodiversity conservation.

Assisted gene flow (AGF) can restore fitness in small plant populations. Due to climate change, current fitness patterns could vary in the future ecological scenario, as highly performant lineages can undergo maladaptation under the new climatic contexts. Peripheral populations have been argued to represent a potential source of species adaptation against climate change, but experimental evidence is poor. This paper considers the consequences of within- and between-population mating between a large core population and the southernmost population, the rare Dianthus guliae , to evaluate optimal AGF design under current and future conditions. We performed experimental self-pollinations and within- and between-population cross-pollinations to generate seed material and test its adaptive value to aridity. Seed germination, seedling growth and survival were measured under current and expected aridity. Effects of population type, pollination treatment and stress treatment on fitness components were analysed by generalized linear models. Relative measures of inbreeding depression and heterosis were taken under different stress treatments. Self-pollination reduced fitness for all the considered traits compared to within- and between-population cross-pollination. Under current aridity regime, the core population expressed higher fitness, and a larger magnitude of inbreeding depression. This indicated the core unit is close to its fitness optimum and could allow for restoring the fitness of the small peripheral population. Contrarily, under increased aridity, the fitness of outbred core lineages decreased, suggesting the rise of maladaptation. In this scenario, AGF from the small peripheral population enhanced the fitness of the core unit, whereas AGF from the core population promoted a fitness loss in the peripheral population. Hence, the small peripheral population could improve fitness of large core units versus climate change, while the contrary could be not true. Integrating reciprocal breeding programmes and fitness analyses under current and predicted ecological conditions can support optimal AGF design in a long-term perspective. [ABSTRACT FROM AUTHOR]

Background: The Chinese mitten crab (Eriocheir sinensis) and the Japanese mitten crab (E. japonica) of the family Varunidae, which are also critical fishery species in their native habitats, including China, Japan, and South Korea, exhibit a rare migration life history for the decapod life cycle. Eriocheir sinensis and E. japonica in South Korea may have originated from speciation or secondary contact in South Korea after speciation of these two species; however, the genetic relationship between these South Korean populations remains unclear, and need to be clarified. Objective: This study examined the population genetic properties of E. sinensis and E. japonica in South Korea to reveal the origin of their co-existence. Methods: Mitochondrial DNA cytochrome c oxidase I sequences of 120 E. sinensis and E. japonica from 6 sampling sites in South Korea were sequenced. Their genetic diversity and haplotype data were compared to previously reported data from the Chinese mainland, Taiwan, Japan, and Russia. Results: The haplotype network, analysis of molecular variance, FST values, Mantel test, and Bayesian skyline plot results of South Korean E. sinensis and E. japonica demonstrated that E. sinensis and E. japonica were isolated from their central populations, Chinese and Japanese populations, by the distance due to the temperature, salinity, ocean current flow, or a geohistorical event. In addition, genetic analysis indicated that the South Korean populations of the two species were marginal populations, exhibiting low genetic diversity, significant negative neutrality values, and radial haplotype networks. The current results also suggest that the Geum River is an overlapping habitat for the natural populations of E. sinensis and E. japonica in South Korea. Further research is required to examine the geohistorical and evolutionary event between E. sinensis and E. japonica in South Korea. Conclusions: The co-existence of E. sinensis and E. japonica in South Korea may have originated from secondary contact after their speciation. [ABSTRACT FROM AUTHOR]

Key message: Lowered pollen and seed quality of B. humilis compared to its widespread tree relatives may threaten its populations at the southwestern edge of the species range. Inability to produce vital propagules is the main threat to the persistence of rare plants at the edges of species distribution. In the present investigation, factors responsible for the low germinability of shrub birch (Betula humilis) seeds in the peripheral populations were analysed. As shrub birch populations are found in phosphorus (P)-limited peatlands with high groundwater levels, the impact of groundwater PO43− ion contents and groundwater table for B. humilis flowering was also studied. Qualities of the B. humilis seeds and pollen were compared to those of tree congeners silver birch (B. pendula) and downy birch (B. pubescens), which usually represent a high reproductive performance. We revealed that the poor germination ability of B. humilis seeds was caused by the high proportions of empty seeds (44.8%) and seeds with partially developed embryos (47.3%). The insufficient numbers of viable seeds may be a consequence of low-quality shrub birch pollen, as only 1.6% of pollen grains sprouted. The germinability of shrub birch pollen was significantly lower than that of Betula tree pollen; thus, there is a risk of pollination of B. humilis female flowers by pollen grains of tree birches. Positive relationships between the P concentrations in seeds and seed germinability were revealed in tree birches, but not in B. humilis. The availability of PO43− ions in the groundwater did not affect B. humilis flowering, but male inflorescences were more numerous in stands with higher groundwater levels. Small, isolated populations present a challenge for conservation. The dueling effects of selection and drift in a limited pool of genetic diversity make the responses of small populations to environmental perturbations erratic and difficult to predict. This is particularly true at the edge of a species range, where populations often persist at the limits of their environmental tolerances. Populations of cisco, Coregonus artedi, in inland lakes have experienced numerous extirpations along the southern edge of their range in recent decades, which are thought to result from environmental degradation and loss of cold, well‐oxygenated habitat as lakes warm. Yet, cisco extirpations do not show a clear latitudinal pattern, suggesting that local environmental factors and potentially local adaptation may influence resilience. Here, we used genomic tools to investigate the nature of this pattern of resilience. We used restriction site‐associated DNA capture (Rapture) sequencing to survey genomic diversity and differentiation in southern inland lake cisco populations and compared the frequency of deleterious mutations that potentially influence fitness across lakes. We also examined haplotype diversity in a region of the major histocompatibility complex involved in stress and immune system response. We correlated these metrics to spatial and environmental factors including latitude, lake size, and measures of oxythermal habitat and found significant relationships between genetic metrics and broad and local factors. High levels of genetic differentiation among populations were punctuated by a phylogeographic break and residual patterns of isolation‐by‐distance. Although the prevalence of deleterious mutations and inbreeding coefficients was significantly correlated with latitude, neutral and non‐neutral genetic diversity were most strongly correlated with lake surface area. Notably, differences among lakes in the availability of estimated oxythermal habitat left no clear population genomic signature. Aim: The centre‐periphery hypothesis (CPH) states that peripheral populations exhibit lower genetic diversity, abundance and size, and higher differentiation compared to central populations, due to a decline in environmental suitability towards range margins. Here, we tested if neutral genetic diversity and functional leaf trait diversity fit the predicted patterns of the CPH. Location: Cerrado, Brazil. Taxon: Qualea grandiflora (Vochysiaceae) and Annona crassiflora (Annonaceae). Methods: Variation in eight functional leaf traits and nine or eight microsatellite loci were analysed in 37 populations (777 individuals) of Q. grandiflora and 21 populations (397 individuals) of A. crassiflora. We tested CPH using three distinct centres of species' range: geographical (centre of species' geographic occurrence), historical (centre of species' refugia) and ecological (centre of species' current suitability areas). Generalised linear regressions were performed between genetic diversity and differentiation, coefficients of variation and means of leaf metamer traits of populations and their distances from each centre. Results: A decrease in allele richness was observed from central towards peripheral populations for ecological (both species) and geographical centres (Q. grandiflora). Overall, the results for both species pointed to a decrease in metamer vigour towards marginal populations, pattern consistent with a decrease in environmental suitability towards periphery as predicted by CPH. Besides geographical and ecological centres, leaf traits fitted CPH considering historical centre. Main Conclusions: Results for leaf traits and genetic diversity of two phylogenetically distant species indicate the consistency of the CPH for Cerrado species. Our results highlight the importance of considering distinct centres and a great number of populations along species' range to better determinate the processes underlying the distribution of genetic diversity and functional traits. The low genetic diversity and metamer vigour in marginal populations can explain the Cerrado endemism of studied species and suggest that the ongoing climatic changes can be critical for their survival. Many species cannot either migrate or adapt at the rate of temperature increases due to climate warming. Therefore, they need active conservation strategies to avoid extinction. Facilitated adaptation actions, such assisted gene flow, aim at the increase of the evolutionary resilience of species affected by global change. In elevational gradients, marginal populations at the lower elevation edges are experiencing earlier snowmelt and higher temperatures, which force them to adapt to the new conditions by modifying their phenology. In this context, advancing the onset of flowering and seed germination times are crucial to ensure reproductive success and increase seedling survival prior to summer drought. Assisted gene flow may bring adaptive alleles and increase genetic diversity that can help throughout ontogeny. The main aim of this work is to assess the effects that different gene flow treatments could have on the desired trait changes in marginal populations. Accordingly, we established a common garden experiment in which we assayed four different gene flow treatments between Silene ciliata Pourr. (Caryophyllaceae) populations located in similar and different elevation edges, belonging to the same and different mountains. As a control treatment, within-population crosses of low elevation edge populations were performed. The resulting seeds were sown and the germination and flowering onset dates of the resulting plants recorded, as well as the seedling survival. Gene flow between populations falling on the same mountain and same elevation and gene flow from high-elevation populations from a different mountain to low-elevation populations advanced seed germination time with respect to control crosses. No significant effects of gene flow on seedling survival were found. All the gene flow treatments delayed the onset of flowering with respect to control crosses and this effect was more pronounced in among-mountain gene flows. The results of this study highlight two important issues that should be thoroughly studied before attempting to apply assisted gene flow in practical conservation situations. Firstly, among-populations gene flow can trigger different responses in crucial traits throughout the ontogeny of plant species. Secondly, the population provenance of gene flow is determinant and plays a significant role on the effects of gene flow.

Abstract Background Temperature is one of the most important abiotic factors limiting plant growth and productivity. Many plants exhibit cold acclimation to prepare for the likelihood of freezing as temperatures decrease towards 0 °C. The physiological mechanisms associated with enabling increased tolerance to sub-zero temperatures vary between species and genotypes. Geographically and climatically diverse populations of Arabidopsis lyrata ssp. petraea were examined for their ability to survive, maintain functional photosynthetic parameters and cellular electrolyte leakage integrity after being exposed to sub-zero temperatures. The duration of cold acclimation prior to sub-zero temperatures was also manipulated (2 and 14 days). Results We found that there was significant natural variation in tolerances to sub-zero temperatures among populations of A. petraea. The origin of the population affected the acclimation response and survival after exposure to sub-zero temperatures. Cold acclimation of plants prior to sub-zero temperatures affected the maximum quantum efficiency of photosystem II (PSII) (F v /F m ) in that plants that were cold acclimated for longer periods had higher values of F v /F m as a result of sub-zero temperatures. The inner immature leaves were better able to recover F v /F m from sub-zero temperatures than mature outer leaves. The Irish population (Leitrim) acclimated faster, in terms of survival and electrolyte leakage than the Norwegian population (Helin). Conclusion The ability to survive, recover photosynthetic processes and cellular electrolyte leakage after exposure to sub-zero temperatures is highly dependent on the duration of cold acclimation.

The evolutionary potential of populations inhabiting marginal areas has been extensively debated and directly affects their conservation value. Gene flow is one of the main factors influencing selection, adaptive potential and thus, local adaptation processes in marginal areas. The effects of differential gene flow provenance are still not well understood, since studies on gene flow between marginal populations have been underrepresented in the literature. This kind of gene flow can be especially beneficial because it can provide both adaptive allelic combinations originated under similar environmental conditions and genetic variation on which selection can act.We conducted a study on the effects of different gene flow provenance on marginal populations of Mediterranean alpine Silene ciliata Pourret (Caryophyllaceae) replicated in three mountain ranges of Central Spain. The delineation of optimal and marginal areas of the species distribution was based on environmental differentiation and relied on the ecological definition of centrality and marginality. We experimentally tested the effect of three different types of pollen‐mediated gene flow on germination rate, seedling size and survival rate in marginal populations and assessed their effects by establishing in situ common gardens. To further assess the evolutionary potential of marginal populations, we performed a reciprocal sowing experiment and measured the same fitness components to determine the extent of local adaptation.We found that gene flow between marginal populations improved germination rate and seedling survival with regard to gene flow from optimal to marginal populations and within marginal populations. In reciprocal sowing experiments, seedling survival rate was higher when the seed source was from marginal areas than when it was from optimal areas in both marginal and optimal sowing sites.Synthesis. Our results suggest that gene flow between marginal populations from similar environmental conditions increases the fitness of the recipient population by increasing genetic diversity and simultaneously providing adaptive alleles generated under similar selective pressures. Results also highlight the adaptive potential of marginal populations as genetic diversity from marginal areas may provide a fitness advantage to the populations in optimal areas. In the main distribution area the genetic pattern of silver birch is dominated by two haplotypes: haplotype A located in the western and north-western Europe, and haplotype C in eastern and southeastern Europe, characterized by high levels of neutral genetic variability within populations, and low differentiation among populations. Information about the amount and structure of genetic variation in the southern marginal areas, representing rear populations left during the expansion of this species from southern glacial refugia, are lacking. The general aim of the study was to investigate the existence of the climatic characteristics typical of the environmental niche of the species, jointly to genetic organization, variation and gene flow, in marginal populations on the Italian Apennines and Greek Southern Rhodope and compare them with populations of the southern part of the main distribution range on the Alps and Balkans. Genetic analysis was performed using nuclear microsatellites loci on 311 trees sampled from 14 populations. Environmental analysis was performed on the multivariate analysis of derived climatic variables. The allelic pattern was analyzed to assess genetic diversity, population diversity and differentiation, population structure and gene flow. The geographic and environmental peripherality did not always match, with some Apennine sites at higher elevation enveloped in the environmental niche. In the peripheral populations on the Apennines, we observed a lower genetic diversity and higher differentiation, with evident genetic barriers detected around these sites. These characteristics were not shown in the marginal Greek populations. Unexpectedly, the southern Italian marginal populations showed genetic links with the Greek and central area of the distribution range. The Greek populations also showed evident gene flow with the Alpine and Balkan areas. The disparity of results in these two marginal areas show that it is not the geographic peripherality or even the ecological marginality that may shape the genetic diversity and structure of marginal populations, but primarily their position as part of the continuous range or as disjunct populations. This outcome suggests different considerations on how to manage their gene pools and the role that these rear populations can play in maintaining the biodiversity of this species.

Populations residing near species' low‐latitude range margins (LLMs) often occur in warmer and drier environments than those in the core range. Thus, their genetic composition could be shaped by climatic drivers that differ from those occurring at higher latitudes, resulting in potentially adaptive variants of conservation value. Such variants could facilitate the adaptation of populations from other portions of the geographical range to similar future conditions anticipated under ongoing climate change. However, very few studies have assessed standing genetic variation at potentially adaptive loci in natural LLM populations. We investigated standing genetic variation at single nucleotide polymorphisms (SNPs) located within 117 candidate genes and its links to putative climatic selection pressures across 19 pedunculate oak (Quercus robur L.) populations distributed along a regional climatic gradient near the species' southern range margin in southeastern Europe. These populations are restricted to floodplain forests along large lowland rivers, whose hydric regime is undergoing significant shifts under modern rapid climate change. The populations showed very weak geographical structure, suggesting extensive genetic connectivity and gene flow or shared ancestry. We identified eight (6.2%) positive FST‐outlier loci, and genotype–environment association analyses revealed consistent associations between SNP allele frequencies and several climatic variables linked to water availability. A total of 61 associations involving 37 SNPs (28.5%) from 35 annotated genes provided important insights into putative functional mechanisms in our system. Our findings provide empirical support for the role of LLM populations as sources of potentially adaptive variation that could enhance species' resilience to climate change‐related pressures. Range marginal populations are often perceived to have lower conservation value compared to those in the core area. The allocation of resources to maintain peripheral populations is therefore often questioned. The sage-leaved rockrose (Cistus salviifolius L.) is a self-incompatible and obligate seeder widely distributed in the Mediterranean area but rare and patchily distributed in Switzerland at its range margin on the southern slopes of the Alps. Here, we combined analysis of genetic diversity with pollinator surveys and field studies of reproductive ecology to compare peripheral Cistus populations in the Alps with range central populations in the Mediterranean. Our results showed no differences in genetic diversity between peripheral and central populations and between fragmented and connected ones at its range margin in the Alps. Although the fragmented populations were visited by more abundant and species richer pollinators (bees and wasps), they showed lower number of seeds and higher self-compatibility compared to the connected ones, which excludes the pollination limitation hypothesis. Overall, our study highlights that peripheral populations of C. salviifolius in the Alps are likely to contribute to maintain genetic diversity, while showing variation in reproductive ecology, and are therefore important for the conservation of this species. In the absence of barriers to dispersal, species' range limits should arise when physiological processes are compromised by unfavorable ecological conditions. This study aims to fill the knowledge gap on the role of physiology in determining plant species range limits. We developed the Physiological Marginality Hypothesis (PMH), a theoretical framework predicting that individuals in ecologically marginal populations are less physiologically efficient than those occurring at the ecological optimum. According to PMH, the probability to reach the optimal physiological efficiency is best shaped by a Gaussian function and curvilinear relationships should exist between physiological traits and ecological gradients. We conducted an analysis on plant physiology studies along altitudinal/latitudinal gradients to investigate which models best describe the relationship between the performance of a given physiological trait and the considered ecological gradients. Data from 54 cases (29 species) concerning photosynthesis, stomatal conductance, carbon isotope composition and photosynthetic water use efficiency were used. Overall, results support PMH in about 70% of cases, suggesting that ecologically marginal individuals can be physiologically less efficient than individuals occurring at the species ecological optimum. Climate warming is likely to shift the range margins of species poleward, but fine‐scale temperature differences near the ground (microclimates) may modify these range shifts. For example, cold‐adapted species may survive in microrefugia when the climate gets warmer. However, it is still largely unknown to what extent cold microclimates govern the local persistence of populations at their warm range margin. We located 99 microrefugia, defined as sites with edge populations of 12 widespread boreal forest understory species (vascular plants, mosses, liverworts and lichens) in an area of ca. 24,000 km2 along the species' southern range margin in central Sweden. Within each population, a logger measured temperature eight times per day during one full year. Using univariate and multivariate analyses, we examined the differences of the populations' microclimates with the mean and range of microclimates in the landscape, and identified the typical climate, vegetation and topographic features of these habitats. Comparison sites were drawn from another logger data set (n = 110), and from high‐resolution microclimate maps. The microrefugia were mainly places characterized by lower summer and autumn maximum temperatures, late snow melt dates and high climate stability. Microrefugia also had higher forest basal area and lower solar radiation in spring and autumn than the landscape average. Although there were common trends across northern species in how microrefugia differed from the landscape average, there were also interspecific differences and some species contributed more than others to the overall results. Our findings provide biologically meaningful criteria to locate and spatially predict potential climate microrefugia in the boreal forest. This opens up the opportunity to protect valuable sites, and adapt forest management, for example, by keeping old‐growth forests at topographically shaded sites. These measures may help to mitigate the loss of genetic and species diversity caused by rear‐edge contractions in a warmer climate. [ABSTRACT FROM AUTHOR]

Abstract The effect of population size on population genetic diversity and structure has rarely been studied jointly with other factors such as the position of a population within the species’ distribution range or the presence of mutualistic partners influencing dispersal. Understanding these determining factors for genetic variation is critical for conservation of relict plants that are generally suffering from genetic deterioration. Working with 16 populations of the vulnerable relict shrub Cneorum tricoccon throughout the majority of its western Mediterranean distribution range, and using nine polymorphic microsatellite markers, we examined the effects of periphery (peripheral vs. central), population size (large vs. small), and seed disperser (introduced carnivores vs. endemic lizards) on the genetic diversity and population structure of the species. Contrasting genetic variation (HE: 0.04–0.476) was found across populations. Peripheral populations showed lower genetic diversity, but this was dependent on population size. Large peripheral populations showed high levels of genetic diversity, whereas small central populations were less diverse. Significant isolation by distance was detected, indicating that the effect of long‐distance gene flow is limited relative to that of genetic drift, probably due to high selfing rates (FIS = 0.155–0.887), restricted pollen flow, and ineffective seed dispersal. Bayesian clustering also supported the strong population differentiation and highly fragmented structure. Contrary to expectations, the type of disperser showed no significant effect on either population genetic diversity or structure. Our results challenge the idea of an effect of periphery per se that can be mainly explained by population size, drawing attention to the need of integrative approaches considering different determinants of genetic variation. Furthermore, the very low genetic diversity observed in several small populations and the strong among‐population differentiation highlight the conservation value of large populations throughout the species’ range, particularly in light of climate change and direct human threats.

Different chromosomal forms of Deschampsia antarctica Desv. (Poaceae), including diploids (2n=26), hypotriploid (2n=36–38) and a genotype with an occasional occurrence of B chromosome (2n=26+0-1B) that originated from southern marginal populations (Argentine Islands region, maritime Antarctic) were studied using molecular cytogenetic, morphometric and biochemical methods. FISH analysis revealed variations in the number of rDNA sites between the diploid and hypotriploid plants. The genome size varied among plants with a different chromosome number and was on average 10.88 pg/2C for diploids and 16.46 pg/2C for hypotriploid. The mean values of leaf length of plants grown in vitro varied within a range of 5.23–9.56 cm. The total phenolic content ranged from 51.10 to 105.40 mg/g, and the total flavonoid content ranged from 1.22 to 4.67 mg/g. The amount of phenolic compounds did not differ significantly between the genotypes, while a variation in the flavonoid content was observed for L59 and DAR12. The diploids did not differ significantly among each other in terms of the number of rDNA loci, but differed slightly in their genome size. The individuals of DAR12 carrying B chromosome were similar to other diploids in terms of their genome size, but statistically differed in leaf length. The hypotriploid had both a greater number of rDNA sites and a larger genome size. No statistical correlations were observed between the genome size and leaf length or genome size and accumulation of phenolic and flavonoid compounds. The results of this study suggest that D. antarctica plants from the southern edge of the range are characterised by the heterogeneity of the studied parameters.

The study of the drivers that shape spatial genetic structure across heterogeneous landscapes is one of the main approaches used to understand population dynamics and responses in changing environments. While the Isolation-by-Distance model (IBD) assumes that genetic differentiation increases among populations with geographical distance, the Isolation-by-Resistance model (IBR) also considers geographical barriers and other landscape features that impede gene flow. On the other hand, the Isolation-by-Environment model (IBE) explains genetic differentiation through environmental differences between populations. Although spatial genetic studies have increased significantly in recent years, plants from alpine ecosystems are highly underrepresented, even though they are great suitable systems to disentangle the role of the different factors that structure genetic variation across environmental gradients. Here, we studied the spatial genetic structure of the Mediterranean alpine specialist Silene ciliata across its southernmost distribution limit. We sampled three populations across an altitudinal gradient from 1850 to 2400 m, and we replicated this sample over three mountain ranges aligned across an E-W axis in the central part of the Iberian Peninsula. We genotyped 20 individuals per population based on eight microsatellite markers and used different landscape genetic tools to infer the role of topographic and environmental factors in shaping observed patterns along the altitudinal gradient. We found a significant genetic structure among the studied Silene ciliata populations which was related to the orography and E-W configuration of the mountain ranges. IBD pattern arose as the main factor shaping population genetic differentiation. Geographical barriers between mountain ranges also affected the spatial genetic structure (IBR pattern). Although environmental variables had a significant effect on population genetic diversity parameters, no IBE pattern was found on genetic structure. Our study reveals that IBD was the driver that best explained the genetic structure, whereas environmental factors also played a role in determining genetic diversity values of this dominant plant of Mediterranean alpine environments.

The marginal populations of the Sahara blue-eyed pond turtle, Mauremys leprosa saharica, in the southern-most species distribution range in the pre-Saharan areas of north-west Africa, are faced with extreme environmental conditions of arid climate and anthropogenic and climate change mediated water and land salinisation. In the current study, we investigated a small and isolated population of M. l. saharica at Sidi El Mehdaoui oasis in the Lower Draa River, southern Morocco, in order to assess its osmo- and iono-regulatory abilities and tolerance to salinity and dehydration. Upon capture, turtles were weighed and measured for shell dimensions and blood and voided urine were taken. Tests of exposure to different levels of water salinity (0%, 35%, and 50% seawater) and maintenance out of water (estivation simulation) were carried out. Osmolalities and Na+, Cl−, K+, and urea concentrations were determined in plasma and voided urine, and glycaemia was measured in blood, before and after tests. Turtles were able to survive in brackish waters with a salinity as high as 24% seawater (8.4 ppt). Their voided urine was hypotonic to plasma, which indicated that they could use their bladder water reserves for osmo- and iono-regulation until the iso-osmocity level beyond which osmotic and ionic anhomeostasy can occur. Experimental tests showed that the osmo- and iono-regulatory capacities of these turtles are relatively limited, and not enough effective to allow them to survive for long-term periods in brackish/saline waters or out of water, because of dehydration indicated by progressive weight loss to a critical threshold. The increased drought, water salinisation and habitat fragmentation related to anthropogenic activities and climate change, represent great threats that can create habitats exceeding the species' threshold for a long-term persistence of the vulnerable small marginal populations of the Saharan pond turtle. Populations at range margins of marine organisms are at the front line of climate-induced changes in abiotic factors and are thus particularly susceptible. Especially, macroalgae that are externally reproducing rely on optimal environmental conditions for gamete release and subsequent fertilisation. However, the effect of climate change–forced decrease in salinity on these critical life stages has been largely overlooked. We tested the impact of forecasted hyposaline conditions on a marginal population of the rockweed Fucus vesiculosus growing at 5.8 PSU on the Finnish coast of the Baltic Sea. We incubated individuals with receptacles for at least seven days at 2.5, 4.1, 5.8, and 7.2 PSU and determined their gamete release and subsequent fertilisation success. We further tested sperm performance at 3.5, 5.0, and 6.1 PSU. Salinity of 2.5 PSU, which is predicted to occur in the region by the end of this century, reduced egg release. In contrast, sperm and antheridia release were not consistently affected by the different salinities, but the size of sperm swells at 3.5 PSU. Because fertilisation success was drastically reduced at 2.5 and 4.1 PSU, we suggest that sperm performance was compromised such that sperm dysfunction hampered fertilisation success. Our results demonstrate that the forecasted hyposalinity negatively affects egg release and sexual reproduction in F. vesiculosus at its northern distribution limit. Although spatial genetic studies have increased significantly in recent years, plants from alpine ecosystems are highly underrepresented, even though they are great suitable systems to disentangle the role of the different factors that structure genetic variation across environmental gradients. Here, we studied the spatial genetic structure of the Mediterranean alpine specialist Silene ciliata across its southernmost distribution limit. We sampled three populations across an altitudinal gradient from 1850 to 2400 m, and we replicated this sample over three mountain ranges aligned across an E-W axis in the central part of the Iberian Peninsula. We genotyped 20 individuals per population based on eight microsatellite markers and used different landscape genetic tools to infer the role of topographic and environmental factors in shaping observed patterns along the altitudinal gradient. We found a significant genetic structure among the studied Silene ciliata populations which was related to the orography and E-W configuration of the mountain ranges. IBD pattern arose as the main factor shaping population genetic differentiation. Geographical barriers between mountain ranges also affected the spatial genetic structure (IBR pattern). Although environmental variables had a significant effect on population genetic diversity parameters, no IBE pattern was found on genetic structure. Our study reveals that IBD was the driver that best explained the genetic structure, whereas environmental factors also played a role in determining genetic diversity values of this dominant plant of Mediterranean alpine environments. • Peripheral populations of Rocky Mountain lodgepole pine should be considered for conservation of genetic diversity. • Lodgepole pine life history includes large scale fire processes. • Reintroducing these fire processes, or their analogs, is appropriate for conservation. • Active management is suggested to emulate natural fire processes, using silviculture. The ability of plant species to persist in changing environments is in part due to inherent genetic variation. However, human perturbations, including climate change, can affect plant demographics in ways that fall outside the range of variability to which they were adapted, and that may threaten the maintenance of genetic diversity. Conservation of genetic diversity can help mitigate the effects of human modification to the environment but it is necessary to determine what populations are, or will be, at risk of local extirpation. Peripheral populations have been identified as important sources of genetic diversity and, when threatened by novel perturbations, should be considered for immediate conservation attention. Peripheral populations of Rocky Mountain lodgepole pine (Pinus contorta var. latifolia) in the Intermountain West serve as an example for conservation of genetic diversity. Lodgepole pine life history attributes indicate a strong evolutionary history with large-scale fire suggesting the importance of stand-replacing disturbances for their population persistence. The practice of fire suppression, superimposed over large-scale adoption of a 'no-action' approach to management, has likely precluded fire from many peripheral populations of lodgepole pine. In addition, recent epidemics of mountain pine beetle (Dendroctonus ponderosae), and climate warming-related extensive and severe fires, that may become more frequent, could affect large areas of lodgepole pine, which further exacerbates the conservation problem. Active management is suggested to restore natural fire processes, or appropriate analogs, to lodgepole pine forests with the goal of maintaining genetic variation in the face of an increasingly uncertain future. Different chromosomal forms of Deschampsia antarctica Desv. (Poaceae), including diploids (2n=26), hypotriploid (2n=36-38) and a genotype with an occasional occurrence of B chromosome (2n=26+0-1B) that originated from southern marginal populations (Argentine Islands region, maritime Antarctic) were studied using molecular cytogenetic, morphometric and biochemical methods. FISH analysis revealed variations in the number of rDNA sites between the diploid and hypotriploid plants. The genome size varied among plants with a different chromosome number and was on average 10.88 pg/2C for diploids and 16.46 pg/2C for hypotriploid. The mean values of leaf length of plants grown in vitro varied within a range of 5.23-9.56 cm. The total phenolic content ranged from 51.10 to 105.40 mg/g, and the total flavonoid content ranged from 1.22 to 4.67 mg/g. The amount of phenolic compounds did not differ significantly between the genotypes, while a variation in the flavonoid content was observed for L59 and DAR12. The diploids did not differ significantly among each other in terms of the number of rDNA loci, but differed slightly in their genome size. The individuals of DAR12 carrying B chromosome were similar to other diploids in terms of their genome size, but statistically differed in leaf length. The hypotriploid had both a greater number of rDNA sites and a larger genome size. No statistical correlations were observed between the genome size and leaf length or genome size and accumulation of phenolic and flavonoid compounds. The performance of populations at the edge of specie’s distribution range may differ substantially from central populations. Here, we develop a modelling framework to estimate ecological niches (i.e. climatic) of four locally endangered plant species and measure the distance of marginal (geographically) populations to the species’ niche centroid in order to analyse whether marginal populations are outside of the optimal ecological niche of each species. Our results show that for three of the four studied species, which have their populations located at the margins of their distribution ranges, are also at the margins of their climate gradients. These results would support the hypothesis that marginality within the set of habitable conditions (i.e. ecological niche) may represent an important determinant on performance of some plant populations, as well as a plausible explanation to the degree of stagnancy or regression experienced by species in those regions where their populations are at the margin of their ecological niche and/or they are restricted to microrefugia with ecological conditions very different from those around them. Finally, this study aims to be a theoretical base from which to advance on, including other types of factors (e.g. biotic interactions, topography, human influence and population fluctuations through time), which will allow for a better understanding of the complex network of factors that occur in marginal plant populations. Background: Hybridisation can be a threat for the survival of a rare species because, in the case of insufficient numbers of appropriate mates, a rare form is much more likely to cross with a widespread taxon. Aims: In the present study, we tested hypotheses concerning the level of hybridisation between endangered Betula humilis and its widespread congeners: B. pendula and B. pubescens as a function of habitat conditions. Methods: We genotyped 312 individuals of three species using AFLP markers. B. humilis specimens were sampled in populations with low and high groundwater levels. Morphological identification of B. pubescens and B. pendula was verified using the Atkinson discriminant function. Results: Altogether, 15 individuals (4.8%) were indicated as putative hybrids. The B. humilis hybrids were found in dry habitats and they could be classified as F1 or F2 generation. Tree hybrids could represent backcrosses to either B. pendula or B. pubescens. Conclusions: Genetic analyses contradicted the idea that hybridisation between B. humilis and its close relatives was extensive. On the other hand, the presence of introgressed individuals in the populations in areas with low groundwater levels implied that pollen swamping might be a threat for declining B. humilis stands. Context Peripheral populations especially those in the leading edge are isolated from rest of the areas in the species distribution range. This can make them less genetically diverse yet genetically distinct from the rest of the populations in the species distribution range. Evaluation of their genetic diversity is thus crucial in understanding the local adaptation potential of a species. Aims We investigated the genetic diversity and genotype by environment interaction at the northern parts of the distribution range of P. avium. Methods Quantitative genetic variation of growth, stem form, and spring phenology were assessed in progenies from 93 plus trees of P. avium selected from 43 locations at the north of the species distribution range in Sweden and tested at two Swedish sites and one Danish site. Results We find large quantitative genetic variation in growth and phenology at the northern part of the distribution range of P. avium. Only a limited genotype by environment interaction was observed with no clear indication of local adaptation at the northern parts of the species distribution. Conclusion We conclude that P. avium harbors a high level of genetic diversity at the north of its distribution range. Present patterns therefore reflect more likely the recent introduction of the species and dispersal dynamics rather than a long-term loss of diversity along South-North ecological clines during the Holocene. With no indications of genetic depletion in growth or phenology, the gene pool in the breeding program is considered suitable for the future propagation of the species in the tested area. Abstract: Areas hosting hotspots of low‐latitude marginal populations of cold‐adapted plant species could be key areas for understanding geographical attributes that result in refugia during climatic shifts as well as the conservation of genetic diversity in the face of climate change. Low‐latitude populations of cold‐adapted plants are important because they may harbour the combination of alleles that foster persistence in a warmer climate. Consequently, identification of areas where arctic‐alpine, circumpolar and circumboreal species reach the low‐latitude ends of their distribution will present a unique opportunity to uncover processes that shaped current biogeographical patterns, as well as prepare for future scenarios. Here, we identify 35 main marginal population hotspots (19 and 16 areas in North America and Europe, respectively) of 183 plant taxa. These hotspots represent areas where southern marginal populations of cold‐adapted species co‐occur. The identification of hotspots was based on geographic overlap of southernmost locations of the target species, in a 50 × 50 km grid. With a threshold of two species in a single grid cell or in two contiguous cells, the analysis revealed that hotspots are in most cases located in the southern portion of major mountain chains. However, hotspots also occur in lowland areas at high latitudes (Fennoscandia, Alaska, Hudson Bay) which do not necessarily correspond to known cold‐ or warm‐stage refugia (e.g. Alps). Rockies and Sierra Nevada both in California and Spain, Apennines, and the southern Scandes, maintain their hotspot status even with more stringent cut‐off thresholds (>3 and >5 species per cell group). From a conservation point of view, our analysis reveals that only a small portion of the hotspots are currently included within protected areas. We discuss the importance of marginal population hotspots to future research on climate change and, finally, outline how conservation strategies can capitalize on the knowledge gained from studying climate change effects on cold‐adapted plants. Because of harsh conditions, suboptimal habitat quality and poor connectivity to other populations, plant populations at the margin of a distribution are expected to be less genetically diverse, but to be more divergent from each other than populations in the centre of a distribution. In northern Europe, northern marginal populations may also be younger than populations further to the south, and may have had less time to accumulate genetic diversity by mutation and gene flow. However, orchids have very small seeds, which are easily dispersed long distances by wind, and orchids are therefore expected to show less differentiation between marginal and central populations than other groups of seed plants. Here, we analysed whether Scandinavian populations of the tetraploid marsh orchid Dactylorhiza majalis subsp. majalis differ from central European populations in genetic diversity patterns. A total of 220 plants from eight central European and ten Scandinavian populations was examined for variation at five nuclear microsatellite loci, nuclear ITS and 13 polymorphic sites in noncoding regions of the plastid genome. The total genetic diversity was slightly lower in Scandinavia than in central Europe, both in plastid and nuclear markers, but the differences were small. Also, the Scandinavian populations were less diverse and somewhat more strongly differentiated from each other than the central European ones. Dactylorhiza majalis subsp. majalis has apparently colonized Scandinavia on multiple independent occasions and from different source areas in the south. Seed flow between Scandinavian populations has still not fully erased the patterns imprinted by early colonization. Our results suggest that marginal populations of orchids may be as important as central ones in preserving genetic diversity through Pleistocene glacial cycles. Peripheral sites usually offer suboptimal conditions for species with wide distributions, where expression of phenotypic variability and potential interspecific hybridization might be enhanced. The Patagonian steppe, the largest and southernmost dryland ecosystem in South America, is characterized by natural rangelands dominated by grasses. Festuca pallescens is a keystone species with a wide distribution in Patagonia over diverse environments reaching the extreme arid zones in the Somuncura plateau, a biogeographical island. Our aim is to study the phylogenetic relationships among Festuca pallescens populations as well as between this species and the sympatric F. argentina in North Patagonia. We analysed fourteen populations along a west-east transect of about 500 km in North Patagonia with three types of molecular markers: ITS, chloroplast trnL-F and eight nuclear microsatellites. Bayesian inferences, maximum parsimony and maximum likelihood analyses with trnL-F and ITS showed that F. pallescens is related to the Patagonian clade within the Festuca phylogeny. However, the easternmost populations of F. pallescens at Somuncura plateau were highly differentiated from the other populations and clustered with F. argentina (a sympatric species of the Asian-American clade). Principal coordinates analyses and Bayesian clustering performed with nuclear microsatellites as well as morphoanatomical traits, showed an intermediate position of one of these easternmost populations with respect to the two species, suggesting admixture. The high genetic variability observed in these peripheral populations highlight their relevance for conservation and might be indicating the existence of evolutionary processes triggering events of speciation in the Patagonian fescues. The current low latitudinal range margins of many extra-tropical plant species consist of small and scattered populations that persist locally in microrefugia. It remains poorly understood how their refugial distribution affects mating patterns and reproductive success. Here we examine flower and acorn production and their determinants in refugial populations of the widespread European forest tree pedunculate oak ( Quercus robur)., We monitored male flower, female flower and acorn production in 159 adult trees from 12 oak stands over 2 years. We related these and derived parameters to a series of ecological and genetic predictor variables extrinsic (stand size, density and isolation as well as elevation, topography and forest cover) or intrinsic (size, phenology and several genotypic measures) to the target tree., Tree fertility was unrelated to extrinsic factors but determined by tree size, although we detected size-independent variation in reproductive investment. Female flower number accurately predicted acorn crop size. Fruit set differed between years, evidencing the existence of pollen limitation at the landscape but not at the local scale. Fruit set also tended to increase with the number of mates of the target tree. We detected no other evidence for genetic constraints on mating., Reproduction was triggered by a combination of small-scale and landscape-scale drivers. Although short-distance mating prevailed, limited pollen flow did not appear to significantly constrain reproductive success. The high intrinsic ability of populations to maintain their reproductive capacity may help explain their successful long-term persistence in an adverse broader environment. [ABSTRACT FROM AUTHOR]
