Background and Aims Pyranoanthocyanins are a type of derivative formed by the cycloaddition reaction of different compounds with anthocyanins during wine maturation and ageing, and show a characteristic pyranic ring. This ring provides a high stability to the pigment, and the ring does not fragment in mass spectrometric experiments. Thus, aglycone ions of these derivatives in tandem mass spectrometry show the same fragmentation pattern as that of the corresponding precursor anthocyanin. The aim of this work was the identification of pyranoanthocyanins present in Rioja aged wines using mass spectrometric features to differentiate among isomeric compounds. Methods and Results The application to three Rioja aged red wines of a methodology that combines column chromatography, solid-phase extraction and high-performance liquid chromatography-diode array detection-electrospray ionisation collision-induced dissociation-tandem mass spectrometry has allowed the identification of 72 pyranoanthocyanin derivatives. Conclusions This paper provides an overview of pyranoanthocyanin derivatives present in aged red wines with the identification of 72 compounds, some of them derivatives that appear in low concentration, and it assesses the importance of mass spectrometric analysis in the differentiation among isomeric derivatives. Significance of the Study The methodology described allows the identification of derivatives at low concentration and the establishment of characteristic fingerprints of native anthocyanins, which provide an effective tool for pyranoanthocyanin identification, allowing the differentiation among isomeric derivatives. In the last two decades, the introduction of tandem mass spectrometry in clinical laboratories has enabled simultaneous testing of numerous acylcarnitines and amino acids from dried blood spots for detecting many aminoacidopathies, organic acidurias and fatty acid oxidation disorders. The expanded newborn screening was introduced in Slovenia in September 2018. Seventeen metabolic diseases have been added to the pre-existing screening panel for congenital hypothyroidism and phenylketonuria, and the newborn screening program was substantially reorganized and upgraded.Tandem mass spectrometry was used for the screening of dried blood spot samples. Next-generation sequencing was introduced for confirmatory testing. Existing heterogeneous hospital information systems were connected to the same laboratory information system to allow barcode identification of samples, creating reports, and providing information necessary for interpreting the results.In t he first y ear of t he expanded newborn screening a total of 15,064 samples w ere screened. Four patients were confirmed positive with additional testing.An expanded newborn screening program was successfully implemented with the first patients diagnosed before severe clinical consequences.Uporaba tandemske masne spektrometrije je v zadnjih dvajsetih letih močno vplivala na diagnostiko vrojenih bolezni presnove, saj omogoča sočasno merjenje številnih acilkarnitinov in aminokislin iz posušenega madeža krvi in s tem prepoznavo različnih aminoacidopatij, organskih acidurij in motenj v metabolizmu maščobnih kislin. Program razširjenega presejanja novorojencev je bil v Sloveniji uveden septembra 2018. Obstoječemu programu presejanja kongenitalne hipotiroze in fenilketonurije je bilo dodanih 17 novih bolezni presnove, ob tem so bile vpeljane določene organizacijske spremembe.Analiza vzorca posušenega madeža krvi poteka s tandemsko masno spektrometrijo. Za potrditveno testiranje je bila vpeljana metoda sekvenciranja naslednje generacije. Obstoječi heterogeni bolnišnični informacijski sistemi so bili povezani v enotni laboratorijski informacijski sistem, kar omogoča kodno identifikacijo vzorca, oblikovanje izvidov in dostop do vseh podatkov, potrebnih za interpretacijo izvida.V prvem letu razširjenega presejanja novorojencev je bilo pregledanih 15.064 vzorcev. Z dodatnimi potrditvenimi testi je bila bolezen potrjena pri štirih bolnikih.Razširjeno presejanje novorojencev je bilo uspešno vzpostavljeno. Prvi bolniki so bili prepoznani, diagnoza jim je bila postavljena pred pojavom kliničnih znakov.

5. Analysis of veterinary drug residues in cheese by ultra-high-performance LC coupled to triple quadrupole MS/ MS.

A simple, reliable, and fast multiresidue method has been developed for the determination of 17 veterinary drugs belonging to several families (macrolides, sulfonamides, and anthelmintics) in cheese at trace levels. Ultra-high-performance LC coupled to MS/ MS has been used for the analysis of these compounds in less than 9 min. Veterinary drug residues have been extracted from cheese samples using a Qu ECh ERS (quick, easy, cheap, effective, rugged, and safe)-based extraction procedure without applying any further clean-up step. Matrix-matched calibration was used for quantification and recoveries were calculated at three concentration levels (10, 50, and 100 μg/kg). The obtained values ranged from 70 to 110% for the selected compounds except for tylosin and josamycin at 100 μg/kg (111.7 and 112.7%, respectively). Intra- and interday precision were also evaluated and RSDs were lower than 25% in all the cases. LOQs ranged from 0.3 μg/kg (for thiabendazole, oxfendazole, mebendazole, josamycin, and fenbendazole) to 10.5 μg/kg (abamectin), whereas decision limit and detection capability ranged from 2.3 (thiabendazole) to 11.3 (abamectin) and 4.2 (thiabendazole) to 14.3 μg/kg (abamectin), respectively. Finally, 13 samples were analyzed and traces of thiabendazole were detected in two different cheeses. [ABSTRACT FROM AUTHOR]
