Although microliter-scale liquid handling with a handheld pipette is a routine task, pipetting nanoliter-scale volumes is challenging due to several technical difficulties including surface tension, adhesion and evaporation effects. We developed a fully automated piezoelectric micropipette with a precision of < 1 nanoliter, improving the efficiency of imaging-based single-cell isolation to above 90%. This improvement is crucial when sorting rare or precious cells, especially in medical applications. The compact piezoelectric micropipette can be integrated into various (bio)chemical workflows. It eliminates plastic tubes, valves, syringes, and pressure tanks. For high-quality phase-contrast illumination of the sample, e.g., cells or tiny droplets, we constructed rings of LEDs arranged concentrically to the micropipette. The same device can be readily used for single-cell printing and nanoliter-scale droplet printing of reagents using either fluorescent or transparent illumination on a microscope. We envision that this new technology will shortly become a standard tool for single-cell manipulations in medical diagnostics, e.g., circulating tumor cell isolation. [ABSTRACT FROM AUTHOR]

The rapid and accurate analysis of micro-samples is a crucial foundation for precision medicine, particularly for early screening and monitoring of cancer, where it holds significant importance. Ultrasound-based multifunctional biocompatible manipulation techniques have been extensively applied in a variety of biomedical fields, providing insights for the development of rapid, cost-effective, and accurate biomarker detection strategies. “Children are not tiny adults” is an adage commonly used in pediatrics to emphasize the fact that children often have different physiological responses to sickness and trauma compared to adults. However, despite widespread acceptance of this concept, diagnostic blood testing is an excellent example of clinical care that is not yet customized to the needs of children, especially newborns. Cumulative blood loss resulting from clinical testing does not typically impact critically ill adult patients, but can quickly escalate in children, leading to iatrogenic anemia and related comorbidities. Moreover, the tests prioritized for rapid, near-patient testing in adults are not always the most clinically relevant tests for children or newborns. The mixing rate is a crucial factor in determining the reaction rate and product distribution in reactors for academic and industrial application. Especially, in pharmaceutical or dangerous chemistry, it is essential to create rapidly homogeneous mixture under the control of a small volume of precious sample. In this study, we propose a microloop reactor that is capable of rapid mixing for homogeneous reaction by utilizing programmable actuated microvalves (PAVs), which can generate the rotary flow rapid mixing in the reactor. The microloop reactor is composed of a stacked layered structure, which is prepared by a soft lithography method. The top layer (fluidic layer) has microchannels for supplying each reagent that is assembled with the bottom layer (control layer). The bottom layer has ultrathin polymer membrane, which can be an on-off valve to precisely control the nanoliter-scale volume of reagents in the reactor. To evaluate mixing performance, we use peroxidase reaction that produces fluorescent by-product (resorufin), thereby observing how fast they are mixed together. We quantify the uniformity of fluorescent intensity throughout the reaction loop, indicating that our proposed microloop reactor exhibits a homogeneous reaction. Droplets of aqueous solutions distributed in an immiscible oil phase are increasingly used and investigated as a means to handle and assay small volumes of samples. The primary attraction of this method is that surface interactions are kept to a minimum, and changes in sample concentration, especially due to adsorption to the walls, are avoided. Microfluidic methods to generate, transport, merge, split and perform reactions in droplets were developed recently. These methods depend on the continuous flow of the two phases involved inside closed microfluidic channels. Alternatively, an electrowetting phenomenon was also exploited to control the movement of droplets between two solid substrates. However, there are some situations where small volume sample transport and assaying are required in open systems. Here, we demonstrate a simple electromechanical probe (tweezers) that is capable of manipulating a small aqueous droplet in a bi-layer oil phase. The tweezer consists of two needles positioned close to each other and uses polarization of the aqueous droplet in an applied electrical field to confine the droplet between the needles with minimal solid contact. Mechanical motion of the tweezer can be used to transport the droplet to various positions. Operations such as aliquoting, merging and transport are demonstrated. Finally, this method was used to perform a DNA amplification assay where droplets of the sample and the amplification mixture are aliquoted separately, mixed and amplified using an in-situ heater. This electromechanical tweezer is of interest in low-throughput, small-volume biological and chemical assays where the investigator requires direct and open access to the samples. The resolving power, chemical sensitivity and non-invasive nature of NMR have made it an established technique for in vivo studies of large organisms both for research and clinical applications. NMR would clearly be beneficial for analysis of entities at the microscopic scale of about 1 nL (the nanoliter scale), typical of early development of mammalian embryos, microtissues and organoids: the scale where the building blocks of complex organisms could be observed. However, the handling of such small samples (about 100 µm) and sensitivity issues have prevented a widespread adoption of NMR. In this article we show how these limitations can be overcome to obtain NMR spectra of a mammalian embryo in its early stage. To achieve this we employ ultra-compact micro-chip technologies in combination with 3D-printed micro-structures. Such device is packaged for use as plug & play sensor and it shows sufficient sensitivity to resolve NMR signals from individual bovine pre-implantation embryos. The embryos in this study are obtained through In Vitro Fertilization (IVF) techniques, transported cryopreserved to the NMR laboratory, and measured shortly after thawing. In less than 1 h these spherical samples of just 130–190 µm produce distinct spectral peaks, largely originating from lipids contained inside them. We further observe how the spectra vary from one sample to another despite their optical and morphological similarities, suggesting that the method can further develop into a non-invasive embryo assay for selection prior to embryo transfer. Abstract: Tracer micronjections are very widely used in brain mapping research. While administration of small quantities/volumes of tracers can easily be achieved through iontophoresis, in the case of pressure injections the volume of the substances injected are much more difficult to control. Instead of using different variables like hydrodynamic conductance, pressure pulses, pressure–temperature-dependent protocols to quantify very small volumes in the nanoliter range out of a higher volume, within the confines of the present study a novel microcapillary design is presented. This microcapillary contains exactly the volume of tracer one intends to inject, therefore the danger of flooding large brain areas or the risk of tracer leakage to neighbouring nuclei are completely eliminated, and in the same time this design assures that very small and circumscribed areas can be labeled and their connections mapped, thus making the experiments more specific. In combination with high precision stereotaxic measurements these small volumes of tracers can yield well targeted and very discrete injection sites that make possible the mapping of individual nuclear subdivisions or delicate nuclei in the brain. The use of molecular techniques to inform diagnosis, prognosis, and treatment design will play an important role in the future of medicine. Each new test, however, represents a new cost to the health care system, and significant effort is required to move new techniques to the clinical setting in the most cost-effective and efficient manner. Consequently, there is a compelling need for technological improvements that will facilitate clinical application of novel assays, by reducing cost and complexity in implementation. This is particularly important in cancer pathology. Here, we present a novel applicator technology that enables staining of individual biopsies in a tissue microarray (TMA) to provide low-cost, multiplexed biomarker testing at the level of intact tissue. The applicator is designed to deliver tens of nanoliters of aqueous reagent to arrayed tissue biopsies kept under a layer of oil-based Liquid Coverslip without contacting the biopsies. A pin consisting of concentric stainless steel electrodes separated by a hydrophobic insulator provides a balance between hydrophobicity and hydrophilicity to hold a reagent droplet on the tip of the pin, whereas a small electrical current passed through the droplet spanning the electrodes is used for drop sensing. This design is more amenable to repeatable manufacturing than a previous prototype, which in initial testing demonstrated successful immunohistochemical and in situ hybridization staining of individual biopsies in a TMA, but was difficult to produce. This new design was tested to investigate the factors affecting its operation, in terms of the volume of reagent picked up and its ability to successfully deliver reagent to the biopsies. The short-term and long-term stabilities of fluorescein are first qualified using a reference standard. Next, we determine the noise contribution of the filler and reader. Abstract: To facilitate drug discovery directed toward platelet-specific targets, we developed a platelet isolation and fluorophore-loading method that yields functionally responsive platelets in which we were able to detect agonist-induced calcium flux using a microfluidics-based screening platform. The platelet preparation protocol was designed to minimize preparation-induced platelet activation and to optimize signal strength. Measurement of platelet activation, as monitored by ratiometric determination of agonist-induced calcium flux in fluor-loaded human platelets, was optimized in a macrosample cuvette format in preparation for detection in a microfluidic chip-based assay. For the microfluidic device used in these studies, a cell density of 1 to 2×106 platelets per milliliter and a nominal flow rate of 5 to 10nl per second provided optimal event resolution of 5 to 20 platelets traversing the detection volume per unit time. Platelets responded in a dose-dependent manner to adenosine diphosphate and protease-activating peptide (PAR) 1 thrombin receptor-activating peptide (TRAP). The work presented here constitutes proof-of-principle experiments demonstrating the enabling application of a microfluidic device to conduct high-throughput signaling studies and drug discovery screening against human platelet targets. Abstract: A MEMS (micro-electro-mechanical systems) micropump with circular bossed membrane designed for nanoliter drug delivery is characterized in this article. A quasistatic model under consideration of low operating frequency is used to characterize this micropump. The mathematical model is an ordinary differential equation that describes the behavior of the micropump by including its key components of bossed membrane and inlet/outlet microvalves. Characterizations of bossed membrane and microvalves are carried out separately in the finite element analysis ANSYS package. The stroke volume of the membrane is calculated within the range that the linear deflection theory is valid. Analysis of the microvalve is a challenging task in microfluidics because it is a coupled field (solid-fluid coupling) problem. To solve the structural (solid) or fluid part separately is impractical in characterizing drug-delivery micropumps. Based on sequential weak solid-fluid coupling in ANSYS/FLOTRAN, the flow rates across the inlet and outlet microvalves are analyzed and simulated. Because the quasistatic equation contains several nonlinear terms, closed-form analytical solution for this equation is impossible; thus MATLAB is used to solve it numerically. The transient flow rate of the micropump is obtained by substituting the pressure in microchamber into the flow rate function of outlet microvalves. Integration of the function over 1 driving cycle and multiplication by the driving frequency provides the drug-delivery rate of the micropump. [Copyright &y& Elsevier]

This report describes the development of a digital microfluidic testing platform and associated clinical assays purposely curated to address current shortcomings in pediatric laboratory testing by using microliter volumes (<50 µL) of samples. The automated platform consists of a small instrument and single-use cartridges, which contain all reagents necessary to prepare the sample and perform the assay. Electrowetting technology is used to precisely manipulate nanoliter-sized droplets of samples and reagents inside the cartridge. To date, we have automated three disparate types of assays (biochemical assays, immunoassays, and molecular assays) on the platform and have developed over two dozen unique tests, each with important clinical application to newborns and pediatric patients. Cell lysis, plasma preparation, magnetic bead washing, thermocycling, incubation, and many other essential functions were all performed on the cartridge without any user intervention. The resulting assays demonstrate performance comparable to standard clinical laboratory assays and are economical due to the reduced hands-on effort required for each assay and lower overall reagent consumption. Genetic analysis of single cells is emerging as a powerful approach for studies of heterogeneous cell populations. Indeed, the notion of homogeneous cell populations is receding as approaches to resolve genetic and phenotypic variation between single cells are applied throughout the life sciences. A key step in single-cell genomic analysis today is the physical isolation of individual cells from heterogeneous populations, particularly microbial populations, which often exhibit high diversity. Here, we detail the construction and use of instrumentation for optical trapping inside microfluidic devices to select individual cells for analysis by methods including nucleic acid sequencing. This approach has unique advantages for analyses of rare community members, cells with irregular morphologies, small quantity samples, and studies that employ advanced optical microscopy., (© 2013 Elsevier Inc.
