Aim: The present investigation aimed to develop niosomes containing gold nanoparticles (Nio-AuNPs) and to evaluate the combinational effect of Nio-AuNPs and x-ray radiation therapy (XRT) on growth inhibition potential and induction of apoptosis in the A549 cell line. Materials & methods: Gold nanoparticles (AuNPs) were synthesized, and niosomes were prepared using the thin-film hydration method. Various techniques were employed to determine their physiochemical characteristics. MTT assay, cell apoptosis analysis and combination index analysis were conducted to evaluate the therapeutic feasibility of Nio-AuNPs combined with XRT. Results: The combination of Nio-AuNPs and XRT resulted in greater cytotoxicity compared with XRT alone or with AuNPs. AbstractRutin and berberine are naturally occurring polyphenols but their usefulness is restricted by their low bioavailability and poor solubility. Thus, by delivering rutin–berberine in the form of niosomes simultaneously and hoped to enhance the permeability through nasal route for the treatment of depression. Thin-film hydration approach was used to generate dual drug-loaded niosomes (rutin–berberine) encapsulating rutin and berberine. Various analytical techniques were used to characterize the morphology, size, compatibility, and leakage of the particles. These techniques included transmission electron microscopy, dynamic light scattering, UV spectrophotometry, and differential scanning calorimetry. The optimized formulation (rutin–berberine optimized niosomal formulation) was subjected to additional characterization for drug release, ex-vivo penetration investigation, confocal laser scanning microscopy, and 2,2-diphenyl-1-picrylhydrazyl assay. The rutin–berberine optimized niosomal formulation analysis showed that the drug release was 76.66 ± 1.24% at 24 hours, the entrapment efficiency was 74.8 ± 2.13% (rutin), and 79.0 ± 3.47% (berberine) and the vesicle size was 72.6 nm with a polydispersion index of 0.208. The antioxidant activity showed 73.67 ± 2.5% which is close to regular ascorbic acid. Studies on permeation ex-vivo revealed that rutin–berberine optimized niosomal formulation had considerably higher permeation (84.89 ± 3.15%) compared to rutin–berberine suspension (37.28 ± 2.33%). Furthermore, rhodamine B-loaded rutin–berberine optimized niosomal formulation appeared to have higher penetration than the control (rhodamine B-solution) according to the confocal laser scanning microscope results of the nasal mucosa. The high recurrence rate of hepatocellular carcinoma (HCC) and poor prognosis after medical treatment reflects the necessity to improve the current chemotherapy protocols, particularly drug delivery methods. Development of targeted and efficient drug delivery systems (DDSs), in all active, passive and stimuli-responsive forms for selective delivery of therapeutic drugs to the tumour site has been extended to improve efficacy and reduce the severe side effects. Recent advances in nanotechnology offer promising breakthroughs in the diagnosis, treatment and monitoring of cancer cells. In this review, the specific design of DDSs based on the different nano-particles and their surface engineering is discussed. This study aimed to develop and evaluate a niosomal formulation loaded with Plumbago zeylanica extract (PZE). The solvent evaporation method was used to prepare niosomes and optimized using the Box–Behnken design. The optimized formulation (PZE-Ns-Opt) was characterized for drug release, DPPH assay, α-amylase inhibition assay, α-glucosidase inhibition assay, and confocal laser scanning microscopy (CLSM) study. PZE-Ns-Opt showed a vesicle size of 253.6 nm, PDI of 0.108, entrapment efficiency of 62.4%, and drug release of 84.01%. The CLSM image of the rat's intestine suggested that the rhodamine red B-loaded PZE-Ns-Opts showed superior penetration compared to the control. Further, the antioxidant activity of the prepared formulation was exhibited as 89.46%±0.016 as compared to PZE (78.10%±0.005). In addition to this, α- amylase activity was inhibited by 95.11%±4.62, 85.88%±2.56, and 89.87%±3.65 by acarbose, PZE, and PZE-Ns-Opt, respectively. In comparison, the α-glucosidase activity was inhibited by 88.47%±1.04, 81.07%±0.50 and 85.78%±0.71 by acarbose, PZE, and PZE-Ns-Opt, respectively. To conclude, the development of PZE-Ns-Opt formulation and its characterization showed the establishment's authenticity. The in vitro antidiabetic, antioxidant studies expressed the protective effect from oxidative stress, CLSM of rat's intestine ability to penetrate to great extent and could be a promising candidate for the management of diabetes mellitus. In the past few decades, considerable attention has been focused on the development of novel drug delivery system (NDDS). The NDDS should ideally fulfil two prerequisites. Firstly, it should deliver the drug at a rate directed by the needs of the body, over the period of treatment. Secondly, it should channel the active entity to the site of action. Conventional dosage forms including prolonged release dosage forms are unable to meet none of these. At present, no available drug delivery system behaves ideally, but sincere attempts have been made to achieve them through various novel approaches in drug delivery. New hexadecylpiperidinium surfactants, containing one or two butylcarbamate fragments, were synthesized. The antimicrobial activity, toxicity, aggregation behavior in aqueous solutions, and solubilization capacity of these surfactants towards the hydrophobic drug ibuprofen were characterized. These surfactants demonstrated a high antimicrobial activity against a wide range of pathogenic bacteria, including both Gram-positive and Gram-negative strains, as well as fungi. By forming mixed-micellar compositions of the cationic surfactant 1-CB(Bu)-P-16 and the nonionic surfactant Brij®35, highly functional and low-toxic formulations were obtained. Furthermore, the transition from mixed micelles to niosomes was accomplished, enhancing their potential as drug delivery systems. Niosomes were found to be less toxic compared to mixed micelles, while also increasing the solubility of ibuprofen in water. The modification of niosomes with cationic surfactants made it possible to increase the stability of the system and improve the solubility of the drug. The data obtained indicate that these new carbamate-containing hexadecylpiperidinium surfactants have significant potential in biomedical applications, particularly in the formulation of advanced drug delivery systems. Triacontanol is a long-chain primary alcohol derived from policosanol, known for its diverse biological activities, including functioning as a plant growth regulator and exhibiting anti-inflammatory and antitumoral effects. However, its application is limited due to its high hydrophobicity, resulting in poor absorption and reduced therapeutic effectiveness. A potential solution to this problem is the use of niosomes. Niosomes are carriers composed of non-ionic surfactants, cholesterol, charge-inducing agents, and a hydration medium. They are effective in encapsulating drugs, improving their solubility and bioavailability. The objective of this study was to optimize and synthesize nano-niosomes for the encapsulation of triacontanol. Niosomes were synthesized using a thin-film hydration method combined with ultrasonication, following a Box–Behnken design. Niosomes were characterized using various techniques including dynamic light scattering, Fourier-transform infrared spectroscopy (FTIR), confocal microscopy, high-resolution scanning electron microscopy, and transmission electron microscopy (TEM). Formulation 14 of niosomes achieved the desired size, polydispersity index (0.198 ± 0.008), and zeta potential (−31.28 ± 1.21). FTIR analysis revealed a characteristic signal in the 3400–300 cm−1 range, indicating intermolecular interactions due to a bifurcated hydrogen bond between cholesterol and S60. Tenofovir is an antiviral drug indicated for the treatment of infections caused by the human immunodeficiency virus (HIV) and hepatitis B virus (HBV). TNF is classified as a BCS Class III drug, indicating its ability to readily dissolve in gastrointestinal fluids with poor permeability across intestinal membranes results in a lower absorption rate, ultimately restricting its bioavailability. The primary challenges associated to enhancing the bioavailability of TNF involve intestinal degradation and efflux transport facilitated by Multidrug resistance protein1. TNF-loaded proniosomes were formulated using 32 factorial design by applying slurry method of preparation with a molar ratio of 2.5:1:1.5 for cholesterol, span 60, and maltodextrin, respectively. Maltodextrin-based polymeric nanoparticles exhibited desirable nano-scale properties, including size, polydispersity index, and zeta potentials, which fell within acceptable ranges. The successful PNFs (T-PN3, T-PN4, and T-PN7) demonstrated TNF entrapment in the range of 92.96 to 96.28%. The hydration volume and hydration time of proniosome-based niosomes for delivering TNF were optimized and the results demonstrated the structural homogeneity of niosomes derived from proniosomes. Transmission electron microscopy (TEM) analysis indicated that the niosomes exhibit a uniform and smooth surface morphology. Successful formulations were further characterized for their powder behaviour by angle of repose, TNF interactions with formulation components by FT-IR and DSC thermal analysis. The T-PN3 and T-PN7 formulations demonstrated a high in-vitro release rate of approximately 99% in PBS. Additionally, the cellular uptake of TNF from successful PNFs in NCI-N87 cells ranged from 81% to 83%, indicating that T-PN3 and T-PN7 exhibited superior performance compared to free TNF and the commercially available tenofovir. Further mechanistic analysis was conducted using MDR1 efflux studies and western blot techniques. There are around 24 distinct lipid vesicles described in the literature that are similar to vesicular systems such as liposomes. Liposome-like structures are formed by combining certain amphiphilic lipids with a suitable stabiliser. Since their discovery and classification, self-assembled liposome-like structures as active drug delivery vehicles captured researchers' curiosity. This comprehensive study included an in-depth literature search using electronic databases such as PubMed, ScienceDirect and Google Scholar, focusing on studies on liposome and liposomes like structure, discussed in literature till 2024, their sizes, benefits, drawback, method of preparation, characterisation and pharmaceutical applications. Pharmacosomes, cubosomes, ethosomes, transethosomes, and genosomes, all liposome-like structures, have the most potential due to their smaller size with high loading capacity, ease of absorption, and ability to treat inflammatory illnesses. Genosomes are futuristic because of its affinity for DNA/gene transport, which is an area of focus in today's treatments. This review will critically analyse the composition, preparation procedures, drug encapsulating technologies, drug loading, release mechanism, and related applications of all liposome-like structures, highlighting their potential benefits with enhanced efficacy over each other and over traditional carriers by paving the way for exploring novel drug delivery systems in the Pharma industry. Cilostazol, an anti-platelet aggregation medicine, is also known to have vasodilation properties and is commonly used for treating muscle soreness and cramps by increasing the muscle oxygen supply. The medication has limited oral bioavailability, is prone to pre-systemic metabolism, and is poorly soluble in aqueous media. A transdermal administration was planned to increase the drug’s solubility and therapeutic efficacy. The current work intended to develop cilostazol niosome-loaded transdermal gel, which was prepared, and with the use of Fourier transform infrared and differential scanning calorimetry analyses, drug-excipient interactions were observed. The medication was formulated utilizing Carbopol-934, Pluronic-F-127, and HPMC gel bases for the transdermally delivered niosomal gels. The produced niosomes had a maximum percentage of drug entrapment at 96.4%, with a particle size of 102 ± 11.30 nm and polydispersity index of 0.29 ± 0.069. The highest percentage of the medication that was entrapped was 96.4%, and the Carbopol-934 gel basis released the major part of the drug under in vitro conditions. A maximum transdermal flux was recorded at 3850.92 μg after 4 h, indicating a 10% increase in cilostazol permeation through rat skin. The flux rate for the niosomal preparation containing the drug ranged from 14.85 to 28.02 μg/cm2 h−1. In comparison to the pure cilostazol-loaded gels, the pharmacokinetics investigation showed that the niosomal gel formulations had considerably greater C max, T max, and AUC0. The niosomes loaded with cilostazol exhibited greater solubility, higher bioavailability, and improved effectiveness. Better therapeutic results may be achieved with systemic and site-directed delivery of cilostazol using the designed transdermal niosomal gel with appropriate molecular tagging modification/(s).

Background: Posaconazole, currently available as a solid oral dosage form, possesses erratic pharmacokinetics that complicate dosing regimens and increase the risk of adverse effects and drug interactions. Hence, innovative strategies, especially topical ones, are necessary to enhance the therapeutic profile and improve patient outcomes. Methodology: The regular 23 factorial design and the concentrations of Oleic acid: Span 80 (2:1) ratio, Cholesterol, and Tween 80 as independent variables were used for the formulation development. The preliminary effect was determined by dependent variables like vesicle size and entrapment efficiency; then the final optimized batch was loaded in 3 % w/w Carbopol gel. Result and Discussion: The optimized batch's vesicle size and entrapment efficiency were 193.34+14.84 nm and 90.03+0.11 %, respectively. Simple Summary: Cancer is the second leading cause of death worldwide, warranting the development of safer, more effective therapies. Liposomes and niosomes can be considered excellent drug delivery systems due to their ability to load all compounds and reduce the toxicity of the loaded drug without reducing its effectiveness. Synechocystis sp. is a unicellular, freshwater blue-green algae strain that has many bioactive compounds that qualify for use in industrial, pharmaceutical, and other fields. This study used nano-liposomes and nano-niosomes to deliver Synechocystis sp. extract against human colon, ovarian, and breast cancer cell lines. The results demonstrated potential activities against human colon, ovarian, and breast cancer cell lines. A total of 22 compounds were identified through the metabolic profiling of an extract, as it is a tool aimed at identifying biologically active compounds in the sample. Subsequently, the molecular docking of these compounds was studied, which is the most effective way to study the compound's effectiveness on a specific disease by determining the binding affinity. The results showed that compounds 21, 6, 7, 8, 12, and 19 have a high degree of correlation. Finally, these results represent a promising step toward developing effective cancer treatments. Future research should translate this approach into successful anticancer drugs through participation in clinical trials. Synechocystis sp. is a unicellular, freshwater cyanobacteria strain that contains many bioactive compounds that qualify its use in industrial, pharmaceutical, and many other fields. This study investigated the potential of nano-liposomes (L) and nano-niosomes (N) for delivering Synechocystis sp. extract against cancer cell lines. Four different types of nanoparticles were prepared using a dry powder formulation and ethanol extract of Synechocystis sp. in both nanovesicles (N1 and N2, respectively) and liposomes (L1 and L2, respectively). Analysis of the formed vesicles using zeta analysis, SEM morphological analysis, and visual examination confirmed their stability and efficiency. L1 and L2 in this investigation had effective diameters of 419 and 847 nm, respectively, with PDI values of 0.24 and 0.27. Furthermore, the zeta potentials were found to range from −31.6 mV to −43.7 mV. Regarding N1 and N2, their effective diameters were 541 nm and 1051 nm, respectively, with PDI values of 0.31 and 0.35, and zeta potentials reported from −31.6 mV to −22.2 mV, respectively. Metabolic profiling tentatively identified 22 metabolites (1–22) from the ethanolic extract. Its effect against representative human cancers was studied in vitro, specifically against colon (Caco2), ovarian (OVCAR4), and breast (MCF7) cancer cell lines. The results showed the potential activities of the prepared N1, N2, L1, and L2 against the three cell lines, where L1 had cytotoxicity IC50 values of 19.56, 33.52, and 9.24 µg/mL compared to 26.27, 56.23, and 19.61 µg/mL for L2 against Caco2, OVCAR4, and MCF7, respectively. On the other hand, N1 exhibited IC50 values of 9.09, 11.42, and 2.38 µg/mL, while N2 showed values of 15.57, 18.17, and 35.31 µg/mL against Caco2, OVCAR4, and MCF7, respectively. Meanwhile, the formulations showed little effect on normal cell lines (FHC, OCE1, and MCF10a). All of the compounds were evaluated in silico against the epidermal growth factor receptor tyrosine kinase (EGFR). The molecular docking results showed that compound 21 (1-hexadecanoyl-2-(9Z-hexadecenoyl)-3-(6′-sulfo-alpha-D-quinovosyl)-sn-glycerol), followed by compounds 6 (Sulfoquinovosyl monoacylgycerol), 7 (3-Hydroxymyristic acid), 8 (Glycolipid PF2), 12 (Palmitoleic acid), and 19 (Glyceryl monostearate), showed the highest binding affinities. These compounds formed good hydrogen bond interactions with the key amino acid Lys721 as the co-crystallized ligand. These results suggest that nano-liposomes and nano-niosomes loaded with Synechocystis sp. extract hold promise for future cancer treatment development. This study investigates the potential synergy effect of Ganoderma lucidum and Lentinus edodes mushroom extracts delivered through niosomes (composed of Span 40, Tween 40, and cholesterol) for anti‐cancer and antimicrobial applications. GC–MS analysis revealed bioactive compounds such as d‐limonene, ascorbic acid, and ergosterol in the extracts, known for their anti‐cancer and antimicrobial effects. The niosomal formulations exhibited improved physical properties, with sizes ranging from 96 to 160 nm and polydispersity index (PDI) values in the range of 0.29–0.54, suggesting that the niosomes had an acceptable size distribution and homogeneity. In cell culture studies, the niosomal formulations demonstrated significant cytotoxic effects on A549 lung cancer cells, with the combination of both extracts leading to a notable decrease in cell viability to 14.86%. Additionally, the combination of G. lucidum and L. edodes extracts in niosomal form showed varying and potent antimicrobial properties against pathogenic bacteria (Pseudomonas aeruginosa and Staphylococcus aureus) and fungi (Candida albicans) with lower minimum inhibitory concentration (MIC) values compared to their free form, indicating a potential synergistic effect. Objective: Zinc Oxide nanoparticles (ZnO NPs) are used widely in nowadays personal care products, especially sunscreens, as a protector against UV irradiation. Yet, they have some reports of potential toxicity. Silica is widely used to cage ZnO NPs to reduce their potential toxicity. Vitamin C derivative, Magnesium Ascorpyl Phosphate (MAP), is a potent antioxidant that can efficiently protect human skin from harmful impacts of UV irradiation and oxidative stress. The combination of silica coated ZnO NPs and MAP nanovesicles could have potential synergistic protective effect against skin photodamage. Methods: Silica coated ZnO NPs and MAP nanovesicles (ethosomes and niosomes) were synthesized, formulated, and evaluated as topical gels. These gel formulations were evaluated in mice for their photoprotective effect against UV irradiation through histopathology and immuno-histochemistry study. Split-face clinical study was conducted to compare the effect of application of silica coated ZnO NPs either alone or combined with MAP nanovesicles. Their photoprotective action was evaluated, using Antera 3D® camera, for melanin level, roughness index and wrinkles depth. Results: Silica coated ZnO NPs when combined with MAP nanovesicles protected mice skin from UV irradiation and decreased the expression of the proinflammatory cytokines, NF-κB. Clinically, silica coated ZnO NPs, alone or combined with MAP nanovesicles, could have significant effect to decrease melanin level, roughness index and wrinkles depth with higher effect for the combination. In the current study, a smart release system responsive to temperature was developed to improve the efficiency of tetracycline (TC) in antibacterial therapy. The nanovesicles designed consist of a non-ionic surfactant, SPAN60, cholesterol and a phase change material (PCM) as a thermoresponsive gating material. Niosomes were prepared using an increasing amount of PCM and characterized in terms of size, zeta potential, colloidal stability and thermoresponsive properties. The vesicles that developed were homogenous in size, had good biocompatibility and stability for up to 3 months and demonstrated thermoresponsive behavior. A low drug leakage was observed at 37 °C, while a rapid release occurred at 42 °C, due to the faster diffusion rate of the drug trough the melted PCM. This controllable drug release capacity allows us to avoid premature drug release, minimizing unwanted and toxic effects and ensuring a long retention time in the nanodevice so that it reaches the infected sites. In addition, TC-loaded niosomes were screened to investigate their antibacterial activity against various Gram-positive and Gram-negative bacteria by minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) assays. An interesting temperature-dependent antibacterial activity was observed against some bacterial strains: the niosomes activity against S. epidermis, for example, was improved by the temperature increase, as suggested by a reduction in MIC values from 112.81 to 14.10 μM observed at 37 and 42 °C, respectively. Taken together, the thermoresponsive platform developed allows us to use lower antibiotic amounts while ensuring therapeutic efficacy and, so, will advance the development of a novel antibacterial agent in clinical practice. The current research focuses on developing and characterizing Itraconazole-loaded niosomes in vitro to improve drug delivery and effectiveness. Niosomes, which are vesicles made of non-ionic surfactants, offer significant benefits by enhancing the absorption of drugs that have low solubility in water. Itraconazole, an antifungal medication with limited water solubility was encapsulated in niosomes using the thin-film hydration technique. The niosomes underwent analysis for particle size, zeta potential, encapsulation efficiency, in vitro drug release and stability. Findings showed that the niosomes had a uniform size distribution, high encapsulation efficiency and released the drug steadily over time. Stability tests demonstrated that the niosomes remained stable under different storage conditions. This study suggests that Itraconazole-loaded niosomes could potentially improve the drug's absorption and effectiveness, setting the stage for future in vivo and clinical investigations. Developing effective therapy for cancer using a natural product like curcumin (CR) is an active area of research. The efficacy of CR against different types of cancers is significantly impeded by its non-specificity, low aqueous solubility, and high metabolism. In this study, novel amphiphilic peptide (AMP) was synthesized and their potential to carry CR into cancer cells via niosomal vesicles was examined. The synthesized AMP was characterized using several techniques such as FAB-MS, HRFAB-MS, 1H-, and 13C-NMR spectroscopy, which was then used to produce niosomal vesicles. CR-loaded AMP niosomal vesicles (CR-AMP-Vesicles) were also characterized in terms of size, zeta potential, PDI, and surface morphology. CR-AMP-Vesicles exhibited a smaller size of about 387.70 nm with 77.32% CR encapsulation and a regulated drug release profile. The AMP and AMP-Vesicles are reported for the first time. Moreover, we are the first to develop AMP-CR-Vesicles and to report the activity of the developed AMP-CR-Vesicles against chronic myeloid leukemia cell lines. The anticancer activity of the formulation was evaluated against chronic myeloid leukemia cell lines which exhibited a promising response with the IC50 of 25 µM. Furthermore, the cytotoxicity of produced vesicles on human normal fibroblast 3T3 cells was studied and found to be non-cytotoxic. The overall results indicate that the developed novel CR-AMP-Vesicles are very effective and promising. The novel formulation has the potential for broader applications along with the fact that the developed niosomes have the potential to substantially reduce the cytotoxicity of bioactive compounds against normal cells while maintaining biological activity. The skin cancer (mostly of melanoma cells) rate has increased in recent years due to genetic and epigenetic reasons. The efficient treatment of skin cancer is an unmet requirement to achieve specific anticancer activity, reduction of the costs and side effects, and precluding resistance of cancer cells. Hence, employment of proper nanocarriers is promising. Niosomal delivery of loaded drugs or compounds is preffered for skin cancer therapy considering targeted delivery, scalability, biocompatibility, stability and high loading capacity, efficiently killing skin tumor cells. The niosomal formulation of natural and synthetic agents has increased the anticancer and antimicrobial effects on the skin surface. Moreover, niosomal formulation of drugs/compounds have significantly decreased the effective dose or concentration and also mitigated side effects. The stable structure, minimized drug leakage, and efficient delivery to the target tissues can be achieved using niosomal formulation. Future studies will contribute to a deeper understanding and development of more stable nanocarriers, such as combining hybrid niosomes with stabilizing substances such as salts and organic structures for effective drug delivery. Background and Objectives: Methicillin-resistant Staphylococcus aureus (MRSA) is a major cause of nosocomial and community acquired infections. Nanoparticles are considered as proper tools to overcome the therapeutic problem of antimicrobial-resistant infections because of the drug concentration increment at the desired location and protection from enzymatic degradation. The goal of this study was to evaluate the effect of the antibacterial and antibiofilm activities of zingerone and niosome containing zingerone against pre-formed biofilm of MRSA isolates. Materials and Methods: 62 MRSA isolates cultured from patients with diabetic ulcers were investigated. Niosomes were synthesized and characterized by X-ray diffraction, zeta potential and scanning electron microscopy (SEM). The size of niosomal particles measured by SEM and zetasizer. Results: The surface charge of prepared niosomes was about -37 mV. The effect of the zingerone and noisome containing zingerone was evaluated against biofilms of MRSA isolates. Also, the antibiofilm activity of prepared niosomes on gene expression of MRSA biofilms was evaluated using Real Time PCR. Our results demonstrated that the niosome containing zingerone had a diameter of 196.1 nm and a -37.3-mV zeta potential. Zingerone removed one and three-day old biofilms of MRSA at the concentration of 1000 µg/ml, while the zingerone-laoded niosomes removed 1, 3-and 5-days old biofilms at the concentration of 250 µg/ml, 250 µg/ml, and 500 µg/ml. Conclusion: The results indicated that niosome containing zingerone eliminated MRSA and its biofilms faster compared with free zingerone and it suggested that zingerone-encapsulated niosomes could be considered as a promising treatment against MRSA and its biofilms. Scientists are seeking to find an effective treatment for tumors that has no side effects. N-Acetyl-l-cysteine (NAC) is a thiol compound extracted from garlic. Current study explores the potential of NAC-loaded niosomes (NAC-NIO) for tumor treatment in mice. NAC-loaded niosomes' efficiency, morphology, UV absorption, size distribution, zeta potential, release, and FTIR analysis were evaluated. For vivo study, 25 male BALB/c mice were divided to five groups: gp1 negative control (receive saline), gp2 positive control (tumor group), gp3 treated with NAC, gp4 treated with NAC-NIO at the same time of tumor injection, and gp5 treated with NAC-NIO after tumor growth (day 14). The impact of NAC-NIO on the tumor treatment was evaluated by measuring tumor size progress, comet assay, oxidative stress parameters (GSH, nitric oxide, MDA), western blot analysis, and histopathological investigation of tissues. NAC-NIO showed 72 ± 3% encapsulation efficiency and zeta potential − 5.95 mV with spherical shape. It was found that oral administration of NAC-NIO in a dose of 50 mg/kg provided significant protection against tumor cells. Our formulation decreases DNA injury significantly (P < 0.05). It was noticed that NAC-NIO can increase oxidative stress levels in tumor tissue. On the other hand, the caspase 3 and caspase 9 gene expression were upregulated significantly (P < 0.001) in mice administrated NAC-NIO compared with all other groups. Histological studies confirmed the protective effect of NAC-NIO against tumor especially for treatment during tumor growth protocol. This article reviews contemporary niosome preparation methods, specifically focusing on the widespread thin film hydration technique and the newly reported and novel ball milling approach. The study compares these methodologies regarding particle size, entrapment efficiency, and drug release characteristics exhibited by the resulting niosomes. In order to collect, evaluate, and summarize data, Google Scholar was the primary resource of choice. The selection criteria were based on the most relevant, highly cited, and up-to-date research available on the subject. Keywords associated with the methods of niosomes preparation formed the basis of the search criterion. Recently published and highly cited papers were considered for inclusion in the final list. The findings of this study indicate that the technique of manufacture of niosomes has a significant impact on niosome production, particularly when it comes to particle size, encapsulation efficiency, and drug release. This review summarized the new and novel niosome preparation methods such as solvent casting and ball milling. Solvent casting improved niosome synthesis encapsulation efficiency. The new and unique ball milling method has improved niosome size distribution and encapsulation efficiency while providing good control over particle size and shape. Schizophrenia and bipolar disorder stand as intense and persistent mental illnesses. This research underscores the development of a niosomal thermoreversible gel for intranasal drug delivery with a focus on precise administration to the olfactory lobe. Utilizing a 32 factorial design, lurasidone hydrochloride niosomes were fabricated through the thin film hydration method. The niosomes underwent assessment to determine their encapsulation efficiency, particle size, zeta potential and polydispersity index whereas thermoreversible niosomal in situ gel based on PF-127 in conjunction with HPMC K4M was characterized for pH, gelation time, temperature responsiveness, in vitro release and rheological characteristics. The results indicated that the optimized batch (F4) illustrated a particle size of 171.4 ± 5.12 nm and an encapsulation efficiency 94.67 ± 0.73%. Optimized niosomal gel (poloxamer 17%) characterized with gelation at 37 °C, pseudoplastic flow and virtuous structural integrity. Both in vitro and ex vivo drug release exhibited sustained release through in situ gel. Abstract Background Periodontal diseases may benefit more from topical treatments with nanoparticles rather than systemic treatments due to advantages such as higher stability and controlled release profile. This study investigated the preparation and characterization of thermosensitive gel formulations containing clindamycin-loaded niosomes and solid lipid nanoparticles (SLNs) loaded with fluconazole (FLZ), as well as their in vitro antibacterial and antifungal effects in the treatment of common microorganisms that cause periodontal diseases. Methods This study loaded niosomes and SLNs with clindamycin and FLZ, respectively, and assessed their loading efficiency, particle size, and zeta potential. The particles were characterized using a variety of methods such as differential scanning calorimetry (DSC), dynamic light scattering (DLS), and Transmission Electron Microscopy (TEM). Thermosensitive gels were formulated by combining these particles and their viscosity, gelation temperature, in-vitro release profile, as well as antibacterial and antifungal effects were evaluated. Results Both types of these nanoparticles were found to be spherical (TEM) with a mean particle size of 243.03 nm in niosomes and 171.97 nm in SLNs (DLS), and respective zeta potentials of -23.3 and -15. The loading rate was 98% in niosomes and 51% in SLNs. The release profiles of niosomal formulations were slower than those of the SLNs. Both formulations allowed the release of the drug by first-order kinetic. Additionally, the gel formulation presented a slower release of both drugs compared to niosomes and SLNs suspensions. Conclusion Thermosensitive gels containing clindamycin-loaded niosomes and/or FLZ-SLNs were found to effectively fight the periodontitis-causing bacteria and fungi.

Abstract Cutaneous leishmaniasis is the most prevalent form of leishmaniasis worldwide. Although various anti-leishmanial regimens have been considered, due to the lack of efficacy or occurrence of adverse reactions, design and development of novel topical delivery systems would be essential. This study aimed to prepare artemether (ART)-loaded niosomes and evaluate their anti-leishmanial effects against Leishmania major. ART-loaded niosomes were prepared through the thin-film hydration technique and characterized in terms of particle size, zeta potential, morphology, differential scanning calorimetry, drug loading, and drug release. Furthermore, anti-leishmanial effect of the preparation was assessed in vitro and in vivo. The prepared ART-loaded niosomes were spherical with an average diameter of about 100 and 300 nm with high encapsulation efficiencies of > 99%. The results of in vitro cytotoxicity revealed that ART-loaded niosomes had significantly higher anti-leishmanial activity, lower general toxicity, and higher selectivity index (SI). Half-maximal inhibitory concentration (IC50) values of ART, ART-loaded niosomes, and liposomal amphotericin B were 39.09, 15.12, and 20 µg/mL, respectively. Also, according to the in vivo study results, ART-loaded niosomes with an average size of 300 nm showed the highest anti-leishmanial effects in animal studies. ART-loaded niosomes would be promising topical drug delivery system for the management of cutaneous leishmaniasis.

Abstract One of the most effective strategies to fight viruses and handle health diseases is vaccination. Recent studies and current applications are moving on antigen, DNA and RNA-based vaccines to overcome the limitations related to the conventional vaccination strategies, such as low safety, necessity of multiple injection, and side effects. However, due to the instability of pristine antigen, RNA and DNA molecules, the use of nanocarriers is required. Among the different nanocarriers proposed for vaccinal applications, three types of nanovesicles were selected and analysed in this review: liposomes, transfersomes and niosomes. PubMed, Scopus and Google Scholar databases were used for searching recent papers on the most frequently used conventional and innovative methods of production of these nanovesicles. Weaknesses and limitations of conventional methods (i.e., multiple post-processing, solvent residue, batch-mode processes) can be overcome using innovative methods, in particular, the ones assisted by supercritical carbon dioxide. SuperSomes process emerged as a promising production technique of solvent-free nanovesicles, since it can be easily scaled-up, works in continuous-mode, and does not require further post-processing steps to obtain the desired products. As a result of the literature analysis, supercritical carbon dioxide assisted methods attracted a lot of interest for nanovesicles production in the vaccinal field. However, despite their numerous advantages, supercritical processes require further studies for the production of liposomes, transfersomes and niosomes with the aim of reaching well-defined technologies suitable for industrial applications and mass production of vaccines.

Novel drug delivery system opens the doors towards nano/micro formulation strategies to overcome the challenges associated with the poorly soluble and permeable drugs. Lipid based nanoparticles are widely accepted that includes liposomes, niosomes and micelles which are FDA approved. Such lipid based drug delivery allows delivery for natural phytoconstituents, biopharmaceutical classification system (BCS) class II and class IV drugs are effectively delivered to improve its solubility, permeability and bioavailability. The article provides the recent advances and application of lipid based dosage form for improvement of therapeutic efficacy.

Abstract Background Niosomes hold promise as drug delivery systems for cancer treatment, with niosome size impacting stability, biodistribution, and effectiveness. This study optimized methotrexate (MTX)-loaded niosome formulation by studying the effects of components and processing conditions on size. The niosomes formulation was made by the thin-film hydration technique. Results The optimized formulation (NIO 17) with a 6:2:2 ratio of span 60, soya PC, and cholesterol achieved 55.05% methotrexate encapsulation, particle size 597.2 nm, PDI 0.49, and zeta potential − 23.3 mV. The compatibility of methotrexate with lipids was confirmed via Fourier transform infrared spectroscopy, and transmission electron microscopy revealed spherical, well-dispersed vesicles. Differential scanning calorimetry indicated methotrexate conversion or entrapment within vesicles. In vitro release exhibited a sustained pattern with an initial burst. NIO 17 showed potent anti-cancer activity against B16-F10 cells (GI50: 38.7176 μg/mL). Ex vivo studies suggest tailoring niosome size (597.2–982.3 nm) to target specific skin depths (0–38 μm) for enhanced localized drug delivery. Conclusions This study demonstrates the potential of methotrexate-loaded niosomes as a novel cancer therapy approach, highlighting the potent anti-cancer activity and transdermal delivery potential of NIO 17. Niosomes are arousing significant interest thanks to their low cost, high biocompatibility, and negligible toxicity. In this work, a supercritical CO2-assisted process was performed at 100 bar and 40 °C to produce niosomes at different Span 80/Tween 80 weight ratios. The formulation of cholesterol and 80:20 Span 80/Tween 80 was selected to encapsulate vancomycin, used as a model active compound, to perform a drug release rate comparison between PEGylated and non-PEGylated niosomes. In both cases, nanometric vesicles were obtained, i.e., 214 ± 59 nm and 254 ± 73 nm for non-PEGylated and PEGylated niosomes, respectively, that were characterized by a high drug encapsulation efficiency (95% for non-PEGylated and 98% for PEGylated niosomes). However, only PEGylated niosomes were able to prolong the vancomycin release time up to 20-fold with respect to untreated drug powder, resulting in a powerful strategy to control the drug release rate. This paper presents, for the first time, evidence for vesicle destruction and payload loss at the stage of purification of niosome dispersions by centrifugation, an important operation in the assembly of vesicular materials. The ability of niosomes of different compositions to reassemble, i.e., to restore the vesicular structure after destruction in the field of centrifugal forces, was demonstrated by dynamic light scattering and fluorescence spectroscopy. The kinetics of reassembly of vesicular structures is determined by the strength of the centrifugal field and the composition of niosomes. In contrast to ternary compositions, where particle size and modality are essentially unchanged after redispersion of the precipitate resulting from centrifugation, niosome dispersions containing anionic dicetyl phosphate includes micron-sized particles after redispersion, which vary in size over a wide range throughout the observation period. The reassembly process is complicated by the presence of charge on the surface of the niosomes. Elastic niosomes - ethosomes have been synthesised which, due to the high deformability of the shells, are less susceptible to destruction in the centrifugal field and retain the contents of the aqueous core. Using the "energy landscape" approximation, it is shown that vesicular structures assembled during hydration and reassembled after their centrifugation occupy different positions in the energetic pathway of their preparation. The results obtained should also be taken into account when determining the entrapment efficiency, since this procedure uses centrifugation to separate the load. It is important to note that the physical stability of niosomes, which is usually considered in terms of the functional activity of particles, is manifest and should be considered at the material preparation stage. Despite past efforts towards therapeutical innovation, cancer remains a highly incident and lethal disease, with current treatments lacking efficiency and leading to severe side effects. Hence, it is imperative to develop new, more efficient, and safer therapies. Bee venom has proven to have multiple and synergistic bioactivities, including antitumor effects. Nevertheless, some toxic effects have been associated with its administration. To tackle these issues, in this work, bee venom-loaded niosomes were developed, for cancer treatment. The vesicles had a small (150 nm) and homogeneous (polydispersity index of 0.162) particle size, and revealed good therapeutic efficacy in in vitro gastric, colorectal, breast, lung, and cervical cancer models (inhibitory concentrations between 12.37 ng/mL and 14.72 ng/mL). Additionally, they also revealed substantial anti-inflammatory activity (inhibitory concentration of 28.98 ng/mL), effects complementary to direct antitumor activity. Niosome safety was also assessed, both in vitro (skin, liver, and kidney cells) and ex vivo (hen's egg chorioallantoic membrane), and results showed that compound encapsulation increased its safety. Hence, small, and homogeneous bee venom-loaded niosomes were successfully developed, with substantial anticancer and anti-inflammatory effects, making them potentially promising primary or adjuvant cancer therapies. Niosomes, vesicles formed by non-ionic surfactants, offer advantages in drug delivery by enhancing solubilization and encapsulating both lipophilic and hydrophilic drugs. This study focuses on ß-sitosterolloaded niosomes designed to protect its anti-diabetic properties. Formulated with Span™ 60, Span™ 80 and cholesterol, the niosomes showed high entrapment efficiency (90%) and favorable release kinetics. Stability studies confirmed their robustness in various conditions. In vivo experiments demonstrated significant improvements in body weight and blood glucose levels in diabetic rats. The purpose of this study was to conduct the kinetic assessment of iontophoretic delivery of niosomal tetracycline-HCl formulated in an electroconductive gel. Tween-80 and Span-80 were used to obtain tetracycline-HCl niosomes with an average diameter of 101.9 ± 3.3 nm, a polydispersity index of 0.247 ± 0.004, a zeta potential of − 34.1 mV, and an entrapment efficiency of 70.08 ± 0.16%. Four different gel preparations, two of which contained niosomal tetracycline-HCl, were transdermally delivered using Franz diffusion cells under the trigger effect of iontophoresis, applied at 0.2, 0.5, and 1 mA/cm2 current density. The control group was the passive diffusion results of the preparation made using a tetracycline-HCl-based drug marketed in Turkey. The control group was compared with the groups that contained (a) tetracycline-HCl in an electroconductive gel, (b) the niosomal tetracycline-HCl formulation in water, and (c) the niosomal tetracycline-HCl formulation in the electroconductive gel. The group with the niosomal formulation in the electroconductive gel displayed the highest increase in iontophoretic transdermal delivery relative to the control group, displaying a 2-, 2.1-, and 2.2-fold increase, respectively, by current density. The experimental results of transdermal delivery using the synergistic effect of niosomal formulation in electroconductive gel and the trigger effect of iontophoresis appeared to divert slightly from zero-order kinetics, demonstrating a statistically significant increase in the rate of controlled transdermal drug delivery. Considering that about 20% of the formulation is transdermally delivered in the first half-hour, the iontophoretic transdermal delivery of niosomal tetracycline-HCl can be efficiently used in local iontophoretic therapy. The male reproductive system is negatively influenced by Al exposure. Al represented a considerable hazard to men's reproduction capabilities. Amygdalin (AMG) and spirulina platensis (SP) have been considered to have a strong antioxidant and repro-protective activity; also, targeted drug delivery systems called niosomes improve the distribution of water-soluble medications like amygdalin and spirulina. Current study targeted to determine the effectiveness of AMG and SP against negative reproductive impact resulted by aluminum chloride (AlCl3) toxicity. Sixty adult male albino rats were separated into 6 groups, including the control group, which received distilled water; AlCl3 group, which received AlCl3; AMG+AlCl3 group, which received AlCl3+AMG; AMGLN+AlCl3 group, which received AlCl3+amygdalin-loaded niosomes; SP+AlCl3 group, which received AlCl3+SP; and SPLN+AlCl3 group, which received AlCl3+spirulina-loaded niosomes. All treatments were orally gavaged daily for 5 weeks, and rats were weighed weekly. At the termination of the experiment, some males (three from each group) were used for fertility traits via mating thirty virgin rat females (in a ratio of 1:2 and 2:3 male:female, respectively) followed by recording of birth weights and litter size (number of pups per each female) at birth to assess males' reproductive capability. Other males were euthanized for collection of serum, epididymal semen samples, and tissue samples for biochemical, sperm evaluation, gene expression, and histopathological measurements. There are a considerable number of negative impacts of AlCl3 on male fertility clarified by declined serum testosterone levels; an increased oxidative stress (MDA, TAC); deteriorated semen quality; down-regulation of CYP11A1, StAR, and HSD-3b gene expressions; and testicular tissue degenerative changes. In addition, litter size (number of pups per each female) and birth weights of pups obtained from mated females were affected. AMG and SP treatments, either in niosomal or conventional form, alleviated the AlCl3 negative effects by reducing oxidative stress; increasing testosterone levels; improving semen quality; upregulating of CYP11A1, StAR, and HSD-3b gene expressions; and reducing degenerative changes of testicular tissue. Besides, negative reproductive effect was diminished as observed by changes in the litter size (number of pups per each female) and birth weights of pups obtained from mated females. AMG and SP treatments (either in niosomal or conventional form), ameliorated the AlCl3 negative effects as they possess powerful antioxidant activity, as well as they have the ability to improve the reproductive activity of affected males. The objective of this study was to prepare and conduct a comparative analysis on 5-fluorouracil (5FU) niosomes stabilized with or without Soluplus, utilizing, respectively, passive loading and active loading techniques. Cholesterol was combined with Span 40 and Tween 40 surfactants to prepare several niosomal formulations by either passive or active methods. Encapsulation efficiency (EE%), loading capacity (LC), mean particle sizes, polydispersity indices (PDI), scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), pH stability, and in vitro drug release in bio relevant media of pH 1.2, 6.8, and 7.4 were employed to evaluate the formulations. Batches made using the active loading approach (B1-B5) demonstrated higher entrapment efficiency (25–40%) (p ˂ 0.05) than those made using the passive loading method (20%) (with the exception of batch A1). In comparison to niosomes made using the passive loading method, the active loading methodology produced niosomes with a lower PDI (˂ 0.25) and particle size (˂ 70 nm). SEM revealed spherical and discrete vesicles for the Soluplus stabilized niosomes. Both approaches produced niosomes with prominent hydrogen bonding as observed in the FTIR study. The formulations were stable and sustained drug release, but the active loading strategy imparted a more sustained release pattern than the passively loaded one. Soluplus stabilized niosomes demonstrated higher (p ˂ 0.05) drug release (60–70%) in all release media. Based on the production of smaller particle sizes and narrower size distribution, the active loading strategy may be a more effective way to load hydrophilic medicines such as 5FU into niosomes. One of the most effective strategies to fight viruses and handle health diseases is vaccination. Recent studies and current applications are moving on antigen, DNA and RNA-based vaccines to overcome the limitations related to the conventional vaccination strategies, such as low safety, necessity of multiple injection, and side effects. However, due to the instability of pristine antigen, RNA and DNA molecules, the use of nanocarriers is required. Among the different nanocarriers proposed for vaccinal applications, three types of nanovesicles were selected and analysed in this review: liposomes, transfersomes and niosomes. PubMed, Scopus and Google Scholar databases were used for searching recent papers on the most frequently used conventional and innovative methods of production of these nanovesicles. Weaknesses and limitations of conventional methods (i.e., multiple post-processing, solvent residue, batch-mode processes) can be overcome using innovative methods, in particular, the ones assisted by supercritical carbon dioxide. SuperSomes process emerged as a promising production technique of solvent-free nanovesicles, since it can be easily scaled-up, works in continuous-mode, and does not require further post-processing steps to obtain the desired products. As a result of the literature analysis, supercritical carbon dioxide assisted methods attracted a lot of interest for nanovesicles production in the vaccinal field. However, despite their numerous advantages, supercritical processes require further studies for the production of liposomes, transfersomes and niosomes with the aim of reaching well-defined technologies suitable for industrial applications and mass production of vaccines. Today, the incorporation of liposomes in gels serves as local depots, thus being a fundamental condition for sustained and controlled drug release to optimize patient's outcomes. Insights into niosome-based gels reveal their potential in overcoming conventional drug delivery challenges, positioning them as promising platforms for controlled and targeted drug release due to enhanced stability. Recent investigations highlight the adaptability of niosomal gels in encapsulating both hydrophobic and hydrophilic drugs, ensuring superior stability and sustained release. The tunable properties of niosomal gels also contributed to optimize bioadhesion and permeation across biological membranes. The primary objective of this work is to present the cutting-edge findings in liposome and niosome-based gel research, highlighting their path in modern drug delivery. Paraquat (PQ) herbicide poisoning is a severe medical problem in developing countries without suitable therapy. This study aimed to investigate the effects of crocin (CCN) and nano crocin (NCCN) on PQ -induced toxicity in the MRC-5 cell line. The results showed that the particle size of NCCN was 140.3 ± 18.0 nm, and the zeta potential of the optimal crocin-loaded niosomes was 23.4 ± 2.8 mV. The NCCN was more effective than CCN in the inhibition of PQ-induced toxicity. Treatment of the MRC-5 cells leads to a decrease in ROS and an increase in SOD, CAT, GPX, and TAC levels in PQ-CCN and PQ-NCCN groups compared with the PQ group. These changes tended to be positively associated with the NCCN compared to CCN. Overall, NCCN was more effective than crocin in treating PQ-induced toxicity in vitro and deserved further preclinical consideration. [ABSTRACT FROM AUTHOR]

Background: Niosomes hold promise as drug delivery systems for cancer treatment, with niosome size impacting stability, biodistribution, and effectiveness. This study optimized methotrexate (MTX)-loaded niosome formulation by studying the effects of components and processing conditions on size. The niosomes formulation was made by the thin-film hydration technique. Results: The optimized formulation (NIO 17) with a 6:2:2 ratio of span 60, soya PC, and cholesterol achieved 55.05% methotrexate encapsulation, particle size 597.2 nm, PDI 0.49, and zeta potential − 23.3 mV. The compatibility of methotrexate with lipids was confirmed via Fourier transform infrared spectroscopy, and transmission electron microscopy revealed spherical, well-dispersed vesicles. Differential scanning calorimetry indicated methotrexate conversion or entrapment within vesicles. In vitro release exhibited a sustained pattern with an initial burst. NIO 17 showed potent anti-cancer activity against B16-F10 cells (GI50: 38.7176 μg/mL). Ex vivo studies suggest tailoring niosome size (597.2–982.3 nm) to target specific skin depths (0–38 μm) for enhanced localized drug delivery. Conclusions: This study demonstrates the potential of methotrexate-loaded niosomes as a novel cancer therapy approach, highlighting the potent anti-cancer activity and transdermal delivery potential of NIO 17. Nanotechnology has helped for decades to transform the agricultural system to make it more effective and to ensure the worlds food supply is sustainable and secure. However, potential toxicological affects, interactions with the biotic or abiotic environment, high cost, unknown life cycles of nanomaterials, and their possible increased bioaccumulation effects are some of the barriers which needs to be addressed. This study evaluates development and potential agricultural application of niosomes nanoparticles loaded with Azadirachta indica seed oil. Niosomes are organic nanomaterials composed of non-ionic surfactants, which are inexpensive, have good encapsulation efficiency, more stability, relatively nontoxic, and biodegradable. Azadirachta indica seed oil loaded niosomes were prepared by thin-film hydration method utilizing different molar ratios of surfactant (Tween 80) and stabilizer (Soy lecithin). The characteristics of niosomes were determined by Fourier Transform Infra-Red Spectroscopy, Dynamic Light Scattering, Optical Microscope, and Transmission Electron Microscopy. Further, the prepared niosomes were statistically optimized using the Box-Behnken experimental design. The optimized condition displayed optimum particle size (<100 nm) and acceptable percentage entrapment efficiencies (> 80%). Anti-microbial efficacy of loaded niosomes were evaluated against plant pathogens Xanthomonas and Pantoea. The prepared niosomes displayed considerable antimicrobial properties and can be potentially used in agriculture for the targeted delivery of natural pesticides (Azadirachta indica seed oil) to combat agricultural diseases. Bupropion (Bup) belongs to the norepinephrine–dopamine reuptake inhibitor (NDRI) class and it is the only FDA-approved drug of its class for the treatment of major depressive disorder (MDD), sold under the name of Wellbutrin. Although bupropion is effective in suppressing the symptoms, its regular use and overdose might lead to seizures and liver failure. Thus, we aimed to nanoformulate bupropion onto a niosomal vesicle to improve its efficacy and achieve the same therapeutic effect at lower scheduled doses. A thin film hydration method was adopted to synthesize and optimize Bup entrapped niosomes using three different surfactants of the sorbitan ester series (Span 20, 40, and 60) in combination with cholesterol. The optimization data determined that the niosome formulated with a cholesterol-to-surfactant ratio of 1:1.5 is the most stable system, with the Bup entrapped niosomes containing Span 20 (Bup@N20C) exhibiting minimal in vitro and in vivo toxicity, and demonstrating the sustained release of Bup in artificial cerebrospinal fluid (ACSF). The Bup@N20C formulation showed increased exploration activity and reduced irregular movements in reserpine-induced depression in the adult zebrafish model, suggesting the potential for mood improvement through the suppression of depression-like behavior which was established by statistical analysis and trajectory data. The Bup@N20C-treated group even surpasses the treatment effect of the positive control group and is comparable to the control group. Hence, it can be inferred that niosomal formulations of Bup represent a promising delivery system capable of achieving the brain delivery of the cargo by bypassing the blood–brain barrier facilitated by their small architectural structure. Transdermal drug delivery has gained significant attention as a non-invasive and convenient method for administering drugs. However, the stratum corneum, the outermost layer of the skin, poses a significant barrier to drug permeation. To overcome this challenge, vesicular carriers have emerged as promising systems for enhancing drug delivery through the skin. This review highlights recent advances in the development of vesicular carriers for transdermal drug delivery. Liposomes, niosomes, transfersomes, ethosomes, and solid lipid nanoparticles are among the commonly used vesicular carriers. These carriers offer advantages such as improved drug solubility, prolonged drug release, and enhanced drug stability. Additionally, they can encapsulate a wide range of drugs, including hydrophilic and lipophilic compounds. Various strategies have been employed to optimize vesicular carriers for transdermal drug delivery. These include modifying the vesicle composition, size, and surface charge to enhance skin penetration. The incorporation of penetration enhancers, such as surfactants, has also been explored to improve drug permeation across the skin. Furthermore, advancements in nanotechnology have led to the development of novel vesicular carriers, such as nanostructured lipid carriers and elastic liposomes. These carriers offer improved drug loading capacity, sustained release profiles, and enhanced skin penetration. Moreover, the use of vesicular carriers has shown promise in delivering a wide range of therapeutic agents, including small molecules, peptides, proteins, and genetic material.
