The vibronic structure and nuclear dynamics in the first five excited singlet electronic states of bithiophene (2T) are investigated here. Specifically, considerations are given to comprehend the first two structureless and broad electronic absorption bands and the role of nonadiabatic coupling in the excited state relaxation mechanism of 2T in the gas phase. Associated potential energy surfaces (PESs) are established by constructing a model vibronic coupling Hamiltonian using 18 vibrational degrees of freedom and extensive ab initio electronic structure calculations. The topographies of these PESs are critically examined, and multiple conical intersections are established. The nuclear dynamics calculations are performed by propagating wave packets on the coupled electronic manifold. The present theoretical results are in good agreement with the experimental observations. It is found that strong nonadiabatic coupling between the S1–S4 and S1–S5 states along totally symmetric modes is predominantly responsible for the structureless and broad first absorption band, and overlapping S2, S3, S4, and S5 states form the second absorption band. Photorelaxation from the highly excited S5 to the lowest S1 state takes place through a cascade of diabatic population transfers among the S1–S4–S5 electronic manifold within the first ∼100 fs. Totally symmetric C=C stretching, C–S stretching, C–H wagging, ring puckering, and inter-ring bending modes collectively drive such relaxation dynamics. [ABSTRACT FROM AUTHOR]

The Random-Phase approximation (RPA) provides an appealing framework for semi-local density functional theory. In its Resolution-of-the-Identity (RI) approach, it is a very accurate and more cost-effective method than most other wavefunction-based correlation methods. For widespread applications, efficient implementations of nuclear gradients for structure optimizations and data sampling of machine learning approaches are required. We report a well scaling implementation of RI-RPA nuclear gradients on massively parallel computers. The approach is applied to two polymorphs of the benzene crystal obtaining very good cohesive and relative energies. Note: Photoelectron imaging of MnO3- to probe its nuclear and electronic structure In this note, we revisit the MnO SB 3 sb SP - sp photoelectron spectrum using photoelectron imaging and reassess the assignment, showing that in fact, MnO SB 3 sb is planar in its ground electronic state. The nuclear and magnetic structures of CaxSr2−xWMnO6 (x = 0.5, 1.0, 1.5) have been investigated by neutron diffraction at 295, 30 or 40 K, and 4 or 5 K. The compounds crystallized in the monoclinic symmetry with a space group P21/n. The nuclear structure consists of ordered WO6 and Mn2+O6 octahedral frameworks, with lattice constant a decreasing more rapidly than b with increasing Ca content. No structural change was found around the magnetic transition temperature, although the Mn octahedron below the magnetic order temperature exhibited a slightly compressed Jahn–Teller distortion (0.1–0.3 Å) along the c axis. A standard magnetic model was used to solve the magnetic structure, with the Mn magnetic moments ferromagnetically coupled along the a axis and anti-ferromagnetically coupled along the b and c axes, resulting in a magnetic propagation vector of k = (0,1/2, 1/2) and a magnetic superlattice of 1a × 2b × 2c. In each crystallographic cell, the Mn1–Mn2 atoms are antiferromagnetically configurated, compensating the total moment to zero by symmetry. A layer-type magnetism along the (111) crystallographic plane was confirmed to couple antiferromagnetically between these planes and ferromagnetically within them. The magnetic structure refinement confirmed an average Mn moment of 4.5 μB. Also, it reveals that the collinear magnetic Mn configuration has three possible spin orientations, namely, (with their standard deviation) [μx = (3.3 ± 0.4) μB, μy = (2.5 ± 0.3) μB, μz = (−0.7 ± 0.7) μB], [μx = (3.0 ± 0.4) μB, μy = (0.5 ± 0.2) μB, μz = (−2.8 ± 0.2) μB, and [μx = (−1.8 ± 0.4) μB, μy = (2.9 ± 0.2) μB, μz = (−2.4 ± 0.3) μB. The first configuration is estimated to be the ground state, while the other magnetic configurations are an optimum local value derived from the refinements. Renal Cell Carcinoma (RCC) is the most common malignant tumor (85%) of kidney cancer and has a complex histological pattern and nuclear structure. The manual diagnosis of kidney cancer or any other cancer from histopathology image depends on the knowledge and experience of pathologists, and the pathologist's experience influences the results. According to studies, the kind of histology in kidney cancer is related to the prognosis and course of treatment. Since the kind of histology, molecular profile, and stage of the disease all affect how the disease is treated, there is an essential need to develop an automated system that can precisely analyze the histopathological images of the disease. This work demonstrates how a deep learning framework can be used to predict and classify associated grades of RCC from provided haematoxylin and eosin (H &E) images. The proposed model focuses on two important tasks- First to capture and extract associated features from the H &E images of five different grades. Second, to classify the new set of unseen H &E images into five separate grades using the obtained features. The proposed architecture has been tested and experimented on two independent datasets containing H &E stained histopathology images. The proposed architecture has been examined using the following performance metrics namely precision, recall, F1 - score, accuracy, Floating-point operations (FLOPs), and the total number of parameters. The obtained results show that the proposed architecture attains better results over seven state-of-the-art deep learning architectures on two different H &E stained histopathology image datasets. Summary: This paper evaluates the seismic performance of the frame‐bent structure of a conventional island main building in a nuclear power plant (NPP) using the CAP1400 NPP project of Rongcheng Shidaowan in Shandong Province as a case study. A 12‐pin 3‐span prototype structural model of a steel reinforced concrete (SRC) frame‐bent main building is established in ABAQUS, and its seismic fragility is analyzed. The results show that under the conditions of an 8‐degree frequent earthquake and an 8‐degree fortification earthquake, the transcendental probability under the four limit states of the structural seismic fragility matrix with Sa as intensity measure (IM) is higher than that with peak ground acceleration (PGA) as IM. Moreover, under an 8‐degree rare earthquake, the structure has completely exceeded the normal service performance level and has reached the service performance level of temporary use and repair. The seismic damage index of the structure demonstrates that the structure's seismic damage does not exceed the medium damage state under a small earthquake. However, under the action of a moderate earthquake, the seismic damage of the structure ranges from moderate to serious damage. Experimental and theoretical studies of β electrons (electrons emitted in β-decay transitions) and their β-electron spectra have recently experienced a rapid expansion. These β spectral shapes have been used to study total β spectra of fission-product nuclei in the quest for explanation of the reactor-flux anomalies, and individual β transitions in search for β spectral shapes sensitive to the effective value of the weak axial coupling gA. In the former case the TAGS (total absorption gamma-ray spectroscopy) can be efficiently used to measure the total β spectral shapes and in the latter case dedicated measurements of the involved forbidden nonunique β transitions have been deployed. The fourth-forbidden nonunique decay transitions 113Cd(1/2+g.s → 113In(9/2+g.s and 115In(9/2+g.s 115Sn(1/2+g.s represent theoretically and experimentally much-studied cases where the total β spectra consist of these single transitions. In these particular cases the TAGS method could be used to assess the effective value of gA. In the present work we have identified five more interesting cases where a total β spectrum consists of a single transition. These spectra correspond to second-forbidden nonunique transitions and are gA and/or sNME dependent, where sNME denotes the so-called small relativistic vector nuclear matrix element. These studies have been performed using the nuclear shell model with well established effective Hamiltonians. With this we target to β transitions that would potentially be of high interest for the TAGS and present and future dedicated β -spectrum experiments. [ABSTRACT FROM AUTHOR]

Study of the nuclear structure of nuclides used for medical diagnosis using positron emission tomography (PET) is presented in this work. These nuclides are 11C, 13N, 15O and 18F, which are unstable by emitting β+ particles. They have half-lives of several minutes, which make them suitable radionuclides for PET. These radionuclides are produced through nuclear reactions using a cyclotron.  Studying the nuclear structure of these light nuclides is performed through the shell model. The full space of the p-shell model is used for the p-shell nuclei, and the full space of the sd-shell model for the sd-shell nucleus. The study includes calculations of magnetic moments and quadrupole electric moments. Calculations of form factors are presented to give predictions for future experimental studies. Protons and neutrons’ effective charges are used to include core-polarization effects. The single-particle states used in the calculations are those of the harmonic oscillator potential. Evaluated nuclear structure and decay data for all nuclei with mass number A=202 (202Os, 202Ir, 202Pt, 202Au, 202Hg, 202Tl, 202Pb, 202Bi, 202Po, 202At, 202Rn, 202Fr and 202Ra), are presented. All available experimental data are compiled and evaluated, and best values for level and γ -ray energies, quantum numbers, lifetimes, γ -ray intensities and transition probabilities, as well as other nuclear properties, are recommended. Inconsistencies and discrepancies that exist in the literature are discussed. A number of computer codes (https://www.nds.iaea.org/public/ensdf%5fpgm/) developed by members of the NSDD network were used during the evaluation process. Experimental nuclear structure and decay data are evaluated for 13 known nuclides of mass number A=63 (Ti, V, Cr, Mn, Fe, Co, Ni, Cu, Zn, Ga, Ge, As, Se). Detailed evaluated level properties and related information are presented and recommended spectroscopic parameters are provided, combining all the data from various reactions and decays. No excited state has been observed for 63Ti, 63As and 63Se, among which the first two nuclides have no halflife, decay modes and spin-parity being measured and reported yet. The ground-state spins and/or parities of 63V, 63Cr, 63Mn, and 63Ge still have not been definitely determined and information for excited states in these nuclides plus 63Fe is limited. Information for 63Ga is mostly from one measurement and no data for 63Ge ε + β + decay have been measured while this decay mode (%100) of the 63Ge parent is known. The 63Cu nuclide is the most extensively studied via more than 28 different reactions and decays, followed by 63Zn and 63Ni. Among all those nuclides, only 63Cu has a complete radioactive decay scheme. This work supersedes earlier full evaluations of A=63 published by 2001Ba27, 1991Ki10, 1979Au10, and 1975Au03. Nuclear envelopathies are rare genetic diseases that compromise the integrity of the nuclear envelope. Patients with a defect in LEM domain nuclear envelope protein 2 (LEMD2) leading to LEMD2‐associated progeroid syndrome are exceedingly scarce in number, yet they exhibit shared clinical features including skeletal abnormalities and a prematurely‐aged appearance. Our study broadens the understanding of LEMD2‐associated progeroid syndrome by detailing its phenotypic and molecular characteristics in the first female and fourth reported case, highlighting a distinct impact on metabolic functions. The patient's history revealed growth delay, facial and skeletal abnormalities, and recurrent abdominal pain crises caused by hepatomegaly. Comparisons with the previously documented cases emphasized similarities in skeletal and facial features while showcasing unique variations, notably in cardiac and hepatic manifestations. In vitro experiments conducted on patient‐derived peripheral blood and urinary epithelial cells and LEMD2‐downregulated HepG2 cells confirmed abnormalities in the structure of the nuclear envelope in all three tissue‐types. Overall, our work offers a comprehensive profile of a patient with LEMD2‐related syndrome, emphasizing the hepatic involvement in the disease and broadening our understanding of clinical and molecular implications. This study not only contributes specific insights into LEMD2‐related conditions but also underscores potential therapeutic paths for disorders affecting nuclear envelope dynamics. Phosphorus (P) and potassium (K) stress significantly affect the growth, physiological characteristics, and nutrient uptake of rice plants. This study investigated the photosynthetic nitrogen (N) metabolism, N uptake, and N utilization of plants under varied P and K supplies. Two local conventional high-yield rice varieties (Shennong 265 and Liaojing 294) were used. These varieties were subjected to the following hydroponic experimental treatments: HPHK (normal P and K concentrations), HPLK (normal P and 1/20 normal K concentration), LPHK (normal K and 1/20 normal P concentration), and LPLK (1/20 normal P and K concentrations). The results showed that the mesophyll cells had a relatively complete nuclear and chloroplast structures, and the antioxidant enzymes of the plants were significantly reduced under the HPHK treatment. Compared to the LP treatments (LPHK and LPLK), the HPHK treatment was found to have the following potential effects: effectively optimize plant configuration; promote leaf development (Pn, E, Ci, and Tr, chlorophyll, and leaf area index); significantly increase the N-metabolism-related enzyme activity of leaves and roots and the accumulation of N in the plant in the main growth stages; and significantly increase the rice yield and N-related efficiency. In conclusion, the HPHK treatment was found to be beneficial in improving the plant configuration, promoting photosynthetic N metabolism, and increasing grain yield and N-related utilization efficiency. Post-neutron mass yield distribution in the epi-cadmium neutron-induced fission of 233U has been carried out by measuring the cumulative yields of various fission products within the mass ranges of 77 to 117 and 123 to 153 using an off-line gamma-ray spectrometric technique. Independent yields of a few fission products were also measured by using the same technique. Charge distribution correction has been applied on cumulative yields to obtain the post-neutron mass yields. The mass yield distribution parameters such as full-width at tenth-maximum of light and heavy mass wings, the average light mass and heavy mass , and the average number of neutrons <ν> were obtained. The spectrum average neutron energy is 1.9 MeV. Thus, the role of excitation energy on the nuclear structure effect was examined by comparing the mass yield data in between the epi-cadmium and thermal neutron–induced fission of 233U. We have developed a finite element model to simulate the penetration of nanoneedles into the cellular nucleus. It is found that the nuclear lamina, the primary supporting structure of the nuclear membrane, plays a crucial role in maintaining the integrity of the nuclear envelope and enhancing stress concentration in the nuclear membrane. Notably, nuclear lamina A exhibits a more pronounced effect compared to nuclear lamina B. Subsequently, we further conducted experiments by controlling the time of osteopontin (OPN) treatment to modify the nuclear lamina density, and the results showed that an increase in nuclear lamina density enhances the probability of nanoneedle penetration into the nuclear membrane. Through employing both simulation and experimental techniques, we have gathered compelling evidence indicating that an augmented density of nuclear lamina A can enhance the penetration of nanoneedles into the nuclear membrane. This article discusses the field of nuclear physics and its various applications and frontiers. It highlights the importance of collaboration between theory and experiment in making breakthroughs in the field. Well-developed α clusters are known to exist in light N=Z nuclei, and their properties are reasonably well described with modern nuclear structure theories. However, 'modestly' developed clusters in medium to heavy nuclei remain little understood, both theoretically and experimentally. Extension of the focus to include modestly developed clusters leads us to a concept of 'generalized clusters' and 'cluster ubiquitousness'. The former includes clusters more weakly bound than an α cluster, such as deuteron, triton and He3 , and even clusters partially broken owing to nuclear medium effects. The latter means the existence of clusters in any nuclei, where cluster development was not previously discussed. Effects of the tensor and the spin–orbit interactions on the coexistence of clusters with independent nucleons are discussed using recent nuclear theoretical models. A mixture of the clusters with shell-like components plays an essential role in the synthesis of elements in the universe and the origin of life, together with an α decay. It is also pointed out that clustering in heavy nuclei may accelerate fission and fusion processes. Future experimental plans using cluster knockout reactions, which have the potential to extract information of 'generalized clusters' in a variety of nuclei including stable and unstable nuclei, are also discussed. There are profound connections between neutrino physics and nuclear experiments. Exceptionally precise measurements of single and double beta-decay spectra illuminate the scale and nature of neutrino mass and may finally answer the question of whether neutrinos are their own anti-matter counterparts. Neutrino–nucleus scattering underpins oscillation experiments and probes nuclear structure, neutrinos offer a rare vantage point into collapsing stars and nuclear fission reactors and techniques pioneered in neutrino nuclear physics experiments are advancing quantum sensing technologies. In this article, we review current and planned efforts at the intersection of neutrino and nuclear experiments. The Facility for Antiproton and Ion Research (FAIR) is in its final construction stage next to the campus of the Gesellschaft für Schwerionenforschung Helmholtzzentrum for heavy-ion research in Darmstadt, Germany. Once it starts its operation, it will be the main nuclear physics research facility in many basic sciences and their applications in Europe for the coming decades. Owing to the ability of the new fragment separator, Super-FRagment Separator, to produce high-intensity radioactive ion beams in the energy range up to about 2 GeV/nucleon, these can be used in various nuclear reactions. This opens a unique opportunity for various nuclear structure studies across a range of fields and scales: from low-energy physics via the investigation of multi-neutron systems and halos to high-density nuclear matter and the equation of state, following heavy-ion collisions, fission and study of short-range correlations in nuclei and hypernuclei. The newly developed reactions with relativistic radioactive beams (R3B) set up at FAIR would be the most suitable and versatile for such studies. An overview of highlighted physics cases foreseen at R3B is given, along with possible future opportunities, at FAIR. One of the overarching goals in nuclear science is to understand how the nuclear chart emerges from the underlying fundamental interactions. The description of the structure of nuclei from first principles, using ab initio methods for the solution of the many-nucleon problem with inputs from chiral effective field theory, has advanced dramatically over the past two decades. We present an overview over the available ab initio tools with a specific emphasis on electromagnetic observables, such as multipole moments and transition strengths. These observables still pose a challenge for ab initio theory and are one of the most exciting domains to exploit synergies with modern experiments. Precise experimental data are vital for the validation of the theory predictions and the refinement of ab initio methods. We discuss some of the past and future experimental efforts highlighting these synergies. Globozoospermia is a form of male infertility characterized by spermatozoa with spherical heads lacking acrosomes. The aim of this study was to evaluate ultrastructural and molecular defects in different types of globozoospermia. Semen samples from 12 infertile patients (9 with complete globozoospermia and 3 with partial globozoospermia) and 10 normozoospermic men (control) were examined by transmission electron microscopy and immunocytochemistry with antibodies against lamin B1. The presence of lamin A and progerin was assessed by reverse transcription-PCR. Whole exome sequencing was performed in three patients. In semen sampleswith complete and partial globozoospermia, lamin B1 was observed at the periphery of sperm nuclei, whereas lamin A and progerin were absent. Nuclear envelope pores were found in spermatozoa from both patient groups, regardless of morphology and chromatin condensation, in contrast to the control group. Non-condensed chromatin was present in 51%-81% of cases of complete globozoospermia and in 36%-79% of cases of partial globozoospermia. Homozygous DPY19L2 and SPATA16 variants were identified in two patients with partial globozoospermia and one patient with complete globozoospermia. An atypical nuclear membrane with abnormal nuclear pore distribution and lamin B1 localization was observed in spermatozoa from patients with both complete and partial globozoospermia. The genetic defects in the DPY19L2 and SPATA16 genes detected in patients from both globozoospermic groups suggest a generalized disruption of nuclear structure in globozoospermia, highlighting the genetic and phenotypic similarities between complete and partial globozoospermia. Although the principal physical behaviour of a material is inherently connected to its fundamental crystal structure, the behaviours observed in the real-world are often driven by the microstructure, which for many polycrystalline materials, equates to the size and shape of the constituent crystal grains. Here we highlight a cutting edge synthesis route to the controlled engineering of grain structures in thin films and the simplification of associated 3-dimensional problems to less complex 2D ones. This has been applied to the actinide ceramic, uranium dioxide, to replicate structures typical in nuclear fission fuel pellets, in order to investigate the oxidation and subsequent transformation of cubic UO2 to orthorhombic U3O8. This article shows how this synthesis approach could be utilised to investigate a range of phenomena, affected by grain morphology, and highlights some unusual results in the oxidation behaviour of UO2, regarding the phase transition to U3O8. A prestressed concrete containment is the enclosure structure of a nuclear reactor that serves as the last physical barrier of a nuclear power plant (NPP) safety defense system. It plays a key role in the prestress time-limited aging analysis (TLAA) required for operating license extensions for NPPs. Considering prestress containment systems without long-term monitoring tendons in a nuclear power plant, the technical route for prestress TLAAs involves analyzing operating license extension regulations and in-service monitoring technical requirements for prestressed tendons performance. Using tendons based on theoretical calculations of prestress loss in a nuclear power plant and the minimum required value of prestress determined by numerical simulation, the theoretical predictive value can be compared with the minimum required value for prestressed steel tendons. This comparison can be used to evaluate the 20-year life extension of the prestress system and provide a reference for aging and life management of NPPs. [ABSTRACT FROM AUTHOR]

Summary: Arabidopsis lamin analogs CROWDED NUCLEIs (CRWNs) are necessary to maintain nuclear structure, genome function, and proper plant growth. However, whether and how CRWNs impact reproduction and genome‐wide epigenetic modifications is unknown. Here, we investigate the role of CRWNs during the development of gametophytes, seeds, and endosperm, using genomic and epigenomic profiling methods.We observed defects in crwn mutant seeds including seed abortion and reduced germination rate. Quadruple crwn null genotypes were rarely transmitted through gametophytes. Because defects in seeds often stem from abnormal endosperm development, we focused on crwn1 crwn2 (crwn1/2) endosperm. These mutant seeds exhibited enlarged chalazal endosperm cysts and increased expression of stress‐related genes and the MADS‐box transcription factor PHERES1 and its targets.Previously, it was shown that PHERES1 expression is regulated by H3K27me3 and that CRWN1 interacts with the PRC2 interactor PWO1. Thus, we tested whether crwn1/2 alters H3K27me3 patterns. We observed a mild loss of H3K27me3 at several hundred loci, which differed between endosperm and leaves.These data indicate that CRWNs are necessary to maintain the H3K27me3 landscape, with tissue‐specific chromatin and transcriptional consequences. Progerin, the protein that causes Hutchinson-Gilford progeria syndrome, triggers nuclear membrane (NM) ruptures and blebs, but the mechanisms are unclear. We suspected that the expression of progerin changes the overall structure of the nuclear lamina. High-resolution microscopy of smooth muscle cells (SMCs) revealed that lamin A and lamin B1 form independent meshworks with uniformly spaced openings (~0.085 μm²). The expression of progerin in SMCs resulted in the formation of an irregular meshwork with clusters of large openings (up to 1.4 μm²). The expression of progerin acted in a dominant-negative fashion to disrupt the morphology of the endogenous lamin B1 meshwork, triggering irregularities and large openings that closely resembled the irregularities and openings in the progerin meshwork. These abnormal meshworks were strongly associated with NM ruptures and blebs. Of note, the progerin meshwork was markedly abnormal in nuclear blebs that were deficient in lamin B1 (~50% of all blebs). That observation suggested that higher levels of lamin B1 expression might normalize the progerin meshwork and prevent NM ruptures and blebs. Indeed, increased lamin B1 expression reversed the morphological abnormalities in the progerin meshwork and markedly reduced the frequency of NM ruptures and blebs. Thus, progerin expression disrupts the overall structure of the nuclear lamina, but that effect--along with NM ruptures and blebs--can be abrogated by increased lamin B1 expression. Nucleon–nucleon short range correlations (SRCs) in nuclei are important part of the nuclear structure and have been a subject of intensive studies in the last years. These objects represent temporary fluctuations in average nuclear matter density that occur when two or more nucleons separated by about 1 fm = 10–13 cm. Inspired by recent theoretical and experimental progress in understanding of SRC we reanalyzed our data on the production of high momentum and mesons emitted from nuclear targets irradiated by a proton beam. One of the basic properties of SRCs is the universality which means that they are equally involved in the creation of different hadrons on different nuclei, reflecting the properties of nuclear matter. So far, the property of universality has been observed in electron–nuclear collisions for the breakup of proton–neutron correlations, as well as in proton–nuclear collisions for the production of cumulative pions with high momenta. Both processes involve only light quarks. In this letter, we report the first observation of SRC universality in kaon production on nuclei in the kinematic region of high energy–momentum transfers thereby extending the investigation of the SRC universality to the sector of strange quarks. Objective(s): Gene therapy is a hopeful approach for treatment of a wide range of life threatening disease from infectious and inherited diseases to cancer. Multifunctional Envelope-type Nano Device (MEND) is a new carrier as non-viral genetic vector. Moreover, associating peptide structures with the nuclear localization signals (NLSs), which contains various functional groups enables them to condense DNA and specifically transfer genetic material to the nucleus. Materials and Methods: In this study, two forms of low molecular weight protamine (LMWP) were used for preparation of MEND carrier. The MEND carriers were then targeted with GE11 ligand to obtain T-MEND structure. The size distribution of the resulting nanoparticles, as well as their transfection efficiency and cytotoxicity, were investigated on the A549 cell line. Results: Results demonstrated that the size of polyplex carrier's formulation by both peptides (VV45 and VV32) was below 200 nm and MEND formulations were between 200-300 nm. T-MEND formulations contained VV32 and VV45 peptides showed slightly higher transfection than similar MEND formulations. Also, MEND formulation showed increased transfection efficiency compared to similar PD complexes. The result of metabolic activity test showed that MEND lipopolyplex did not represent any remarkable cytotoxicity. Conclusion: It can be concluded that multifunctional carriers designed based on LMWP are considered as the safe carrier for gene delivery. Presence of protamine and targeted ligand in the nanoparticulate structure did not increase the risk of cytotoxicity of carriers. So, MEND and T-MEND lipoplexes showed low cytotoxicity and acceptable transfection efficiency at the level of PEI 25 kDa. [ABSTRACT FROM AUTHOR]

Stem cell loss in aging and disease is associated with nuclear deformation. Yet, how nuclear shape influences stem cell homeostasis is poorly understood. We investigated this connection using Drosophila germline stem cells, as survival of these stem cells is compromised by dysfunction of the nuclear lamina, the extensive protein network that lines the inner nuclear membrane and gives shape to the nucleus. To induce nuclear distortion in germline stem cells, we used the GAL4-UAS system to increase expression of the permanently farnesylated nuclear lamina protein, Kugelkern, a rate limiting factor for nuclear growth. We show that elevated Kugelkern levels cause severe nuclear distortion in germline stem cells, including extensive thickening and lobulation of the nuclear envelope and nuclear lamina, as well as alteration of internal nuclear compartments. Despite these changes, germline stem cell number, proliferation, and female fertility are preserved, even as females age. Collectively, these data demonstrate that disruption of nuclear architecture does not cause a failure of germline stem cell survival or homeostasis, revealing that nuclear deformation does not invariably promote stem cell loss. Key message: We used marker-free technologies to study chromatin at cellular resolution. Our results show asymmetric chromatin distribution, explore chromatin dynamics during mitosis, and reveal structural differences between trichoblast and atrichoblast cell. The shapes, sizes, and structural organizations of plant nuclei vary considerably among cell types, tissues, and species. This diversity is dependent on various factors, including cellular function, developmental stage, and environmental or physiological conditions. The differences in nuclear structure reflect the state of chromatin, which, in turn, controls gene expression and regulates cell fate. To examine the interrelationship between nuclear structure, cell morphology, and tissue-specific cell proliferation and differentiation processes, we conducted multiple visualizations of H3K4me1, H3K9me2, 4′,6-diamidino-2-phenylindole, 5-ethynyl 2′-deoxyuridine, and SCRI Renaissance 2200, followed by subsequent quantitative analysis of individual cells and nuclei. By assigning cylindrical coordinates to the nuclei in the iRoCS toolbox, we were able to construct in situ digital three-dimensional chromatin maps for all the tissue layers of individual roots. A detailed analysis of the nuclei features of H3K4me1 and H3K9me2 in the mitotic and the elongation zones in trichoblast and atrichoblast cells at the root apical meristem revealed cell type-specific chromatin dynamics with asymmetric distribution of euchromatin and heterochromatin marks that may be associated with cell cycle and cell differentiation characteristics of specific cells. Furthermore, the spatial distribution of nuclei stained with 5-ethynyl 2′-deoxyuridine in the epidermis and cortex tissues suggests short-range coordination of cell division and nuclear migration in a linear sequence through an unknown regulatory mechanism. Linear-scaling techniques for Kohn–Sham density functional theory are essential to describe the ground state properties of extended systems. Still, these techniques often rely on the localization of the density matrix or accurate embedding approaches, limiting their applicability. In contrast, stochastic density functional theory (sDFT) achieves linear- and sub-linear scaling by statistically sampling the ground state density without relying on embedding or imposing localization. In return, ground state observables, such as the forces on the nuclei, fluctuate in sDFT, making optimizing the nuclear structure a highly non-trivial problem. In this work, we combine the most recent noise-reduction schemes for sDFT with stochastic optimization algorithms to perform structure optimization within sDFT. We compare the performance of the stochastic gradient descent approach and its variations (stochastic gradient descent with momentum) with stochastic optimization techniques that rely on the Hessian, such as the stochastic Broyden–Fletcher–Goldfarb–Shanno algorithm. We further provide a detailed assessment of the computational efficiency and its dependence on the optimization parameters of each method for determining the ground state structure of bulk silicon with varying supercell dimensions. Optical quantum memories are key elements in modern quantum technologies to reliably store and retrieve quantum information. At present, they are conceptually limited to the optical wavelength regime. Recent advancements in x-ray quantum optics render an extension of optical quantum memory protocols to ultrashort wavelengths possible, thereby establishing quantum photonics at x-ray energies. Here, we introduce an x-ray quantum memory protocol that utilizes mechanically driven nuclear resonant 57Fe absorbers to form a comb structure in the nuclear absorption spectrum by using the Doppler effect. This room-temperature nuclear frequency comb enables us to control the waveform of x-ray photon wave packets to a high level of accuracy and fidelity using solely mechanical motions. This tunable, robust, and highly flexible system offers a versatile platform for a compact solid-state quantum memory at room temperature for hard x-rays. [ABSTRACT FROM AUTHOR]

The measurements of lifetimes and electromagnetic moments of the isomeric states in some of the nuclei around A ≈ 135 carried out at BARC-TIFR Pelletron Linac Facility at TIFR, Mumbai, are highlighted. The results from these measurements, along with similar studies reported in the literature, provide a wealth of nuclear structure information in Ba, La, and Ce isotopes in the A ≈ 135 region. Comparison of the measured moments with the calculations gives a unique testing ground for the predictive power of the nuclear models. Nuclear isomers, the excited meta-stable states of nuclei, offer profound insights into nuclear structure. This article reviews the intersection of nuclear isomer research with mass spectrometry methodologies, particularly focusing on novel capabilities of the multiple-reflection time-of-flight mass spectrometry (MR-TOF-MS) technique. Through a comprehensive examination of established methods for isomer identification and characterization, alongside the technical principles underlying MR-TOF-MS, this review discusses the pivotal role of mass spectrometry in advancing our understanding of nuclear isomers. The operational principles and recent developments in MR-TOF-MS technology are explained and exemplified through case studies from prominent research facilities. Furthermore, this work discusses ongoing efforts to enhance sensitivity, resolution, and measurement capabilities in MR-TOF-MS, promising continued advancements in nuclear physics research and applications. The longer-lived excited nuclear states, referred as nuclear isomers, exist due to the hindered decays owing to their peculiar nucleonic structural surroundings. Some of these conditions, being exceptionally rare and limited to achieve, elevate certain isomers to the status of extreme and unusual isomers among their kin. For example, the E5 coupling of single-particle orbitals is rare and so are E5 decaying isomers. This review delves into some of such remarkable isomers scattered across the nuclear landscape while highlighting the possibilities to find more of them. Unique properties of some of them, harbor the potential for transformative applications in medicine and energy. An exciting example is that of the lowest energy isomer known so far in 229 Th, which may help realize the dream of an ultra-precise nuclear clock in the coming decade. These isomers also offer an insight into the extremes of nuclear structure associated with them, which leads to their unusual status in energy, half-life, spin etc. The review attempts to highlight isomers with high-multipolarities, high-spins, high-energies, longest half-lives, extremely low energy, etc. A lack of theoretical understanding of the decay rates, half-lives and moments of these isomers is also pointed out. Laser spectroscopy is an indispensable tool in radioactive ion beam facilities. It is used for the study of the nuclear structure of radioactive isotopes, as well as for the production and purification of these radioactive beams. Over the years, a rich dataset of electromagnetic moments, nuclear spins, and changes in the mean-square charge radii of the nuclear ground states and long-lived nuclear isomers has been collected. In addition to these properties commonly measured with laser spectroscopy techniques, this technique has been used for the discovery of new isomers as well. In this work, we give a short overview of the laser spectroscopy of nuclear isomers and illustrate how these measurements have so far contributed to deepening our knowledge of nuclear structure. Quantum tunnelling plays a crucial role in heavy-ion fusion reactions at sub-barrier energies, especially in the context of nuclear physics and astrophysics. The nuclear structure of the colliding nuclei and nucleon transfer processes represent intrinsic degrees of freedom. They are coupled to the relative ion motion and, in general, increase the probability of tunnelling. The influence of couplings to nucleon transfer channels relatively to inelastic excitations, on heavy-ion fusion cross sections, is one of the still open problems in this field. We present a new analysis of several systems, based on the combined observation of the energy-weighted excitation functions E σ in relation to their first energy derivatives d (E σ) / d E . The relation between d (E σ) / d E and E σ removes the basic differences due to the varying Coulomb barrier when comparing different systems. We show that, depending on the nuclear structure and/or the presence of strong transfer channels, this representation reveals characteristic features below the barrier. The possible presence of cross section oscillations makes this analysis less clear for light- or medium-light systems. [ABSTRACT FROM AUTHOR]

Lamin A/C, essential inner nuclear membrane proteins, have been linked to progeria, a disease of accelerated aging, and many other diseases, which include cardiac disorder. Lamin A/C mutation and its phosphorylation are associated with altering nuclear shape and size. The role of lamin A/C in regulating normal cardiac function was reported earlier. In the present study, we hypothesized that Doxorubicin (Dox) may alter total lamin A/C expression and phosphorylation, thereby taking part in cardiac injury. An in vitro cellular injury model was generated with Dox (0.1–10.0 μM) treatment on cardiomyoblast cells (H9c2) to prove our hypothesis. Increased size and irregular (ameboid) nucleus shape were observed in H9c2 cells after Dox treatment. Similarly, we have observed a significant increase in cell death on increasing the Dox concentration. The expression of lamin A/C and its phosphorylation at serine 22 significantly decreased and increased, respectively in H9c2 cells and rat hearts after Dox exposure. Phosphorylation led to depolymerization of the lamin A/C in the inner nuclear membrane and was evidenced by their presence throughout the nucleoplasm as observed by immunocytochemistry techniques. Thinning and perforation on the walls of the nuclear membrane were observed in Dox-treated H9c2 cells. LMNA-overexpression in H9c2 protected the cells from Dox-induced cell death, reversing all changes described above. Further, improvement of lamin A/C levels was observed in Dox-treated H9c2 cells when treated with Purvalanol A, a CDK1 inhibitor and N-acetylcysteine, an antioxidant. The study provides new insight regarding Dox-induced cardiac injury with the involvement of lamin A/C and alteration of inner nuclear membrane structure. Rhodomonas (Cryptophyceae) and species assigned to this genus have undergone numerous taxonomic revisions. This also applies to R. marina studied here as it was originally assigned as a species of Cryptomonas and later considered a variation of R. baltica, the type species. Despite being described more than 130 years ago, R. marina still lacks a comprehensive characterization. Light and electron microscopy were employed to delineate a strain from western Greenland. The living cells were 18 µm long and 9 µm wide, elliptical in shape with a pointed to rounded posterior and truncated anterior in lateral view. Two sub-equal flagella emerged from a vestibulum, where also a furrow extended. In transmission electron microscopy, the furrow was associated with a tubular gullet and the pyrenoid embedded in a deeply lobed chloroplast. The chloroplast contained DNA in perforations and was surrounded by starch grains. A tubular nucleomorph was enclosed within the pyrenoid matrix. In scanning electron microscopy, the inner periplast consisted of rectangular plates with rounded edges and posteriorly these were replaced by a sheet-like structure. The water-soluble pigment was Crypto-Phycoerythrin type I (Cr-PE 545). A phylogenetic inference based on SSU rDNA confirmed the identity of strain S18 as a species of Rhodomonas as it clustered with congeners but also Rhinomonas, Storeatula, and Pyrenomonas. These genera formed a monophyletic clade separated from a diverse assemblage of other cryptophyte genera. To further explore the phylogeny of R. marina a concatenated phylogenetic analysis based on the SSU rDNA-ITS1-5.8S rDNA-ITS2-LSU rDNA region was performed but included only closely related species. The secondary structure of nuclear internal transcribed spacer 2 was predicted and compared to similar structures in related species. Non-isolated structures have strong destructive effects and poor isolation effects when encountering earthquakes. Setting isolation bearings can prolong the natural vibration period of the structure, reduce the horizontal seismic response of the structure under the influence of variables such as acceleration, base reaction, and inter story displacement, and enhance the overall seismic performance of the structure. The new material—epoxy plate thick layer rubber isolation bearing—has unique advantages compared to other bearings, such as effective energy absorption, simple construction, and low cost. This study establishes a three-dimensional isolated nuclear power plant containment structure based on the principle of similarity ratio, and compares and analyzes the acceleration, base reaction, and displacement responses of non-isolated and isolated structures. At the same time, the incremental dynamic analysis method (IDA) is used to analyze the seismic vulnerability of the structure, and the isolation performance of the nuclear containment structure using epoxy plate thick layer rubber isolation bearings is comprehensively and deeply explored. The results show that the epoxy plate thick layer rubber isolation bearing effectively prolongs the natural vibration period of the structure, reduces the horizontal seismic response of the structure, reduces the dome acceleration response by 66.55%, and reduces the base horizontal shear force by 55.51%. Therefore, setting epoxy plate thick layer rubber isolation bearings in the isolation layer can effectively enhance the seismic performance of the structure, thereby improving the redundancy of the nuclear power plant containment structure. Over 50% of nuclear power plants (NPPs) worldwide have operated for over three decades, leading to a surge in decommissioning projects. This study addresses the gap in current guidelines by analyzing risks in nuclear decommissioning. Using the fuzzy-AHP technique, tasks within dismantling radioactive concrete structures are prioritized. Findings reveal structural and human-related risks across five main cutting tasks. Collision emerges as a significant concern, particularly during wire saw installation and concrete block hoisting hole creation. Subcategory risk priorities highlight variations in risk across tasks, with jamming, falling, and falling objects identified as top concerns during wire saw transportation. This study emphasizes the importance of comprehensive risk assessment in enhancing safety during decommissioning. It underscores the need to consider both physical risks and risks to personnel throughout the process. By prioritizing safety, stakeholders can ensure worker safety and operational efficiency while minimizing hazards. This research contributes to standardized safety protocols for nuclear decommissioning worldwide, aligning with sustainable energy practices. The outcomes offer practical insights for safety manual development and decision-making processes. Current practices for structural analysis of extremely large‐molecular‐weight polysaccharides via solution‐state nuclear magnetic resonance (NMR) spectroscopy incorporate partial depolymerization protocols that enable polysaccharide solubilization in suitable solvents. Non‐specific depolymerization techniques utilized for glycosidic bond cleavage, such as chemical degradation or ultrasonication, potentially generate structural fragments that can complicate complete and accurate characterization of polysaccharide structures. Utilization of appropriate enzymes for polysaccharide degradation, on the other hand, requires prior structural knowledge and optimal enzyme activity conditions that are not available to an analyst working with novel or unknown compounds. Herein, we describe an application of a permethylation strategy that allows the complete dissolution of intact polysaccharides for NMR structural characterization. This approach is utilized for NMR analysis of Xylella fastidiosa extracellular polysaccharide (EPS), which is essential for the virulence of the plant pathogen that affects multiple commercial crops and is responsible for multibillion dollar losses each year. Role of effective interaction potential in shears band structure in 2 0 4 At has been investigated in more detail than in previous studies. A semi-classical geometric model is used to obtain the interaction strengths between valence protons and neutron holes constituting the shears blades. The value produces overall good agreement with this region's nuclei. Band crossing is observed in the spin-parity range 1 6 + – 2 3 + . Configurations of the Magnetic Rotational (MR) band are assigned in the light of the Shears mechanism with the Principal Axis Cranking (SPAC) model. Experimental rotational frequencies and the deduced ratio of B (M 1) / B (E 2) are well reproduced by SPAC calculations, suggesting the dipole band in 2 0 4 At can undoubtedly be interpreted as the MR band resulting from the effective interaction of repulsive potential. The pseudo-spin symmetry (PSS) provides an important angle to understand nuclear microscopic structure and the novel phenomena found in unstable nuclei. The relativistic Hartree–Fock (RHF) theory, that takes the important degrees of freedom associated with the π -meson and ρ -tensor (ρ -T) couplings into account, provides an appropriate description of the PSS restoration in realistic nuclei, particularly for the pseudo-spin (PS) doublets with high angular momenta ( l ˜ ). The investigations of the PSS within the RHF theory are recalled in this paper by focusing on the effects of the Fock terms. Aiming at common artificial shell closures appearing in previous relativistic mean-field calculations, the mechanism responsible for the PSS restoration of high- l ˜ orbits is stressed, revealing the manifestation of nuclear in-medium effects on the PSS, and thus, providing qualitative guidance on modeling the in-medium balance between nuclear attractions and repulsions. Moreover, the essential role played by the ρ -T coupling, that contributes mainly via the Fock terms, is introduced as combined with the relations between the PSS and various nuclear phenomena, including the shell structure and the evolution, novel halo and bubble-like phenomena, and the superheavy magicity. As the consequences of the nuclear force in complicated nuclear many-body systems, the PSS itself and the mechanism therein can not only deepen our understanding of nuclear microscopic structure and relevant phenomena, but also provide special insight into the nature of the nuclear force, which can further enrich our knowledge of nuclear physics. The discovery of neutron ushered in a paradigm shift in our understanding of the subatomic world. The electrically neutral neutron does not experience Coulomb repulsion produced by the positively charged protons inside the nucleus. As a result, a neutron can penetrate deep into the nucleus and thus is a powerful probe to unravel the nuclear structure. In 1920, Rutherford prophetically predicted the existence of an electrically neutral particle inside the nucleus. It took more than a decade of painstaking experimentation to finally discover it in 1932. The historical aspects of a scientific discovery, for one reason or another, often, do not find their place in the normal discourse at both school and college levels. Studying the historical development of an idea can inspire, excite curiosity among, and familiarize students about how science works. In this article, we have tried to show how the relevant historical information can be integrated in traditional instructions while discussing the discovery of neutron which may enhance the curriculum outcomes and increase scientific literacy. We have discussed two important experiments that hinted at the existence of a new particle inside the nucleus. The arguments that led Chadwick to the ultimate discovery of neutron are also presented.
