Along the shoreline of the White Sea, due to the post-glacial uplift of the coast, some water bodies with stable stratification have been formed. They have been classified as meromictic as they are at different stages of isolation from the Sea. As separation progresses, significant changes occur in the water column, including the composition of chromophoric dissolved organic matter (CDOM) and the structure of the aquatic microbial community. In this work, we searched for optical proxies of euxinia (anoxic conditions with accumulated hydrogen sulfide) in the water column of the meromictic lagoon on Zeleny Cape. The lagoon is separated from the White Sea basin by a shallow threshold that completely isolates the lagoon during low tide, but marine water enters the lagoon during high tide. The ecosystem in the lagoon is characterized by the marine salinity of water and a high organic matter content in the bottom water and sediments. In this study, spectral methods were used to obtain the depth distribution of CDOM, chlorophyll, and bacteriochlorophyll in the lagoon with strong water stratification and euxinic conditions in the bottom water. The measured optical CDOM characteristics were compared with hydrochemical data (water salinity, Eh, pH, dissolved oxygen), phytoplankton (oxygenic phototrophs), and green sulfur bacteria (anoxygenic phototrophs) distribution along the water column. The spectroscopic methods showed to have the advantages of not requiring water sample pre-treatment and allowing rapid sensing of CDOM and photosynthetic pigments at each horizon. In the current study, soil samples were gathered from different places where petrol and diesel filling stations were located for isolation of photosynthetic bacteria under anaerobic conditions using the paraffin wax-overlay pour plate method with Biebl and Pfennig's medium. The three isolated strains were named Rhodopseudomonas palustris SMR 001 (Mallapur), Rhodopseudomonas palustris NR MPPR (Nacahram) and Rhodopseudomonas faecalis N Raju MPPR (Karolbagh). The morphologies of the bacteria were examined with a scanning electron microscope (SEM). The phylogenetic relationship between R. palustris strains was examined by means of 16S rRNA gene sequence analysis using NCBI-BLAST search and a phylogenetic tree. The sequenced data for R. palustris were deposited with the National Centre for Biotechnology Research (NCBI). The total amino acids produced by the isolated bacteria were determined by HPLC. A total of 14 amino acids and their derivatives were produced by the R. palustris SMR 001 strain. Among these, carnosine was found in the highest concentration (8553.2 ng/mL), followed by isoleucine (1818.044 ng/mL) and anserine (109.5 ng/mL), while R. palustris NR MPPR was found to produce 12 amino acids. Thirteen amino acids and their derivatives were found to be produced from R. faecalis N Raju MPPR, for which the concentration of carnosine (21601.056 ng/mL) was found to be the highest, followed by isoleucine (2032.6 ng/mL) and anserine (227.4 ng/mL). Sericulture, often known as silk farming is the practice of rearing silkworms for production of raw silk. One of the major impediments faced by the sericulture community is the silkworm diseases that cause significant crop losses. In the present study, the efficacy of phototrophic bacteria as feed supplements in improving the economic traits as well as disease resistance in mulberry silkworm, Bombyx morí L. was assessed. A total of three phototrophic bacteria were isolated and identified as belonging to the genera Maríchromatium, Rhodobacter and Rhodopseudomonas based on 16S rRNA gene sequencing analysis. Their morphological, physiological and biochemical characters were studied, grown under ambient conditions and biomass was harvested. Mulberry fortified with the aforementioned phototrophic bacteria in solitude and combinations at varying concentrations (0.5, 1, 2 and 5%) were found to be innocuous to the mulberry silkworm. The best results were observed when mulberry leaf was supplemented with Rhodopseudomonas sp. at 2% concentration which improved survival, cocoon weight, shell weight, shell%, filament length and non-breakable filament length by 5.18, 0.27, 3.86, 3.5, 1.5 and 1.2%, respectively under normal rearing conditions. Under Staphylococcus sp. infected conditions, the aforesaid diet also enhanced survival in silkworms by 16%. On the other hand, the same diet didn't exhibit any discernible influence on survival against fungal, viral, or microsporidian infections in silkworm. [ABSTRACT FROM AUTHOR]

Rhodovulum spp. are anoxygenic phototrophic purple bacteria with versatile metabolisms, including the ability to obtain electrons from minerals in their environment to drive photosynthesis, a relatively novel process called phototrophic extracellular electron uptake (pEEU). A total of 15 strains of Rhodovulum sulfidophilum were isolated from a marine estuary to observe these metabolisms in marine phototrophs. One representative strain, Rhodovulum sulfidophilum strain AB26, can perform phototrophic iron oxidation (photoferrotrophy) and couples carbon dioxide fixation to pEEU. Here, we reclassify two R. sulfidophilum isolates, strainAB26 and strain AB19, as Rhodovulum visakhapatnamense using taxonomic re-evaluation based on 16S and pufM phylogenetic analyses. The strain AB26 genome consists of 4,380,746 base-pairs, including two plasmids, and encodes 4296 predicted protein-coding genes. Strain AB26 contains 22 histidine kinases, 20 response regulators, and dedicates ~16% of its genome to transport. Transcriptomic data under aerobic, photoheterotrophy, photoautotrophy, and pEEU reveals how gene expression varies between metabolisms in a novel R. visakhapatnamense strain. Genome comparison led by transcriptomic data under pEEU reveals potential pEEU-relevant genes both unique to R. visakhapatnamense strains and shared within the R. sulfidophilum genomes. With these data we identify potential pEEU-important transcripts and how speciation may affect molecular mechanisms of pEEU in Rhodovulum species from the same environment. [ABSTRACT FROM AUTHOR]

Heliomicrobium modesticaldum has been used as a model organism for the Heliobacteria, the only phototrophic family in the Firmicutes. It is a moderately thermophilic anoxygenic phototrophic bacterium that is capable of fermentative growth in the dark. The genetic manipulation of H. modesticaldum is still in its infancy. Methods to introduce genes through the use of exogenous plasmids and to delete genes from the chromosome through the use of the native CRISPR/Cas system have been developed in the last several years. To expand our genetic toolkit, it was necessary to control gene expression. In this study, we analyzed constitutive and inducible promoters developed for clostridia for their use in H. modesticaldum and further tested two reporters, adhB and lacZ, as indicators of promoter strength. Alcohol dehydrogenase (AdhB) was unsuitable as a reporter in this species due to high endogenous activity and/or low activity of the reporter, but a thermostable LacZ worked well as a reporter. A set of constitutive promoters previously reported to work in Clostridium thermocellum was found to be reliable for controlling the expression of the lacZ reporter gene in H. modesticaldum at a range of activities spanning an order of magnitude. An anhydrotetracycline-inducible promoter was created by inserting tetO operators into a strong constitutive promoter, but it was not fully repressible. The implementation of a xylose-inducible promoter resulted in complete repression of β-gal in the absence of xylose, and reliable expression tunable through the concentration of xylose added to the culture. Pollutants of inorganic nature (acids, alkalis, mineral salts of different composition, metals) change the course of biological processes of environmental purification, but their influence on the physiological properties of phototrophic sulfur bacteria has not been studied enough. The usage of nitrite ions as an electron donor of anoxygenic photosynthesis by cells of phototrophic green and purple sulfur bacteria Chlorobium limicola IMV K-8, Thiocapsa sp. Ya-2003 and Lamprocystis sp. Ya-2003, isolated from Yavorivske Lake, under the influence of the most widespread inorganic pollutants – hydro- and dihydrophosphates, sulfates, chlorides and chlorates, has been studied. It is shown that KH2PO4, K2HPO4, Na2SO4, NaCl and KClO3, present in the van Niel medium with 4.2 mM NaNO2 at concentrations that are 0.5, 1.0, 2.0, 3.0, 4.0 times different from the maximum permissible concentrations (MPC), influenced the biomass accumulation and nitrite ions oxidation by phototrophic green and purple sulfur bacteria. In media with hydro- and dihydrophosphate ions at concentrations 4.0 times higher than the MPC, inhibition of bacterial growth was up to 1.7 times lower than in the control. The biomass accumulation by bacteria in media with chloride and chlorate ions at concentrations 3.0–4.0 times higher than MPC was 2.0–2.8 times lower compared to the control. In the medium with Na2SO4 at concentrations 2.0–4.0 times higher than MPC, the biomass was 2.0–4.0 times lower than in the control. Nitrites’ oxidation by all strains in the media with the studied pollutants was slowed down. The residual content of nitrite ions in media with hydro- and dihydrophosphate, chloride and chlorate ions at their concentrations 4.0 times higher than MPC, exceeded the NO2– content in the control variants up to 1.7 times. If in the medium without pollutants the cells of C. limicola IMV K-8, Thiocapsa sp. Ya-2003 and Lamprocystis sp. Ya-2003 strains oxidized 72.7%, 72.2% and 71.4%, respectively, of nitrite ions present in the medium, then in the medium with sulfate ions at concentration 4.0 times higher than the MPC, bacteria oxidized nitrite ions only at 39.6%, 34.4% and 27.0%, respectively. Oxidation of a lower quantity of nitrites by phototrophic bacteria in the media with inorganic pollutants led to the production by them of a lower quantity of nitrates. The content of NO3– in the media with hydro-, dihydrophosphate and chlorate ions at all concentrations was up to 1.9 times lower than in the control. In media with sulfate ions at concentrations 2.0–4.0 times higher than MPC and chloride at concentration 4.0 times higher than MPC, the content of nitrate ions was 2.1–4.3 and 2.0 times, respectively, lower than in the control variants. Inorganic pollutants stimulated the synthesis of intracellular carbohydrates in C. limicola IMV K-8. If the content of intracellular glucose in cells grown in the medium without pollutants was 10.3 mg/g dry cell weight, then in cells grown in media with K2HPO4, KH2PO4, Na2SO4, NaCl and KClO3 at concentrations 4.0 times higher than MPC, its content increased by 12.2%, 10.7%, 51.6%, 17.1% and 35.9%, respectively. The glycogen content in the cells grown in the medium without pollutants was 45.1 mg/g dry cell weight. Hydro- and dihydrophosphate, chloride and chlorate ions at concentrations 4.0 times higher than MPC stimulated glycogen synthesis in cells by 47.5%, 57.6%, 67.4% and 74.6%, respectively. The glycogen content in cells grown in the medium with Na2SO4 at concentrations 3.0 and 4.0 times higher than MPC increased by 102.9% and 107.5%, respectively. Therefore, it is established that pollutants of inorganic nature affect the physiological properties of photosynthetic sulfur bacteria and thus change the course of biological processes of environment purification, in particular, from nitrite ions.

Application of three combinations of lactic acid bacteria (Lactobacillus plantarum, Lactobacillus rhamnosus), phototrophic bacteria (Rhodopseudomonas palustris) and yeast (Saccharomyces cerevisiae) with sugar cane molasses, marked as: EM1, EM5 and EM AGRO, against the phytopathogenic fungi of tomato and pepper: Fusarium oxysporum, Alternaria alternata, Botrytis cinerea, Colletotrichum sp., Verticilium dahliae and Pythium aphanidermatum was evaluated in vitro and in vivo. A combination of bacteria and yeast named EM5 showed the highest mycelium growth inhibition against B. cinerea (38.4%) in a double agar diffusion test. In a microdilution test, the combination EM1 showed the highest inhibitory effect on B. cinerea (MIC 1x10-3 ml/ml), while EM5 showed a similar inhibitory effect towards F. oxysporum, A. alternata and Colletotrichum sp. (MIC 10 ml/ml). The use of EM1 (in concentrations 10 and 100 µl/ml) and EM AGRO (10 µl/ml) is recommended for tomato seedling protection. EM1 (100 µl/ml), EM5 and EM AGRO (10 µl/ml) are recommended for pepper seedling protection.

Photosynthetic bacteria (PSB) possess versatile metabolic abilities and are widely applied in environmental bioremediation, bioenergy production and agriculture. In this review, we summarize examples of purple non-sulfur bacteria (PNSB) through biofertilization, biostimulation and biocontrol mechanisms to promote plant growth. They include improvement of nutrient acquisition, production of phytohormones, induction of immune system responses, interaction with resident microbial community. It has also been reported that PNSB can produce an endogenous 5-aminolevulinic acid (5-ALA) to alleviate abiotic stress in plants. Under biotic stress, these bacteria can trigger induced systemic resistance (ISR) of plants against pathogens. The nutrient elements in soil are significantly increased by PNSB inoculation, thus improving fertility. We share experiences of researching and developing an elite PNSB inoculant (Rhodopseudomonas palustris PS3), including strategies for screening and verifying beneficial bacteria as well as the establishment of optimal fermentation and formulation processes for commercialization. The effectiveness of PS3 inoculants for various crops under field conditions, including conventional and organic farming, is presented. We also discuss the underlying plant growth-promoting mechanisms of this bacterium from both microbial and plant viewpoints. This review improves our understanding of the application of PNSB in sustainable crop production and could inspire the development of diverse inoculants to overcome the changes in agricultural environments created by climate change. Abstract Background Phototrophic purple non-sulfur bacteria (PNSB) have gained attention for their ability to produce a valuable clean energy source in the form biohydrogen via photofermentation of a wide variety of organic wastes. For maturation of these phototrophic bioprocesses towards commercial feasibility, development of suitable immobilisation materials is required to allow continuous production from a stable pool of catalytic biomass in which energy is not diverted towards biomass accumulation, and optimal hydrogen production rates are realised. Here, the application of transparent polyvinyl-alcohol (PVA) cryogel beads to immobilisation of Rhodopseudomonas palustris for long-term hydrogen production is described. PVA cryogel properties are characterised and demonstrated to be well suited to the purpose of continuous photofermentation. Finally, analysis of the long-term biocompatibility of the material is illustrated. Results The addition of glycerol co-solvent induces favourable light transmission properties in normally opaque PVA cryogels, especially well-suited to the near-infrared light requirements of PNSB. Material characterisation showed high mechanical resilience, low resistance to diffusion of substrates and high biocompatibility of the material and immobilisation process. The glycerol co-solvent in transparent cryogels offered additional benefit by reinforcing physical interactions to the extent that only a single freeze–thaw cycle was required to form durable cryogels, extending utility beyond only phototrophic bioprocesses. In contrast, conventional PVA cryogels require multiple cycles which compromise viability of entrapped organisms. Hydrogen production studies of immobilised Rhodopseudomonas palustris in batch photobioreactors showed higher specific hydrogen production rates which continued longer than planktonic cultures. Continuous cultivation yielded hydrogen production for at least 67 days from immobilised bacteria, demonstrating the suitability of PVA cryogel immobilisation for long-term phototrophic bioprocesses. Imaged organisms immobilised in cryogels showed a monolithic structure to PVA cryogels, and demonstrated a living, stable, photofermentative population after long-term immobilisation. Conclusion Transparent PVA cryogels offer ideal properties as an immobilisation matrix for phototrophic bacteria and present a low-cost photobioreactor technology for the further advancement of biohydrogen from waste as a sustainable energy source, as well as development of alternative photo-bioprocesses exploiting the unique capabilities of purple non-sulfur bacteria.

Summary: A plant‐associated phototrophic bacterium, R. palustris strain PS3, was inoculated into a soil‐based MFC to generate electricity. We evaluated the performance of this soil‐based microbial fuel cell (MFC) and elucidated the essential factors that contributed to power generation. PS3 showed the potential to enhance power generation, especially when the apparatus was operated in a sealed chamber with illumination. We deduced that the improved power performance was due to the enhanced electron transport through the living electrode that was grown as a PS3 biofilm via photoheterotrophic metabolism. In addition, we suggested that the interplay between phototrophic fixation of ambient CO2 and anaerobic oxidation of ferrous iron in soil was also involved in the increased power output. We implemented CMOS (complementary metal‐oxide‐semiconductor) technology with the soil‐based MFC to harvest energy in a more efficient and stable manner. Abstract—Illumination of purple phototrophic bacteria in the carotenoid absorption band of light harvesting complexes often leads to low energy efficiency of absorbed light. This is due to singlet–triplet fission of carotenoid excitation. In the present study, the nature of this process was explored with a phototrophic bacterium Thermochromatium tepidum as an example. It was demonstrated by time-resolved EPR spectroscopy and magnetic field modulation of the fluorescence yield that the concept of intramolecular excitation fission that was developed in several publications was not confirmed experimentally. Evidence of the intermolecular character of excitation fission that involves two carotenoid molecules of the light-harvesting pigment–protein complexes, LH1-RC and LH2, has been obtained. Abstract Shrimp cultivation has been faced with pathogenic bacterial infections, such as luminous vibriosis and acute hepatopancreatic necrosis disease (AHPND) caused by Vibrio harveyi and Vibrio parahaemolyticus , respectively. Biodegradable compounds that act to prevent both serious shrimp diseases are to be explored for producing safe shrimp. The probiotic purple nonsulfur bacteria (PNSB) are safe for use in producing antivibrio compounds against shrimp pathogenic vibrios. The probiotic PNSB, Rhodobacter sphaeroides (SS15, TKW17) and Afifella marina STW181, released antivibrio compounds inhibiting various shrimp pathogenic Vibrio spp. in the optimum conditions that were similar to the conditions of shrimp cultivation. The antivibrio compounds by these PNSB were stable over the wide pH range from 3 to 10, and at elevated temperatures up to 121 °C for 20 min; they were also quite sensitive to lipase, pronase and α-chymotrypsin. All the antivibrio compounds had bactericidal activity as evidenced by damaged cells with many holes, and they also showed bacteriolytic activity with the highest found for the strain STW181. One of the purified antivibrio compounds produced by the strain SS15, a low molecular weight (<3000 Da) cationic compound containing NH 2 group(s), was the most effective in inhibiting AHPND-causing V. parahaemolyticus SR2. Regarding toxicity to brine shrimp, the extract from water layer in ethyl acetate extraction of strain SS15 (extract-SS15) containing antivibrio peptides had LC50 at 6.79 ± 0.63 mg mL−1. In challenge tests the extract-SS15 at 2.68 mg mL−1 adversely affected water quality; however, it could significantly decrease the mortality of white shrimp infected with the virulent AHPND strain SR2. All the tested probiotic PNSB produced antivibrio compounds that are biodegradable with great potential to control serious shrimp pathogenic vibrios for sustainable shrimp cultivation. Abstract This study aimed to investigate the possibility to remove mercury (Hg) using the live or dead biomass of Hg resistant purple nonsulfur bacteria (PNSB) for their potential application to clean up shrimp ponds and during cultivation. The efficiency of Hg2+ removal by three PNSB strains; Rhodovulum sulfidophilum SRW1–5, and Afifella marina strains SSS2-1 and SSW15-1, grown with 2 different growth conditions (microaerobic light and aerobic dark conditions) were tested (2 mg/L HgCl 2 , 4.5 mg dry cell weight/mL, 30 min). It was found that the efficiency to remove Hg2+ by the dead cells of all strains was significantly higher than for live cells; and the most effective strain was SSS2-1. The highest Hg2+ removal under the optimum conditions using a 4 mg/L initial HgCl 2 concentration by live and dead cells grown with both incubation conditions were approximately 87% and 95%, respectively. Under both incubation conditions, Hg2+ biosoption by strain SSS2-1 fitted the Freundlich model for live cells and the Langmuir model for dead cells. The kinetics of Hg2+ biosorption by both cell types of strain SSS2-1 suggests the process as a pseudo-second order kinetic model. This study demonstrates that biomass of the strain SSS2-1 has great potential for its biosorption to remove Hg2+ from contaminated areas like shrimp ponds. This study aimed to investigate the potential of Rhodopseudomonas palustris C1 and Rubrivivax benzoatilyticus C31 to ameliorate As toxicity and to reduce As uptake in rice. Strain C1 was superior to strain C31 for siderophore production. The mixed culture (1: 1) was most effective in reducing the toxicity of As species [As(III) and/or As(V), each 30 mg/l] by yielding maximal germination index that related to α- and β-amylase activities in two Thai rice cultivars (HomNil: HN and PathumThani 1: PT). Arsenic toxicity to the seed germination followed the order: mixed As species > As(III) > As(V); and the toxicity was reduced in inoculated sets, particularly with a mixed culture. The mixed culture significantly enhanced rice growth under As stress in both rice cultivars as indicated by an increase in the production of chlorophyll a and b, and also supporting the non-enzymatic (carotenoids, lipid oxidation, and nitric oxide) and enzymatic (superoxide dismutase, ascorbate peroxidase, catalase, and glutathione reductase) activities. These were concomitant with productions of 5-aminolevulinic acid, indole-3-acetic acid, exopolymeric substances, and siderophores which significantly reduced As accumulation in treated rice. It can be concluded that the mixed culture has great potential to ameliorate rice from As toxicity by preventing As species entry into rice for enhancing rice growth and also for reducing As accumulation to produce safe rice from rice grown in contaminated paddy fields. Background: The extract Lycogen™ from the phototrophic bacterium Rhodobacter sphaeroides (WL-APD911) has attracted significant attention because of its promising potential as a bioactive mixture, attributed in part to its anti-inflammatory properties and anti-oxidative activity. Objective: This study aims to investigate the components of Lycogen™ and its anti-inflammatory properties and anti-oxidative activity. Design and results: The mutant strain R. sphaeroides (WL-APD911) whose carotenoid 1,2-hydratase gene has been altered by chemical mutagenesis was used for the production of a new carotenoid. The strain was grown at 30°C on Luria–Bertani (LB) agar plates. After a 4-day culture period, the mutant strain displayed a 3.5-fold increase in carotenoid content, relative to the wild type. In the DPPH test, Lycogen™ showed more potent anti-oxidative activity than lycopene from the wild-type strain. Primary skin irritation test with hamsters showed no irritation response in hamster skins after 30 days of treatment with 0.2% Lycogen™. Chemical investigations of Lycogen™ using nuclear magnetic resonance (NMR) 1H, 13C, and COSY/DQCOSY spectra have identified spheroidenone and methoxyneurosporene. Quantitative analysis of these identified compounds based on spectral intensities indicates that spheroidenone and methoxyneurosporene are major components (approximately 1:1); very small quantities of other derivatives are also present in the sample. Conclusions: In this study, we identified the major carotenoid compounds contained in Lycogen™, including spheroidenone and methoxyneurosporene by high-resolution NMR spectroscopy analysis. The carotenoid content of this mutant strain of R. sphaeroides was 3.5-fold higher than that in normal strain. Furthermore, Lycogen™ from the mutant strain is more potent than lycopene from the wild-type strain and does not cause irritation in hamster skins. These findings suggest that this mutant strain has the potential to be used as an enriched carotenoid source.

Bitter-1 is a shallow hypersaline soda lake in Kulunda Steppe (Altai region, Russia). During a study period between 2005 and 2016, the salinity in the littoral area of the lake fluctuated within the range from 85 to 400 g/L (in July of each year). Light-dependent nitrogen fixation occurred in this lake up to the salt-saturating conditions. The rates increased with a decrease in salinity, both under environmental conditions and in laboratory simulations. The salinities below 100 g/L were favorable for light-dependent nitrogen fixation, while the process was dramatically inhibited above 200 g/L salts. The analysis of nifH genes in environmental samples and in enrichment cultures of diazotrophic phototrophs suggested that anaerobic fermenting and sulfate-reducing bacteria could participate in the dark nitrogen fixation process up to soda-saturating conditions. However, we cannot exclude the possibility that haloalkaliphilic nonheterocystous cyanobacteria (Euhalothece sp. and Geitlerinema sp.) and anoxygenic purple sulfur bacteria (Ectothiorhodospira sp.) might also play a role in the process at light conditions. The heterocystous cyanobacterium Nodularia sp. develops at low salinity (below 80 g/L) that is not characteristic for Bitter-1 Lake and thus does not make a significant contribution to the nitrogen fixation in this lake. This study was aimed to screen 22 purple nonsulfur bacteria (PNSB) isolates with the ability to secrete proteolytic enzymes and antivibrio compounds, including affecting factors on proteolytic enzyme production of the selected PNSB. Use of overlay diffusion method under aerobic dark conditions found that only 12 PNSB isolates (54.55%) were able to inhibit shrimp pathogenic Vibrio spp.; while 18 isolates (81.82%) could liquefy gelatin under conditions of aerobic dark and microaerobic light. Twenty-time freeze-dried culture supernatant concentrates collected from 12 PNSB grown under the microaerobic light conditions, with the use of agar well diffusion found that only strain PS342 was capable to inhibit all 6 tested shrimp pathogenic vibrios. Among 18 proteolytic PNSB, only 5 strains showed high activity of gelatin liquefaction and this included strain PS342. The strain PS342 was identified using 16 S rRNA gene as Rhodovulum sulfidophilum . Glutamate-malate (GM) medium supplemented with 1.5% NaCl was the most suitable medium by giving µ max as 0.336 h −1 . Use of central composite design, the maximal proteolytic activity was an average of 14.52 unit/ml in the suitable medium containing 1% gelatin under aerobic-dark conditions with the optimal speed at 150 rpm. The results of the verify test showed that optimum conditions (pH 7.90, 1.30% NaCl and 29.50 °C) for the proteolytic activity (15.40 unit/ml) were very close to real conditions for shrimp cultivation. Purple phototrophic bacteria (PPB) have been recently proposed as a key potential mechanism for accumulative biotechnologies for wastewater treatment with total nutrient recovery, low greenhouse gas emissions, and a neutral to positive energy balance. Purple phototrophic bacteria have a complex metabolism which can be regulated for process control and optimization. Since microbial processes governing PPB metabolism differ from traditional processes used for wastewater treatment (e.g., aerobic and anaerobic functional groups in ASM and ADM1), a model basis has to be developed to be used as a framework for further detailed modelling under specific situations. This work presents a mixed population phototrophic model for domestic wastewater treatment in anaerobic conditions. The model includes photoheterotrophy, which is divided into acetate consumption and other organics consumption, chemoheterotrophy (including simplified fermentation and anaerobic oxidation) and photoautotrophy (using hydrogen as an electron donor), as microbial processes, as well as hydrolysis and biomass decay as biochemical processes, and is single-biomass based. The main processes have been evaluated through targeted batch experiments, and the key kinetic and stoichiometric parameters have been determined. Bacteria that utilize sunlight to supplement metabolic activity are now being described in a range of ecosystems. While it is likely that phototrophy provides an important competitive advantage, the contribution that these microorganisms make to the bioenergetics of polar marine ecosystems is unknown. In this minireview, we discuss recent advances in our understanding of phototrophic bacteria and highlight the need for future research.

A potential increase in the activity of Cyanobacteria and other phototrophic groups, e.g., Rhodobacterales, was observed and associated with UVR, suggesting the presence of photo-activated repair mechanisms to resist high levels of solar radiation. In addition, the percentage contribution of cyanobacterial sequences in the afternoon was similar to those recorded in the morning. The shifts in the contribution by Cyanobacteria also influenced the rate of change in nitrate, nitrite, and phosphate, highlighted by a high level of nitrate accumulation during hours of high radiation and temperature associated with nitrifying bacteria activity. We did not detect ammonia or nitrite oxidizing bacteria in situ, but both functional groups (Nitrosomona and Nitrospira) appeared mainly in pyrolibraries generated from dark incubations. In total, our results reveal that both the structure and the diversity of the active bacteria community was extremely dynamic through the day, and showed marked shifts in composition that influenced nutrient recycling, highlighting how abiotic variation affects potential ecosystem functioning. The present and commonly used batch culture system ( BCS) as adopted by many small-scale tropical hatcheries is beset by poor rotifer production and sudden crashes. This study aimed to produce nutritive rotifers and evaluate their performance based on the BCS by using phototrophic bacteria ( PB) that can be easily and cheaply cultured from palm oil mill effluent ( POME), an agro-industrial byproduct usually discarded as waste. Brachionus rotundiformis given a sole diet of POME-grown PB ( Rhodovulum sulfidophilum) grew as well as on the commercially produced microalgae, Nannochloropsis. Production, growth rate and fecundity of rotifers fed condensed bacterial cells ( bPB) and culture broth of bacteria grown in POME ( cPB) were evaluated. The best performance in terms of the stated parameters was obtained for rotifers fed 200 mL of cPB in 3 L of culture water; this media sustained a mean rotifer density of 600-900 individuals mL−1 after 3-6 days of culture. The biochemical composition of rotifers fed PB was comparable to those fed microalgae, except that the former contained more polyunsaturated fatty acids. [ABSTRACT FROM AUTHOR]

Microbial-mat communities in the effluent channels of Octopus and Mushroom Springs within the Lower Geyser Basin at Yellowstone National Park have been studied for nearly 50 years. The emphasis has mostly focused on the chlorophototrophic bacterial organisms of the phyla Cyanobacteria and Chloroflexi. In contrast, the diversity and metabolic functions of the heterotrophic community in the microoxic/anoxic region of the mat are not well understood. In this study we analyzed the orange-colored undermat of the microbial community of Mushroom Spring using metagenomic and rRNA-amplicon (iTag) analyses. Our analyses disclosed a highly diverse community exhibiting a high degree of unevenness, strongly dominated by a single taxon, the filamentous anoxygenic phototroph, Roseiflexus spp. The second most abundant organisms belonged to the Thermotogae, which have been hypothesized to be a major source of H2 from fermentation that could enable photomixotrophic metabolism by Chloroflexus and Roseiflexus spp. Other abundant organisms include two members of the Armatimonadetes (OP10); Thermocrinis sp.; and phototrophic and heterotrophic members of the Chloroflexi. Further, an Atribacteria (OP9/JS1) member; a sulfate-reducing Thermodesulfovibrio sp.; a Planctomycetes member; a member of the EM3 group tentatively affiliated with the Thermotogae, as well as a putative member of the Arminicenantes (OP8) represented ≥1% of the reads. Archaea were not abundant in the iTag analysis, and no metagenomic bin representing an archaeon was identified. A high microdiversity of 16S rRNA gene sequences was identified for the dominant taxon, Roseiflexus spp. Previous studies demonstrated that highly similar Synechococcus variants in the upper layer of the mats represent ecological species populations with specific ecological adaptations. Se estudiaron las comunidades estratificadas de microorganismos fotosintéticos que se encuentran en el Salar de Llamará ubicado en el desierto de Atacama, norte de Chile, mediante métodos microscópicos y espectrofotométricos. El espesor de la zona fótica de los tapetes descritos varió entre 8 y 30 mm lo cual podría atribuirse a la granulometría y la composición mineralógica de los sedimentos. Se diferencian tres tipos de tapetes. El primero con una única capa pigmentada de color verde; el segundo con capas de colores verde y naranja y un tercero en el que se observa, además de las capas verde y naranja, una de color púrpura. En uno de los sitios muestreados no se encontraron capas pigmentadas. Debajo de la zona pigmentada el sedimento es de color blanco, excepto en uno de los sectores donde se observó una coloración negra atribuible a sulfuro de hierro. Los microorganismos predominantes de la capa naranja fueron diatomeas y cianobacterias unicelulares principalmente de los grupos Cyanothece y Synechococcus. Las cianobacterias filamentosas Microccoleus sp. y Oscillatoria sp. fueron las más abundantes en la capa verde. No se observaron diatomeas en los sitios estudiados donde la salinidad del agua intersticial osciló entre 12 y 33 %. En la capa verde de estos sitios predominaron las cianobacterias cocoides, de los grupos Synechococcus, Cyanothece y Gloeocapsa y del género Gloeobacter, sobre las cianobacterias filamentosas. La capa púrpura estuvo compuesta principalmente por bacterias fototróficas anoxigénicas similares a células de los géneros Chromatium y Thiocapsa. Los espectros de absorción revelaron que la clorofila a es el pigmento más abundante en la mayoría de las muestras analizadas. Los valores integrados de clorofila a y bacterioclorofila a alcanzaron 230 y 144 mg m-2 en el espesor de la zona pigmentada, respectivamente. También se detectaron abundantes microorganismos no fotosintéticos en los tapetes incluyendo cocos y bacilos no identificados. En todos los tapetes muestreados en el Salar se encontraron bacterias reductoras de sulfato.Stratified photosynthetic microbial mats are described from the Salar de Llamará, a salt flat basin located in the Atacama desert of northern Chile. Microscopic and spectrophotometric techniques were used. The thickness of the photic zone of these communities spans 8 to 30 mm. This is probably due to the grain size and mineralogical composition of associated sediments. Three different types of mats were recognized. A first one was characterized by a green pigmented layer; a second with orange and green coloured layers, and the third with orange and green layers and an additional purple layer. At one sampling site, no pigmented layers were present. Sediments underlying the mats were white, but in one site, black coloured sediments were observed; this dark colour is probably the result of iron sulphide precipitation. Predominant microorganisms in the orange pigmented layers were diatoms and unicellular cyanobacteria, mainly from the Cyanothece and Synechococcus groups. Filamentous cyanobacteria Microleus sp. and Oscillatoria sp. were the most abundant in the green layer. When interstitial brines reached salinities between 12 and 33 %, no diatoms were observed, and the coccoidal cyanobacteria from the Synechococcus, Cyanothece and Gloeocapsa groups and genus Gloeobacter predominated over filamentous Cyanobacteria in the green layer. The purple layer was built primarily of anoxygenic phototrophic bacteria similar to cells of the genera Chromatium and Thiocapsa. Absorption spectra revealed that chlorophyll a is the most abundant pigment in most of analyzed samples. Integrated values of chlorophyll a and bacteriochlorophyll a reached values of up to 230 and 144 mg m-2 along all of the pigmented zone, respectively. Abundant non-photosynthetic microorganisms were found in the mats, including unidentified cocci and bacilli. Sulphate reducing bacteria were present in all the sampled mats.

Previously, we presented data that indicated microbial sulfide oxidation would out-compete strictly chemical, abiotic sulfide oxidation reactions under nearly all conditions relevant to extant ecosystems (Luther et al., 2011). In particular, we showed how anaerobic microbial sulfide oxidation rates were several orders of magnitude higher than even metal catalyzed aerobic sulfide oxidation processes. The fact that biotic anaerobic sulfide oxidation is kinetically superior to abiotic reactions implies that nearly all anaerobic and sulfidic environments should host microbial populations that oxidize sulfide at appreciable rates. This was likely an important biogeochemical process during long stretches of euxinia in the oceans suggested by the geologic record. In particular, phototrophic sulfide oxidation allows the utilization of carbon dioxide as the electron acceptor suggesting that this process should be particularly widespread rather than relying on the presence of other chemical oxidants. Using the Chesapeake Bay as an example, we argue that phototrophic sulfide oxidation may be more important in many environments than is currently appreciated. Finally, we present methodological considerations to assist other groups that wish to study this process.

Six phototrophic microbial mat communities from different geothermal springs (YNP) were studied using metagenome sequencing and geochemical analyses. The primary goals of this work were to determine differences in community composition of high-temperature phototrophic mats distributed across the Yellowstone geothermal ecosystem, and to identify metabolic attributes of predominant organisms present in these communities that may correlate with environmental attributes important in niche differentiation. Random shotgun metagenome sequences from six phototrophic communities (average~ 53 Mbp/site) were subjected to multiple taxonomic, phylogenetic and functional analyses. All methods, including G+C content distribution, MEGAN analyses and oligonucleotide frequency-based clustering, provided strong support for the dominant community members present in each site. Cyanobacteria were only observed in non-sulfidic sites; de novo assemblies were obtained for Synechococcus-like populations at Chocolate Pots (CP_7) and Fischerella-like populations at White Creek (WC_6). Chloroflexi-like sequences (esp. Roseiflexus and/or Chloroflexus spp.) were observed in all six samples and contained genes involved in bacteriochlorophyll biosynthesis and the 3-hydroxypropionate carbon fixation pathway. Other major sequence assemblies were obtained for a Chlorobiales population from CP_7 (proposed family Thermochlorobacteriaceae), and an anoxygenic, sulfur-oxidizing Thermochromatium-like (Gamma-proteobacteria) population from Bath Lake Vista Annex (BLVA_20). Additional sequence coverage is necessary to establish more complete assemblies of other novel bacteria in these sites (e.g., Bacteroidetes and Firmicutes); however, current assemblies suggested that several of these organisms play important roles in heterotrophic and fermentative metabolisms. During the past decades, tremendous advances have been made in the possibilities to study the diversity of microbial communities in the environment. The development of methods to study these communities on the basis of 16S rRNA gene sequences analysis was a first step into the molecular analysis of environmental communities and the study of biodiversity in natural habitats. A new dimension in this field was reached with the introduction of functional genes of ecological importance and the establishment of genetic tools to study the diversity of functional microbial groups and their responses to environmental factors. Functional gene approaches are excellent tools to study the diversity of a particular function and to demonstrate changes in the composition of prokaryote communities contributing to this function. The phylogeny of many functional genes largely correlates with that of the 16S rRNA gene, and microbial species may be identified on the basis of functional gene sequences. Functional genes are perfectly suited to link culture-based microbiological work with environmental molecular genetic studies. In this review, the development of functional gene studies in environmental microbiology is highlighted with examples of genes relevant for important ecophysiological functions. Examples are presented for bacterial photosynthesis and two types of anoxygenic phototrophic bacteria, with genes of the Fenna-Matthews-Olson-protein (fmoA) as target for the green sulfur bacteria and of two reaction center proteins (pufLM) for the phototrophic purple bacteria, with genes of adenosine-5′phosphosulfate (APS) reductase (aprA), sulfate thioesterase (soxB) and dissimilatory sulfite reductase (dsrAB) for sulfur oxidizing and sulfate reducing bacteria, with genes of ammonia monooxygenase (amoA) for nitrifying/ammonia-oxidizing bacteria, with genes of particulate nitrate reductase and nitrite reductases (narH/G, nirS, nirK) for denitrifying bacteria and with genes of methane monooxygenase (pmoA) for methane oxidizing bacteria.

The spatial distribution and composition of anoxygenic phototrophic bacteria in the enriched bacterial communities from different depths of karst lakes Kirkilai and Ramunelis was studied using spectrophotometric analysis, as well as microbiological and molecular methods. In Lake Kirkilai, the highest bacterial abundance was measured in the metalimnion and near the bottom (up to 10.7 × 10 cell/mL); in Lake Ramunelis it was in the anoxic hypolimnion (up to 22.4 × 10 cell/mL). Increased water mineralization (0.5-1.2 g/L) with the domination of SO and Ca ions created favorable conditions for the development of sulfate-reducing bacteria; hydrogen sulfide produced as a result of their life activity facilitated the development of sulfur-oxidizing bacteria. The pigment analysis of phototrophic green and purple sulfur bacteria showed the domination of green sulfur bacteria in the enrichment culture. The results of phylogenetic analysis showed that Chlorobium limicola dominated in the enrichment culture for the green sulfur bacteria, whereas purple nonsulfur bacteria of the genus Rhodopseudomonas dominated in the enrichment culture for the purple sulfur bacteria. Survival of marble goby larvae fed either Rhodovulum sulfidophilum, a phototrophic bacterium cultured from palm oil mill effluent ( pPB), or microalgae ( Nannochloropsis sp.) was evaluated at two salinities. Larvae directly fed pPB had survival of 0-29% at 5 g L−1 salinity and 0-19% at 10 g L−1 salinity, whereas larvae directly fed microalgae suffered complete mortality after 20 days of culture at both salinities. However, larvae indirectly fed pPB or microalgae, i.e. via rotifers (Days 1-30) and Artemia nauplii (Days 21-30) cultured solely from pPB or microalgae, showed improved survival of 35-55% or 44-49% at 5 g L−1 salinity respectively. In all experiments, fish larvae reared at 5 g L−1 salinity showed significantly higher ( P < 0.01) mean survival than those reared at 10 g L−1 salinity. The survival of larvae fed the bacterial-based diet was higher compared with microalgal diet used in previous studies. The pPB had higher total polyunsaturated fatty acids and docosahexaenoic acid ( DHA) than the microalgae, which had very high eicosapentaenoic acid ( EPA). Larvae with very high ratios of DHA/ EPA (>11) or/and ARA (arachidonic acid)/ EPA (>5), attributable to their given diet, however suffered the highest mortality. [ABSTRACT FROM AUTHOR]

Time-resolved electron paramagnetic resonance was used to study the properties of carotenoid triplet states populated in LH2 light-harvesting complexes of phototrophic bacteria Allochromatium minutissimum, Rhodopseudomonas palustris, and in carotenoid films free of bacteriochlorophyll. The study was performed on purified LH2 preparations not contaminated by reaction centers, and under selective pigment excitation. The obtained results enable a conclusion that the carotenoid triplet states, both in LH2 complexes and films, are populated in the process of homofission of singlet excitation into two triplets, which involves only carotenoid molecules. It is observed that the fission process in magnetic field leads to predominant population of the T spin sublevel of the triplet. One molecular spin sublevel of the triplet is demonstrated to possess an increased probability of intersystem crossing to the ground state, independent of the carotenoid configuration. Pigment composition of the LH2 protein heterodimers is discussed, and a conclusion of the possible presence of two interacting carotenoid molecules is made. [ABSTRACT FROM AUTHOR]

Bacteria that utilize sunlight to supplement metabolic activity are now being described in a range of ecosystems. While it is likely that phototrophy provides an important competitive advantage, the contribution that these microorganisms make to the bioenergetics of polar marine ecosystems is unknown.
