Parasitoids of different species frequently develop in the same host, a phenomenon referred to as "multiparasitism". Although the outcomes of multiparasitism have been well-documented in the literature, the underlying mechanisms, particularly the substances injected by a female parasitoid along with her egg(s) into a host during parasitism, remain relatively unexplored. Previous work on parasitoids associated with the cabbage white butterfly, Pieris brassicae (Lepidoptera: Pieridae) has shown that the larva of the solitary parasitoid Hyposoter ebeninus (Hymenoptera: Ichneumonidae) has a higher survival but a longer development time when competing with the gregarious parasitoid Cotesia glomerata (Hymenoptera: Braconidae). In this study, we hypothesize that the maternal factors injected by C. glomerata are responsible for the effect on the performance of H. ebeninus larvae. This hypothesis was tested using P. brassicae caterpillars first parasitized with H. ebeninus and then injected with C. glomerata maternal factors, or parasitized by both parasitoids. Our results suggest that C. glomerata maternal factors are at least partially responsible for the reduction in H. ebeninus developmental mortality (likely through effects on the immune response of the host caterpillar), but does not seem to affect its development time. We discuss these results and the current knowledge of maternal-factor-mediated parasitoid interactions.

The venom gland is a ubiquitous organ in Hymenoptera. In insect parasitoids, the venom gland has been shown to have multiple functions including regulation of host immune response, host paralysis, host castration and developmental alteration. However, the role played by the venom gland has been mainly studied in parasitoids developing in larval or pupal hosts while little is known for parasitoids developing in insect eggs. We conducted the first extensive characterization of the venom of the endoparasitoid Ooencyrtus telenomicida (Vassiliev), a species that develops in eggs of the stink bug Nezara viridula (L.). In particular we investigated the structure of the venom apparatus, its functional role and conducted a transcriptomic analysis of the venom gland. We found that injection of O. telenomicida venom induces: 1) a melanized-like process in N. viridula host eggs (host-parasitoid interaction), 2) impairment of the larval development of the competitor Trissolcus basalis (Wollaston) (parasitoid-parasitoid interaction). The O. telenomicida venom gland transcriptome reveals a majority of digestive enzymes (peptidases and glycosylases) and oxidoreductases (laccases) among the most expressed genes. The former enzymes are likely to be involved in degradation of the host resources for the specific benefit of the O. telenomicida offspring. In turn, alteration of host resources caused by these enzymes may negatively affect the larval development of the competitor T. basalis . We hypothesize that the melanization process induced by venom injection could be related to the presence of laccases, which are multicopper oxidases that belong to the phenoloxidases group. This work contributed to a better understanding of the venom in insect parasitoids and allowed to identify candidate genes whose functional role can be investigated in future studies. In many cooperatively breeding animals, subordinate group members have lower reproductive capacity than dominant group members. Theory suggests subordinates may downregulate their reproductive capacity because dominants punish subordinates who maintain high fertility. However, there is little direct experimental evidence that dominants cause physiological suppression in subordinates. Here, we experimentally test how social interactions influence subordinate reproductive hormones in Polistes dominula paper wasps. Polistes dominula queens commonly found nests in cooperative groups where the dominant queen is more fertile than the subordinate queen. In this study, we randomly assigned wasps to cooperative groups, assessed dominance behaviour during group formation, then measured levels of juvenile hormone (JH), a hormone that mediates Polistes fertility. Within three hours, lowest ranking subordinates had less JH than dominants or solitary controls, indicating that group formation caused rapid JH reduction in low-ranking subordinates. In a second experiment, we measured the behavioural consequences of experimentally increasing subordinate JH. Subordinates with high JH-titres received significantly more aggression than control subordinates or subordinates from groups where the dominant's JH was increased. These results suggest that dominants aggressively punished subordinates who attempted to maintain high fertility. Low-ranked subordinates may rapidly downregulate reproductive capacity to reduce costly social interactions with dominants. Many animals adapt to rapidly changing social environments by using social information to modulate their hormone titres. Modulation of hormone titres, as proposed by the challenge hypothesis, provides a mechanism by which individuals can match their behaviour to their current social environment and thereby avoid costs associated with prolonged high hormone titres. Thus far, most work on social responsiveness of hormone titres has focused on androgens in vertebrates. However, there is mounting evidence that insect hormones, especially juvenile hormone, may respond to social stimuli in ways that parallel androgens in vertebrates. This review will integrate work on social modulation of hormone titres in vertebrates and insects. First we review how the hormone-mediated trade-off between fecundity and life span may be a key selective force favouring socially responsive hormone titres in both systems. Then we review theoretical and empirical work in vertebrates and insects that address how factors such as social instability, dominance rank and mating system influence hormone titre responsiveness. These studies illustrate that (1) juvenile hormone (JH) is responsive to social stimuli in a range of insect taxa and (2) JH responses match key predictions of the challenge hypothesis. We conclude that there are strong similarities in endocrine responsiveness across vertebrate and insect taxa. The challenge hypothesis provides a useful conceptual framework for hypothesis-driven research in insect endocrinology. In addition, exploring areas of convergence and divergence across vertebrates and insects may help clarify how selection has shaped patterns of endocrine responsiveness. [Copyright &y& Elsevier]

Ongoing studies by our group showed that the outcome of the intrinsic competition between two solitary egg parasitoids, Trissolcus basalis (Wollaston) (Hymenoptera: Scelionidae) and Ooencyrtus telenomicida (Vassiliev) (Hymenoptera: Encyrtidae), is dominated by O. telenomicida. In this article we investigated the role played by the ovipositing O. telenomicida female in the suppression of a T. basalis competitor. Laboratory experiments were conducted by allowing an O. telenomicida female to puncture the eggs of Nezara viridula (L.) (Heteroptera: Pentatomidae) with her ovipositor (= no oviposition) or to parasitize them. The results show that O. telenomicida relies on some physiological mechanisms to mediate its interspecific intrinsic competition with T. basalis. In fact, the emergence of T. basalis was strongly reduced in host eggs that were parasitized either before or after being punctured by O. telenomicida at fixed time intervals (5, 15, 30, or 45 h). The low percentage of emergence of T. basalis (ranging from approximately 4-20%) was a consequence of the delay and growth rate reduction of larval development. Furthermore, the percentage of eclosion of N. viridula nymphs was negatively affected by the O. telenomicida female's punctures (96% from healthy host eggs, 4% from punctured host eggs). Host eggs punctured or oviposited in by O. telenomicida showed alterations in the ooplasm including some melanized-like areas near the hole made with the ovipositor; such alterations indicate that the adult parasitoid releases substances that affect the host eggs survival. These results suggest that the O. telenomicida female influences both the physiological interspecific parasitoid-parasitoid interaction, as well as the host-parasitoid interaction, providing, for the first time in egg parasitoids, evidence that physiological suppression of some competitive egg parasitoids is mediated by the ovipositing female. Abstract: In many vertebrate societies, subordinate females exhibit down-regulated reproductive physiologies relative to those of dominants, a condition commonly termed physiological suppression. Research into the causes of physiological suppression has focused principally on the role of the subordinate''s social environment (typically the presence of the dominant female and/or an absence of unrelated males within the group), while few studies have considered the additional role that the physical environment may play. Here we present new evidence from wild Damaraland mole-rats, Fukomys damarensis, revealing that physiological suppression among subordinate females eases markedly during the annual rains (a time when ecological constraints on dispersal are relaxed), despite the continued presence of the dominant female and in groups that contain no new immigrant males. Subordinate females showed substantially higher pituitary sensitivities to GnRH challenge during the wet period than the dry, a contrast that cannot be attributed to between-female differences (as it holds for paired within-female comparisons), associated changes in body mass (as our analyses control for this), or concomitant reductions in physiological stress (as their urinary cortisol concentrations were actually higher in the wet period). We suggest that our findings reflect selection for the maintenance of reproductive readiness among subordinate females during high rainfall periods, given the increased likelihood of encountering dispersal and/or mating opportunities with extra-group males when ecological constraints on dispersal are relaxed. These findings reveal new complexity in the processes that regulate physiological suppression, suggesting a key role in some species for changes in the physical as well as social environment. Abstract: Tephritid fruit fly parasitoid guilds are dominated by solitary koinobiont species that attack different host stages, but most emerge as adults from host puparia. Previous studies suggest intrinsic competitive superiority by the egg-attacking parasitoid Fopius arisanus (Sonan) against all larval-attacking parasitoids in Hawaii. In this study, we tested the early-acting competitive superiority prediction in relation to the co-evolutionary history of competition between an egg–larval parasitoid (Fopius ceratitivorus Wharton), and each of three larval parasitoids [Psyttalia concolor (Szépligeti), Diachasmimorpha kraussii (Fullaway), and Diachasmimorpha longicaudata (Ashmead)]. F. ceratitivorus and P. concolor share a common origin (eastern Africa), while D. kraussii is an Australian species, and D. longicaudata is from Southeast Asia. The outcomes of intrinsic competition between the egg-attacking parasitoid and each of the three larval-attacking parasitoids within their common host, the Mediterranean fruit fly Ceratitis capitata (Wiedemann) were compared. F. ceratitivorus invariably eliminated the co-evolved P. concolor through physiological suppression of the later-attacking parasitoid’s egg development, providing evidence that supports the early-acting-superiority hypothesis. However, F. ceratitivorus was unable to suppress development of the two non co-evolved larval parasitoids. Instead, the larvae of both later-acting parasitoid species physically killed F. ceratitivorus larvae inside the host. The results suggest that co-evolutionary history influences competitive superiority. The evolution of inter-specific competition and its implications for biological control are discussed. Abstract: Precocious larvae, clonally produced together with reproductive siblings in the polyembryonic parasitoid Copidosoma floridanum, are known to physically attack competitors in multiparasitized hosts. In this study, we show that physiological suppression by C. floridanum, as well as precocious larval activity, causes death of the larval parasitoid Glyptapanteles pallipes. Approximately 70% of the hosts multiparasitized by C. floridanum and G. pallipes produced C. floridanum offspring, irrespective of the interval of multiparasitism. G. pallipes eggs or larvae died even in multiparasitized hosts that did not contain precocious larvae of C. floridanum. An injection of C. floridanum-parasitized or multiparasitized-host hemolymph into G. pallipes singly-parasitized hosts paralyzed almost all G. pallipes larvae within 70h. In vitro analysis showed that the hemolymph factor toxic to G. pallipes eggs and larvae was present in C. floridanum-parasitized hosts through their larval stages. Heating or proteinase treatment reduced its toxicity, suggesting that the factor is a protein. [Copyright &y& Elsevier]

The Escherichia coli aspartase gene aspA has been expressed in the fungus Aspergillus nidulans using the powerful constitutive gpdA promoter and trpC terminator, both from A. nidulans. Multiple, but not single, copies of aspA overcome nutritional deficiencies resulting from the loss of catabolic NAD-linked glutamate dehydrogenase. They also circumvent certain nutritional deficiencies resulting from loss of the positive-acting regulatory gene product mediating nitrogen metabolite repression. Both of these cases of physiological suppression involve the aspartase-catalyzed catabolism of aspartate to ammonium plus fumarate. No physiological evidence for the opposite reaction leading to aspartate synthesis was obtained as multiple copies of aspA did not affect the phenotype resulting from the loss of anabolic NADP-linked glutamate dehydrogenase. The use of vectors containing aspA and recipients lacking NAD-linked glutamate dehydrogenase is an efficient means of selecting multicopy transformants in A. nidulans and also offers the possibility to select strains having increased aspartase levels from original transformants. This study evaluated the influence of the phenolic compounds carvacrol (CAR) and thymol (THY) on some physiological characteristics and on the modulation of the secretion of some staphylococcal virulence factors, that is, coagulase and enterotoxin. This study also investigated possible mechanisms for the establishment of the anti-staphylococcal activity of these compounds. Sublethal concentrations (0.3 and 0.15 μL/mL) of CAR and THY inhibited the activity of the enzymes coagulase and lipase and led to a decrease in salt tolerance. At the tested sublethal concentrations, both CAR and THY led to a total suppression of enterotoxin production. The loss of a 260-nm-absorbing material and an efflux of potassium ions occurred immediately after the addition of CAR and THY at 0.6 and 1.2 μL/mL and increased up to 120 min of exposure. Electron microscopy of cells exposed to CAR and THY (0.6 μL/mL) revealed that individual cells appeared to be deformed, with projections of cellular material. The observations of leakage of cellular material and an altered cell surface suggest that gross damage to a cell's cytoplasmic membrane, which results in a disruption in protein secretion, could be responsible for the anti-staphylococcal properties of CAR and THY.
