1. A Current Overview of the Papaya meleira virus, an Unusual Plant Virus

Abstract: Papaya meleira virus (PMeV) is the causal agent of papaya sticky disease, which is characterized by a spontaneous exudation of fluid and aqueous latex from the papaya fruit and leaves. The latex oxidizes after atmospheric exposure, resulting in a sticky feature on the fruit from which the name of the disease originates. PMeV is an isometric virus particle with a double-stranded RNA (dsRNA) genome of approximately 12 Kb. Unusual for a plant virus, PMeV particles are localized on and linked to the polymers present in the latex. The ability of the PMeV to inhabit such a hostile environment demonstrates an intriguing interaction of the virus with the papaya. A hypersensitivity response is triggered against PMeV infection, and there is a reduction in the proteolytic activity of papaya latex during sticky disease. In papaya leaf tissues, stress responsive proteins, mostly calreticulin and proteasome-related proteins, are up regulated and proteins related to metabolism are down-regulated. Additionally, PMeV modifies the transcription of several miRNAs involved in the modulation of genes related to the ubiquitin-proteasome system. Until now, no PMeV resistant papaya genotype has been identified and roguing is the only viral control strategy available. However, a single inoculation of papaya plants with PMeV dsRNA delayed the progress of viral infection.

Abstract: We report the complete genome sequencing of a novel member of the genus Vitivirus in the family Betaflexiviridae (subfamily Trivirinae) infecting pineapple. The full genome of this virus was obtained from total RNAs extracted from pineapple leaf samples collected in Reunion Island, using a combination of high-throughput sequencing technologies. It is 6,757 nt long, excluding the poly(A) tail, and shares all the hallmarks of vitiviruses. Phylogenetic analyses performed on the replication-associated protein and capsid protein gene sequences unambiguously place this new virus, for which we propose the name “pineapple vitivirus A”, in the Vitivirus genus.

Abstract: International audience; Cucumber vein yellowing virus (CVYV) is an emerging virus on cucurbits in the Mediterranean Basin, against which few resistance sources are available, particularly in melon. The melon accession PI 164323 displays complete resistance to isolate CVYV-Esp, and accession HSD 2458 presents a tolerance, i.e. very mild symptoms in spite of virus accumulation in inoculated plants. The resistance is controlled by a dominant allele Cvy-1 1 , while the tolerance is controlled by a recessive allele cvy-2, independent from Cvy-1 1 . Before introducing the resistance or tolerance in commercial cultivars through a long breeding process, it is important to estimate their specificity and durability. Upon inoculation with eight molecularly diverse CVYV isolates, the resistance was found to be isolate-specific since many CVYV isolates induced necrosis on PI 164323, whereas the tolerance presented a broader range. A resistance-breaking isolate inducing severe mosaic on PI 164323 was obtained. This isolate differed from the parental strain by a single amino acid change in the VPg coding region. An infectious CVYV cDNA clone was obtained, and the effect of the mutation in the VPg cistron on resistance to PI 164323 was confirmed by reverse genetics. This represents the first determinant for resistance-breaking in an ipomovirus. Our results indicate that the use of the Cvy-1 1 allele alone will not provide durable resistance to CVYV and that, if used in the field, it should be combined with other control methods such as cultural practices and pyramiding of resistance genes to achieve long-lasting resistance against CVYV.

Abstract: International audience; In this study, we looked for genetic factors in the pepper (Capsicum annuum) germplasm that control the number of potato virus Y (PVY) particles entering the plant (i.e. effective population size at inoculation) and the PVY accumulation at the systemic level (i.e. census population size). Using genotyping-by-sequencing (GBS) in a core collection of 256 pepper accessions, we obtained 10 307 single nucleotide polymorphisms (SNPs) covering the whole genome. Genome-wide association studies (GWAS) detected seven SNPs significantly associated with the virus population size at inoculation and/or systemic level on chromosomes 4, 6, 9 and 12. Two SNPs on chromosome 4 associated with both PVY population sizes map closely to the major resistance gene pvr2 encoding the eukaryotic initiation factor 4E. No obvious candidates for resistance were identified in the confidence intervals for the other chromosomes. SNPs detected on chromosomes 6 and 12 colocalized with resistance quantitative trait loci (QTLs) previously identified with a biparental population. These results show the efficiency of GBS and GWAS in C. annuum, indicate highly consistent results between GWAS and classical QTL mapping, and suggest that resistance QTLs identified with a biparental population are representative of a much larger collection of pepper accessions. Moreover, the resistance alleles at these different loci were more frequently combined than expected by chance in the core collection, indicating widespread pyramiding of resistance QTLs and widespread combination of resistance QTLs and major effect genes. Such pyramiding may increase resistance efficiency and/or durability.

Abstract: Background as of March 2019As a result of the health monitoring carried out by the members of ANSES's Expert Committee and our ANSES employees, we became aware of an alert concerning an emerging tomato virus: tomato brown rugose fruit virus (ToBRFV).Tomato brown rugose fruit virus is an emerging tobamovirus first reported in 2014 in Israel and 2015 in Jordan on greenhouse tomatoes. In 2018, it was also reported in Mexico (where its eradication is under way), and in the United States where it was eradicated in the same year. Within the European Union (EU), it was reported in Germany in 2018 (North Rhine-Westphalia where its eradication is under way) and in Italy (Sicily) in early 2019. This virus was also the subject of an alert in the Bulletin de Santé du Végétal (BSV) for the Provence-Alpes-Côte d'Azur (PACA) region in December 2018. It was added to the alert list of the European and Mediterranean Plant Protection Organization (EPPO) in January 2019 and was the subject of an alert sheet issued by ANSES's Laboratory for Plant Health (LSV_2019_03_0027).The main host plants of ToBRFV are tomato (Solanum lycopersicum) and pepper (Capsicum annuum). The damage observed on greenhouse tomato plants includes chlorosis, mosaic and mottling on the leaves and necrotic spots on peduncles, calyces and flower stalks. Fruits show yellowor brown spots with characteristic rugose symptoms, and may be deformed and ripen irregularly, making them non-marketable. The incidence of the disease varies between 10 and 100% of fruit depending on the site. Similar symptoms have also been observed on Capsicum annuum fruits inMexico. Viruses belonging to the genus Tobamovirus have a particularly effective means of spread. Transmission is mechanical, through simple direct contact between plants or via hands, work tools, clothing, pollinating insects, birds or irrigation water. Seeds can also transmit tobamoviruses. Lastly, these viruses are highly stable and can survive for several months on inert media without any loss of infectivity. These epidemiological characteristics make tobamoviruses a formidable threat to highdensity crops such as those grown in greenhouses.The long-lasting resistance carried by the Tm-2 and Tm-2 2 genes deployed in tomato varieties in production since the 1970s has led to a considerable reduction in the negative impacts associated with other tobamoviruses (tomato mosaic virus and tobacco mosaic virus). However, these genes are ineffective against ToBRFV. The lack of resistance in tomatoes therefore particularly jeopardises this production sector, which has so far been spared.1.2 Purpose of the requestIn view of the importance of the greenhouse tomato production sector in France, the high infectivity of this virus and the susceptibility of the tomato, the succession of reports at production sites on several continents and especially within the EU, and the lack of regulatory measures against thisvirus, which could lead to certain high risk pathways, you are asked to carry out an express risk assessment of ToBRFV in order to:(i) assess the probability of introduction and spread of ToBRFV and the scale of the potential economic impacts in metropolitan France, (ii) propose possible management measures in the event of an unacceptable risk to the production sector.; Contexte en mars 2019Grâce à la veille sanitaire réalisée par les membres du Comité d’Experts Spécialisé de l’Anses et nos agents Anses, une alerte nous est parvenue concernant un virus émergent de la tomate : le Tomato brown rugose fruit virus (ToBRFV).Le Tomato brown rugose fruit virus est un tobamovirus émergent dont les premiers signalements datent de 2014 en Israël et de 2015 en Jordanie sur des tomates produites sous serre. En 2018, il a été également signalé au Mexique (où il est en cours d’éradication) et aux Etats-Unis où il a étééradiqué la même année. Les dégâts observés sur tomate en production sous serre incluent des chloroses, des mosaïques et des marbrures sur les feuilles, ainsi que des taches nécrotiques sur les pédoncules, calices et pédoncules floraux. Les fruits présentent des taches jaunes ou brunes, avec des symptômes de rugosité caractéristiques, et peuvent être déformés et avoir une maturation irrégulière, et ainsi devenir non commercialisables. L’incidence de la maladie varie entre 10 et 100%de fruits atteints selon les sites. Des symptômes similaires ont également été observés sur les fruits de Capsicum annuum au Mexique.Le mode de dissémination des virus appartenant au genre Tobamovirus est particulièrement efficace. La transmission est mécanique par simple contact direct entre les plantes ou via les mains, les outils de travail, les vêtements, les insectes pollinisateurs, les oiseaux et l’eau d’irrigation. Lessemences peuvent également transmettre les tobamovirus. Enfin, ces virus sont très stables : ils peuvent en effet survivre plusieurs mois sur des supports inertes sans perte de pouvoir infectieux. Ces caractéristiques épidémiologiques rendent les tobamovirus redoutables dans les cultures à haute densité de plantation comme les cultures conduites sous serre.La résistance durable portée par les gènes Tm-2 et Tm-22 et déployée dans les variétés de tomates de production depuis les années 1970 a permis une réduction considérable des impacts négatifs liés à d’autres tobamovirus (tomato mosaic virus et tobacco mosaic virus principalement). Or, ces gènes sont inefficaces contre le ToBRFV. L’absence de résistance chez la tomate met donc particulièrement en péril cette filière de production jusque-là épargnée.Objet de la saisine : Compte tenu de l’importance de la filière de production de tomates sous serre en France, du fort caractère infectieux de ce virus et de la sensibilité de la tomate, de la succession des signalements dans des sites de production sur plusieurs continents et plus particulièrement au sein de l’UE et de l’absence de mesure réglementaire vis-à-vis de ce virus qui pourrait engendrer des filières d’entrée à risque, il vous est demandé de réaliser une évaluation de risque simplifiée du ToBRFV afin de : (i) évaluer la probabilité d’introduction et de dissémination du ToBRFV et l’ampleur des impacts économiques potentiels sur le territoire français métropolitain,(ii) proposer des options de mesures de gestion en cas de risque inacceptable pour la filière de production.

Abstract: Translation initiation factors 4E (eIF4E) are the main source of resistance to potyvirus. We systematically assessed tomato single and double knock-out (KO) mutants of members of the eIF4E-coding gene family for resistance to Pepper veinal mottle virus (PVMV), a major constraint to tomato production. We show that the KO mutant of eIF4E2 has partial resistance to PVMV isolate IC, with plants harboring weak symptoms and low virus loads at the systemic level. The causal effect of eIF4E2 loss-of-function on resistance was confirmed on a progeny segregating for the KO mutation. The eIF4E2 KO mutant was resistant to six of the eight PVMV isolates tested and no resistance to other potyviruses was observed. This is the first evidence that mutation of eIF4E2 is in itself conferring resistance to a potyvirus and 3D protein modelling suggests that the eIF4E2 gene could be converted into a functional resistance allele.

Abstract: Cucurbits are widely grown vegetables in different provinces of Turkey. However their production is restricted by pests and pathogens including different plant viruses, among which Zucchini yellow mosaic virus (ZYMV) is particularly damaging. Infections by this virus during the early stages of plant development causes deformation of fruits and significant yield losses. During 2009-2014, 221 cucurbits samples were collected from fields and greenhouses from different provinces of Turkey. Serological testing was performed by double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) for presence of Watermelon mosaic virus (WMV), ZYMV, Cucumber mosaic virus (CMV), Papaya ring spot virus (PRSV), cucumber green mottle mosaic virus (CGMMV) and Squash mosaic virus (SqMV). For molecular analaysis of ZYMV, full-length CP nucleotide sequences were obtained for 45 ZYMV isolates and compared with sequences from worldwide isolates available in databases. The CP coding regions of Turkish ZYMV were 837 nt long and encoded 279 amino acids (aa). Forty two isolates from different regions were classified in the molecular subgroup Al, the most frequent in the Mediterranean Basin, Europe and Africa. Three isolates collected from Antalya region were classified in subgroup A4 and showed 99 % sequence identity with isolates from the same subgroup that have emerged recently in France.

Abstract: Cucumber vein yellowing virus (CVYV) is emerging throughout the Mediterranean Basin, where it causes important damages on cucumber and melon crops. It has been suggested that CVYV originated from the Middle East and has spread only recently to other areas. In this work, an isolate from Sudan was characterized and surveys performed in that country between 1992 and 2012 revealed a long‐term presence of CVYV with a high molecular variability, showing that the virus has long been endemic in Sub‐Saharan Africa. Comparison of the full‐length sequences of 11 CVYV isolates from different geographic origins revealed recombination events in CVYV populations from the Mediterranean Basin and the Middle East, and evidence for different selection pressures along the genome. These results shed a new light on the evolution of CVYV.

Abstract: Strategies to manage plant disease—from use of resistant varieties to crop rotation, elimination of reservoirs, landscape planning, surveillance, quarantine, risk modeling, and anticipation of disease emergences—all rely on knowledge of pathogen host range. However, awareness of the multitude of factors that influence the outcome of plant–microorganism interactions, the spatial and temporal dynamics of these factors, and the diversity of any given pathogen makes it increasingly challenging to define simple, all-purpose rules to circumscribe the host range of a pathogen. For bacteria, fungi, oomycetes, and viruses, we illustrate that host range is often an overlapping continuum—more so than the separation of discrete pathotypes—and that host jumps are common. By setting the mechanisms of plant–pathogen interactions into the scales of contemporary land use and Earth history, we propose a framework to assess the frontiers of host range for practical applications and research on pathogen evolution.

Abstract: International audience; In spite of their widespread occurrence, only few host jumps by plant viruses have been evidenced and the molecular bases of even fewer have been determined. A combination of three independent approaches, (i) experimental evolution followed by reverse genetics analysis, (ii) positive selection analysis and (iii) locus-by-locus analysis of molecular variance (AMOVA) allowed reconstructing the Potato virus Y (PVY; genus Potyvirus, family Potyviridae) jump to pepper (Capsicum annuum), probably from other solanaceous plants. Synthetic chimeras between infectious cDNA clones of two PVY isolates with contrasted levels of adaptation to C. annuum showed that the P3 and, to a lower extent, the CI cistron played important roles in infectivity towards C. annuum. The three analytical approaches pinpointed a single nonsynonymous substitution in the P3 and P3N-PIPO cistrons that evolved several times independently and conferred adaptation to C. annuum. In addition to increasing our knowledge of host jumps in plant viruses, this study illustrates also the efficiency of locus-by-locus AMOVA and combined approaches to reconstruct adaptive mutations in the genome of RNA viruses.

Abstract: Papaya meleira virus (PMeV) is the causal agent of papaya sticky disease, which is characterized by a spontaneous exudation of fluid and aqueous latex from the papaya fruit and leaves. The latex oxidizes after atmospheric exposure, resulting in a sticky feature on the fruit from which the name of the disease originates. PMeV is an isometric virus particle with a double-stranded RNA (dsRNA) genome of approximately 12 Kb. Unusual for a plant virus, PMeV particles are localized on and linked to the polymers present in the latex. The ability of the PMeV to inhabit such a hostile environment demonstrates an intriguing interaction of the virus with the papaya. A hypersensitivity response is triggered against PMeV infection, and there is a reduction in the proteolytic activity of papaya latex during sticky disease. In papaya leaf tissues, stress responsive proteins, mostly calreticulin and proteasome-related proteins, are up regulated and proteins related to metabolism are down-regulated. Additionally, PMeV modifies the transcription of several miRNAs involved in the modulation of genes related to the ubiquitin-proteasome system. Until now, no PMeV resistant papaya genotype has been identified and roguing is the only viral control strategy available. However, a single inoculation of papaya plants with PMeV dsRNA delayed the progress of viral infection.

Abstract: paper originating from a presentation made at the 13th International Plant Virus Epidemiology Symposium held at Avignon, France June 6-10, 2016; The “Papaya ringspot virus (PRSV) cluster” of cucurbit-infecting potyviruses contains five acknowledged species that have similar biological, serological and molecular properties. Additional data suggest there are other uncharacterized species from various locations in the world that likely belong to the PRSV cluster including a new PRSV-like virus reported from Sudan in 2003. Molecular and biological data indicated that the virus from Sudan belongs to a new species, tentatively named wild melon vein banding virus (WMVBV). The complete nucleotide sequence of a second virus from Sudan revealed it was a divergent relative of Moroccan watermelon mosaic virus (MWMV). Based on sequence similarity this virus was determined to be a distinct species and tentatively named Sudan watermelon mosaic virus (SuWMV). Molecular analyses indicate that SuWMV is a recombinant between WMVBV- and MWMV-related viruses. Based on surveys performed in Sudan between 1992 and 2012, SuWMV appeared 10 times more frequent than WMVBV in that country (14.6% vs. 1.5% of the samples tested). The geographic structure and molecular diversity patterns of the putative and acknowledged species suggest that the PRSV-like cluster originated in the Old World about 3600 years ago, with an important diversification in Africa.

Abstract: Recent advances in high-throughput sequencing technologies and bioinformatics have generated huge new opportunities for discovering and diagnosing plant viruses and viroids. Plant virology has undoubtedly benefited from these new methodologies, but at the same time, faces now substantial bottlenecks, namely the biological characterization of the newly discovered viruses and the analysis of their impact at the biosecurity, commercial, regulatory, and scientific levels. This paper proposes a scaled and progressive scientific framework for efficient biological characterization and risk assessment when a previously known or a new plant virus is detected by next generation sequencing (NGS) technologies. Four case studies are also presented to illustrate the need for such a framework, and to discuss the scenarios.

Abstract: The impact of flower strips on the colonization of crops by the aphid Aphis gossypii and its natural enemies was studied from 2013 et 2015 within the framework of the AGATH project, funded by the Ministère de l’Agriculture, de l’Alimentation et de la Forêt (Casdar) and led by the Ctifl. A mix of flowers, tailored by the INRA (French National Institute for Agricultural Reasearch) to attract natural enemies of aphids in the proximity of melon crops without having a negative impact on the crops, was assessed in a network of plots throughout the three main melon production areas. The attractivity of this mix of flowers to natural enemies was characterized. The density of A. gossypii and of arthropod predator and/or parasitoid populations was compared in melon plots with these flower strips and in identical plots in terms of crop management but with bare soil.; L’effet de bandes fleuries sur la colonisation des cultures par le puceron Aphis gossypii et ses ennemis naturels a été étudié dans le cadre du projet AGATH, soutenu par le ministère de l’Agriculture, de l’Alimentation et de la Forêt (Casdar) et porté par le Ctifl de 2013 à 2015. Un mélange fleuri conçu par l’Inra pour attirer les ennemis naturels de pucerons à proximité des parcelles de melon sans exercer en parallèle d’effet délétère sur les cultures a été évalué dans un réseau de parcelles réparties dans les trois principaux bassins de production de melons. L’attractivité de ce mélange fleuri vis-à-vis des principaux ennemis naturels de pucerons a été caractérisée. Les densités des populations d’A. gossypii et d’arthropodes prédateurs et/ou parasitoïdes de pucerons mesurées dans des parcelles de melon bordées par ce mélange fleuri et dans des parcelles identiques en termes d’itinéraire technique mais bordées par une surface de sol nu ont été comparées.

Abstract: Plant pathogens can play a role in the competitive interactions between plant species and have been understudied in native prairies, which are declining globally, and in Conservation Reserve Program (CRP) lands in the United States. Barley/Cereal yellow dwarf virus (B/CYDV) are among the most economically important disease-causing agents of small grain cereal crops, such as wheat, and are known to infect over 150 Poaceae species, including many of the grass species present in prairies and CRP lands. Field surveys of Poaceae species were conducted in endangered Palouse Prairie and CRP habitats of southeastern Washington and adjacent northern Idaho, USA from 2010 to 2012 to examine for the presence of B/CYDV among plant hosts and aphid vectors. Viral species were identified via cloning and sequencing. Landscape, soil and climate data were retrieved from USDA-NASS and USDA-NRCS databases. Analyses were conducted to examine effects of diverse agroecological and environmental factors on virus prevalence. A total of 2271 grass samples representing 30 species were collected; 28 of these were infected with BYDV in at least one location. BYDV infection was detected at every CRP and prairie remnant sampled, with an overall infection of 46%. BYDV-SGV and BYDV-PAV were the only two B/CYDV species encountered, with BYDV-SGV being more prevalent. Sampling time (season) and host plant identity were the main variables explaining variation in virus prevalence among sites. BYDV was more prevalent in perennial compared to annual grass species. Aphids were encountered only once suggesting non-colonizing aphids, potentially from neighboring cereal fields, are responsible for disease spread in these habitats. BYDV prevalence increased in sampled habitats as cereal crop cover increased within a 1-km radius of a habitat patch. Results demonstrate moderate to high and persistent prevalence of BYDV in an endangered grassland habitat. Species composition and susceptibility to pathogens should be considered when creating seed mixes for CRP sites, especially in relation to agricultural crops and diseases in a region. Future work exploring host abundance, competence and habitat utilization by vectors is required to fully elucidate BYDV ecology and epidemiology in grassland habitats.

Abstract: Cacao swollen shoot virus (CSSV) is a major pathogen of cacao (Theobroma cacao) in Africa, and long-standing efforts to limit its spread by the culling of infected trees have had very limited success. CSSV is a particularly difficult virus to study, as it has a very narrow host range, limited to several tropical tree species. Furthermore, the virus is not mechanically transmissible, and its insect vector can only be used with difficulty. Thus, the only efficient means to infect cacao plants that have been experimentally described so far are by particle bombardment or the agroinoculation of cacao plants with an infectious clone. We have genetically transformed three non-host species with an infectious form of the CSSV genome: two experimental hosts widely used in plant virology (Nicotiana tabacum and N. benthamiana) and the model species Arabidopsis thaliana. In transformed plants of all three species, the CSSV genome was able to replicate, and, in tobacco, CSSV particles could be observed by immunosorbent electron microscopy, demonstrating that the complete virus cycle could be completed in a non-host plant. These results will greatly facilitate the preliminary testing of CSSV control strategies using plants that are easy to raise and to transform genetically.

Abstract: International audience; Several potyviruses affect lettuce (Lactuca sativa) and chicory (Cichorium spp.) crops worldwide and are important constraints for production because of the direct losses that they induce and/or because of their seed transmission. Here, the molecular and biological properties are described of two potyviruses that were recently isolated from lettuce plants showing mosaic or strong necrotic symptoms in an experimental field in southeastern France. The first potyvirus belongs to the species Endive necrotic mosaic virus and is present in a large number of wild plant species, especially Tragopogon pratensis. It is unable to infect lettuce cultivars with a resistance to Turnip mosaic virus that is present in many European cultivars and probably conferred by the Tu gene. The second potyvirus belongs to the tentative species lettuce Italian necrotic virus and was not observed in wild plants. It infected all tested lettuce cultivars. Wild accessions of Lactuca serriola, Lactuca saligna, Lactuca virosa and Lactuca perennis were identified as resistant to one or the other potyvirus and could be used for resistance breeding in lettuce. No resistance against these two potyviruses was observed in the tested Cichorium endivia cultivars. In contrast, all tested Cichorium intybus cultivars or accessions were resistant.

Abstract: paper originating from a presentation made at the 13th International Plant Virus Epidemiology Symposium held at Avignon, France June 6-10, 2016; Ornamental plants constitute a largely unknown and potentially important source of pathogens affecting not only ornamental plants, but also major crop species. We have carried out studies using high-throughput sequencing of 21-24 nt RNAs from potentially virus-infected ornamental plants, followed by assembly of sequence scaffolds, to identify the virus and viroid genomes present in a panel of 67 plant samples representing 46 species belonging to the main sectors of the ornamental plant industry (cut flowers, pot plants, bulbs). A pilot study demonstrated that samples could be pooled (5 samples per pool), and the overall process simplified without loss of detection of important known pathogens. In a full-scale study, pools of 5 samples were organized in a 5 × 5 matrix to facilitate attribution of a sequence to a precise sample directly from analysis of the matrix. In the total of 67 samples analyzed in the two studies, partial sequences suggesting the presence of 25 previously unknown viruses and viroids were detected, including all types of virus and viroid genomes, and also showed four cases of known viruses infecting previously undescribed hosts. Furthermore, two types of potential mis-assembly were analyzed, and were shown to not affect the conclusions regarding the presence of the pathogens identified, but show that mis-assembly can affect the results when the objective is determining complete bona fide viral genome sequences. These results clearly confirm that ornamental plants constitute a potential source of unknown viruses and viroids that could have a major impact on agriculture, and that sequencing siRNAs of potentially virus- or viroid-infected ornamental plants is an effective means for screening for the presence of potentially important pathogens.

Abstract: paper originating from a presentation made at the 13th International Plant Virus Epidemiology Symposium held at Avignon, France June 6-10, 2016; Cultivar choice is at the heart of cropping systems and resistant cultivars should be at the heart of disease management strategies whenever available. They are the easiest, most efficient and environmentally friendly way of combating viral diseases at the farm level. Among the melon genetic resources, Vat is a unique gene conferring resistance to both the melon aphid Aphis gossypii and the viruses it carries. The ‘virus side’ of this pleiotropic phenotype is seldom regarded as an asset for virus control. Indeed, the effect of Vat on virus epidemics in the field is expected to vary according to the composition of aphid populations in the environment and long-term studies are needed to draw a correct trend. Therefore, the first objective of the study was to re-evaluate the potential of Vat to reduce viral diseases in melon crops. The second objective was to investigate the potential of Vat to exert a selection pressure on virus populations. We monitored the epidemics of Cucurbit aphid-borne yellows virus (CABYV), Cucumber mosaic virus (CMV), Watermelon mosaic virus (WMV) and Zucchini yellow mosaic virus (ZYMV) in two melon lines having a common genetic background, a resistant line (R) and a susceptible line (S), in eight field trials conducted in southeastern France between 2011 and 2015. Vat had limited impact if any on WMV epidemics probably because A. gossypii is not the main vector of WMV in the field, but a favorable impact on CMV, yet of variable intensity probably related to the importance of A. gossypii in the total aphid population. Vat had a significant impact on CABYV epidemics with mean incidence reduction exceeding 50% in some trials. There was no effect of Vat on the structure of virus populations, both for the non-persistent WMV transmitted by numerous aphid species and for the persistent CABYV transmitted predominantly by A. gossypii.

Abstract: The viral protein genome-linked (VPg) of potyviruses is a protein covalently linked to the 5′ end of viral RNA. It interacts with eIF4E, a component of the cellular translation initiation complex. It has been suggested that the 5′ RNA-linked VPg could mimic the cellular mRNA cap, promoting synthesis of viral proteins. Here, we report evidence for recruitment of the plant eIF4E by Lettuce mosaic virus (LMV, potyvirus) particles via the 5′ RNA-linked VPg. Analysis of the viral population was performed by enzyme-linked immunosorbent assay–based tests, either with crude extracts of LMV-infected tissues or purified viral particles. In both cases, LMV-VPg and LMV-eIF4E subpopulations could be detected. After reaching a maximum within the first 2 weeks postinoculation, these populations decreased and very few labeled particles were found later than 3 weeks postinoculation. The central domain of VPg (CD-VPg) was found to be exposed at the surface of the particles. Using a purified recombinant lettuce eIF4E and CD-VPg–specific antibodies, we demonstrate that the plant factor binds to the VPg via its central domain. Moreover, the plant eIF4E factor could be imaged at one end of the particles purified from LMV plant extracts, by immunoredox atomic force microscopy coupled to scanning electrochemical microscopy. We discuss the biological significance of these results.

Abstract: UMR BFP - Equipe Virologie; The genomes of two novel viruses were assembled from 454 pyrosequencing data obtained from vanilla leaves from La Réunion. Based on genome organization and homologies, one agent was unambiguously classified as a member of the genus Potexvirus and named vanilla virus X (VVX). The second one, vanilla latent virus (VLV), is phylogenetically close to three unclassified members of the family Alphaflexiviridae with similarity to allexiviruses, and despite the presence of an additional 8-kDa open reading frame, we propose to include VLV as a new member of the genus Allexivirus. Both VVX and VLV were mechanically transmitted to vanilla plants, resulting in asymptomatic infections.

Abstract: Tomato mild yellow leaf curl Aragua virus (ToMYLCV) is a begomovirus first reported infecting tomato (Solanum lycopersicum) and milkweed (Euphorbia heterophylla) in Venezuela. In this study, a ToMYLCV isolate (Zulia-219) was completely sequenced and its host range was evaluated. The DNA-A and DNA-B components of isolate Zulia-219 showed 93 and 85% nucleotide sequence identity with the respective counterparts of the ToMYLCV type strain. According to current demarcation criteria for begomovirus species, Zulia-219 is a new strain of ToMYLCV. Interestingly, tomato plants inoculated with ToMYLCV Zulia-219 displayed severe symptoms, including severe chlorotic leaf curling, in contrast to mild symptoms associated with the type strain of this begomovirus. These results indicate potential risks associated with this new ToMYLCV strain for tomato production in Venezuela.
