Abstract Background Phosphonates are the main components in the global phosphorus redox cycle. Little is known about phosphonate metabolism in freshwater ecosystems, although rapid consumption of phosphonates has been observed frequently. Cyanobacteria are often the dominant primary producers in freshwaters; yet, only a few strains of cyanobacteria encode phosphonate-degrading (C-P lyase) gene clusters. The phycosphere is defined as the microenvironment in which extensive phytoplankton and heterotrophic bacteria interactions occur. It has been demonstrated that phytoplankton may recruit phycospheric bacteria based on their own needs. Therefore, the establishment of a phycospheric community rich in phosphonate-degrading-bacteria likely facilitates cyanobacterial proliferation, especially in waters with scarce phosphorus. We characterized the distribution of heterotrophic phosphonate-degrading bacteria in field Microcystis bloom samples and in laboratory cyanobacteria “phycospheres” by qPCR and metagenomic analyses. The role of phosphonate-degrading phycospheric bacteria in cyanobacterial proliferation was determined through coculturing of heterotrophic bacteria with an axenic Microcystis aeruginosa strain and by metatranscriptomic analysis using field Microcystis aggregate samples. Results Abundant bacteria that carry C-P lyase clusters were identified in plankton samples from freshwater Lakes Dianchi and Taihu during Microcystis bloom periods. Metagenomic analysis of 162 non-axenic laboratory strains of cyanobacteria (consortia cultures containing heterotrophic bacteria) showed that 20% (128/647) of high-quality bins from eighty of these consortia encode intact C-P lyase clusters, with an abundance ranging up to nearly 13%. Phycospheric bacterial phosphonate catabolism genes were expressed continually across bloom seasons, as demonstrated through metatranscriptomic analysis using sixteen field Microcystis aggregate samples. Coculturing experiments revealed that although Microcystis cultures did not catabolize methylphosphonate when axenic, they demonstrated sustained growth when cocultured with phosphonate-utilizing phycospheric bacteria in medium containing methylphosphonate as the sole source of phosphorus. Conclusions The recruitment of heterotrophic phosphonate-degrading phycospheric bacteria by cyanobacteria is a hedge against phosphorus scarcity by facilitating phosphonate availability. Cyanobacterial consortia are likely primary contributors to aquatic phosphonate mineralization, thereby facilitating sustained cyanobacterial growth, and even bloom maintenance, in phosphate-deficient waters. Energy (photosynthetically active [PAR] and ultraviolet [UVR] radiation) and matter (organic and inorganic nutrients) fluxes regulate the ecosystem's stability. However, the mechanisms underpinning the potential interplay between resistance and resilience to shifts in nutrient inputs and UVR are poorly understood. To assess how the UVR × nutrients interaction alters ecosystem stability, we exposed in situ a microbial food web from an oligotrophic ecosystem to: (1) two light (UVR + PAR and PAR), and (2) four nutrient (ambient concentrations, phosphorus [P], carbon [C] and C × P addition) treatments for three weeks. During this period, we quantified the community composition and biomass, sestonic P and C:P ratio, primary [PP] and bacterial [BP] production, community [CR] and bacterial [BR] respiration, excreted organic carbon [EOC], as well as the commensalistic phytoplankton-bacteria interaction (i.e. bacterial carbon demand [BCD]:EOC ratio) and the metabolic balance of the ecosystem (i.e. [PP:R] ratio). The stability of all response variables under the four environmental scenarios tested (i.e. UVR, UVR × C, UVR × P, and UVR × C × P) was quantified by means of the resistance and resilience indexes. The microbial community was dominated by phototrophs during the experimental period regardless of the treatment considered. The most complex scenario, i.e. UVR × C × P, decreased the resistance for all variables, except for BR and the PP:R ratio. Despite that PP:R ratio showed the highest resistance under such scenario, it was >1 in all environmental scenarios (i.e. net autotrophic), except under the UVR × C interaction, where, concomitant with increased resilience, the balance shifted towards net heterotrophy (PP:R < 1). Under the UVR × C × P scenario, the metabolic balance of the ecosystem proved strongly resistant due mainly to high resistance of bacterial respiration and a firm stability of the commensalistic interaction. Our results evidence that the high resilience of phototrophs (favoring their predominance over mixo- and heterotrophs) may lead to the maintenance of the autotrophic nature and carbon (C) sink capacity of the ecosystem. While many studies have examined the impact of oil on phytoplankton or bacteria, very few considered the effects on the biological complex formed by phytoplankton and their associated phytoplankton-attached (PA) and free-living (FL) bacteria. However, associated bacteria can affect the physiology of phytoplankton and influence their stress responses. In this study, we monitored the growth of Heterocapsa sp., an armoured dinoflagellate, exposed to crude oil, Corexit dispersant, or both. Growth of Heterocapsa sp. is unaffected by crude oil up to 25 ppm, a concentration similar to the lower range measured on Florida beaches after the Deepwater Horizon oil spill. The PA bacteria community was resistant to exposure, whereas the FL community shifted towards oil degraders; both responses could contribute to Heterocapsa sp. oil resistance. The growth rate of Heterocapsa sp. decreased significantly only when exposed to dispersed oil at 25 ppm, indicating a synergistic effect of dispersant on oil toxicity in this organism. For the first time, we demonstrated the decoupling of the responses of the PA and FL bacteria communities after exposure to an environmental stress, in this case oil and dispersant. Our findings suggest new directions to explore in the understanding of interactions between unicellular eukaryotes and prokaryotes. Significance and Impact of the Study In the environment, oil spills have the capacity to modify phytoplankton communities, with important consequences on the food web and the carbon cycle. We are just beginning to understand the oil resistance of phytoplankton species, making it difficult to predict community response. In this study we highlighted the strong resistance of Heterocapsa sp. to oil, which could be associated with its resilient attached bacteria and oil degradation by the free-living bacteria. A large number of tailings ponds formed by slag accumulation have become serious environmental hazards. Spatially high potential energy and long-term accumulation may result in gradient-changing seepage pollution. The assemblages of phytoplankton and bacteria are widely used as assessment indicators. In this study, we investigate the changes in phytoplankton and bacterial assemblages in tailing pollution. The results showed that there are temporal and spatial variabilities in seepage pollution. The abundance and diversity of phytoplankton and bacteria decreased with increasing pollution. However, Synedra acus (diatom) and Polynucleobacter (bacteria) were positively correlated with pollution levels (r = 0.37, P < 0.05; r = 0.24, P < 0.05). Heavy metals are the main contributors to bacterial changes (16.46%), while nutrients are for algae (13.24%). Tailings pond pollution reduced the number of phytoplankton and bacterial linkages. However, more pollution broke the originally independent modules of phytoplankton and bacteria, and they produced more positive correlations (79.39%; 87.68%). Microcystis sp. and Limnobacter were the key nodes of the co-occurrence network in the polluted areas. Exploring the interactions between bacteria and phytoplankton within different pollution levels could provide insights into biological interaction patterns and the bioremediation of tailings ponds. Phytoplankton-bacteria interactions drive the surface ocean sulfur cycle and local climatic processes through the production and exchange of a key compound: dimethylsulfoniopropionate (DMSP). Despite their large-scale implications, these interactions remain unquantified at the cellular-scale. Here we use secondary-ion mass spectrometry to provide the first visualization of DMSP at sub-cellular levels, tracking the fate of a stable sulfur isotope ( 34 S) from its incorporation by microalgae as inorganic sulfate to its biosynthesis and exudation as DMSP, and finally its uptake and degradation by bacteria. Our results identify for the first time the storage locations of DMSP in microalgae, with high enrichments present in vacuoles, cytoplasm and chloroplasts. In addition, we quantify DMSP incorporation at the single-cell level, with DMSP-degrading bacteria containing seven times more 34 S than the control strain. This study provides an unprecedented methodology to label, retain, and image small diffusible molecules, which can be transposable to other symbiotic systems.
