Agricultural environments are becoming increasingly contaminated with plastic pollution. Plastics in the environment can also provide a unique habitat for microbial biofilm, termed the 'plastisphere', which can also support the persistence of human pathogens such as Salmonella. Human enteric Salmonella enterica serovar Typhimurium can enter agricultural environments via flooding or from irrigation with contaminated water. Using soil mesocosms we quantified the ability of S. Typhimurium to persist on microplastic beads in two agriculturally relevant soils, under ambient and repeat flood scenarios. S. Typhimurium persisted in the plastisphere for 35 days in both podzol and loamy soils; while during multiple flood events was able to survive in the plastisphere for up to 21 days. S. Typhimurium could dissociate from the plastisphere during flooding events and migrate through soil in leachate, and importantly could colonise new plastic particles in the soil, suggesting that plastic pollution in agricultural soils can aid S. Typhimurium persistence and facilitate further dissemination within the environment. The potential for increased survival of enteric human pathogens in agricultural and food production environments due to plastic contamination poses a significant public health risk, particularly in potato or root vegetable systems where there is the potential for direct contact with crops. Microplastics (MPs) and antibiotic resistance genes (ARGs) in the environment are the major subject of concern across the globe as they jeopardize not only aquatic life but human health as well. MPs are potentially competent to adsorb various micropollutants (heavy metals), bacterial communities, and biofilm formation which comply with the requirement of efficient sorption of antibiotics and transfer of ARGs. MPs adsorb antibiotics through physicochemical interactions, leading to the formation of biofilm-enriched bacteria on the surfaces of MPs that further enhances the abundance of antibiotic resistance genes (ARGs) and facilitates their dispersal. Therefore, this review recapitulates the interactive forces responsible for antibiotics sorption on MPs and also the intricate relationship between MPs, antibiotics, and microbial hosts of ARGs. Furthermore, the role of the wastewater treatment plants (WWTPs), which serve as a formidable connecting link between MPs and the ecosystem, is discussed in relation to dissemination of antibiotics, ARGs, and antibiotic-resistant bacteria (ARB) to get a clear perception of the contribution of WWTPs in drawing together MPs with antibiotics, ARB, and ARGs. Abstract Background Antibiotics and microplastics are two major aquatic pollutants that have been associated to antibiotic resistance selection in the environment and are considered a risk to human health. However, little is known about the interaction of these pollutants at environmental concentrations and the response of the microbial communities in the plastisphere to sub-lethal antibiotic pollution. Here, we describe the bacterial dynamics underlying this response in surface water bacteria at the community, resistome and mobilome level using a combination of methods (next-generation sequencing and qPCR), sequencing targets (16S rRNA gene, pre-clinical and clinical class 1 integron cassettes and metagenomes), technologies (short and long read sequencing), and assembly approaches (non-assembled reads, genome assembly, bacteriophage and plasmid assembly). Results Our results show a shift in the microbial community response to antibiotics in the plastisphere microbiome compared to surface water communities and describe the bacterial subpopulations that respond differently to antibiotic and microplastic pollution. The plastisphere showed an increased tolerance to antibiotics and selected different antibiotic resistance bacteria (ARB) and antibiotic resistance genes (ARGs). Several metagenome assembled genomes (MAGs) derived from the antibiotic-exposed plastisphere contained ARGs, virulence factors, and genes involved in plasmid conjugation. These include Comamonas, Chryseobacterium, the opportunistic pathogen Stenotrophomonas maltophilia, and other MAGs belonging to genera that have been associated to human infections, such as Achromobacter. The abundance of the integron-associated ciprofloxacin resistance gene aac(6’)-Ib-cr increased under ciprofloxacin exposure in both freshwater microbial communities and in the plastisphere. Regarding the antibiotic mobilome, although no significant changes in ARG load in class 1 integrons and plasmids were observed in polluted samples, we identified three ARG-containing viral contigs that were integrated into MAGs as prophages. Conclusions This study illustrates how the selective nature of the plastisphere influences bacterial response to antibiotics at sub-lethal selective pressure. The microbial changes identified here help define the selective role of the plastisphere and its impact on the maintenance of environmental antibiotic resistance in combination with other anthropogenic pollutants. Plastics are offering a new niche for microorganisms colonizing their surface, the so-called "plastisphere," in which diversity and community structure remain to be characterized and compared across ocean pelagic regions. Here, we compared the bacterial diversity of microorganisms living on plastic marine debris (PMD) and the surrounding free-living (FL) and organic particle-attached (PA) lifestyles sampled during the Tara expeditions in two of the most plastic polluted zones in the world ocean, i.e., the North Pacific gyre and the Mediterranean Sea. The 16S rRNA gene sequencing analysis confirmed that PMD are a new anthropogenic ocean habitat for marine microbes at the ocean-basin-scale, with clear niche partitioning compared to FL and PA lifestyles. At an ocean-basin-scale, the composition of the plastisphere communities was mainly driven by environmental selection, rather than polymer types or dispersal effect. A plastisphere "core microbiome" could be identified, mainly dominated by Rhodobacteraceae and Cyanobacteria. Predicted functions indicated the dominance of carbon, nitrogen and sulfur metabolisms on PMD that open new questions on the role of the plastisphere in a large number of important ecological processes in the marine ecosystem. Microplastics (MPs) are of increasing concern due to their role as reservoirs for antibiotic resistance genes (ARGs) and pathogens. To date, few studies have explored the influence of anthropogenic activities on ARGs and mobile genetic elements (MGEs) within various riverine MPs, in comparison to their natural counterparts. Here an in-situ incubation was conducted along heavily anthropogenically-impacted Houxi River to characterize the geographical pattern of antibiotic resistome, mobilome and pathogens inhabiting MPs- and leaf-biofilms. The metagenomics result showed a clear urbanization-driven profile in the distribution of ARGs, MGEs and pathogens, with their abundances sharply increasing 4.77 to 19.90 times from sparsely to densely populated regions. The significant correlation between human fecal marker crAssphage and ARG (R2 = 0.67, P=0.003) indicated the influence of anthropogenic activity on ARG proliferation in plastisphere and natural leaf surfaces. And mantel tests and random forest analysis revealed the impact of 17 socio-environmental factors, e.g., population density, antibiotic concentrations, and pore volume of materials, on the dissemination of ARGs. Partial least squares-path modeling further unveiled that intensifying human activities not only directly boosted ARGs abundance but also exerted a comparable indirect impact on ARGs propagation. Furthermore, the polyvinylchloride plastisphere created a pathogen-friendly habitat, harboring higher abundances of ARGs and MGEs, while polylactic acid are not likely to serve as vectors for pathogens in river, with a lower resistome risk score than that in leaf-biofilms. This study highlights the diverse ecological risks associated with the dissemination of ARGs and pathogens in varied MPs, offering insights for the policymaking of usage and control of plastics within urbanization.

Upon entering the marine environment, plastics are colonized by a plethora of microorganisms to form a plastisphere, influencing the fate and transport of the plastic debris and the health of marine ecosystems. The assembly of marine plastisphere is generally believed to be dominated by stochastic processes. However, it remains elusive whether microbial interaction in the assembly of plastisphere microbial communities is conserved or not. We analyzed the plastisphere microbiomes of 137 plastic debris samples from intertidal zones at different geographical locations and habitats (seagrass, coral, mangrove, beach, and open ocean) and compared them with the surrounding sediment and seawater microbiomes. Microbial community structures of the plastisphere from different locations were more similar to each other but differed substantially from the surrounding sediment and water microbiomes, implying a common mechanism of plastisphere assembly. We used different machine learning algorithms (Multinomial Logistic Regression, Support Vector Machine, Decision Trees, Random Forest, and Artificial Neural Networks) to classify plastic debris samples with high sensitivity based on the microbiome composition. Eukaryotic and prokaryotic phototrophic organisms such as green algae, diatoms, and cyanobacteria, were found to be enriched on the plastic surfaces. Network analysis revealed the central role of the phototrophic organisms in the formation and sustenance of the plastispheres. We found that phototrophs served as core members interacting strongly with heterotrophic organisms in marine plastisphere, irrespective of the sampling location, habitats, and polymer types. This would explain the stochastic assembly of the plastisphere along with conserved properties driven by the phototrophs in the surrounding environment. Our results highlight the importance of phototrophic organisms in shaping the marine plastisphere microbial communities.

Microplastics (MPs) have emerged as a pressing global pollution issue and have drawn significant attention in the field of environmental science in recent years. Plastics are widely used in aquaculture, leading to the accumulation of MPs in aquaculture environments, referred to as aquaculture-derived MPs (AD-MPs). This review summarizes the pollution status of MPs in aquaculture environments, their adverse impacts on aquaculture, and the potential benefits of MPs serving as substrates for nitrogen-cycling microbiota to improve water quality in aquaculture. Our study underscores the severity of microplastic pollution in aquaculture, with the microplastic content reaching 362.8 items L−1 in water and up to 124,884 items kg−1 in sediment. However, these levels vary significantly depending on the type of aquaculture system and the specific location of aquaculture activities, with MPs being most abundant in estuary aquaculture systems. The primary sources of MPs in aquaculture environments include plastic nets, packaging and other plastic tools used in aquaculture. Consequently, most of the MPs present are white fibers composed of polypropylene and polyethylene. These MPs accumulate in aquatic products such as fish, shrimp, crabs, and shellfish, with abundances reaching 61.75 items per individual. This accumulation leads to toxic and combined effects on cultured animals and poses a risk to human health through the food chain. However, the surfaces of MPs (the plastisphere) also facilitate a series of chemical reactions, including biological nitrogen fixation, nitrification and denitrification, by serving as a substrate that attracts nitrogen-cycling microorganisms. These findings highlight the potential positive effects of MPs in aquaculture. To mitigate the ecological risks arising from MPs in aquaculture and address their potential threats to human health, it is essential to conduct long-term, sustainable assessments of MPs in aquaculture water sources. Special attention needs to be given to the effects of nitrogen-cycling biofilms on the surface of MPs and their potential benefits to aquaculture water quality.

Frequently used protective items during the pandemics are prepared from plastics products, such as polypropylene, polyurethane, polyacrylonitrile, polystyrene, polycarbonate, polyethylene, and polyester. Globally more than 57 million pounds of personal protective equipment along with other biomedical plastic waste are entering into the oceans per annum. Lots of economic benefits can be obtained by recycling the polymeric wastes by converting them into useful products through, reduced manufacture cost, bio converted products, and cleaner environment by developing a circular waste management model. Personal protective equipment's are prepared from different plastic products and the outer most surface of these free-floating plastics presents in the aquatic environment serve as colonization support to diverse microbial populations known as plastisphere. Microbial colonization starts with an initial attachment under different environmental conditions, subsequently by developing a permanent association on the plastic surface. The community structure of microorganisms in the plastisphere can be significantly influenced by the depth of water in the aquatic ecosystem where microplastics present and the nutrients availability. As microorganisms have been deeply investigated for their role in the biodegradation of several pollutants, the present review article focuses on microbial diversity on plastisphere in the aquatic ecosystem, the underlying mechanism of colonization, and their potential role in microplastic degradation. Background: Plastic waste is a global environmental issue that impacts the well-being of humans, animals, plants, and microorganisms. Microplastic contamination has been previously reported at Kung Wiman Beach, located in Chanthaburi province along with the Eastern Gulf of Thailand. Our research aimed to study the microbial population of the sand and plastisphere and isolate microorganisms with potential plastic degradation activity. Methods: Plastic and sand samples were collected from Kung Wiman Beach for microbial isolation on agar plates. The plastic samples were identified by Fourier-transform infrared spectroscopy. Plastic degradation properties were evaluated by observing the halo zone on mineral salts medium (MSM) supplemented with emulsified plastics, including polystyrene (PS), polylactic acid (PLA), polyvinyl chloride (PVC), and bis (2-hydroxyethyl) terephthalate (BHET). Bacteria and fungi were identified by analyzing nucleotide sequence analysis of the 16S rRNA and internal transcribed spacer (ITS) regions, respectively. 16S and ITS microbiomes analysis was conducted on the total DNA extracted from each sample to assess the microbial communities. Results: Of 16 plastic samples, five were identified as polypropylene (PP), four as polystyrene (PS), four as polyethylene terephthalate (PET), two as high-density polyethylene (HDPE), and one sample remained unidentified. Only 27 bacterial and 38 fungal isolates were found to have the ability to degrade PLA or BHET on MSM agar. However, none showed degradation capabilities for PS or PVC on MSM agar. Notably, Planococcus sp. PP5 showed the highest hydrolysis capacity of 1.64 ± 0.12. The 16S rRNA analysis revealed 13 bacterial genera, with seven showing plastic degradation abilities: Salipiger, Planococcus, Psychrobacter, Shewanella, Jonesia, Bacillus, and Kocuria. This study reports, for the first time of the BHET-degrading properties of the genera Planococcus and Jonesia. Additionally, The ITS analysis identified nine fungal genera, five of which demonstrated plastic degradation abilities: Aspergillus, Penicillium, Peacilomyces, Absidia, and Cochliobolus. Microbial community composition analysis and linear discriminant analysis effect size revealed certain dominant microbial groups in the plastic and sand samples that were absent under culture-dependent conditions. Furthermore, 16S and ITS amplicon microbiome analysis revealed microbial groups were significantly different in the plastic and sand samples collected. Conclusions: We reported on the microbial communities found on the plastisphere at Kung Wiman Beach and isolated and identified microbes with the capacity to degrade PLA and BHET. The increasing number of plastic particles in the environment gives rise to a unique ecological niche called the plastisphere, which is characterized by a dynamic assemblage of microorganisms comprising autotrophs, heterotrophs, predators, and pathogens. This paper reviews the formation, characteristics, and factors influencing the terrestrial and aquatic plastisphere. The terrestrial and aquatic plastisphere forms when hitchhiking microorganisms, often bacteria, in the environment adhere to the surfaces of plastic particles and alter the surface properties of the particles for subsequent colonization of increasingly tightly clinging microorganisms. The terrestrial plastisphere is not as mobile as the aquatic plastisphere and is defined by the characteristics of the soil anchoring it. The microorganisms on the plastisphere are often defined by the dominant microorganisms in the surroundings, particularly those of the phyla Proteobacteria, Actinobacteria, Bacteroidota, Firmicutes, Chloroflexi, Acidobacteria, Cyanobacteria, Ascomycota, Basidiomycota, Chytridiomycota, Ciliophora, Ochrophyta, and Chlorophyta. However, the compositions and dominant microbial species on different plastic surfaces vary widely as they change with time, plastic properties, and biotic and abiotic environmental factors. Temporal changes in the plastisphere are due to succession. Plastic properties, particularly the type, surface properties, size, color, degree of aging, and chemical leaching, also affect the composition of the plastisphere. Biotic environmental factors comprising the dominant ambient microorganisms and interspecies interactions, together with the abiotic ones, including pH, temperature, nutrient availability, soil properties, salinity, and light, significantly shape the plastisphere. Although accumulating evidence indicates that endangered animals suffer from plastic pollution, this has been largely overlooked. Here, we explored the bacteria and eukaryotes living in the plastics gathered from the natural habitat of the highly endangered crocodile lizard. The results demonstrated that the bacterial and eukaryotic communities on plastics formed a unique ecosystem that exhibited lower diversity than those in the surrounding water and soil. However, microbes displayed a more complex and stable network on plastic than that in water or soil, implying unique mechanisms of stabilization. These mechanisms enhanced their resilience and contributed to the provision of stable ecological services. Eukaryotes formed a simpler and smaller network than bacteria, indicating different survival strategies. The bacteria residing on the plastics played a significant role in carbon transformation and sequestration, which likely impacted carbon cycling in the habitat. Furthermore, microbial exchange between plastics and the crocodile lizard was observed, suggesting that plastisphere serves as a mobile gene bank for the exchange of information, including potentially harmful substances. Overall, microbes on plastic appear to significantly impact the crocodile lizard and its natural habitat via various pathways. These results provided novel insights into risks evaluation of plastic pollution and valuable guidance for government efforts in plastic pollutant control in nature reserves.

Plastic pollution in coastal areas, particularly in subtropical and tropical regions, remains a pervasive environmental issue. Marine plastic debris provides an artificial surface that rapidly accumulates a dynamic microbial biofilm upon entering the marine ecosystem. Especially the early stages of colonization are critical in shaping the microbial community. This study investigates the early microbial colonization, in less than a week, on five different plastic polymers in Caribbean coastal waters through 16S rRNA gene amplicon sequencing. We discovered shared bacterial taxa among the various plastic polymers and sampling timepoints, with dominant orders being Flavobacteriales, Rhodobacterales, Rhizobiales, and Pseudomonadales. Statistical analysis confirmed significant differences in community composition between the two sampling points, with polystyrene exhibiting a distinct microbial community on day 6 compared to polyethylene, polypropylene, and nylon. We found the same for polyethylene compared to nylon and polyethylene-terephthalate. Further examination identified 47 genera responsible for these differences, primarily belonging to the phyla Proteobacteria and Bacteroidota. Our data indicate an influence of both environmentally related stochastic processes and plastic-related specific factors during early colonization. Interestingly, we noticed an increase in the relative abundance of hydrocarbon and potentially plastic-degrading bacteria (PDB) from 12.4 to 34.5 % between the first and sixth day, suggesting their vital role in shaping the epiplastic community. Notably, some identified PDB have been reported to degrade the specific polymers studied, thus the monitored increase in relative abundance supports their role in plastic degradation. However, more research is required to fully understand their functioning and potential role in the epiplastic community. Our study provides insights into the prokaryotic colonization of marine plastics in the Caribbean basin, where to date studies have been limited despite high pollution rates.

Plastic pollution is increasingly found in agricultural environments, where it contaminates soil and crops. Microbial biofilms rapidly colonise environmental plastics, such as the plastic mulches used in agricultural systems, which provide a unique environment for microbial plastisphere communities. Human pathogens can also persist in the plastisphere, and enter agricultural environments via flooding or irrigation with contaminated water. In this study we examined whether Salmonella Typhimurium and Vibrio cholerae can be transferred from the plastisphere on plastic mulch to the surface of ready-to-eat crop plants, and subsequently persist on the leaf surface. Both S. Typhimurium and V. cholerae were able to persist for 14 days on fragments of plastic mulch adhering to the surface of leaves of both basil and spinach. Importantly, within 24 h both pathogens were capable of dissociating from the surface of the plastic and were transferred onto the surface of both basil and spinach leaves. This poses a further risk to food safety and human health, as even removal of adhering plastics and washing of these ready-to-eat crops would not completely remove these pathogens. As the need for more intensive food production increases, so too does the use of plastic mulches in agronomic systems. Therefore, there is now an urgent need to understand the unquantified co-pollutant pathogen risk of contaminating agricultural and food production systems with plastic pollution.

Plastic pollution in aquatic environments poses significant concerns due to its potential to serve as a refuge for aquatic pathogens. However, the role of plastic surfaces and microbial biofilm interfaces in facilitating pathogen development remains poorly understood. In this study, a microcosm setup was employed to investigate the interactions between plastics and the microbial community and examine the differences in bacterial community composition and potential pathogen occurrences between the plastisphere-biofilm and surrounding seawater. Community composition analysis combined with SEM observations over time indicated that biofilm extracellular polymeric substance formation over 14 days had a link with the relative abundance and succession patterns of pathogen taxa. Colony clusters were observed on biofilms from day 7 and coincided with higher bacterial pathogen dominance. On day 14, pathogen abundance overall decreased with a potentially degrading biofilm. Pseudomonas and Pseudoalteromonas were the dominant potential pathogen groups observed in the microcosm. When further subjected to chemical treatment as an imposed environmental stress over time, biofilm-associated Psuedoalteromonas sharply increased in abundance after three days of exposure, but quickly diminished by 14 days in favor of genera such as Acinetobacter, Pseudomonas, and Staphylococcus. These results suggest that environmental plastisphere-biofilms can promote the early selection, enrichment, and spread of pathogenic bacteria in the aquatic environment and could be later worsened under chemical and long-term pressure. This study provided new insights into the succession of pathogens in plastisphere biofilms, contributing to the understanding of pathogen risks involved in emerging plastisphere biofilms in light of global plastic pollution. Microplastics (MPs) are a significant threat to soils. However, there is scarce information on the impact of MPs on soil properties, particularly in volcanic ashderived soils. The objective of this study was to evaluate the impact of polyamide (PA) and low-density polyethylene (LDPE) MPs on the biological and chemical characteristics of an Andisol from central Chile. Twenty-one parameters were evaluated, including pH, electrical conductivity (EC), total organic carbon (TOC), dissolved organic carbon (DOC), total phosphorus (TP), available phosphorous (AP), available nitrogen (AN), inorganic nitrogen forms (NH4 +, NO2 - and NO3), carbohydrates (CHO), polyphenols (POLs), humic substances, soil basal respiration (SBR) and activities of soil enzymes such as dehydrogenase, ß-glucosidase, phosphatase and urease. For this, a microcosm was set up in clay pots in an incubation chamber at 21°C and 60% soil moisture, with the addition of PA and LDPE at doses of 1% and 3% w/w; a control treatment consisting of microcosm without MPs was also included. After 6 weeks of incubation, PA addition resulted in an increase in TOC and NH4 + by up to 32% and 26%, respectively, and a decrease in NO3 - by 22%. AP decreased by 15%-19% with the addition of PA and LDPE. Similarly, acid phosphatase and ß-glucosidase activities decreased by 15% and 26% with PA and LDPE, respectively. The distance-based linear model (DistLM) was used to analyse relationships in chemical and biological datasets. The analysis revealed that TOC and TP were primary components in the best model for predicting microbiological changes (R2 = .469, AICs = 16.026), indicating that MPs accumulation affects soil carbon cycling and P content. Overall, the results show that MPs addition alters soil chemical and microbiological properties in Andisols, with varying effects depending on the type and dose of MPs, with the highest dose (3%) producing the most marked negative effects. Introduction: Once dispersed in water, plastic materials become promptly colonized by biofilm-forming microorganisms, commonly known as plastisphere. The different plastic materials, either derived from petrochemical hydrocarbons (i.e., LDPE and PET) or biodegradable (PLA and Mb), were used by opportunistic aquatic microorganisms as adhesion surfaces rather than carbon sources. The Mb-associated microorganisms (i.e. mostly members of the family Burkholderiaceae) were likely able to degrade the starch residues on the polymer surfaces, although the Mb matrix maintained its original chemical structure and morphology. Overall, our findings provide insights into the complex interactions between aquatic microorganisms and plastic materials found in lake waters, highlighting the importance of understanding the plastisphere dynamics to better manage the fate of plastic debris in the environment. Microplastics (MPs), which result from the breakdown of plastic waste, have become ubiquitous in various environmental compartments. The "plastisphere", referring to the unique bacterial communities inhabiting plastic debris, includes pathogens and antibiotic resistance genes. Wastewater treatment plants (WWTPs) are hotspots for plastisphere formation, but significant releases of MPs still occur. This study investigates the microbial communities on polystyrene (PS) MPs through in situ deployment across primary, secondary, and tertiary WWTP stages. Biofilms formed on the PS MPs exhibited greater bacterial diversity than background waters. Certain genera acted as pioneers in the biofilms, attracting and facilitating the accumulation of other microbes from background waters. The biofilms formed on the MPs became more resistant to treatment processes compared to freely floating bacteria. This study sheds light on the evolution of microbial communities on MPs within WWTPs and their roles as carriers of microbes in effluents, with implications for environmental and public health. Microplastic pollution has become a global environmental challenge, with significant impacts on ecosystems and human health. Microbes have emerged as a promising tool in the combating against microplastic contamination. However, the complex relationship between microbes and microplastics presents both opportunities and challenges, leading to a nuanced understanding of their applications in degradation. This paper provides critical insights into the multifaceted roles of different microorganisms in microplastic degradation. It begins by highlighting the ‘good’ aspects, where several strains of microorganisms show the potential to break down microplastics through enzymatic activities and the formation of biofilms. Conversely, the ‘bad’ aspects of microbial involvement in microplastic degradation are examined. Microorganisms can facilitate the transport and bioaccumulation of microplastics in various ecosystems, potentially exacerbating their harmful effects. The ‘ugly’ side of microplastic degradation includes the production of harmful byproducts during microbial breakdown, raising concerns about secondary pollution and toxicity. The concept of plastisphere is discussed in this context, focusing on the phototrophs, photoheterotrophs and heterotrophs. Novel technologies involving microbial degradation of microplastics are also explained. The work emphasises the need for a comprehensive and balanced approach regarding the application of microorganisms in microplastic degradation and remediation.

Plastic waste has become a significant global ecological concern due to its substantial harmful impacts. They are eventually colonized and transformed by diverse microbial communities known as the ‘‘plastisphere”. Only a limited group of microbes have been identified so far due to the prevalence of non-culturable strains that inhabit polymer substrates. We investigated the community diversity and predictive functionality of the plastisphere microbiota using a high-throughput next-generation sequencing (NGS) approach to unveil their plastic degrading potential. Samples were collected from four different plastic-enriched disposal sites adjacent to Kolkata, West Bengal, India, followed by processing, gDNA extraction, library preparation, and sequencing. The most prevalent phyla were Proteobacteria, followed by Bacteroidetes, Actinobacteria, Cyanobacteria, Firmicutes, and Verrucomicrobia. The functional profiling revealed that genes associated with metabolism, cellular processes, and signalling were most prevalent, followed by poorly characterized information storage and processing. In this study, we predicted the existence of a beneficial microbiome associated with bioremediation and plastic degradation, which suggests the potential utilization of plastics as primary carbon sources. Our findings also highlighted the existence of promising microbial enzymes associated with the biodegradation of several plastic substrates.Furthermore, our research unveiled the enriched distribution of beneficial microbiome in the studied metagenome, which offers a diverse prospect. This investigation establishes a connection between the structure of microbial communities and diverse genes actively engaged in the biodegradation of plastic waste within plastic disposal sites. These beneficial degraders can be investigated further for broad-spectrum applications in plastic bioremediation. Subsequent explorations of their plastic degrading enzymes will provide profound contributions to plastic pollution mitigation.

With a growing number of research reports on microplastics (MPs), there is increasing concern regarding MPs-induced contamination in soil ecological systems. Notwithstanding, the interaction between the plastisphere and rhizosphere microbial hotspots in soil-plant systems, as well as the diversity and composition of plastisphere microbial communities in such systems, remain largely unexplored. This study evaluated the response of rhizosphere bacterial communities to MPs at three growth stages of pepper and examined the bacterial communities present on MPs (plastisphere). The 16 S rRNA revealed that, under the stress of MPs, the Chao1 and Shannon index of the pepper soil bacterial community decreased. Meanwhile the relative abundance of Actinobacteriota was decreased, and that of Proteobacteria was increased. Furthermore, the plastisphere serves as a unique microbial habitat (niche) that recruits the colonization of specific bacterial groups, including potential plastic-degrading bacteria and potential pathogens (e.g., Massilia and Pseudomonas). Simultaneously, the plastisphere recruits specific bacteria that may impact the rhizosphere soil bacterial communities, thus indirectly affecting plant growth. Functional prediction using PICRUSt2 revealed higher activity in the plastisphere for Metabolism of terpenoids and polyketides, Human diseases, and Xenobiotics biodegradation and metabolism. Notably, the human diseases metabolic pathway exhibited increased activity, suggesting potential ecological risks associated with pathogens. These results highlighted that the plastisphere serves as a unique microbial habitat (niche) in the soil ecological systems, recruiting specific bacteria and potentially interfering with the surrounding soil microbial community, thereby influencing the functional characteristics of the soil ecological systems.

Biodegradable plastics (BDPs) have attracted extensive attention as an alternative to conventional plastics. BDPs could be mineralized by composting, while the quality of compost affected by the presence of BDPs and the residual microplastics (MPs) has not been well evaluated. This study aimed to explore the MPs release potential and environmental implications of commercial BDPs (aromatic−aliphatic copolymer) films in uncontrolled composting. Results showed that the molecular weight of BDPs decreased by >60% within 60 d. However, the non-extracted organic matter and wet-sieving measurements indicated that MPs continuously released and accumulated during regular composting. The average MPs release potential (0.1–5 mm) was 134.6 ± 18.1 particles/mg (BDPs), which resulted in 103–104 particles/g dw in compost. The plastisphere of MPs showed a significantly higher (0.95–16.76 times) abundance of antibiotic resistance genes (ARGs), which resulted in the rising (1.34–2.24 times) of ARGs in compost heaps, in comparison to the control groups. Overall, BDPs promote the spread of ARGs through the selective enrichment of bacteria and horizontal transfer from released MPs. These findings confirmed that BDPs could enhance the release potential of MPs and the dissemination of ARGs, which would promote the holistic understanding and environmental risk of BDPs.

Plastic waste accumulation in marine environments has complex, unintended impacts on ecology that cross levels of community organization. To measure succession in polyolefin-colonizing marine bacterial communities, an in situ time-series experiment was conducted in the oligotrophic coastal waters of the Bermuda Platform. Our goals were to identify polyolefin colonizing taxa and isolate bacterial cultures for future studies of the biochemistry of microbe- plastic interactions. HDPE, LDPE, PP, and glass coupons were incubated in surface seawater for 11 weeks and sampled at two-week intervals. 16S rDNA sequencing and ATR-FTIR/HIM were used to assess biofilm community structure and chemical changes in polymer surfaces. The dominant colonizing taxa were previously reported cosmopolitan colonizers of surfaces in marine environments, which were highly similar among the different plastic types. However, significant differences in rare community composition were observed between plastic types, potentially indicating specific interactions based on surface chemistry. Unexpectedly, a major transition in community composition occurred in all material treatments between days 42 and 56 (p < 0.01). Before the transition, Alteromonadaceae, Marinomonadaceae, Saccharospirillaceae, Vibrionaceae, Thalassospiraceae, and Flavobacteriaceae were the dominant colonizers. Following the transition, the relative abundance of these taxa declined, while Hyphomonadaceae, Rhodobacteraceae and Saprospiraceae increased. Over the course of the incubation, 8,641 colonizing taxa were observed, of which 25 were significantly enriched on specific polyolefins. Seven enriched taxa from families known to include hydrocarbon degraders (Hyphomonadaceae, Parvularculaceae and Rhodobacteraceae) and one n-alkane degrader (Ketobacter sp.). This so-called ‘plastisphere’ community can also include human pathogens, particularly if the plastic has been exposed to faecal contamination (e.g., from wastewater or organic manures and livestock faeces). The plastisphere is hypothesised to facilitate the survival and dissemination of pathogens, and therefore plastics in agricultural systems could play a significant role in transferring human pathogens to crops, particularly as microplastics adhering to ready to eat crops are difficult to remove by washing. In this paper we critically discuss the pathways for human pathogens associated with microplastics to interact with crop leaves and roots, and the potential for the transfer, adherence, and uptake of human pathogens from the plastisphere to plants. In a paradigm shift in plastic wastes due to the COVID-19 pandemic, wetlands such as mangroves are threatened by a new form of pollution, plastics, on top of the eutrophication of estuarine waters due to nitrogen and phosphorus wastes/effluents that lead to cyanobacterial proliferation. Both plastic and nutrient pollution lead to prosperity of cyanotoxin-producing cyanobacteria that flourish in both and disperse leading to the detriment of fauna and flora in the mangrove ecosystem due to resulting toxicities. Although cyanotoxins are still a relatively poorly studied phenomenon in mangroves, their presence does create a focus of attention due to biofilm formation and the resultant flotation and sinking properties that are linked to cyanobacterial mats on plastic debris. Sri Lanka, being the first country in the world to conserve all its mangrove wetlands, does have a responsibility to prevent the invasion of plastics to this protected ecosystem, and binding with the Ramsar Convention, precluding plastic waste and their concomitant footprint, is a task at hand to the relative authorities. The path ahead mandates that we study the properties of plastics for cyanobacterial proliferation, biofilm formation, the fates of such plastics (flotation, dispersal and sinking), the cyanotoxin production changes that are attributed—or linked—to plastic pollution and the resultant impacts on mangrove ecosystems. Cyanotoxins are long-lived, and it is paramount that we find the necessary mechanisms to eliminate or curtail their production in mangrove ecosystems while establishing surveillance and monitoring of both the producers and the harmful agents. Cyanobacteria although vehicles for nitrogen fixation and replenishing of nutrients to an N-depleted ecosystem such as the mangroves, could lead to enhancements in cyanotoxins production. However, this phenomenon remains ambiguous and poorly studied in applied phycology in relation to mangroves. “New normal” plastics are lodged mostly on the surfaces of bark, prop roots, and pneumatophores, which are the localities where the highest level of new nitrogen is fixed, and this may lead to the proliferation of N-fixing, cyanotoxin-producing cyanobacteria, which may have repercussions on both flora and fauna of mangroves. Therefore, it is crucial that we monitor plastic pollution and find mechanisms for sanitizing plastics-imprinted mangroves to lessen the harmful footprint resulting from plastic overload.

Coastal habitats have been suggested to serve as a sink for unaccounted plastic debris, i.e., “missing plastic” in the sea, and hence, a hotspot of plastic pollution in the marine and coastal environments. Although the accumulation of plastic debris may pose significant threats to coastal ecosystems, we know little about the fate of these plastic debris and their ecological impacts due to the lack of studies on plastic-microbe interactions in coastal habitats, especially for the tropical marine and coastal environments. In this study, we collected plastic debris from 14 sites consisting of various coastal ecosystems (seagrass meadows, mangrove forests, and beaches), and marine ecosystem (coral reef) around Singapore and characterized the prokaryotic and eukaryotic microbial communities colonized on them. Our results showed that the composition of plastisphere communities in these intertidal ecosystems was predominantly influenced by the sediment than by the plastic materials. Compared with surrounding sediment and seawater, the plastic debris enriched potential plastic degraders, such as Muricauda, Halomonas, and Brevundimonas. The plastic debris was also found to host taxa that play significant roles in biogeochemical cycles (e.g., cyanobacteria, Erythrobacter), hygienically relevant bacteria (e.g., Chryseobacterium, Brevundimonas), and potential pathogens that may negatively impact the health of coastal ecosystems (e.g., Thraustochytriaceae, Labyrinthulaceae, Flavobacterium). Taken together, our study provides valuable insights into the plastic-microbe interactions in tropical coastal and marine ecosystems, highlighting the urgent need for plastisphere studies to understand the fate and ecological impacts of plastic debris accumulated in coastal habitats.

The polyethylene (PE) film mulching as a water conservation technology has been widely used in dryland agriculture, yet the long-term mulching has led to increasing accumulation of secondary pollutants in soils. The decomposition of PE film-sourced pollutants is directly associated with the enrichment of specific bacterial communities. We therefore hypothesized that plant biomass may act as an organic media to mediate the pollutant decomposition via reshaping bacterial communities. To validate this hypothesis, plant biomass (dried maize straw and living clover) was embedded at the underlying surface of PE film, to track the changes in the composition and function of bacterial communities in maize field across two years. The results indicated that both dry crop straw and alive clover massively promoted the α-diversity and abundance of dominant bacteria at plastisphere, relative to bulk soil. Bacterial communities tended to be clustered at plastisphere, forming the bacteria islands to enrich pollutant-degrading bacteria, such as Sphingobacterium, Arthrobacter and Paracoccus. The microbial colonization of plastic substrates made of polyvinylchloride (PVC) and polyethylene (PE) was studied in Tethys and Road Bays (Ross Sea, Antarctica) in order to evaluate the metabolic profiles of the plastisphere community in comparison with those of the surrounding waters. PVC and PE panels, mounted on stainless steel structures, were deployed in the austral summer 2017 at 5 and 20 m and recovered one year later at four different stations (Amorphous Glacier-AG was potentially impacted by the ice-melting process, and its control site was within Tethys Bay-TB; Road Bay-RB, close to the wastewater plant of the Italian research station Mario Zucchelli and its control site Punta Stocchino-PTS). Additional panels were settled in Road Bay at 5 m and recovered after three months to follow time variability in the microbial colonization process. At the same times and depths as plastic substrates, water samples were also collected. Carbon substrates' utilization rates were determined on scraped microbial biofilm and water samples, with a fluorimetric assay based on 96-well Biolog Ecoplates. Complex carbon sources, carbohydrate and amines were the organic substrates that mostly fuelled the community metabolism in the RB area, while in the TB area, in addition to carbohydrates, phosphate carbon compounds and amino acids were also actively utilized. Within Road Bay, small differences in the physiological profiles were found, with higher metabolic rates in the biofilm community after 3 months' deployment (late austral summer period) compared to 12 months, suggesting that autumn to spring period conditions negatively affected foulers' metabolism. Moreover, different metabolic profiles between the plastisphere and the pelagic microbial community were observed; this last utilized a higher number of carbon sources, while plastic substrates were colonized by a more specialized community. Higher carbon substrate utilization rates were recorded at RB and AG stations, receiving organic supply from anthropic activity or ice melting sources, respectively, compared to their control sites. Plastics are accumulating in the world's oceans, while ocean waters are becoming acidified by increased CO2. We compared metagenome of biofilms on tethered plastic bottles in subtidal waters off Japan naturally enriched in CO2, compared to normal ambient CO2 levels. Extending from an earlier amplicon study of bacteria, we used metagenomics to provide direct insights into changes in the full range of functional genes and the entire taxonomic tree of life in the context of the changing plastisphere. We found changes in the taxonomic community composition of all branches of life. This included a large increase in diatom relative abundance across the treatments but a decrease in diatom diversity. Network complexity among families decreased with acidification, showing overall simplification of biofilm integration. With acidification, there was decreased prevalence of genes associated with cell–cell interactions and antibiotic resistance, decreased detoxification genes, and increased stress tolerance genes. There were few nutrient cycling gene changes, suggesting that the role of plastisphere biofilms in nutrient processes within an acidified ocean may not change greatly. Our results suggest that as ocean CO2 increases, the plastisphere will undergo broad-ranging changes in both functional and taxonomic composition, especially the ecologically important diatom group, with possible wider implications for ocean ecology. Numerous microorganisms and other invertebrates that are able to degrade polyethylene (PE) have been reported. However, studies on PE biodegradation are still limited due to its extreme stability and the lack of explicit insights into the mechanisms and efficient enzymes involved in its metabolism by microorganisms. In this review, current studies of PE biodegradation, including the fundamental stages, important microorganisms and enzymes, and functional microbial consortia, were examined. Considering the bottlenecks in the construction of PE-degrading consortia, a combination of top-down and bottom-up approaches is proposed to identify the mechanisms and metabolites of PE degradation, related enzymes, and efficient synthetic microbial consortia. In addition, the exploration of the plastisphere based on omics tools is proposed as a future principal research direction for the construction of synthetic microbial consortia for PE degradation. One of the most discussed issues in environmental science and society is plastic pollution of natural ecosystems and its impact on living organisms. This is evidenced by the flow of numerous publications that have appeared in recent years. The results of the studies confirm that the pollution of aquatic ecosystems with plastic waste is considered one of the most severe global environmental problems. The wide distribution of photooxidation and biological degradation products of micro- and nano-sized plastic particles (MP and NP) in the World Ocean is of particular concern, which make the main contribution to biological effects. Catastrophic environmental consequences of marine ecosystems contamination with microplastics are associated not only with economic damage, but also with human health and the biosecurity of mariculture. Human-made plastisphere is a new ecosystem that plays an increasingly significant role in the life of marine microorganisms. Biofilms formed on the rough and hydrophobic surfaces of the plastisphere consist of various taxa of microorganisms and differ in composition from the surrounding marine communities. In addition, they act as depositories and carriers of intra- and extracellular antibiotic resistance genes (ARGs). The purpose of the review is to summarize current information on environmental pollution by microplastics in the context of its role in the conservation and spread of pathogenic bacteria and antibiotic resistance genes in natural environments with an emphasis on marine ecosystems.

AbstractPlastic wastes have a negative impact on marine environments; however, they can be used as carbon sources and habitats by certain microbes. Microbes in the marine plastisphere can migrate worldwide through the ocean and cause serious environmental problems when they encounter suitable environments. Therefore, efforts to investigate the microbes inhabiting the marine plastisphere are increasing. In the present study, fungal strains were isolated from plastic wastes buried in Korean sea sands and mudflats and identified using molecular and morphological analyses. Five species were identified that were previously unrecorded from South Korea: Cladosporium funiculosum, Neosetophoma poaceicola, Neosetophoma rosigena, Parasarocladium gamsii, and Trichoderma fomiticola. Their molecular phylogenies and morphological characteristics are described in this study.

Plastics are associated with a worldwide pollution crisis, with strong negative impacts on both terrestrial and aquatic ecosystems. In marine environments, various organisms are colonizing plastic debris, but few studies have focused on fungal communities despite their non-trivial ecological roles in the marine environment. In this study, different types of plastics (biodegradable and conventional) immersed in marine natural environments and under laboratory controlled settings were collected after long-term colonization. Using a metabarcoding approach targeting two genetic markers, namely, the ITS2 region and the V4 hypervariable region of the 18S rRNA gene, we highlighted that fungal communities associated with plastic polymers were distinct from those found in the surrounding seawater. They also differed significantly between sampling locations and the nature of immersed polymers, indicating that fungal colonization was impacted by the sites and types of plastics, with clear dissimilarities between conventional and biodegradable polymers. Specifically for the conventional PVC polymer (Polyvinyl chloride), we also observed the successive stages of biofilm development and maturation after long-term immersion in seawater. A noticeable change in the fungal communities was observed around 30–40 days in natural settings, suggesting a colonization dynamic likely associated with a transition from biofilm formation to distinct communities likely associated with biofouling. Complex microbial communities colonize plastic substrates over time, strongly influencing their fate and potential impacts on marine ecosystems. Among the first colonizers, diatoms play an important role in the development of this 'plastiphere'. We investigated 936 biofouling samples and the factors influencing diatom communities associated with plastic colonization. These factors included geographic location (up to 800 km apart), duration of substrate submersion (1 to 52 weeks), plastics (5 polymer types) and impact of artificial ageing with UV light. Diatom communities colonizing plastic debris were primarily determined by their geographic location and submersion time, with the strongest changes occurring within two weeks of submersion. Several taxa were identified as early colonizers (e.g. Cylindrotheca, Navicula and Nitzschia spp.) with known strong adhesion capabilities. To a lesser extent, plastic-type and UV-ageing significantly affected community composition, with 14 taxa showing substrate-specificity. In this study, we examined the toxicity potential of the epiplastic microalgal community that developed on low-density polyethylene (LD-PE) plastic debris found in two distinct regions of the Monastir Bay (Tunisia): the coast exposed to anthropogenic discharges and the open sea in front of the Kuriat Islands. Concentrations of potentially toxic elements (PTEs) accumulated in sediments and plastisphere were compared in order to determine their toxicity potential to biological life. The collected plastispheres were predominantly composed of cyanobacteria, chlorophytes, and diatoms. Diatoms display a relatively high diversity (25 species). At all stations, potentially harmful microalgae (PHM) were more abundant in the plastisphere than in seawater and the coastal zone seems to harbour increased number of potentially harmful cyanobacteria within the plastisphere. At the offshore station S1, the PHM community was dominated by the potentially harmful diatoms belonging to the genus Pseudo-nitzschia. Phormidium sp. was the main potentially harmful cyanobacterium identified in the plastisphere of S1. PTEs concentration in the plastisphere was higher than in sediment and ranking with very high contamination factors at all sites according to the sequence Pb > Cu > Cd > Ni > Zn. The highest accumulation of PTEs in the plastisphere was recorded near harbors and industrial zones with important human interference. This work shows that plastisphere can be a threat to vulnerable species not only because it can contain PHM but also because it can accumulate PTEs. [ABSTRACT FROM AUTHOR]
