CXCL12 and CXCR4 proteins and mRNAs were monitored in the dorsal root ganglia (DRGs) of lumbar (L4–L5) and cervical (C7–C8) spinal segments of naïve rats, rats subjected to sham operation, and those undergoing unilateral complete sciatic nerve transection (CSNT) on post-operation day 7 (POD7). Immunohistochemical, Western blot, and RT-PCR analyses revealed bilaterally increased levels of CXCR4 protein and mRNA in both lumbar and cervical DRG neurons after CSNT. Similarly, CXCL12 protein levels increased, and CXCL12 mRNA was upregulated primarily in lumbar DRGs ipsilateral to the nerve lesion. Intrathecal application of the CXCR4 inhibitor AMD3100 following CSNT reduced CXCL12 and CXCR4 protein levels in cervical DRG neurons, as well as the length of afferent axons regenerated distal to the ulnar nerve crush. Furthermore, treatment with the CXCR4 inhibitor decreased levels of activated Signal Transducer and Activator of Transcription 3 (STAT3), a critical transforming factor in the neuronal regeneration program. Administration of IL-6 increased CXCR4 levels, whereas the JAK2-dependent STAT3 phosphorylation inhibitor (AG490) conversely decreased CXCR4 levels. This indicates a link between the CXCL12/CXCR4 signaling axis and IL-6-induced activation of STAT3 in the sciatic nerve injury-induced pro-regenerative state of cervical DRG neurons. The role of CXCR4 signaling in the axon-promoting state of DRG neurons was confirmed through in vitro cultivation of primary sensory neurons in a medium supplemented with CXCL12, with or without AMD3100. Purpose of Review: Describe the rationale for preconditioning MSCs prior to use as therapy and the state-of-the-art of using preconditioning of MSCs in clinical settings. Recent Findings: Mounting preclinical data supports preconditioning of mesenchymal stromal cells (MSCs) to enhance their therapeutic efficacy. Most research has focused on cytokine priming and hypoxic preconditioning, while other approaches, such as glycoengineering, remain relatively understudied. Despite strong preclinical data, clinical evidence supporting preconditioning strategies are limited to six Phase I clinical trials (most of them in progress). Summary: Here, we succinctly discuss the rationale for preconditioning using cytokines, hypoxia, and glycoengineering, while elaborating on the respective clinical experiences. Overall, we note that preconditioning is highly dependent on the desired application, and therefore requires elucidating the mechanism of action of the MSCs used for therapy. Preconditioning may also help mitigate heterogeneity of MSC lots. Based on the remarkable safety profile of MSCs, even when used in allogeneic settings, the role of preconditioning prior to their final formulation might be the key to reach expected therapeutic outcomes.

Canola (Brassica napus L. var. oleifera Moech) is one of the main oilseeds for edible and industrial purposes. For the evaluation of the physiological quality of a lot of seeds, the tetrazolium test has been considered as a promising alternative, due to the speed and efficiency in the characterization of viability. The present study aimed to adapt the methodology of the tetrazolium test to evaluate the quality of canola seeds. Alth B4 and Hyola 433 hybrids were used. The study involved the characterization of the initial profile of the lots by determining the moisture content, weight of a thousand seeds, first count, germination test, germination speed index, emergence, initial stand, emergency speed index. Pre-conditioning of the seeds was carried out, defining the soaking curve and the seeds placed to soak for periods of 10, 12, 14, 16, 18 and 22 hours, by the methods of hydration on paper and direct immersion in water and the adequacy of the tetrazolium test methodology as a function of different concentrations (0.075%; 0.5% and 1.0%) and immersion periods of 2, 4 and 6 hours. The results indicated that the lowest concentrations and lowest imbibition periods present a higher percentage of viability for canola seeds. It was concluded that the tetrazolium test is efficient to evaluate the viability in canola seeds with pre-conditioning of the seeds in hydration in paper for 10 hours at 25 °C, followed by the total removal of the tegument, in the tetrazolium solution of 0.075% per 2 hours at 30°C. What is the role of soil moisture in maintaining the continental tropical convergence zone (CTCZ) during the active phase of the monsoon? This question has been addressed in this study using soil moisture datasets from ERA5 reanalysis and is validated with other datasets. The analysis shows that, like rainfall, soil moisture also shows prominent intraseasonal oscillation. Furthermore, the sub-seasonal and seasonal features of soil moisture are different from each other. During the summer monsoon season, the seasonal mean maximum soil moisture is found over western coastal regions, central parts of India, and the northeastern Indian subcontinent. However, during the sub-seasonal active phases of the monsoon, the maximum positive soil moisture anomaly was found in northwestern parts of India. Our analysis also suggests soil moisture plays a pre-conditioning role during active phases of the monsoon over the monsoon core zone of India. It is found that the pre-conditioning depends on that region's soil type (texture) and climate patterns. Analysis of the moist static energy (MSE) budget during the active phases shows that soil moisture feedback affects the boundary layer MSE and rainfall. We also evaluate the pre-conditioning feedback in the Climate Forecast System (CFS) model-free run to understand the well-known CFS model rainfall bias. CFS runs do not show the realistic pre-conditioning role of soil moisture and its feedback on the rainfall as in observations. Thus, the rainfall dry bias in CFS operational forecasts may be partly due to this error in soil moisture-rainfall feedback. Another important conclusion from this study is that the soil texture has an important role in changing the soil–moisture and rainfall intraseasonal pattern over the region. Using a WRF regional model experiment, we find that the pre-conditioning of soil moisture over sandy soil texture leads to increased precipitation during the active phase of the monsoon. Pre-exercise passive heating attenuates muscle damage caused by eccentric exercise in rats where the induction of heat shock proteins (HSPs) confers a myoprotective effect. We investigated whether pre-exercise hot water immersion (HWI) confers similar benefits in humans. Eleven recreational male athletes were immersed in 41°C water up to 60 min or until rectal temperatures reached 39.5°C. After a 6 h rest, the participants performed an eccentric downhill run for 1 h at −4% gradient to induce muscle damage. An endurance capacity test at 75% VO2max was conducted 18 h later. The control trial was similar except that participants were immersed at 34°C. Blood samples were collected to assess HSPs levels, creatine kinase, and lactate dehydrogenase activities. Plasma eHSP70 was higher post-immersion in HWI trials (1.3 ± 0.4 vs 1.1 ± 0.4; p = 0.005). Plasma eHSP27 was higher before (p = 0.049) and after (p = 0.015) endurance test in HWI. Leukocytic p-HSP27 was increased 18 h after HWI (0.97 ± 0.14 vs 0.67 ± 0.11; p = 0.04). Creatine kinase and lactate dehydrogenase activities were increased by 3-fold and 1.5-fold, respectively, after endurance test in HWI but did not differ across trials (p > 0.05). Mean heart rates were higher during eccentric run and endurance test in HWI as compared to control (p < 0.05). Endurance capacity was similar between trials (57.3 ± 11.5 min vs 55.0 ± 13.5 min; p = 0.564). Resource fluxes at the stream–riparian interface are a vital contributor to both systems' energy budgets. The effect of distinct litter exposure patterns—direction of the riparia–stream movement and duration of exposure at each habitat—however, remains to be elucidated. In this field experiment, oak leaves in fine and coarse mesh bags were either exposed to a stream-to-riparia or riparia-to-stream movement sequence for distinct periods (2:6, 4:4, or 6:2 weeks). After 8 weeks, ash-free mass loss, microbial activity, and fungal biomass were compared in leaves undergoing inverse movement sequences (e.g., 2-week exposure to the riparian area at the beginning vs. end of the colonization period). Mass loss in coarse mesh bags was negatively affected when leaves were previously exposed to a short (2 weeks) terrestrial pre-conditioning period, despite higher microbial activity and fungal biomass, when compared to the inverse movement. This effect on mass loss was neutralized by longer terrestrial exposures that likely allowed for a more thorough conditioning of the leaves, through extended leaching and terrestrial microbial colonization. Our results suggest that terrestrial pre-conditioning periods of < 2 weeks lead to litter-quality legacy effects in tough leaves, to which aquatic communities respond through lower substrate degradation efficiency, hindering stream decomposition. Contrastingly, oak aquatic pre-conditioning, regardless of duration, provides riparian communities with a high-quality resource, promoting litter processing through grazing behavior. As climate-induced hydrological shifts may result in altered provision/quality of detritus subsidies at the stream–riparia interface, we suggest that assessments of decomposition dynamics should consider the entire litter conditioning history. Abstract Mesenchymal stem/stromal cells (MSCs)‐based therapy brings the reassuring capability to regenerative medicine through their self‐renewal and multilineage potency. Also, they secret a diversity of mediators, which are complicated in moderation of deregulated immune responses, and yielding angiogenesis in vivo. Nonetheless, MSCs may lose biological performance after procurement and prolonged expansion in vitro. Also, following transplantation and migration to target tissue, they encounter a harsh milieu accompanied by death signals because of the lack of proper tensegrity structure between the cells and matrix. Accordingly, pre-conditioning of MSCs is strongly suggested to upgrade their performances in vivo, leading to more favored transplantation efficacy in regenerative medicine. Indeed, MSCs ex vivo pre-conditioning by hypoxia, inflammatory stimulus, or other factors/conditions may stimulate their survival, proliferation, migration, exosome secretion, and pro-angiogenic and anti-inflammatory characteristics in vivo. In this review, we deliver an overview of the pre-conditioning methods that are considered a strategy for improving the therapeutic efficacy of MSCs in organ failures, in particular, renal, heart, lung, and liver.

Machinability of Ti-6Al-4V, despite its numerous applications, is challenging owing to its poor thermal conductivity, severe chemical affinity and strain hardening characteristics. Although various coating techniques and expensive lubrication strategies have been tried, a significant improvement in tool life characteristics are yet to be achieved. Hence this work aims at suggesting a novel cost-effective and ecofriendly tool treatment technique by exploring the impact of a dual combination of an AlSi-10Mg coating developed by an emerging pre-conditioning technique with the incorporation of micro-textures on tool face to observe significant improvements in tool life and machinability characteristics of the alloy. It was analyzed from the results that while the micro-textures played a vital role in reducing the severe contact stress and machining temperature due to its improved heat transfer by convection, AlSi-10Mg film effectively shielded the cutting edges of the tool by providing a dampening effect along the edges, thereby ensuring improved tool life and enhanced machinability characteristics at the same time. The proposed tool combination was observed to be successful in significantly reducing the flank wear by 35%, 43% and 30% where 75, 253, and 371 µm were observed at 80, 100, and 120 m/min as compared to 117, 449, and 531 µm in the case of uncoated non-textured tool at the same operating conditions. Surface roughness of Ti-6Al-4V also reduced by 25%, 34%, and 37% where 1394, 1168, and 872 nm as compared to plain tool where 1840, 1770, and 1370 nm were recorded. Moreover, average chip curl diameter was observed to be 2.62, 3.36, and 6.49 mm as compared to chips generated by uncoated tools which recorded values of 4.33, 11.9, and 18.87 mm at 80, 100, and 120 m/min respectively. This study presents a novel approach to rock pre-conditioning to promote the sustainability of low-grade ore mining applications such as in-situ recovery and cave mining. The proposed method involves a two-stage hybrid approach, utilizing soundless cracking demolition agents (SCDAs) to initiate radial fractures in a predrilled host rock, followed by hydraulic stimulation to extend the fractures. SCDA injection in the first stage creates multiple radial fractures around the injection well. However, the extent of fractures is limited to the near vicinity of the injection well. To overcome this limitation, the second stage involves the application of hydraulic stimulation to extend the initiated fractures, which produces a greater fracture density compared to pure hydraulic stimulation. The concept was assessed using a fully coupled hydro-mechanical discrete element model that simulated the hybrid fracturing method on crystalline rock at the grain scale. The results indicate that the proposed method can create a high density of fractures around the injection well. Additionally, we identify and evaluate the key factors affecting the performance of the proposed method, including rock mass heterogeneity, stress anisotropy, and pre-existing defects, providing valuable insights for further experimental design and execution. Overall, the study offers promising results for a potential solution to enhance the efficiency of low-grade ore mining through the hybrid rock pre-conditioning method. In multiuser massive Multiple Input Multiple Output (MIMO) systems, a large amount of antennas are deployed at the Base Station (BS). In this case, the Minimum Mean Square Error (MMSE) detector with soft-output can achieve the near-optimal performance at the cost of a large-scale matrix inversion operation. The optimization algorithms such as Gradient Descent (GD) method have received a lot of attention to realize the MMSE detection efficiently without a large scale matrix inversion operation. However, they converge slowly when the condition number of the MMSE filtering matrix (the coefficient matrix) increases, which can compromise the efficiency of their implementation. Moreover, their soft information computation also involves a large-scale matrix-matrix multiplication operation. In this paper, a low-complexity soft-output signal detector based on Adaptive Pre-conditioned Gradient Descent (APGD-SOD) method is proposed to realize the MMSE detection with soft-output for uplink multiuser massive MIMO systems. In the proposed detector, an Adaptive Pre-conditioner (AP) matrix obtained through the Quasi-Newton Symmetric Rank One (QN-SR1) update in each iteration is used to accelerate the convergence of the GD method. The QN-SR1 update supports the intuitive notion that for the quadractic problem one should strive to make the pre-conditioner matrix close to the inverse of the coefficient matrix, since then the condition number would be close to unity and the convergence would be rapid. By expanding the signal model of the massive MIMO system and exploiting the channel hardening property of massive MIMO systems, the computational complexity of the soft information is simplified. The proposed AP matrix is applied to the GD method as a showcase. However, it also can be used by Conjugate Gradient (CG) method due to its generality. It is demonstrated that the proposed detector is robust and its convergence rate is superlinear. Simulation results show that the proposed detector converges at most four iterations. Simulation results also show that the proposed approach achieves a better trade-off between the complexity and the performance than several existing detectors and achieves a near-optimal performance of the MMSE detector with soft-output at four iterations without a complicated large scale matrix inversion operation, which entails a big challenge for the efficient implementation.

The therapeutic effectiveness of stem cells after transplantation is hampered by the hypoxic milieu of chronic wounds. Prior research has established antioxidant priming as a thorough plan to improve stem cell performance. The purpose of this study was to ascertain how caffeic acid (CA) priming affected the ability of human adipose-derived stem cells (hASCs) to function under hypoxic stress. In order to study the cytoprotective properties of CA, hASCs were primed with CA in CoCl2 hypoxic conditions. Microscopy was used to assess cell morphology, while XTT, Trypan Blue, X-gal, LDH, Live Dead, scratch wound healing, and ROS assays were used to analyze viability, senescence, cell death, proliferation, and reactive oxygen species prevalence in the cells. According to our findings, CA priming enhances hASCs' ability to survive and regenerate in a hypoxic microenvironment more effectively than untreated hASCs. Our in-vitro research suggested that pre-treatment with CA of hASCs could be a unique way to enhance their therapeutic efficacy and ability to survive in hypoxic microenvironments.

Carbonation curing cement-based materials can improve the properties of cement products while permanently sequestering CO2, which is considered to be a very promising carbon reduction technology. In this paper, cement pastes with different pore structures were prepared by changing the water-cement ratio (w/c =0. 35, w/c =0. 4, w/c =0. 45) and pre-conditioning time (2, 4, 6, 8, 16, 24, 48 and 72 h), and the influences of pore structure and water content on the carbonation reaction of cement paste were studied. The results show that the carbon sequestration rate of cement paste with different water-cement ratios reaches the maximum when the pre-conditioning time is 8 h, and the carbon sequestration rate increases with the increase of water-cement ratio. At the same time, for cement paste with early hydration (24 h standard curing +8 h pre-conditioning), the influence of water-cement ratio on the pore structure is much greater than the degree of hydration. The cement paste with initial water-cement ratio of 0. 45 undergoes 8 h pre-conditioning and then undergoes 8 h carbonization curing. Its carbon sequestration rate, carbonation depth and compressive strength reach 20. 35% (mass fraction), 12. 49 mm and 61. 99 MPa, respectively, which increases the compressive strength by 14. 27% compared with the strength of the 28 d standard curing test block (54. 25 MPa). The main products of carbonation reaction are CaCO3 and silica gel. The filling of CaCO3 optimizes the pore structure of cement paste, significantly reduces the pores larger than 50 nm, and the main crystal forms of CaCO3 are calcite and aragonite. This study analyzes the fracture patterns generated from the high-energy release caused by commercially available explosives and the current capability of numerical methods to replicate this. The mechanics of rock fracture and fragmentation are first studied using the Hybrid Stress Blasting Model (HSBM) at the lab-scale through comparison with experimental results available in the literature. Following this, an in-house experimental blast campaign was undertaken with a detailed examination of an unconfined, 605 mm diameter cylindrical sample and a pressurised (confined), 700 mm diameter cylindrical sample. The post-blast samples were dissected and fractures were visually mapped before comparing the fracture intensity results to the output of the HSBM, which captures stress-wave-induced damage but not gas loading, for these test cases. This paper makes contributions to the literature in three fundamental areas. Firstly, the capability of numerical methods to capture the phenomenology of blasting on rock fracture and fragmentation was validated and the limitations were discussed when looking to use this as a design tool for practical blasting operations. Secondly, the experimental campaign provides detailed insights into the response of cementitious grout materials to internal, blast-induced loading that can be applied in various fields such as mining and construction. Finally, the impact of confining pressure on blast damage was investigated and details were provided for how this can be captured in numerical predictions. It was found that certain aspects of the material response can be well predicted through numerical analysis, namely, the damage radius and the number of dominant fractures. However, it also indicated a shortcoming in the explicit comparison of fracture intensity measures such as P21, with the use of damage metrics appearing more appropriate. The grout-based testing methodology discussed in this work provides an efficient means for gathering data on the impacts of individually controlled aspects of a blast in both unconfined, and confined environments. The permeability of rocks is important in a range of geoscientific applications, including CO 2 sequestration, geothermal energy extraction, and in situ mineral recovery. This work presents an investigation of the change in permeability in porphyry rock samples due to blast-induced fracturing. Two samples were analysed before and after exposure to stress waves induced by the detonation of an explosive charge. Micro-computed tomography was used to image the interior of the samples at a pixel resolution of 10.3 μ m . The images were segmented into void, matrix, and grain to help quantify the differences in the rock samples. Following this, they were binarised as void or solid and the cumulant lattice Boltzmann method (LBM) was applied to simulate the flow of fluid through the connected void space. A correction required with the use of inlet and outlet reservoirs in computational permeability assessment was also proposed. Interrogation of the steady-state flow field allowed the pre- and post-loading permeability to be extracted. Conclusions were then drawn as to the effectiveness of blasting for enhancing fluid accessibility via the generation of microfractures in the rock matrix within the vicinity of a detonated charge. This paper makes contributions in three fundamental areas relating to the numerical assessment of permeability and the enhancement of fluid accessibility in low-porosity rocks. Firstly, a correction factor was proposed to account for the reservoirs commonly imposed on digitised rock samples when investigating sample permeability through numerical methods. Secondly, it validates the benefits of the LBM in handling complex geometries that would be intractable with conventional computational fluid dynamics methods that require body-fitted meshing. This is done with a novel implementation of the cumulant LBM in the open-source TCLB code. Finally, the improvement in fluid accessibility in low-permeability rock samples was shown through the assessment of multiple regions within two blasted samples. It was found that the blast-induced loading can generate extended microfractures that results in multiple orders of magnitude of permeability enhancement if the target rock possesses existing weaknesses and/or mineralisation. Human bone marrow mesenchymal stem cell derived-extracellular vesicles (HBMSC-EV) are known for their regenerative and anti-inflammatory effects in animal models of myocardial ischemia. However, it is not known whether the efficacy of the EVs can be modulated by pre-conditioning of HBMSC by exposing them to either starvation or hypoxia prior to EV collection. HBMSC-EVs were isolated following normoxia starvation (NS), normoxia non-starvation (NNS), hypoxia starvation (HS), or hypoxia non-starvation (HNS) pre-conditioning. The HBMSC-EVs were characterized by nanoparticle tracking analysis, electron microscopy, Western blot, and proteomic analysis. Comparative proteomic profiling revealed that starvation pre-conditioning led to a smaller variety of proteins expressed, with the associated lesser effect of normoxia versus hypoxia pre-conditioning. In the absence of starvation, normoxia and hypoxia pre-conditioning led to disparate HBMSC-EV proteomic profiles. HNS HBMSC-EV was found to have the greatest variety of proteins overall, with 74 unique proteins, the greatest number of redox proteins, and pathway analysis suggestive of improved angiogenic properties. Future HBMSC-EV studies in the treatment of cardiovascular disease may achieve the most therapeutic benefits from hypoxia non-starved pre-conditioned HBMSC. Mesenchymal stromal cells (MSCs) represent a promising treatment for immune-related diseases due to their diverse immunomodulatory paracrine functions. However, progress of culture-expanded MSCs is hindered by inconsistent cell function, poor localization, and insufficient retention when administered as suspended cell injections, thus placing spatiotemporal dosing constraints on therapeutic functions. To address these limitations, we introduce the combination of in vitro interferon-gamma (IFN-γ) priming, a key stimulator of MSC immunosuppressive potency, and thermoresponsive cultureware to harvest cultured MSCs as directly transplantable scaffold-free immunosuppressive cell sheets. Here, we demonstrate that MSC sheets produced with IFN-γ priming upregulate expression of immunosuppressive factors indoleamine 2,3-dioxygenase (IDO-1), interleukin-10 (IL-10), programmed death ligand-1 (PD-L1), and prostaglandin E2 (PGE2) in both dose- and duration-dependent manners. In addition, IFN-γ primed MSC sheets showed increased ability to inhibit T-cell proliferation via indirect and direct contact, specifically related to increased IDO-1 and PGE2 concentrations. Furthermore, this study's use of human clinical-grade single-cell-derived clonal bone marrow-derived MSCs, contributes to the future translatability and clinical relevancy of the produced sheets. Sesbania aculeata is a multipurpose legume crop grown primarily for green manuring in the rice-based cropping system. Besides this, it is an industrial crop and is also used as food in many parts of the world. The present work reports for optimization of various parameters (growth medium, plant growth regulators, pre-conditioning, orientation of explant, and presence of thiol compounds) affecting in vitro regeneration using mature cotyledon explants. The 5-day-old mature cotyledon explants excised from seedlings grown on Murashige and Skoog (MS) salts and Gamborg (B5) vitamins medium containing 15 μM 6-benzylaminopurine were cultured with its adaxial side facing on medium containing 2.5 μM 6-benzylaminopurine and 50 mg/L thiourea and produced multiple shoots (7 ‒ 8) in 100% cultures within 28 days. Healthy shoots were rooted on half-strength Murashige and Skoog (MS) salts and full-strength vitamins medium augmented with 2.5 μM indole-3-butyric acid.

Induced pluripotent stem cell (iPSC) derived mesenchymal stem cells (iMSCs) represent a promising source of progenitor cells for approaches in the field of bone regeneration. Bone formation is a multi-step process in which osteogenesis and angiogenesis are both involved. Many reports show that the secretome of mesenchymal stromal stem cells (MSCs) influences the microenvironment upon injury, promoting cytoprotection, angiogenesis, and tissue repair of the damaged area. However, the effects of iPSC-derived MSCs secretome on angiogenesis have seldom been investigated. In the present study, the angiogenic properties of IFN-γ pre-conditioned iMSC secretomes were analyzed. We detected a higher expression of the pro-angiogenic genes and proteins of iMSCs and their secretome under IFN-γ and hypoxic stimulation (IFN-H). Tube formation and wound healing assays revealed a higher angiogenic potential of HUVECs in the presence of IFN-γ conditioned iMSC secretome. Sprouting assays demonstrated that within Coll/HA scaffolds, HUVECs spheroids formed significantly more and longer sprouts in the presence of IFN-γ conditioned iMSC secretome. Through gene expression analyses, pro-angiogenic genes (FLT-1, KDR, MET, TIMP-1, HIF-1α, IL-8, and VCAM-1) in HUVECs showed a significant up-regulation and down-regulation of two anti-angiogenic genes (TIMP-4 and IGFBP-1) compared to the data obtained in the other groups. Our results demonstrate that the iMSC secretome, pre-conditioned under inflammatory and hypoxic conditions, induced the highest angiogenic properties of HUVECs. Mesenchymal stem/stromal cells (MSCs) therapy has been a cornerstone of regenerative medicine in humans and animals since their identification in 1968. MSCs can interact and modulate the activity of practically all cellular components of the immune response, either through cell-cell contact or paracrine secretion of soluble mediators, which makes them an attractive alternative to conventional therapies for the treatment of chronic inflammatory and immune-mediated diseases. Many of the mechanisms described as necessary for MSCs to modulate the immune/inflammatory response appear to be dependent on the animal species and source. Although there is evidence demonstrating an in vitro immunomodulatory effect of MSCs, there are disparate results between the beneficial effect of MSCs in preclinical models and their actual use in clinical diseases. This discordance might be due to cells' limited survival or impaired function in the inflammatory environment after transplantation. This limited efficacy may be due to several factors, including the small amount of MSCs inoculated, MSC administration late in the course of the disease, low MSC survival rates in vivo, cryopreservation and thawing effects, and impaired MSC potency/biological activity. Multiple physical and chemical pre-conditioning strategies can enhance the survival rate and potency of MSCs; this paper focuses on hypoxic conditions, with inflammatory cytokines, or with different pattern recognition receptor ligands. These different pre-conditioning strategies can modify MSCs metabolism, gene expression, proliferation, and survivability after transplantation.

Quantifying the mechanical behavior of skin has been foundational in applications of cosmetics, surgical techniques, forensic science, and protective clothing development. However, previous puncture studies have lacked consistent and physiological boundary conditions of skin. To determine natural skin tension, excision of in situ porcine skin resulted in significantly different diameter reduction (shrinkage) in leg (19.5 %) and abdominal skin (38.4 %) compared to flank skin (28.5 %) (p = 0.047). To examine effects of initial tension and pre-conditioning, five conditions of initial tension (as percentage of diameter increase) and pre-conditioning were tested in quasistatic puncture with a 5 mm spherical impactor using an electrohydraulic load frame and custom clamping apparatus. Samples with less than 5 % initial tension resulted in significantly greater (p = 0.011) force at failure (279.2 N) compared to samples with greater than 25 % initial tension (195.1 N). Eight pre-conditioning cycles of 15 mm displacement reduced hysteresis by 45 %. The coefficient of variance was substantially reduced for force, force normalized by cutis thickness, displacement, stiffness, and strain energy up to 46 %. Pre-conditioned samples at physiological initial tension (14–25 %) resulted in significantly greater (p = 0.03) normalized forces at failure (278.3 N/mm) compared to non-conditioned samples of the same initial tension (234.4 N/mm). Pre-conditioned samples with 14–25 % initial tension, representing physiological boundary conditions, resulted in the most appropriate failure thresholds with the least variation. For in vitro puncture studies, the magnitude of applied initial tension should be defined based on anatomical location, through a shrinkage experimentation, to match natural tension of skin. The review focuses on the neuroprotective mechanisms of therapeutic hypothermia from the standpoint of metabolic depression and genomic reprogramming of neurons that develop when brain temperature decreases.The concept of hypothermic pre-conditioning based on the development of typical nonspecific reactions for the formation of the cytoprotective phenotype of neurons due to potentially dangerous stimuli, such as ischemia, reperfusion, and hypothermia, was used to explain the effects of low temperatures. The data confirming the role of therapeutic cerebral hypothermia as a technique of selective brain exposure to mild cold for the neuroprotection and correction of temperature balance disorders are shown.The approach to therapeutic hypothermia as a hypothermic pre-conditioning allows to significantly expand the scope of its use in various procedural variants.

The study was conducted to understand the effects of a short (10 min) and a long (30 min) duration warm-up on subsequent readiness to exercise and movement during simulated rugby league match play. Methods: Using a randomised cross-over design, 13 male rugby players (age: 23.6 ± 4.1 y) completed 10- or 30-min warm-up immediately before 2 × 23 min rugby league movement simulation protocol. Results: Total distance, high- and low speed running and tympanic temperature (ES = 0.56 to 20.8) were all higher in the 30 min warm-up, with differences in relative distance and heart rate unclear (ES = -0.36 to 0.06). Differences in participants' readiness to exercise after the warm-ups were unclear (ES = 0.25). Differences between trials for movement characteristics (ES = -0.13 to -0.32), RPE (ES = -0.13 to 0.04) and B[La] after the simulation were mostly unclear, with only trivial changes in high-speed running (ES = 0.08) and a lower heart rate (ES = -0.26) between the two playing bouts after the 30 min warm-up trial. Lipopolysacharide (LPS) pre-conditioning (PC), has been shown to exert protective effects against cytotoxic effects. Therefore, we hypothesized, the tolerance produced by LPS PC will be resulted by the alterations and modifications in gene and protein expression. With reference to the results of MTT assays, AO/PI staining, and Annexin V-FITC analyses of LPS concentration (0.7815–50 μg/mL) and time-dependent (12–72 h) experiments, the pre-exposure to 3 μg/mL LPS for 12 h protected the differentiated PC12 cells against 0.75 mg/mL LPS apoptotic concentration. LPS-treated cells secreted more inflammatory cytokines like IL-1α, IL-1β, IL-2, IL-3, IL-4, IL-6, IL-17, IFN-γ, and TNF-α than LPS-PC cells. The production of inflammatory mediators ROS and NO was also higher in the LPS-induced cells compared to LPS-PC cells. Conversely, anti-inflammatory cytokines (like IL-10, IL-13, CNTF, and IL-1Ra) were upregulated in the LPS-PC cells but not in the LPS-induced cells. Meanwhile, the LPS initiated caspase-8 which in turn activates effector caspase 3/7. When the activities of caspases in the LPS-induced cells were inhibited using z-VADfmk and z-DEVDfmk, the expressions of c-MYC and Hsp70 were increased, but p53 was reduced. The potential molecules associated with protective and destructive effect was measured by RT2 Profiler PCR array to elucidate the signaling pathways and suggested inhibition NF-κB/caspase-3 signaling pathway regulates the cytoprotective genes and proto-oncogenes. In conclusion, this study provides a basis for future research to better understand the molecular mechanism underlying LPS pre-conditioning /TLR4 pre-activation and its functional role in offering cytoprotective response in neuronal environment.

Before cracking, subjecting water chestnuts to pre-conditioning is a common practice. However, the existing practice of pre-conditioning has no scientific rationale and therefore leads to low flour recovery and poor quality. Thus, the aim of the present research work was to optimize the pre-conditioning process for water chestnuts and to analyze the efficacy of optimized process as a means to increase the resistant starch content and reduce the glycemic index in water chestnut flour. Central composite rotatable design was used to determine the effect of water chestnut:water ratio, water temperature and pre-conditioning time on yield and brightness (L* value) of flour. Optimization of pre-conditioning process was done by response surface of desirability function. Regression models for both the product responses were highly significant (p 0.95). Scanning electron microscopy (SEM) showed compact, smooth and oval shaped starch granules with crystalline network in preconditioned water chestnut flour. FTIR studies also indicated higher absorbance ratio of 1047/1022 cm−1 for preconditioned water chestnut flour which further authenticated the SEM findings. Subjecting water chestnuts to optimized conditions of pre-conditioning (i.e., water chestnut:water ratio 1:1.5; water temperature 87.85̊C and pre-condition time 45 min), also increased the resistant starch content and lowered the glycemic index and glycemic load of the water chestnut flour.

The CBC and the transcript abundance of cytokines and their receptors expressed in leukocytes were measured from calves at two early timepoints, and again after diagnosis with bovine respiratory disease (BRD). Pre-conditioning by hydraulic fracturing is a vital work in block caving mining of hard-rock metal mine, and breakdown pressure is one of the important parameters in rock hydraulic fracturing process, which determines the equipment selection and job-parameter control. Based on numerical simulation method, a pre-holed cylindrical hard rock sample was built for simulation of hydraulic fracturing process to investigate the effect of stress condition, rock properties and water injection rate on breakdown pressure. The results show that the breakdown pressure is increased with the confining stress, and the breakdown pressure is 2.28 times confining stress based on rock tension strength. The variation of rock tension strength affects the breakdown pressure and a positive linear relation exists. Pre-formed notch could obviously reduce the breakdown pressure and control the orientation of hydraulic fracture. And the borehole diameter has a reverse influence on rock breakdown pressure. Fibrosis is driven by a misdirected cell response causing the overproduction of extracellular matrix and tissue dysfunction. Numerous pharmacological strategies have attempted to prevent fibrosis but have attained limited efficacy with some detrimental side effects. While stem cell treatments have provided more encouraging results, they have exhibited high variability and have not always improved tissue function. To enhance stem cell efficacy, we evaluated whether mechanical memory could direct cell response. We hypothesized that mechanically pre-conditioning on a soft matrix (soft priming) will delay adipose-derived stem cell (ASC) transition to a pro-fibrotic phenotype, expanding their regenerative potential, and improving healing in a complex tissue environment. Primary ASCs isolated from rat and human subcutaneous fat exhibited mechanical memory, demonstrated by a delayed cell response to stiffness following two weeks of soft priming including decreased cell area, actin coherency, and extracellular matrix production compared to cells on stiff substrates. Soft primed ASCs injected into our rat model of post-traumatic elbow contracture decreased histological evidence of anterior capsule fibrosis and increased elbow range-of-motion when evaluated by joint mechanics. These findings suggest that exploiting mechanical memory by strategically controlling the culture environment during cell expansion may improve the efficacy of stem cell-based therapies targeting fibrosis. This study explores whether an on-water kayak-specific resistance exercise (KSRE) performed during the warm-up period can induce postactivation potentiation (PAP) to improve subsequent kayak-sprint performance. The KSRE involves stringing several tennis balls around the boat's hull to induce drag while paddling. In a laboratory setting, 10 well-trained male (N = 6) and female (N = 4) kayakers performed three repetitions of the single-arm seated-row (SASR) at 91% one-repetition maximum to induce PAP and were assessed for their peak power output while performing SASR on the Keiser machine at two-minute intervals up to 16 min. Each individual's latency period for the onset of PAP during this exercise was recorded. In separate sessions, athletes performed in random, a 30 m kayak-sprint in a swimming pool setting after either control (no pre-performance loading) or experimental (post-PAP induction via the KSRE taking into account the individual' onset of PAP latencies period obtained in the laboratory) conditions. Criterion measures of boat velocities of the third and fourth stroke from the start and 30 m sprint times, assessed using video, were not significantly different between control and experimental conditions. The translation of increased power output elicited via PAP using land-based resistance exercises into water-based resistance exercises to improve kayak-sprint performance remains a challenge. Land salinization, resulting from the ongoing climate change phenomena, is having an increasing impact on coastal ecosystems like salt marshes. Although halophyte species can live and thrive in high salinities, they experience differences in their salt tolerance range, being this a determining factor in the plant distribution and frequency throughout marshes. Furthermore, intraspecific variation to NaCl response is observed in high-ranging halophyte species at a population level. The present study aims to determine if the environmental history, namely heavy metal pre-conditioning, can have a meaningful influence on salinity tolerance mechanisms of Spartina patens, a highly disperse grass invader in the Mediterranean marshes. For this purpose, individuals from pristine and heavy metal contaminated marsh populations were exposed to a high-ranging salinity gradient, and their intraspecific biophysical and biochemical feedbacks were analyzed. When comparing the tolerance mechanisms of both populations, S. patens from the contaminated marsh appeared to be more resilient and tolerant to salt stress, this was particularly present at the high salinities. Consequently, as the salinity increases in the environment, the heavy metal contaminated marsh may experience a more resilient and better adapted S. patens community. Therefore, the heavy metal pre-conditioning of salt mash populations appears to be able to create intraspecific physiological variations at the population level that can have a great influence on marsh plant distribution outcome.

These variations are not explained by currently used algorithms. In this paper, we present a new hypothesis: individual metabolic processes, through the oxygen window (OW) or Inherent Unsaturation of tissues, modulate the presence and volume of static metabolic bubbles (SMB) that in turn act as precursors of circulating VGE after a dive.MethodsWe derive a coherent system of assumptions to describe static gas bubbles, located on the vessel endothelium at hydrophobic sites, that would be activated during decompression and become the source of VGE. We first refer to the OW and show that it creates a local tissue unsaturation that can generate and stabilize static gas phases in the diver at the surface. We then use Non-extensive thermodynamics to derive an equilibrium equation that avoids any geometrical description. The final equation links the SMB volume directly to the metabolism.Results and DiscussionOur model introduces a stable population of small gas pockets of an intermediate size between the nanobubbles nucleating on the active sites and the VGE detected in the venous blood. The resulting equation, when checked against our own previously published data and the relevant scientific literature, supports both individual variation and the induced differences observed in pre-conditioning experiments. It also explains the variability in VGE counts based on age, fitness, type and frequency of physical activities. This study presents an anhydrobiotic engineering approach aiming at conferring a high degree of desiccation tolerance to the Gram-negative endophyte Kosakonia radicincitans. In particular, pre-conditioning of bacteria under high salinities provides a remarkable positive influence on drying survival. The endophytic bacteria accumulate exogenous hydroxyectoine > 500 µmol g−1 dry weight cells exerted by osmotic stress at 4% NaCl. Microfermentation research demonstrated that hydroxyectoine provides positive effects on reducing the lag phase duration and alleviates the dissolved oxygen consumption under high salinity conditions. Beyond the amassing of hydroxyectoine, this work provides evidence supporting the notion that hydroxyectoine can produce significant changes in the endogenous bacterial metabolome during the exponential growth phase at high-osmolarity. Metabolome changes include alterations on tricarboxylic acid cycle, novo-synthesis of specific intracellular metabolites such as mannitol, myo-inositol and trehalose, and fold changes on amino acids such as l-leucine, l-asparagine, l-serine, l-methionine and l-proline. The significant fold change of l-aspartate suggests a potential acidic proteome at high-osmolarity environments, extending the knowledge of salt-stressed bacterial endophytes. Thus, these findings place the metabolic salt stress response and the hydroxyectoine accumulation by K. radicincitans into a physiological context, paving the way into the interaction between cellular phenotype associated with salt stress tolerance and drying survival capacity of Gram-negative endophytes. It has recently been revealed that during the aorta-clamped period, D-Ala2, D-Leu5-Enkephalin (DADLE) infusion can protect the spinal cord against ischemia and reperfusion (I/R) injury. However, the protective effects of DADLE administration prior to ischemia or at the time of early reperfusion have not yet been investigated. Drug pre- or post-conditioning can serve as a more valuable clinical strategy. Therefore, the present study was designed to investigate the neuroprotective effect of DADLE infusion at different time intervals in order to determine the optimum time point for ischemic spinal cord protection. A total of 40 New Zealand white rabbits were randomly divided into 5 groups: Sham-operated (Sham), normal saline pre-conditioning (NS), DADLE per-conditioning (Dper), DADLE pre-conditioning (Dpre) and DADLE post-conditioning (Dpost). All animals were subjected to spinal cord ischemia for 30 min followed by 48 h reperfusion. Hind limb motor functions were assessed according to the Tarlov criterion when the animals regained consciousness, 6, 24 and 48 h after reperfusion. Histological analysis and the number of viable α-motor neurons were also used to assess the extent of spinal cord injury. Compared with the NS group, the Tarlov scores and the number of normal neurons were significantly higher in the Dper group (P<0.05), which were consistent with the results of a previous study. In addition, the paraplegia rate and loss of normal motor neurons were lower in the DADLE per- and post-conditioning groups compared with the DADLE pre-conditioning; however, these were not statistically significant. DADLE 0.05 mg/kg administration at three time points all mitigated normal motor neuron injury in the anterior horn and decreased the paraplegia rates in rabbits. Introduction: Today, the production of biological nanoparticles has expanded significantly in different industries. Although their side effects have not yet been fully evaluated on brain tissue. In addition, by applying a stressor stimulus less than the threshold for injury to the tissue (pre-preparation), such as physical activity, it can increase the resistance of the tissue to more powerful stimuli. Therefore, the purpose of this study was to investigate the effect of high dose doses of biological nanoparticles with two different types of training prediction on structural changes in brain tissue of male Wistar rats. Methods: A total of 30 male Wistar rats were divided into 6 groups including healthy control, silver biological nanoparticles, aerobic exercise protocols, training protocols 2 (anaerobes), biological biological nanoparticles + exercise protocols 1, biological nanoparticles Silver + Practice Protocol 2 was done. Intraperitoneal injection of silver nanoparticles after 10 weeks of aerobic and anaerobic treatments was performed on 10% of body weight of each rat in 5 times. After 48 hours after the last injection, rats were indifferent and sample was taken. The specimens were then photographed and stained with Hematoxylin-Eosin stained with optical microscopy. Results: The results of this study showed that aerobic training and anaerobic exercise training had a significant effect on the distance traveled in the progressive endurance test on rodent treadmill (P=0.000) and body weight (P=0.000). Intraperitoneal injection of high-dose silver nanoparticles also caused inflammation and mild inflammation of brain tissue in untreated rats, and the white matter and gray matter of nerve cells did not change. On the other hand, it was reduced trained groups and in the aerobic training group it was more significant than anaerobic. Conclusion: Pre-conditioning of aerobic training can be effective in reducing the degree of brain tissue damage caused by the injection of silver biologic nanoparticles. [ABSTRACT FROM AUTHOR]

The present work focused on the utilisation of jackfruit flesh flour mixed with a legume (pigeon pea) and cereal (rice) flour in different proportions to produce extrudates using twinscrew extrusion cooking. Preliminary trials were carried out for selection of variables, and extrusion cooking was carried out using a twin-screw extruder at extrusion parameters namely temperature: 130°C for two different heating zones, die diameter: 4 mm, and screw speed: 270 rpm. In the finished product, the ash content increased from 1.86 to 2.93% with a concomitant reduction in fat from 1.38 to 0.02% and protein content from 13.50 to 11.56% with the addition of jackfruit flesh flour in the extrudates. The hardness of extrudates increased while a reduction in expansion ratio was observed with the addition of jackfruit flesh flour. Microstructural analysis indicated the absence of uniform structure of starch granules in all the extrudates. Sensory evaluation revealed that extruded sample C (containing 5% jackfruit flesh flour) had the highest overall acceptability. The results indicated that composite flour (jackfruit flesh, rice and pigeon pea) in the ratios of 5:47.5:47.5 could be used to produce quality extrudates. [ABSTRACT FROM AUTHOR]

Purpose: Epigallocatechin 3-gallate (EGCG), a green tea polyphenol, has been shown to have anti-oxidant and anti-inflammatory effects in vitro and in vivo. The aim of this study was to investigate the effects and mechanism of EGCG on isolated pancreatic islets as pre-conditioning for pancreatic islet transplantation. Methods: The pancreatic islets were divided into two groups: an islet culture medium group (control) and an islet culture medium with EGCG (100 µM) group. We investigated the islet viability, Nrf2 expression, reactive oxygen species (ROS) production, and heme oxygenase-1 (HO-1) mRNA. Five hundred islet equivalents after 12 h of culture for the EGCG 100 µM and control group were transplanted under the kidney capsule of streptozotocin-induced diabetic ICR mice. Results: The cell viability and insulin secretion ability in the EGCG group were preserved, and the nuclear translocation of Nrf2 was increased in the EGCG group (p < 0.01). While the HO-1 mRNA levels were also higher in the EGCG group than in the control group (p < 0.05), the ROS production was lower (p < 0.01). An in vivo functional assessment showed that the blood glucose level had decreased in the EGCG group after transplantation (p < 0.01). The relationship between mycorrhiza functioning and composition of arbuscular mycorrhizal (AM) fungal communities is an important but experimentally still rather little explored topic. The main aim of this study was thus to link magnitude of plant benefits from AM symbiosis in different abiotic contexts with quantitative changes in AM fungal community composition. A synthetic AM fungal community inoculated to the model host plant Medicago truncatula was exposed to four different abiotic contexts, namely drought, elevated phosphorus availability, and shading, as compared to standard cultivation conditions, for two cultivation cycles. Growth and phosphorus uptake of the host plants was evaluated along with the quantitative composition of the synthetic AM fungal community. Abiotic context consistently influenced mycorrhiza functioning in terms of plant benefits, and the effects were clearly linked to the P requirement of non-inoculated control plants. In contrast, the abiotic context only had a small and transient effect on the quantitative AM fungal community composition. Our findings suggest no relationship between the degree of mutualism in AM symbiosis and the relative abundances of AM fungal species in communities in our simplified model system. The observed progressive dominance of one AM fungal species indicates an important role of different growth rates of AM fungal species for the establishment of AM fungal communities in simplified systems such as agroecosystems. An investigation studied the impact of spectral composition on plant responses to heat stress. Petunia × hybrida seedlings were grown under nine different light qualities, including the modifying and constant compositions of blue and red LEDs light, as well as Fluorescent and white LED lamps. After 28 days, the seedlings were exposed to hyperthermia (40 °C / 35 °C, 4 days). The results showed that the highest survival rate (100 %), and the lowest leaf chlorotic area (14 %) were observed among seedlings grown under modifying spectral compositions with a gradual increase in red light, which also showed the highest change rate of increased Chl a/ b ratio (262 %) and lowest change rate of increased total Chl/ Car ratio (45 %). Furthermore, seedlings grown under a gradual increase in red light showed the highest relative water content (82 %) and the lowest change rate of root water content (1.2 %). Moreover, the highest proline and lignin contents, along with the lowest electrolyte leakage, 7.22 µg g−1 FW, 104 mg g−1 FW, and 25 %, respectively, were observed in seedlings exposed to a gradual increase in red light after heat stress compared to other lighting treatments. Also, after heat stress, seedlings grown under a gradual increase in red light showed the highest contents of studied enzymatic antioxidants. Our investigation highlighted a link between spectral composition during the seedling stage and heat stress tolerance of examined plants. Background: Liver transplantation is the second most common transplant procedure in the United States. A leading cause of post-transplantation organ dysfunction is I/R injury. During I/R injury, the serine/threonine kinase Akt is activated, stimulating downstream mediators to promote cellular survival. Due to the cellular effects of Akt, therapeutic manipulation of the Akt pathway can help reduce cellular damage during hepatic I/R that occurs during liver transplantation. Objective: A full description of therapeutic options available that target Akt to reduce hepatic I/R injury has not been addressed within the literature. The purpose of this review is to illuminate advances in the manipulation of Akt that can be used to therapeutically target I/R injury in the liver. Methods: An in depth literature review was performed using the Scopus and PubMed databases. A total of 75 published articles were utilized for this manuscript. Terminology searched includes a combination of “hepatic ischemia/reperfusion injury”, “Akt/PKB”, “preconditioning” and “postconditioning.” Results: Four principal methods that reduce I/R injury include hepatic pre- and postconditioning, pharmacological intervention and future miRNA/gene therapy. Discussed therapies used serum alanine aminotransferase levels, liver histology and phosphorylation of downstream mediators to confirm the Akt protective effect. Conclusion: The activation of Akt from the reviewed therapies has resulted in predictable reduction in hepatocyte damage using the previously mentioned measurements. In a clinical setting, these therapies could potentially be used in combination to achieve better outcomes in hepatic transplant patients. Evidence supporting reduced I/R injury through Akt activation warrants further studies in human clinical trials.

Remote ischemic pre-conditioning (rIPC) has emerged as a potential mechanism to reduce ischemia-reperfusion injury. Clinical data, however, have been mixed, and its physiological basis remains unclear, although it appears to involve release of circulating factor(s) and/or neural pathways. Here, the authors demonstrate that adenosine receptor activation is an important step in initiating human pre-conditioning; that pre-conditioning liberates circulating cardioprotective factor(s); and that exogenous adenosine infusion is able to recapitulate release of this factor. However, blockade of adenosine receptors in ischemic tissue does not block the protection afforded by pre-conditioning. These data have important implications for defining the physiology of human pre-conditioning and its translation to future clinical trials.
