ABSTRACT: This study aimed to determine the effect of circulating progesterone (P4) concentrations produced by a corpus luteum (CL) or released by an intravaginal P4 implant (IPI) on GnRH-induced LH release, ovulatory response, and subsequent CL development, after treatment with 100 μg of gonadorelin acetate (GnRH challenge). Nonlactating multiparous Holstein cows were synchronized and GnRH was used to induce ovulation (d −7). Over 4 replicates, cows that ovulated (n = 87) were randomly assigned to a 2 × 2 factorial arrangement (presence or absence of CL and insertion or not of an IPI at GnRH challenge), creating 4 groups: CL_IPI, CL_NoIPI, NoCL_IPI, and NoCL_NoIPI. On d −1.5, NoCL_IPI and NoCL_NoIPI received 2 doses of 0.53 mg of cloprostenol sodium (PGF2α), 24 h apart to regress CL. On d 0, cows were treated with 100 μg of GnRH and, simultaneously, cows from IPI groups received a 2-g IPI maintained for the next 14 d. Diameter of dominant follicle, ovulatory response, and subsequent CL volume were assessed by ultrasonography on d −1.5, 0, 2, 7, and 14. Blood samples were collected on d −1.5, 0, 1, 2, 3, 5, 7, and 14 for analysis of circulating P4 and at 0, 1, 2, 4, and 6 h after GnRH challenge for analysis of circulating LH. In a subset of cows (n = 34), the development of the new CL was evaluated daily, from d 5 to 14. The presence of CL at the time of GnRH challenge affected the LH peak and ovulatory response (CL: 5.3 ng/mL and 58.1%; NoCL: 13.2 ng/mL and 95.5%, respectively). However, despite producing a rapid increase in circulating P4, IPI insertion did not affect LH concentration or ovulation. Regardless of group, ovulatory response was positively correlated with LH peak and negatively correlated with circulating P4 on d 0. Moreover, new CL development and function were negatively affected by the presence of CL and by the IPI insertion. In summary, circulating P4 produced by a CL exerted a suppressive effect on GnRH-induced LH release and subsequent ovulation of a 7-d-old dominant follicle, whereas the IPI insertion at the time of GnRH had no effect on LH concentration or ovulation. Finally, elevated circulating P4, either from CL or exogenously released by the IPI, compromised the development and function of the new CL, inducing short cycles in cows without CL at the time of GnRH treatment.

The fertility response following induction of follicular wave emergence by follicle ablation combined withprogesterone based ovulation synchronization in postpartum anestrous buffaloes was assessed. Postpartum anestrousbuffaloes (n=24) with absence of overt estrus signs categorized as treatment group (Group 1; n=15) and controlgroup (Group 2; n=9). Group 1 was subjected to transvaginal ultrasound-guided follicle ablation on a random dayof the estrous cycle (Day=0) and insertion of CIDR. Seven days later, CIDR was removed and prostaglandin F2αanalogue was given intramuscularly. GnRH analogue was injected on day 9.5 and fixed-time AI was done 12 h and24 h later. Ultrasonography was performed to follow ovarian structures on different occasions and on day 38 postovulationfor detecting pregnancy. Group 2 did not receive any treatment however, estrus detection in this groupwas done daily by bull parading and visual monitoring followed by AI if found in standing estrus. In treatmentgroup, the follicular wave emergence occurred by 24 h following ablation in 60% buffaloes and in 40% remaininganimals by 48 h. The estrus exhibition was 100 % in group 1 versus 22.2 % in group 2. Mean interval from CIDRremoval to ovulation was 81.2±3.08 h and from wave emergence to ovulation was 9.92±0.18 days. The first servicepregnancy rate in treatment group was higher than control (66.67% vs 22.2%). The induction of follicular waveemergence through ultrasound guided follicle ablation and progesterone based ovulation synchronization can beused to improve pregnancy rates in anestrous buffaloes.

In this study, the second use of an intravaginal progesterone-releasing device or controlled intravaginal drug release device (CIDR) was evaluated. After a first use of 11 days, the CIDR was again used for either nine or 12 days with 200 or 300 IU equine chorionic gonadotrophin (eCG) being injected on its removal. Sixty-four ewes were randomly distributed to four treatments (n=16/group): CIDR9+eCG200, CIDR9+eCG300, CIDR12+eCG200, and CIDR12+eCG300. The eCG was administered intramuscularly on withdrawal of the device. Thus, the experiment was a completely randomized design with a 2×2 factorial arrangement of treatments. Oestrus presentation did not differ between treatments (P =0.29). However, with the dose of 200 IU of eCG, oestrus presentation tended to increase (P =0.08). The onset and duration of oestrus, percentage of gestation, and return to oestrus did not differ between treatments (P >0.05). Progesterone concentration in serum was greater (P < 0.05) in ewes treated with CIDR12+eCG300. Prolificacy was greatest (1.44) with the CIDR12+eCG300 treatment and was different (P = 0.001) from the treatments CIDR9+eCG200 (1.21) and CIDR9+eCG300 (1.20), but not from the CIDR12+eCG200 treated ewes (1.31). The CIDR12+eCG300 treatement produced the highest percentage of twin births (45.8%) (P =0.001). Leaving the device in place for 12 days increased (P =0.001) the incidence of twin births. Use of the CIDR for a second time synchronized oestrus in ewes successfully with better fertility being obtained when the device was left in place for 12 days, and 300 IU of eCG was injected on its removal. Follicular wave synchronization (FWS) before ovum pick-up (OPU) is one of the strategies used to improve the efficiency of in vitro embryo production (IVP). This study aimed to evaluate the effect of FWS on the total follicular number, cumulus-oocyte complex (COC) recovery, and in vitro embryo development in Angus cows (n = 33) subjected to OPU with 14-day intersession intervals. Additionally, it was also evaluated the presence of carryover effects given the short intersession interval used. The experiment was run as a 2-treatment (FWS vs. Control) x 2-period (1 vs. 2) crossover design. Animals in the FWS group received an intravaginal progesterone implant (1gr), estradiol benzoate (2 mg), and D-cloprostenol (150 μg) on day 0 and the OPU was performed on day 5. Control group animals did not receive any hormone treatment. The FWS increased the number of 6–10 mm follicles (P = 0.05), but it decreased the COC recovery rate (P < 0.01). The FWS did not affect the total or frozen embryo numbers (P = 0.49 and P = 0.17; respectively), but it increased the total blastocyst cell number (P < 0.01). A carryover effect was found on the total and < 6 mm follicles number (P = 0.10 and P < 0.01; respectively), and on the regular, atretic, viable, and total number of COC (P = 0.01, P = 0.08, P = 0.02 and P < 0.01; respectively). We concluded that the FWS increased the quality of embryos after OPU with 14-day intersession intervals in Angus cows and that this kind of OPU/IVP scheme enabled the existence of a carryover effect, especially on the follicle number and COC morphology. The objectives of this study were 1) to monitor corpus luteum (CL) dynamics after two different protocols of ovulation induction in prepubertal Nellore heifers, and 2) to determine differences in luteal function. Fifty-seven heifers (weight 289.61 ± 32.28 kg, BCS 5.66 ± 0.65, age 17.47 ± 0.81 months) were divided into two groups: GP4+GnRH received a progesterone (P4) device of 3rd use for 10 days, followed by the administration of 0.02 mg buserelin acetate (GnRH) 48 h after removal of the device, and GGnRH received only GnRH. The CLs formed were monitored by ultrasonography every 2 days until their functional regression (decrease in the color Doppler signal and serum P4 concentration < 1 ng/mL), determining their diameter and area, numerical pixel value (NPV), pixel heterogeneity, and vascularization percentage. The peak systolic velocity, end diastolic velocity, resistivity index and pulsatility index (PI) of the ovarian artery and serum P4 concentration were also measured. A lifespan of the CL of more than 16 days was classified as normal-function and of less than 16 days as premature regression. The variables were compared between treatments, CL categories (normal-functional, prematurely regressed or non-functional), days of evaluation, and their interactions using the MIXED procedure of the SAS program (p ≤ 0.05). Three animals of each group (6/57 = 11%) did not respond to treatment, corresponding to an ovulation rate of 89%. There was a higher percentage of normal-function CLs in GP4+GnRH (81%) and a higher percentage of non-functional CLs in GGnRH (52%; P4 concentration < 1 ng/mL in all assessments). Normal-function CLs exhibited a greater area, vascularization percentage and P4 concentration than prematurely regressed and non-functional CLs. Lower diameter, area, NPV and P4 concentration were observed for non-functional CLs, but there was no difference in vascularization percentage compared to prematurely regressed CLs. Progesterone concentration was efficient in diagnosing CL function and was positively correlated with CL area (r = 0.62; p < 0.001) and vascularization percentage (r = 0.38; p < 0.001). Diameter and PI were important for the early diagnosis of non-functional and prematurely regressed CLs, respectively. In conclusion, luteal function differed for the first CL that develops after ovulation induction in prepubertal heifers. Ultrasonographic parameters (diameter, area, NPV, vascularization percentage, and PI) can be used to predict CL function. [ABSTRACT FROM AUTHOR]

Abstract: Objectives were to compare circulating progesterone (P4) profile and pregnancies per AI (P/AI) of two commercial intravaginal P4 devices (PRID-Delta® vs CIDR®). In Experiment 1, ovariectomized dairy cows (PRID-Delta, n =6 vs CIDR, n =6) were sampled throughout 7 days to measure circulating P4. In Experiment 2 (PRID-Delta, n =399 vs CIDR, n =375), cows were assigned to treatments, as follows: D0, an intravaginal P4 device containing 1.38g of P4 (CIDR) or 1.55g of P4 (PRID-Delta); D6: 25mg PGF2α (Dinoprost) and P4 devices were removed 24h later. Insemination was performed at 56h after P4 removal. Cows visually detected in estrus between days 18 and 24 after 1st synchronized AI were re-inseminated. PRID-Delta produced greater circulating P4 compared to CIDR, particularly within 4 days after insertion (P <0.01). The logistic regression analysis indicated a tendency for improved P/AI at 1st AI in PRID-Delta cows compared to CIDR (36% vs 31%, P =0.10). More cows were detected in estrus in the following cycle nearly 21d after 1st AI when treated with PRID-Delta (28% vs 16%), but P/AI in the returning-natural estrus breedings did not differ (PRID-Delta=56% vs CIDR=55%; P =0.91). As a result, final cumulative P/AI was greater in cows receiving PRID-Delta (46% vs 37%, P =0.02). These results indicate that PRID-Delta seem to maintain greater circulating P4 levels as compared to CIDR in non-lactating dairy cows. This might explain potential benefits in fertility of dairy cows found in Experiment 2. Underlying physiological consequence of greater circulating P4 during synchronization programs in lactating cows in terms of oocyte quality and other reproductive structures warrants further investigation.
