La membrane biologique qui enveloppe les globules gras du lait (ou milk fat globule membrane, MFGM) constitue une interface protectrice pour le coeur de triglycérides et apporte des lipides et des protéines bioactifs au consommateur, en particulier le nouveau-né allaité. Cependant, l’assemblage des lipides polaires qui constituent la MFGM ainsi que ses propriétés biophysiques sont encore mal connus, notamment en rapport avec la présence de cholestérol ou avec l’historique thermique auquel le lait peut être soumis pendant sa mise en oeuvre en industrie alimentaire ou pendant sa pasteurisation et son stockage en lactarium.En particulier, les lipides polaires des laits de mammifères possèdent une proportion significative d’espèces à haute température de transition de phase gel/liquide désordonné, telles que la sphingomyéline (~30% w/w des lipides polaires totaux; température de fusion = 34°C – Murthy et al., 2015). Ces espèces seraient responsables de la formation de domaines lipidiques à la surface des globules gras du lait (Lopez et al., 2010 ; Guyomarc’h et al., 2014). L’objectif de ces travaux était donc de décrire l’organisation latérale des lipides polaires laitiers lorsqu’ils sont assemblés sous la forme d’une bicouche hydratée, modèle de la membrane du globule gras du lait. Dans ce contexte, la microscopie à force atomique (AFM) se révèle un instrument performant, permettant de quantifier des hétérogénéités structurales et mécaniques au sein des membranes lipidiques, avec une résolution nanométrique et dans des conditions physiologiquement pertinentes.

International audience; The phenomenon of protein superstructural polymorphism has become the subject of increased research activity. Besides the relevance to explain the existence of multiple prion strains, such activity is partly driven by the recent finding that in many age-related neurodegenerative diseases highly ordered self-associated forms of peptides and proteins might be the structural basis of prion-like processes and strains giving rise to different disease phenotypes. Biophysical studies of prion strains have been hindered by a lack of tools to characterize inherently noncrystalline, heterogeneous and insoluble proteins. A description of the pressure response of prion quaternary structures might change this picture. This is because applying pressure induces quaternary structural changes of PrP, such as misfolding and self-assembly. From the thermodynamics of these processes, structural features in terms of associated volume changes can then be deduced. We suggest that conformation-enciphered prion strains can be distinguished in terms of voids in the interfaces of the constituting PrP protomers and thus in their volumetric properties.

Many proteins are able to self-assembly into well-defined supramolecular structures. We have previously shown that two food globular proteins with highly homologous tertiary structures and opposite electric charges, i.e. lysozyme and a-lactalbumin, interact under specific conditions of temperature and pH and generate spherical microparticles. The objective of this study was to investigate the stability of the lysozyme - a-lactalbumin microparticles during lyophilisation, one example of food processes. Solutions of protein microparticles without and with addition of protective molecules such as sucrose or matlodextrin were freeze-dried under conservative conditions in a pilot plant and samples were removed at each step of the process: after freezing, sublimation and desorption. The morphology and the particle size distribution of the protein microparticles were investigated by direct optical observation under a microscope and by using a Coulter counter. The freezing (and thawing) appeared to be the most damaging steps with the formation of clusters or agglomerates of microparticles. Addition of sucrose or maltodextrin sharply reduced the formation of particles agglomerates after freezing by limiting the cryoconcentration of the particles and trapping them in an amorphous matrix. Maltodextrin seemed to be a more efficient protective molecule than sucrose for preserving the stability of the protein microparticles during the freeze-drying process.

The paper deals with the estimation of biophysical variables in the frame of a precision farming project. N-trials were performed on wheat crops for 2 years, providing a calibration and a validation dataset. Airborne radiometric measurements were acquired over the trials. Biophysical variables were retrieved from inversion of the radiometric signal through radiative transfer models. To minimize the number of solutions and constrain the inversion, a priori knowledge about the parameters was introduced in the cost function. Using this technique, green leaf area index and leaf chlorophyll content were estimated with a 10% accuracy.

[Departement_IRSTEA]GMA [TR1_IRSTEA]11 - VERSEAU / TRANSFEAU; International audience; The Alpilles-ReSeDA program was initiated to develop and test methods for interpreting remote sensing data that could lead to a better evaluation of soil and vegetation processes. This article presents the experiment that was set up in order to acquire the necessary data to achieve this goal. Intensive measurements were performed for almost one year over a small agricultural region in the South of France (20 kilometers square). To capture the main processes controlling land-atmosphere exchanges, the local climate was fully characterized, and surface energy fluxes, vegetation biomass, vegetation structure, soil moisture profiles, surface soil moisture, surface temperature and soil temperature were monitored. Additional plant physiological measurements and a full characterization of physical soil parameters were also carried out. After presenting the different types of measurements, examples are given in order to illustrate the variability of soils and plant processes in the area in response to the experienced climate.; Le programme Alpilles-ReSeDA a été mis en place pour développer et tester des méthodes permettant une meilleure utilisation des données de télédétection pour le suivi du fonctionnement des sols et des cultures. Cet article présente l'expérimentation qui a été réalisée pour acquérir un jeu de données permettant cette analyse. Des mesures intensives ont été réalisées pendant presque une année sur une petite région agricole du Sud de la France (20 kilomètres carrés). De façon à suivre l'ensemble des processus contrôlant les échanges surface-atmosphère, l'ensemble des paramètres climatiques locaux ont été mesurés, ainsi que les flux d'énergie de surface, les caractéristiques de structure de la végétation et du sol l'humidité et les températures du sol, la température de surface. Des mesures des paramètres physiologiques des plantes et des caractéristiques physiques des sols ont également été entreprises. Après avoir présenté les différents types de mesures réalisées, des exemples présentant la variabilité des couverts végétaux et des sols dans la zone d'étude sont présentés.
